Den foreliggende opfindelse angår hidtil ukendte mellemprodukter til fremstilling af antibakterielle forbindelser tilhørende den gruppe, der sædvanligvis kaldes cephalosporiner samt en fremgangsmåde til 5 fremstilling deraf.
Selve patentlitteraturen rummer et stort antal beskrivelser vedrørende fremstillingen af cephalosporiner ved omsætning med en silyleret kerne (såsom 7-aminocepha-losporansyre (7-ACA) eller 7-aminodesacetoxycephalosporan-10 syre) af en sidekædesyre i form af dens syrechlorid. Når denne syre indeholder en fri aminogrupper, er denne gruppe fortrinsvis blokeret, såsom ved protonisering, og der anvendes således f.eks. 2-phenylglycylchlorid,hydrochlorid til fremstilling af cephalexin. Kernens 4-carboxylgruppe 15 kan blokeres ved silylering eller ved esterificering.
Nog]e eksempler på sådanne patentskrifter er U.S.A. patentskrifter nr. 3.671.449, 3.694.437, 3.741.959, 3.957.773, 3.965.098, 4.051,131 og britisk patentskrift nr. 1.073.530. I mange tilfælde fjernes 3-acetoxygruppen i 20 7-ACA før acylering med en heterocyklisk thiol, f.eks. ceforanid (U.S.A. patentskrift nr. 4.100.346 og den deri omtalte kendte teknik), cefatrizin (U.S.A. patentskrift nr. 3.867.380), cefaparol (U.S.A. patentskrift nr.

1. I en trehalset kolbe, som er forsynet med omrører, temperaturregulator, termometer og nitrogentilførselsrør, anbringes 18 g (0,066 mol) 7-aminocephalosporansyre (som fortrinsvis har været omkrystalliseret ved hjælp af 5 toluensulfonsyremetoden) og 300 ml 0,1M pH 6,4 phosphatpuffer (20,7 g monobasisk natriumphosphat O,lH20 + 8,5 g vandfri dibasisk natriumphosphat q.s. til 2 liter).
2. Under omrøring af den i trin 1 beskrevne blanding tilsættes 1,5 g natriumbisulfit og 16 g (0,078 mol 10 dinatrium-l-carboxymethyl-5-mercaptotetrazol.
3. Under fortsat omrøring bobles nitrogen gennem blandingen i 10 minutter.
4. Under fortsat omrøring og nitrogentilførsel opvarmes opslæmningen over en 20 minutters periode til 15 56°C. I dette tidsrum tilsættes 6,5 g natriumbicarbonat i små portioner.
5. Under fortsat omrøring og nitrogentilførsel holdes j temperatur af opløsningen på 56°C i 4 timer. pH-værdien bør forblive mellem 6,2 og 6,6.
20 6. Reaktionsblandingen afkøles på et isbad til 5°C.
7. 50 ml af en 1:1 phosphorsyre/vandopløsning sættes til blandingen eller koncentreret HC1 til en pH-værdi på 2,0 til 3,0.
8. Produktet opsamles ved filtrering. Filterkagen 25 vaskes med 20 ml koldt vand efterfulgt af 200 ml kold methanol.
b) 1. 2,72_g 7-aminocephalosporansyre og 1,16 g 5-mer- capto-1-methyl-lH-tetrazol suspenderes i 14 ml vandfri 30 acetonitril. Dernæst tilsættes 4,25 g bortrifluorid-di-ethylether komplekssalt og opløses deri. Opløsningen opvarmes til 50°C i 2 timer for at lade reaktionen forløbe. Reaktionsopløsningen afkøles, 14 ml vand tilsættes, og pH-værdien indstilles på 4,0 med ammoniakvand under 35 isafkølede betingelser. De udfældede krystaller frafiltreres, vaskes med 5 ml vand og dernæst med 5 ml acetone og tørres derefter til dannelse af 3,00 g (91,5% udbytte) 7-amino-3-[5-(1-methyl-l,2,3,4-tetrazolyl)- 7

Ved tilsætning af 1,25 ml 12N saltsyre under 1 ovenfor til reaktionsopløsningen, omrøring i 2 timer under isafkølede betingelser og frafiltrering af de udfældede krystaller, vaskning med to 5 ml portioner acetone og tør- 35 ring, opnåedes 3,20 g (88,0% udbytte) af hydrochloridsalt-et af 7-amino-3-[5-(l-methyl-l,2,3,4-tetrazolyl)-thio-methyl]-Δ3-cephem-4-carboxylsyre, smeltepunkt 184-186°C. dek.

