Description
- Die vorliegende Erfindung betrifft die Verwendung von speziellen Ethylaminderivaten als antihypertensive Arzneistoffe.
- Hochdruck ist ein verbreitetes Leiden, dessen Häufigkeit mit dem Alter zunimmt. Ferner stellt Hochdruck einen wichtigen Faktor bei Gehirnschlag und Herzleiden dar, die ebenfalls eine häufige Todesursache sind. Demgemäß werden antihypertensive Arzneistoffe als besonders wichtige Arzneimittel für geriatrische Erkrankungen angesehen.
- Bei 90% oder mehr der Hochdruckpatienten ist die Ursache des Hochdrucks nicht klar. Die Patienten müssen während ihrer gesamten Lebensdauer einen antihypertensiven Arzneistoff einnehmen. Daher muß der antihypertensive Arzneistoff sehr sicher sein und eine zuverlässig pharmazeutische Wirkung gegen Hochdruck entfalten sowie eine lang anhaltende Wirkung zeigen.
- In letzter Zeit wurde über Serotonin-antagonistische Antihypertensiva berichtet; vergl. z. B. Journal of Cardiovascular Pharmacology, Bd. 7 (Suppl. 7), S. 110-111 (1985). Derartige Antihypertensiva sind jedoch im Hinblick auf ihre antihypertensive Wirkungsweise und ihre niedrige Aktivität nicht immer zufriedenstellend. Von derartigen Arzneimitteln wurde berichtet, daß sie auch gegen verschiedene Krankheiten, die durch Serotonin hervorgerufen werden, beispielsweise Thrombose, Geschwüre und dergl., wirken, daß aber die Wirksamkeit gegen diese Erkrankungen nicht klar nachgewiesen worden ist. Ferner sind zahlreiche dieser Antihypertensiva und ihrer synthetischen Zwischenprodukte in organischen Lösungsmitteln ziemlich unlöslich oder müssen durch komplizierte und aufwendige Verfahren hergestellt werden.
- Erfindungsgemäß wird die Verwendung von speziellen Ethylaminderivaten der nachstehenden Formel zur Herstellung von antihypertensiv wirksamen Arzneimitteln bereitgestellt. Derartige Verbindungen weisen eine Serotonin-antagonistische Wirkung und eine antihypertensive Wirkung auf und lassen sich in einfacher und vorteilhafter Weise großtechnisch herstellen.
- In der vorstehenden Formel (I) gelten folgende Definitionen:
- Die in der Formel durch Q wiedergegebene Struktur entspricht einer der folgenden Formeln:
- worin E ein Stickstoffatom, -NH oder substituiertes -NH, ein Sauerstoffatom oder ein Schwefelatom bedeutet, Y, Z¹, Z², Z³ und Z&sup4; unabhängig voneinander unter Wasserstoff und Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, die mit einem oder mehreren Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder Phosphoratomen substituiert sind, Aralkylgruppen mit 6 bis 12 Kohlenstoffatomen, Aralkylgruppen mit 6 bis 12 Kohlenstoffatomen, die mit einem oder mehreren Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder Phosphoratomen substituiert sind, Arylgruppen mit 6 bis 12 Kohlenstoffatomen und Arylgruppen mit 6 bis 12 Kohlenstoffatomen, die mit einem oder mehreren Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder Phosphoratomen substituiert sind, ausgewählt sind, Y gegebenenfalls an X gebunden ist, wobei eine Brückengruppe gebildet wird, in der die Brücke durch eine Alkylenbrücke oder eine Alkylenbrücke, die substituiert ist mit einem oder mehreren Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder Phosphoratomen und/oder mindestens ein Sauerstoff-, Schwefel- oder Stickstoffatom gebildet wird, und Z¹ und Z², Z² und Z³ oder Z³ und Z&sup4; gegebenenfalls aneinander gebunden sind, wobei eine Brückengruppe gebildet wird, in der die Brücke durch eine Alkylenbrücke oder eine Alkylenbrücke, die substituiert ist mit einem oder mehr Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder Phosphoratomen und/oder mindestens ein Sauerstoff-, Schwefel- oder Stickstoffatom gebildet wird, Spezielle Beispiele für den Substituenten Q sind nachstehend angegeben:
- C bedeutet Wasserstoff, Methyl, Ethyl, Propyl, Butyl, Pentyl oder Hexyl oder ist an A unter Bildung einer Methylen-, Ethylen- oder Propylenbrücke gebunden.
- Die in der Formel durch die kombinierte Gruppe -A-B wiedergegebene Struktur ist unter folgenden Formeln ausgewählt:
- Aus den vorstehenden Formeln ist ersichtlich, daß A eine der folgenden Bedeutungen haben kann:
- worin eine der Bindungen mit der Gruppe C in Formel I verknüpft ist oder ein Wasserstoffatom trägt. B kann andererseits als gegebenenfalls substituierter Aryl- oder Aralkylrest angesehen werden, der an A über einen Substituenten, wie Sulfonyl oder Carbonyl, gebunden ist.
- Bei der verknüpfenden Gruppe X handelt es sich um eine organische Gruppe, die unter den folgenden Formeln (VIII-XI) ausgewählt ist:
- worin n eine ganze Zahl von 0 bis 9 bedeutet, Z&sup5; und Z&sup6; unabhängig voneinander für Wasserstoff, Alkyl oder Aralkyl oder Alkyl oder Aralkyl, das mit einem oder mehreren Heteroatomen und/oder mit einem oder mehreren Substituenten, die gegebenenfalls ein oder mehrere Heteroatome enthalten, substituiert ist, stehen, Z&sup7; eine Alkylgruppe mit 1 bis 20 Kohlenstoffatomen oder eine Alkylgruppe mit 1 bis 20 Kohlenstoffatomen ist, die substituiert ist mit einem oder mehreren Heteroatomen und/oder mit einem oder mehreren Substituenten, die gegebenenfalls ein oder mehrere Heteroatome enthalten, und Z&sup8; Wasserstoff, eine Cyan-, Amino-, Nitro-, Carboxyl-, Ester- oder Aminocarbonylgruppe ist. Beim Ester von Z&sup8; kann es sich beispielsweise um Alkoxycarbonyl oder Alkoxysulfonyl handeln.
