Description
- Die vorliegende Erfindung betrifft oberflächenmodifizierte Substrate und ein Verfahren für ihre Herstellung.
- Es ist bekannt, daß chemische Stoffe mit biologischer Aktivität auf die Oberfläche eines Substrats gebunden werden können, wenn funktionelle Gruppen auf der Substratoberfläche durch Oberflächenmodifikation zur Verfügung gestellt werden. Solche funktionellen Gruppen auf der Substratoberfläche können zur ionischen Interaktion geladen oder mit funktionellen Gruppen des chemischen Stoffes kovalent umgesetzt werden. Die Herstellung der Trägermatrix für immobilisierte Enzyme durch Adsorption eines Polyamins auf einem Substratmaterial und Kontaktieren des Polyamins mi einem bifunktionellen Reagenz zur Vernetzung und zur Stabilisierung des Polyamins, wobei eine Oberfläche mit funktionellen Gruppen zur Verfügung gestellt wird, mit denen Enzyme kovalent verbunden werden können, wurde in US PS 4,268,423 (Rohrbach et al.) beschrieben. Die in diesem Fall verwendeten Vernetzungsmittel sind Dialdehyde und Diisocyanate.
- In der US PS 4,229,838 (Mano et al.) wird eine Gefäßprothese mit antithrombogenen Eigenschaften hergestellt durch Adsorption eines bestimmten Polyamins, Polyethylenimin, auf der porösen Oberfläche, Vernetzung mit einem bifunktionellen Mittel wie Dialdehyd und Quaternisierung der verbleibenden Aminogruppen, um eine positiv geladene Oberfläche zu erhalten, auf der negativ geladenes Heparin adsorbiert werden kann.
- US PS 4,565,740 (Gölander und Larsson) und EP-A-0086- 187 beschreiben oberflächenmodifizierte Substrate, hergestelle durch Adsorption eines Komplexes eines polymeren kationischen oberflächenaktiven Mittels mit primären wie auch sekundären und/oder terciaren Aminostickstoff, vorzugsweise Polyethylenimin oder Polyamide, und eines Dialdehyds mit 1 - 4 Kohlenstoffatomen zwischen den Aldehydgruppen auf der Oberfläche und ggf. Adsorption verschiedener zusätzlicher Schichten polymerer anionischer und oberflächenaktiver Mittel mittels Zwischenschichten des Komplexes. Die derart modifizierte Oberfläche kann ene biologisch aktive Verbindung durch ionische oder kovalente Bindung adsorbieren.
- Im allgemeinen isn erwünscht, daß eine Oberflächenmodifikation zu hoher Dichte reaktiver funktioneller Gruppen führt. Darüber hinaus sollte die Reaktion reproduzierbar und unter normalen Reaktionsbedingungen leicht durchführbar sein, wobei die modifizierte Oberfläche in dem Sinne stabil sein soll, daß keine Ablösung von den Schichten der Oberflächenmodifikation erfolgen sollte. Bisher war es schwierig, oberflächenmodifizierte Substrate nach dem Verfahren der zuletzt erwähnten Vorveröffentlichung zur Verfügung zu stellen, die allen diesen Anforderungen in befriedigendem Umfang genügen.
- Es wurde nunmehr gefunden, daß eine überraschend bessere Oberflächenmodifikation durchgeführt werden kann, indem auf die Substratoberfläche ene Schicht eines Polyamins adsorbiert wird, das ein hohes mittleres Molekulargewicht aufweist.
- Das Polyamin wird durch Vernetzung mit Crotonaldehyd stabilisert, der ein Monoaldehyd mit einer C-C Doppelbindung in Konjugation mit der Aldehydfunktion ist. Dann wird eine Schicht eines anionischen Polysaccharids auf der Oberfläche des vernetzten Polyamins adsorbiert, bei Bedarf gefolgt von einer oder mehreren weiteren alternierenden Schichten vernetzten Polyamins und anionischen Polysaccharids. Schließlich wir in der anionischen Polysaccharidschicht oder auf der äußersten anionischen Polysaccharidschicht eine Schicht eines nicht vernetzten Polyamins mit freien Primären Aminogruppen adsorbiert.
- Die vorliegende Erfindung stellt ebenfalls eine oberflächenmodifiziertes Substrat zur Verfügung, das
- (i) eine Schicht elnes Polyamins hohen mittleren Molekulargewichts, das mit Crotonaldehyd vernetzt ist, adsorbiert auf der Oberfläche des Substrats;
- (ii) eine Schicht eines anionischen Polysaccharids, adsorbiert auf der Oberfläche des vernetzten Polyamins;
- (iii) bei Bedarf eine oder mehrere zusätzliche sich abwechselnde Schichten von dem genannten vernetzten Polyamin (i) und dem anionischen Polysaccharid (ii); und
- (iv) eine Schicht eines nicht vernetzten Polyamins, das freie primäre Aminogruppen zur Verfügung stellt, adsorbiert auf der oder auf der äußersten Schicht anionischen Polysaccharids, aufweist.
