Description
- Die vorliegende Erfindung betrifft ein neues Histamin-Derivat, ein Verfahren zu dessen Herstellung und seine therapeutische Anwendung.
- EP-A-0 214 058 beschreibt eine pharmazeutische Zusammensetzung, die als wirksames Prinzip ein Histamin-Derivat der Formel
- enthält, worin R&sub1;, R&sub2; und R&sub4; jeweils ein Wasserstoff-Atom oder eine Methyl-Gruppe bezeichnen, oder R&sub1; und R&sub2; zusammengenommen eine Methylen-Gruppe bilden und R&sub3; ein Wasserstoff- Atom oder eine Methyl- oder Carboxy-Gruppe bedeutet, mit der Maßgabe, daß R&sub1;, R&sub2;, R&sub3; und R&sub4; nicht gleichzeitig Wasserstoff bezeichnen, sowie ein pharmazeutisch verträgliches Vehikel oder eine pharmazeutisch verträgliche Trägersubstanz.
- Dieses Dokument betrifft insbesondere Verbindungen, die an sich bereits bekannt sind, doch wurden bei diesen agonistische Eigenschaften gegenüber den H&sub3;-Rezeptoren entdeckt, die die Freisetzung und Synthese von Histamin regulieren. Unerwarteterweise wurde nun gefunden, daß α,β-Dimethylhistamin, eine von der allgemeinen Formel I umfaßte, bislang aber nicht beschriebene Verbindung eine agonistische Wirkung auf die H&sub3;-Rezeptoren besitzt, die selektiver und derjenigen anderer Verbindungen mit verwandten Strukturen überlegen ist.
- Die Erfindung betrifft somit α,β-Dimethylhistamin der Formel
- in Form des Racemats, des optischen Isomers oder der Diastereomerenmischung, sowie dessen pharmazeutisch annehmbare Säureadditionssalze.
- Die Verbindung, die in ihrer Seitenkette zwei asymmetrische Kohlenstoff-Atome aufweist, existiert in Form von vier optischen Isomeren.
- Ein Verfahren zur Herstellung der erfindungsgemäßen Verbindung ist dadurch gekennzeichnet, daß man ein Alkyl(mit C&sub1;-C&sub6;)-2-amino-3-(1H-imidazol-4-yl)butyrat mit Hilfe eines Metallhydrids zu 2-Amino-3-(1H-imidazol-4-yl)butanol reduziert, dieses Butanol-Derivat mit Hilfe eines Chlorierungsmittels in 2-Amino-1-chlor-3-(1H-imidazol-4-yl)butan umwandelt und man dieses chlorierte Derivat katalytisch reduziert.
- Das Alkyl- (insbesondere Methyl-) 2-Amino-3-(1H-imidazol- 4-yl)butyrat, von dem bei diesem Verfahren ausgegangen wird, läßt sich beispielsweise ausgehend von 1-Triphenylmethyl-1H- imidazol-4-ylcarbaldehyd nach J.L. Kelley, C.A. Miller und Ed.W. McLean, J.Med.Chem. 20, 721 (1977), herstellen, und die letztere Verbindung ist erhältlich ausgehend von 1H-Imidazol-4-ylmethanol nach P. Dziuron und W. Schunack, Arch.Pharm. (Weinheim, D.) 306, 347 (1973) und M. Barnabé und A. Burger, J.Med.Chem. 14, 883 (1971).
- Die Erfindung betrifft auch ein Verfahren zur stereospezifischen Herstellung der Verbindung, dadurch gekennzeichnet, daß man ein Alkyl(mit C&sub1;-C&sub6;)-(1H-imidazol-4-yl)carboxylat, dessen Imidazol-Rest geschützt ist (insbesondere durch eine Triphenylmethyl-Gruppe), in erythro-α,β-Dimethylhistamin umwandelt und man die Enantiomeren (+) und (-) dieser Verbindung mittels einer optisch aktiven Säure trennt.
- Das folgende Beispiel veranschaulicht die Synthese der erfindungsgemäßen Verbindung in Form des Racemats, insbesondere ausgehend von Stufe i).
- 52,06 g (0,5 mol) Formamidinacetat und 45,04 g (0,5 mol) 1,3-Dihydroxypropan-2-on werden nach und nach in 250 ml flüssigen Ammoniak gegeben. Diese Mischung wird in einen Stahlautoklaven gegeben und 12 h lang bei 40ºC, 10 bar (1 MPa) reagierengelassen. Anschließend wird der Ammoniak durch Verdampfen von der Reaktionsmischung abgetrennt. Der resultierende gelbe Sirup wird in 500 ml 2-Propanol gelöst und mit trockenem HCl-Gas auf pH 2 gebracht. Der gebildete voluminöse Niederschlag wird filtriert und mit 500 ml sehr heißem 2-Propanol in drei Portionen extrahiert. Zusatz von 200 ml Diethylether zu den vereinigten Filtraten und Halten bei -25ºC ergibt 57,1 g (Ausb.: 84,9%) an 1H-Imidazol-4-yl- methanol-chlorhydrat in Form hellbrauner Kristalle.

