Description
Die Erfindung betrifft Fluorenverbindungen und Verfahren zur Herstel
lung verschiedener Fluorenverbindungen.
Der Ausdruck "Fluoren" bezeichnet die tricyclische Verbindung der
Strukturformel:
In den hier verwendeten chemischen Namen wird die Position von
Substituenten am Fluoren durch Angabe der entsprechenden Ziffer
angezeigt.
Falls nicht anders angegeben, bedeutet der Ausdruck "Fluorenyl" den 9-
Fluorenyl-Rest.
Für Fluoren und verschiedene Derivate von Fluoren sind weite Anwen
dungsbereiche offenbart worden. Die US-PS 40 16 289 offenbart, daß
gewisse Fluorenderivate beim Herstellen von Medikamenten zum Ge
brauch bei der Behandlung von Viruserkrankungen nützlich sind. Die
US-PS 34 26 069 offenbart, daß gewisse N-Alkanoyl-amino-alkyl-fluorenderi
vate als Monomere zur Herstellung von Polyamiden, Polyharnstoffen und
dergleichen nützlich sind. Die US-PS 31 14 737 offenbart, daß gewisse
Fluorenylmetallocene als Katalysatoren für die wäßrige Polymerisation von
Vinylmonomeren, wie Styrol oder Acrylate, nützlich sind. Anch die US-PS
48 92 851 offenbart ein Fluorenylmetallocen als Katalysator zum Polyme
risieren eines Olefins.
Eine Anfgabe der vorliegenden Erfindung besteht darin, neue Fluorende
rivate bereitzustellen. Eine andere Aufgabe der vorliegenden Erfindung
ist es, neue Verfahren zur Herstellung von Fluorenderivaten bereitzu
stellen. Andere Aspekte, Aufgaben und Vorteile der vorliegenden Erfin
dung ergeben sich aus der folgenden Beschreibung.
Gemäß einem Aspekt stellt die vorliegende Erfindung Methoden zur
Herstellung von 1-Methylfluoren bereit. Eine bevorzugte Ansführungsform
betrifft die Umsetzung von Fluoranthen zu 1-Methylfluoren.
Ein anderer Aspekt der Erfindung umfaßt Methoden zur Herstellung von
4-Methylfluoren. Eine bevorzugte Ausführungsform betrifft die Umsetzung
von Phenanthren zu 4-Methylfluoren.
Ein weiterer Aspekt der vorliegenden Erfindung betrifft eine Methode
zur Herstellung von tert.-butylsubstituierten Fluorenen durch Umsetzung
von carborylsubstituierten Fluorenen mit Trimethylaluminium.
Zwei verschiedene Wege sind zur Herstellung von 1-Methylfluoren verfüg
bar. In beiden Fallen umfaßt eine bevorzugte Ausführungsform die
Herstellung des 1-Methylfluorens aus einem Ausgangsmaterial, das als
Fluoranthen bekannt ist.
Einer der Wege umfaßt letztlich die Reduktion von 1-Hydroxymethyl
fluorenon mit Wasserstoff in Anwesenheit eines Palladium-Kohlenstoff-
Katalysators unter Bildung von 1-Methylfluoren. Der andere Weg umfaßt
letztlich die Reduktion von 1-Hydroxymethylfluoren mit Wasserstoff in
Anwesenheit eines Palladium-Kohlenstoff-Katalysators unter Bildung von
1-Methylfluoren.
