La secuencia transportadora puede unirse por ejemplo (por ejemplo directamente) de forma N-terminal o (por ejemplo, directamente) de forma C-terminal a la secuencia de (poli)-péptidos inhibidora del inhibidor de JNK. La secuencia transportadora y la secuencia de (poli)péptidos inhibidora también puede estar separada, por ejemplo, puede estar separada por secuencias intermedias. Se contempla también que la secuencia transportadora pueda disponerse por completo en cualquier lado de la molécula inhibidora de JNK que la
45 secuencia de (poli)-péptidos inhibidora, en particular si el inhibidor de JNK es una molécula más compleja (por ejemplo que comprende varios dominios, sea un conjugado multimérico, etc.). También se contempla que la secuencia transportadora y la secuencia de (poli)-péptidos inhibidora pueden superponerse siempre y cuando la actividad inhibidora de JNK se mantenga. Ejemplos de esta superposición se dan posteriormente.
Las secuencias transportadoras para su uso con el inhibidor de JNK de la presente invención se pueden seleccionar de, sin estar limitadas a éstas, secuencias transportadoras derivadas de HIV TAT (HIV), por ejemplo proteínas nativas tales como la proteína TAT (por ejemplo como la descrita en la patente US Nº 5.804.604, col. 2, l.64 -col. 118, l. 25, en particular col. 3, l. 23 -38 y en la US Nº 5.674.980, col. 2, l.64 -col. 116, l. 36, en particular col. 3, l. 23 -38), HSV VP22 (Herpes simplex) (descrita por ejemplo en WO 97/05265; Elliott y O’Hare, 55 Cell 88:223-233 (1997)), proteínas no virales (Jackson et al, Proc. Natl. Acad. Sci. US 89:10691-10695 (1992)), secuencias transportadoras derivadas de Antennapedia, particularmente de Drosophila antennapedia (por ejemplo, la secuencia portadora de antennapedia de la misma), FGF, lactoferrina, etc., o derivados de péptidos básicos, por ejemplo péptidos que tienen una longitud de 5 a 15 aminoácidos, de preferencia 10 a 12 aminoácidos y que comprendan al menos 80%, muy preferiblemente 85% o incluso 90% aminoácidos básicos, tales como arginina, lisina y/o histidina, o se pueden seleccionar de, por ejemplo, secuencias de péptidos ricas en arginina, tales como RRRRRRRRR (Rg; SEQ ID NO: 152), RRRRRRRR (R8; SEQ ID NO: 153), RRRRRRR (R7; SEQ ID NO: 154), RRRRRR (R6, SEQ ID NO: 155), RRRRR (R5, SEQ ID NO: 156), etc., de VP22, de proteínas o péptidos PTD-4, de RGD-K16, de proteínas o péptidos PEPT1/2 o PEPT1/2, de proteínas o péptidos SynB3 o SynB3, de inhibidores de PC, de proteínas o péptidos derivados de P21 o de proteínas o péptidos

Ejemplos de secuencias transportadoras para su uso en el inhibidor de JNK de la presente invención son en particular, sin limitarse a las mismas, secuencias transportadoras básicas derivadas de la proteína HIV-1 TAT. Preferentemente, la secuencia transportadora básica de la proteína TAT de VIH-1 puede incluir secuencias de 5 la proteína TAT del virus de la inmunodeficiencia humana VIH-1, por ejemplo, como se describe en, por ejemplo, patentes US Nº 5.804.604 y 5.674.980. En este contexto, la proteína HIV-1 TAT de longitud completa tiene 86 residuos de aminoácido codificados por dos exones del gen HIV TAT. Los aminoácidos de TAT 1-72 son codificados por el exón 1, mientras que los aminoácidos 73-86 son codificados por el exón 2. La proteína TAT de longitud completa se caracteriza por una región básica que contiene dos lisinas y seis argininas (aminoácidos 49-57) y una región rica en cisteína que contiene siete residuos de cisteína (aminoácidos 22-37). Al igual que en otras proteínas reguladoras, la región N-terminal puede estar implicada en la protección frente a proteasas intracelulares (Bachmair, A. et al., Cell 56: 1019-1032 (1989)). Las secuencias transportadoras TAT a emplear preferentemente en el inhibidor de JNK de la presente invención se caracterizan por la presencia de la secuencia de aminoácidos de la región básica de TAT (aminoácidos 49-57 de la proteína TAT de origen natural); la ausencia de la secuencia de aminoácidos de la región rica en cisteína de TAT (aminoácidos 22-36 de la proteína TAT de origen natural) y la ausencia del dominio carboxi-terminal codificado por el exón 2 de TAT (aminoácidos 73-86 de la proteína TAT de origen natural). Muy preferiblemente, la secuencia transportadora en el inhibidor de JNK de la presente invención se puede seleccionar de una secuencia de aminoácidos que contenga los residuos 48-57 ó 49 a 57 de TAT o variantes de los mismos.
