Description
- Die vorliegende Erfindung betrifft Ketonderivate und Arzneimittel, in denen Ketonderivate oder pharmakologisch annehmbare Salze davon einen Wirkstoff darstellen, insbesondere blutbildende Mittel.

US-A-5.059.620 offenbart die antiarrhythmische Aktivität von ausgewählten Arylsubstituierten Aminomethylbenzolderivaten. -
EP-A-0071535 offenbart 2/3-substituierte 4-(Thienylacetamido)phenoxyhydroxypropylamine, die als Beta-blockierende blutdrucksenkende Mittel wirksam sind. -
FR-A-2.185.391 US-A-3.963.756 betreffen Derivate von 6-Methyl-2H-pyran-2,4(3H)-dion als Mittel zur Behandlung von Bluthochdruck. - Als zu den Ketonderivaten in Zusammenhang mit der vorliegenden Erfindung analoge Verbindungen werden Beispiele von 3-(1-Phenyl-2-acetyl)-4-hydroxypyron-Analoga und 3-(1-Phenyl-2-acetyl)-4-hydroxycumarin-Analoga angeführt, aber diese unterschieden sich von den Ketonderivaten in Zusammenhang mit der vorliegenden Erfindung darin, dass sie keine Amidbindung aufweisen.
- In Bezug auf die Anwendung von Lactonen sind die beispielsweise in
JP-A-05-043568 EP-A-0508690 beschriebenen Verbindungen als entzündungshemmende Mittel mit Phospholipase-A2-hemmender Aktivität bekannt; die in Archive der Pharmazie 316 (2), 115–120, Weinhelm, Deutschland (1983), beschriebenen Verbindungen sind als gerinnungshemmende Mittel bekannt; und die in J. Antibiot. 47(2), 143–147 (1994), beschriebenen Verbindungen sind als Anti-AIDS-Arzneimittel mit HIV-Protease-hemmender Aktivität bekannt. In Bezug auf die Anwendung von Lactamen sind die in Chem. Pharm. Bull. 32(10), 4197–4204 (1984), beschriebenen Verbindungen als Arzneimittel mit antimikrobieller Aktivität bekannt, und die in Antibiot. 33(2), 173–181 (1980), beschriebenen Verbindungen sind als anaerobe Antibiotika bekannt. -
EP-A-0841063 , das als Stand der Technik gemäß Artikel 54(3) EPÜ verfügbar ist, schlägt ein blutbildendes Mittel vor, das ein Ketonderivat als Wirkstoff aufweist. - Die vorliegende Erfindung hat das Ziel, Ketonderivate mit herausragender blutbildender Wirkung bereitzustellen.
- Das zuvor genannte Ziel wird durch die vorliegende Erfindung wie untenstehend beschrieben erreicht.
- Spezifisch stellt die vorliegende Erfindung ein Ketonderivat der folgenden allgemeinen Formel (I) bereit: (worin A für die allgemeine Formel (II) oder (III) steht, m die ganze Zahl 1 oder 2 ist, R1 bis R11 unabhängig voneinander ein Wasserstoffatom oder eine Substituentengruppe sind und n = 0 oder 1 ist), insbesondere ein Ketonderivat oder ein pharmakologisch annehmbares Salz des Ketonderivats, worin R1 bis R7 unabhängig voneinander ein Wasserstoffatom oder ein aus der Gruppe, die ein Fluoratom, ein Chloratom, ein Bromatom, C1- bis C19-Alkylgruppen, Alkenylgruppen mit bis zu 19 Kohlenstoffatomen, C6- bis C12-Arylgruppen, C6-12-Aryl-C1-19-Alkylgruppen, C1-19-Alkyl-C6-12-Arylgruppen, C6-12-Aryl-C2-19-Alkenylgruppen, C2-19-Alkenyl-C6-12-Arylgruppen, eine Hydroxylgruppe, C1-10-Alkoxygrupen, eine Nitrogrup pe, Carboxylgruppe, C2-10-Alkoxycarbonylgruppen und eine Cyanogruppe umfasst, ausgewählter Substituent sind oder R1 und R2 zusammen -CH=CH-CH=CH- sind, R5 und R6 zusammen -CH=CH-CH=CH- oder -(CH2)q- sind (wobei q eine ganze Zahl im Bereich 2 bis 5 ist) oder R6 und R7 zusammen -(CH2)r- sind (wobei r eine ganze Zahl im Bereich 2 bis 5 ist),
