Claims (18)
1. Glucoamylasevariant, som omfatter en substitution i mindst en position svarende til position 79 i det modne polypeptid ifølge SEQ ID NO: 2, hvor varianten har glucoamylaseaktivitet, og hvor varianten er valgt fra gruppen bestående af: a) et polypeptid, som udviser mindst 85% sekvensidentitet med det modne polypeptid ifølge SEQ ID NO: 2; b) et polypeptid, som kodes for af et polynukleotid, som udviser mindst 85% identitet med det modne polypeptids kodningssekvens ifølge SEQ ID NO: 1; og (c) et fragment af det modne polypeptid ifølge SEQ ID NO: 2, som har glucoamylaseaktivitet,
2. Variant ifølge krav 1, hvor antallet af substitutioner er 1 -20, f.eks. 1 -10 og 1 -5, såsom 1, 2. 3, 4, 5, 6, 7, 8, 9 eller 10 substitutioner.
3. Variant ifølge et hvilket som helst af de foregående krav, hvor varianten omfatter en substitution valgt blandt K79V, K79A, K79G, K79I, K79L, K79S, K79T.

9. Anvendelse af et polypeptid ifølge et hvilket som helst af kravene 1-6 til brygning.
10. Fremgangsmåde til fremstilling af et fermenteringsprodukt ud fra stivelsesholdigt materiale, hvilken fremgangsmåde omfatter følgende trin: (a) at gøre stivelsesholdigt materiale flydende i nærværelse af en alfa-amylase; (b) at forsukre materialet, som er gjort flydende; og (c) at fermentere med en fermenterende organisme; hvor trin (a) og/eller trin (b) udføres ved anvendelse af mindst en glucoamylase ifølge et hvilket som helst af kravene 1-6.
11. Fremgangsmåde til fremstilling af et fermenteringsprodukt ud fra stivelsesholdigt materiale, hvilken fremgangsmåde omfatter følgende trin: (a) at forsukre stivelsesholdigt materiale ved en temperatur, som er lavere end den laveste gelatineringstemperatur for det stivelsesholdige materiale; og (b) at fermentere med en fermenterende organisme, hvor trin (a) udføres ved anvendelse af mindst en glucoamylase ifølge et hvilket som helst af kravene 1-6.
12. Isoleret polynukleotid, som koder for polypeptidet ifølge et hvilket som helst af kravene 1-6.
13. Nukleinsyrekonstruktion eller ekspressionsvektor, som omfatter polynukleotidet ifølge krav 12, hvilket polynukleotid er operativt forbundet til en eller flere kontrolsekvenser, der styrer fremstillingen af polypeptidet i en ekspressionsvært.
14. Rekombinant værtscelle, som omfatter polynukleotidet ifølge krav 12, hvilket polynukleotid er operativt forbundet til en eller flere kontrolsekvenser, der styrer fremstillingen af polypeptidet.
