Description
第1の局面において、本発明は、阻害性の(ポリ)ペプチド配列を含んでいるJNK阻害剤に関する。当該阻害性の(ポリ)ペプチド配列は、以下の一般式:

X8は、F、fおよびwのアミノ酸から選択されるアミノ酸であり、
X1、X2、X3、X5、X7およびX8からなる群から選択されるアミノ酸の少なくとも1、2、3、4、5または6つがD−アミノ酸であることを前提としており、好ましくは、X3、X5、X7およびX8からなる群から選択されるアミノ酸の少なくとも1、2、3または4つがD−アミノ酸であることを前提としている。

(c)配列番号1の配列内のX1、X2、X3、X5、X7およびX8ならびに配列番号2〜27におけるそれぞれの位置からなる群から選択されるアミノ酸に対応するそれぞれの位置に少なくとも1、少なくとも2、少なくとも3、少なくとも4、少なくとも5または6つのD−アミノ酸を含んでおり、より好ましくは、少なくとも1、少なくとも2、少なくとも3または4つのD−アミノ酸を示しており、
(d)JNK活性を依然として阻害する(すなわち、本明細書に規定されるようなJNK阻害剤であることを前提としている。
(a)当該阻害性の(ポリ)ペプチドは、RPTTLNLF(配列番号191)、KRPTTLNLF(配列番号192)、RRPTTLNLF、および/またはRPKRPTTLNLF(配列番号193)からなる群から選択される。
(b)当該トランスポータ配列は、表2に開示されているトランスポータ配列またはそれらのバリアント/断片からなる群から好ましく選択され、配列番号31〜34および46〜151の配列、またはそれらのバリアントもしくは断片のそれぞれからなる群からより好ましく選択される。
さらなる局面において、本発明は、本発明のJNK阻害剤に対して産生される抗体の製造(すなわち、本発明のJNK阻害剤を認識する抗体を製造する方法)に関する。抗体を製造する方法は、当該分野において極めて周知である。

本発明の種々の実施形態および局面を例示する特定の実施例を以下に記載する。しかし、本発明の範囲は、本明細書に記載する特定の実施形態に限定されない。当業者であれば、上述の説明、添付の図面、および以下の実施例を参照することによって、本明細書に記載の実施形態およびその種々の変更も明らかになるであろう。なお、このような全ての変更も添付の特許請求の範囲の範囲内に含まれる。
本発明を説明するための実施例として、配列番号172の配列を有しているJNK阻害剤の合成を以下に詳述する。当業者は、本発明の他のJNK阻害剤の合成についても、以下に詳述する合成法を使用し且つ容易に適用可能なことを理解する。
特記する場合を除き、エアフィルタリング環境下、室温(22℃±7℃)において上記JNK阻害剤の作製を行った。合成のスケールを説明すると、上記樹脂上の開始アミノ酸は0.7mmolであり、精製後に得られるペプチドの予想生成量は約1gであった。機械的攪拌および/または窒素バブリングを行いながら、フリットディスクを設けた30mL〜50mLの容量のリアクタ内でペプチドの合成を手動で行った。
まず、窒素の存在下で、p−メチルベンズヒドリルアミド樹脂(MBHA−樹脂)を、ジクロロメタン/ジメチルホルムアミド/ジイソプロピルエチルアミンを用いて洗浄した。洗浄後、PyBOB(ベンゾトリアゾール−1−イル−オキシ−トリス−ピロリジノ−ホスホニウム−ヘキサフルオロフォスフェイト)/ジイソプロピルエチルアミン/1−ヒドロキシベンゾトリアゾール中で、上記p−メチルベンズヒドリルアミド樹脂をRinkアミドリンカー(p−[Fmox−2,4−ジメトキシベンジル]−フェノキシ酢酸)に結合させて、Fmoc−Rinkアミド−MBHA樹脂を作製した。
以下のサイクルを行って、上記樹脂にアミノ酸をカップリングさせた。
上記Fmoc−Rinkアミド−MBHA樹脂を35%(v/v)のピペリジン/ジメチルホルムアミド中で洗浄した後、ジメチルホルムアミド中で洗浄して、脱保護を行った。上記脱保護の反応を約16分間行った。上記樹脂に対して、ジメチルホルムアミド/ジクロロメタン(50/50)中のFmoc−保護アミノ酸(例えば、2等量のアミノ酸およびHOBt(1−ヒドロキシベンゾトリアゾール))を添加した後、カップリング剤として2等量のジイソプロピルカルボジイミド(DIC)を添加した。添加した上述のアミノ酸に応じて、カップリング反応は1時間〜一晩の間行った。0.5mL/100mgのペプチド−樹脂に基づいてボリュームを計算し、各サイクル後に調整した。上記カップリング後、上記樹脂をDMFで3回洗浄した。