Die Reduktion von 1-Hydroxymethylfluorenon oder 1-Hydroxymethylfluo
ren mit Wasserstoff kann unter allen geeigneten Reaktionsbedingungen
ausgeführt werden. Allgemein wird die Reaktion in Anwesenheit eines
geeigneten flüssigen Verdünnungsmittels, z. B. eines Lösungsmittels für die
Fluoren- oder Fluorenonverbindung, durchgeführt. Beispiele sind Benzol,
Toluol, Xylol oder Tetrahydrofuran. Alle geeigneten Palladium-Kohlen
stoff-Katalysatoren können eingesetzt werden. Solche Katalysatoren enthal
ten typischerweise 1 bis 20 Gew.-% Palladium. Die Bedingungen von
Druck und Temperatur können leicht vom Fachmann für die optimale
Herstellung ausgewählt werden. Die Reaktion wird typischerweise unter
solchen Bedingungen durchgeführt, daß der Wasserstoff einen Gesamt
druck zur Folge hat, der größer als der Luftdruck ist. Das Reaktionsp
rodukt kann nach bekannten Methoden isoliert und gereinigt werden.
Der Weg, der die Reduktion von 1-Hydroxymethylfluorenon umfaßt,
erlaubt die Herstellung von 1-Methylfluoren aus Fluoranthen in drei
Reaktionsschritten. Im einzelnen wird das Fluoranthen mit Wasserstoffper
oxid und Essigsäure unter geeigneten Reaktionsbedingungen zum 1-Gar
bonsäurefluorenon umgesetzt. Das 1-Carbonsäurefluorenon wird unter
Verwendung eines "gemischten Hydrids", d. h. ein Lithiumaluminiumhy
drid/Aluminiumchlorid-Katalysator unter Bildung des 1-Methoxyfluorenons
reduziert. Die Reduktion mittels Lithiumaluminiumhydrid/Aluminiumchlo
rid-Katalysator wird im allgemeinen in Anwesenheit eines geeigneten,
flüssigen Verdünnungsmittels durchgeführt, typischerweise ein Kohlenwas
serstoff oder ein Ether, wie Diethylether und/oder Tetrahydrofuran.
Das Molverhältnis von Lithiumalumniumhydrid zu Aluminiumchlorid kann
- nach Maßgabe der gewünschten Ergebnisse - über einen weiten Bereich
variieren. Typischerweise liegt das Molverhältnis von Lithiumalumini
umhydrid zu Aluminiumchlorid im Bereich von etwa 3 : 1 bis etwa 1 : 1.
Der "gemischte Hydrid"-Katalysator wird im allgemeinen in einem Mol
verhältnis von etwa 2 : 1 (Lithiumaluminiumhydrid zu Aluminiumtrichlorid)
verwendet.
Das Verfahren zur Herstellung des 1-Methylfluorens durch Reduktion von
1-Hydroxymethylfluoren umfaßt vier Stufen, wenn man Fluoranthen als
Ausgangsmaterial verwendet. Die erste Stufe betrifft die Umsetzung von
Fluoranthen zu 1-Carbonsäurefluorenon, bevorzugt durch Reaktion von
Fluoranthen mit Wasserstoffperoxid in Essigsäure. Das erhaltene 1-Car
bonsäurefluorenon wird dann mit Wasserstoff unter Verwendung eines
Palladium-Kohlenstoff-Katalysators in der gleichen Weise reduziert, wie
vorher beschrieben. Das resultierende 1-Garbonsäurefluoren wird dann
unter Verwendung eines Lithiumaluminiumhydrid/Aluminiumchlorid-Kataly
sators zu 1-Hydroxymethylfluoren reduziert, das dann mit Wasserstoff in
Anwesenheit eines Palladium-Kohlenstoff-Katalysators reduziert wird.
Zwei verschiedene Wege sind auch zur Herstellung des 4-Methylfluorens
verfügbar. Ein Weg umfaßt die Reduktion von 4-Hydroxymethylfluoren
mit Wasserstoff in Anwesenheit eines Palladium-Kohlenstoff-Katalysators.
Der andere Weg umfaßt die Reduktion von 4-Hydroxymethylfluorenon
mit Wasserstoff in Anwesenheit eines Palladium-Kohlenstoff-Katalysators.
Diese Reduktionen können in der gleichen Weise ausgeführt werden, wie
vorher für die vergleichbare Reduktion beschrieben, die die Herstellung
von 1-Methylfluoren betrifft.
