Den foreliggende opfindelse angår en særlig fremgangsmåde til fremstilling af cis-diklor-trans-dihy-droxy-bis-(isopropylamin)-platin(IV) i hovedsagelig ren form. Forbindelsen er nyttig til kemoterapeutisk behandling 5 af cancer og maligne neoplasmer.
I britisk patentskrift nr. 1578323 er der beskrevet koordinationsforbindelser af platin med strukturen

10 Platinet er fortrinsvis til stede som Pt^+, således at der fremkommer et neutralt kompleks med to hydroxyl-og to halogenidligander.
Skønt andre grupper R end hydrogen normalt ikke foretrækkes, kan de bruges og kan udgøres af lavere alkyl-15 grupper såsom metyl eller ætyl, eller en opløseliggørende gruppe såsom en sulfonsyregruppe. 25 Egnede aminforbindelser med grenet kæde, der kan bruges som grupperne A og B, angives at være isopropylamin, isobutylamin, isoamylamin og 2-aminohexan.
En forbindelse der falder inden for kravenes rammer i britisk patentskrift nr. 1578323 er en sådan hvor 3Q A og B i den almene formel begge er isopropylamin og X
og Y begge er klorid, idet komplekset således er cis-diklor-trans-dihydroxy-bis-(isopropylamin)-platin(IV). Den fremstillingsmetode der er angivet i det nævnte patentskrift består i at man opvarmer en opslæmning af det tilsvarende 35 cis-diamindiklorplatin(II)-kompleks med en hydrogenperoxyd-opløsning, kogning i 1/2 time indtil der fremkommer gul farve, afkøling, filtrering, vask af remanensen med vand og æter og tørring i luft. Det resulterende råprodukt om- 3 150474 krystalliseres fra en vand/hydrogenperoxyd-opløsningsmid-delblanding og tørres i vakuum. Af tallene for elementær analyse, det infrarøde absorptionsspektrum og røntgenkrystallografi af en enkelt krystal blev der tilskrevet produktet 5 formlen for det tilsvarende l:l-hydrat.
Senere arbejde har vist at komplekset, fremstillet og omkrystalliseret som beskrevet ovenfor, faktisk eksisterer som et addukt med hydrogenperoxyd i mængdeforholdet 2 molekyler kompleks til 1 molekyle H202, men tilstedevæ-10 reisen af hydrogenperoxyd anses som ufordelagtig ud fra et farmakologisk synspunkt.
Det vides nemlig at indførelse af hydrogenperoxyd i legemet bør undgås på grund af muligheden for beskadigelse af DNA. Den cytotoxiske virkning af den kendte antitu-15 morforbindelse cisplatin (diammindiklorplatin) skyldes direkte samvirke med cellulært DNA, men forbindelsen spalter ikke specifikt DNA. Antitumorvirkningen af platin(IV)-forbindelsen skyldes muligvis reduktion in vivo til biologisk aktive platin(II)-species, men kan også tænkes at bero 20 på en evne til i modsætning til cisplatin at kunne spalte DNA. Den hypotese har været fremsat at forbindelsen CHIP [cis-cis-trans-PtIVCl2-[(CH^)2CHNH2]2(OH)2] er i stand til at frembringe hydroxyl- eller superoxydradikaler, der så kunne spalte DNA; det viste sig imidlertid at forbindel-25 sen CHIP selv var ude af stand til at frembringe et radikal som potentielt kunne føre til beskadigelse af DNA; en iagttagen spaltning skyldtes faktisk det peroxyd som var bundet . til forbindelsen som et addukt fra fremstillingen deraf.
Der foreligger følgelig tvivl om hvorvidt CHIP i 30 og for sig er i stand til at spalte DNA, og en sådan spaltningsreaktion må undgås hvis det overhovedet er muligt (idet den indtil andet er bevist i teorien lige så vel kan give carcinogen virkning som antitumorvirkning).
Den konklusion kan ikke afvises, at tidligere under-35 søgelser der viste DNA-brud fremkaldt af CHIP har anvendt prøver af forbindelsen indeholdende hydrogenperoxyd og at den rene form af CHIP ikke kan spalte DNA.