R8 und R9 jeweils unabhängig voneinander ein Wasserstoffatom oder ein aus der Gruppe, die C1-6-Alkylgruppen, Alkenylgruppen mit bis zu 6 Kohlenstoffatomen, C6-12-Arylgruppen, C6-12-Aryl-C1-19-Alkylgruppen, C1-19-Alkyl-C6-12-Arylgruppen, C6-12-Aryl-C2-19-Alkenylgruppen, C2-9-Alkenyl-C6-12-Arylgruppen, C2-10-Alkoxycarbonylgruppen umfasst, ausgewählter Substituent sind oder R8 und R9 zusammen -(CH2)4- oder -(CH2)5- sind, R10 ein Wasserstoffatom oder ein aus einem Fluoratom, Chloratom oder Bromatom ausgewählter Substituent ist, R11 ein Wasserstoffatom oder ein aus einer C1-6-Alkylgruppe oder C6-12-Arylgruppe ausgewählter Substituent ist oder R10 und R11 zusammen -CH=CH-CH=CH- oder -(CH2)t- sind (wobei t eine ganze Zahl im Bereich 2 bis 5 ist); und stellt solche Ketonderivate zur Verwendung als Arzneimittel, insbesondere blutbildende Mittel, bereit, die die Ketonderivate der allgemeinen Formel (I) und pharmakologisch annehmbare Salze davon enthalten. -
1 ist ein Graph, der die blutbildende Wirkung in Beispiel 11 veranschaulicht. - Nun werden bevorzugte Ausführungsformen der vorliegenden Erfindung beschrieben.
- Wenn Z in der allgemeinen Formel (I) der vorliegenden Erfindung ein Wasserstoffatom ist, sind die Keto-Enol-Tautomere der folgenden Formeln umfasst.
- Hier können die C1- bis C19-Alkylgruppen der allgemeinen Formel (I) unverzweigt, verzweigt oder zyklisch sein, und Beispiele dafür umfassen Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, sec-Butyl, tert-Butyl, n-Pentyl, Isopentyl, Neopentyl, tert-Pentyl, n-Hexyl, n-Heptyl, n-Octyl, n-Nonyl, n-Decyl, n-Dodecyl, n-Tetradecyl, n-Pentadecyl, n-Hexadecyl, n-Heptadecyl, 15-Methylhexadecyl, 7-Methylhexadecyl, n-Octadecyl, n-Nonadecyl, Eicosyl, Tetracosyl, 24-Methyltetracoxyl, Cyclopropyl, Cyclobutyl und Cyclohexyl. Die C1- bis C19-Alkenylgruppen können unverzweigt oder verzweigt sein und umfassen Isomere in Bezug auf die Doppelbindung (E- oder Z-Isomere). Beispiele umfassen Ethenyl, 2-Propenyl, 2-Butenyl, 2-Pentenyl, 2-Hexenyl, 2-Heptenyl, 2-Octenyl, 2-Nonenyl, 2-Decenyl, 3-Butenyl, 3-Pentenyl, 3-Hexenyl, 3-Heptenyl, 3-Octenyl, 3-Nonenyl, 3-Decenyl, 4-Pentenyl, 4-Hexenyl, 4-Heptenyl, 4-Octenyl, 4-Nonenyl, 4-Decenyl, 1,3-Butadienyl, 1,3-Pentadienyl, 1,3-Hexadienyl, 1,3-Heptadienyl, 1,3-Octadienyl, 1,3-Nonadienyl, 1,3-Decadienyl, 1,4-Pentadienyl, 1,4-Hexadienyl, 1,4-Heptadienyl, 1,4-Octadienyl, 1,4-Nonadienyl, 1,4-Decadienyl, 1,3,5-Hexatrienyl, 1,3,5-Heptatrienyl, 1,3,5-Octatrienyl, 1,3,5-Nonatrienyl und 1,3,5-Decatrienyl.
- Die Arylgruppen können mit einem oder mehreren Halogenatomen, wie z.B. einem Chloratom, Bromatom oder Fluoratom, einer Hydroxygruppe, einer Nitrogruppe, Methoxy-, Ethoxy- oder einer anderen Alkoxygruppe, einer Carboxylgruppe, einer Carbomethoxy-, Carboethoxy- oder einer anderen Carboalkoxygruppe, einer Cya nogruppe, einer Trifluormethylgruppe, einer Methylthio- oder einer anderen Alkylthiogruppe oder einer Phenylthiogruppe substituiert sein.