In einer bevorzugten Ansführungsform wird das 4-Methylfluoren aus
Phenanthren hergestellt. Das Phenanthren wird zu 2,2′-Dicarbonsäure
biphenyl durch Verwendung von Wasserstoffperoxid in Essigsäure oxidiert.
Das Biphenylprodukt wird dann unter Verwendung von Schwefelsäure
gemäß J. Am. Chem. Soc. 64, 2845 (1942) unter Bildung von 4-Car
bonsäurefluorenon oxidiert. Das 4-Carbonsäurefluorenon kann dann unter
Verwendung des Lithiumaluminiumhydrid/Aluminiumchlorid "gemischten
Hydrid"-Katalysators, wie oben beschrieben zu 4-Hydroxymethylfluorenon,
reduziert werden, das durch Reduktion mit Wasserstoff in Anwesenheit
eines Palladium-Kohlenstoff-Katalysators in das 4-Methylfluoren überführt
wird. In einem alternativen Weg kann das 4-Carbonsäurefluorenon mit
Wasserstoff in Anwesenheit eines Palladium-Kohlenstoff-Katalysators zu 4-
Carbonsäurefluoren reduziert werden, und diese Verbindung kann mit Li
thiumaluminiumhydrid/Aluminiumchlorid zu 4-Hydroxymethylfluoren redu
ziert werden, das dann seinerseits mit Wasserstoff in Anwesenheit eines
Palladium-Kohlenstoff-Katalysators zu 4-Methylfluoren reduziert werden
kann.
Tert.-butylsubstituierte Fluorenverbindungen können aus carboxylsubstitu
ierten Fluorenverbindungen durch Umsetzung der Carboxylgruppe mit
Trimethylaluminium hergestellt werden. Der Begriff "carboxylsubstituierte
Fluorenverbindungen" bezeichnet hierbei Fluorenverbindungen mit einem
Substituent der einen Carbonylrest besitzt, dessen Kohlenstoff mit einer
beliebigen Position des Fluorenrings verknüpft ist. Beispiele umfassen
Carbonsäuren und Acetylreste.
Bei dieser Reaktion wird typischerweise das carboxylsubstituierte Fluoren
in einer geeigneten Flüssigkeit gelöst und dann eine geeignete Menge
Trimethylaluminium zugegeben. Die angewendeten Temperatur- und
Druckbedingungen können nach Maßgabe der gewünschten Ergebnisse
über einen weiten Bereich variieren. Vorzugsweise wird die Reaktion
unter Rückfluß für eine Dauer von 1 bis 20 Stunden durchgeführt, wobei
eine Dauer von 5 bis 10 Stunden besonders bevorzugt ist. Das erhaltene
Produkt kann in bekannter Weise abgetrennt und gereinigt werden. Die
Menge des verwendeten Trimethylaluminiums kann nach Maßgabe der
gewünschten Ergebnisse etwas variieren, abhängig von der verwendeten
speziellen Fluorenverbindung und der gewünschten Ausbeute. Handelt es
sich bei der Fluorenverbindung um ein carbonsäuresubstituiertes Fluoren,
beträgt das Molverhältnis von Trimethylaluminium zu Fluorenverbindung
vorzugsweise mindestens etwa 5 : 1. Wenn die Fluorenverbindung eine
Acetylverbindung ist, kann andererseits ein Molverhältnis von nur etwa
2 : 1 von Trimethylaluminium zu Fluorenverbindung zufriedenstellend sein.
Die Beispiele erläutern die Erfindung.
Zwei verschiedene Reaktionsschemas sind verwendet worden, um 1-
Methylfluoren aus Fluoranthen darzustellen. Die Reaktionsschemas kön
nen durch das folgende Flußdiagramm veranschaulicht werden. Beide
Schemas umfassen die Verwendung von 1-Carbonsäurefluorenon als
Ausgangsmaterial.