上記カップリングの完遂度を、1級アミンにニンヒドリン試験(またはKaiser試験1)を行い、2級アミンにchloranyl試験2を行うことで検査した。ある場合では、信頼性の調整のために、上記chloranyl試験とニンヒドリン試験とを組み合わせ得る。上記カップリングの検査によってカップリング反応が未完了であることが示された場合、ジメチルホルムアミド/ジクロロメタンおよびジイソプロピルエチルアミン中において、より低過剰(0.5等量〜1等量)のアミノ酸、PYBOP、HOBTの存在下でカップリング処理を繰り返す。上記樹脂の機能性を測定した結果、当該樹脂の初期のロード量に応じて、概して0.6meq/g〜0.2meg/gであった。最後のアミノ酸をカップリングした後、上記ペプチド−樹脂を従来通り脱保護し、真空状態のオーブンで30℃において乾燥させる前にDCMを用いて5回洗浄した。上記ペプチド−樹脂を乾燥させた後、固相合成による生成率を、上記樹脂の初期のロード量から算出した理論上の重量増加と比較した上記ペプチド樹脂の重量増加の比率として、算出した。このように算出した上記生成率は、100%に近い値であった。
TFA/K試薬とも呼ばれるトリフルオロ酢酸/1,2−エタンジチオール/チオアニソール/水/フェノール(88/2.2/4.4/4.4/7 v/v)の混合物中において、上記樹脂からの上記ペプチドの切り出しを室温において4時間行った。この際の反応ボリュームは、1mL/100mgのペプチド樹脂であった。上記樹脂を上記試薬に添加している間において、混合温度が30℃未満となるように管理した。
フリットディスクを用いた濾過により、上記樹脂から上記ペプチドを抽出した。ロータベーパー(rotavapor)で体積を1/3にまで濃縮した後、上記ペプチドを冷却したt−ブチルメチルエーテルを使用して沈殿させ、濾過した。その後、このようにして得られた粗製ペプチドを真空中、30℃において乾燥させた。
その後、逆相HPLCを使用して、上記粗製ペプチドの純度が95%以上となるように精製を行った。上記精製後、ロータベーパーで上記粗製ペプチドの精製分画を濃縮し、凍結乾燥させた。
配列番号172の配列を有している精製ペプチドの濃縮および凍結乾燥したプールを、水に溶解させ、Dowexアセテート50−100メッシュ樹脂(Dowex acetate, 50−100 mesh resin)上でイオン交換クロマトグラフィーにより精製した。
以下に標準的な操作手順を詳述し、本発明のJNK阻害剤の阻害効果の測定方法を記載する。この方法より、JNKによるc−Jun特異的な基質のリン酸化を低下させる候補化合物の能力を、インビトロにおいて、非放射性標準化アッセイにより測定することが可能になった。さらに、JNK用に選択した化合物の阻害効果(IC50)およびKiの決定方法を説明する。この方法は、候補化合物がJNK活性を阻害するか否かの検証に好適である。当業者は、下記の方法を特定の目的および必要に応じて適合させる方法を理解する。

3.1 取込み実験の材料および方法
(a)細胞株
本実験に使用する細胞株として、HL−60(参照番号CCL−240、ATCC、ロット116523)を使用した。
2008年4月4日に56℃で30分間デコンプリメントした10%FBS(参照番号a64906−0098、PAA社、ロットA15−151)、1mMのピルビン酸ナトリウム(参照番号S8636、Sigma社、ロット番号56K2386)、およびペニシリン(100ユニット/ml)/ストレプトマイシン(100μg/ml)(参照番号P4333、Sigma社、ロット番号106K2321)を、2008年5月5日に補充した、RPMI(参照番号21875−091、Invitrogen社、ロット8296)またはDMEM(参照番号41965、Invitrogen社、ロット13481)。
PBS 10X(参照番号70011、Invitrogen社、ロット8277):滅菌H2Oを使用して1Xまで希釈。
トリプシン−0.05%EDTA(参照番号L−11660、PAA社、ロットL66007−1194)。
6ウェル培養プレート(参照番号140675、Nunc社、ロット番号102613)。

Ripaリーシスバッファ:10mMのNaH2PO4 pH7.2、150mMのNaCl、1%トリトンX−100、1mMのEDTA pH8.0、200μMのNa3VO2、0.1%SDS、1Xプロテアーゼ阻害剤カクテル(参照番号11873580001、Roche社、ロット番号13732700)。
