Description
- Diese Erfindung betrifft eine neue Verwendung von bekannten Peptidderivaten der unten folgenden allgemeinen Formel (I) oder deren pharmazeutisch verträgliche Salze für die Herstellung eines Medikamentes zur Behandlung und/oder Prävention von Entzündungen, speziell Arthritis.
- Peptidderivate der unten folgenden allgemeinen Formel (I) sind bereits aus der EP-A-0 336 230 bekannt. Es ist auch bekannt aus dieser Literaturstelle, daß Peptidderivate der Formel (I) pharmakologische Wirkungen besitzen, wie Substanz P- Antagonismus und Neurokinin A-Antagonismus und nützlich zur Herstellung von Medikamenten zur Verwendung in der Behandlung und Prävention von Asthma sind.
- Aus der EP-A-0 394 989 sind Peptidverbindungen mit einer völlig verschiedenen Grundstruktur bekannt, die nützlich in der Herstellung von Medikamenten zur Verwendung in der Behandlung von entzündlichen Erkrankungen sind, wie der rheumatoiden Arthritis. Außerdem nimmt man an, daß diese Peptidverbindungen Antagonisten der Substanz P und der Neurokinine A und B sind, und daher nützlich in der Behandlung oder Prävention von Tachikinin-vermittelten Erkrankungen sind, wie Entzündungen.
- Es ist ein Ziel der vorliegenden Erfindung Verbindungen zu finden, die nützlicher zu Behandlung oder Prävention inflammatorischer Erkrankungen sind, wie denjenigen, die in der EP-A-0 394 989 offenbart sind.
- Überraschender Weise fand der Anmelder, daß die Peptidderivate der allgemeinen Formel (I), die in der EP-A-0 336 230 offenbart wurden, eine starke antiinflammatorische Wirkung besitzen und daher für die Herstellung eines Medikamentes zur Prävention und/oder Behandlung von Entzündungen insbesondere Arthritis verwendet werden können.
- Das Thema der vorliegenden Erfindung besteht in der Verwendung der Peptidderivate folgender allgemeinen Formel:
- worin R¹ Wasserstoff, Phenyl(C&sub1;-C&sub6;)alkoxycarbonyl, (C&sub1;-C&sub6;)- Alkanoyl, (C&sub1;&sub5;-C&sub2;&sub0;)Alkanoyl, Benzoyl, Thienyl(C&sub1;-C&sub6;)alkanoyl, Phenyl(C&sub3;-C&sub6;)alkenyl, substituiert mit einer (C&sub2;-C&sub6;)- Alkenylgruppe, oder Phenyl(C&sub1;-C&sub6;)alkanoyl, substituiert mit einer (C&sub1;-C&sub6;)Alkylgruppe, ist;
- R² Hydroxy ist und
- R³ Carboxy oder (C&sub1;-C&sub6;)Alkoxycarbonyl ist, oder
- R² und R³ miteinander verbunden sind, um eine Gruppe der Formel: -O- - darzustellen;
- R&sup4; Hydroxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder (C&sub1;-C&sub6;)Alkanoyloxy ist;
- R&sup5; Hydroxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder (C&sub1;-C&sub6;)Alkanoyloxy ist;
- R&sup6; Hydroxy, (C&sub1;-C&sub6;))Alkoxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder (C&sub1;-C&sub6;)Alkanoyloxy ist; und
- eine Einfachbindung oder eine Doppelbindung darstellt,
- oder eines pharmazeutisch verträglichen Salzes davon zur Herstellung eines Medikamentes zur Prävention und/oder Behandlung von Entzündungen.
- Die Peptidderivate der obigen allgemeinen Formal (I), die in der vorliegenden Erfindung verwendet werden, besitzen eine starke antiinflammatorische Wirkung, wie aus den in der unten folgenden Tabelle auf Seite 7 zusammengefaßten Testergebnissen ersichtlich ist.