Zur Herstellung des 1-Carbonsäurefluorenons, d. h. Formel 1, wurden 20,2
g (0,1 mol) Fluoranthen in 150 ml Essigsäure bei 90° C gelöst. Bei dieser
Temperatur wurden dann 200 ml 30% wäßriges H2O2 schrittweise zu
gegeben. Dann wurde das Reaktionsgemisch für weitere drei Stunden bei
dieser Temperatur gerührt. Zu Beginn der Reaktion wurde ein schwach
gelber Niederschlag gebildet, der nach einiger Zeit verschwand. Dann
wurde das Reaktionsgemisch auf 0° C im Eisbad gekühlt. Ein orange
farbener Niederschlag wurde gebildet und abfiltriert. Das Filtrat wurde
in kalte, verdünnte, wäßrige HCl gegossen. Ein orange-gelber Nieder
schlag wurde gebildet, der zweimal mit H2O gewaschen wurde und dann
in einer wäßrigen NH3-Lösung gelöst wurde, um das nicht umgesetzte
Fluoranthen zu entfernen. Dann wurde das Gemisch filtriert. Beim
Neutralisieren des Filtrats mit verdünnter HCl wurde ein oranger Nieder
schlag gebildet. Der Niederschlag, 1-Carbonsäurefluorenon, wurde abfil
triert und getrocknet. Die gebildete Menge war 13,4 g.
Etwa 0,76 g (0,02 mmol) LiAlH4 wurden in einem Gemisch aus 75 ml
Diethylether und 25 ml Tetrahydrofuran (getrocknet über LiAlH4) sus
pendiert. Das Gemisch wurde auf 0° C in einem Eisbad gekühlt. Dann
wurden 1,33 g (0,01 mmol) AlCl3 in kleinen Portionen zugefügt, und das
Gemisch wurde bei Raumtemperatur 15 Minuten gerührt. Dann wurden
4,2 g (0,02 mmol) des Carbonsäurefluorenons, gelöst in 400 ml Tetrahy
drofuran, über einen Tropftrichter zugefügt, während das Reaktionsge
misch unter Rückfluß erhitzt wurde. Das Rühren wurde für zusätzliche
30 Minuten beibehalten. Dann wurde das Reaktionsgemisch auf Zimmer
temperatur gekühlt, und nicht umgesetztes LiAlH4 wurde mit einer
wäßrigen Lösung von HCl zerstört. Die organische Phase wurde zwei
Mal mit Wasser gewaschen und über Na2SO4 getrocknet. Dann wurde
das Lösungsmittel mit einem Verdampfer entfernt. Der Feststoff, d. h. 1-
Hydroxymethylfluorenon (Formel 2), wurde in einer Menge von 3,2 g
isoliert.
Das rohe 1-Hydroxymethylfluorenon kann ohne weitere Reinigung ver
wendet werden. 2 g Palladium auf Kohlenstoff-Katalysator, der etwa 10
Gew.-% Pd enthielt, wurden in einem Kolben eingewogen, und 4,2 g
(0,02 mmol) des isolierten 1-Hydroxymethylfluorenons wurden in 250 ml
Tetrahydrofuran gelöst und in den Kolben gegeben. Die Hydrierung
wurde bei Raumtemperatur unter einem schwachen Überdruck von H2
durchgeführt bis 1350 ml H2 verbraucht wurden. Das Reaktionsgemisch
wurde filtriert und das Lösungsmittel des Filtrats wurde im Vakuum
entfernt. Der cremefarbene Rückstand wurde mit Pentan extrahiert, die
Lösung wurde über Silica-Gel filtriert und das Lösungsmittel im Vakuum
entfernt. Das erhaltene Produkt, 1-Methylfluoren, war ein farbloser
Feststoff und wurde in quantitativer Ausbeute gebildet.