倒立顕微鏡(Axiovert 40 CFL;Zeiss社;20X)を使用して、細胞の数を数えるととともに、当該細胞の観察を行った。Fusion Alpha Plate読取器(Perkin Elmer社)を使用して、蛍光の読み取りを行った。
懸濁細胞において、FITCで標識化したペプチドの細胞内移行を研究した。ポリ−DL−リジンでコーティングしたディッシュに、1×106細胞/mlの密度で細胞を播種した。その後、既知濃度のペプチドを添加する前に、上記プレートを37℃、5%CO2、および100%の相対湿度下で24時間インキュベートした。上記ペプチドの添加後、上記細胞を37℃、5%CO2、および100%の相対湿度下で30分間、1時間、6時間、または24時間インキュベートした。その後、細胞表面に吸着したペプチドを除去するために、上記細胞を酸性バッファ(0.2Mのグリシン、0.15MのNaCl、pH3.0)で2回洗浄した(Kameyama et al., (2007), Biopolymers, 88, 98-107を参照)。塩基性アミノ酸がリッチなペプチドは細胞表面に強く吸着され、ペプチドの内在化がしばしば過大評価されるという結果を導く場合が多くなるため、上記酸性バッファを使用した。このように酸性バッファを使用した細胞の洗浄を採用して、細胞表面に吸着したペプチドを除去した。Fab/細胞浸透性ペプチドコンジュゲートの細胞内への取り込みを判定する際に上記酸性バッファによる洗浄を行い、その後、PBSにより2回洗浄した。RIPAリーシスバッファを添加することにより、上記細胞を破壊した。その後、バックグランドの除去およびタンパク質含有量のノーマライズを行った後に、内在化したペプチドの相対量を蛍光に基づいて決定した。すなわち、本方法の工程は以下のものである:1.細胞培養、2.酸性洗浄および細胞抽出物、3.蛍光プレート読取器を使用したペプチドの内在化の分析。
上記6ウェル培養プレートは3mlのPoly−D−Lys(Sigma社、P9011;PBS中25μg/ml)でコーティングされており、上記24ウェルプレートは600μlのPoly−D−Lys(Sigma社、P9011;PBS中25μg/ml)でコーティングされており、上記96ウェルプレートは125μlのPoly−D−Lys(Sigma社、P9011;PBS中25μg/ml)でコーティングされており、各プレートは、37℃、5%CO2、および100%の相対湿度下で4時間インキュベートされる。
上記抽出物を氷上で冷却する。
懸濁細胞(または上記皿に十分に接着していない細胞):
上記細胞を「Falcon15ml」に移す。細胞を最大限に回収するため、上記ディッシュを1mlのPBSで洗浄する。
最大で2400rpmの回転数で2分間回転させて上記細胞を回収。
上記細胞を1mlの冷PBS中に懸濁する。
コーティングを施した“エッペンドルフチューブ”(1mlのポリDーLysで4時間コーティングを行い、1mlのPBSで2回洗浄を行ったエッペンドルフチューブ)に上記細胞を移す。
1mlの冷却した酸性洗浄バッファで3回洗浄し、最大2400rpmの回転数で2分間遠心分離させる。
“エッペンドルフ”中での細胞の拡散に注意する。
1mlの冷却したPBSで2回洗浄して、中和させる。
50μlのリーシスRIPAバッファを添加する。
撹拌しながら、氷上で30分間〜1時間インキュベートする。
接着細胞:
3ml、1ml、または200μl(それぞれ、6ウェルプレート、24ウェルプレート、または96ウェルプレートの場合)の冷却した酸性洗浄バッファで3回洗浄する。ディッシュから剥がれる細胞の発生に注意する。
1ml(6ウェルプレート、24ウェルプレート、または96ウェルプレートの場合)のPBSで2回洗浄して、中和させる。
50μlのリーシスRIPAバッファを添加する。
撹拌しながら、氷上で30分間〜1時間インキュベートする。
冷却したヘラで上記細胞をそぎ取る。24ウェルプレートおよび96ウェルプレートの場合、4000rpmの回転数および4℃の温度で15分間、直接、遠心分離にかけて、細胞残屑を除去した。その後、得られた上清(24ウェルプレートの場合は100ml、96ウェルプレートの場合は50ml)を、暗くした96ウェルプレートに直接的に移した。上記プレートの読取を蛍光プレート読取器(Fusion Alpha、Perkin Elmer社)を使用して行った。
上記のライセートを、コーティングを行った“エッペンドルフ”(1mlのポリD−Lysを使用して4時間コーティングを行い、1mlのPBSで2時間洗浄したエッペンドルフ)内に移す。
