Den foreliggende opfindelse angår hidtil ukendte penem-forbindelser, fremgangsmåde til fremstilling heraf og i farmaceutiske og veterinære præparater indeholdende disse forbindelser.

De omhandlede forbindelser er kvaternære ammoniumcarboxy-later med den i krav 1 angivne formel I og deres farmaceutiske eller veterinære acceptable salte.
10 Den foreliggende opfindelse omfatter alle mulige geometriske og optiske isomere af forbindelserne med formlen I, enten i form af isomere blandinger eller i form af de enkelte adskilte isomere.
15 Som allerede nævnt omfatter opfindelsen også farmaceutisk eller veterinært acceptable salte af forbindelserne med formel I. Disse salte kan både være salte med syrer, som f.eks. med eddikesyre, citronsyre, vinsyre, fumarsyre eller methansulfonsyre og salte med baser, som f.eks. alka- 20 li- eller jordalkalimetalhydroxider, specielt natrium- og kaliumhydroxider eller med organiske baser, som f.eks. triethylamin, pyridin, benzylamin eller collidin, herunder aminosyrer, som f.eks. lysin eller procain.
25 Opfindelsen omfatter også indre salte, dvs. zwitterioner.

Omsætningen mellem en forbindelse med formlen II og en forbindelse med formlen III kan udføres i et passende organisk, fortrinsvis aprotisk opløsningsmiddel, der f.eks. kan være tetrahydrofuran, dimethylformamid, acetone eller 25 en halogeneret carbonhydrid som dichlormethan.
Omsætningstemperaturen kan fortrinsvis variere mellem ca.
-100 °C og ca. +40 °C, fortrinsvis mellem -70 °C og +15 °C.

Ifølge en foretrukken fremgangsmåde omsættes forbindelsen med formlen IV med det passende sulfonylanhydrid eller sulfonylhalogenid i nærvær af et overskud, sædvanligvis i en mængde lig med eller to gange den molære ækvivalente 20 mængde af den Ønskede forbindelse med formlen II: i dette tilfælde isoleres forbindelsen med formlen III endog ikke fra reaktionsblandingen, fordi den reagerer in situ med forbindelsen med formlen II.
25 Den foretrukne fremgangsmåde udføres sædvanligvis under anvendelse af tør dichlormethan som opløsningsmiddel ved temperaturer fra ca. -70 °C til +25 °C.
Når en forbindelse med formlen II omsættes med en forbin-30 delse med formlen III, hvor L betegner halogen, kan tilstedeværelsen af et sølvsalt, specielt et som er opløseligt i reaktionsmediet, f.eks. AgC104, være en fordel.
En forbindelse med formlen III, hvori L betegner halogen, 35 kan fremstilles ud fra den tilsvarende carbinol-precursor med formlen IV efter en modificeret Mitsunobu-reaktion, hvor carbinolen omsættes med organisk aminhydrohalogenid DK 170341 B1 11 som f.eks. methoxyaminhydrochlorid, pyridinhydrochlorid, pyridinhydrobromid og det foruddannede kompleks opnået ud fra diethylazodicarboxylat og triphenylphosphin, idet omsætningen udføres f.eks. i tetrahydrofuran eller methy-5 lenchlorid, fortrinsvis omkring stuetemperatur.
Alternativt kan ved en modificeret Mitsunobu-reaktion et zinkhalogenid, som f.eks. zinkchlorid, zinkbromid og zinkiodid, anvendes i stedet for det organiske aminhydro-10 halogenid under betingelser, der i det væsentlige svarer til de, der er beskrevet i J. Org. Chem., 1984, 49, 3027.
Alternativt kan en forbindelse med formlen III, hvori L betegner halogen, fås ud fra carbinolen med formel IV ved 15 mere almindelig metoder, der indebærer omsætning med uorganiske halogenider som f.eks. S0C12, PClg, PC13, PBr3, P0C13, P0Br3.
En yderligere metode, der er velkendt i litteraturen, 20 nemlig omsætningen med PPhg i CCl^, kan anvendes til fremstilling af forbindelsen med formel III, hvori L betegner chlorid.
En forbindelse med formlen III, hvor L betegner iod, kan 25 fremstilles ved en halogenid-ombytningsreaktion ud fra en forbindelse med formel III, hvor L betegner chlor eller brom og natriumiodid, idet denne omsætning fortrinsvis udføres med acetone ved temperaturer fra 0 °C til +60 °C (tilbagesvalingstemperatur).