- Das erfindungsgemäße Verfahren kann geeigneterweise durchgeführt werden, indem das Substrat in Kontakt mit einer wässrigen Lösung des Polyamins bei einem pH-Wert von 8 - 10, insbesondere pH 9 in Kontakt gebracht wird. Die Konzentration der Ausgangspolyaminlösung liegt im Bereich von 1 - 10 Gew.%, insbesondere 5 Gew.%, von der 1 ml auf ein Endvolumen von 500 - 2000 ml, insbesondere 1000 ml verdünnt werden kann. Diese fertige Lösung kann ebenfalls von 100 - 1000 ul, insbesondere 340 ul Crotonaldehyd enthalten. Alternativ hierzu wird das Substrat zunächst mit einer Lösung von Polyamin der genannten Konzentration und des genannten pH- Werts und dann mit einer Lösung des Crotonaldehyds in genannte Konzentration und bei genanntem pH-Wert behandelt. Die Temperatur ist nicht entscheidend; bevorzug wird die Behandlung jedoch bei Zimmertemperatur durchgeführt.
- Nach Spülen mit Wasser wird das Substrat mit einer Lösung eines anionischen Polysaccharids behandelt, die von 10 mg - 500 mg, vorzugsweise 100 mg des Polysaccharids in einem Volumen von 1000 ml enthält. Diese Stufe wird bei einer Temperatur im Bereich von 40 - 70º C, vorzugsweise 55º C und einem pH-Wert von 1 - 5, vorzugsweise 3 durchgeführt.
- Nach einer weiteren Spülung mit Wasser kann diese erste Stufe ein oder mehrere Male wiederholt werden; wenn schließlich eine Schicht Polysaccharid adsorbiert worden ist, wird das Substrat min einer Polyaminlösung einer Konzentration von 1 - 20 mal, vorzugsweise 10 mal der zuvor erwähnten bei der genannten Temperatur und dem genannten pH- Wert behandel.
- Das Polyamin ist vorzugsweise ein polymeres aliphatisches Amin, insbesondere Polyethylenimin mit hohem mittleren Molekulargewicht, wobei jedes Polyamin mit hohem mittleren Molekulargewicht und freien primären Aminogruppen verwendbar ist.
- Das anionische Polysaccharid ist vorzugsweise ein sulfatiertes Polysaccharid, insbesondere Dextransulfat.
- Die aminierte Oberfläche kann zusätzlich weiter durch Reduktion mit einem geeigneten Reduktionsmittel wie Natriumcyanborhydrid stabilisiert werden.
- Die erfindungsgemäße modifizierte Oberfläche besitzt freie primäre Amingruppen, durch die chemische Stoffe entweder ionisch oder kovalent gebunden werden können.
- Insbesondere aldehydhaltige chemische Stoffe können durch Bildung Schiff'scher Basen gebunden werden, ggf. gefolgt durch eine Stabilisierungsreaktion wie einer Reduktion zur Umwandlung der Schiff'schen Basen in sekundäre Amine.
- Erfindungsgemäß können folgende Vorteile erreicht werden:
- - Höhere Dichte der reakniven Aminofunktionen, mit dem Ergebnis höherer biologischer Aktivität bei Anbindung von biologisch aktivem Material;
- - deutlich niedrigere Konzentration von Vernetzungsmittel im Vergleich zum Stand der Technik;
- - hohe Stabilität der fertigen Oberfläche.
- Die erhöhte biologische Aktivität ergibt sich aus den folgenden Beispielen, die die Dichte der Bindungszentren als Grad der Heparinisierung zeigt, die erzielt wird, nachdem die Oberflächenmodifikation gemäß Erfindung im Vergleich zum Beispiel aus der US PS 4,565,740 (Gölander und Larsson) sowie EP-A-0 086 187 durchgeführt wurde:
- a) Vergleich der Ergebnisse, die erhalten werden, wenn erfindungsgemäß mit niederen Konzentrationen von Vernetzungsmittel gearbeitet wird; und
- b) Vergleich der Ergebnisse, die mit diesen beiden Methoden erhalten werden, wenn mit höheren Konzentrationen an Vernetzungsmittel gemäß Stand der Technik gearbeitet wird:
- In den Beispielen werden Polyethylen(PE)rohre zunächst mit Kaliumpermanganat in konzentrierter Schwefelsäure geätzt, worauf die Rohre mit folgenden Lösungen kontaktiert werden:
- 1a) 1 ml 5 % Polymin SN*, 340 ul Crotonaldehyd in 1 l 0,17 M Boratpuffer mit dem pH-Wert 9
- 2a) 1 ml 5 % Polymin SN* und 1,54 ml einer Lösung mit Gehalt an 25 Gew.% Glutaraldehyd in 1 l 0,17 M Boratpuffer mit dem pH-Wert 9
- 3) 0,1 g Dextransulfat* in 1 l einer 0,15 M Natriumchloridlösung, pH-Wert 3 bei 55º C
- 4) 10 ml 5 % Polymin SN* in 1 l Wasser, pH-Wert 9
- 5) 0,25 Heparin, abgebaut mit Salpetrigsäure (gemäß US PS 4,613,665, (Larm)) und 0,025 g Natriumcyanborhydrid in 1 l 0,15 M Natriumchloridlösung vom pH-Wert 3,9 bei 55º C
- 6) 0,17 M Boratpuffer mit pH-Wert 9.