その後、上記の溶解細胞を4℃の条件下で、1000gで30分間遠心分離して、細胞残屑を除去した。
上澄を回収し、コーティングを行った“エッペンドルフ”(1mlのポリD−Lysを使用して4時間コーティングを行い、1mlのPBSで2時間洗浄したエッペンドルフ)内で−80℃においてこれを保存する。
製造者による使用説明に従って、標準BACアッセイ(キット番号23225、Pierce社)を使用して各タンパク質抽出物の内容を決定した。
10μlの各サンプルを蛍光プレート読取器(Fusion Alpha、Perkin Elmer社)で読み取り、バックグラウンドを除去し、タンパク質濃度によるノーマライズを行った後に、当該各サンプルの相対蛍光を決定した。
FITCにより標識化されたTAT由来のトランスポータ構築物の、HL−60細胞株の細胞内への時間依存的な内在化(摂取)は、配列番号52〜96、43、および45〜47の配列を有しているトランスポータペプチドの配列を使用して、上記のように行われた。上記トランスポータペプチドの配列を表4に示す。
図6に示すように、24時間のインキュベーション後、共通配列rXXXrXXXr(配列番号31)を有している全てのトランスポータが、上記L−TATトランスポータ(配列番号43)よりも高い内在化能を示した。10mMのr3−L−TAT由来トランスポータの存在下で、Hela細部を96ウェルプレート内で24時間培養した。その後、上記Hela細胞を酸性バッファ(0.2Mのグリシン、0.15MのNaCl、pH3.0)で2回洗浄し、PBSで2回洗浄した。そして、RIPAリーシスバッファを添加して細胞を破壊した。その後、各抽出物の蛍光強度を(Fusion Alphaプレート読取器(PerkinElmer社)を使用して)読み取り、バックグラウンドの除去を行って、内在化したペプチドの相対量を決定した。
・24時間のインキュベーション後、共通配列rXXXrXXXr(配列番号31)を有している全トランスポータ(可能性のある配列の選択については、表2を参照)が、上記L−TATトランスポータ(配列番号43)よりも高い内在化能を示した(図6)。上記実験結果により、上記共通配列rXXXrXXXr(配列番号31)の有効性が十分に立証された。
・一つの位置が、最も高いトランスポータ活性およびに重要であること(図6):第2の位置におけるY(配列番号142に対応する配列91)。
以下に手順を詳述し、リガンド誘導分泌後に、ヒト細胞(血液、WBC、PBMC、精製した一次リンパ球、細胞株、...)から放出された数種類のヒトサイトカインの放出量をどのように測定したかを記載する。
・ 96ウェルプレート:
上記上澄の回収用(参照番号82.1581、Sarstedt社)。

アッセイ希釈剤:PBS 1X+10%FBS(参照番号:A15−151、PAA、56℃で30分間デコンプリメンテーション)。

・吸光度読取:
上記吸光度をFusion Alpha Plate読取器(Perkin Elmer社)で読み取った。
・反復ピペット、デジタルピペット、または多重チャンネルピペット。
サンプルの調製
サンプルとして、培養したヒト細胞(通常は、全血、WBC、PMBC、WBCの精製サブタイプ、癌性細胞株)から得られた培養培地上澄を使用する。遠心分離(400g、5分間、4℃)により上記サンプルから粒子状物質を取り除き、この直後に、上記サンプルのアッセイを行うか、または上記サンプルを−20℃以下で保存する。凍結融解を繰り返し行うことは避ける。使用の1時間前に、凍結させた上記サンプルを解凍し、遠心分離器にかける。工程11において、上記サンプルをアッセイ希釈剤中で上記プレートにおいて直接的に希釈する(アッセイ希釈剤を添加した後に上記サンプルを添加し、ピペットによる吸い上げ下げを行う)。
凍結乾燥したスタンダードを室温まで温めた後、材料損失を避けるようにバイアルを慎重に開ける。そして、上記凍結乾燥させたスタンダードを、企図する量の脱イオン水でもどし、スタンダードのストックを作製する。希釈を行う前に上記スタンダードを少なくとも15分間平衡化させる。穏やかにボルテックスして、混合する。上述のように上記凍結乾燥させたスタンダードを脱イオン水でもどした直後に、スタンダードのストックを各バイアル当たり50μlとなるようにポリプロピレン製バイアルに等分し、−20℃で最大6か月間凍結させる。必要に応じて、これら等分/凍結を行う前に、2℃〜8℃で最大で8時間保存する。なお、上記脱イオン水でもどしたスタンダードを室温で静置することは避ける。
