Description
- Die vorliegende Erfindung betrifft das Gebiet der biologisch aktiven Peptide. Diese Erfindung betrifft insbesondere biologisch aktive Peptide, welche die Aminosäure D-Tryptophan (D-Trp) enthalten.
- Es ist bekannt, daß durch Einbau oder Einführung eines D-Tryptophan-Restes in eine biologisch aktive Peptidkette die Aktivität dieser Kette erhöht wird. Außerdem wird durch einen solchen Einbau oder eine solche Einführung die biologische Aktivität verlängert. Die länger anhaltende und verbesserte Wirksamkeit solcher Peptide hängt wahrscheinlich zusammen mit der erhöhten Beständigkeit gegen Abbau durch Peptidasen.
- Beispiele für D-Tryptophan enthaltende Peptide sind die LHRH-Agonisten, wie sie von D.H. Coy et al. in "Journal of Medical Chemistry", Band 19, S. 423 (1976), von W. Koenig et al. in "Peptide Chemistry" (1987), von T. Shiba und S. Sakakibara (eds.), Osaka, in "Protein Research Foundation", Osaka (1988), S. 591, von B.J.A. Furr et al. in "Journal of Endocrinol. Invest.", Band 11, S. 535 (1988), beschrieben sind. Beispiele für D-Tryptophan enthaltende Somatostatin-Analoga, beispielsweise die Peptide Octreotid und Vapreotid, werden von R. Deghenghi in "Biomedicine and Pharmacotherapy", Band 42, S. 585 (1988), beschrieben. Weitere Beispiele für ein D-Tryptophan enthaltendes Peptid sind die synthetischen Antagonisten der Substanz P, wie sie von D. Regoli et al. in "European Journal of Pharmacology", Band 99, S. 193 (1984), beschrieben werden, und GHRP-6, beschrieben von C.Y. Bowers et al. in "Endocrinology, Band 114. S. 1537 (1984).
- Peptide, die Tryptophan enthalten, unterliegen dem Abbau nach der "Kynurenin-Reaktion". Bei dieser Reaktion baut das Enzym Tryptophan-Pyrrolase (d.h. Indolamin-2,3- dioxygenase) den Pyrrolring von Tryptophan ab. Kynurenin und andere Zerfallsprodukte werden durch diesen Abbau erzeugt. Einige der Zerfallsprodukte haben sich als toxisch erwiesen, wenn sie in erhöhten Konzentrationen vorhanden sind, wie von R.M. Silver et al. in "The New England Journal of Medicine", Band 322, S. 874 (1990), berichtet.
- D-Tryptophan enthaltende Peptide unterliegen einem Abbau durch Sauerstoff und andere reaktionsfähige Radikale, wie von R. Geiger und W. Koenig in "The Peptides", Academic Press, Band 3, S. 82, New York (1981), berichtet. Das D-Tryptophan in der Peptid-Kette kann mit aktiven oder aktivierten Gruppen reagieren, wenn Peptide formuliert werden zu bestimmten pharmazeutischen Zusammensetzungen mit kontrollierter Freigabe, beispielsweise solchen auf der Basis von Polymilchsäure-Polyglycolsäure-Polymeren. Es wird angenommen, daß dieser Abbau entweder durch Wärme oder durch Anwesenheit von Katalysatoren erleichtert wird. Es ist auch möglich, daß Radiolyse-Produkte, die während der ionisierenden Sterilisierung dieser pharmazeutischen Zusammensetzungen gebildet werden, den Abbau von D-Tryptophan erleichtern. Der Abbau von D-Tryptophan und der gleichzeitig auftretende Abbau der D-Tryptophan enthaltenden pharmazeutischen Verbindung sind eindeutig ein unerwünschter Effekt.
- Benötigt wird ein Derivat von D-Tryptophan, das die oben erläuterte, länger anhaltende biologische Aktivität beibehält, während es gleichzeitig beständig ist gegen Abbau durch Indolamin-Dioxygenase, Sauerstoff oder andere reaktionsfähige Radikale bzw. Reste. Es ist natürlich wesentlich, daß ein solches Derivat von D-Tryptophan seine biologische Aktivität beibehält,verglichen mit natürliches D-Tryptophan enthaltenden bioaktiven Peptiden.
