Description
- TECHNISCHES GEBIET
- Die vorliegende Erfindung betrifft ein Aerosolpräparat, das beim Sprühen einen Sorbet-ähnlichen Feststoff auf der besprühten Stelle bildet und über einer verstärkte Kühlwirkung verfügt.
- EINSCHLÄGIGER STAND DER TECHNIK
- Um den durch Quetschung, Verstauchung, Muskelermüdung, usw. hervorgerufenen Schmerz oder durch Fußpilz, Insektenbiss, usw. hervorgerufenen Juckreiz rasch zu stillen, ist es wirksam, den angegriffenen Teil zu kühlen. In der Vergangenheit sind für derartige Symptome die Aerosolpräparate häufig zur Anwendung gelangt, die beim Versprühen keinen Sorbet-ähnlichen Feststoff bilden. Durch diese Aerosolpräparate kann jedoch keine dauerhafte schmerzstillende oder den Juckreiz stillende Wirkung auf dem angegriffenen Teil erzielt werden, da derartige Aerosolpräparate auf der Haut keine dauerhafte kühlende Wirkung haben. Die Beschreibung der WO 90/11068 und die Japanische Patentanmeldung Kokai 4-103526 offenbaren Aerosolpräparate, die ein Sorbet-ähnliches Schaumgel bilden, um diese Nachteile zu überwinden. Allerdings erfordern die Aerosolpräparate bekannter Ausführung komplizierte Prozeduren für die Herstellung des Konzentrats, wie beispielsweise Prozeduren des Erhitzens und Emulgierens, und es besteht bei ihnen ein Problem des geringen Wirkungsgrads in der Herstellung.
- Darüber hinaus kommt es darauf an, dass die bekannten Aerosolpräparate Wasser enthalten, das beim Versprühen einen Sorbet-ähnlichen Feststoff bildet, da die anhaltende Kühlwirkung dieser Präparate durch Gefrieren des darin enthaltenden Wassers erzielt werden kann. Dementsprechend ist es unmöglich, dass in den bekannten Präparaten in Wasser labile Medikamente oder stark lipophile Medikamente enthalten sind.
- OFFENBARUNG DER ERFINDUNG
- Als Ergebnis umfangreicher Forschungsarbeiten zur Lösung der vorgenannten Aufgaben haben die Erfinder der vorliegenden Erfindung festgestellt, dass ein Aerosolpräparat, das einen niederen Alkohol, einen höheren Alkohol und ein verflüssigtes Gas aufweist, keine Emulgierungsprozedur für die Herstellung benötigt und eine dauerhafte Kühlwirkung hat, da das Aerosolpräparat einen Sorbet-ähnlichen festen Stoff beim Versprühen bildet, wodurch das Ziel der Erfindung erreicht wurde.
- Das bedeutet, die vorliegende Erfindung betrifft ein Aerosolpräparat, das beim Versprühen einen Sorbet-ähnlichen Feststoff bildet, der ein Konzentrat mit einem Gehalt eines niederen Alkohols mit 1 bis 3 Kohlenstoffatomen aufweist sowie einen vorgegebenen höheren Alkohol mit mindestens 12 Kohlenstoffatomen und ein verflüssigtes Gas enthaltendes Treibmittel.
- Alle bekannten Aerosolpräparate bilden beim Versprühen durch Gefrieren des darin enthaltenden Wassers einen Feststoff, während im Gegensatz dazu das Aerosolpräparat der vorliegenden Erfindung nicht immer notwendigerweise Wasser enthält und dadurch gekennzeichnet ist, dass das Aerosolpräparat unter völlig neuen Bedingungen beim Versprühen einen Feststof fbildet.
- Entsprechend dem Aerosolpräparat der vorliegenden Erfindung bildet der höhere Alkohol, der durch die Verdampfungswärme des verflüssigten Gases, einschließlich des niederen Alkohols gekühlt wird, einen Sorbet-ähnlichen Feststoff beim Versprühen.
