Description
- Die vorliegende Erfindung betrifft 1,3,5-Triazinderivate der allgemeinen Formel (I)
- worin
- R für einen C&sub5;-C&sub1;&sub2; Cycloalkylrest steht&sub1; welcher durch C&sub1;- C&sub4;-Alkylreste mono- oder polysubstituiert sein kann, oder eine Gruppe der Formel (II), (III) oder (IV).
- worin R2 gleich C&sub1;-C&sub9;-Alkyl ist,
- n eine ganze Zahl von 0 bis 3 bedeutet,
- R&sub3; ein Wasserstoffatom oder einen Methylrest darstellt,
- A für C&sub5;-C&sub8;-Cycloalkyl oder C&sub4;-C&sub8;-Alkyl steht,
- B gleich C&sub1;-C&sub4;-Alkyl ist,
- m eine ganze Zahl von 1 bis 10 bedeutet, und
- R' und R'', die gleich oder unterschiedlich sind, dieselbe Bedeutung wie R haben oder Wasserstoff, ein Alkalimetall oder eine Ammoniumgruppe darstellen, die gegebenenfalls durch Alkyl- oder Hydroxylalkylreste substituiert ist, oder C&sub1;-C&sub1;&sub8;-Alkyl bedeuten, sowie ein Verfahren zu ihrer Herstellung und ihre Verwendung als Lichtschutzmittel.
- Bekanntlich übt die ultraviolette Strahlung im Sonnenlicht eine schädliche Einwirkung auf das Hautgewebe aus und bewirkt einen Abbau von Polymeren. Durch die Verwendung bestimmter chemischer Komponenten, sogenannter Sonnenschutzfaktoren, die den UV-Anteil der Sonnenstrahlung absorbieren können, ist es möglich, die schädlichen Effekte der Strahlung, Hautalterung und Alterung synthetischer Polymermaterialien zu verhindern oder zumindest zu verlangsamen.
- Zahlreiche Substanzen sind als schützende Wirkstoffe untersucht und in Experimenten verwendet worden, und es existiert ausführliche Patentliteratur zu diesem Thema, worin Substanzen verschiedener chemischer Klassen vorgeschlagen werden, welche die Fähigkeit besitzen, Strahlung im ultravioletten Bereich zu absorbieren, insbesondere die gefährlichste Strahlung zwischen 290 und 320 nm, sogenanntes UVB.
- Von diesen Substanzen kommen jedoch nur relativ wenige für die praktische Anwendung in Frage, so z. B. Ester von p-Methoxyzimtsäure und p-Dimethylaminobenzoesäure, Benzotriazole und Hydroxybenzophenone.
- (In SOFW, 115 (1989) 21. November No. 18, wird die Substanz Octyltriazon (Uvinul T 150) als brauchbarer UVB- Absorber beschrieben.)
- Ein gemeinsamer Nachteil all dieser Substanzen ist ihre geringe Absorptionsfähigkeit, so daß es notig ist, relativ große Mengen zu verwenden, um die beste Lichtschutzkapazität zu erreichen, d. h. ihr sogenannter Sonnenschutzfaktor ist relativ niedrig. Der Lichtschutzfaktor (SPF) ist eine Maßeinheit des Lichtschutzvermögens eines Sonnenschutzes oder einer kosmetischen Formulierung, welche einen oder mehrere Sonnenschutzfaktoren enthält. Er ist direkt mit der spezifischen Extinktion verknüpft und wird in vivo durch Tests am Menschen bestimmt.
- Diese Tests sind sehr zeitaufwendig und teuer, so daß viele Anläufe unternommen wurden Techniken zu entwickeln, die geeignet sind, den SPF in vitro zu bestimmen. Eine Methode, den SPF in vitro zu bestimmen, die hinreichend zuverlässige Ergebnisse lieferte, ist die von B.L. Diffey und J. Robson in J. Soc. Cosmet. Chem. 40, 127-133 (1989) beschriebene Methode. Diese Methode besteht in der Verwendung eines synthetischen Materials, Transpor, als einen Ersatz für menschliche Haut und in der spektroradiometrischen Messung der UV-Strahlung von einem Sonnenschutz, der auf das oben genannte Substrat aufgetragen wird. Die so erhaltenen Ergebnisse mit Sonnenschutzsubstanzen, die schon bekannt sind und bereits in Kosmetika verwendet werden, sind dabei den experimentell in vivo erhaltenen Werten sehr ähnlich.