- In ihren bevorzugten Ausführungsformen betrifft die vorliegende Erfindung die Verwendung eines Peptidderivates der allgemeinen Formel (I) worin:
- R¹ 3-(2-Pentylphenyl)propanoyl ist,
- R² und R³ miteinander verbunden sind, um eine Gruppe der Formel: -O- - darzustellen,
- R&sup4; Hydroxy ist,
- R&sup5; Hydroxy ist,
- R&sup6; Hydroxy ist und
- eine Doppelbindung ist, oder eines pharmazeutisch verträglichen Salzes davon, speziell der Verbindung
- Entsprechend einer weiteren bevorzugten Ausführungsform betrifft die vorliegende Erfindung die Verwendung des Peptidderivates der allgemeinen Formel (I), wie sie oben definiert ist, oder eines pharmazeutisch verträglichen Salzes davon für die Herstellung eines Medikamentes zur Prävention und/oder Behandlung von Arthritis.
- Geeignete pharmazeutisch verträgliche Salze der Verbindung (I) sind konventionelle nichtgiftige Salze und können einschließen: Ein Salz mit einer Base oder ein Säureadditionssalz wie ein Salz mit einer anorganischen Base, z. B. Alkalimetallsalz (z. B. Lithiumsalz, Natriumsalz und Kaliumsalz), einem Erdalkalimetallsalz (z. B. Calciumsalz und Magnesiumsalz), einem Ammoniumsalz; einem Salz mit einer organischen Base, z. B., einem organischen Aminsalz (z. B. Triethylaminsalz, Pyridinsalz, Picolinsalz, Ethanolaminsalz, Triethanolaminsalz, Dicyclohexylaminsalz und N,N'-Dibenzylethylendiaminsalz); ein anorganisches Säureadditionssalz (z. B. Hydrochlorid, Hydrobromid, Sulfat und Phosphat; ein organisches Carbonsäure- oder Sulfonsäureadditionssalz (z. B. Formiat, Acetat, Trifluoracetat, Maleat, Tartrat, Methansulfonat, Benzolsulfonat und p- Toluolsulfonat); ein Salz mit einer basischen oder sauren Aminosäure (z. B. Arginin, Asparaginsäure und Glutaminsäure).
- Die Verbindung (I) oder ein pharmazeutisch verträgliches Salz davon, die entsprechend der vorliegenden Erfindung verwendet wird, besitzt antiinflammatorische Wirkungen und ist als solche von Wert als aktiver Bestandteil für die Verwendung in antiinflammatorischen Zusammensetzungen.
- Diese antiinflammatorischen Zusammensetzungen sind wirksam in der Prävention und Behandlung von Entzündungen, wie verschiedene Typen der Arthritis artikulärer Rheumatismus (z. B. rheumatoide Arthritis), Osteoarthrose (z. B. Osteoarthrose des Knies), Periarthritis humeroscarpularis, zerviko-omo-brachiales Syndrom oder ähnliches.
- Die Verbindung (I) oder pharmazeutisch verträgliche Salze davon, die als aktiver Bestandteil in diesen Zusammensetzungen verwendet wird, kann als solche verabreicht werden. Sie wird jedoch im allgemeinen in Form von pharmazeutisch verträglichen Präparaten verabreicht.
- Die oben erwähnten pharmazeutischen Präparate können eine Anzahl von Formen einnehmen, wie orale Präparate [z. B. Lösungen, Emulsionen, Suspensionen, Kapseln, Granulate, Pulver, Tabletten und Sirups], Injektionen, Zäpfchen oder ähnliches.
- Solche Präparate können durch bekannte pharmazeutische Verfahren unter Verwendung von geeigneten Exzipienten hergestellt werden, wie Sucrose, Stärke, Manitol, Sorbitol, Lactose, Glucose, Cellulose, Talk, Calciumphosphat und Calciumcarbonat; Bindemitteln wie Cellulose, Methylcellulose, Hydroxypropylcellulose, Polypropylpyrrolidon, Gelatine, Gummiarabicum, Polyethylenglycol, Sucrose und Stärke; Desintegratoren wie Stärke, Carboxymethylcellulose, Hydroxypropylstärke, Natriumbicarbonat, Calciumphosphat und Calciumcitrat; Schmiermittel wie Magnesiumstearat, Talk und Natriumlaurylsulfat; Geschmackstoffe wie Zitronensäure, Menthol, Glycin und Orangenpulver; Konservierungsmittel wie Natriumbenzoat, Natriumbisulfit, Benzylalkohol, Methylparaben und Propylparaben, Stabilisatoren wie Zitronensäure, Natriumcitrat und Essigsäure; Suspensionsmittel wie Methylcellulose, Polyvinylpyrrolidon und Aluminiumstearat; Dispergiermittel wie Hydroxypropylcellulose, Polysorbat 80 und Carboxymethylcellulose; Lösungsmittel wie Wasser, isotonische Mittel wie Natriumchlorid; Basiswachse wie Kakaobutter, Polyethylenglycol und weißes Softparaffin usw.