c) På samme måde som ved fremgangsmåden b ovenfor omdannes 5-mercapto-l-carboxymethyl-l,2,3,4-tetrazol til 7-amino-3-[5-(1-carboxymethyl-l,2,3,4-tetrazolyl)thiome-5 thyl]-ceph-3-em-4-carboxylsyre, der smelter ved 183°C j under dekomponering.
De i de følgende eksempler anvendte syrechlorider kan i erstattes af en lang række andre syrechlorider til fremstilling af konventionelle cephalosporiner. Eksempel 1
Fremstilling af bis-trimethylsilyl-7-aminodecephalosporan-syreester
En suspension af 7-ADCA (også kaldet 7-aminodesacet-05 oxycephalosporansyre eller 7-aminodecephalosporansyre) (10 g, 46,68 mmol) i tør methylenchlorid (100 ml) behandledes med trimethylchlorsilan (11,8 g, 13,7 ml, 108 mmol) (TMCS) efterfulgt af triethylamin (10,86 g, 14,4 ml, 107 mmol) (TEA) dråbevis i løbet af 30 minutter. Reaktions-10 blandingen analyseredes dernæst for fuldstændig silylering ved NMR analyse. NMR indicerede et integralforhold mellem -C02SiMe3 og 7-NHSiMe3 på 468:462. Der opnåedes således 100% omdannelse.

Fremstilling af trimethylsilyloxycarbonyl-7-aminodecepha-losporansyre-TMS-ester
Reaktionsblandingen af bis-trimethylsilyl-7-aminode-cephalosporansyreester forgassedes dernæst med carbondi-20 oxid ved 25°C i 4 timer under omrøring og analyseredes for fuldstændig carbonylering ved NMR. Der opnåedes 95% omdannelse.
Eksempel 3 25 Fremstilling af 7(D-o-amino-p-hydroxyphenylacetamido)-3- -methyl-3-cephem-4-carboxylsyre (Cefadroxil) DMF kompleks fra trimethy1silyloxycarbonyl-7-aminodecephalo sporansyre-TMS-ester
Trimethylsilyloxycarbonyl-7-aminodecephalosporansyre-30 -TMS-blanding (46,68 mmol) indeholdende triethylamin HC1 omrørtes og afkøledes til 5°C. Opslæmningen behandledes med propylenoxid (3,7 ml, 52,7 mmol). D-(-)-2-(4'-hydroxy-phenyl)glycylchlorid HC1 hemidioxan opløsningsmiddel (13,7 g, 48,7 mmol) i fem portioner tilsattes ved 5°C i 35 løbet af 3 timer under god omrøring. Blandingen omrørtes yderligere ved 5°C i 2 timer. Intet fast syrechlorid blev tilbage i reaktionsblandingen. Den endelige acylerings- 30

blanding behandledes med methanol (5 ml) efterfulgt af isvand (60 ml). pH-værdien indstilledes til 2,3 med triethylamin, medens temperaturen holdtes på 5°C. Den vandige fase fraskiltes, poleringsfiltreredes på et filter, 05 der forud var belagt med diatoméjord ("Dicalite") og vaskedes med vand (15 ml). Filtratet og vaskevæsken indstilledes til pH-værdi 4,5 med triethylamin og isopropanol (100 ml), og N,N-dimethylformamid (220 ml) tilsattes. Blandingen podedes med 10 mg cefadroxil-DMF-kompleks og 10 fik lov til at krystallisere ved 25°C i 7 timer under kraftig omrøring. Produktet opsamledes og vaskedes med dimethylformamid (DMF; 20 ml) og acetone (2 x 20 ml), og der opnåedes 11,51 g hvidt krystallinsk cefadroxil-DMF-kompleks i et udbytte på 55,96%. NMR og IR var identiske 15 med en standardprøve. NMR viste 1,9 mol DMF pr. mol cefa-droxil.

Fremstilling af 7-(D-a-amino-a-phenylacetamido)-3-methyl-20 -3-cephem-4-carboxylsyre ·'(cephalexin)
Trimethylsilyloxycarbonyl-7-aminodecephalosporansyre--TMS-ester (46,68 mmol) indeholdende triethylamin HC1 orn-rørtes og afkøledes til 5°C. Opslæmningen behandledes med propylenoxid (3,7 ml, 52,7 mmol). D-(-)-phenylglycylchlo-25 rid HC1 (10,2 g, 47,5 mmol) tilsattes i fem portioner ved 5°C i løbet af 5 timer under god omrøring. Blandingen om-rørtes yderligere i 2 timer ved 5°C. Tyndtlagskromatografi (TLC) viste ufuldstændig acylering. Reaktionsblandingen opvarmedes derpå til 25°C og omrørtes i 1 time. Den ende-30 lige acyleringsblanding med behandledes med vand (50 ml). pH-værdien indstilledes til 1,4 under omrøring ved 25°C i 20 minuttér. Den vandige fase fraskiltes, poleringsfiltreredes med diatoméjord ("Dicalite"), og filterkagen vaskedes med vand (15 ml). DMF (10 ml) sattes til den rige, 35 polerede vandige portion. Den vandige opløsning opvarmedes derefter til 60-63°C og behandledes med triethylamin (11 ml) i løbet af 15 minutter til opretholdelse af pH- 31

Cephaloglycin fremstilledes ved at følge fremgangsmålo derne 1, 2 og 3, idet den deri anvendte 7-aminodecephalo-sporansyre (7-ADCA) erstattedes af en ækvimolær vægtmængde 7-aminocephalosporansyre (7-ACA).