Forbindelserne med formel II, V, VI, VII og VIII er kendte forbindelser og kan fremstilles ud fra kendte forbindelser på kendte måder.

De omhandlede forbindelser med formel I er kraftige, bredspektrede antibakterielle midler. I forhold til andre penem-forbindelser, f.eks. natriumsaltene af penem-forbindelser med formlen IV eller forbindelserne med formel 20 ir hvori Q betegner en amin i stedet for kvaternært ammonium, er de sædvanligvis i besiddelse af betydelig forøget in vitro aktivitet over for både Gram-positive og .Gram-negative stammer.
25 Herudover udviser de i forhold til andre penem-forbindelser usædvanlige forlængede plasmaniveauer og, dersom de afprøves in vivo efter parenteral administrering, udviser de en særdeles høj terapeutisk effektivitet ved behandling af infektioner, der er forårsaget af både Gram-posi-30 tive og Gram-negative bakterier, idet deres toxicitet på den anden side er ganske ringe.

20 intravenøst eller intramuskulært (som opløsninger eller suspensioner), idet den intravenøse administreringsmåde foretrækkes i alvorlige situationer; ved inhalation i form af aerosoler eller opløsninger til tågedannelse; in-travaginalt i form f.eks. af stave eller topisk i form af 25 lotioner, cremer og salver. Den daglige dosis ligger i området fra ca. 0,5 til ca.
100 mg pr. kg legemsvægt, idet er tages hensyn til dyre- DK 170341 Bl 22 arten, til alderen, vægten og tilstanden af den patient, der skal behandles, og af hyppigheden og administreringsvejen.
5 En foretrukken administreringsmåde for de omhandlede forbindelser er den parenterale: i dette tilfælde administreres forbindelserne f.eks. til voksne mennesker i en mængde på fra ca. 250 mg til ca. 1000 mg pr. dosis, fortrinsvis ca. 500 mg pr. dosis i 1 til 4 gange om dagen, 10 idet forbindelsen er opløst i et passende opløsningsmiddel som f.eks. sterilt vand eller lidocain-hydrochlorid-opløsning til intramuskulære injektioner, og sterilt vand, fysiologisk saltvand, dextroseopløsning eller almindelige intravenøse væsker eller elektrolyter til in-15 travenøse injektioner. Yderligere kan de omhandlede forbindelser administreres profylaktisk, dvs. som rensende eller overfladedesinficerende præparater f.eks. i en koncentration på ca. 0,2 til 1 vægt-% af forbindelsen blandet med, suspenderet eller opløst i et almindeligt inert 20 tørt eller vandigt bærestof, der anvendes til afvaskning eller spray.
De kan også anvendes som fodertilskud til dyrefoder.
25 Følgende eksempler illustrerer opfindelsen nærmere, idet dog eksemplerne 10 og 11 illustrerer fremstilling af mellemprodukter .
EKSEMPEL 1 30 (5R,6S)-6-[(IR)-hydroxyethyl]-2-[4-(N-methylpyrrolidino-methyl)phenyl]-penem-3-carboxylat (forbindelse 1)
En opløsning af (3S)-[(lR)-tert-butyldimethylsilyloxy-35 ethyl]-(4R)-[4-(tert-butyldiphenylsilyloxyraethyl)-phe nyl] -acetylthio-1-[(a-triphenylphosphoranyliden)allyloxy-carbonylmethyl]azetidin-2-on (1,6 g) i tør destilleret DK 170341 B1 23 tetrahydrofuran (16 ml) behandles med eddikesyre (0,95 ml) og tetrabutylammoniumfluoridtrihydrat (1,25 g).
