Definitions
- the invention relates to a sample dispenser for applying liquid samples to a sample receiving area.
- sample dispensers which have a capillary with which a liquid sample can be drawn in and dispensed to a sample area.
- the sample area must have an absorbent material that sucks the sample liquid out of the capillary.
- the capillary of this autosampler is inserted into a thin spring leaf similar to a fountain pen.
- the smallest amounts can be recorded very precisely, since the amount is exactly defined by the volume of the capillary.
- the discharge of the sample quantity taken up in the capillary causes considerable problems, since the absorbent material forming the sample receiving area must have a very strong suction effect, since otherwise residues of the sample liquid remain in the capillary.
- These sample dispensers with capillaries can therefore only be used for special sample collection areas and it is also difficult to use such sample dispensers in an automated process in which it is not always checked whether the sample liquid taken up in the capillary is completely transferred to the sample collection area.
- a membrane comb which consists of a porous material, such as a nylon membrane coated with hydroxyl groups.
- a sample amount of approximately 0.2 to 0.3 microliters should be applied to each individual tooth of this membrane comb.
- the sample material is, for example, DNA in formamide.
- Samplers in the form of tips are also known, with which it should be possible to meter viscous media.
- these samplers have not proven themselves in the above-described application for the metered transfer of formamide, since no drops or only the smallest drops with different amounts remain attached to them.
- WO 93/09872 describes a plate with reaction vessels (e.g. FIG. 5), which is basically similar to a microtiter plate.
- a metering device is provided for supplying chemicals.
- This measuring device has protrusions to which drops of chemicals can adhere. These protrusions are partly shown as hollow cylindrical bodies.
- WO 98/57747 shows a further autosampler which is designed as a rod with a wettable tip and non-wettable side surfaces.
- the rod can basically have any cross-section (oval, polygonal, ...) and that it can consist of glass or another material composition.
- DE 30 16 682 A1 relates to a blood serum applicator.
- the applicator described therein has a porous section with which the blood serum is transferred.
- FIG. 1A to FIG. 2B shows a sample dispenser which has recesses in which the sample is taken up by means of capillary forces.
- No. 5,882,930 describes a sampler which is formed from a transfer element on which pins are arranged in a regular grid. Sample droplets may adhere to the free tips of the pens.
- the pins preferably have a diameter of 30 ⁇ m to 100 ⁇ m and are cylindrical.
- WO 98/5852 describes a sample applicator with pins which are fastened to a holder structure. The pens are so thin that the tip is enclosed by the drop.
- the invention has for its object to provide a sampler for applying liquid samples to a sample receiving area, which is simple in structure, can safely accommodate small sample volumes ( ⁇ 1 microliter) and can be safely transferred to a sample receiving area, and which is suitable for automatic operation , in which not every sample intake and every sample delivery can be checked individually.
- the sample dispenser according to the invention for applying liquid samples to a sample receiving area is formed from a sample dispenser body which has a sample receiving surface.
- the sample receiving area is delimited by a circumferential edge and is provided with a flat recess such that when the sample receiving area is immersed, a droplet of a liquid sample of a predetermined size adheres to the sample receiving area and is released onto the sample receiving area by placing the sample receiving area on it.
- the desired quantities of liquid samples can be safely taken up only by immersing the sample receiving area in the liquid sample and released by placing them on the sample receiving area.
- the sample receiving area preferably has a certain absorbency, which, however, can be significantly lower than is necessary when using the sample dispensers with capillary described at the beginning.
- the sample dispenser according to the invention is very simple, with it the desired amount of sample to be transferred is determined by the recess in the sample receiving surface and it is surprisingly reproducible.
- the amount of sample is influenced by the physical properties, in particular the surface tension, of the sample liquid. The greater the surface tension of the sample liquid, the larger the liquid droplets transferred with the sample dispenser according to the invention. When dimensioning the recess of the The type of sample liquid must therefore also be taken into account.
- samplers e.g. Pipette tips, steel capillaries, glass capillaries and various samplers with long grooves or notches. With all these samplers, the problems described at the outset existed when transferring the sample liquid to the sample receiving area.
- a sampler was tested which has a flat sample receiving surface with a predetermined size, the surface being roughened.
- this embodiment has allowed a safe transfer of the sample liquid to the sample receiving area, this has taken sample droplets of various sizes when immersed in the sample liquid, so that even with this sample dispenser it is not possible to transfer a reproducible amount of sample.
- FIG. 1 shows a sample dispenser according to the invention according to a first embodiment in side view
- FIG. 2 shows the sampler according to FIG. 1 in longitudinal section
- FIG. 4 shows the tip of the sampler according to FIG. 1 in an enlarged sectional view
- FIG. 5 the tip of the sampler according to FIG. 1 in a perspective view looking obliquely from below
- 6 shows a sample dispenser according to a second embodiment in side view
- FIG. 7 shows the sampler according to FIG. 7 in longitudinal section
- FIG. 8 shows the sample dispenser from FIG. 6 in a perspective view looking obliquely from above
- FIG. 9 is an enlarged sectional view of the tip of the sampler from FIG. 6,
- Fig. 10 is an enlarged view of the tip of Fig. 6 in side view
- FIG. 11 shows the tip of the sampler from FIG. 6 in a perspective view looking obliquely from below.
