Claims (5)
1. Fremgangsmåde til syntetisering af et oligonukleotid omfattende: sekventielt at kondensere og oxidere en nukleosidphosphoramidit-forbindelse i nærvær af en syre-/azolkompleksforbindelse under anvendelse af et udgangsråmateriale, dvs. hydrofobisk gruppebundet nukleosid repræsenteret ved formel (1):
(l) hvor R1 betegner en alkylengruppe med 1 til 12 carbonatomer; R2 betegner en alkylengruppe med 1 til 22 carbonatomer; R3 og R4 hver især uafhængigt betegner en alkylgruppe med 1 til 22 carbonatomer, eller R3 og R4 betegner en divalent gruppe, i hvilken de er bundet til hinanden for at danne en hete-rocyklus sammen med tilstødende nitrogenatomer og R2, hvor heterocyklus-sen er repræsenteret ved Formel (2):
(2) hvor R7 betegner en alkylgruppe med 1 til 6 carbonatomer; m betegner et helt tal på 0 eller 1; og y betegner et helt tal på 0 til 3; R5 betegner en enkeltbinding eller en alkylengruppe med 1 til 22 carbonatomer; R6 hver især uafhængigt betegner en alkylgruppe med 6 til 30 carbonatomer; n betegner et helt tal på 2 til 6; X betegner et hydrogenatom, en hy-droxylgruppe, eller en hydroxylgruppe beskyttet af en beskyttelsesgruppe; og Z betegner en adenylgruppe, en guanylgruppe, en cytosylgruppe, en thymi-nylgruppe, en uracilgruppe eller et derivat af nukleinsyrebasen med en polargruppe eventuelt beskyttet af en beskyttelsesgruppe; hvor en kondensationsreaktion udføres ved på forhånd at opløse den hydro-fobiske gruppebundne nukleosid eller hydrofobiske gruppebundne oligonu-kleotid og nukleosidphosphoramidit-forbindelsen i et ikke-polært opløsningsmiddel, og at bringe den resulterende opløsning i kontakt med syre-/azol-kompleksforbindelsen eller en opløsning, der indeholder kompleksforbindelsen.
2. Fremgangsmåde til syntetisering af en oligonukleotid ifølge krav 1, hvor en opløsning til opløsning af den hydrofobiske gruppebundne nukleosid eller den hydrofobiske gruppebundne oligonukleotid og nukleosidphosphoramidit-forbindelsen i et ikke-polært opløsningsmiddel og en opløsning til opløsning af syre-/azol-kompleksforbindelsen i et ikke polært opløsningsmiddel fremstilles separat og blandes.
3. Fremgangsmåde til syntetisering af en oligonukleotid ifølge krav 1 eller 2, hvor en opløsning til opløsning af den hydrofobiske gruppebundne nukleosid eller den hydrofobiske gruppebundne oligonukleotid og nukleosidphospho-ramidit-forbindelsen i et ikke-polært opløsningsmiddel og en opløsning til opløsning af syre-/azol-kompleksforbindelsen i et ikke-polært opløsningsmiddel ledes ind i en strømningsvej med henblik på kontinuerligt at føre kondensationsreaktionen ind i strømningsvejen.
4. Fremgangsmåde til syntetisering af en oligonukleotid ifølge et af kravene 1 til 3, hvor udgangsråmaterialet af den hydrofobiske gruppebundne nukleosid er en forbindelse repræsenteret ved Formel (3):
(3) hvor R6 hver især uafhængigt betegner en alkylgruppe med 18 til 30 carbon-atomer; og R1, X, Z og y betegner det samme som ovenfor.
5. Fremgangsmåde til syntetisering af en oligonukleotid ifølge krav 4, hvor R6 i Formel (3) er en octadecylgruppe.
