Abstract

Настоящее изобретение относится к очищенным белкам, последовательностям ДНК, кодирующим их экспрессию, и рекомбинантным молекулам ДНК, включая клетки-хозяева, трансформированные с их помощью, для трансформирования растений кофе с целью подавить экспрессию ферментов, необходимых для синтеза этилена. Последовательности ДНК и рекомбинантные молекулы ДНК характеризуются тем, что они кодируют экспрессию ферментов АЦК синтазы (аминоциклопропан-1-карбоксилат синтазы) или АЦК оксидазы, которые участвуют в процессе биосинтеза этилена в растениях кофе. Растения кофе трансформируют векторами, содержащими последовательности ДНК АЦК синтазы и/или АЦК оксидазы, которые кодируют экспрессию соответствующей РНК, которая является антисенсом мРНК для АЦК синтазы и/или АЦК оксидазы. Полученная антисенсная мРНК связывается с мРНК, соответствующей АЦК синтазе и/или АЦК оксидазе, тем самым инактивируя мРНК, кодирующую один или более ферментов в процессе биосинтеза этилена. Указанные последовательности ДНК также могут быть использованы для предотвращения синтеза АЦК синтазы или АЦК оксидазы путем косупрессии. Итогом любой реализации является то, что трансформированные растения становятся не способными синтезировать этилен, причем на другие аспекты их метаболизма влияния не оказывается.Международная заявка была опубликована вместе с отчетом о международном поиске.