- Die hier verwendeten Ausdrücke "biologischer Effekt" oder "pharmakologischer Effekt" beziehen sich auf den qualitativen Effekt, den ein bioaktives Peptid auf lebendes Gewebe hat. So hat beispielsweise LHRH, ein ein Luteinisierungshormon freisetzendes Hormon den biologischen Effekt, Zellen der Vorderlappen-Hypophyse zu veranlassen, das Lutenisierungshormon freizusetzen. Im Gegensatz dazu wird der Ausdruck "Potenz" bzw. "Stärke" in seinem konventionellen Sinne verwendet und bezieht sich auf den Grad und die Dauer der Bioaktivität eines gegebenen Peptids.
- Unter Anwendung dieser vorstehend definierten Ausdrücke wird benötigt ein Tryptophan enthaltendes bioaktives Peptid, das gegen oxidativen Abbau und gegen Angriff durch ein reaktionsfähiges Radikal (Rest) beständig ist bei gleichzeitiger Aufrechterhaltung der gleichen biologischen Aktivität und einer ähnlichen oder höheren Potenz bzw. Stärke als sie die derzeit verfügbaren analogen Peptide ergeben.
- Erfindungsgemäß wurde nun ein Derivat von D-Tryptophan gefunden, das einem biologisch aktiven Peptid, dem das Derivat einverleibt worden ist, eine verbesserte Beständigkeit gegen oxidative Abbaureaktionen des Tryptophan-Derivats verleiht unter gleichzeitiger Aufrechterhaltung der biologischen Aktivität und des pharmakologischen Effekts, wie sie Peptide aufweisen, denen unverändertes D- Tryptophan einverleibt worden ist. Die vorliegende Erfindung bezieht sich insbesondere auf biologisch aktive Peptide, denen mindestens ein D-Tryptophan einverleibt worden ist, bei dem eine niedere Alkylgruppe, vorzugsweise eine Alkylgruppe mit 1 bis 3 Kohlenstoffatomen, in die 2-Position substituiert (eingebaut) ist. Peptide, denen solche substituierten D-Tryptophane einverleibt worden sind, sind stabiler in Gegenwart von reaktionsfähigen Radikalen (Resten) oder wenn Pharmazeutika, die solche Peptide enthalten, ionisierender Strahlung ausgesetzt werden.
- Die erfindungsgemäßen biologisch aktiven Peptide sind durch die folgende Formel charakterisiert:
- A-D-2-ALK-TRP-B
- worin alk eine niedere Alkylgruppe, vorzugsweise eine solche mit 1 bis 3 Kohlenstoffatomen, bedeutet. A ist die SEQUENCE ID Nr. 1, worin der Glutaminsäurerest in der SEQUENCE-Position 1 Pyro-Glu oder D-Phe-Cys-Phe ist; die SEQUENCE ID Nr. 2, worin der Phenylalaninrest in der SEQUENCE-Position 1 D-Phe ist, der Prolin-Rest in der SEQUENCE-Position 4 D-Prolin ist und die Tryptophan-Reste in den SEQUENCE-Positionen 7 und 9 sind D-Tryptophan; Arg-D- Trp-N-methyl-Phe; oder His sind; und B ist die SEQUENCE ID Nr. 3, worin der Glycin-Rest in der SEQUENCE-Position 4 Gly-NH&sub2;; Leu-Arg-Pro-NHCH&sub2;CH&sub3;; oder Lys-Thr-Cys- NHCH(CH&sub2;OH)CHOH(CH&sub3;) ist; die SEQUENCE ID Nr. 4, worin der Tryptophan-Rest in der SEQUENCE-Position 4 Trp-NH&sub2;; Met- NH&sub2;; oder Leu-Met-NH&sub2; ist; die SEQUENCE ID Nr. 5, worin der Phenylalanin-Rest in der SEQUENCE-Position 3 D-Phe ist und der Lysin-Rest in der SEQUENCE-Position 4 Lys-NH&sub2; ist; mit der Maßgabe, daß dann, wenn A die SEQUENCE ID Nr. 1 darstellt, B die SEQUENCE ID Nr. 3 ist, worin der Glycin- Rest in der SEQUENCE-Position 4 Gly-NH&sub2; oder Leu-Arg-Pro- NHCH&sub2;CH&sub3; ist; daß dann, wenn A D-Phe-Cys-Phe darstellt, B Lys-Thr-Cys-NHCH(CH&sub2;OH)CHOH(CH&sub3;) ist, worin die Cystein- Einheiten durch ein cyclisches Disulfid miteinander verbunden sind; daß dann, wenn A D-Phe-Cys-Tyr darstellt, B die SEQUENCE ID Nr. 4 ist, worin die Cystein-Einheiten durch ein cyclisches Disulfid miteinander verbunden sind und der Tryptophan-Rest in der SEQUENCE-Position 4 Trp-NH&sub2; ist; daß dann, wenn A die SEQUENCE ID Nr. 6 darstellt, worin der Prolin-Rest in der SEQUENCE-Position 1 D-Pro ist und die Tryptophan-Reste in den SEQUENCE-Positionen 4 und 6 D-Trp sind, B Met-NH&sub2; ist; daß dann, wenn A Arg-D-Trp-n- methyl-Phe darstellt, B Leu-Met-NH&sub2; ist; oder daß dann, wenn A His darstellt, B die SEQUENCE ID Nr. 5 ist, worin der Phenylalanin-Rest in der SEQUENCE-Position 3 D-Phe ist und der Lysin-Rest in der SEQUENCE-Position 4 Lys-NH&sub2; ist.