- Die Dosierung für die pharmazeutische Zusammensetzung dieser Erfindung hängt ab vom Alter, Körpergewicht, dem klinischen Zustand des Patienten, dem Verfahren der Verabreichung und anderen Umständen. Im allgemeinen jedoch wird die aktive Verbindung (I) oder ihr pharmazeutisch verträgliches Salz in Dosen von 1 bis 300 mg, bevorzugt 10 bis 100 mg entweder oral oder parenteral verabreicht (z. B. intraartikulär, intravenös).
- Die folgenden Testbeispiele beabsichtigen, die hervorragende antiinflammatorische Wirkung der Verbindung (I) oder ihrer pharmazeutisch verträglichen Salze, die entsprechend der vorliegenden Erfindung verwendet werden, zu demonstrieren.
- Die Verbindung der folgenden Strukturformel: (Im folgenden kurz als Tetraydro-WS9326A bezeichnet)
- Acht Wochen alte männliche Wistar-Ratten wurden verwendet. Jede Ratte wurde mit 0,2 ml pentobarbitalnatrium (50 mg/ml) in physiologischer Salzlösung i.p. anästhesiert und dann intravenös mit 2 ml/kg Evans-Blau (10 mg/ml) in physiologischer Salzlösung dosiert. Danach wurden 0,1 ml 2%-iges Carrageenin in physiologischer Salzlösung in die Gelenkhöhle des rechten Knies verabreicht. Nach vier Stunden wurde das Tier getötet und enthäutet, um den Gelenkregion freizulegen, und auf das Auslaufen von Evans-Blau untersucht. Dann wurde physiologische Salzlösung (0,15 ml) in die Gelenkhöhle injiziert. Nach dem Einschneiden wurden 5 ul Synovial-Flüssigkeit gesammelt. Die erhaltene Synovial-Flüssigkeit wurde mit physiologischer Salzlösung (40 ul) verdünnt, und unter Verwendung eines Zellzählers wurden die Zellen unter einem Mikroskop gezählt.
- Die Testverbindung (Tetrahydro-WS9326A) wurde in 0,1 % Methylcellulose in physiologischer Salzlösung suspendiert, und 0,1 ml dieser Suspension wurden in das Kniegelenk 15 Minuten vor der Carrageeninverabreichung injiziert. Testergebnis Dosis Anzahl der Tiere Anzahl der Zellen in der Synovialflüssigkeit (x 10&sup4;/ml) % Inhibition Träger (0,1%-ige Methylcellulose in physiologischer Salzlösung) nur
- Eine injizierbare Suspension, die die Testverbindung und die anderen unten erwähnten Bestandteile enthielt, wurde in die Kniegelenke von Kaninchen (6 männliche und 6 weibliche) und Hunden (3 männliche und 3 weibliche) zweimal die Woche für vier Wochen verabreicht. Die Dosis betrug 8 mg/kg für die Kaninchen und 12 mg/kg für die Hunde. Die Verabreichung der Testverbindung verursachte weder den Tod der Kaninchen noch der Hunde.

- Die folgenden Bestandteile wurde routinemäßig vermischt und pulverisiert, um ein Pulver zu ergeben.
- Tetrahydro-WS9326A 30,0 (Gew.-%)
- Lactose 69,4
- Polyoxyl-40-Stearat 0,1
- Hydroxypropoylcellulose 0,5
- Die folgenden Bestandteile wurde routinemäßig vermischt und komprimiert, um Tabletten bereitzustellen. Rezept der Tabletten Tetrahydro-WS9326A Lactose Croscarmellose-Natrium, Typ A Hydroxypropylcellulose Polyoxyl-40-Stearat Magnesiumstearat 420 mg/Tablette
- Falls erforderlich, können die so erhaltenen Tabletten filmbeschichtet oder enteralbeschichtet werden, um beschichtete film- oder enteralbeschichtete Tabletten bereitzustellen.