- Zu einer gerührten Lösung von 18,8 g (140 mmol) 1H-Imidazol- 4-ylmethanol in 140 ml Dimethylformamid und 50 ml Triethylamin werden nach und nach 40,5 g (145 mmol) Triphenylmethylchlorid in 300 ml Dimethylformamid gegeben. Nach 2 h Rühren bei Umgebungstemperatur wird die Reaktionsmischung auf 2 kg zerstoßenes Eis gegossen. Der Niederschlag wird mit Wasser gewaschen, und die Verunreinigungen werden durch Digerieren mit 400 ml heißem Dioxan entfernt. Nach Aufarbeiten, Waschen mit Diethylether und Trocknen unter Vakuum werden 41 g (Ausb.: 86%) 1-Triphenylmethyl-1H-imidazol-4-ylmethanol in Form eines weißen Pulvers erhalten.

- 68,1 g (0,2 mol) 1-Triphenylmethyl-1H-imidazol-4-ylmethanol und 177,9 g (2 mol) Mangandioxid werden 3 h lang in 1,3 l Dioxan am Rückfluß erhitzt. Die heiße Suspension wird über 100 ml Celite auf einem Büchner-Trichter filtriert und mit 0,5 l heißem Dioxan in drei Portionen gewaschen. Die Lösung wird unter Vakuum eingedampft, und der feste weiße Rückstand wird unter Vakuum getrocknet. Es werden 49,1 g (72,5%) Produkt erhalten.

- Zu einer frisch bereiteten Lösung von 290 mmol Methylmagnesiumiodid in 700 ml Diethylether werden 49,1 g (145 mmol) 1-Triphenylmethyl-1H-imidazol-4-ylcarbaldehyd in 200 ml Tetrahydrofuran gegeben. Die Mischung wird 2 h lang bei Umgebungstemperatur reagieren gelassen, dann wird eine Lösung von 19,4 g (0,36 mol) Ammoniumchlorid in 70 ml Wasser zugesetzt. Nach Rühren über Nacht wird der Niederschlag durch Filtration abgetrennt, und es wird mit 300 ml Tetrahydrofuran gewaschen. Die vereinigten Filtrate werden mit 300 ml Diethylether verdünnt und mit Waser gewaschen. Die organische Phase wird über Natriumsulfat getrocknet. Nach Verdampfen erhält man 46,5 g (Ausb.: 90,4%) 1-(1-Triphenylmethyl-1H-imidazol-4-yl)ethanol in Form eines weißen Pulvers. Schmp.: 156-157ºC.

- 46,1 g (130 mmol) 1-(1-Triphenylmethyl-1H-imidazol-4-yl)ethanol werden in 500 ml 2N Salzsäure bei 70ºC im Laufe einer Stunde gelöst. Der Niederschlag wird durch Filtration abgetrennt. Die Lösung wird mit 300 ml Diethylether in zwei Portionen gewaschen und unter Vakuum eingedampft, um das Chlorhydrat von 1-(1H-Imidazol-4-yl)ethanol in Form eines leicht gelben Öls zu erhalten. Diesem Öl werden bei 0ºC nach und nach 100 ml Thionylchlorid zugesetzt, und man läßt die Lösung eine Nacht lang bei Umgebungstemperatur reagieren. Verdampfen und Kristallisieren aus Diethylether/Ethylenglycoldimethylether ergeben 19,1 g (88%) 1-Chlor-1-(1H-imidazol-4-yl)ethan-chlorhydrat. Gelbe Kristalle, Schmp.: 101-104 ºC.