- Die folgenden Verbindungen sind biologisch aktive erfindungsgemäße Peptide, die mindestens ein erfindungsgemäßes C-2-substituiertes D-Tryptophan enthalten.
- A. SEQUENCE ID Nr. 7, worin der Glutaminsäure-Rest in der SEQUENCE-Position 1 Pyro-Glu ist, der Tryptophan-Rest in der SEQUENCE-Position 6 D-2-methyl-Trp ist und der Glycin-Rest in der SEQUENCE-Position 10 Pro-Gly-NH&sub2; ist.
- B. SEQUENCE ID Nr. 8, worin der Gluaminsäure-Rest in der SEQUENCE-Position 1 Pyro-Glu ist, der Tryptophan-Rest in der SEQUENCE-Position 6 D-2-methyl-Trp ist und der Prolin- Rest in der SEQUENCE-Position 9 Pro-NHCH&sub2;CH&sub3; ist.
- C. SEQUENCE ID Nr. 9, worin der Prolin-Rest in der SEQUENCE-Position 1 D-Pro ist, der Tryptophan-Rest in der SEQUENCE-Position 4 D-Trp ist, der Tryptophan-Rest in der SEQUENCE-Position 6 D-Trp ist, der Tryptophan-Rest in der SEQUENCE-Position 7 D-2-ethyl-Trp ist und der Methionin- Rest in der SEQUENCE-Position 8 Met-NH&sub2; ist.

- Die Verbindungen A und B sind Analoga des natürlichen Peptids SEQUENCE ID Nr. 14, worin der Glutaminsäure-Rest in der SEQUENCE-Position 1 Pyro-Glu ist und der Glycin- Rest in der Position 10 Gly-NH&sub2; ist. SEQUENCE ID Nr. 14 ist ein das Luteinisierungs-Hormon freisetzendes Hormon (LH-RH), ein neurohumorales Hormon, das im Hypothalamus gebildet wird, das die Sekretion des LH-Lutenisierungs- Hormons durch die Vorderlappen-Hypophyse stimuliert. Die Verbindungen A und B gehören deshalb zur Klasse der LHRH- Agonisten und sind auch jeweils wie folgt definiert:
- [D-2-methyl-Trp&sup6;]LHRH und [Des-Gly¹&sup0;-D-2-methyl-Trp&sup6;-Proethylamid &sup9;]LHRH.
- Die Verbindungen C und D sind Antagonisten der Substanz P. Die Substanz P ist ein Neurotransmitter, der von sensorischen Neutronen verwendet wird, welche Schmerz-Antworten oder andere schädliche Stimuli an das Zentralnervensystem weiterleiten. Die Verbindungen C und D weisen deshalb analgetische und antiinflammatorische Effekte auf. Die Peptide E und F sind Analoga (Agonisten) von Somatostatin und weisen als solche eine antisekretorische und antitumorale Aktivität auf. Das Peptid G ist ein Analogon von GHRP (einem das Wachstumshormon freisetzenden Peptid), welches die Freisetzung des Wachstumshormons stimuliert.