Im zweiten Weg wird das 1-Carbonsäurefluorenon durch Verwendung des
Palladium-Kohlenstoff-Katalysators in der gleichen Weise reduziert, wie
für die Umsetzung des 1-Hydroxymethylfluorenons zu 1-Methylfluoren
beschrieben. Eine quantitative Ausbeute von 1-Carbonsäurefluoren, d. h.
Formel 3, wurde erhalten. Das Volumen von verbrauchtem Wasserstoff
war 960 ml. Dieses Produkt wurde dann zu 1-Hydroxymethylfluoren, d. h.
Formel 4, reduziert durch Verwendung des LiAlH4/AlCl3 "gemischten
Hydrid"-Katalysators, wie für die Herstellung von 1-Hydroxymethylfluore
non beschrieben. Das 1-Hydroxymethylfluoren wurde dann unter Ver
wendung des Palladium-Kohlenstoff-Katalysators und Wasserstoff unter
Bildung von 1-Methylfluoren reduziert.
Etwa 2 g (0,01 mmol) 1-Carbonsäurefluoren wurden in 50 ml Toluol
suspendiert. Dann wurden 4,6 ml, d. h. etwa 0,05 mol AlMe3 zu der
Lösung zugegeben, und das Reaktionsgemisch wurde für 10 Stunden
unter Rückfluß gekocht. Beim Erhitzen bildete das Reaktionsgemisch eine
homogene Lösung. Das Reaktionsgemisch wurde auf Raumtemperatur
gekühlt und dann in eisgekühlte, verdünnte wäßrige HCl gegossen. Die
organische Schicht wurde abgetrennt, mit H2O gewaschen und über
Na2SO4 getrocknet. Dann wurde das Lösungsmittel im Vakuum entfernt.
Der farblose Rückstand wurde mit Pentan extrahiert, die Lösung über
Silica-Gel filtriert und das Lösungsmittel im Vakuum entfernt. Die
Ausbeute von 1-tert.-Butylfluoren, Formel 6, war quantitativ.
In dieser Reaktion wurde 2-Acetylfluoren mit Trimethylaluminium umge
setzt. Die Methylierung war analog zu der in Beispiel II beschriebenen
Umwandlung von Verbindung 3 in Verbindung 6. Jedoch war in diesem
Fall nur ein zweifacher Überschuß von AlMe3 notwendig. Das 2-tert.-
Butylfluoren wurde als weißer Feststoff in quantitativer Ausbeute gebildet.
Zwei verschiedene Reaktionsschemas sind verwendet worden, um 4-
Methylfluoren, d. h. Formel 15, herzustellen. Die Schemas können wie
folgt zusammengefaßt werden:
Beide Schemas benötigen 4-Carbonsäurefluorenon, Formel 11, als Aus
gangsmaterial. Diese Verbindung wurde aus Phenanthren durch Anwen
dung eines ähnlichen Verfahrens gebildet, wie das in J. Org. Chem 21,
243 (1956) offenbarte, mit der Ausnahme, daß kein Acetanhydrid ver
wendet wurde. Statt dessen wurden Wasserstoffperoxid und Essigsäure ver
wendet, wobei in 67%iger Ausbeute 2,2′-Dicarbonsäurebiphenyl, d. h.
Formel 10, erhalten wurde.
Das Biphenylprodukt der Formel 10 wurde dann unter Verwendung von
Schwefelsäure gemäß J. Am. Chem. Soc. 64, 2845 (1942) oxidiert, wobei
4-Carbonsäurefluorenon der Formel 11 in einer Ausbeute von 82%
erhalten wurde.
Die Verbindung von Formel 11 wurde unter Verwendung von LiAlH4
und AlCl3 in der gleichen Weise reduziert, wie in Beispiel I. Die Reak
tion führte zu einer 80%igen Ausbeute an 4-Hydroxymethylfluorenon
(Formel 14), das dann unter Verwendung von Wasserstoff und dem
vorher beschriebenen Palladium-Kohlenstoff-Katalysator reduziert wurde.