- Es ist somit ersichtlich, daß X lediglich aus einer gegebenenfalls substituierten Alkylengruppe (Formel XI) bestehen kann oder lediglich eine Brücke über ein Stickstoffund/oder Schwefelatom (Formel VIII) bilden kann oder eine Kombination dieser beiden Möglichkeiten (Formel IX) darstellen kann.
- Das erfindungsgemäß verwendete Ethylaminderivat kann in Form eines Salzes vorliegen. Wenn das Salz des Derivats erfindungsgemäß einem antihypertensiven Arzneimittel einverleibt wird, soll es selbstverständlich in Form eines pharmazeutisch verträglichen Salzes vorliegen.
- Das vorstehend definierte Ethylaminderivat erweist sich vorteilhaft als Antihypertensivum zur Behandlung von Säugetieren, einschließlich Menschen, mit Hochdruck. Das Antihypertensivum kann peroral in Form einer Tablette, Kapsel oder Elixiers oder parenteral in Form einer sterilen Lösung oder Suspension verabreicht werden, um den Blutdruck zu verringern. Das Ethylaminderivat kann den Patienten oder Tieren, die behandelt werden sollen, im allgemeinen mehrmals jeweils in einer Einheitsdosis von 0,2-500 mg/Patient oder Tier verabreicht werden. Demgemäß kann die Gesamtdosis des Derivats 1-2000 mg/Patient oder Tier/Tag betragen. Selbstverständlich kann die Dosierungsmenge je nach dem Krankheitszustand, dem Gewicht des Patienten oder Tiers und anderen Faktoren, die vom Fachmann in geeigneter Weise in Betracht gezogen werden, verändert werden.
- Das Ethylaminderivat kann auch zusammen mit einem weiteren antihypertensiven Mittel verabreicht werden. Als Beispiele für derartige Mittel, die zusammen mit dem erfindungsgemäßen Derivat eingesetzt werden können, lassen sich α&sub1;-Antagonisten, wie Prozosin, Calcium-Antagonisten, wie Nifedipin, Nicardipin, Diltiagen und Verapamil, oder Konvertase-Inhibitoren, wie Captoril oder Enalapril, erwähnen.
- Die vorerwähnten typischen Kombinationen können auf herkömmliche Weise zu einer pharmazeutischen Zusammensetzung zubereitet werden. Etwa 0,2 bis etwa 500 mg des Ethylaminderivats oder eines physiologisch verträglichen Salzes oder eines Gemisches davon werden mit einem physiologisch verträglichen Vehikel, Träger, Streckmittel, Bindemittel, Antiseptikum, Stabilisator, Geschmacksstoff und dergl., in Mengen, wie sie für herkömmliche pharmazeutische Präparate erforderlich sind, vermischt.
- Beispiele für pharmazeutische Additive zur Herstellung von Tabletten, Kapseln und dergl. sind: Bindemittel, wie Traganth, Gummi arabicum, Maisstärke oder Gelatine; Vehikel, wie feinkristalline Cellulose; Streckmittel, wie Maisstärke, vorgelierte Stärke oder Alginsäure; Süßungsmittel, wie Saccharose, Lactose oder Saccharin; Geschmackstoffe, wie Pfefferminz, Öl aus Gaulthenia adenothrix Maxim oder Kirsche. Wenn das Präparat in Form einer Kapsel vorliegt, kann es zusätzlich einen flüssigen Träger, beispielsweise ein Fett oder Öl, neben den vorerwähnten Materialien enthalten. Verschiedene weitere Materialien können eingesetzt werden, um beschichtete Pillen zu bilden oder um die physikalische Form des Präparats durch ein abweichendes Verfahren zu verändern. Beispielsweise können Tabletten mit Shellack, Zucker oder beiden Bestandteilen beschichtet werden. Ein Sirup oder Elixier kann den Wirkstoff zusammen mit Saccharose als Süßungsmittel, Methyl- oder Propylparaben als Antiseptikum, einen Farbstoff und Kirsch- oder Orangenessenz als Geschmackstoff enthalten.
- Eine sterile Zusammensetzung für Injektionszwecke kann auf herkömmliche Weise hergestellt werden, indem man beispielsweise den Wirkstoff in einem Träger, wie destilliertem Wasser für Injektionszwecke, zusammen mit einem natürlichen pflanzlichen Öl, wie Sesamöl, Kokosöl, Erdnußöl oder Baumwollsamenöl, oder einem synthetischen Fettträger, wie Ethyloleat, löst oder suspendiert. Gegebenenfalls können ein Puffer, ein Antiseptikum, ein Antioxidationsmittel oder dergl. der Zusammensetzung einverleibt werden.
- Nachstehend wird die Erfindung unter Bezugnahme auf die folgenden Beispiele näher erläutert.
- 20 ml einer Dichlorethanlösung von 21,0 g (200 mMol) Triethylamin wurden tropfenweise unter Eiskühlung zu 80 ml einer Dichlorethanlösung mit einem Gehalt an 22,1 g (100 mMol) 2-Nitrobenzolsulfonylchlorid und 20,4 g (100 mMol) 2- Bromethylaminhydrobromid gegeben. Nach Beendigung der tropfenweisen Zugabe wurde das gesamte Gemisch über Nacht bei Raumtemperatur gerührt. Nach der Umsetzung wurde das Reaktionsgemisch zweimal mit 100 ml 1 n Salzsäurelösung und sodann einmal mit 100 ml Wasser gewaschen. Sodann wurde die organische Phase mit wasserfreiem Natriumsulfat getrocknet.
- Nach Eindampfen des Lösungsmittels unter vermindertem Druck erhielt man die vorstehende Titelverbindung (a) in kristalliner Form.
- Ausbeute: 29,5 g (95,4 mMol), 95%.