- 2-Methyl-tryptophan ist bekannt (vgl. H.N. Rydon, "J. Chem. Soc." 1948, 705) und die homologen alkylierten Derivate werden zweckmäßig hergestellt aus den entsprechenden 2-Alkyl-indolen nach bekannten Verfahren (vgl. J.P. Li et al, "Synthesis" (1), 73, 1988). Die Auftrennung der racemischen Tryptophan-Derivate in die optischen Antipoden zur Bildung der erfindungsgemäßen D-Enantiomeren kann auch nach den verschiedensten Verfahren erzielt werden (vgl. "Amino Acids, Peptides and Proteins", Band 16, S. 18-20, The Royal Society of Chemistry, London, 1985). Zur Herstellung der erfindungsgemäßen Peptide können sowohl die Lösungsphasen- als auch die Festphasen-Methode der Peptidsynthese angewendet werden (vgl. R. Geiger et al., "The Peptides", Academic Press, New York, 1981). Wenn die Festphasen-Methode angewendet wird, werden vorzugsweise Peptid-Synthesizer, beispielsweise das Applied Biosystem 430A, Bioresearch Sam 9500 oder Beckman-Modell 990 bevorzugt verwendet.
- Bei dieser Herstellungsweise wird die erste Aminosäure an das Benzhydrylamin-Harz gebunden und die restlichen geschützten Aminosäuren werden dann stufenweise unter Anwendung der Standard-Verfahren, wie sie von den Herstellern der Synthesizer empfohlen werden, gekuppelt. So werden beispielsweise Aminosäure-Kupplungen durchgeführt durch Verwendung von symmetrischen Anhydriden in dem Applied Biosystems-Synthesizer und von Diisopropylcarbodiimid in den Bioresearch- oder Beckman-Vorrichtungen. Die Aminosäurederivate werden durch tert-Butoxy-carbonyl-Gruppen an der α-Aminofunktion während der Synthese geschützt. Die in der Aminosäure in der Seitenkette vorhandenen funktionellen Gruppen werden vorher geschützt, beispielsweise durch Acetyl(Ac), Benzoyl (Bz), t-Butyloxycarbonyl (Boc), Benzyloxymethyl (Bom), Benzyl (Bzl), Benzyloxycarbonyl (Z), Formyl (For), p-Nitrophenylester (ONp), Tosyl (Tos) und dgl. Die funktionellen Gruppen von Histidin werden beispielsweise geschützt durch Benzyloxymethyl (His(Bom)), Tosyl (His(Tos)), die funktionellen Gruppen von Tryptophan werden beispielsweise geschützt durch Formyl (Trp(For)), diejenigen von Serin werden beispielsweise geschützt durch Benzyl (Ser(Bzl)), diejenigen von Tyrosin werden beispielsweise geschützt durch 2-Br-benzyloxycarbonyl (Tyr(2- Br-Z)), diejenigen von Arginin werden beispielsweise geschützt durch Tosyl (Arg(Tos)), diejenigen von Leucin werden beispielsweise geschützt durch O-Benzyl-p-tosyl (Leu(O-Bzl-p-Tos)), diejenigen von Prolin werden beispielsweise geschützt durch O-Benzyl-HCl (Pro(O-Bzl-HCl)), diejenigen von Glycin werden beispielsweise geschützt durch O-Benzyl-HCl (Gly(O-Bzl-HCl)), diejenigen von Cystein werden beispielsweise geschützt durch 4-Methyl-benzyl (Cys(4-Me-Bzl)), diejenigen von Lysin werden beispielsweise geschützt durch Benzyloxycarbonyl (Lys(Z)), diejenigen von Threonin werden beispielsweise geschützt durch Benzyl-OH (Thr(Bzl-OH)), diejenigen von Valin werden beispielsweise geschützt durch O-Benzyl-tosyl (Val(O-Bzl- p-Tos)), diejenigen von Glutaminsäure werden beispielsweise geschützt durch O-Benzyl (Glu(O-Bzl)), diejenigen von Methionin werden beispielsweise geschützt durch P- Nitrophenylester (Me(Onp)) und diejenigen von Alanin werden beispielsweise geschützt durch O-Benzyl-HCl (Ala(O- Bzl-HCl))
- Die Boc-Schutzgruppen an der α-Aminosäurefunktion werden in jeder Stufe entfernt durch Behandlung mit 60 %iger Trifluoressigsäure ("TFA") in Dichlormethan. Die Abspaltung von Trp und Met enthaltenden Peptiden von dem Harz oder unter gleichzeitiger Entfernung aller Seitenketten-Schutzgruppen wird erzielt wie von J.P. Tam et al. in "J. Am. Chem. Soc.", Band 105, S. 6442 (1983), beschrieben. Die rohen Peptide werden nach der HF-Spaltung an einer Sephadex G-50 F-Säule in 50 %iger Essigsäure oder durch präparative Umkehrphasen-HPLC unter Verwendung von Gradienten von Acetonitril und Wasser, das 0,1 % Trifluoressigsäure enthält, gereinigt.