使用直前に、Diluent試薬中で希釈倍率が2倍毎の段階的希釈法を使用して10ポイントの標準曲線を調製した。4000pg/mlの高スタンダードが推奨される。
ビオチン/SAv試薬の1工程インキュベーション。必要な量の検出用抗体をアッセイ希釈剤(Assay Diluent)に添加する。使用前15分以内に、必要な量の酵素試薬を添加して、ボルテックスまたは混合を十分に行う。推奨される希釈物を得るため、ロット固有の取扱説明書/分析証明書を参照。使用後、残留して動作しているディテクタを廃棄する。
1.コーティング用バッファ中に希釈した捕捉抗体を各ウェル当たり100μL使用して、PVCマイクロタイタープレートのウェルをコーティングする。推奨される抗体のコーティング用希釈物を得るため、ロット固有の説明書/分析証明書を参照。
2.上記PVCマイクロタイタープレートを接着性プラスチックで覆い、4℃の温度下で一晩かけてインキュベートする。
3.上記コーティング溶液を取り除き、上記プレートのウェルに150μlの洗浄バッファを注入して洗浄する。
4.上記プレートを浴槽上で軽く叩いて、コーティング溶液または洗浄液を取り除く。
5.上述のプロセスを2回繰り返し行うことで、あわせて3回の洗浄を行う。
6.最後の洗浄後、上記プレートを紙タオルで軽く叩いて、残留する洗浄バッファを取り除く。
7.コーティングを行った上記ウェル内において、各ウェル当たり100μlのDiluent試薬を添加して、残留しているタンパク質結合部位をブロッキングする。
8.上記プレートを接着性プラスチックで覆い、室温で1時間インキュベートする。
9.上記インキュベーションの最中に、スタンダードの調製を開始する。
10.洗浄バッファ150μlを使用して、工程3のように洗浄を1回行う。これにより、上記プレートは、サンプル添加への準備ができた状態である。
11.各ウェルに対して、アッセイ希釈剤中に適切に希釈したサンプルを50μlづつ添加する。正確な定量結果を得るために、未知試料のシグナルを標準曲線のものと常に比較する。スタンダード(3重)とブランクとは各サイトカインを存在させて試験を行い、正確性を確保する。
12.上記プレートを接着性プラスチックで覆い、室温で2時間インキュベートする。
13.上記工程3と同様に、上記プレートを150μlの洗浄バッファで4回洗浄する。
14.推奨される希釈(ロット固有の説明書/分析証明を参照)において、50μlのディテクタミックス(アッセイ希釈剤中の検出抗体+二次ストレプタビジン−HRP抗体)を各ウェルに添加する。
15.上記プレートを接着性プラスチックで覆い、遮光した状態で、室温において1時間インキュベートする。
16.上記工程3にように、上記プレートを150μlの洗浄バッファで6回洗浄する。
17.各ウェルに50μlのDAKO TMB溶液を添加し、上記プレートを密閉せずに室温で15〜20分間暗闇でインキュベートする。
18.停止液50μlを各ウェルに添加する。上記プレートを優しくタップし、完全な混合を確保する。
19.プレートミキサーにより、上記プレートを500rpmの回転数で5分間混合する。
20.波長450nmで光学濃度を読み取る(プログラム:Fusion Alpha Plate読取器上でサイトカイン_ELISA)。
各スタンダード対照(スタンダードコントロール)と各サンプルについて、3回繰り返した読取の平均をとる。ゼロ標準の光学濃度(O.D)の平均を差し引く。O.D対数に対応するサイトカイン対数をプロットした標準曲線を引き、回帰分析により最もよく適合する線を決定する。サンプルを希釈した場合、上記標準曲線から読み取れる濃度値は、希釈係数を乗じた値になることが求められる。標準曲線は、分析対象の各サンプルのセットに対して作成する必要がある。Grugg試験を使用して、異常値データを避ける。その後、標準偏差の2倍の間隔内にないデータを廃棄する。ポジティブ対照(ポジティブコントロール)が予め確認されたようなデータを示す場合、独立した(independent)実験を考慮に入れる。これら独立した実験をプールする(N>3)。
データは、サイトカイン放出量(単位:pg/ml)、または阻害剤の処理を行わない誘導条件と比較した百分率(%)で示す。