- 11,9 g (55 mmol) Acetamidomalonsäurediethylester werden einer Lösung von 105 mmol Natriumethylat in 100 ml Ethanol zugesetzt. Nach 2 h Rühren wird die Lösung auf 0ºC gekühlt, und es werden 8,4 g (50 mmol) 1-Chlor-1-(1H-imidazol-4-yl)ethan-chlorhydrat zugesetzt. Die Mischung wird 2 h lang gerührt, unter Vakuum eingeengt und in 100 ml Salzsäure auf 2% gelöst. Diese Lösung wird mit 50 ml Diethylether extrahiert, durch Zugabe von Natriumcarbonat auf pH 8 gebracht und mit 200 ml Methylenchlorid in vier Portionen extrahiert. Die Methylenchlorid-Phasen werden vereinigt, über Natriumsulfat getrocknet und unter Vakuum eingedampft, um 13,8 g (88,7%) 2-Acetamido-2-[1-(1H-imidazol-4-yl)ethyl]propan-1,3- disäurediethylester in Form eines hellbraunen Öls zu ergeben.

Nach Abkühlen und Eindampfen unter Vakuum erhält man 9,8 g (96%) 2-Amino- 3-(1H-imidazol-4-yl)buttersäure-dichlorhydrat in Form eines Schaums.

- 9,40 g (38,9 mmol) 2-Amino-3-(1H-imidazol-4-yl)buttersäuredichlorhydrat werden in 100 ml Methanol gelöst. Während vierstündigen Erhitzens am Rückfluß läßt man HCl-Gas durch die Lösung strömen. Nach Abkühlen wird überschüssige Salzsäure unter Vakuum abgezogen. Man erhält 9,8 g (98%) 2-Amino-3-(1H-imidazol-4-yl)buttersäuremethyl-dichlorhydrat in Form eines weißen Feststoffes.

- 7,7 g (30 mmol) 2-Amino-3-(1H-imidazol-4-yl)buttersäuremethylester-dichlorhydrat werden bei 0ºC einer gerührten Suspension von 3,4 g (90 mmol) Lithiumaluminiumhydrid in 125 ml Tetrahydrofuran zugegeben. Anschließend wird die Mischung 3 h lang am Rückfluß erhitzt, auf 0ºC gekühlt und durch Zugabe von 6,5 ml Wasser in 15 ml Tetrahydrofuran hydrolysiert. Es wird mit 20 ml einer 5N Natriumhydroxid- Lösung gerührt, um einen grobkörnigen Niederschlag zu erhalten, der filtriert und mit 100 ml Ethanol in drei Portionen extrahiert wird. Einengen der Fraktionen ergibt ein Öl, das zur Abtrennung mineralischen Materials mit wasserfreiem Ethanol behandelt wird. Nach Filtration und Verdampfen erhält man 4g (Ausb.: 85,9%) 2-Amino-3-(1H-imidazol-4- yl)butanol, das in sein öliges Dichlorhydrat überführt wird. Zur Analyse wird eine Probe entnommen und in das Dipikrat überführt, das aus einer Mischung von Ethanol/Wasser umkristallisiert wird. Gelbe Kristalle, Schmp.: 168ºC.

Schrittweise Zugabe von 200 ml Chloroform ergibt 2-Amino-1-chlor-3-(1H-imidazol-4-yl)butandichlorhydrat. Man erhält 2,87 g (Ausb.: 88,2%) eines hygroskopischen Niederschlags.

Durch Zugabe von Petrolether kristallisiert die Titelverbindung in Form ihres Dichlorhydrats. - Das folgende Beispiel veranschaulicht die stereospezifische Synthese der erfindungsgemäßen Verbindung.
- 112,09 g (1 mol) 1H-Imidazol-4-ylcarbonsäure werden in 1,3 l Methanol gegeben. Während vierstündigen Erhitzens am Rückfluß läßt man HCl-Gas durch die Lösung strömen. Nach Abkühlen auf Umgebungstemperatur und Eindampfen auf 300 ml erhält man 98 g des Chlorhydrats der Titelverbindung in Form weißer Kristalle. Abermaliges Eindampfen ergibt 46,5 g zusätzliches Produkt (insgesamt 144,5 g; Ausb. 88,9%). Eine Analysenprobe wird aus Methanol umkristallisiert, Schmp.: 173-174ºC.