Efter 6 timers omrøring ved stuetemperatur fjernes opløs-5 ningsmidlet i vakuum, og remanensen udrystes mellem fortyndet vandig NaHCO^ og ethylacetat.
Remanensen fra det organiske lag renses ved Si02~kromatograf i, hvilket giver (3S)-[(lR)-tert-butyldimethylsilyl-10 oxyethyl]-(4R)-[4-(hydroxymethyl)phenyl]-acetylthio-Ι Ε (a-triphenylphosphoranyliden)allyloxycarbonylmethyl]aze-tidin-2-on (1,0 g) som et skum.
En opløsning af dette materiale i toluen (30 ml) opvarmes 15 7 timer med tilbagesvaling; opløsningsmidlet fjernedes herefter, og remanensen hældtes gennem en SK^-søjle (EtOAc-cyclohexan som elueringsmidler). Der opnåedes al-lyl (5R,6S)-6-[(IR)-tert-butyldimethylsilyloxyethyl]-2- [4-(hydroxymethyl)phenyl]-penem-3-carboxylat (375 mg) som 20 voksagtig substans; IR; » (CH„C10) 1790, 1710 cm-"*·; NMR (60 MHz, CDClg + D20) = δ 0,08 (6H, s, SiMe2), 0,88 (9H, s, SiBu1), 1,28 (3H, d, J = 6 Hz, CH3-CH), 3,68 (IH, dd, J = 1,5 og 6 Hz, H-6), 4,2 (IH, m; CH^CH), 4,5-4,7 (4H, m, ArCH2 + C02CH2), 5,2 og 5,35 (2H, hvert m, =CH2), 25 5,65 (IH, d, J - 1,5 Hz, H-5), 5,9 (IH, m, CHgCH-CHg), 7- 7,5 (4H, m, Ar) ppm.
En opløsning af ovennævnte mellemprodukt (225 mg) i tør ethanolfri dichlormethan (10 ml), afkølet ved -50 °C un-30 der argon, behandledes efter hinanden med N-methylpyrro-lidin (0,25 ml) og trifluormethansulfonsyreanhydrid (0,125 ml). Man lod reaktionsblandingens temperatur stige til -20 °C, og efter 20 minutters forløb bratkøledes med 0,1 M vandig saltsyre. Det organiske lag fraskiltes, der 35 vaskedes med saltvand, tørredes og inddampedes til en remanens, der tritureredes i ethylacetat-ethylether-blan-dinger, hvorved opnåedes allyl (5R,6S)-6-[(lR)-tert-bu- DK 170341 B1 24 tyldimethylsilyloxyethyl]-2-[4-(N-methylpyrrolidinome-thyl)phenyl]-penem-3-carboxylat som et pulver (150 mg); IR: » „ (CHClo film) 1785, 1710 cm-1.
luaX o 5 Dette materiale optoges i tetrahydrofuran (9 ml) og eddikesyre (0,18 ml), og tetrabutylammoniumfluoridtrihydrat (0,3 g) tilsattes. Den klare opløsning henstod 30 timer ved stuetemperatur, hvorefter den koncentreredes og ledtes gennem en silicagel-søjle pakket med CH2C12.

Ammoniumsaltene elueredes med en gradient af MeCN i CH2C12; herefter elueredes forbindelsen med vandig MeCN.
De produktholdige fraktioner udsaltedes (NaCl), og den fraskilte vandige fase ekstraheredes med frisk MeCN.
15 MeCN-fasen og MeCN-ekstrakterne opsamledes, der tørredes (MgS04) og inddampedes til opnåelse af salte (Cl-, acetat) af allyl (5R,6S)-6-[(lR)-hydroxyethyl]-2-[4-(N-me-thyl-N-pyrrolidinomethyl)phenyl]-penem-3-carboxylat (90 mg); IR »max (film) 3400-3200, 1785, 1710 cm-1).

Ovennævnte forbindelse (90 mg) i en 1:1 tetrahydrofuran-dichlormethan-blanding (6 ml) behandledes efter hinanden med eddikesyre (0,03 ml), triphenylphosphin (9 mg) og te-trakis (triphenylphosphin) Pd(0) (9 mg).