- In der vorliegenden Erfindung bedeutet der niedere Alkohol, der 1 bis 3 Kohlenstoffatome hat, einen geradkettigen oder verzweigten Alkohol und beispielsweise Methanol, Ethanol, denaturiertes Ethanol, Propanol, Isopropanol, usw. und bevorzugt Ethanol. Die Menge des niederen Alkohols beträgt 0,3% bis 65 Gew.% bezogen auf die Gesamtmenge des Präparats und mehr bevorzugt 1,5% bis 50 Gew.% und am meisten bevorzugt 15% bis 35 Gew.%. Wenn die Menge des niederen Alkohols kleiner ist als 0,3 Gew.%, kann das Konzentrat nicht ohne weiteres mit dem Treibmittel homogen gemischt werden. Wenn die Menge des niederen Alkohols mehr als 65 Gew.% beträgt, wird das Kältegefühl vermindert, da die Treibmittelmenge relativ verringert ist.
- Der höhere Alkohol der vorliegenden Erfindung, der mindestens 12 Kohlenstoffatome hat, ist: Laurylalkohol, Hexadecylalkohol, Stearylalkohol, Cetylstearylalkohol, Behenylalkohol, Wollwachsalkohol oder hydrierter Wollwachsalkohol. Diese können allein oder in Mischung miteinander verwendet werden, wobei Stearylalkohol oder Hexadecylalkohol bevorzugt sind.
- Die Menge des höheren Alkohols beträgt 0,005% bis 14 Gew.% bezogen auf die Gesamtmenge des Präparats und mehr bevorzugt 0,015% bis 10 Gew.% und am meisten bevorzugt 0,5% bis 5 Gew.%. Wenn die Menge des höheren Alkohols kleiner ist als 0,005 Gew.%, wird das anhaltende Kältegefühl vermindert, da das Aerosolpräparat nicht ohne weiteres beim Versprühen den Feststoff bilden kann. Wenn die Menge des höheren Alkohols andererseits mehr als 14 Gew.% beträgt, wird die Herstellungsprozedur kompliziert, da der höhere Alkohol nicht ohne weiteres aufgelöst werden kann.
- Der höhere Alkohol muss in dem niederen Alkohol und dem verflüssigten Gas absolut aufgelöst sein, um so ein homogenes System zu ergeben, wodurch die Wirkung der vorliegenden Erfindung erzielt werden kann. Zur Lösung der Aufgabe ist es möglich, wenn der höhere Alkohol nicht ausreichend aufgelöst werden kann, dass andere Lösemittel oder solubilisierende Mittel enthalten sind (z.B. Polyoxyethylenalkylether, Polyoxyethylen-hydriertes Ricinusöl, Diisopropylazipat, Isopropylmyristat, 1,3-Butylenglykol, Propylenglykol, Polyethylenglykol, Glycerin, Milchsäre, Natriumhydroxid, usw.).
- Das verflüssigte Gas ist Dimethylether. Die Menge des verflüssigten Gases beträgt bevorzugt 50% bis 85 Gew.% bezogen auf die Gesamtmenge des Präparats.
- Wenn die Bedingungen, unter denen das Aerosolpräparat der vorliegenden Erfindung einen Sorbet-ähnlichen Feststoff bildet, zwischen dem Konzentrat und dem Treibmittel des Aerosolpräparats unterschieden werden, ist es notwendig, dass mindestens 6 Gew.% des niederen Alkohols in dem Konzentrat enthalten sind, der 1 bis 3 Kohlenstoffatome enthält, mindestens 1 Gew.% des höheren Alkohols in dem Konzentrat enthalten sind, der mindestens 12 Kohlenstoffatome enthält, und 0,5% bis 20 Gewichtsteile des verflüssigten Gases bezogen auf 1 Gewichtsteil des Konzentrats enthalten sind.