- Eine Lösung von 151,79g (1,5 mol) Triethylamin in 1 l Methylenchlorid wird mit 97,55 g (0,6 mol) 1H-Imidazol-4-ylcarbonsäuremethylester-chlorhydrat versetzt. Nach einstündigem Rühren bei Umgebungstemperatur wird die Mischung nach und nach mit einer Lösung von 183,99 g (0,66 mol) Triphenylmethylchlorid in 500 ml Methylenchlorid versetzt, und man läßt 1 h lang reagieren. Nach Beendigung der Reaktion wird die Mischung 4mal mit 200 ml Wasser gewaschen, über Natriumsulfat getrocknet und zur Trockene eingedampft. Das erhaltene Öl kristallisiert nach einer Nacht. Nach Pulverisieren und 3 Waschungen mit 200 ml heißem Diethylether erhält man 210,2 g (95,1%) der Titelverbindung in Form hellgelber Kristalle. Eine Probe wird zur Analyse umkristallisiert; weiße Kristalle, Schmp.: 147-152ºC.

- 202,65 g (0,55 mol) 1-Triphenylmethyl-1H-imidazol-4-ylcarbonsäuremethylester werden in 0,8 l trockenem Toluol bei 85ºC gelöst. 44 g (1,1 mol) Natriumhydrid (60% Dispersion in Mineralöl) werden zugegeben. Die Mischung wird unter Rühren innerhalb 2 h mit 96,92 g (1,1 mol) Ethylacetat versetzt. Man läßt die Mischung 16 h lang bei 70ºC reagieren. Danach wird das Toluol unter vermindertem Druck abgezogen, und die erhaltene Mischung wird ohne Reinigung im nächsten Schritt verwendet. Gleichwohl wird eine kleine Menge des öligen braunen Rückstands für die Analyse entnommen, in Methylenchlorid gelöst, es wird mit einer wäßrigen Lösung von Ammoniumchlorid und Wasser gewaschen, über Natriumsulfat getrocknet und eingedampft, um ein Öl zu erhalten. Eine Kristallisation aus Diethylether und Umkristallisieren aus Ethanol/Diethylether ergibt die Titelverbindung in Form weißer Kristalle; Schmp.: 136-139ºC.

Nach Rühren und Zugabe von Diethylether erhält man 115,3 g der Titelverbindung [59,5% bezogen auf b)] in Form hellgelber Kristalle. Eine Probe wird zur Analyse aus Ethanol umkristallisiert; weiße Kristalle, Schmp.: 164-165ºC.

Die Mischung wird unter Rühren nach und nach mit 71,47 g (0,3 mol) 2-Phosphonopropionsäuretriethylester versetzt und weitere 2 h bei Umgebungstemperatur gehalten. Es werden 30,5 g (86,5 mmol) 1-(Triphenylmethyl-1H-imidazol-4-yl)ethanon zugesetzt und man hält 16 h lang am Rückfluß. Nach Eindampfen und Lösen in 1 l Chloroform/Isopropanol (3:1) wird 3mal mit Wasser gewaschen. Nach Trocknen über Natriumsulfat und Eindampfen erhält man ein Öl, das hauptsächlich aus (E)-2-Methyl-3- (1-triphenylmethyl-1H-imidazol-4-yl)-2-butensäureethylester besteht. Beim Behandeln des Rückstands mit Diethylether erhält man 10,9 g eines Feststoffs, der hauptsächlich aus dem (Z)-Isomer besteht. Die Reinigung auf einer Chromatographiesäule (Kieselgel 63-200 um, Elutionsmittel: Petrolether/Diethylether 2:3) ergibt 5,6 g (14,8%) der Titelverbindung als weißen Feststoff. Eine Probe wird zur Analyse aus einer Mischung von Diethylether/Ethanol umkristallisiert und ergibt farblose Kristalle, Schmp.: 166-168ºC.

- 125 ml Chloroform werden einer Lösung des oben erhaltenen erythro-3-(1H-Imidazol-4-yl)-2-methylbutansäure-chlorhydrats in 20 ml konzentrierter Schwefelsäure zugesetzt. Die Mischung wird gerührt, und es werden 5,85 g (90 mmol) Natriumazid bei 0ºC während 1 h zugesetzt. Anschließend wird die Reaktionsmischung 14 h lang bei 45ºC gehalten. Nach Zugabe von Eis wird die organische Schicht abgetrennt. Die wäßrige Phase wird auf pH 8,5 gebracht und zur Trockene eingedampft. Das erhaltene feste Material wird in einer Soxhlet-Apparatur mit Isobutanol behandelt, um die Titelverbindung in Form eines rohen Öls zu erhalten, das durch Säulenchromatographie gereinigt wird (Kieselgel 63-200 um, Elutionsmittel: Chloroform/Ammoniak-gesättigtes Methanol 85:15), um 2,32 g [85,9% bezogen auf f)] erythro-[3-(1H-Imidazol-4-yl)but-2-yl]amin in Form eines farblosen Öls zu erhalten. Man erhält 0,79 g (42,5%) der Titelverbindung in Form weißer Kristalle, Schmp.: 181ºC.