- Beispiele für C1- bis C10-Alkoxygruppen umfassen Methoxy, Ethoxy, Propoxy, Butoxy, Pentoxy, Hexoxy, Heptoxy und Octoxy. Beispiele für C2- bis C10-Alkoxycarbonylgruppen umfassen Methoxycarbonyl, Ethoxycarbonyl, Propoxycarbonyl, Butoxycarbonyl, Pentoxycarbonyl, Hexoxycarbonyl und Heptoxycarbonyl.
- X steht in der allgemeinen Formel (I) für O, S, CH2 oder NH, wobei O oder CH2 jedoch bevorzugt sind.
- Noch bevorzugter sind R1 bis R11 in der allgemeinen Formel (I) unabhängig voneinander ein Wasserstoffatom, ein Fluoratom, ein Chloratom, ein Bromatom oder eine C1- bis C5-Alkylgruppe, wobei die C1- bis C5-Alkylgruppe unverzweigt oder verzweigt sein kann und Beispiele dafür Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, Isobutyl, sec-Butyl, tert-Butyl, n-Pentyl, Isopentyl, Neopentyl und tert-Pentyl umfassen.
- Beispiele für pharmakologisch annehmbare Salze umfassen Basen- und Säureadditionssalze. Basenadditionssalze sind Salze, die die biologische Wirksamkeit und die Eigenschaften der freien Säure beibehalten, ohne biologisch oder auf andere Weise unerwünscht zu sein, und umfassen Salze, die von anorganischen Basen erhalten werden, wie z.B. Natrium-, Kalium-, Lithium-, Ammonium-, Calcium- und Magnesiumsalze. Sie umfassen selbstverständlich auch Salze, die von organischen Basen erhalten werden. Beispielsweise umfassen sie Salze, die von substituierten Aminen, wie z.B. primären, sekundären, tertiären, natürlichen substituierten und zyklischen Aminen, sowie basischen Ionenaustauschharzen erhalten werden, wie z.B. Isopropylamin, Trimethylamin, Diethylamin, Tripropylamin, Ethanolamin, 2-Dimethylaminoethanol, 2-Diethylaminoethanol, Trimethamin, Dicyclohexylamin, Lysin, Arginin, Histidin, Coffein, Procain, Hydrabamin, Cholin, Betain, Ethylendiamin, Glucosamin, Methylglucamin, Theobromin, Purin, Piperazin, Piperidin, N-Ethylpiperidin, Ornithin und Polyaminharz. Säureadditionssalze sind Salze, die die biologische Wirksamkeit und die Eigenschaften der freien Base beibehalten, ohne biologisch oder auf andere Weise unerwünscht zu sein, und umfassen anorganische Säuresalze, wie z.B. Hydrochlorid, Sulfat, Nitrat, Hydrobromid, Hydrofluorborat, Phosphat und Perchlorat, sowie organische Säuresalze, wie z.B. Oxalat, Tartrat, Lactat und Acetat. Die pharmakologisch annehmbaren Salze der vorliegenden Erfindung sind jedoch nicht auf diese beschränkt.
- Unter den Verbindungen der vorliegenden Erfindung befinden sich, wenn ein asymmetrisches Kohlenstoffatom im Molekül vorhanden ist, optische Isomere und, wenn zumindest zwei asymmetrische Kohlenstoffatome vorhanden sind, Diastereomere. Die vorliegende Erfindung umfasst diese optischen Isomere und Diastereomere. Weiters umfasst die Erfindung Stereoisomere.
- Die Verbindungen der vorliegenden Erfindung können durch eine Kombination von bekannten Verfahren hergestellt werden. Die Synthese kann beispielsweise durch die Reaktion einer Verbindung der Formel (V) (in Formel (V) ist Q beispielsweise ein Chloratom oder eine Hydroxylgruppe) mit einer Verbindung der Formel (IV) durchgeführt werden, wobei das Syntheseverfahren nicht auf dieses bestimmte Verfahren beschränkt ist.