本実施例の手順を以下に詳述し、LPSの投与を6時間受けたヒトPMA分化THP1細胞から如何にしてサイトカイン生成を誘導したかを説明する。本実施例は、サイトカインの刺激誘導放出を減衰させることに関する、本発明にかかるJNK阻害剤(特に、配列番号172の配列を有しているJNK阻害剤)の能力を試験するために行った。サイトカイン放出の読取のために、異なるリガンドを使用してTHP1細胞をex−vivoにおいて刺激した。非毒性のドーズ下では、JNK阻害剤の有効性は、非処理のサンプルと比較したサイトカインレベルの低下で示され、ELISA法によりモニタリングされる。化合物の毒性は、トレタゾリウム塩(tretazolium salt(MTS))からホルマリンへの還元(紫色を生じさせる。)に基づき評価した。
a.材料
・細胞株:THP−1(参照番号TIB−202、ATCC、ロット57731475)。
・培養培地、試薬、およびプレート
− 以下の成分を補充したRMPI(参照番号21875−091、Invitrogen社):
10%FBS(参照番号A15−151、PAA社):56℃で30分間のデコンプリメンテーション。
10mMのHepes(参照番号H0887、Sigma社)。
50Mの−メルカプトエタノール(参照番号63690、Fluka社:14.3Mのストック):−20℃でストックしたPBSに対して、50mMのアリコート560lを添加。
1mMのピルビン酸ナトリウム(参照番号S8636、Sigma社)。
ペニシリン(100ユニット/ml)/ストレプトマイシン(100g/ml)(参照番号P4333、Sigma社)。
その後、上記RPMI培地を0.22Mのフィルタ(参照番号SCGPU05RE、Millipore社)で濾過する。
− PBS 10X(参照番号70011、Invitrogen社):滅菌H2Oを使用して1Xまで希釈する。
− DMSO:参照番号41444、Fluka社。
− PMA(ホルボール 12−ミリステート−13アセテート、参照番号P1585、Sigma社、濃度1mM=616.8ug/ml、−20℃のDMSO中)。RPMI中100nMの終濃度で(10mlの培地中において1μl)直接使用する。
− LPSultrapure(リポ多糖体、参照番号tlrl−eklps、Invivogen社、濃度5mg/ml):LPSのストック溶液:4℃において、PBS中3g/ml。RPMI培地中40ng/mlの4X濃度溶液を調製するために直接使用する(最低限、1800l/プレート;5プレート:LPS3g/mlを125l+9250l RPMI)。
− 96ウェルプレート
接着細胞培養用(参照番号167008、Nunc社)
上澄回収用(参照番号82.1581、Sarstedt社)
ELISA用(F96 maxisorp、参照番号442404、Nunc社)
コーティング溶液:ポリD−リジン(参照番号P9011、Sigma社):PBS1X中、25g/mlの最終希釈物。
・ELISA試薬およびキット
− コーティングバッファELISA:0.1MのNaCarbonate pH9.5(1リットルのH2O中、7.13gのNaHCO3(参照番号71627、Fluka社)および1.59gのNa2CO3(参照番号71345、Fluka社)、NaOHを含む9.5までのpH 濃縮)
− 洗浄バッファELISA:PBS 1X+0.01%Tween20(参照番号P1379、Sigma社、ロット094K0052)(=1リットルのPBS1Xを調製し、磁性撹拌器を使用して撹拌を行いながら100μlのTween20をゆっくりと添加する)。
− アッセイ希釈剤:PBS1X+10%FBS(参照番号A15−151、PAA社、56℃で30分間デコンプリメンテーション)。
− DAKO TMB(参照番号S1599、DAKO社):市販の基質溶液。
− 停止液:1MのH3PO4(200mlの場合、177mlのH2O+23mlのH3PO4 85%(参照番号345245、Aldrich社))。
− TNF− :キット ヒトTNF ELISAセット、BD OptEIA(参照番号555212、DB社)。
・細胞毒性測定:CellTiter96試薬(参照番号G3581、Promega社)。
・コントロール(対照)化合物:SP600125(参照番号ALX−270−339−M025、Alexis社、濃度:20mM DMSO)。
・吸光度の読取:Fusion Alpha Plate読取器(Perkin Elmer社)で吸光度を読み取った。
・連続分注ピペット、デジタルピペット、またはマルチチャンネルピペット
TopSeal−A:96ウェルマイクロプレートシール(参照番号600585、PerkinElmer社)。
ウェルのコーティング
上記プレートを200lのポリD−リジン(1x)でコーティングした。そして、37℃の温度条件下で、5%CO2、100%の相対湿度下で、インキュベーションを2時間行った。