- Polymin SN* ist ein Polyethylenimin von BASF, wobei Polymin ein eingetragenes Warenzeichen ist. Das verwendete Dextransulfat* ist erhältlich voh Pharmacia.
- Die PE-Rohre wurden mit obigen Lösungen in der folgenden Reihenfolge behandeln und zwischen jeder Behandlungsstufe mit Wasser gespült:
- Im Test gemäß Erfindung:
- 1a, 3, 1a, 3, 4, 5 und 6.
- Im Test gemäß Stand der Technik:
- 2a, 3, 2a, 3, 4, 5 und 6.
- Die Rohre wurden dann mit einer Lösung von Albumin über 24 Stunden gespült; dann wurden die folgenden Ergebnisse hinsichtlich der Heparinaktivität erzielt: Aufnahme von Antithrombin, gemessen als Thrombininhibition IU/cm² Vernetzungsmittel vor Spülung nach Albuminspülung Crotonaldehyd Glutaraldehyd
- Dies zeigt deutlich, daß eine dramatisch erhöhte Heparinaktivität erzielt wird, wenn das erfindungsgemäße Verfahren mit dem Vernetzungsmittel gemäß Erfindung durchgeführt wird, verglichen mit Verwendung eines Vernetzungsmittels gemäß Stand der Technik, obgleich Crotonaldehyd bei einer verhältnismäßig niederen Konzentration verwendet wurde.
- Die Rohre wurden wie in Beispiel 1 mit dem Unterschied behandelt, daß die Lösungen 1a und 2a ersetzt wurden durch 1b bzw 2b wie folgt:
- 1b) 1 ml 5 % Polyimin SN* und 4,12 ml Crotonaldehyd in 1 l 0,17 M Boratpuffer vom pH-Wert 9
- 2b) 1 ml 5 % Polyimin SN* und 20 ml Glutaraldehyd (25 %ige Lösung) in 1 l 0,17 M Boranpuffer vom pH-Wert 9
- Dieses ergab die folgenden Ergebnisse bezüglich Heparinaktivität: Aufnahme von Antithrombin, gemessen als Thrombininhibition IU/cm² Vernetzungsmittel vor Spülung nach Albuminspülung Crotonaldehyd Glutaraldehyd
- Diese Ergebnisse zeigen überraschend bessere Aktivität, wenn das erfindungsgemäße Vernetzungsmittel bei Konzentrationen gemäß Verfahren nach dem Stand der Technik verwendet wird.
- Darüber hinaus erkennt man, wenn Beispiel 1 mit Beispiel 2 verglichen wird, daß die Kombination des spezifischen Vernetzungsmittels gemäß Erfindung, nämlich Crotonaldehyd, und die zehnfach niedrigere Konzentration gemäß vorliegender Erfindung zu einer überraschend höheren Aktivität führt, wenn eine biologisch aktive Substanz auf die erfindungsgemäß modifizierte Oberfläche aufgebracht wird.
- Auf die erfindungsgemäß behandelte Oberfläche können chemische Stoffe mit funktionellen Gruppen, die mit Aminogruppen reagieren können, ohne Verlust biologischer Aktivität gebunden werden. EIN Beispiel für solche Stoffe sind Heparinfragmente mit Aldehydgruppen, die hergestellt werden durch Abbau von Heparin mit salpetiger Säure gemäß US PS 4,613,605 (Larm), um heparinisierte Oberflächen herzustellen.
- Natürlich können auch andere chemische Stoffe als Heparin mit anderen biologischen Aktivitäten und mit Gruppen, die mit Aminogruppen reagieren können, oder die aktiviert wurden, um mit Aminogruppen zu reagieren, durch die erfindungsgemäße Oberflächenmodifizierung auf die Oberfläche gebunden werden.
- Bei Bedarf können aie freien primären Aminogruppen auf der Oberfläche, die durch die Endschicht Polyamin zur Verfügung gestellt werden, quaternisiert werden, so daß eine negativ geladene, biologisch aktive Verbindung ionisch gebunden werden kann.