- Das Aerosolpräparat der vorliegenden Erfindung kann durch Auflösen des niederen Alkohols und des höheren Alkohols hergestellt werden, um ein homogenes System zu liefern und nach Erfordernis durch Zusetzen solcher anderen Komponenten, die das homogene System nicht zerstören, sowie Einfüllen des resultierenden Konzentrats zusammen mit dem verflüssigten Gas in einen Aerosolbehälter. Beispiele für die Aerosolbehälter schließen Behälter ein, die aus Metallen oder Kunststoffen hergestellt sind, wie sie in der Regel zur Anwendung gelangen. Hinsichtlich der anhaltenden Kühlwirkung ist vorzugsweise eine geeignete Wassermenge enthalten, so dass eine andere Komponente das homogen System nicht zerstört. Im Falle der Zugabe von Wasser zu dem Konzentrat können nicht mehr als 90 Gew.% Wasser bezogen auf die Gesamtmenge des Konzentrats zugesetzt werden, wobei jedoch bevorzugt 20% bis 60 Gew.% Wasser dem Konzentrat hinsichtlich der leichteren Bemessung des Präparats zugesetzt werden.
- Um die schmerz- und juckreizstillende Wirkung auf dem angegriffenen Teil zu verbessern, kann das Aerosolpräparat der vorliegenden Erfindung solche medikamentenwirksame Bestandteile enthalten, die die Wirkung der vorliegenden Erfindung nicht beeinträchtigen, wie beispielsweise entzündungshemmende und schmerzstillende Mittel (z.B. Indomethacin, Methylsalicylat, Monoglykolsalicylat, Ketoprofen, Flurbiprofen, Piroxicam, Diclofenac, Ibuprofen, Mephenamsäure, Dexamethasonacetat, usw.), Antipruriginosa (z.B. Crotamiton, Ichthammol, Mochthamol, Thymolsäure, usw.), antimykotische Mittel (z.B. Amorolfin-hydrochlorid, Undecylensäure, Pentachlorphenol, Clotrimazol, Tolnaftat, Trichomycin, Miconazolnitrat, Lanoconazol, Sulconazolnitrat, Oxyconazolnitrat, Bifonazol, usw.), Antihistaminika (z.B. Diphenhydramin, Diphenhydramin-hydrochlorid, Isothipendyl-hydrochlorid, usw.), örtliche Anästhetika (z.B. Lidocain, Dibucain-hydrochlorid, usw.), Antibiotika (z.B. Kaliumiodid, Chlorhexidingluconat, Acrinol, Benzalkoniumchloid, usw.), Antiphlogistika (z.B. Penicillin, Tetracyclin-hydrochlorid, Fradiomycin, Kanamycin, usw.), kühlende Mittel (z.B. 1-Menthol, Kampfer, Pfefferminzöl, usw.) und dergleichen. Die Mengen dieser medikamentenwirksamen Komponente sind individuell verschieden, betragen bevorzugt jedoch 0,001% bis 10 Gew.% bezogen auf die Gesamtmenge des Präparats.
- Das Aerosolpräparat der vorliegenden Erfindung kann nach Erfordernis alle beliebigen Additive enthalten, die für normale Aerosolpräparate verwendbar sind, so lange sie die Wirkung der vorliegenden Erfindung nicht beeinträchtigen, wie beispielsweise Antioxidantien (z.B. Dibutylhydroxytoluol, usw.), perkutane Absorptionsvermittler (z.B. Glycerinester von Fettsäuren, Fette, aliphatische Carboxylate, Nicotinate, Azon, Alkylenglykolester von Fettsäure, usw.), Parfüme, Farbstoffe und dergleichen.
- GEWERBLICHE NUTZBARKEIT
- Die vorliegende Erfindung ermöglicht die Bereitstellung eines Aerosolpräparats, das beim Versprühen einen Sorbet-ähnlichen Feststoff bildet, keine komplizierte Prozedur des Emulgierens für die Herstellung erfordert und eine Kühlwirkung hat, die mit derjenigen von Erzeugnissen bekannter Ausführung vergleichbar ist.
- BESTE AUSFÜHRUNGSFORM DER ERFINDUNG
- Die vorliegende Erfindung wird detaillierter mit Hilfe der folgenden Beispiele und des Testbeispiels veranschaulicht.
- BEISPIEL 1
- Es wurden 4,76 Gewichtsteile Stearylalkohol und 28,53 Gewichtsteile Ethanol gemischt, gerührt und homogen aufgelöst, um ein Konzentrat zu ergeben. Das Konzentrat wurde in einen druckfesten Behälter eingefüllt und auf den Behälter ein Ventil aufgesetzt und in diesen 0,07 Gewichtsteile Propan, 1,93 Gewichtsteile n-Butan, 0,93 Gewichtsteile Isobutan und 63,78 Gewichtsteile Dimethylether eingefüllt wurden. Auf den Behälter wurde ein Sprühkopf aufgesetzt, um ein Aerosolpräparat zu erhalten.