Efter 10 minutters omrøring gentoges tilsætningen af HOAc, PPhg og katalysatoren, og efter yderligere 15 minutters forløb fjernedes opløsningsmidlet i vakuum. Remanensen opløstes i demineraliseret vand og rensedes ved 30 omvendt fasekromatografi (LiChroprep RP-18 Merck), der først elueredes med vand, herefter med en gradient i MeCN (fra 95:5 til 1:1) og til slut med H20-MeCN-Et0H (4:6:1).
De passende fraktioner (TLC på Si02, i-Pr0H/H20/H0Ac 5:1:1, langsomt løbende plet) blev slået sammen og fryse-35 tørret, hvorved der opnåedes 40 mg af titelforbindelsen; IR: (KBr) 3400, 177°. 1620 cm”1? En opløsning af allyl (5R,6S)-[tert-butyldimethylsilyl-20 oxyethyl]-2-[4-(hydroxymethyl)phenyl]-penem-3-carboxylat (300 mg) fremstillet som beskrevet i eksempel 1 i tør, ethanolfri dichlormethan (16 ml) behandledes med pyridin (1,4 ml) og trifluormethansulfonsyreanhydrid (0,8 ml) ved -40 °C under nitrogen. Efter 30 minutters forløb tilsat-25 tes 0,1 M vandig saltsyre (10 ml), det organiske lag fraskiltes, vaskedes med en yderligere mængde 0,1 N saltsyre, tørredes og inddampedes.
Den fremkomne remanens (410 mg) opløstes i tetrahydrofu-30 ran (20 ml) og omrørtes 24 timer ved stuetemperatur i nærvær af eddikesyre (1 ml) og tetrabutylammoniumfluorid-trihydrat (1,6 g). Fjernelse af opløsningsmidlet og rensning ved hjælp af silicagel-søjlekromatografi (C^C^, derefter C^C^-MeCN, dernæst MeCN-^O) gav ally lesteren 35 af titelforbindelsen (250 mg); IR: » (KBr): 3400, « mox 1780, 1705 cm ? UV: (EtOH) 256 og 336 um.

En blanding af kalium-2-hydroxymethylbenzoat (9,45 g) og chloracetone (5 ml) i tør dimethyl formamid (100 ml) omrørtes 3 timer ved stuetemperatur. Ethylacetat og saltop- DK 170341 B1 27 løsning sattes derefter til, og det organiske lag tørredes efter yderligere vask med vand og inddampedes, hvorved acetonyl-2-hydroxymethylbenzoat (10 g) opnåedes som et hvidt pulver. En opløsning af dette materiale (1 g) i 5 dimethylformamid (20 ml) omrørtes 3 timer i nærværelse af tert-butyldiphenylsilylchlorid (2,32 ml) og imidazol (0,864 g).
Reaktionsblandingen udrystedes mellem ethylacetat og van-10 dig saltsyre; den tørrede organiske fase inddampedes, og remanensen tritureredes i N-hexan, hvorved der opnåedes 2-(tert-butyldiphenylsilyloxymethyl)benzoat (0,94 g), som et hvidt fast stof. IR; » (CHC1«) 1740 sh, 1725 cm""·*·;

Til denne forbindelse (0,78 g) opløst i acetonitril (25 ml) sattes en opløsning af N/10 NaOH (26,3 ml). Blandingen syrnedes med fortyndet saltsyre under omrøring, det organiske lag fraskiltes, vaskedes 2 gange med saltopløsning, tørredes og inddampedes.

Den faste remanens tritureredes i n-hexan og filtreredes, hvorved der opnåedes 2-(tert-butyldiphenylsilyloxyme-thyl)benzoesyre (0,54 g) som et hvidt fast stof med smeltepunkt 130-132 °C.

Denne forbindelse, der opløstes i tør ethanolfri dichlor-methan (30 ml), omrørtes 6 timer ved stuetemperatur i nærværelse af thionylchlorid (0,5 ml). Reaktionsblandingen inddampedes i vakuum fra benzen (2 gange), hvorved 35 der opnåedes 2-(tert-butyldiphenylsilyloxymethyl)benzoyl-chlorid, der straks anvendtes i det efterfølgende trin.