- Im Patent DE 3,206,398 werden neue Sonnenschutzsubstanzen beschrieben, die von 5-Triazin abstammen und durch Reaktion von Dichlortriazin mit Estern von p- Aminobenzoesäure erhalten werden. Diese Substanzen absorbieren sehr intensiv im UVB-Bereich und haben demzufolge einen wesentlich größeren SPF als bisher bekannte Sonnenschutzsubstanzen.
- Es wurde nun gefunden, daß die erfindungsgemäßen Substanzen einen unerwartet hohen SPF aufweisen, höhere Werte als die Produkte nach DE 3,206,398. Eine weiterere Aufgabe der Erfindung ist demnach die Verwendung der Substanzen der Formel (I) als Sonnenschutzsubstanzen und Lichtstabilisatoren.
- Die erfindungsgemäßen Verbindungen können zweckmäßigerweise zur Lichtstabilisation synthetischer Polymere gegen Sonnenstrahlung zwischen 290 und 320 nm Wellenlänge verwendet werden, um Photodegradation und Veränderungen zu vermeiden. Es wurde auch gefunden, daß die Verbindungen der Formel (I) eine überraschend hohe Schutzaktivität auf der Haut gegenüber dem schädigenden Teil der Sonnenstrahlung aufweisen.
- Die erfindungsgemäßen Verbindungen können durch Reaktion von symmetrischem Trichlortriazin oder Tribromtriazin mit Estern von p-Aminobenzoesäure der Formel (V) hergestellt werden,
- wobei R&sub4; der Definition der oben definierten R, R' und R'' entspricht.
- Geeignete Lösungsmittel, in denen die Reaktion ausgeführt werden kann, sind diejenigen, die inert gegenüber den Reagenzien sind, wie z. B. Acetonitril, Ketone wie Aceton und Methylethylketon, Ether wie z. B. Diethylether, Diisopropylether, Tetrahydrofuran und Dioxan sowie aliphatische oder aromatische Kohlenwasserstoffe wie Pentan, Heptan, Cyclohexan, Benzol, Toluol, Xylol oder Mixturen davon.
- Die Reaktion kann in An- oder Abwesenheit von Säureakzeptoren durchgeführt werden, z. B. Hydroxiden von Alkalimetallen oder Erdalkalimetallen oder Bicarbonat oder Carbonaten von Alkalimetallen in molaren Verhältnissen von 3 - 4 mol des Esters von p- Aminobenzoesäure pro ein Mol von Trichlortriazin.
- Wenn die Substituenten R, R' und R'' sich voneinander unterscheiden, wird die Reaktion zwischen dem Trihalogentriazin und den verschiedenen Estern in Stufen ausgeführt, gegebenenfalls unter Isolierung und erforderlichenfalls unter Reinigung der Zwischenprodukte, vor der folgenden Reaktion mit weiterem Ester von p- Aminobenzoesäure; diese Synthesemethoden und Reaktionen von Trihalogentriazin zur Substitution von drei Halogenatomen durch identische oder unterschiedliche Aminradikale sind jedoch gut bekannt und in der technischen Literatur häufig beschrieben, besonders in der sich mit bestimmten Typen von Farbstoffen und optischen Aufhellern beschäftigenden Literatur.
- Einige der Ester der Formel (V) sind bekannt und andere, die neuartig sind, werden durch bekannte Methoden hergestellt.
- Die erfindungsgemäßen Verbindungen besitzen zusätzlich zu der sehr großen Absorption im UVB-Bereich noch andere Charakteristika, die als nötig für eine praktische Anwendung erachtet werden, so z. B. Lichtstabilität, Hitzestabilität, Ungiftigkeit usw. Tatsächlich sollte ein optimaler UVB-Absorber folgende Eigenschaften besitzen:
- 1. Eine hohe spezifische Extinktion E' bei 290 - 320 nm, was zu geringeren Anwendungsdosen und deshalb zu Rentabilität sowie einem minimalen toxikologischen Risiko führt.
- 2. Lichtstabilität.
- 3. Hitzestabilität.
- 4. Oxidationsstabilität.
- 5. Stabilität bei verschiedenen pH-Werten.
