Description
- Diese Erfindung betrifft die Bereitstellung von antimikrobiellen Mundspülmitteln für die topische Anwendung von in Wasser löslichen antimikrobiellen Bestandteilen auf die inneren Oberflächen des Mundes eines Warmblüters, einschließlich des Menschen. Diese Erfindung betrifft insbesondere eine solche Zusammensetzung, die in Form einer relativ niederviskosen, fließfähigen Flüssigkeit aufgebracht werden kann und beim Kontakt mit der warmen Oberfläche eines solchen Wesens jedoch schnell zu einem relativ hochviskosen, im wesentlichen nicht-fließfähigen Gel wird.
- Das Problem der Bereitstellung einer solchen Zusammensetzung ist in der CA-A-1 072 413 und der US-A-4 188 373 angesprochen worden, wobei die CA-A-1 072 413 eine Zusammensetzung beschreibt, die mit geeigneten Mengen an Geliermitteln formuliert worden ist, wobei die Hauptkomponente ein Poloxamer (z.B. Pluronic F- 127) ist, das bei Raumtemperatur flüssig ist und beim Aufwärmen auf eine Temperatur oberhalb von 26ºC ein Gel bildet. Die Konzentration des Polymers ist so eingestellt, daß sich die gewünschte Sol-Gel-Übergangstemperatur ergibt. Eine Art der Aufbringung ist die Formulierung von nicht-toxischen Zusammensetzungen für therapeutische, prophylaktische oder kosmetische Behandlung von Menschen oder Tieren zur Aufbringung auf den Körper oder in Körperhöhlen. In der US-A-4 188 373 ist eine pharmazeutische Zusammensetzung zur medizinischen Behandlung von Schleimhaut- oder Augenkrankheiten beschrieben, die 10 bis 26 Gew. -% eines Polyoxyethylen/Polyoxypropylen-Blockcopolymers (z.B. Pluronic F-127) umfaßt, das eine Gelübergangstemperatur von 25 bis 40 ºC aufweist.
- Die vorliegende Erfindung umfaßt die Verwendung eines einzigen wasserlöslichen, nichtionischen Blockcopolymers aus Ethylenoxid und Propylenoxid mit der Formel HO(C&sub2;H&sub4;O)a (C&sub3;H&sub6;O)b (C&sub2;H&sub4;O)CH, wobei die Ethylenoxideinheiten 65 bis 75 Gew.-% des Copolymers umfassen und das Copolymer ein durchschnittliches Molekulargewicht von 11 000 bis 13 000 aufweist, zur Herstellung eines antimikrobiellen Mundspülmittels in einer Menge von 10 bis 17 Gew.-%, bezogen auf diese Zusammensetzung, wobei das Copolymer in einer solchen Menge geliefert wird, daß die Zusammensetzung bei Temperaturen unterhalb von 26,7 ºC (80 ºF) fließfähig ist und beim Kontakt mit der Oberfläche des Mundraumes ein Gel bildet.
- Vorzugsweise enthält die Zusammensetzung 12 bis 17 Gew.-% des Copolymers.
- Das Ethylenoxid/Propylenoxid-Blockcopolymer der erfindungsgemäßen Zusammensetzung ist ausgewählt aus einer Gruppe von wasserlöslichen Polyalkylenglykol-Blockcopolymeren, die generisch als Poloxamere bekannt sind.