Enhver reaktion der er så fundamental for cellekemien som spaltning af DNA må antages at være skadelig indtil det modsatte er bevist, og når en påvist gunstig effekt af et antitumormiddel beror på en reaktion med DNA, der 5 ikke resulterer i beskadigelse af DNA, må enhver forbindelse som medfører DNA-spaltning betragtes med stor skepsis og skal så vidt muligt undgås. Derfor foretrækkes peroxyd-frit CHIP, en praktisk betegnelse for cis-diklor-trans-dihydroxy-bis-(isopropylamin)-platin(IV).
10 Forsøg på at fjerne hydrogenperoxydet ad fysisk vej er imidlertid stedse mislykkedes på grund af nedbrydning af komplekset, hvorved der sker tab af hydroxy-liganderne.
Det har derfor hidtil været anset for umuligt at fremstille og isolere komplekset i i hovedsagen ren form, dvs. frit 15 for konstaterbare mængder af hydrogenperoxyd.
Det har imidlertid nu vist sig at komplekset alligevel kan fremstilles i i det væsentlige ren form.

Cl^\ ^ NH2CH(CH3)2 OH 25 og denne koordinationsforbindelse er værdifuld til kemoterapeutisk behandling af cancer og maligne neoplasmer.
Fremgangsmåden ifølge opfindelsen til fremstilling af cis-diklor-trans-dihydroxy-bis-(isopropylamin)-platin(IV) 30 i i det væsentlige ren form er ejendommelig ved at man omsætter cis-diklor-di(isopropylamin)-platin(II) med hydrogenperoxyd, isolerer det derved dannede produkt, opløser produktet i Ν,Ν-dimetylacetamid, isolerer det resulterende produkt som udgøres af et 1:1 addukt af nævnte forbindelse 35 med Ν,Ν-dimetylacetamid og derefter fjerner N,N-dimetyl-acetamidet under vakuum ved en temperatur på under 80°C.
Ifølge opfindelsen er temperaturen fortrinsvis under 50°C.