2時間後、上記ウェルを200lのPBS 1Xで2回洗浄した(直ちに使用するか、または使用するまで200lのPBS 1xの存在下で30℃の温度で静置した(但し、静置期間は3日を超えないものとした))。
3日後、顕微鏡を使用して上述の接着細胞を観察した。PMA含む上記培地を吸引して、PMAを含まない100lの新しいRPMI培地を代わりに供給した(PBS 1xによる洗浄工程は行わなかった)。
6時間後、100lの上澄を新しい96ウェルプレートへ移した。そして、上記96ウェルプレートを封止し、ELSA法(実施例4を参照)によるサイトカイン分泌の分析を行うまで−20℃で保存した。
上記データの分析をELISA法で説明したように行う(実施例4を参照)。簡単に説明すると、ELISA法の場合:各スタンダードコントロールおよび各サンプルについて、3重の読取をしてその平均をとる。そして、ゼロスタンダードの光学濃度(O.D)の平均を差し引く。O.Dの対数に対応するサイトカイン濃度の対数をプロットする標準曲線を引き、回帰分析により最もよく適合する線を決定する。サンプルを希釈していた場合、上記標準曲線から読み取れる濃度値は希釈係数を乗じた値であることが求められる。標準曲線は、アッセイ対象のサンプルの各セットについて作成すべきである。Grugg試験を利用して、異常値データを避けた。標準偏差の2倍の間隔範囲の外にあるデータは廃棄した。ポジティブコントロールが以前に観察されたようなデータを示した場合は、独立した実験を考慮に入れる。上記独立した実験はプールされている(N>3)。
クエン酸ナトリウムを含む、予め標識化された真空管に接続した静脈穿刺を利用して、麻酔をかけたラットまたは健康な志願人の全血を採取した。上記管を7、8回反転させて採取した全血とクエン酸ナトリウムを穏やかに混合させた後、得られたミックスを、刺激を与えるまで室温に保持した。配列番号172に示す配列を有しているJNK阻害剤を、PBS中6倍の濃縮度で調製し、30μl/ウェルの分量で上記ミックスを96ウェルプレートに添加した。全血をPBS中に1:2の比率で希釈し、PBSのみ又は上記配列番号172の配列を有している上記JNK阻害剤が予め添加されている各ウェルに120μlの希釈血液を添加した。Stuart OrbitalインキュベータSI500の回転数を85rpmに設定し、全血を37℃で60分間インキュベートした。活性化因子(LPS)は、6倍の濃縮度で調製したものを30μl/ウェルの分量で使用した。培養を60分間行った後、上記全血にLPSを添加し、ピペットによる吸い上げ下げを行って混合した。その後、得られたミックスを撹拌(85rpm)させながら37℃で4時間保存した。4時間のインキュベーションを行った後、上記プレートを予め冷却した遠心分離機にかけ、約770gおよび4℃で15分間遠心分離を行った。最後に上澄を回収し、サイトカインの測定を行うまで−20℃で保存した。その後、標準的なElisaキット(例えば、R&D Systems社製のDuoSet ElisasまたはBD Biosciences社製のBD Opteia Set Elisa)を使用して、サイトカイン(IL−6、IL−2、IFNγ、およびTNFα)を測定した。測定結果を、上記サイトカインの、上澄中のpg/mlとして示す。
配列番号196、197、および172の配列を有しているJNK阻害剤(終濃度:0.1mM)を、ヒト血清中(PBS 1x中で10%および50%)で消化した。Tugyi et al.(Proc Natl Acad Sci USA, 2005, 413-418)に開示されているように実験を行った。消化されずに残った完全なペプチドをUPLC−MSで定量化した。2つの別箇のアッセイを行う以外は同一の方法で、上記配列番号196、197、および172の配列の安定性を算定した。上記配列番号196の配列を有しているJNK阻害剤は6時間以内に完全に分解されてアミノ酸残基になった一方、上記配列番号172の配列を有しているJNK阻害剤は14日後に漸く完全に分解された。上記配列番号197の配列を有しているJNK阻害剤は30日後も安定したままであった。