- Das Copolymer weist vorzugsweise ein durchschnittliches Molekulargewicht von 12 000 bis 13 000 auf. Das Polaxamer der Wahl ist als Poloxamer 407 bezeichnet und weist ein durchschnittliches Molekulargewicht von etwa 12 500 auf. Die Poly(ethylenoxid)- blöcke machen im Durchschnitt 67 Mol aus, d.h. etwa 69 Gew.-%. Poloxamer 407, wie es von dem Hersteller beschrieben ist, ist nicht toxisch, zeigt in Rattenstudien eine LD&sub5;&sub0; von 15,4 g/kg und in Kaninchenstudien eine akute Dermaltoxizität von größer als 2 g/kg. Eine wäßrige Lösung von 20 Gew.-% Poloxamer 407 wurde durch den Hersteller als nicht-reizend in einer Draize-Kaninchenaugen-Reizungsuntersuchung befunden. Tabelle 1 gibt einige der physikalischen Eigenschaften dieses bevorzugten Poloxamers 407 an, wie es durch die BASF Wyandotte Corporation von Parsippany, New Jersey, unter dem Warenzeichen PLURONIC F-127 verkauft wird. - Poloxamer 407 ist nahezu geschmackslos und geruchslos und hat daher Verwendung bei der Solubilisierung von aromatischen Verbindungen in im Mundraum anwendbaren Hygieneprodukten gefunden wie in wäßrigen/alkoholischen Mundspülmitteln. Üblicherweise ist in diesen Zusammensetzungen die Konzentration recht niedrig (niedriger als 10 Gew.-% und im allgemeinen niedriger als 1 Gew.-%) und daher zeigen solche Zusammensetzungen nicht die hierin beschriebenen Selbstgelierungseigenschaften. Die Poloxamerverbindungen sind tatsächlich als bei der Bildung von Gelen brauchbar bekannt und in einer Broschüre des zuvor genannten BASF Unternehmens mit dem Titel "Pluronic und Tetronic Blockcopolymers Surfactants", Seiten 16 bis 17 (1987) in dieser Hinsicht beschrieben. In einer weiteren Veröffentlichung der BASF mit dem Titel "Technical Data of Pluronic Polyols" wird das empfohlene Verfahren zur Bildung von Gelen aus Pluronic F-127 Lösungen beschrieben. Es ist angegeben, daß sich mit Konzentrationen mit einem Minimum von 20 Gew.-% einer solchen Verbindung Gele bilden und gebildet werden können, indem das Pluronic F-127 bei 80 ºC in der Lösung gelöst wird und auf Raumtemperatur abgekühlt wird, um das Gel zu bilden. Alternativ kann das Pluronic F-127 in einer Wasserlösung bei 5 bis 10 ºC gelöst werden und dann auf Raumtemperatur gebracht werden, woraufhin es ein Ringgel bildet. Es ist klar, daß diese Lehren einen Fachmann nicht dazu führen, Pluronic F-127 zu verwenden, um die Aufgaben dieser Erfindung zu lösen, da solche Lehren für die Aufgaben dieser Erfindung vollständig schädlich sind; es ist gelehrt, daß das Ergebnis eine gelierte Lösung bei Raumtemperatur ist, d.h. eine, die nicht in Form einer fließbaren Flüssigkeit abgefüllt, gelagert oder abgegeben werden kann.
- Stattdessen ist nun gefunden worden, daß unter Verwendung von Poloxamer 407 in Konzentrationen, die vom Stand der Technik nicht gelehrt sind, nämlich größer als 10 % und weniger als 17 % und vorzugsweise 12 bis 17 %, eine wäßrige Einphasenlösung erzeugt wird, die bei Raumtemperatur flüssig und fließfähig ist und in nur ein paar Sekunden geliert, wenn sie auf etwa 80 ºF (26,7 ºC) erwärmt wird.
- Während der Mechanismus für das Phänomen der Gelierung bei der gewünschten Temperatur nicht vollständig klar ist, wird angenommen, daß ein solches Verhalten insgesamt von der Wahl eines spezifischen engen Bereichs von Definitionsparametern für das Polymer abhängt, z.B. dem Molekulargewicht und den Anteilen von Ethylenoxid- zu Propylenoxideinheiten zusammen mit der Wahl eines engen Bandes von Konzentrationen des Polymers in der Zusammensetzung.
- Zusätzlich zu antimikrobiell wirkenden Bestandteilen können die erfindungsgemäßen Mundspülmittel auch antibiotische Mittel, z.B. Triclosan, Chlorhexidin, Alexidin, Cetylpryridiniumchlorid oder Sanguinarin, sowie essentielle Öle, Fluoride, die Antikarieseigenschaften liefern, und Adstringentien wie Zinkverbindungen enthalten. Einige typische antimikrobielle Mittel sind mit größerer Besonderheit in der ebenfalls übertragenen und anhängigen Britischen Patentanmeldung 8801773, veröffentlicht als GB-A- 2 200 551A am 10. August 1988, angegeben. Solche wirksamen Bestandteile können auch Adstringenzsalze umfassen, die einen dünnen Schutzfilm auf der Oberfläche von Körperzellen bilden und daher die Empfindlichkeit der Zellen gegen über externen Reizungen verringern, die durch mechanische, thermische oder chemische Einwirkung verursacht werden können. Beispiele von Adstringens verbindungen, die in oral angewendeten Zusammensetzungen verwendet werden, umfassen Zinksalze wie Zinkchlorid und Zinkcitrat, die in Wasser löslich sind. Solche wirksamen Bestandteile können auch bestimmte Verbindungen zum Zweck der topischen Deodorierung umfassen wie beispielsweise Chlorophylline. Außerdem können solche wirksamen Bestandteile Fluoride als Antikariesmittel umfassen wie beispielsweise Natriumfluorid, Zinn(II)fluorid und Natriummonofluorophosphat.
- Zusätzlich zu solchen wirksamen Bestandteilen werden typischerweise weitere Komponenten in oral anwendbaren wäßrigen Zusammensetzungen verwendet, einschließlich solche zusätzlichen Komponenten wie Alkohol, Geschmacksstoff, Feuchthaltemittel und Tenside.