- Die folgenden Beispiele dienen lediglich Erläuterungszwecken, die vorliegende Erfindung ist darauf jedoch nicht beschränkt.
- SEQUENCE ID Nr. 7, worin der Glutaminsäure-Rest in der SEQUENCE-Position 1 Pyro-Glu ist, der Tryptophan-Rest in der SEQUENCE-Position 6 D-2-methyl-Trp ist, und der Glycin-Rest in der SEQUENCE-Position 10 Gly-NH&sub2; ist.
- Die Schutzgruppen für die Seitenketten sind Tosyl (Tos) für Arginin und Histidin und Bromobenzyloxycarbonyl (2-Br-Z) für Tyrosin. Das Benzhydrylamin-Harz (2,2 g) (Bachem ) wurde zu 1 % mit Prolin vernetzt und die verwendete Vorrichtung war eine Beckman-Vorrichtung Modell 990. Die durch Boc (tert-Butyloxycarbonyl) geschützten Aminosäuren werden mit Dicyclohexylcarbodiimid gekuppelt. Die Boc-Gruppen werden durch Trifluoressigsäure in Methylenchlorid entfernt.
- Die Synthese ergab 4,07 g des Decapeptid-Harzes (98 % theoretische Gewichtszunahme). Ein Teil dieses Harzes (1,5 g) wurde bei 0ºC 30 min lang mit HF (24 ml) und Anisol (8 ml) gerührt. Der HF wurde dann so schnell wie möglich (innerhalb etwa 60 min) im Vakuum entfernt und zu dem so erhaltenen Rückstand wurde EtOAc zugegeben. Das feste Material wurde abfiltriert, mit EtOAc gewaschen, getrocknet und mit 2 M AcOH extrahiert. Die Lyophilisierung ergab ein weißes Pulver, das durch Gelfitration an einer Sephadex G-25 (fein)-Säule (2,5 x 95 cm) durch Elution mit 2 M AcOH gereinigt wurde. Der Eluat-Anteil, der dem Haupt-Peak entsprach, wurde dann getrocknet und an einer Sephadex G-25 (fein)-Säule (2,5 x 95 cm), die vorher mit der unteren Phase und danach mit der oberen Phase des folgenden Zweiphasen-Lösungsmittelgemisches n-BuOH/AcOH-H&sub2;O (4:1:5) äquilibriert worden war, weiter eluiert. Die Elution mit der oberen Phase ergab einen Haupt-Peak und das Peptid aus diesem Bereich wurde gesammelt, zur Trockne eingeengt und mit verdünnter AcOH lyophilisiert, wobei man das Titel- Peptid in Form eines weißen Pulvers erhielt. Die Aminosäure-Analyse stimmte mit der gewünschten Struktur überein.
- SEQUENCE ID Nr. 8, worin der Glutaminsäure-Rest in der SEQUENCE-Position 1 Pyro-Glu ist, der Tryptophan-Rest in der SEQUENCE-Position 6 D-2-methyl-Trp ist, und der Prolin-Rest in der SEQUENCE-Position 9 Pro-NHCH&sub2;CH&sub3; ist.
- Das Peptid wurde zusammengebaut aus einem zu 1 % vernetzten Pro-Merrifield-Harz (2,0 g, 1,0 mmol Pro) unter Anwendung der gleichen Bedingungen und Schutzgruppen wie sie in Beispiel 1 verwendet worden waren, jedoch mit der Ausnahme, daß für die Imidazol-Gruppe von Histidin eine Dinitrophenol-Schutzgruppe verwendet wurde. Das erhaltene Peptid-Harz (3,45 g) wurde mit Ethylamin (20 ml, 0ºC) 6 h lang gerührt und das überschüssige Amin wurde im Vakuum entfernt. Das geschützte Peptid-Harz wurde mit MeOH extrahiert und durch Zugabe eines großen Überschusses an EtOAc ausgefällt, wobei man 1,36 g Material erhielt. Das erhaltene Produkt wurde mit HF-Anisol behandelt und von der Schutzgruppe befreit und das nach dieser Behandlung erhaltene rohe Peptid wurde durch Gelfiltration und anschließende Verteilungschromatographie gereinigt, wobei man das obengenannte homogene Peptid erhielt. Die Aminosäure-Analyse stimmte mit der gewünschten Struktur überein.