- In Bezug auf die Synthese von Verbindung (IV) kann die Herstellung der Lactone (wenn X ein Sauerstoffatom ist) durch die beispielsweise in J. Chem. Soc. Perkin Trans. I, 121–129 (1987); J. Org. Chem. 59, 488–490 (1994); Bull. Chem. Soc. Japan 52, 3601–3605 (1979); J. Chem. Soc. Perkin Trans. I, 1225–1231 (1987); Tetrahedron Lett. 5143–5146 (1983), und Chem. Pharm. Bull. 38, 94–98 (1990), offenbarten Verfahren erfolgen, und die Herstellung der Lactame (die Verbindungen, bei denen X = NH ist) kann durch die in Chem. Pharm. Bull. 32(10), 4197–4204 (1984); Pharmazie 43(7), 473–474 (1988); Monatsh. Chem. 123(1–2), 93–98 (1992); J. Inorg. Biochem. 24(3), 167–181 (1985); J. Am. Chem. Soc. 107(18), 5219–5224 (1985), und J. Org. Chem. 50(8), 1344–1346 (1985), offenbarten Verfahren erfolgen.
- Wenn ein therapeutisches Mittel, das eine wirksame Menge eines Ketons der vorliegenden Erfindung enthält, klinisch verabreicht wird, wird es oral oder parenteral verabreicht. Die Verabreichungsformen umfassen Tabletten, Dragees, Pillen, Kapseln, Pulver, Pastillen, Lösungen, Suppositorien und Injektionen, wobei diese durch Com poundieren mit medizinisch zulässigen Füllstoffen hergestellt werden können. Die folgenden können als Beispiele für Füllstoffe verwendet werden. Es gibt medizinisch zulässige Füllstoffe, wie z.B. Lactose, Saccharose, Glucose, Sorbit, Mannit, Kartoffelstärke, Amylopectin, verschiedene andere Stärkearten, Cellulosederivate (z.B. Carboxymethylcellulose und Hydroxyethylcellulose), Gelatine, Magnesiumstearat, Polyvinylalkohol, Polethylenglykolwachs, Gummiarabikum, Talk, Titandioxid, Olivenöl, Erdnussöl, Sesamöl und andere Arten von pflanzlichen Ölen, flüssiges Paraffin, neutrale Fettbase, Ethanol, Propylenglykol, physiologische Salzlösung, sterilisiertes Wasser, Glycerin, Färbemittel, Geschmacksstoffe, Verdickungsmittel, Stabilisatoren, isotonische Mittel und Puffer.
- Die Bezeichnung "blutbildendes Mittel" bezieht sich hierin auf ein Arzneimittel, das, wenn es Menschen oder Tieren verabreicht wird, die Produktion von Blutplättchen, roten Blutkörperchen und weißen Blutkörperchen im Körper fördert und eingesetzt wird, um eine durch Krebschemotherapie, Strahlentherapie, Knochenmarktransplantation und Arzneimitteltherapie oder durch eine immunologische Anomalie oder Anämie, wie z.B. renale Anämie, hämorrhagische Anämie, hämolytische Anämie oder Mangelanämie, hervorgerufene Zytopenie zu verhindern oder zu behandeln. Weiters können die blutbildenden Mittel der vorliegenden Erfindung auf dem Gebiet der Behandlung von durch eine Infektionskrankheit, eine Viruserkrankung oder Ernährungsstörungen hervorgerufener aplastischer Anämie, Thrombozytopenie und Hypoleukozytose oder idiopathischer thrombozytopenischer Purpura eingesetzt werden. Weiters kann es auch für gelagertes Eigenblut eingesetzt werden.
- Es ist auch möglich, die blutbildenden Mittel der vorliegenden Erfindung beispielsweise in Kombination mit EPO, das ein Mittel zur Bildung von roten Blutkörperchen ist, oder mit G-CSF, das ein Mittel zur Bildung von weißen Blutkörperchen ist, zur Prävention oder Behandlung einer durch Krebschemotherapie, Strahlentherapie, Knochenmarktransplantation und Arzneimitteltherapie oder durch immunologische Anomalien oder eine Anämie, wie z.B. renale Anämie, hämorrhagische Anämie, hämolytische Anämie, Mangelanämie, hervorgerufenen Hypozytose zu verwenden.
- Die Menge des therapeutischen Mittels der vorliegenden Erfindung, die zum Einsatz kommt, variiert in Abhängigkeit von den Symptomen, dem Körpergewicht, dem Alter und der Verabreichungsform, wobei einem Erwachsenen gewöhnlicherweise 0,01 mg bis 2000 mg pro Tag verabreicht werden können.

