- Plasmid pSGS480 was transiently transfected into CHO cells and the cells were grown at 28°C for 7 days. The presence of CR(del C)-Fc protein in these cells and the conditioned media was examined by Western blot analysis.

- the mouse spleen cells were fused with FL653 myeloma cell at a 1spleen :6 myeloma ratio and were plated at 100,000, 33,000and 11,000 cells per well into 96 well tissue culture plates in selection media. Wells positive for growth were screened by FACS and ELISA a week later. Two fusions were performed.

- Antigens CR-del-C-Fc and CR-EGF-fc, control hu IgG1 fusion protein were diluted in binding buffer to 500ng/ml. 100 ⁇ l were added per well and incubated for 1 hr at 37°C our overnight at 4°C. The liquid was decanted and the plate inverted and blotted until dry. 250 ⁇ l/well blocking buffer was then added, followed by incubation for 30 min. at 37° C. Again, the liquid was decanted and the plate inverted and blotted until dry. Supernatants were diluted 1:50 in wash buffer, and plated at 50 ⁇ l/well, followed by incubation for 1 hour at room temperature.
- Plates were washed 3X vigorously with 250 ⁇ l/well wash buffer. Then 100 ⁇ l/well 2' antibody diluted in wash buffer at 1:10,000 was added, followed by incubation for 30 min. at room temperature. Plates were then washed 3X vigorously with 250 ⁇ l/well wash buffer, then substrate added at 100 ⁇ l/well. Color was permitted to develop until sufficiently dark, then 100 ⁇ l/well stop solution was added and the plates read for absorbance at 450nm.

Abstract
Claims (21)
- Anticorps qui lie spécifiquement un épitope de Cripto compris dans le domaine s'étendant des résidus d'acides aminés de l'acide aminé 46 à l'acide aminé 62 de SEQ ID N° 1 ou SEQ ID N°2.
- Anticorps selon la revendication 1, l'anticorps liant un épitope choisi dans le groupe des épitopes auxquels les anticorps produits par les hybridomes choisis dans le groupe consistant en A10B2.18 et B3F6.17 se lient.
- Anticorps selon la revendication 1 ou 2, l'anticorps étant capable d'intemaliser Cripto.
- Anticorps selon l'une quelconque des revendications 1 à 3, qui est un fragment d'anticorps choisi dans le groupe consistant en un fragment Fab, un fragment Fab' et un fragment F(ab)2.
- Anticorps selon l'une quelconques des revendications 1 à 3, qui est un anticorps de pleine longueur.
- Anticorps selon l'une quelconques des revendications 1 à 3, qui est un anticorps simple chaîne.
- Anticorps selon l'une quelconques des revendications 1 à 6, qui est conjugué à un agent chimiothérapeutique.
- Anticorps selon l'une quelconques des revendications 1 à 6, pour une utilisation en thérapie en combinaison avec un agent chimiothérapeutique non conjugué.
- Anticorps selon la revendication 7, dans lequel l'agent chimiothérapeutique est choisi dans le groupe consistant en une pro-drogue activée par une tumeur, un radionuclide et une toxine choisie dans le groupe consistant en la ricine, la toxine diphtérique et l'exotoxine de Pseudomonas.
- Anticorps selon la revendication 7 ou 9, dans lequel l'agent chimiothérapeutique est un maytansinoïde.
- Anticorps selon l'une quelconque des revendications 1 à 10, qui est humain.
- Anticorps selon l'une quelconque des revendications 1 à 11, qui est monoclonal.
- Anticorps selon l'une quelconque des revendications 1 à 12, qui est humanisé.
- Composition pharmaceutique comprenant au moins un des anticorps selon l'une des revendications 1 à 7 et 9 à 13 et éventuellement un support.
- Composition pharmaceutique conformément à la revendication 14, comprenant en outre un agent chimiothérapeutique non conjugué.
- Composition pharmaceutique selon la revendication 14 ou 15, dans laquelle l'anticorps est B3F6.17 humanisé.
- Utilisation d'un anticorps de l'une quelconque des revendications 1 à 13 ou d'une composition de l'une quelconque des revendications 14 à 16, pour réduire la croissance d'une tumeur in vitro.
- Utilisation d'un anticorps de l'une quelconque des revendications 1 à 13 ou d'une composition de l'une quelconque des revendications 14 à 16, pour la préparation d'une composition pharmaceutique destinée à réduire la croissance d'une tumeur in vivo.
- Utilisation selon la revendication 17 ou 18, dans laquelle la cellule tumorale est choisie dans le groupe consistant en des cellules de tumeur de sein, testicule, colon, poumon, ovaire, vessie, utérus, col de l'utérus, pancréas et estomac.
- Utilisation d'un anticorps de l'une quelconque des revendications 1 à 13 ou d'une composition de l'une quelconque des revendications 14 à 16, pour la préparation d'une composition pharmaceutique pour le traitement de la prolifération de cellules indésirables.
- Procédé pour moduler la croissance de cellules tumorales in vitro dans un échantillon, comprenant l'étape d'addition à l'échantillon, de l'anticorps selon l'une quelconque des revendications 1 à 13 ou de la composition selon l'une quelconque des revendications 14 à 16.