EKSEMPEL 7 10 (5R, 6S) - 6- [ (IR) -hydroxyethyl] - 2- [3 - (N-methylpyrrolidino) -propyl]penem-3-carboxylat (forbindelse 31) METODE A 15
Til en opløsning af allyl (5R,6S)-6-[(lR)-tert-butyldime-thylsilyloxyethyl] -2- (3-hydroxypropyl )penem-3-carboxylat (400 mg) i tør ethanolfri dichlormethan (15 ml) afkølet ved -30 °C under nitrogen sattes efter hinanden N-methyl-20 pyrrolidin (0,47 ml) og trifluormethansulfonsyreanhydrid (0,36 ml).
Badet fjernedes, og reaktionsblandingen hens tod ved stuetemperatur, indtil det meste af udgangsmaterialet var 25 forsvundet (kontrol med TLC). Yderligere dichlormethan og 0,1 M vandig saltsyre tilsattes under omrøring, og den fraskilte organiske fase vaskedes med saltopløsning, tørredes og inddampedes, hvorved der opnåedes en olieagtig remanens, der for det meste bestod af salte (chlorid, 30 triflag) af allyl (5R,6S)-6-[(lR)-tert-butyldimethylsi- lyloxyethyl] - 2- [3 - (N-methylpyrrolidinopropyl ] -penem-3-carboxylat.
Uden rensning sattes dette materiale til en opløsning af 35 tetrabutylammoniumfluoridtrihydrat (1,8 g) i tetrahydro- furan (15 ml) og eddikesyre (1 ml), og desilyleringen kontrolleredes med TLC. Når omsætningen var løbet til en DK 170341 B1 34 de, fjernedes opløsningsmidlet i vakuum, og remanensen rensedes ved flashkromatografi (CHC^-MeCN og derefter MeCN-H20).
5 Den fremstillede forbindelse (0,27 g), allylesteren af titelforbindelsen, omrørtes 30 minutter under nitrogen i en blanding af triphenylphosphin (50 mg) og tetrakis (triphenylphosphin) palladium (50 mg) i dichlormethan-te-trahydrofuran (7 ml + 7 ml) og eddikesyre (0,5 ml).

Det meste af opløsningsmidlet fjernedes i 25 vakuum, og remanensen udrystedes mellem ethylacetat og vand.
Det tørrede organiske lag koncentreredes til opnåelse af en remanens, der rensedes ved kortvejs-flashkromatografi, 30 hvorved man opnåede allyl (5R,6S)-6-[(lR)-tert-butyldime- thylsilyloxyethyl]-2-(3-iodpropyl)penem-3-carboxylat som en sirup (220 mg); IR: » (film) 1790, 1710 og 1580 ^ ΙΠοΧ cm . Til en opløsning af ovennævnte materiale i tør te-trahydrofuran (30 ml) sattes N-methylpyrrolidin (0,1 ml) 35 og sølvperchlorat (165 mg). Reaktionsblandingen omrørtes 6 timer ved 0 °C, og herefter lod man temperaturen stige til stuetemperatur i en nitrogenatmosfære i mørke. Opløs- DK 170341 B1 35 ningsmidlet fjernedes, og remanensen udrystedes mellem dichlormethan og 0,1 N saltsyre ved 5 °C. Det organiske lag fraskiltes, filtreredes og inddampedes, hvorved der opnåedes en remanens, der bestod af salte (hovedsageligt 5 chlorid) af allyl (5R,6S)-6-[(lR)-tert-butyldimethylsi-lyloxyethyl]-2-[3-(N-methylpyrrolidino)propyl]penem-3-carboxylat.
Dette mellemprodukt oparbejdedes ved desilylering og de-10 allylering som beskrevet under fremgangsmåde A ovenfor, hvorved der opnåedes en identisk prøve af titelforbindelsen.

Efter 15 minutters forløb tilsattes dichlormethan (250 ml); det 20 organiske lag vaskedes med vand (2 x 200 ml), tørredes (Na2S04 og inddampedes, hvorved opnåedes 4,7 g urenset sølv-(3S)-[(IR)-tert-butyldimethylsilyloxyethyl]-1-(1-al-lyloxycarbonyl-l-triphenylphosphoranylidenmethyl)-2-oxo-azetidinyl-4-thiolat som et brunt skum.