- 6. Gute Löslichkeit, Emulgierbarkeit oder Löslichkeit in Ausgangssubstanzen, die gewöhnlich als Bestandteile von Hautkosmetika verwendet werden.
- 7. Vernachlässigbare Giftigkeit.
- 8. Keine signifikante Farbe oder Geruch.
- 9. Relativ großes Molekulargewicht, was eine geringere Wahrscheinlichkeit der Absorption durch die Haut bedeutet und deshalb eine größere Sicherheit aus toxokologischer Sicht beinhaltet.
- 10. Verträglichkeit mit den vielen Substanzen, die normalerweise in dermatologischen Formulierung verwendet werden.
- Nach einer bevorzugten Ausführungsform der Erfindung werden die Verbindungen der Formel (I) enthaltenden Gemische zum Schutze der Haut gegenüber den schädigenden Effekten der Sonnenstrahlung verwendet.
- Die erfindungsgemäßen Verbindungen können natürlich - gegebenenfalls auch in Kombination mit anderen Stabilisatoren - zu Kosmetika und auch zu synthetischen Polymeren generell in Mengen zwischen 0,05 und 10 Gew% vorzugsweise jedoch von 0,1 bis 5 Gew% des Polymers oder des Kosmetikums hinzugefügt werden.
- Die Kosmetika können zu verschiedenen Typen gehören und können für verschiedene Zwecke verwendet werden.
- Normalerweise liegen sie in Form von Salben, Cremes, Lotionen und Emulsionen vor.
- Die Substanzen der Formel (I) werden hinzugefügt, um entweder die Formulierung selbst zu schützen, so z. B. um unerwünschte Entfärbung zu vermeiden, oder um die mit dem Kosmetikum behandelte Epidermis gegen die schädlichen Effekte der UVB-Strahlung zu schützen, welche Sonnenbrand verursacht und die Alterung der Haut beschleunigt, so daß sie vorzeitig trocken, faltig oder schuppig wird.
- Die folgenden Beispiele beschreiben die vorliegende Erfindung.
- 4,6 g Cyanchlorid und 17,5 g von Cyclohexyl-p- aminobenzoat werden zu 150 ml Xylol gegeben. Das Reaktionsgemisch wird 6 Stunden unter Rückfluß gerührt.
- 100 ml Oktan werden hinzugefügt und das Gemisch wird auf 0-5ºC gekühlt. Das weiße kristalline Produkt wird abfiltriert, gewaschen und im Vakuum getrocknet. Dies ergibt 16 g von 2,4,6-Tris-[p-(cyclohexyloxycarbonyl)-anilino]-s-triazin.
- Schmelzpunkt: 171 - 173 ºC
- UV (Methanol) : E' = 1691 bei 312 nm
- Die in Tabelle 1 aufgelisteten Substanzen werden in Übereinstimmung mit dem in Beispiel 1 beschriebenen Verfahren hergestellt. Tabelle 1
- 6,7 g Natriumbicarbonat werden zu einer Lösung von 13,8 g Cyanchlorid in 150 ml Aceton, gekühlt auf 0ºC, hinzugefügt. Eine Lösung von 27,7 g Di-[2-(4-tert.- octylphenoxy)-ethyl]-4-aminobenzoat in 100 ml Aceton wird langsam zu dem erhaltenen Gemisch hinzugefügt, wobei die Temperatur bei 0 - 2 ºC gehalten wird.
- Das Gemisch wird für 30 Minuten bei 0º gerührt, dann werden 50 ml Wasser langsam hinzugefügt. Es ergibt sich eine sehr dichte Suspension, die für weitere 30 Minuten bei 0ºC gerührt und dann filtriert wird. Der Filterkuchen wird viele Male mit Wasser und dann mit Aceton gewaschen und anschließend bei 40º0 im Vakuum getrocknet. Dies ergibt 36 g von 2-[p-[2-(4-tert.-Octylphenoxy)ethoxycarbonyl]-anilino]-4,6-dichlor-s-triazin.
- Schmelzpunkt 249 - 251 ºC.