- Alkohole werden in denaturierter Form in Konzentrationen von 5 bis 30 % für die verschiedenen Zwecke der Erhöhung der Wirkung eines Geschmacksstoffs, Erhöhung der antimikrobiellen Effizienz oder zur Vereinfachung der Löslichkeit anderer Bestandteile verwendet. Geeignete Alkohole sind beispielsweise Ethanol oder Isopropanol.
- Geschmacksstoffe die in Mundspülmitteln verwendet werden, umfassen beispielsweise Bestandteile wie Eukalyptol, Menthol, Thymol, Methylsalicylat zusammen mit Geschmacksmodifizierungsmitteln. Außerdem sind bestiinmte minzartige Geschmacksstoffe und zimtartige Geschmacksstoffe verwendet worden, einschließlich beispielsweise Pfefferminz, Spearminz oder Nelken. Häufig werden auch Süßstoffe wie Saccharinverbindungen verwendet.
- Feuchthaltemittel werden hauptsächlich verwendet, um Kristallisation um Verschlüsse herum zu verhindern. Glycerinsorbit, Polyethylenglykol und Polypropylenglykol sind im allgemeinen die Feuchthaltemittel der Wahl.
- Außerdem sind, wie oben beschrieben, Tenside verwendet worden, hauptsächlich, bei der Solubilisierung von einigen der anderen Bestandteile zu helfen und eine Schaumwirkung, falls gewünscht, zu liefern. Zusätzlich zu den oben beschriebenen Poloxamerverbindungen können solche Tenside andere nichtionische Verbindungen wie Polyethylenfettsäureester und Sorbitanmonostearat sowie kationische Verbindungen wie Cetylpyridiniumchlorid oder anionische Verbindungen wie Natriumlaurylsulfat enthalten.
- Die folgenden Beispiele veranschaulichen die Natur der vorliegenden Erfindung weiter. Alle Mengen und Anteile beziehen sich auf das Gewicht.
- Es wird ein antimikrobielles Mundspülmittel hergestellt, das Chlorhexidindigluconat als den wirksamen antimikrobiellen Bestandteil umfaßt. Bestandteil Gewichtsprozent entionisiertes Wasser Ethanol (190º) Natrium-Saccharin Hibitane (20 % Chlorhexidindicgluconat) Pluronic F-127 Geschmacksstoff 89-180 FD&C Blau Nr. 1 (0,1 % Lsg.) FD&C Gelb Nr. 5 (0,1 % Lsg.)
- Diese Zusammensetzung ist bei Temperaturen unterhalb von 82 ºF (27,8 ºC) eine fließfähige Flüssigkeit und, wenn sie auf 82 ºF erhitzt wird, geliert sie in weniger als einer Minute. Die Probe geliert beim Kontakt mit der Mundoberfläche.
- Es wird ein antimikrobielles Mundspülmittel hergestellt, das Cetylpyridiniumchlorid als den wirksamen antimikrobiellen Bestandteil umfaßt. Bestandteil Gewichtsprozent entionisiertes Wasser Natrium-Saccharin Ethanol (190º) Cetylpyridiniumchlorid Pluronic F-127 Geschmacksstoff 89-180 FD&C Blau Nr. 1 (0,1 % Lsg.) FD&C Gelb Nr. 5 (0,1 % Lsg.)
- Wiederum ist diese Zusammensetzung unterhalb von 82 ºF eine fließfähige Flüssigkeit und geliert in weniger als einer Minute, wenn sie auf 82 ºF (27,6 ºC) erhitzt wird. Die Probe geliert beim Kontakt mit der Mundoberfläche. Dementsprechend beeinträchtigt die Verwendung von alternativen antimikrobiellen Mitteln nicht das gewünschte Gelierungsphänomen.
Claims (1)
1. Verwendung eines einzigen wasserlöslichen, nichtionischen
Blockcopolymers aus Ethylenoxid und Propylenoxid mit der
Formel HO(C&sub2;H&sub4;O)a(C&sub3;H&sub6;O)b(C&sub2;H&sub4;O)CH, wobei die
Ethylenoxideinheiten 65 bis 75 Gew.-% des Copolymers umfassen und das
Copolymer ein durchschnittliches Molekulargewicht von 11 000
bis 13 000 aufweist, bei der Herstellung eines
antimikrobiellen Mundspülmittels in einer Menge von 10 bis 17 Gew.-%,
bezogen auf das Mundspülmittel, wobei das Copolymer in einer
solchen Menge geliefert wird, daß die Zusammensetzung bei
Temperaturen unterhalb von 26,7 ºC (80 ºF) fließfähig ist
und bei Kontakt mit der Oberfläche des Mundraumes ein Gel
bildet.