- Wie im Stand der Technik gemäß US PS 4,565,740 beschrieben ist, kann das Substrat aus beliebigem Material wie Metall, insbesondere rostfreiem Stahl, Aluminium, Glas oder einem polymeren Material, insbesondere Polyvinylchlorid, Polyurethan, Silikongummi, Polyetrafluorethylen, Polystyrol und/oder Polyolefinen, die üblicherweise für die Herstellung medizinischer Artikel, insbesondere für Blutkontakt, verwendet werden, bestehen. Zu Beispielen solcher Artikel zählen Rohre, Schläuche, Katheter, Oxygeneratoren, Filter, intravaskulare Sonden, Blutpumpen und ähnliche.
Claims (15)
1. Verfahren zum Modifizieren der Oberfläche eines
festen Substrats mit den Verfahrensstufen:
(a) Adsorbieren eines Polyamins hohen mittleren
Molekulargewichts und Vernetzen des Polyamins mit Crotonaldehyd auf der
Oberfläche des Substrats, wobei der Crotonaldehyd entweder
gleichzeitig mit oder getrennt von dem Polyamin zugesetzt
wird;
(b) nachfolgendes Adsorbieren einer Schicht eines
anionischen Polysaccharids auf der Oberfläche des vernetzten
Polyamins;
(c) bedarfsweise Wiederholung der Stufen (a) und (b)
ein- oder mehrmalig; und
(d) Adsorbieren einer Schicht nicht vernetzten Polyamins,
das freie primäre Aminogruppen zur Verfügung stellt, auf der
anionischen Polysaccharidschicht oder auf der äußersten
anionischen Polysaccharidschicht.
2. Verfahren nach Anspruch 1, mit der weiteren Stufe
der Quaternisierung der freien primären Amingruppen.
3. Verfahren nach Anspruch 1, wobei das Substrat
Metall, Glas und/oder ein polymeres Material ist.
4. Verfahren nach Anspruch 3, wobei das Substrat
rostfreier Stahl oder Aluminium ist.
5. Verfahren nach Anspruch 3, wobei das Substrat
Polyvinylchlorid, Polyurethan, Silikongummi,
Polytetrafluorethylen, Polystyrol und/oder Polyolefin ist.
6. Verfahren nach einem der vorhergehenden Ansprüche,
wobei das Polysaccharid Dextransulfat ist.
7. Verfahren nach einem der vorhergehenden Ansprüche,
wobei das Polyamin Polyethylenimin ist.
8. Verfahren nach Anspruch 1 oder nach einem der
Ansprüche 3 bis 7 unser Rückbeziehung auf Anspruch 1 mit dem
weiteren Schrift der Verbindung einer biologisch aktiven
chemischen Einheit mit funktionellen Gruppen, die mit den
primären Aminogruppen reaktiv sind, mit der nicht vernetzten
Polyaminschicht.
9. Verfahren nach Anspruch 8, wobei die chemische
Einheit Heparin ist.
10. Verfahren nach Anspruch 2 mit der weiteren Stufe
der ionischen Bindung einer negativ geladenen biologisch
aktiven Verbindung mit der nicht vernetzten, quaternisierten
Polyaminschicht.
11. Oberflächenmodifiziertes Substrat mit
(i) einer Schicht eines Polyamins hohen mittleren
Molekülargewichts, das mit auf der Oberfläche des Substrats
absorbiertem Crotonaldehyd vernetzt wurde;
(ii) einer Schicht eines auf der Oberfläche des vernetzten
Polyamins absorbierten anionischen Polysaccharids;
(iii) bedarfsweise zwei oder mehr zusätzlichen wechselnden
Schichten des vernetzten Polyamins (i) und des anionischen
Polysaccharids (ii); und
(iv) einer Schicht nicht vernetzten Polyamins, das freie
primäre Aminogruppen zur Verfügung stellt, die auf der oder
auf der äußersten Schicht anionischem Polysaccharids
absorbiert ist.
12. Substrat nach Anspruch 11, das des weiteren
(v) eine biologisch aktive chemische Einheit enthält, die an
das nicht vernetzte Polyamin kovalent gebunden ist.
13. Substrat nach Anspruch 12, wobei die biologisch
aktive chemische Einheit Heparin ist.
14. Substrat nach Anspruch 11, wobei die Schicht (iv)
eine Schicht eines nicht vernetzten Polyamins mit freien
Aminogruppen ist, die quaternisiert wurden.
15. Substrat nach Anspruch 14, das zusätzlich
(v) eine biologisch aktive Verbindung aufweist, die ionisch
mit der nicht vernetzten quaternisierten Polyaminschicht
verbunden ist.