- Ein Gemisch mit einem Gehalt an 9,27 g (30 mMol) (2-Nitrobenzolsulfonyl)-aminoethylbromid, 7,31 g (30 mMol) 4- Fluorbenzoylpiperidin-hydrochlorid, 8,48 g (80 mMol) Natriumcarbonat und 100 ml Methylisobutylketon wurde über Nacht in einem Ölbad gerührt.
- Nach der Umsetzung wurde das Reaktionsgemisch mit 50 ml Wasser gewaschen, sodann wurde die organische Phase isoliert und das Lösungsmittel unter vermindertem Druck abgedampft.
- Nach säulenchromatographischer Reinigung an Kieselgel (Elutionsmittel: Chloroform/Methanol = 5/1) wurde das erhaltene Produkt mit 4 n Chlorwasserstoff/Dioxan-Lösung in das Hydrochlorid übergeführt.
- Ausbeute: 10,5 g (22,2 mMol), 76%.

- Eine Suspension von 1-[2-(2-Nitrobenzolsulfonyl)-aminoethyl]-4-(4-fluorbenzoyl)-piperidin (vergl. Beispiel 1) in 160 ml Essigsäure wurde auf 90ºC erwärmt. Sodann wurden innerhalb von 10 Minuten 8 g Eisen (Pulver) allmählich zugegeben. Die Reaktionslösung wurde dunkel. Anschließend wurde weitere 10 Minuten gerührt, wobei weiße Kristalle ausfielen. Nach dem Abkühlen wurde die Reaktionslösung abfiltriert. Der Rückstand wurde mit Methanol gewaschen. Nach dem Abdampfen des Lösungsmittels aus dem erhaltenen Filtrat wurde der Rückstand mit einer 4 n Chlorwasserstoff/Dioxan-Lösung versetzt. Man erhielt die Titelverbindung in Form des Hydrochlorids.
- Ausbeute: 65,8 g (13,8 mMol), 71%.