En opløsning af e-bromtoluensyre (1,08 g) i tør benzen (10 ml) omrørtes 2 timer med thionylchlorid (0,73 ml) og 30 tør DMF (5 dråber). Reaktionsblandingen inddampedes til tørhed, optoges i toluen og inddampedes atter. Den uren-sede forbindelse omkrystalliseredes med n-hexan (10 ml, aktivt kul), hvorved der opnåedes ren a-bromtoluensyre-chlorid (0,95 g) som hvide små blade.

Herefter fjernedes opløsningsmidlet i vakuum, ether tilsattes, og suspensionen omrørtes 1 time ved stuetempera-30 tur.
Det gule faste materiale opsamledes herefter, hvilket gav 100 mg allyl (5R,6S)-6-[(lR)-tert-butyldimethylsilyloxy-ethyl]-2-{4-[3-hydroxypyridino)methyl]phenyl}penem-3-oar-35 boxylat, bromidsalt; NMR (90 MHz, CDCl^) δ ppm 0,12 (6H, S), 0,82 (9H, s), 1,20 (3H, d, J = 6,0 Hz), 3,70 (IH, dd, J = 1,6 og 5,0 Hz), 4,18 (IH, dd, J = 5,0 og 6,0 Hz), DK 170341 B1 40 4,45 (2H, m), 5,02 (IH, d, J « 10,0 Hz), 5,14 (IH, d, J = 17,0 Hz), 5,61 (IH, d, J = 1,6 Hz), 5,5-5,7 (IH, m), 5,82 (2H, s), 7,50 (4H, s), 7,4-7,7 (IH, m), 7,8-8,0 (IH, m), 8,2-8,4 (IH, m), 8,68 (IH, m), 9,26 (IH, br, s).

Denne forbindelse opløstes i tør TMF og omrørtes natten over i nærværelse af eddikesyre (100 ml) og tetrabutylam-moniumfluoridtrihydrat (125 mg).
10 Reaktionsblandingen inddampedes herefter i vakuum og rensedes delvis på silicagel, der elueredes med acetoni-tril/vand-blandinger. Opløsningen inddampedes herefter i vakuum, hvilket gav allyl (5R,6S)-6-[(lR)-hydroxyethyl]- 2-{4-[(3-hydroxypyridino)methyl]phenyl}penem-3-carboxy-15 lat, bromidsalt som en gul olie (80 mg).
Denne forbindelse opløstes i THF/C^Cl.
Eddikesyre (0,8 ml), triphenylphosphin (80 mg) og heref-20 ter (PhgP)4Pd (30 mg) tilsattes herefter efter hinanden.
Omsætningen, der udførtes som angivet i de tidligere eksempler, gav 20 mg af titelforbindelsen; IR; » (KBr)

25 EKSEMPEL 13 (5R,6S)-6-[(lR)-hydroxyethyl]-2-{4-[(4-ethansulfonat-1-pyridino)methyl]phenyl}penem-3-carboxylsyre (forbindelse 30 55/a)
En blanding af allyl (5R,6S)-6-[(lR)-hydroxyethyl]-2-[4-(brommethyl)phenyl]penem-3-carboxylat (100 mg) og natri-um-4-pyridinethansulfonat (145 mg) i tør DMF omrørtes i 35 48 timer ved stuetemperatur.