- Untenstehend werden Ausführungsformen der vorliegenden Erfindung detaillierter durch die Bereitstellung von Referenzbeispielen und Beispielen und unter Bezugnahme auf
1 erläutert, wobei1 einen Graph darstellt, der die blutbildende Wirkung in Beispiel 11 zeigt. - Beispiel 1
- Synthese von Verbindung 1
- 4-Nitrophenylessigsäure (7,2 g, 40 mmol) und dann Dicyclohexylcarbodiimid (8,6 g, 42 mmol) wurden bei Raumtemperatur zu einer Dichlorethan-Suspension (100 ml) von 4-Hydroxy-6-methyl-2-pyron (5,0 g, 40 mmol) zugesetzt, dann wurde 4-Dimethylaminopyridin (0,97 g, 8,0 mmol) zugesetzt, und 1 h lang wurde Rühren bei Raumtemperatur durchgeführt, danach weitere 16 h bei 50 °C. Nach der Rückkehr zu Raumtemperatur wurden der erzeugte nicht-lösliche Dicyclohexylharnstoff abfiltriert, und die Reaktionslösung wurde mit verdünnter Salzsäure gewaschen, wonach die wässrige Phase mit Dichlormethan extrahiert wurde. Nach dem Einengen der Lösung wurde Natriumhydrogencarbonat zu dem erhaltenen Rückstand zugesetzt, um das Natriumsalz zu bilden, und Waschen wurde mit Ethylacetat durchgeführt.

Tetrahydrofuran (50 ml), Methanol (15 ml) und konzentrierte Salzsäure (5 ml) wurden zu der erhaltenen Verbindung (1,00 g, 3,46 mmol) zugesetzt, wonach auch Pd-C (5 %, feucht, 200 mg) zugesetzt und die Atmosphäre durch Wasserstoff ersetzt wurde. In 3 h war das Rohmaterial aufgebraucht, und der Wasserstoff wurde durch Argon ersetzt. Der Niederschlag wurde abfiltriert, mit heißem Methanol gewaschen und das Filtrat eingeengt. Als der erhaltene Feststoff aus Methanol umkristallisiert wurde, wurde 3-(4-Aminophenylacetyl)-4-hydroxy-6-methyl-2-pyronhydrochlorid (760 mg, 74 %) erhalten. 2-Thiophenacetylchlorid (0,17 ml, 1,5 mmol) wurde zu einer THF-Lösung (10 ml) der erhaltenen Verbindung (400 mg, 1,35 mmol) zugesetzt, und dann wurde Triethylamin (0,52 ml, 4,1 mmol) zugesetzt, und Rühren wurde 1 h lang bei Raumtemperatur durchgeführt. Danach wurde verdünnte Salzsäure zugesetzt und eine Extraktion mit Dichlormethan durchgeführt. Die organische Phase wurde mit wasserfreiem Natriumsulfat getrocknet, das Lösungsmittel wurde abdestilliert, wonach Trennung und Reinigung mittels Säulenchromatographie durchgeführt wurden. Als eine Umkristallisierung aus Ethanol durchgeführt wurde, wurde Verbindung 1 (280 mg, 54 %) in Form von hellgelben Kristallen erhalten.

- Synthese von Verbindung 2
- 4-Nitrobenzaldehyd (4,5 g, 29,7 mmol) und eine Reihe von Piperidin-Tropfen wurden zu einer Benzol-Lösung (40 ml) von Dehydroessigsäure (5,00 g, 29,7 mmol) bei Raumtemperatur zugesetzt, wonach 6 h lang rückflusserhitzt wurde, während das Wasser durch Azeotropie entfernt wurde. Der Großteil des Benzols wurde direkt abdestilliert, und dann wurde unter reduziertem Druck eine Destillation zur Trockene durchgeführt. Danach wurde, als eine Umkristallisierung aus Ethanol durchgeführt wurde, 4-Hydroxy-6-methyl-3-(4-nitrophenyl)acryloyl-2-pyron (7,3 g, 82 %) in Form von orangen Kristallen erhalten.