コントロールの動物を犠牲にし、そのリンパ節(LN)を採り、完全RPMI培地中で保存した。保存後、5mlのピストンを使用して、70μmフィルタ上で、上記LNを上記完全RPMI培地と混ぜて破砕した。数滴の培地を添加してストレーナーを濡れた状態に保った。450gおよび4℃の条件下で細胞を7分間遠心分離した。沈殿物を5mlの新しい培地中に再懸濁した。再懸濁後、細胞を再度、セルストレーナーに通した。細胞を1400rmpの回転数および4℃の条件下で10分間にわたり再度遠心分離しつつ、1アリコート分の細胞の数を数えた。細胞をMACSバッファ(107細胞当たり80μlのMACSバッファ)中に再懸濁した。1000万個の細胞当たり10μlの抗ラットMHCマイクロビーズを添加し、細胞を4℃〜8℃で15分間インキュベートした。そして、上記細胞を15mlのMACSバッファで洗浄し、700gおよび4℃の条件下で7分間遠心分離した。108個の細胞当たり500μlのMACSバッファ中に沈殿物を再懸濁した。動物ごとに、上記MACSセパレータの磁界内にLSカラム(LS column)を1つ設置した。上記LSカラムを先ず3mlのMACSバッファでリンスした。上記LSカラムの下方において1つの管を氷中に設置して、細胞=T細胞を回収した(ネガティブセレクションを行い、溶出物を回収した)。細胞懸濁物を添加し、溶出物を氷上で回収した。カラムを3mlのMACSバッファで3回洗浄した。溶出したT細胞を、700gおよび4℃下で7分間遠心分離した。再懸濁させた細胞の数を数え、200000細胞/ウェルの密度で100μlの完全培地中に播種した。2μg/mLのCD3抗体を使用する実験の前日に、プレートに予めコーティングを施し、実験日に当該プレートをPBSで3回洗浄した。リガンドによる活性化の1時間前に、2倍に濃縮した100μlの(ポリ)ペプチドJNK阻害剤(配列番号172)で細胞を処理した。(ポリ)ペプチドJNK阻害剤(配列番号172)を用いた前処理の1時間後に、細胞を2μg/mLの抗CD28抗体で24時間刺激した。この24時間の刺激付与の後、上澄を回収し、分析を行うまで−20℃で保存した。その後、標準のElisaキットを使用してサイトカインを測定した。測定結果を、上澄における、上記サイトカインのpg/mlとして示した。
クエン酸ナトリウムを含み、予め標識化された真空管に接続された静脈穿刺を使用して、健康状態の志願者からヒト全血を採取した。上記管を7、8回反転させて混合した後、刺激を与えるまで室温に保持する。1,2ml−96ウェルプレートに、350μlのRPMI+P/Sを添加した。RPMI+P/S(各ウェル当たり50μl)中において、配列番号172の10倍濃縮物を調製した。この50μlを1,2ml−96ウェルプレートに添加した。その後、培地のみ又はJNK阻害剤を予め添加した各ウェルに、50μlの全血を添加した。全血を37℃および5%CO2の条件下で60分間インキュベートした。RPMI+P/S中に希釈した、ウェルあたり50μlのリガンドを、最終希釈の10倍濃度となるように調製した。上記60分間のインキュベーション後、リガンドを添加した。そして、ピペットで上記全血の吸い上げ下げして、上記ウェル内の混ぜ合わせを行った。上記全血は、37℃で3日間インキュベーションを行った(各ウェル内の吸い上げ下げを1日1回行い、当該ウェル内の混ぜ合わせをした)。上記インキュベーションの終了時に、プレートの混ぜ合わせを行い、その後、予め冷却した遠心分離機を使用して、2500rpmおよび4℃の条件下で遠心分離を15分間行った。その後、標準のElisaキットを使用してサイトカインを測定した。測定結果を、上記上澄における、上記サイトカインのpg/mlで示す。

- 配列番号2〜27のいずれか1つから選択される阻害性のペプチド配列を含んでいる、ペプチドである、JNK阻害剤。
- 配列番号8のペプチド配列を含んでいる、請求項1に記載のJNK阻害剤。
- トランスポータ配列を含んでいる、請求項1または2に記載のJNK阻害剤。
- 上記阻害性のペプチド配列および上記トランスポータ配列は重なり合っている、請求項3に記載のJNK阻害剤。
- 上記トランスポータ配列は、配列番号28〜30のいずれか1つにしたがう、D−およびL−アミノ酸が交互になっている配列を含んでいる、請求項3または4に記載のJNK阻害剤。
- 上記トランスポータ配列は配列番号31〜170のいずれか1つから選択される、請求項3〜5のいずれか1項に記載のJNK阻害剤。
- 上記トランスポータ配列は、配列番号31〜34、46、47および52〜151のいいずれか1つから選択される、請求項3〜6のいずれか1項に記載のJNK阻害剤。
- 上記トランスポータ配列は、上記阻害性のペプチド配列のC末端またはN末端に直接に置かれている、請求項3〜7のいずれか1項に記載のJNK阻害剤。
- 配列番号171〜190のいずれか1つにしたがう配列を含んでいる、請求項3〜8のいずれか1項に記載のJNK阻害剤。