- BEISPIEL 2
- Es wurden 0,31 Gewichtsteile Indomethacin, 4,11 Gewichtsteile Diisopropylazipat, 1,23 Gewichtsteile Polyoxyethylenalkylether, 16,47 Gewichtsteile denaturiertes Ethanol, 13,39 Gewichtsteile gereinigtes Wasser und 1,23 Gewichtsteile Cetylallcohol gemischt, gerührt und homogen aufgelöst, um ein Konzentrat zu ergeben. Das Konzentrat wurde in einen druckfesten Behälter eingefüllt und auf den Behälter ein Ventil aufgesetzt und in diesen 63,26 Gewichtsteile Dimethylether gefüllt. Es wurde ein Sprühkopf auf den Behälter aufgesetzt, um ein Aerosolpräparat zu erhalten.
- BEISPIEL 3
- Es wurden 0,35 Gewichtsteile Miconazolnitrat, 1,76 Gewichtsteile Diisopropylazipat, 1,05 Gewichtsteile Isopropylmyristat, 0,70 Gewichtsteile Glycerin, 15,80 Gewichtsteile denaturiertes Ethanol und 8,74 Gewichtsteile gereinigtes Wasser, 0,53 Gewichtsteile Stearylalkohol, 0,53 Gewichtsteile Cetylalkohol, 0,70 Gewichtsteile Polyoxyethylenalkylether und 0,35 Gewichtsteile Polyoxyethylenhydriertes Ricinusöl gemischt, gerührt und homogen aufgelöst, um ein Konzentrat zu ergeben. Das Konzentrat wurde in einen druckfesten Behälter eingefüllt und auf den Behälter ein Ventil aufgesetzt und in diesen 69,49 Gewichtsteile Dimethylether gefüllt. Es wurde ein Sprühkopf auf den Behälter aufgesetzt, um ein Aerosolpräparat zu erhalten.
- VERGLEICHSBEISPIEL 1
- Nach der gleichen Methode wie in Beispiel 1 unter Verwendung der Rezeptur von Beispiel 1, worin Stearylalkohol durch Ethanol ersetzt wurde, wurde ein Aerosolpräparat erhalten.
- VERGLEICHSBEISPIEL 2
- Nach der gleichen Methode wie in Beispiel 2 unter Verwendung der Rezeptur von Beispiel 2, worin Polyoxyethylenalkylether und Cetylalkohol durch denaturiertes Ethanol bzw. gereinigtes Wasser ersetzt wurden, wurde ein Aerosolpräparat zum Vergleich erhalten.
- VERGLEICHSBEISPIEL 3
- Es wurden 0,36 Gewichtsteile Miconazolnitrat, 1,79 Gewichtsteile Diisopropylazipat, 1,07 Gewichtsteile Isopropylmyristat, 0,71 Gewichtsteile Glycerin, 17,06 Gewichtsteile denaturiertes Ethanol und 8,27 Gewichtsteile gereinigtes Wasser gemischt, gerührt und homogen aufgelöst, um ein Konzentrat zu ergeben. Das Konzentrat wurde in einen druckfesten Behälter gefüllt und ein Ventil auf den Behälter aufgesetzt und in diesen 70,74 Gewichtsteile Dimethylether gefüllt. Es wurde ein Sprühkopf auf den Behälter aufgesetzt, um ein Aerosolpräparat für den Vergleich zu ergeben.