- Unter Anwendung der vorstehend beschriebenen Verfahren mit geeigneten Modifikationen, die dem Fachmann auf dem Gebiet allgemein bekannt sind, wurden die folgenden Peptide synthetisiert:
- SEQUENCE ID Nr. 9, worin der Prolin-Rest in der SEQUENCE- Position 1 D-Prolin ist, die Tryptophan-Reste in den SEQUENCE-Positionen 4 und 6 D-Trp sind, der Tryptophan-Rest in der SEQUENCE-Position 7 D-2-ethyl-Trp ist und der Methionin-Rest in der SEQUENCE-Position 8 Met-NH&sub2; ist.
- SEQUENCE ID Nr. 10, worin der Tryptophan-Rest in der SEQUENCE-Position 2 D-Trp ist, der Phenylalanin-Rest in der SEQUENCE-Position 3 N-methyl-Phe ist, der Tryptophan- Rest in der SEQUENCE-Position 4 D-2-isopropyl-Trp ist und der Methionin-Rest in der SEQUENCE-Position 6 Meth-NH&sub2; ist.
- SEQUENCE ID Nr. 11, worin der Phenylalanin-Rest in der SEQUENCE-Position 1 D-Phe ist, der Tryptophan-Rest in der SEQUENCE-Position 4 D-2-methyl-Trp ist und der Cystein-Rest in der SEQUENCE-Position 7 Cys- NHCH(CH&sub2;OH)CHOHCH&sub3; (cyclisches Disulfid) ist.
- SEQUENCE ID Nr. 12, worin der Phenylalanin-Rest in der SEQUENCE-Position 1 D-Phe ist, der Tryptophan-Rest in der SEQUENCE-Position 4 D-2-methyl-Trp ist und der Tryptophan-Rest in der SEQUENCE-Position 8 Trp-NH&sub2; (cyclisches Disulfid) ist.
- SEQUENCE ID Nr. 13, worin der Tryptophan-Rest in der SEQUENCE-Position 2 D-2-methyl-Trp ist, der Phenylalanin- Rest in der SEQUENCE-Position 5 D-Phe ist und der Lysin- Rest in der SEQUENCE-Position 6 Lys-NH&sub2; ist.
- Obgleich die obengenannten erfindungsgemäßen Beispiele spezifische Ausführungsformen derselben beschreiben, wird angenommen, daß die Substitution mit einem D-2- Alkyl-tryptophan in einem bioaktiven Peptid, das mindestens einen Tryptophan-Rest enthält, ebenfalls zu bioaktiven Peptiden führt, welche die obengenannten Vorteile und Vorzüge ergeben.
- Die Einarbeitung eines D-2-Alkyl-tryptophans in bioaktive Peptide, wie vorstehend beschrieben, stellt eine Methode zur Verlängerung und Beibehaltung der Aktivität dieser Peptide dar. Wenn analoge bioaktive Peptide, die nicht durch ein D-2-Alkyl-tryptophan substituiert sind, verschiedenen Behandlungsbedingungen und Substanzen ausgesetzt werden, kann die Aktivität dieser Peptide in nachteiliger Weise beeinflußt werden. Bei Sterilisierungsverfahren, wie sie in der pharmazeutischen Industrie angewendet werden, können bioaktive Verbindungen einer ionisierenden Strahlung ausgesetzt sein. Diese Strahlung kann bewirken, daß sich reaktionsfähige Radikale (Reste) bilden. Tryptophan, das Peptide enthält, sind besonders empfindlich gegenüber dem Angriff durch solche Radikale (Reste) und ein solcher Angriff kann das Peptid unwirksam oder möglicherweise toxisch machen. Außerdem können verschiedene Formulierungsverbindungen, wie z.B. Polymilchsäure- Polyglycolsäure-Polymere, aktive oder aktivierte Gruppen enthalten, die ebenfalls Tryptophan enthaltende bioaktive Peptide angreifen können. Die vorliegende Erfindung stellt ein Verfahren zum Schützen eines Tryptophan enthaltenden bioaktiven Peptids gegenüber diesen Gefahren bei der Herstellung dar, wobei gleichzeitig auch die Beständigkeit der Peptide gegen oxidativen Abbau nach Beendigung der Formulierung erhöht wird. Es wird angenommen, daß die Anwesenheit der Alkylgruppe in der Position Nr. 2 des Tryptophans die Stabilität des Pyrrol-Ringes erhöht, an dem ein Angriff durch reaktionsfähige Radikale (Reste) und aktive oder aktivierte Gruppen erfolgt.