- 30 ml einer Chloroformlösung mit einem Gehalt an 3,65 g (9,00 mMol) 1-[2-(2-Aminobenzolsulfonyl)-aminoethyl]-4-(4- fluorbenzoyl)-piperidin, 0,16 g (0,90 mMol) 4-Piperidinopyridin und 0,91 g (9,00 mMol) Triethylamin wurden tropfenweise mit 0,976 g (9,00 mMol) Chlorameisensäureethylester versetzt. Nach beendet er Umsetzung wurde das Reaktionsgemisch 3 Stunden gerührt und sodann mit 50 ml 1 n Salzsäure und einmal mit 50 ml Wasser in der angegebenen Reihenfolge extrahiert. Sodann wurde die erhaltene organische Phase mit wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde abgedampft und der Rückstand durch Säulenchromatographie (Elutionsmittel: Essigsäureethylester/Ethanol = 20/1) gereinigt. Man erhielt die Titelverbindung in Form eines öligen Produkts.
- Ausbeute: 3,18 g (6,67 mMol), 74,1%.

- 20 ml einer Pyridinlösung von 1,10 g (2,30 mMol) 4-[2- (2-Ethoxycarbonylaminobenzolsulfonyl)-aminoethyl]-1-(4-fluorbenzoyl)-piperidin (vergl. Beispiel 3) wurden 15 Stunden unter Rückfluß erwärmt. Nach Beendigung der Umsetzung wurde das Lösungsmittel abgedampft. Der Rückstand wurde zur Kristallisation mit Dichlorethan-Ether versetzt. Die erhaltenen Kristalle wurden abfiltriert, mit Ether gewaschen und sodann unter vermindertem Druck getrocknet. Man erhielt die Titelverbindung.
- Ausbeute: 0,31 g (0,72 mMol), 32%.