- VERGLEICHSBEISPIEL 4 (Ein Aerosolpräparat, das für die Herstellung eine Emulgierungsprozedur erfordert)
- Es wurden 0,31 Gewichtsteile Indomethacin, 4,07 Gewichtsteile Diisopropylazipat, 1,22 Gewichtsteile Polyoxyethylensorbitanmonostearat, 0,81 Gewichtsteile Polyoxyethylensorbitanmonostearat und 1,22 Gewichtsteile Sorbitanmonostearat unter Erhitzen aufgelöst und mit 15,94 Gewichtsteilen heißem gereinigtem Wasser gründlich gemischt. Nach dem Kühlen unter Rühren wurde die Mischung mit 14,23 Gewichtsteilen denaturiertem Ethanol gemischt und homogen dispergiert, um ein Konzentrat zu ergeben. Das Konzentrat wurde in einen druckfesten Behälter gefüllt und ein Ventil auf den Behälter aufgesetzt und in diesen 62,20 Gewichtsteile Dimethylether gefüllt. Es wurde ein Sprühkopf auf den Behälter aufgesetzt, um ein Aerosolpräparat zu ergeben.
- TESTBEISPIEL 1
- Es wurde ein Thermoelement an einem Folienstück mit Hilfe von Klebbändern befestigt und die Folie auf einem bei 33°C geregelten thermostatischen Wasserbad ausgestrichen. Die in den Beispielen 1 bis 3 und den Vergleichsbeispielen 1 bis 4 erhaltenen Aerosolpräparate wurden jeweils auf das Thermoelement für 3 Sekunden gesprüht. Die Eigenschaften des Aerosolpräparats wurden beim Versprühen festgestellt und die von dem Thermoelement in Abhängigkeit von der Zeit angezeigten Werte gemessen. Die Ergebnisse sind in Tabelle 1 dargestellt.
- Die Aerosolpräparate der Beispiele 1 bis 3 und des Vergleichsbeispiels 4 bildeten beim Versprühen einen Sorbet-ähnlichen Feststoff, wobei jedoch die der Vergleichsbeispiele 1 bis 3 keinerlei Sorbet-ähnlichen Feststoff bildeten. Anhand der Temperaturänderung auf dem Thermoelement betrugen die Temperaturen durch die Aerosolpräparate der Vergleichsbeispiele 1 bis 3 mindestens 30°C bei 30 Sekunden nach dem Sprühen und kehrten zu der ursprünglichen Temperatur nach 45 Sekunden nach dem Sprühen zurück, während im Gegensatz dazu die Aerosolpräparate der Beispiele 1 bis 3 und des Vergleichsbeispiels 4 eine bleibende Kühlwirkung zeigten.

- Aerosolpräparat, das beim Sprühen einen Sorbet-ähnlichen Feststoff bildet, aufweisend: (a) ein Konzentrat, enthaltend: 0,3% bis 65 Gew.% bezogen auf die Gesamtmenge des Präparats eines niederen Alkohols mit 1 bis 3 Kohlenstoffatomen und 0,005% bis 14 Gew.% bezogen auf die Gesamtmenge des Präparats eines höheren Alkohols mit mindestens 12 Kohlenstoffatomen, ausgewählt aus: Laurylalkohol, Hexadecylalkohol, Stearylalkohol, Cetylstearylalkohol, Behenylalkohol, Wollwachsalkohole, hydrierte Wollwachsalkohole und Mischungen davon; und (b) ein Treibmittel, enthaltend 50% bis 85 Gew.% bezogen auf die Gesamtmenge des Präparats Dimethylether.
- Aerosolpräparat nach Anspruch 1, worin der niedere Alkohol mit 1 bis 3 Kohlenstoffatomen in einer Menge von mindestens 6 Gew.% bezogen auf das Konzentrat vorliegt, der höhere Alkohol mit mindestens 12 Kohlenstoffatomen in einer Menge von mindestens 1 Gew.% bezogen auf das Konzentrat vorliegt und Dimethylether in einer Menge von 0,5 bis 20 Gewichtsteilen bezogen auf das Gewicht des Konzentrats vorliegt.
- Aerosolpräparat nach Anspruch 1, worin der niedere Alkohol Ethanol ist.
- Aerosolpräparat nach Anspruch 1, worin der höhere Alkohol mindestens ein Alkohol ist, der ausgewählt ist aus der Gruppe, bestehend aus Stearylalkohol und Hexadecylalkohol.
- Aerosolpräparat nach Anspruch 1, worin das Konzentrat 20% bis 60 Gew.% Wasser enthält.