Herefter inddampedes blandingen i vakuum, og remanensen rensedes ved omvendt fasekromatografi, hvorved der efter frysetørring opnåedes allyl (5R,6S)-6-[(lR)-hydroxy-ethyl]-2-{4-[(4-ethansulfonat-1-pyridino)methyl]phenyl}-5 penem-3-carboxylat; IR (KBr): 3400, 1790, 1710 cm"1; UV (H20/10% CHgCN): 336 nm; NMR (200 MHz, D20) 6 ppm 1,30 (3H, d, J = 6,4 Hz), 3,34 (4H, m), 4,05 (IH, dd, J = 1,5 og 5,6 Hz), 4,26 (IH, dq, J = 5,6 og 6,4 Hz), 5,04 (IH, d, J = 16,2 Hz), 5,6-5,8 (IH, m), 5,78 (2H, s), 5,80 (IH, 10 d, J = 1,5 Hz), 7,46-7,54 (4H, ABq, J = 8,4 Hz), 7,98 (2H, d, J = 6,7 Hz), 8,77 (2H, d, J = 6,7 Hz).
Behandling af denne forbindelse med HOAc/Ph^P/(Ph^P)^Pd under samme betingelser som angivet i eksempel 2 gav en 15 prøve (20 mg) af titelforbindelsen. IR (KBr): 3400, 1775, 1700 cm"1; UV (H20): 330, 252 nm; NMR (200 MHz, D20, t = 45 °C) δ ppm 1,56 (3H, d, J = 6,5 Hz), 3,60 (4H, m), 4,27 (IH, dd, J = 1,5 og 5,8 Hz), 4,51 (IH, dq, J = 5,8 og 6,5 Hz), 6,02 (2H, s), 6,05 (IH, d, J - 1,5 Hz), 7,69-7,79 20 (4H, ABq, J * 8,3 Hz), 8,24 (2H, d, J - 6,8 Hz), 9,00 (2H, d, J = 6,8 Hz).
EKSEMPEL 14 25 Ved at arbejde som beskrevet ovenfor i eksempel 13 og erstatte natrium-4-pyridinethansulfonat med 3-aminopyridin og natrium-4-pyridinmethancarboxylat fremstilledes henholdsvis : 1 (5R,6S)-6-[(IR)-hydroxyethyl]-2-{4-[(3-amino-pyridino)me- thyl]phenyl}-penem-3-carboxylat (forbindelse 65) IR (KBr): 3400 (bred), 1765 cm"1 og 35 (5R,6S)-6-[(IR)-hydroxyethyl]-2-{4-[(4-methancarboxylat- 1-pyridino)methyl]phenyl}penem-3-carboxylsyre (forbindelse 77) DK 170341 B1 42 IR (KBr): 3400, 1765, 1735 cm"1.
EKSEMPEL 15 5 (5R,6S)-6-[(IR)-hydroxyethyl]-2-[4-(3-carboxymethylpyri- , dino)methylphenyl]-penem-3-carboxylat
Ved at arbejde som beskrevet i eksempel 2, og erstatte pyridin med med 3-allyloxycarbonylmethyl-pyridin opnåedes 10 titelforbindelsen.

Ved at arbejde som beskrevet i eksempel 2, og erstatte 20 pyridin med 4-allyloxycarbonylpyridin opnåedes titelforbindelsen.

Claims (5)
1. Penem-forbindelser med den almene formel I med 5 (5R,6S)-konfiguration OH JL /V XV--T ?/- A-CH?-Q+
10 CR) jr 2 (I) f-N ( 0 co2<-> 0 15 hvori A betegner phenylen, 0Λ . alkylen eller Cn . alky-, (+ ) lenoxycarbony1, og Q' ' betegner en /r1 + / 2 —R - gruppe, hvori 20 ^R3 (i) R1 betegner C^_4 alkyl eventuelt substitueret med en carboxygruppe, og R2 og R^ sammen med nitrogenatomet betegner en pyrrolidinyl-, piperidinyl- el- 25 ler piperazinylgruppe; eller (ii) R^, R2 og Rg betegner sammen med nitrogenatomet en eventuelt substitueret pyridylgruppe, hvor substi-tuenteme er en eller to C^_4 alkylgrupper eller en 30 carbamoyl-, hydroxy-, amino-, carboxy-, carboxyme- thyl-, hydroxymethansulfonat- eller ethansulfonat-gruppe; forudsat at når A betegner C1-C4 alkylen, betegner R^, R2 35 ©S R3 sammen med nitrogenatomet ikke en pyridylgruppe, eller et farmaceutisk eller veterinært acceptabelt salt DK 170341 B1 44 heraf.