- Ein mit einem Argonballon und zwei Tropftrichtern ausgerüsteter Dreihalskolben wurde mit Argon gespült und sodann mit 6,6 g (90,17 mMol) Natriumamid versetzt. Nach erneutem Spülen mit Argon wurden 30 ml 1,2-Dimethoxyethan zugesetzt. 25 g (0,14 Mol) 3,4-Dimethoxyphenylacetonitril wurden in einen weiteren Kolben gegeben, der sodann mit Argon gespült und mit 25 ml 1,2-Dimethoxyethan versetzt wurde. Die Lösung wurde mit einem Zylinder in den Tropftrichter gebracht. Die 3,4-Dimethoxyphenylacetonitrillösung wurde tropfenweise unter Rühren und Eiskühlung zur Natriumamidsuspension gegeben. Sodann wurde das Eisbad entfernt und das Produkt eine weitere Stunde gerührt. Anschließend wurde die Reaktionslösung wieder in das Eisbad gebracht und tropfenweise mit einer 1,2-Dimethoxyethanlösung (17 ml) mit einem Gehalt an 17,4 ml (0,19 Mol) Isopropylbromid versetzt. Anschließend wurde das gesamte Produkt über Nacht bei Raumtemperatur gerührt. Sodann wurde das Reaktionsgemisch in Eiswasser gegossen, mit Ether extrahiert und mit 1 n wäßriger Natriumhydroxidlösung, Wasser, wäßriger 5% Citronensäurelösung und Wasser in der angegebenen Reihenfolge gewaschen. Die Etherphase wurde mit wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck abgedampft. Man erhielt 29,11 g eines öligen Produkts. Eine geringe Menge an Ethanol wurde zur Kristallisation zugesetzt. Das Produkt wurde umkristallisiert. Man erhielt 2-(3,4-Dimethoxyphenyl)-2-isopropylacetonitril.
- Ausbeute: 19,2 g (8,77 mMol), 63%.
- 0,32 g (8,2 mMol) Natriumamid wurden in einen mit Argon gespülten Dreihalskolben gegeben. Nach erneutem Spülen mit Argon wurden 5 ml 1,2-Dimethoxyethan zugesetzt. 1,49 g (6,8 mMol) 2-(3,4-Dimethoxyphenyl)-2-isopropylacetonitril wurden in einen weiteren Kolben gegeben, der sodann mit Argon gespült und mit 10 ml 1,2-Dimethoxyethan versetzt wurde. Dieses Produkt wurde mittels eines Zylinders tropfenweise in die vorstehend hergestellte Natriumamidsuspension gegeben. Nach einiger Zeit wurde das Reaktionsgemisch weiß und trüb. Anschließend wurde das Gemisch mit einer Spritze entnommen und tropfenweise zu 1,2-Dimethoxyethan (50 ml) mit einem Gehalt an 2,5 ml (26,3 mMol) 1-Brom-3-chlorpropan, das in einem weiteren, mit Argon gespülten Kolben hergestellt worden war, gegeben. Das gesamte Produkt wurde über Nacht bei Raumtemperatur gerührt. Anschließend wurde das Reaktionsgemisch mit Wasser versetzt, mit Chloroform extrahiert und mit wasserfreiem Magnesiumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels unter vermindertem Druck erhielt man 2,12 g eines Rohprodukts. Durch Dünnschichtchromatographie (SiO&sub2;/CHCl&sub3;) wurde im Reaktionsgemisch nicht-umgesetztes Ausgangsmaterial festgestellt, jedoch wurde dieses Reaktionsgemisch ohne weitere Reinigung in der nächsten Stufe eingesetzt.
- 50 ml Toluol und 2,12 g rohes 5-Chlor-2-(3,4-dimethoxyphenyl)-2-isopropylvaleronitril wurden zu einem Gemisch aus 2,6 g (10,7 mMol) 4-(4-Fluorbenzoyl)-piperidin-hydrochlorid und 0,9 g (8,5 mMol) wasserfreiem Natriumcarbonat gegeben und unter Rückfluß erwärmt. Die Umsetzung wurde dünnschichtchromatographisch verfolgt. Die Erwärmung wurde zum gegebenen Zeitpunkt abgebrochen. Das Reaktionsgemisch wurde mit Wasser versetzt und sodann mit Chloroform extrahiert. Nach Trocknen mit wasserfreiem Magnesiumsulfat wurde das Lösungsmittel unter vermindertem Druck abgedampft. Das erhaltene Rohprodukt wurde durch Kieselgel-Säulenchromatographie (CHCl&sub3;/MeOH) gereinigt. Man erhielt die Titelverbindung.