- 12,8 g Methyl-p-aminobenzoat werden zu 20,7 g dieses Zwischenproduktes in 200 ml Xylol hinzugefügt und das Gemisch wird für 5 Stunden unter Rückfluß gerührt. Das Xylol wird dann abdestilliert und der Rückstand in Isopropanol auskristallisiert. Dies ergibt 19 g von [2-(4-tert.-Octylphenoxy)-ethoxycarbonyl]-anilino]-4,6- bis[p-(methoxycarbonyl)-anilino]-s-triazin.
- Schmelzpunkt: 157 - 160 ºC
- UV (Methanol) : E' = 1636 bei 312,6 nm.
- Unter der Verwendung eines ähnlichen wie in Beispiel 8 beschriebenen Verfahrens werden die Substanzen von Tabelle 2 hergestellt. Tabelle 2 Tabelle 2 (Fortsetzung)
- Der SPF einiger in den Beispielen beschrieber Substanzen wurde unter Verwendung der in B.L. Diffey und J. Robson in J. Soc. Cosmet. Chem. 40 127 - 133 (1989) bechriebenen Methode bestimmt.
- Die erhaltenen Resultate sind in der folgenden Tabelle 3 aufgelistet. Tabelle 3
- Substanz A ist das in Patent DE 3,206,398 beschriebene Beispiel 1.
- Eine Mischung aus 10 g Cyclodimeticon/Dimeticoncopolymer (Dow Corning Q2-3225), 10 g Cyclometicon (Dow Corning 344), 0,5 g Polysorbat 20 (Tween 20) und 3 g der Substanz aus Beispiel 3 wird angesetzt. Dieses Gemisch wird zu einer vorher angesetzten Lösung aus 0,2 g von 1,1'- Methylen-bis-3-(3'-hydroxymethyl-2,4-dioxoimidazolidinyl)-Harnstoff sowie 0,05 g Methylparaben und 73,25 g Wasser hinzugefügt.

1. Verbindungen der allgemeinen Formel (1)
worin
R für einen C&sub5;-C&sub1;&sub2;-Cycloalkylrest steht, welcher durch
C&sub1;-C&sub4;-Alkylreste mono- oder polysubstituiert sein
kann, oder eine Gruppe der Formel (II), (III) oder
(IV):
worin R&sub2; gleich C&sub1;-C&sub9;-Alkyl ist,
n eine ganze Zahl von 0 bis 3 bedeutet,
R&sub3; ein Wasserstoffatom oder einen Methylrest
darstellt,
A für C&sub5;-C&sub8;-Cycloalkyl oder C&sub4;-C&sub8;-Alkyl steht,
B gleich C&sub1;-C&sub4;-Alkyl ist,
m eine ganze Zahl von 1 bis 10 bedeutet, und
R' und R", die gleich oder unterschiedlich sind, die
selbe Bedeutung wie R haben oder Wasserstoff, ein
Alkalimetall, einen Ammoniumrest, der gegebenenfalls
durch Alkyl- oder Hydroxyalkyl-Reste substituiert
ist, oder C&sub1;-C&sub1;&sub8;-Alkyl bedeuten.

3. Verfahren zur Herstellung der Verbindungen nach
Anspruch 1, dadurch gekennzeichnet, daß eine
Verbindung der allgemeinen Formel (VI)
worin X für Chlor oder Brom steht, mit einem Ester
der 4-Aminobenzoesäure der allgemeinen Formel (V)
worin R&sub4; gemäß der obigen Definition für R, R' und R"
ist, umgesetzt wird.
4. Verwendung der Verbindungen nach Anspruch 1 als
Stabilisatoren für synthetische Polymere.
5. Verwendung der Verbindungen nach Anspruch 1 als für
die Dermatologie geeignete Mittel.
6. Verwendung der Verbindungen nach Anspruch 1 als
Sonnenschutz und Desaktivatoren für freie Radikale
zur Herstellung von kosmetischen und dermatologischen
Zusammensetzungen, welche für den Schutz der Haut
verwendet werden können.
7. Kosmetische und dermatologische Zusammensetzungen,
welche eine Verbindung nach den Ansprüchen 1 oder 2
als aktiven Wirkstoff enthalten.
8. Kosmetische und dermatologische Zusammensetzungen
nach Anspruch 7, welche 0,05 bis 10 Gew% des aktiven
Wirkstoffes aufweisen.
9. Kosmetische und dermatologische Zusammensetzungen
nach Anspruch 7, welche 0,1 bis 5 Gew% des aktiven
Wirkstoffes aufweisen.
