Claims (15)

  1. MUTANTE DELTA-9-ELONGASER OG ANVENDELSE HERAF TIL FREMSTILLING AF POLYUMÆTTEDE FEDTSYRER
    1. Isoleret polynukleotid, der omfatter: (a) en nukleotidsekvens, der koder for et mutant polypeptid, der har delta-9-elongaseaktivitet, og som har en aminosyresekvens som vist i SEQ ID NO: 22, hvor SEQ ID NO: 22 adskiller sig fra SEQ ID NO: 10 ved mindst én aminosyremutation, hvilken eller hvilke mutationer er udvalgt fra gruppen bestående af: i) en L35G-mutation; ii) en L35F-mutation; iii) en L35M-mutation; iv) en L35G-mutation og mindst én anden mutation, der er udvalgt fra gruppen bestående af: S9A, S9D, S9G, S9I, S9K, S9Q, Q12K, A21D, A21T, A21V, V32F, Y84C, L108G, G127L, W132T, M143N, M143W, L161T, L161Y, W168G, I179M, I179R, C236N, Q244N, A254W og A254Y; v) L35G-, A21V-, L108G- og I179R-mutationer; vi) L35G-, W132T- og I179R-mutationer; vii) L35G-, S9D-, Y84C- og I179R-mutationer; viii) L35G-, Y84C-, I179R- og Q244N-mutationer; ix) L35G-, A21V-, W132T-, I179R- og Q244N-mutationer; x) L35M- og Q107E-mutationer; xi) K58R-og I257T-mutationer; xii) en D98G-mutation og xiii) L130M- samt V243A-mutationer; eller (b) et komplement af nukleotidsekvensen fra del (a), hvor komplementet og nukleotidsekvensen består af det samme antal nukleotider og er 100 % komplimentære.
  2. 2. Isoleret polynukleotid ifølge krav 1, hvor nukleotidsekvensen koder for et mutant polypeptid, der er udvalgt fra gruppen bestående af SEQ ID NO: 59, SEQ ID NO: 29, SEQ ID NO: 32, SEQ ID NO: 35, SEQ ID NO: 38, SEQ ID NO: 41, SEQ ID NO: 62, SEQ ID NO: 87, SEQ ID NO: 101, SEQ ID NO: 104, SEQ ID NO: 107 og SEQ ID NO: 110.
  3. 3. Isoleret polynukleotid ifølge krav 2, hvor nukleotidsekvensen koder for et mutant polypeptid som vist i SEQ ID NO: 59.
  4. 4. Mutant polypeptid, der har delta-9-elongaseaktivitet, der er kodet for af det isolerede polynukleotid ifølge krav 1, hvor proteinsekvensen fortrinsvis er udvalgt fra gruppen bestående af: SEQ ID NO; 59, SEQ ID NO: 29, SEQ ID NO: 32, SEQ ID NO: 35, SEQ ID NO: 38, SEQ ID NO: 41, SEQ ID NO: 62, SEQ ID NO: 87, SEQ ID NO: 101, SEQ ID NO: 104, SEQ ID NO: 107 og SEQ ID NO: 110.
  5. 5. Mutant polypeptid ifølge krav 4, hvor den procentvise substratomdannelse af linolsyre til eicosadiensyre af delta-9-elongasen er mindst 110 % sammenlignet med den procentvise substratomdannelse af linolsyre til eicosadiensyre af polypeptidet som vist i SEQ ID NO: 10, hvor mere fortrinsvis den procentvise substratomdannelse af linolsyre til eicosadiensyre er mindst 120 % sammenlignet med den procentvise substratomdannelse af linolsyre til eicosadiensyre af polypeptidet som visti SEQ ID NO: 10.
  6. 6. Rekombinant konstrukt, der omfatter det isolerede polynukleotid ifølge krav 1, og som er operativt forbundet med mindst én regulatorisk sekvens.
  7. 7. Celle transformeret med det rekombinante konstrukt ifølge krav 6.
  8. 8. Transformeret celle ifølge krav 7, hvor cellen er udvalgt fra gruppen bestående af: planter, bakterier, gær, alger, euglenoider, stramenopiler, oomyceter og svampe.
  9. 9. Transformeret celle ifølge krav 8, hvor cellen er en olieholdig gær, der producerer mindst omkring 25 % af sin tørcellevægt som olie.