- Zu 100 ml destilliertem Wasser zur Injektion wurde 1 g Carboxymethylcellulosenatrium gegeben, und die Mischung wurde gerührt. Zu der resultierenden Lösung wurden 40 mg Polysorbat 80 gegeben, und nach dem Mischen 1 g Tetrahydro-WS9326A darin dispergiert, um eine injizierbare Suspension bereitzustellen.
- Zu 50 ml Macrogol 400 wurde 1 g Tetrahydro-WS9326A gegeben, und die Mischung wurde gerührt. Die resultierende Lösung wurde mit destilliertem Wasser zur Injektion auf 100 ml aufgefüllt, um eine injizierbare Lösung bereitzustellen.
- Nachdem Tetrahydro-WS9326A in einer Lösung von Polysorbat 80 in destilliertem Wasser zur Injektion dispergiert worden war, wurde eine Lösung von Natriumchlorid, Benzylalkohol und Carboxymethylcellulosenatrium in destilliertem Wasser für die Injektion hinzugegeben. 5 ml der resultierten Suspension wurden in eine Ampulle gefüllt, die dann durch Schmelzen versiegelt wurde, um eine injizierbare Suspension, die die folgenden Bestandteile enthielt, bereitzustellen.
- Tetrahydro-WS9326A 10 mg
- Polysorbat 80 2 mg
- Carboxymethylcellulosenatrium 7,5 mg
- Benzylalkohol 9 mg
- Natriumchlorid 9 mg
- Destilliertes Wasser zur Injektion q.s. Referenz:
- Tetrahydro-WS9326A wurde erhalten durch Unterwerfen der Verbindung der obigen Strukturformel der Reduktion mit 10 %igem Pd-C.
Claims (4)
1. Verwendung eines Peptidderivates der allgemeinen Formel:
worin R¹ Wasserstoff, Phenyl(C&sub1;-C&sub6;)alkoxycarbonyl, (C&sub1;-C&sub6;)-
Alkanoyl, (C&sub1;&sub5;-C&sub2;&sub0;)Alkanoyl, Benzoyl,
Thienyl(C&sub1;-C&sub6;)alkanoyl, Phenyl(C&sub3;-C&sub6;)alkenyl, substituiert mit einer (C&sub2;-C&sub6;)-
Alkenylgruppe, oder Phenyl(C&sub1;-C&sub6;)alkanoyl, substituiert mit
einer (C&sub1;-C&sub6;)Alkylgruppe, ist;
R² Hydroxy ist und
R³ Carboxy oder (C&sub1;-C&sub6;)Alkoxycarbonyl ist, oder
R² und R³ miteinander verbunden sind, um eine Gruppe der
Formel:
-O-
-darzustellen;
R&sup4; Hydroxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder (C&sub1;-C&sub6;)Alkanoyloxy ist;
R&sup5; Hydroxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder (C&sub1;-C&sub6;)Alkanoyloxy ist;
R&sup6; Hydroxy, (C&sub1;-C&sub6;) )Alkoxy, Phenyl(C&sub1;-C&sub6;)alkoxy oder
(C&sub1;-C&sub6;)Alkanoyloxy ist; und
eine Einfachbindung oder eine Doppelbindung
darstellt,
oder eines pharmazeutisch verträglichen Salzes davon zur
Herstellung eines Medikamentes zur Prävention und/oder
Behandlung von Entzündungen.
2. Verwendung nach Anspruch 1, worin das Peptidderivat die
Verbindung (I), worin
R¹ 3-(2-Pentylphenyl)propanoyl ist,
R² und R³ miteinander verbunden sind, um eine Gruppe der
Formel: -O- - darzustellen,
R&sup4; Hydroxy ist,
R&sup5; Hydroxy ist,
R&sup6; Hydroxy ist und
eine Doppelbindung ist,
oder ein pharmazeutisch verträgliches Salz davon ist.
3. Verwendung nach Anspruch 2, worin das Peptidderivat eine
Verbindung der Formel ist:
4. Verwendung eines Peptidderivates nach Anspruch 1 oder eines
pharmazeutisch verträglichen Salzes davon für die
Herstellung eines Medikamentes zur Prävention und/oder
Behandlung von Arthritis.