- 2,2 ml (20 mMol) Thionylchlorid wurden tropfenweise zu einer Methylenchloridlösung (30 ml) mit einem Gehalt an 3 g (20 mMol) 4-Phenyl-1-butanol gegeben und über Nacht bei Raumtemperatur gerührt. Die Reaktionslösung wurde mit Wasser, 50 % wäßriger Bicarbonatlösung, Wasser, 1 n Salzsäure und Wasser in der angegebenen Reihenfolge gewaschen. Anschließend wurde das Lösungsmittel mit wasserfreiem Magnesiumsulfat getrocknet. Nach Destillation unter vermindertem Druck erhielt man 3,62 g rohes 4-Phenylbutylchlorid. Dieses Produkt wurde ohne weitere Reinigung bei der nächsten Umsetzung eingesetzt.
- Eine geeignete Menge an Toluol, 1,8 g (12 mMol) Natriumjodid und 31 g rohes Phenylbutylchlorid wurden zu einer Mischung aus 3 g (12,3 mMol) 4-(4-Fluorbenzoyl)-piperidin-hydrochlorid und 2,6 g (24,5 mMol) wasserfreiem Natriumcarbonat gegeben und unter Rückfluß erwärmt. Nach Umsetzung über Nacht wurde die Erwärmung abgebrochen. Das Reaktionsgemisch wurde mit Wasser versetzt und sodann mit Chloroform extrahiert. Der erhaltene Extrakt wurde mit wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde unter vermindertem Druck eingedampft. Das auf diese Weise erhaltene Rohprodukt wurde durch Kieselgel-Säulenchromatographie gereinigt. Man erhielt die Titelverbindung.

- 20 ml einer Isopropylalkohollösung von 6,25 g (50,0 mMol) 2-Aminothiophenol wurden zu 20 ml einer Isopropylalkohollösung von 2,97 g (55,0 mMol) Natriummethoxid unter einem Stickstoffstrom gegeben und vermischt. Nach 30-minütigem Rühren wurde das Reaktionsgemisch auf 0ºC gekühlt und tropfenweise mit 10 ml einer Toluollösung von 1-Brom-3-chlorpropan versetzt. Nach beendeter Umsetzung wurde das gesamte Produkt 1 Stunde bei Raumtemperatur gerührt und sodann 1 Stunde unter Rückfluß erwärmt. Sodann wurden 60 ml 1 n Natriumhydroxid zugesetzt. Anschließend wurde das Reaktionsgemisch mit 50 ml Ether extrahiert. Das Lösungsmittel wurde abgedampft. Man erhielt ein Rohprodukt der Titelverbindung.

- 10 ml einer Dichlormethanlösung von 3,33 g (20,0 mMol) Cinnamoylchlorid wurden tropfenweise zu 20 ml einer Dichlormethanlösung mit einem Gehalt an 4,02 g (20,0 mMol) 1-Chlor- 3-(2-aminophenylthio)-propan und 2,02 g (20,0 mMol) Triethylamin bei Raumtemperatur gegeben. Nach Beendigung der tropfenweisen Zugabe wurde das gesamte Produkt über Nacht gerührt. Nach zweimaligem Waschen mit 30 ml 1 n Salzsäure und einmaligem Waschen mit Wasser wurde die organische Phase mit wasserfreiem Magnesiumsulfat getrocknet. Das Lösungsmittel wurde abgedampft und der Rückstand aus Dichlormethan-Ether-Hexan umkristallisiert. Man erhielt die Titelverbindung.

- Die gleiche Reaktion wie in Beispiel 1 wurde unter Verwendung von 3,17 g (9,58 mMol) 1-Chlor-3-[2-(cinnamoylamino)phenylthio]-propan, 2,33 g (9,58 mMol) 4-(4-Chlorbenzoyl)-piperidin-hydrochlorid, 2,76 g (20,0 mMol) Kaliumcarbonat und 30 ml Isoamylalkohol als Ausgangsmaterialien durchgeführt. Das erhaltene Produkt wurde säulenchromatographisch gereinigt (Laufmittel: Chloroform/Methanol = 40/1). Man erhielt die Titelverbindung.

- Die gleiche Reaktion wie in Beispiel 1 wurde unter Verwendung von 3,73 g (18,5 mMol) 1-Chlor-3-(2-aminophenylthio)propan, 4,52 g (18,5 mMol) 4-(4-Fluorbenzoyl)-piperidin-hydrochlorid, 6,90 g (50,0 mMol) Kaliumcarbonat und 50 ml Butanol als Ausgangsmaterialien durchgeführt. Das Produkt wurde durch Kieselgel-Chromatographie (Laufmittel: Chloroform/Methanol = 40/1) gereinigt. Man erhielt die Titelverbindung.