- Tetrahydrofuran (250 ml), Methanol (75 ml) und konzentrierte Salzsäure (25 ml) wurden zu der erhaltenen Verbindung (5,00 g, 16,6 mmol) zugesetzt, danach wurde Pd-C (5 %, umfassend 100 % Wasser, 700 mg) zugesetzt, und die Atmosphäre wurde durch Wasserstoff ersetzt. Nach 17-stündigem Rühren wurde der Wasserstoff durch Argon ersetzt. Der Niederschlag wurde abfiltriert, mit heißem Methanol gewaschen, und das Filtrat wurde eingeengt. Als der erhaltene Feststoff aus Ethanol umkristallisiert wurde, wurde 3-(4-Aminophenylpropionyl)-4-hydroxy-6-methyl-2-pyronhydrochlorid (3,66 g, 71 %) erhalten. 2-Thiophenacetylchlorid (0,35 ml, 2,8 mmol) wurde zu einer THF-Lösung (10 ml) der erhaltenen Verbindung (800 mg, 2,58 mmol) zugesetzt, dann wurde Triethylamin (1,07 ml, 7,7 mmol) zugesetzt, wonach 2-stündiges Rühren bei Raumtemperatur erfolgte, danach weitere 4 h bei 60 °C. Dann wurde verdünnte Salzsäure zugesetzt und eine Extraktion mit Dichlormethan durchgeführt. Die organische Phase wurde mit wasserfreiem Natriumsulfat getrocknet, das Lösungsmittel wurde abdestilliert, wonach Trennung und Reinigung mittels Säulenchromatographie durchgeführt wurden. - Synthese von Verbindung 3
- 3-Nitrophenylessigsäure (7,20 g, 39,6 mmol) und dann Dicyclohexylcarbodiimid (8,6 g, 42 mmol) wurden zu einer 1,2-Dichlorethan-Suspension (100 ml) von 4-Hydroxy-6-methyl-2-pyron (5,00 g, 39,6 mmol) bei Raumtemperatur zugesetzt, wonach 4-Dimethylaminopyridin (0,24 g, 2,0 mmol) zugesetzt wurde und 1-stündiges Rühren bei 60 °C durchgeführt wurde, wonach weiteres 4-Dimethylaminopyridin (0,48 g, 4,0 mmol) zugesetzt wurde und 6-stündiges Rühren bei 60 °C durchgeführt wurde. Während die Lösung noch warm war, wurde der Dicyclohexylharnstoff abfiltriert, und die Reaktionslösung wurde mit verdünnter Salzsäure (50 ml) gewaschen, wonach die wässrige Phase mit Dichlormethan extrahiert wurde. Nach dem Trocknen mit wasserfreiem Natriumsulfat, wurde das Lösungsmittel abdestilliert, und Trennung und Reinigung wurden mittels Säulenchromatographie durchgeführt. Als aus Ethanol umkristallisiert wurde, wurde 4-Hydroxy-6-methyl-3-(3-nitrophenylacetyl)-2-pyron (6,60 g, 58 %) in Form von weißen Kristallen erhalten.

Der Dicyclohexylharnstoff wurde abfiltriert, während die Lösung noch warm war, wonach die Reaktionslösung mit verdünnter Salzsäure (50 ml) gewaschen wurde, wonach die wässrige Phase mit Dichlormethan extrahiert wurde. Nach dem Trocknen mit wasserfreiem Natriumsulfat wurde das Lösungsmittel abdestilliert, und Trennung und Reinigung erfolgten mittels Säulenchromatographie. Bei Durchführen einer Umkristallisierung aus Ethanol wurde 3-Hydroxy-2-(4-nitrophenylacetyl)-2-cyclohexenon (8,2 g, 67 %) in Form von hellgelben Kristallen erhalten. Tetrahydrofuran (100 ml), Methanol (35 ml) und konzentrierte Salzsäure (10 ml) wurden zu der erhaltenen Verbindung (2,00 g, 7,3 mmol) zugesetzt, danach wurde Pd-C (5 %, umfassend 100 % Wasser, 300 mg) zugesetzt, und die Atmosphäre wurde durch Wasserstoff ersetzt. Das Rohmaterial war nach 4 h verbraucht, und dann wurde der Wasserstoff durch Argon ersetzt. Der Niederschlag wurde abfiltriert, mit heißem Methanol gewaschen, und das Filtrat wurde eingeengt. Tetrahydrofuran (50 ml), Methanol (18 ml) und konzentrierte Salzsäure (5 ml) wurden zu der erhaltenen Verbindung (1,00 g, 3,1 mmol) zugesetzt, danach wurde Pd-C (5 %, umfassend 100 % Wasser, 150 mg) zugesetzt, und die Atmosphäre wurde durch Wasserstoff ersetzt. Das Rohmaterial war nach 4 h verbraucht, und dann wurde der Wasserstoff durch Argon ersetzt.