25 Preferentemente, la secuencia transportadora en un inhibidor de JNK dado de la presente invención tiene también D-aminoácidos, por ejemplo para mejorar la estabilidad frente a proteasas. Se prefieren en particular las secuencias transportadoras que exhiben un orden específico de D-y L-aminoácidos alternantes. Este orden de D-y L-aminoácidos alternantes (el motivo) puede seguir – sin estar limitado a esto – el patrón de cualquiera de las SEQ ID NO: 28-30:
donde: d es un D-aminoácido; L es un L-aminoácido;
35 a es 0-3, de preferencia 0-2, muy preferiblemente 0, 1, 2 ó 3, todavía más preferiblemente 0, 1, ó 2 y muy preferiblemente 1; l, m y n son independientemente unos de otros 1 ó 2, de preferencia 1; x e y son independientemente entre sí 0, 1 ó 2, de preferencia 1.
Este orden de D-y L-aminoácidos (motivo) se hace relevante cuando la secuencia transportadora es sintetizada, es decir, mientras la secuencia de aminoácidos (es decir el tipo de residuos de cadena lateral) permanece sin alterar, los isómeros respectivos se alternan. Por ejemplo, una secuencia transportadora conocida derivada de HIV TAT es RKKRRQRRR (SEQ ID NO: 43). Aplicar el orden de D-/L-aminoácidos de SEQ ID NO: 30 a la misma produciría rKKRrQRRr (SEQ ID NO: 46).
45 En una realización particular, la secuencia transportadora del inhibidor de JNK de la presente invención puede comprender al menos una secuencia de acuerdo con rXXXrXXXr (SEQ ID NO: 31), donde: r representa una arginina D-enantiomérica; X es cualquier L-aminoácido (incluyendo glicina); y en donde cada X puede seleccionarse individual e independientemente de cualquier otra X dentro de la SEQ ID NO: 31. De preferencia, al menos 4 de dichos 6 X L-aminoácidos dentro de la SEQ ID NO: 31 son K o R. En otra realización, el inhibidor de JNK de acuerdo con la presente invención comprende la secuencia transportadora rX1X2X3rX4X5X6r (SEQ ID NO: 32), donde X1 es K, X2 es K, X3 es R y X4, X5 y X6 son cualquier L-aminoácido (incluyendo glicina) seleccionado independientemente entre sí. De forma similar, la
55 secuencia transportadora del inhibidor de JNK de acuerdo con la presente invención puede comprender la secuencia rX1X2X3rX4X5X6r (SEQ ID NO: 33), donde X4 es Q, X5 es R, X 6 es R y X1, X2 y X3 son cualquier L-aminoácido (incluyendo glicina) seleccionado independientemente entre sí. Se prefieren particularmente los anticuerpos policlonales o monoclonales que reconocen un (poli)-péptido que comprende la SEQ ID NO: 172, pero que no reconocen (o al menos reconocen en un menor grado, por ejemplo, al menos de un orden de magnitud) un (poli)-péptido que comprende la secuencia RKKRRQRRRRPKRPATLNLF (SEQ ID NO: 199).
5 La presente invención se refiere también a una célula aislada de acuerdo con el método especificado arriba de aislar una célula que produce un anticuerpo que reconoce un inhibidor de JNK de acuerdo con la presente invención, donde la célula produce un anticuerpo que, preferentemente, reconoce al menos un (poli)-péptido seleccionado de cualquiera de las SEQ ID NO: 1, 3-20, 22-24 y 27, pero no reconoce esencialmente el mismo (poli)-péptido con L-aminoácidos en lugar de los D-aminoácidos en la secuencia que corresponde a la SEQ ID NO: 1 (por ejemplo, sí reconoce un (poli)-péptido que comprende la secuencia RPkRPaTLNLf (SEQ ID NO: 8), pero no reconoce (o al menos en un menor grado, por ejemplo, al menos de un orden de magnitud) un (poli)-péptido que comprende la secuencia RPKRPTTLNLF (SEQ ID NO: 193).