- 1,8 g (6,7 mMol) Phosphortribromid wurden zu einer Chloroformlösung (20 ml) von 3,3 g (20 inMol) 5-Phenyl-1-pentanol gegeben und 4 Stunden unter Rückfluß erwärmt. Bei dünnschichtchromatographischer Verfolgung der Umsetzung fanden sich immer noch Ausgangsmaterialien im Reaktionsgemisch, so daß weitere 0,2 g Phosphortribromid zugesetzt wurden und weiter unter Rückfluß erwärmt wurde. Anschließend wurde die Reaktionslösung mit Wasser gewaschen und mit wasserfreiem Magnesiumsulfat getrocknet. Nach dem Abdampfen des Lösungsmittels unter vermindertem Druck erhielt man 4,30 g rohes 5-Phenylpentylbromid.
- 50 ml Isopropylalkohol und 2,26 g rohes 5-Phenylpentylbromid wurden zu einer Mischung aus 2,4 g (9,9 mMol) 4-(4- Fluorbenzoyl)-piperidin-hydrochlorid und 2,1 g (19,8 mMol) wasserfreiem Natriumcarbonat gegeben und unter Rückfluß erwärmt. Die Umsetzung wurde über Nacht fortgesetzt. Anschließend wurde der Erwärmungsvorgang abgebrochen. Das Reaktionsgemisch wurde unter vermindertem Druck eingeengt und mit Wasser versetzt. Sodann wurde mit Chloroform extrahiert. Der erhaltene Extrakt wurde mit wasserfreiem Magnesiumsulfat getrocknet. Das nach dem Abdampfen des Lösungsmittels unter vermindertem Druck erhaltene Rohprodukt wurde durch Kieselgel-Säulenchromatographie gereinigt. Man erhielt die Titelverbindung.

- Männliche SD-Ratten (Alter 8-12 Wochen, Gewicht etwa 300 g) wurden getötet. Zur Entnahme der Aorta wurde ein Abdomenschnitt durchgeführt. Die Aorta wurde in 2,5 Stücke geschnitten und in ein Magnusrohr eingehängt, das bei 37±1ºC gehalten wurde und 20 ml K.S. Linger-Lösung enthielt, in die ein Gasgemisch aus 95% O&sub2; und 5% CO&sub2; eingeleitet wurde. Die Proben wurden mit einem isotonischen Transducer verbunden. Eine Aufzeichnung wurde unter einer Belastung von 1 g durchgeführt. Die Serotoninkonzentration betrug 1 · 10&supmin;&sup7; m bis 1 · 10&supmin;&sup4; m. Die Konzentration des zu testenden Produkts betrug 1 · 10&supmin;&sup4; in bis 1 · 10&supmin;³ m. Aus der Serotonin-Dosis-Reaktionskurve wurde der pA&sub2;-Wert gemäß dem Verfahren von Van Rossem et al. berechnet.
- Die Affinität des 5-HT&sub2;-Rezeptors wurde durch einen Rezeptorbindungstest auf der Basis der inikrosomalen Fraktion von Rindercortex bewertet.
- Die Ergebnisse sind in Tabelle 1 zusammengestellt.

- Sechs männliche SHR-Ratten, die ausreichend akklimatisiert waren und bei denen Hochdruck festgestellt worden war (spontan hypertensive Ratten, Gewicht 400-440 g) wurden als Testtiere verwendet.
- 1 ml physiologische Kochsalzlösung mit einem Gehalt an dem zu testenden Produkt (10 mg/kg) in 2,5% Niocol und 2,5% Ethanol wurde auf einmal intravenös injiziert. Der Blutdruck des Testtieres wurde gemäß dem geschlossenen Blutdruck-Meßverfahren (blutlos) gemessen.