Dann wurde Dicyclohexylcarbodiimid (460 mg, 2,2 mmol) zugesetzt, und 30-minütiges Rühren wurde durchgeführt. Nach der Rückkehr zu Raumtemperatur und 15-stündigem Rühren wurde verdünnte Salzsäure zugesetzt und eine Extraktion mit Dichlormethan durchgeführt. Die organische Phase wurde mit wasserfreiem Natriumsulfat getrocknet und das Lösungsmittel abdestilliert, wonach Trennung und Reinigung mittels Säulenchromatographie durchgeführt wurden. Bei Durchführung einer Umkristallisierung aus Ethanol wurde Verbindung 8 (156 mg, 22 %) in Form von weißen Kristallen erhalten.

- Beispiel 9
- Dicyclohexylcarbodiimid (980 mg, 4,7 mmol) und dann Triethylamin (0,60 ml, 4,2 mmol) wurden zu einer Dichlormethan-Lösung (20 ml) von Tetronsäure (450 mg, 4,50 mmol) und 4-(2-Thiophen-2-ylacetylamino)phenylessigsäure (900 mg, 4,51 mmol), das aus Thiophenacetylchlorid und 4-Aminophenylessigsäure synthetisiert worden war, zugesetzt, und nach 30-minütigem Rühren wurde 4-Dimethylaminopyridin (82 mg, 0,67 mmol) zugesetzt, und das Rühren wurde weitere 24 h lang durchgeführt. Der Dicyclohexylharnstoff wurde abfiltriert, und die Reaktionslösung wurde mit verdünnter Salzsäure gewaschen, wonach die wässrige Phase mit Dichlormethan extrahiert wurde. Nach dem Trocknen mit wasserfreiem Natriumsulfat wurde das Lösungsmittel abdestilliert, und Trennung und Reinigung wurden mittel Säulenchromatographie durchgeführt. Bei Durchführung einer Umkristallisierung aus Ethanol wurde Verbindung 9 (660 mg, 41 %) in Form von weißen Kristallen erhalten.

- Eine Dichlormethan-Lösung (20 ml) von 3-(4-Aminophenyl)-4-hydroxy-6-methyl-2-pyronhydrochlorid (700 mg, 2,4 mmol) und 3-Methylthiophenessigsäure (390 mg, 2,4 mmol), die aus 3-Methylthiophen-2-aldehyd synthetisiert worden war, wurde eisgekühlt, und Triethylamin (0,7 ml, 4,8 mmol) wurde zugesetzt. Dann wurden Dicyclohexylcarbodiimid (515 mg, 2,5 mmol) und eine kleine Menge Dimethylaminopyridin zugesetzt, und 20-stündiges Rühren bei Raumtemperatur wurde durchgeführt. Verdünnte Salzsäure wurde zugesetzt und eine Extraktion mit Dichlormethan durchgeführt. Die organische Phase wurde mit wasserfreiem Natriumsulfat getrocknet und das Lösungsmittel abdestilliert. Der Rückstand wurde mittels Säulenchromatographie getrennt und gereinigt, und bei Durchführung einer Umkristallisierung aus Ethanol wurde Verbindung 10 (352 mg, 37 %) in Form von weißen Kristallen erhalten.

- Blutbildende Wirkung in gewöhnlichen Mäusen
- Verbindung 1 wurde als Testarzneimittel über eine Dauer von vier aufeinander folgenden Tagen hinweg C57BL/6-Mäusen (6 Wochen alt, männlich) (n = 6) intravenös in einer Dosis von 10 mg/kg verabreicht, und 7 Tage nach Beginn der Verabreichung wurde die Anzahl der Blutkörperchen im peripheren Blut gemessen.

1 zeigt den Anstieg des Prozentsatzes bezogen auf eine Kontrollgruppe (als 100 % angenommen). - Aus den Ergebnissen geht deutlich hervor, dass das Testarzneimittel die Anzahl der roten Blutkörperchen signifikant steigert, was den Wert der im vorliegenden Patent beschriebenen Verbindungen für medizinische Anwendungen zeigt.
- Potenzial der gewerblichen Anwendbarkeit
- Die Ketone der vorliegenden Erfindung rufen einen signifikanten Anstieg der roten Blutkörperchen und anderer Zellen hervor und sind als herausragende blutbildende Mittel in der Medizin wirksam, insbesondere zur Prävention und Behandlung einer durch Krebschemotherapie, Strahlentherapie, Knochenmarktransplantation oder Arzneimitteltherapie sowie durch eine immunologische Anomalie oder Anämie hervorgerufenen Zytopenie.