- the sampler according to the invention has a sampler body 1 formed from an aluminum alloy, which consists of a rod-shaped one
- the tip 4 has the shape of a cylinder.
- the length of the shaft region 2 is approximately 20 mm, that of the transition region 3 approximately 19 mm and the length of the tip 4 approximately 2 mm.
- the diameter of the tip is approx. 1 mm.
- the free end face of the tip 4 forms a sample receiving surface 5.
- the sample receiving surface 5 is circular (FIGS. 3, 5) so that it is delimited by a circular edge 6.
- the sample receiving surface 5 is provided with a conical recess 7, which is concentric with a longitudinal axis 8 of the sample dispenser, the conical surfaces formed by the conical recess ending at the edge 6 delimiting the sample receiving surface 5.
- the angle ⁇ , which the conical recess encloses, is in the range from 90 ° to 150 ° and is 120 ° in the exemplary embodiment shown. Such a recess can easily be made with a drilling tool.
- the recess 7 produced with a drilling tool has smooth inner surfaces which, apart from deburring the edges, have no post-processing require. It has been shown that a smoothly designed sample receiving surface 5 is advantageous, since this allows a better transfer of the sample liquid from the sampler to a sample receiving area. If the recess 7 is produced by a method other than drilling, in which the surface is roughened, it is expedient for the surface to be smoothed, for example by polishing.
- a bore 9 is made concentrically to the longitudinal axis 8, which extends into the transition area 3.
- a threaded hole 10 is made perpendicular to the hole 9.
- a holding rod of an actuating device e.g. a robot for the amplification or sequencing of DNAs and are fixed by means of a screw screwed into the threaded bore 10.
- the sample dispenser according to the invention is attached to the actuating device with the sample receiving surface 5 facing downward.
- the sample dispenser with its sample receiving surface 5 is immersed in a sample reservoir, as a result of which a sample droplet is deposited on the sample receiving surface 5, the size of which is determined by the sample receiving surface 5 and the recess 7.
- the sample droplet adheres to the sample receiving surface 5 by wetting forces, which are usually Van der Waals forces.
- the sample dispenser is then placed with its sample receiving surface 5 on a sample receiving area, with its sample receiving surface 5 being pressed onto the sample receiving area. As a result, the sample droplet adhering to the sample receiving surface 5 is completely transferred to the sample receiving area.
- the sample receiving area soft and elastic, for example by providing a corresponding base, so that it is pressed into the recess 7 when the sampler is pressed on and completely absorbs the sample liquid contained therein.
- the cleaning of the autosampler according to the invention takes place by pressing into a moistened foam material, whereby the recess 7 and the surfaces of the autosampler adjoining it are cleaned.
- the foam material The material is open-celled and it is moistened with water. However, it is also possible to add a detergent to the water.
- FIGS. 6 to 11 show a second exemplary embodiment of the sampler according to the invention, which essentially corresponds to the first exemplary embodiment, which is why the same parts are designated with the same reference numerals.
- the only difference from the first exemplary embodiment is that the cylinder of the tip 4 has a larger diameter, namely 1.5 mm, and the tip 4 is flattened into two parallel surfaces 11 in the region of the lateral surface of the cylinder adjacent to the sample receiving surface 5 (FIG . 9 to 11).
- the two flattened surfaces 11 are arranged at a distance d of approximately 1 mm. This results in an approximately rectangular sample receiving surface 5 with short bent-out side edges in the view from below (FIG. 11).
- the second exemplary embodiment has a larger sample receiving surface 5 compared to the first exemplary embodiment, which is why larger sample droplets are also recorded with it.
- the distance between the parallel surfaces 11 is not greater than the diameter of the sample receiving surface 5 of the first exemplary embodiment, which is why the sampler according to the second exemplary embodiment also in spatially narrowly defined sample receiving areas, such as e.g. the teeth of membrane combs known from DE 197 00 626 A1 can be applied.
- the sample receiving area 5 is delimited by a circumferential edge, so that its area is clearly defined, which also defines the sample receiving volume in addition to the other parameters such as the depth and shape of the recess 7 and the type of sample liquid .
- the peripheral edge can be formed by a body edge, in which adjacent body surfaces of the sampler body abut one another at a right angle or at a smaller angle.
- the edge can also be formed, for example, by a narrow ring web on a flat or curved surface.
- a further essential aspect of the invention is the recess, since on the one hand, due to the adhesive forces and the surface tension of the sample liquid, a safe absorption of a predetermined amount of sample liquid is guaranteed and, on the other hand, the secure transfer to a sample receiving area is ensured.