- Aus den vorstehenden Ergebnissen ist ersichtlich, daß die erfindungsgemäßen Ethylaminderivate sowohl eine Serotonin-antagonistische Aktivität als auch eine antihypertensive Aktivität besitzen und daher als antihypertensive Wirkstoffe verwendet werden können.
Claims (6)
1. Verwendung eines Ethylaminderivats der Formel (I) oder
eines Salzes davon zur Herstellung eines antihypertensiv
wirksamen Arzneimittels
worin die Struktur der kombinierten Gruppe -A-B unter den folgenden ausgewählt ist:
wobei die Gruppe A durch eine der folgenden Strukturen
dargestellt ist:
worin
eine der Bindungen mit der Gruppe C in Formel I verknüpft ist
oder ein Wasserstoffatom trägt,
C Wasserstoff, Methyl, Ethyl, Propyl, Butyl, Pentyl oder
Hexyl ist oder C an A unter Bildung einer Methylen-, Ethylen-
oder Trimethylen-Brücke gebunden ist, Q unter den folgenden
Gruppen (II-VII) ausgewählt ist:
worin E ein Stickstoffatom, -NH oder substituiertes -NH, ein
Sauerstoffatom oder ein Schwefelatom bedeutet, Y, Z¹, Z², Z³

und Arylgruppen mit 6 bis 12 Kohlenstoffatomen, die mit einem
oder mehr Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder
Phosphoratomen substituiert sind, ausgewählt sind, Y
gegebenenfalls an X gebunden ist, wobei eine Brückengruppe
gebildet wird, in der die Brücke durch eine Alkylenbrücke
oder eine Alkylenbrücke, die substituiert ist mit einem oder
mehreren Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder
Phosphoratomen und/oder mindestens ein Sauerstoff-, Schwefel-
oder Stickstoffatom gebildet wird, und Z¹ und Z², Z² und Z³
oder Z³ und Z&sup4; gegebenenfalls aneinander gebunden sind, wobei
eine Brückengruppe gebildet wird, in der die Brücke durch
eine Alkylenbrücke oder eine Alkylenbrücke, die substituiert
ist mit einem oder mehr Halogen-, Sauerstoff-, Schwefel-,
Stickstoff- oder Phosphoratomen und/oder mindestens ein
Sauerstoff-, Schwefel- oder Stickstoffatom gebildet wird
X eine unter den folgenden (VIII) bis (XI) ausgewählte
organische Gruppe ist:
worin n eine ganze Zahl von 0 bis 9 bedeutet, Z&sup5; und Z&sup6;
unabhängig voneinander für Wasserstoff, Alkyl oder Aralkyl

20 Kohlenstoffatomen ist, die substituiert ist mit einem oder
mehr Heteroatomen und/oder mit einem oder mehr Substituenten,
die gegebenenfalls ein oder mehr Heteroatome enthalten, und
Z&sup8; Wasserstoff, eine Cyan-, Amino-, Nitro-, Carboxyl-, Ester-
oder Aminocarbonylgruppe ist.
2. Verwendung nach Anspruch 1, wobei Z¹, Z², Z³ und Z&sup4;
unabhängig voneinander unter Wasserstoffatomen,
Halogenatomen, Hydroxyl-, Amino-, Hydroxyamino-, Nitro-,
Cyan-, Alkyl-, Aralkyl-, Aryl-, Alkoxy-, Aralkyloxy-,
Aryloxy-, Alkylamino-, Aralkylamino-, Arylamino-, Acylamino-,
Acyloxy-, Aminosulfonylgruppen ausgewählt sind und irgendeine
dieser Alkyl-, Aralkyl-, Aryl-, Alkoxy-, Aralkyloxy-,
Aryloxy-, Alkylamino-, Aralkylamino-, Arylamino-, Acylamino-,
Acyloxy- und Aminosulfonylgruppen mit einem oder mehr
Halogen-, Sauerstoff-, Schwefel-, Stickstoff- oder
Phosphoratomen substituiert ist.

Ethylaminderivat zusammen mit einem geeigneten Trägermaterial
verwendet wird.
6. Verwendung nach einem der Ansprüche 1 bis 4, wobei das
Ethylaminderivat zusammen mit mindestens einem der folgenden
verwendet wird: einem Diureticum, einem Calcium-Antagonisten,
einem β-Blocker, einem α-Blocker und einem Konversions-Enzym-
Inhibitor (converting enzyine inhibitor) verwendet wird.