  10. 10. Transformeret celle ifølge krav 9, hvor den olieholdige gær endvidere omfatter mindst ét rekombinant DNA-konstrukt, der omfatter et isoleret polynukleotid, der er operativt bundet til mindst én regulatorisk sekvens, hvilket rekombinant DNA-konstrukt koder for et polypeptid, der er udvalgt fra gruppen bestående af: delta-4-desaturase, delta-5-desaturase, delta-8-desaturase, delta-6-desaturase, delta-9-desaturase, delta-12-desaturase, delta-15-desaturase, delta-17-desaturase, Ci4/]6-elongase, Ci6/is-elongase, Cis/20-elongase og C20/22-elongase.
  11. 11. Transformeret celle ifølge krav 10, hvor olie, der er produceret af den olieholdige gær, omfatter en langkædet polyumættet fedtsyre, der er udvalgt fra gruppen bestående af: arachidonsyre, eicosadiensyre, eicosapentaensyre, eicosatetraensyre, eicosatriensyre, dihomo-y-linolensyre, docosatetraensyre, docosapentaensyre og docosahexaensyre.
  12. 12. Transformeret celle ifølge et hvilket som helst af kravene 8 til 11, hvor gæren er en olieholdig gær, der er udvalgt fra gruppen bestående af: Yarrowia, Candida, Rhodotorula, Rhodosporidium, Cryptococcus, Trichosporon og Lipomyces.
  13. 13. Transformeret celle ifølge krav 12, hvor cellen er en Yarrowia lipolytica.
  14. 14. Fremgangsmåde til fremstilling af en polyumættet fedtsyre, hvilken fremgangsmåde omfatter: a) tilvejebringelse af en olieholdig gær, der omfatter: i) et rekombinant konstrukt, der er operativt bundet til mindst én regulatorisk sekvens, hvor det rekombinante konstrukt omfatter et isoleret polynukleotid, der koder for et mutant polypeptid, der har delta-9-elongaseaktivitet og har en amino syre sekvens som vist i SEQ ID NO: 22, hvor SEQ ID NO: 22 adskiller sig fra SEQ ID NO: 10 ved mindst én amino syremutation, hvilke(n) mutation(er) er udvalgt fra gruppen bestående af: (a) en L35G-mutation; (b) en L35F-mutation; (c) en L35M-mutation; (d) en L35G-mutation og mindst én anden mutation, der er udvalgt fra gruppen bestående af: S9A, S9D, S9G, S9I, S9K, S9Q, Q12K, A21D, A21T, A21V, V32F, Y84C, L108G, G127L, W132T, M143N, M143W, L161T, L161Y, W168G, I179M, I179R, C236N, Q244N, A254W og A254Y; (e) L35G, A21V, L108G og I179R-mutationer; (Γ) L35G-, W132T- og I179R-mutationer; (g) L35G-, S9D-, Y84C- og I179R-mutationer; (h) L35G-, Y84C-, I179R- og Q244N-mutationer; (i) L35G-, A21V-, W132T-, I179R- og Q244N-mutationer; (j) L35M- og Q107E-mutationer; (k) K58R- og I257T-mutationer; (l) en D98G-mutation og (m) L130M- samt V243A-mutationer, og ii) en kilde til substratfedtsyre, der er udvalgt fra gruppen bestående af linolsyre og alpha-linolensyre; b) dyrkning af gæren fra trin (a) under forhold, hvor det rekombinante konstrukt, der koder for det mutante polypeptid med delta-9-elongaseaktivitet, udtrykkes, og substratfedtsyren omdannes til produktfedtsyre, hvor linolsyre omdannes til eicosadiensyre, og alpha-linolensyre omdannes til eicosatriensyre og; c) eventuelt indvinding af produktfedtsyren fra trin (b), hvor produktfedtsyren fortrinsvis igen indvindes i en oprenset olie.
  15. 15. Rekombinant mikrobiel værtscelle, der producerer en olie, der omfatter mindst 22,5 vægtprocent eicosapentaensyre målt som en vægtprocent af tørcellevægten, hvilken rekombinant mikrobiel værtscelle omfatter mindst ét mutant delta-9-elongasepolypeptid, hvor det mutante delta-9-elongasepolypeptid omfatter en aminosyresekvens som vist i SEQ ID NO: 59, hvor værtscellen omfatter et ekspressionskonstrukt, der indeholder en polynukleotidsekvens, der koder for det mutante polypeptid.