Eine quantitative Ausbeute von 4-Methylfluoren resultierte.
Die Verbindung der Formel 11 wurde unter Verwendung von Wasserstoff
und dem vorher beschriebenen Palladium-Kohlenstoff-Katalysator redu
ziert. Die Reaktion erbrachte eine quantitative Ausbeute an 4-Carbonsäu
refluoren der Formel 12. Reduktion dieser Säure mit LiAlH4 und AlCl3
ergab eine 80%ige Ausbeute an 4-Hydroxymethylfluoren der Formel 13.
Dieses Produkt wurde dann unter Verwendung von Wasserstoff und dem
Palladium-Kohlenstoff-Katalysator reduziert, wobei 4-Methylfluoren in
quantitativer Ausbeute gebildet wurde.
4-Carbonsäurefluoren wurde mit Trimethylaluminium umgesetzt wie
allgemein in Beispiel II beschrieben. Hierbei wurde in 60%iger Ausbeute
4-tert.-Butylfluoren erhalten.
Claims (11)
1. Tert.-Butylfluorene der allgemeinen Formel I,
wobei sich die tert.-Butylgruppe in 1-, 2- oder 4-Stellung befindet.
2. Verfahren zur Herstellung von tert.-butylsubstituierten Fluorenen,
dadurch gekennzeichnet, daß man das entsprechende carboxylsub
stituierte Fluoren mit Trimethylaluminium umsetzt.
3. Verfahren nach Anspruch 2, dadurch gekennzeichnet, daß das 1-
carboxylsubstituierte Fluoren durch Reduktion von 1-Carboxyfluorenon
mit H2 in Gegenwart eines Palladium-Kohlenstoff-Katalysators erhal
ten worden ist.
4. Verfahren nach Anspruch 3, dadurch gekennzeichnet, daß das 1-
Carboxyfluorenon durch Umsetzung von Fluoranthen mit H2O2 und
Essigsäure erhalten worden ist.
5. Verfahren zur Herstellung von 1- oder 4-Methylfluoren, dadurch
gekennzeichnet, daß man 1- oder 4-Hydroxymethylfluorenon oder 1-
oder 4-Hydroxymethylfluoren der Reduktion mit Wasserstoff in
Anwesenheit eines Palladium-Kohlenstoff-Katalysators unterwirft.
6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß das 1- oder
4-Hydroxymethylfluorenon oder das 1- oder 4-Hydroxymethylfluoren
durch Reduktion des 1- oder 4-Carbonsäurefluorenons bzw. des 1-
oder 4-Carbonsäurefluorens mit LiAlH4 und AlCl3 erhalten worden
ist.
7. Verfahren nach Anspruch 6, dadurch gekennzeichnet, daß das 1- oder
4-Carbonsäurefluoren durch Reduktion von 1- oder 4-Carbonsäu
refluorenon mit H2 in Anwesenheit eines Palladium-Kohlenstoff-
Katalysators erhalten worden ist.
8. Verfahren nach Anspruch 6 oder 7, dadurch gekennzeichnet, daß das
1-Carbonsäurefluorenon durch Umsetzen von Fluoranthen mit H2O2
und Essigsäure erhalten worden ist.
9. Verfahren nach Anspruch 6 oder 7, dadurch gekennzeichnet, daß das
4-Carbonsäurefluorenon durch Umsetzung von 2,2′-Dicarbonsäurebi
phenyl mit Schwefelsäure erhalten worden ist.
10. Verfahren nach Anspruch 9, dadurch gekennzeichnet, daß das 2,2′-Di
carbonsäurebiphenyl aus Phenanthren erhalten worden ist.
11. Verfahren nach Anspruch 10, dadurch gekennzeichnet, daß das 2,2′-
Dicarbonsäurebiphenyl durch Umsetzung von Phenanthren mit H2O2
und Essigsäure erhalten worden ist.