Det har vist sig at anvendelse af dimetylacetamid ved fremgangsmåden ifølge opfindelsen til tilvejebringelse af det beskrevne mellemprodukt, altså 1:1-adduktet, er velegnet med hensyn til at fjerne hydrogenperoxyd,hvor simple 5 omkrystallisationer under anvendelse af almindelige opløsningsmidler er mislykkedes. Som nævnt foran lykkedes det ikke med vand; det samme gælder metanol trods det meget store numeriske overskud af opløsningsmiddelmolekyler i forhold til hydro-genperoxydmolekyler, skønt man ville have ventet at i det 10 mindste det ene af disse opløsningsmidler ville være velegnet på grund af deres polaritet og deres blandbarhed med hydrogenperoxyd.
Det som mellemprodukt fremkomne 1:1 addukt af cis-diklor-trans-dihydroxy-bis-(isopropylamin)-platin(IV) med 15 dimetylacetamid er i sig selv et nyt produkt som er isoler-bart, stabilt og karakteriserbart.
Fremgangsmåden ifølge opfindelsen skal i det følgende belyses nærmere ved et eksempel som angår (a) fremstilling af rå cis-diklor-trans-dihydroxy-bis-(isopropylamin)-20 Pt(IV), og (b) fremstilling af det som mellemprodukt dannede addukt med dimetylacetamid og påfølgende isolation af det i hovedsagen rene slutprodukt.
Eksempel 25 a) Fremstilling af råproduktet 26,8 g omkrystalliseret cis-Pt-(i-C3H7NH2)2Cl2 °P“ slæmmedes i 50 ml varmt vand og der tilsattes under omrøring 100 rumfang (100 ml) vandigt hydrogenperoxyd. Opslæmningen 30 kogtes i 1/2 time indtil den var gul og afkøledes derefter og filtreredes. Remanensen vaskedes med vand, derpå med æter og tørredes derpå i luft. Udbyttet var 10,5 g (35%).
Beregnet for hemiperhydratet med bruttoformlen PtCgN2Cl203H27: C 16,55 H 4,87 N 6,43 O 11,03 Cl 16,28 35 Fundet: C 16,59 H 4,90 N 6,48 0 10,80 Cl 16,20%
Den infrarøde hydroxyl-strækningsabsorption (vO-H) optræder ved 3515 cm Der blev ikke analyseret for Pt på dette trin og forbindelsen havde intet smeltepunkt i 150474 6 traditionel forstand; der sker sønderdeling som vist i kurverne (fig. 8) over termisk gravimetrisk analyse (TGA).
Som det fremgår begynder sønderdelingen ved ca. 150°C og ved 500°C er der kun platinmetal tilbage, repræsenterende 5 ca. 44% af den oprindelige vægt.
b) Fremstilling af adduktet og isolation af det i hovedsa-gen rene produkt._ 11 g af det på den ovenfor beskrevne måde fremstille-10 de råprodukt opløstes i ca. 1 liter N,N-dimetylacetamid og opløsningen filtreredes gennem en glassinter (porøsitet 4) til fjernelse af alt det tilstedeværende uopløselige materiale. Opløsningen afkøledes til 5°C i 2 timer, podedes til forebyggelse af overmætning og opbevaredes ved -10°C 15 i nogle dage. De resulterende krystaller frafiltreredes, vaskedes med æter og tørredes.
Fjernelse af Ν,Ν-dimetylacetamid til frembringelse af i hovedsagen rent produkt udførtes ved opvarmning af krystallerne til en temperatur på 50°C eller derunder ved 20 et tryk på 0,1 torr i nogle dage, eller indtil fraværet af Ν,Ν-dimetylacetamid bekræftedes ved termisk gravimetrisk analyse og/eller gas-væskekromatografi.
Opløsningen af råproduktet fra eksemplets afsnit a) i Ν,Ν-dimetylacetamid kan udføres ved stuetemperatur, 25 men det foretrækkes at bruge svagt forhøjet temperatur, dvs. ikke over ca. 40°C.
Yderligere mulige rensningstrin omfatter omkrystallisation af råproduktet (fra trin a) ovenfor) fra en tynd hydrogenperoxydopløsning, efterfulgt af tørring ved ca.
30 50°C i vakuum samt yderligere filtrering af N,N-dimetylacet- amidopløsningen (fra trin b)) gennem et silikagelleje (70-230 mesh) til fjernelse af eventuelle polyhydroxyplatinpro-dukter.
Analysedata til karakterisering af det tidligere 35 kendte hydrogenperoxydaddukt, forbindelsen ifølge den foreliggende opfindelse og Ν,Ν-dimetylacetamidadduktet er som beskrevet nedenfor og belyst nærmere ved tegningen, på hvilken 7 150474 fig. 1 viser IR-spektrum for det kendte hydrogenper-oxydaddukt, fig. 2 IR-spektret for det ny dimetylacetamid- addukt, 5 fig. 3 IR-spektrum for den ved fremgangsmåden ifølge opfindelsen fremstillede forbindelse, fig. 4 røntgenpulver-diffraktionsmønster for det kendte hydrogenperoxydaddukt, fig. 5 røntgenpulver-diffraktionsmønster for det 10 nye N,N-dimetylacetamidaddukt, fig. 6 røntgenpulver-diffraktionsmønster for den ved fremgangsmåden ifølge opfindelsen fremstillede forbindelse, fig. 7 kromatogrammer af forskellige forbindelser, 15 jfr. nedenfor, fig. 8 resultatet af termiske gravimetriske analyser af forskellige forbindelser jfr. nedenfor, og fig. 9 en stereoskopisk gengivelse af N,N-dimetyl-acetamidadduktet.
20 Cis,trans,cis-[PtCl2(OH)2(iPrNH2)2] - den ved frem gangsmåden ifølge opfindelsen fremstillede forbindelse -var i relation til de efterfølgende analyser fremstillet som enkeltportioner A, B og C, der analyseredes hver for sig, og som desuden sammenblandedes omhyggeligt til en sam-25 let portion X der ligeledes analyseredes. Analysemetoden var som følger:
En afvejet prøve af vedkommende produkt (ca. 20 mg) indsprøjtes som en opløsning i analyseapparatet, der viser resultaterne som kurver. Arealet under kurven er et mål 30 for den tilstedeværende mængde. Der er således fejlmuligheder ved afvejningen, på opløsningsstadiet og iboende fejl i apparatet. I praksis anses en fejl på -2% for ventet og acceptabel. Analysedata var som følger: 35 8 150474