Nach Eindampfen und erneutem Lösen in 20 ml Wasser und 20 ml Methylenchlorid wird 4mal mit Methylenchlorid extrahiert. Die wäßrige Lösung wird eingedampft und die überschüssige Bromwasserstoffsäure wird durch Lösen in Ethanol und anschließendes mehrmaliges Eindampfen abgetrennt. Nach Zusatz von Diethylether und Isopropanol gefolgt von Rühren erhält man 0,18 g (72%) (-)-erythro-[3-(1H-Imidazol-4-yl)but-2-yl]amin-dibromhydrat in Form weißer Kristalle, die im Bereich von 207-230ºC schmelzen.

Man löst in trockenem Ethanol und gibt eine äquivalente Menge Kalium-t-butylat zu, um nach Filtration und Eindampfen 0,17 g (1,2 mmol) erythro-[3-(1H- Imidazol-4-yl)but-2-yl]amin in Form der freien Base zu erhalten. Die Base wird in 10 ml einer heißen Lösung von Ethanol/Wasser (1:1) gelöst, und es wird eine heiße Lösung von 1,07 g (2,5 mmol) (-)-2R,3R-Di-O-(4-toluyl)weinsäure in 30 ml Ethanol/Wasser (1:1) zugesetzt. Man beläßt 3 Tage bei Umgebungstemperatur, dann werden die reultierenden weißen Kristalle aus Ethanol/Wasser (1:1) umkristallisiert, bis konstanter Schmelzpunkt und Drehwert erhalten werden. Man erhält 0,21 g [11,3% bezogen auf Edukt von Schritt i)]) weiße Kristalle, Schmp.: 181ºC.