15 La presente invención contempla también generar anticuerpos contra las secuencias transportadoras específicas, permitiendo así identificar por ejemplo inhibidores de JNK como los descritos en la tabla 3. En consecuencia, todos los aspectos (anticuerpos monoclonales o policlonales; métodos para generar los mismos, células que producen los mismos, etc.) descritos arriba para anticuerpos que reconocen un inhibidor de JNK de la presente invención (en particular al menos un (poli)-péptido que comprende o consiste en una secuencia seleccionada de cualquiera de las SEQ ID NO: 1, 3-20, 22-24 y 27) se pueden aplicar también en el contexto de (poli)péptidos que comprendan o consistan en una secuencia seleccionada de cualquiera de las SEQ ID NO: 31-34 y 46-151. Ciertamente, la secuencia de referencia que no debe ser reconocida (o al menos en un menor grado, por ejemplo, de al menos un orden de magnitud) es en este escenario nuevamente la misma secuencia, sin embargo con L-aminoácidos en lugar de D-aminoácidos en el tramo de la secuencia
25 transportadora respectivo.
Son bien conocidos en la técnica métodos para ensayar anticuerpos (monoclonales y/o policlonales) en relación a sus afinidades de unión. Una posibilidad, entre otras, es caracterizar la afinidad de unión de un anticuerpo mediante un ELISA-sándwich usando el péptido objetivo así como controles negativos (por ejemplo, el mismo péptido con L-aminoácidos únicamente). El límite de ELISA puede calcularse – sin estar limitado a –en réplicas blanco como sigue:
límite de ELISA = promedio (control negativo) + (3x desviación estándar de control negativo).
35 Si el valor de muestra es menor que o igual al límite de ELISA, el anticuerpo probado puede considerarse que no tiene afinidad al péptido objetivo. Si el valor de muestra excede el límite de ELISA, se puede considerar que el anticuerpo probado tiene afinidad por el péptido objetivo. Más aún, cuanto más alto sea el valor de muestra, más fuerte es la afinidad del anticuerpo probado para el objetivo.
Un servicio comercial que ofrece la producción de anticuerpos monoclonales o policlonales es por ejemplo Eurogentec (Seraing, Bélgica).
Ejemplos
45 A continuación se presentan ejemplos particulares que ilustran varias realizaciones y aspectos de la invención.
Ejemplo 1: Síntesis del inhibidor de JNK de SEQ ID NO: 172
Como ejemplo ilustrativo, se muestra a continuación la síntesis del inhibidor de JNK de SEQ ID NO: 172. El experto en la técnica sabrá que dicha síntesis también se puede usar y adaptarse fácilmente a la síntesis de cualquier otro inhibidor de JNK de acuerdo con la presente invención. El inhibidor de JNK de SEQ ID NO: 172 se fabricó mediante síntesis de péptidos en fase sólida usando la estrategia de Fmoc (9-fluorenilmetiloxicarbonilo). El enlazador entre el péptido y la resina fue el enlazador de amida Rink (ácido p-[Fmoc-2,3-dimetoxibencil]fenoxiacético). El péptido se sintetizó mediante ciclos sucesivos
55 de acoplamiento de Fmoc-aminoácidos y desprotección con Fmoc. Al final de la síntesis, el péptido completado fue cortado con ácido trifluoroacético (TFA) directamente para producir la amida C-terminal cruda, la cual se purificó después mediante HPLC de fase inversa preparativa. Las fracciones purificadas se agruparon en un lote homogéneo, que se trató mediante cromatografía de intercambio iónico para obtener su sal acetato. El péptido fue después liofilizado.
Excepto cuando se indique, la fabricación tuvo lugar a temperatura ambiente (22ºC±7ºC) en un ambiente de aire filtrado. La escala de síntesis fue 0,7 mmol del aminoácido de partida en la resina para un rendimiento esperado de aproximadamente 1 g de péptido purificado. La síntesis se llevó a cabo manualmente en un reactor