Fugtighedsindhold: <0,25 vægt% (alle portioner) ved termisk gravimetrisk analyse (TGA). Den foretages ved at en prøve af forbindelsen opvarmes progressivt og dens vægtændring 25 e opnoteres efterhånden som komponenterne evaporerer.
Nedenstående vises resultaterne af TGA for fem forskellige prøver af cis-diklor-trans-dihydroxy-bis-(isopro-pylamin)-platin(IV) fremstillet som netop beskrevet, udtrykt ^ som % vægttab og % slutvægt. Til sammenligning vises de teoretiske værdier for CHIP.I^C^.

Fig. 4, 5 og 6 peger på samme konklusion, idet bevisligheden i dette tilfælde er røntgenpulver-diffraktionsmønstrene . På alle figurerne er 20vinklen anført og også adskillelsen af planerne i Å.
Fig. 7 viser de mængder Ν,Ν-dimetylacetamid (DMA), 25 der er til stede i portionerne A, B, C og X, målt ved gas-væskekromatografi.
For et givet område ville et i sammenligning med standarden lignende respons hos A, B, C eller X repræsentere en DMA-koncentration på ca. 1 vægt%.
30 Fig. 8 viser resultaterne af termisk gravimetrisk analyse af forskellige forbindelser. På denne figur repræsenterer kurve 1 den tidligere kendte hydrogenperoxydaddukt-forbindelse, 35 kurve 2 den nye N,N-dimetylacetamid-adduktforbindel- se og kurve 3 den ved fremgangsmåden ifølge opfindelsen fremstillede forbindelse.

Sporene er arbitrært forskudt på "vægt(%)"-aksen af hensyn til en simpel gengivelse.
Det ses at kurve 1 viser et større vægttab ved ca.
160°C på grund af tabet af trans-OH-grupper såvel som HgOg, 5 mens kurverne 2 og 3 viser i det væsentlige samme vægttab ved den temperatur på grund af at der kun sker tab af trans-OH-grupper. I kurve 2 repræsenterer den skrånende skulder op til en temperatur på 150°C tab af N,N-dimetylacetamid, og derefter ses det at de forbindelser der repræsenteres 10 af kurverne 2 og 3 er i alt væsentlige identiske.
Den i fig. 9 viste stereoskopiske .gengivelse er fremkommet ved røntgenkrystallografisk undersøgelse af en enkelt krystal af Ν,Ν-dimetylacetamidadduktet. Forsøg på at isolere en enkelt krystal af den ved fremgangsmåden 15 ifølge opfindelsen fremstillede, i det væsentlige rene forbindelse ud fra en enkelt krystal af Ν,Ν-dimetylacetamidadduktet resulterede i en agglomeration af mikrokrystalli-ter, og røntgenpulvermønsteret for dette er vist i fig. 6.
Den i det væsentlige rene forbindelse cis-diklor-trans-20 dihydroxy-bis-(isopropylamin)-Pt(IV) kan indgives på samme måde som beskrevet i britisk patentskrift nr.
1578323 for det kendte hydrogenperoxydaddukts vedkommende.