- Die erfindungsgemäße Verbindung wurde Gegenstand einer pharmakologischen Untersuchung.
- Die Verbindung wurde in Form des Racemats der Stereoisomeren untersucht. Im folgenden die durchgeführten Tests:
- a) Hemmung der Histamin-Freisetzung, induziert durch Depolarisation von Hirnrindenschnitten der Ratte nach der von Arrang et al., Nature 302, 832, 1983, beschriebenen Methode. Die durch die erfindungsgemäße Verbindung hervorgerufene maximale Hemmung ist identisch mit der durch Histamin (das ein vollständiger Agonist ist) hervorgerufenen, und ihre Konzentration bei 50% Hemmung (CI&sub5;&sub0;) ist wie folgt:
- - "Racemat" 12 10&supmin;&sup9; M
- - erythro-Form 6 10&supmin;&sup9; M
- - (+)-erythro-Isomer (3,4 ± 2,0) 10&supmin;&sup9; M
- - (-)-erythro-Isomer (48 ± 31) 10&supmin;&sup9; M
- - threo-Form 200 10&supmin;&sup9; M
- Es zeigt sich somit, daß die Wirkung der erfindungsgemäßen Verbindung auf die H&sub3;-Rezeptoren selektiv in einem der vier Stereoisomeren liegt, nämlich dem (+)-erythro-Enantiomer. Die Wirkung des (-)-erythro-Enantiomers ist wahrscheinlich geringer als dem oben gefundenen Wert entspricht, da die Verbindung durch einen kleinen Anteil (um 15%) (+)-erythro- Enantiomer verunreinigt ist.
- b) Kontraktion des isolierten Meerschweinchenkrummdarms:
- Die Verbindung ist ein vollständiger Agonist, zeigt aber nur weniger als 1/1000 der Wirkung von Histamin.
- c) Kontraktion des isolierten Meerschweinchenherzohrs:
- Die Verbindung ist ein partieller Agonist (maximale Wirkung: 60% der von Histamin), etwa tausendfach weniger wirksam als Histamin.
- Die Ergebnisse sind in folgender Tabelle zusammengestellt. TABELLE Wirkung der erfindungsgemäßen Verbindung auf die drei histaminergen Rezeptorklassen im Vergleich zu der von Histamin. Histamin α,β-Dimethylhistamin "Racemat" erythro-Form (+)-erythro (-)-erythro
- Außerdem hat sich die Verbindung als in vivo wirksam erwiesen, da sie in einer oralen Dosis von 20 mg/kg (Racemat) die Histamin-Synthese im Rattenhirn maximal hemmt (bestimmt nach der von Arrang et al., Nature 327, 117, 1987, beschriebenen Methode).
- Die Studie zeigt, daß die erfindungsgemäße Verbindung sehr starke und vor allem sehr selektive H&sub3;-Wirkung zeigt. Tatsächlich ist ihre Wirkung auf die Rezeptoren H&sub1; und H&sub2; vernachlässigbar, 10 bis 100mal schwächer als die der besten unter den gemäß EP-A-0 214 058 bekannten Verbindungen, während ihre H&sub3;-Wirkung und ihre Selektivität überlegen sind. Sie kann somit ein sehr potenter und sehr selektiver H&sub3;-Agonist sein, der in der Therapie unter verschärften Sicherheitsbedingungen verwendbar ist.
- Daher ist sie geeignet, die histaminerge Reizleitung im Verdauungstrakt, im Nerven-, kardiovaskulären und Immunsystem zu vermindern. Somit ist sie in der Therapie verwendbar als Arzneimittel mit sedativer, schlafregulierender, antikonvulsiver, hypothalamus-/hypophysensekretionsregulierender, antidepressiver, gehirnkreislaufmodulierender Wirkung etc..
- Zudem ist durch die Stimulierung z.B. der H&sub3;-Rezeptoren der Lunge eine Hemmung der Freisetzung von Entzündungsbotenstoffen bei verschiedenen allergischen Erkrankungen (z.B. Asthma) zu erwarten.
- In der Gastroenterologie kann die Hemmung der Histamin- Freisetzung im Magen sekretions- und geschwürhemmende Wirkung ausüben. Eine Veränderung bei der Freisetzung von Botenstoffen der Immunantwort kann letztere modifizieren.
- Aufgrund dieser Eigenschaften kann die erfindungsgemäße Verbindung in Form des Racemats oder des wirksamsten Stereoisomers das wirksame Prinzip von pharmazeutischen Zusammensetzungen darstellen, die auch ein pharmazeutisch verträgliches Vehikel oder eine pharmazeutisch verträgliche Trägersubstanz umfassen. Ihre Wirkungsweise, ihre verschiedenen pharmakologischen Wirkungen und ihre geringe Toxizität am Tier lassen an Anwendungen in der Human- und Veterinärmedizin denken, mit Dosen in der Größenordnung von 0,1 bis 10 mg/kg, insbesondere oral oder parenteral.
- Sie kann insbesondere dargereicht werden in Form von Tabletten, Dragées, Gelatinekapseln, Aerosolen, injizierbaren Lösungen oder Suppositorien.
Claims (6)
1. α,β-Dimethylhistamin in Form des Racemats, des optischen
Isomers oder der Diastereomerenmischung, sowie dessen
pharmazeutisch annehmbare Säureadditionssalze.
2. Erythro-α,β-Dimethylhistamin sowie dessen pharmazeutisch
annehmbare Säureadditionssalze.
3. (+)-Isomer von erythro-α,β-Dimethylhistamin sowie dessen
pharmazeutisch annehmbare Säureadditionssalze.
4. Pharmazeutische Zusammensetzung, enthaltend eine
Verbindung nach einem der Ansprüche 1 bis 3 und ein
pharmazeutisch verträgliches Vehikel oder eine pharmazeutisch
verträgliche Trägersubstanz.
5. Verfahren zur Herstellung der Verbindung nach Anspruch
1, dadurch gekennzeichnet, daß man ein Alkyl(mit C&sub1;-C&sub6;)-
2-amino-3-(1H-imidazol-4-yl)butyrat mit Hilfe eines
Metallhydrids zu 2-Amino-3-(1H-imidazol-4-yl)butanol
reduziert, dieses Butanol-Derivat mit Hilfe eines
Chlorierungsmittels in 2-Amino-1-chlor-3-(1H-imidazol-
4-yl)butan umwandelt und man dieses chlorierte Derivat
katalytisch reduziert.
6. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß
das Chlorierungsmittel Thionylchlorid ist.
