Claims (17)

1. Antibiotika-frit selektionssystem omfattende en bakteriecelle der mangler et araD-gen, i hvilket en vektor som bærer et araD-gen er blevet tilføjet som en selektionsmarkør, hvor fra araD-genet af vektoren er udtrykt en aktiv L-ribulose-5-phosphat 4-epimerase.
2. Antibiotika-frit selektionssystem ifølge krav 1, hvor bakteriecellen er en Escherichia coli -celle.
3. Antibiotika-frit selektionssystem ifølge krav 2, hvor en mutation i araD-genet af vektoren introducerer en stopcodon i codon 8 af araD-genet, hvor fra araD-genet af vektoren er udtrykt en aktiv L-ribulose-5-phosphat 4-epimerase.
4. Antibiotika-frit selektionssystem ifølge krav 2, hvor E. coli er en E. coli-stamme JM109.
5. Antibiotika-frit selektionssystem ifølge krav 2 eller 3, hvor E. coli er en E. coli-stamme DH5 alpha-Tl.
6. Antibiotika-frit selektionssystem ifølge krav 2 eller 3, hvor E. coli er en E. coli-stamme AGI.
7. Antibiotika-frit selektionssystem ifølge krav 2, hvor E. co//'-stammen er stamme DH5 alpha-Tl som mangler araD-genet og ulaF-genet.
8. Antibiotika-frit selektionssystem ifølge krav 2, hvor E. co//'-stammen er stamme AGI som mangler araD-genet og sgbE-genet.
9. Antibiotika-frit selektionssystem ifølge et hvilket som helst af kravene 5-6, hvor E. co//'-stammen mangler araD-genet, u/aF-genet, og sgbE-genet.
10. Vektor omfattende et araD-gen med en mutation som genererer en stopcodon i codon 8 af araD-genet som en selektionsmarkør.
11. Vektor ifølge krav 10, hvor vektoren er en ekspressionsvektor omfattende: (a) en DNA-sekvens som koder for et kerne-forankringsprotein operativt forbundet til en heterolog promoter, kerne-forankringsproteinet omfattende (i) et DNA-bindingsdomæne som binder sig til en specifik DNA-sekvens, og (ii) et funktionelt domæne som binder sig til en kernekomponent, eller en funktionel ækvivalent deraf; og (b) en multimeriseret DNA-sekvens som danner et bindingssted til kerneforankringsproteinet, hvor vektoren mangler et papilloma-virus replikationsoprindelsessted.
12. Vektor ifølge krav 11, hvor (a) kerne-forankringsproteinet er E2-proteinet af Bovine Papilloma Virus type 1 (BPV), og (b) den multimeriserede DNA-sekvens er multiple bindingssteder for BPV E2-proteinet inkorporeret i vektoren som en klynge, hvor bindingsstederne kan være som hoved-til-hale strukturer eller kan inkluderes i vektoren med mellemrum anbragt positionering, hvor vektoren mangler et papilloma-virus replikationsoprindelsessted.
13. Vektor ifølge krav 12, yderligere omfattende en deletion i den multimeriserede DNA-sekvens.
14. Vektor ifølge krav 12, yderligere omfattende en mutation i Shine-Dalgarno-sekvensen af araD-genet.
15. Fremgangsmåde til udvælgelse af celler transformeret med et plasmid indeholdende et araD-gen som en selektionsmarkør og interessegenet, fremgangsmåden omfattende at indsætte plasmidet i den araD-deficiente værtscelle og dyrke cellerne i et dyrkningsmedium indeholdende arabinose, hvor fra araD-genet af plasmidet er udtrykt en aktiv L-ribulose-5-phosphat 4-epimerase.
16. Fremgangsmåde ifølge krav 15, hvor cellen er en Escherichia coli -celle.
17. Fremgangsmåde ifølge krav 16, hvor araD-genet af plasmidet muteres genererende en stopcodon i codon 8 af araD-genet, hvor fra araD-genet af plasmidet der udtrykkes en aktiv L-ribulose-5-phosphat 4-epimerase.