Claims (12)
- Ketonderivat der folgenden allgemeinen Formel (I) (worin A die allgemeine Formel (II) oder (III) aufweist, m die ganze Zahl 1 oder 2 ist, R1 bis R7 unabhängig voneinander ein Wasserstoffatom oder ein aus der Gruppe, die ein Fluoratom, ein Chloratom, ein Bromatom, C1-19-Alkylgruppen, Alkenylgruppen mit bis zu 19 Kohlenstoffatomen, C6-12-Arylgruppen, C6-12-Aryl-C1-19-Alkylgruppen, C1-19-Alkyl-C6-12-Arylgruppen, C6-12-Aryl-C2-19-Alkenylgruppen, C2-19-Alkenyl-C6-12-Arylgruppen, die Hydroxylgruppe, C1-10-Alkoxygrupen, die Nitrogruppe, Carboxylgruppe, C2-10-Alkoxycarbonylgruppen und die Cyanogruppe umfasst, ausgewählter Substituent sind oder R1 und R2 zusammen -CH=CH-CH=CH- sind, R5 und R6 zusammen -CH=CH-CH=CH- oder -(CH2)q- sind (wobei q eine ganze Zahl im Bereich 2 bis 5 ist) oder R6 und R7 zusammen -(CH2)r- sind (wobei r eine ganze Zahl im Bereich 2 bis 5 ist), R8 und R9 jeweils unabhängig voneinander ein Wasserstoffatom oder ein aus der Gruppe, die C1-6-Alkylgruppen, Alkenylgruppen mit bis zu 6 Kohlenstoffatomen, C6-12-Arylgruppen, C12-Aryl-C1-19-Alkylgruppen, C1-19-Alkyl-C6-12-Arylgruppen, C6-12-Aryl-C2-19-Alkenylgruppen, C2-19-Alkenyl-C6-12-Arylgruppen, C2-10-Alkoxycarbonylgruppen umfasst, ausgewählter Substituent sind oder R8 und R9 zusammen -(CH2)4- oder -(CH2)5- sind, R10 ein Wasserstoffatom oder ein aus einem Fluoratom, Chloratom oder Bromatom ausgewählter Substituent ist, R11 ein Wasserstoffatom oder ein aus einer C1-6-Alkylgruppe oder C6-12-Arylgruppe ausgewählter Substituent ist oder R10 und R11 zusammen -CH=CH-CH=CH- oder -(CH2)t- sind (wobei t eine ganze Zahl im Bereich 2 bis 5 ist), X = O, S, CH2 oder NH ist, Z ein Wasserstoffatom oder eine Methylgruppe ist, Y = O oder S ist, n = 0 oder 1 ist) oder ein pharmakologisch annehmbares Salz davon.
- Ketonderivat nach Anspruch 1 oder ein pharmakologisch annehmbares Salz davon, worin R1 bis R7 jeweils unabhängig voneinander ein Wasserstoffatom, ein Fluoratom, ein Chloratom, ein Bromatom, eine C1-5-Alkylgruppe oder eine Phenylgruppe sind.
- Ketonderivat nach Anspruch 1 oder ein pharmakologisch annehmbares Salz davon, worin Z ein Wasserstoffatom ist.
- Ketonderivat nach Anspruch 1 oder ein pharmakologisch annehmbares Salz davon, worin X = O oder CH2 ist.
- Ketonderivat nach einem der vorangegangenen Ansprüche oder ein pharmakologisch annehmbares Salz davon zur Verwendung als Arzneimittel.
- Ketonderivat nach einem der Ansprüche 1 bis 6 oder ein pharmakologisch annehmbares Salz davon zur Verwendung als blutbildendes Mittel.
- Ketonderivat nach einem der Ansprüche 1 bis 6 oder ein pharmakologisch annehmbares Salz davon zur Verwendung in der Behandlung und Prävention von Zytopenie.
- Verwendung eines Ketonderivats nach einem der Ansprüche 1 bis 6 oder eines pharmakologisch annehmbaren Salzes davon zur Herstellung eines blutbildenden Mittels.
- Verwendung eines Ketonderivats nach einem der Ansprüche 1 bis 6 oder eines pharmakologisch annehmbaren Salzes davon zur Herstellung eines blutbildenden Mittels zur Behandlung und Prävention von Zytopenie.
- Pharmazeutische Zusammensetzung, umfassend ein Ketonderivat nach einem der Ansprüche 1 bis 6 oder ein pharmakologisch annehmbares Salz davon sowie ein pharmakologisch annehmbares Additiv.
