Claims (18)
1. Fremgangsmåde til måling af ethanolaminphosphat, omfattende: et første trin med tilsætning af et enzym, som kan katalysere en reaktion, der danner acetaldehyd ud fra ethanolaminphosphat, til en prøve og udføre en første enzymatisk reaktion til dannelse af acetaldehyd, phosphorsyre og ammoniak; og et andet trin med kvantificering af mindst en af det/den resulterende acetaldehyd, phosphorsyre og ammoniak til bestemmelse af en mængde af ethanolaminphosphat i måleprøven.
2. Målefremgangsmåde ifølge krav 1, hvor det andet trin omfatter: et undertrin med tilsætning til måleprøven efter den første enzymatiske reaktion, af acetaldehyd-dehydrogenase (ALDH) og oxideret nicotinamid-adenindinukleotid (NAD+) og udførelse af en anden enzymatisk reaktion til dannelse af reduceret nicotinamid-adenindinukleotid (NADH); og et andet undertrin med spektrokemisk kvantificering af det reducerede nicotinamid-adenindinukleotid (NADH) til kvantificering af acetaldehyd i måleprøven efter den første enzymatiske reaktion.
3. Målefremgangsmåde ifølge krav 1 eller 2, hvor der i første trin yderligere tilsættes et coenzym til måleprøven.
4. Målefremgangsmåde ifølge krav 3, hvor coenzymet er pyridoxalphosphat.
5. Målefremgangsmåde ifølge et hvilket som helst af kravene 1-4, hvor enzymet til anvendelse i det første trin er et protein, som har en aminosyrese-kvensregion med et GabT-domæne bestående af følgende aminosyrese-kvens: GXXXBADEBQXGFZRXG, hvor X betyder en aminosyre, B betegner en forgrenet aminosyre udvalgt fra gruppen bestående af I, L og V, og Z betyder G eller A.
6. Målefremgangsmåde ifølge et hvilket som helst af kravene 1-4, hvor enzymet til anvendelse i det første trin er: (1) et protein bestående af en aminosyresekvens betegnet med SEQ ID NO:1 eller 2, (2) et protein bestående af en aminosyresekvens, hvor, i aminosyresekven-sen betegnet med SEQ ID NO:1 eller 2, en aminosyresekvens er blevet dele-teret, substitueret eller tilsat, eller (3) et protein bestående af en aminosyresekvens med mindst 90 % homologi med aminosyresekvensen betegnet med SEQ ID NO:1 eller 2.
7. Program til muliggørelse af, at en computer udfører fremgangsmåden til måling af ethanolaminphosphat ifølge et hvilket som helst af kravene 1-6.
8. Program ifølge krav 7, yderligere omfattende et trin med bestemmelse, baseret på en måleværdi af ethanolaminphosphat i måleprøven, af hvorvidt et individ, som har tilvejebragt måleprøven, er en depressiv patient, og et andet trin med udlæsning af et således opnået bestemmelsesresultat.
9. Registreringsmedium, der er læsbart af computeren med programmet ifølge krav 7 eller 8 lagret deri.
10. Målesystem med programmet ifølge krav 7 eller 8 inkorporeret deri.
11. Anvendelse af et reagens omfattende et enzym, som kan katalysere en reaktion, der danner acetaldehyd ud fra ethanolaminphosphat som et in vitro-ethanolaminphosphat-målereagens.
12. Anvendelse af reagenset ifølge krav 11, hvor reagenset yderligere omfatter et coenzym for enzymet.
13. Anvendelse af reagentet ifølge krav 12, hvor coenzymet er pyridoxalphosphat.
14. Anvendelse af reagenset ifølge et hvilket som helst af kravene 11-13, hvor enzymet er et protein, som har en aminosyresekvensregion med et GabT-domæne bestående af følgende aminosyresekvens: GXXXBAD EBQXG FZRXG, hvor X betyder en aminosyre, B betegner en forgrenet aminosyre udvalgt fra gruppen bestående af I, L og V, og Z betyder G eller A.
15. Middel omfattende ethanolaminphosphat-målereagenset ifølge et hvilket som helst af kravene 11-14 til anvendelse som et middel til diagnosticering af depression.
16. Anvendelse af et kit omfattende en beholder med et enzym, som kan katalysere en reaktion, der danner acetaldehyd ud fra ethanolaminphosphat, indeholdt separat deri til måling af ethanolaminphosphat in vitro.
17. Anvendelse ifølge krav 16, hvor kittet yderligere omfatter en anden beholder med et coenzym til enzymet, hvilket coenzym er indeholdt separat i den anden beholder.
18. Anvendelse ifølge krav 16 eller 17, hvor enzymet er et protein med en aminosyresekvensregion med et GabT-domæne bestående af følgende ami-nosyresekvens: GXXXBADEBQXGFZRXG, hvor X betyder en aminosyre, B betegner en forgrenet aminosyre udvalgt fra gruppen bestående af I, L og V, og Z betyder G eller A.