- Obgleich die hier beschriebene Erfindung so berechnet ist, daß sie die obengenannten Ziele erfüllt, sei darauf hingewiesen, daß zahlreiche Ausführungsformen und Modifikationen vom Fachmann auf diesem Gebiet entworfen werden können.

Claims (13)
1. Bioaktive Peptide, die D-Tryptophan-Reste
enthalten, in denen mindestens ein D-Tryptophan-Rest durch D-2-
Alkyl-Tryptophan, das eine niedere Alkylgruppe als
Substituent in der Position Nr. 2 aufweist, ersetzt ist, wobei
das genannte D-2-Alkyl-tryptophan dem Peptid eine erhöhte
Beständigkeit gegen Oxidation verleiht unter
gleichzeitiger Aufrechterhaltung im wesentlichen der gleichen
pharmakologischen Wirkung wie analoge bioaktive Peptide, in
denen der mindestens eine Tryptophan-Rest nicht ersetzt
ist.
2. Bioaktive Peptide nach Anspruch 1, worin die
niedere Alkylgruppe 1 bis 3 Kohlenstoffatome enthält.
3. Bioaktive Peptide nach Anspruch 2, worin die

Rest D-Pro ist und die Tryptophan-Reste in den Positionen
4 und 6 D-Trp sind, bedeutet, B für Met-NH&sub2; steht;
dann, wenn A Arg-D-Trp-N-methyl-Phe bedeutet, B für Leu-
Met-NH&sub2; steht; und
dann, wenn A His bedeutet, B für
steht,
worin der Phenylalanin-Rest D-Phe und der Lysin-Rest Lys-
NH&sub2; sind;
wobei die genannten Peptide eine erhöhte Beständigkeit
gegen ionisierte Sterilisierung, gegen reaktionsfähige
Radikale (Reste) und Oxidation aufweisen bei gleichzeitiger
Aufrechterhaltung eines ähnlichen pharmakologischen
Effekts wie analoges D-Tryptophan enthaltende Peptide, die
nicht mindestens ein D-2-Alkyl-Tryptophan enthalten.
5. Bioaktive Peptide nach Anspruch 4, worin die
niedere Alkylgruppe 1 bis 3 Kohlenstoffatome enthält.
6. Bioaktive Peptide nach Anspruch 5, worin die
Alkylgruppe Methyl, Ethyl oder Isopropyl ist.
7. Bioaktive Peptide nach Anspruch 4, worin das
genannte Peptid ist
worin der Glutaminsäure-Rest in der Position 1
Pyro-Glu ist, der Tryptophan-Rest in der Position 6 D-2-
methyl-Trp ist und der Glycin-Rest in der Position 10 Gly-
NH&sub2; ist;
worin der
Glutaminsäure-Rest in der Position 1 Pyro-Glu ist, der
Tryptophan-Rest in der Position 6 D-2-methyl-Trp ist und
der Prolin-Rest in der Position 9 Pro-NHCH&sub2;CH&sub3; ist;
worin der Prolin-Rest
in der Position 1 D-Prolin ist, die Tryptophan-Reste in
den Positionen 4 und 6 D-Trp sind, der Tryptophan-Rest in
der Position 7 D-2-ethyl-Trp ist und der Methionin-Rest in
der Position 8 Met-NH&sub2; ist;
worin der Tryptophan-Rest in der Position 2 D-Trp ist, der
Phenylalanin-Rest in der Position 3 N-methyl-Phe ist, der
Tryptophan-Rest in der Position 4 D-2-isopropyl-Trp ist
und der Methionin-Rest in der Position 6 Met-NH&sub2; ist;
worin der Tryptophan-Rest in der
Position 2 D-2-methyl-Trp ist, der Phenylalanin-Rest in
der Position 5 D-Phe ist und der Lysin-Rest in der
Position 6 Lys-NH&sub2; ist.
8. Bioaktive Peptide nach Anspruch 4, worin das
Peptid ist
worin der
Phenylalanin-Rest in der Position 1 D-Phe ist, der
Tryptophan-Rest in der Position 4 D-2-methyl-Trp ist und der
Cystein-Rest in der Position 7 Cys-NHCH(CH&sub2;OH)CHOH(CH&sub3;) ist
und worin die Cystein-Einheiten durch eine cyclische
Disulfid-Bindung miteinander verbunden sind; oder
worin der Phenylalanin-Rest in der Position 1 D-Phe ist,
der Tryptophan-Rest in der Position 4 D-2-methyl-Trp ist
und der Tryptophan-Rest in der Position 8 Trp-NH&sub2; ist,
worin die Cystein-Einheiten mittels einer cyclischen
Disulfid-Bindung miteinander verbunden sind.
9. Bioaktive Peptide, die D-2-Alkyl-tryptophan-Reste
enthalten, worin die genannten Peptide sind
worin der
Glutaminsäure-Rest in der Position 1 Pyro-Glu ist, der
Tryptophan-Rest in der Position 6 D-2-methyl-Trp ist, und
der Glycin-Rest in der Position 10 Gly-NH&sub2; ist;
worin der
Glutaminsäure-Rest in der Position 1 Pyro-Glu ist, der Tryptophan-
Rest in der Position 6 D-2-methyl-Trp ist und der Prolin-
Rest in der Position 9 Pro-NHCH&sub2;CH&sub3; ist;
worin der Prolin-Rest in
der Position 1 D-Prolin ist, die Tryptophan-Reste in den
Positionen 4 und 6 D-Trp sind, der Tryptophan-Rest in der
Position 7 D-2-ethyl-Trp ist und der Methionin-Rest in der
Position 8 Met-NH&sub2; ist;
worin der
Tryptophan-Rest in der Position 2 D-Trp ist, der
Phenylalanin-Rest in der Position 3 N-methyl-Phe ist, der
Tryptophan-Rest in der Position 4 D-2-isopropyl-Trp ist
und der Methionin-Rest in der Position 6 Met-NH&sub2; ist;
worin der Phenylalanin-Rest
in der Position 1 D-Phe ist, der Tryptophan-Rest in der
Position 4 D-2-methyl-Trp ist und der Cystein-Rest in der
Position 7 Cys-NHCH(CH&sub2;OH)CHOH(CH&sub3;) ist und worin die
Cystein-Einheiten durch eine cyclische Disulfid-Bindung
miteinander verbunden sind;
Trp, worin der Phenylalanin-Rest in der Position 1 D-Phe
ist, der Tryptophan-Rest in der Position 4 D-2-methyl-Trp
ist und der Tryptophan-Rest in der Position 8 Trp-NH&sub2; ist,
worin die Cystein-Einheiten mittels einer cyclischen
Disulfid-Bindung miteinander verbunden sind; und
worin der Tryptophan-Rest in der
Position 2 D-2-methyl-Trp ist, der Phenylalanin-Rest in
der Position 5 D-Phe ist und der Lysin-Rest in der
Position 6 Lys-NH&sub2; ist.
10. Verfahren zur Verlängerung der pharmakologischen
Aktivität eines bioaktiven Peptids, das einen oder mehr D-
Tryptophan-Reste enthält, das umfaßt den Ersatz mindestens
eines D-Tryptophan-Restes des genannten Peptids durch
einen D-2-Alkyl-tryptophan-Rest, um so die Beständigkeit
der genannten Peptide gegen Abbau durch Sauerstoff,
reaktionsfähige Radikale (Reste) und andere
reaktionsfähige Gruppen zu erhöhen bei gleichzeitiger
Aufrechterhaltung eines ähnlichen biologischen Effekts wie analoge
bioaktive Peptide, in denen der Tryptophan-Rest nicht in
dieser Weise ersetzt ist.
11. Verfahren zur Aufrechterhaltung der
pharmakologischen Wirksamkeit eines bioaktiven Peptids, das einen oder
mehr Tryptophan-Reste enthält, das sterilisiert oder
aktiven oder aktivierten Gruppen während der Herstellung
ausgesetzt werden solle das umfaßt den Ersatz mindestens
eines
Tryptophan-Restes des genannten Peptids durch einen D-
2-Alkyl-tryptophan-Rest, um so die Beständigkeit des
genannten Peptids gegen die Wirkungen der Sterilisierung
oder der Einwirkung von aktiven oder aktivierten Gruppen
zu erhöhen wie bei analogen bioaktiven Peptiden, bei denen
der mindestens eine Tryptophan-Rest nicht in der genannten
Weise ersetzt ist.
12. Verfahren nach Anspruch 11, worin das bioaktive
Peptid durch Einwirkenlassen von ionisierender Strahlung
sterilisiert wird.
13. Verfahren nach Anspruch 11, worin das Peptid
aktiven oder aktivierten Gruppen ausgesetzt wird, die in
Substanzen mit kontrollierter Freisetzung auf der Basis
von Polymilchsäure-Polyglycolsäure-Polymeren vorhanden
sind.
