Description
- Die Erfindung bezieht sich auf antimikrobiell wirkende Zusammensetzungen, enthaltend das Magnesiumsulfataddukt von 2,2'-Dithiobis-pyridin-1,1'-dioxid und ein wasserlösliches Zinksalz; genauer bezieht sie sich auf antimikrobielle Zusammensetzungen, enthaltend ein wasserlösliches, nicht ionisches Pyrithionderivat chemisch bekannt als das Magnesiumsulfataddukt von 2,2'-Dithiobis-pyridin-1,1'-dioxid (im folgenden als "Magnesiumsulfataddukt" bezeichnet) und ein wasserlösliches Zinksalz. Insbesondere wurde beobachtet, daß die Gegenwart eines wasserlöslichen Zinksalzes die antimikrobielle Wirksamkeit des Magnesiumsulfataddukts gegen bestimmte Typen von Mikroorganismen, wie etwa Pseudomonas aeruginosa, zu einem unerwarteten Ausmaß zu erhöhen scheint.
- US-A-41 61 526 offenbart die Vermeidung oder die Entfernung der Verfärbung in Pyrithion, Pyrithionsalz oder Dipyrithion- Zusammensetzungen, unter Verwendung eines Zinksalzes. Aus US- A-38 90 434 sind Haarshampoozusammensetzungen enthaltend Addukte von bis-(-2-Pyridyl-1-oxid) disulfid bekannt, die antimikrobielle Effekte zeigen.
- Die antimikrobiellen Zusammensetzungen nach der vorliegenden Erfindung können einverleibt werden in verschiedene nützliche therapeutische Zusammensetzungen und Reinigungszusammensetzungen, wie chirurgische Einreibungszusammensetzungen, Hautdesinfektionsmittel, Mundspülungen, Deodorants und Krankenhausreinigern.
- Das Magnesiumsulfataddukt nach der vorliegenden Erfindung ist ein wohl bekanntes, breitwirkendes antimikrobielles Agens. Zum Beispiel eine gut bekannte, kommerziell verfügbare Form wird von Olin Chemicals, Stanford, Conn., USA, unter dem Handelsnamen "OMADINE MDS", in der Trihydratform, verkauft.
- Während das Magnesimsulfataddukt, angewendet nach der vorliegenden Erfindung, eine breite antimikrobielle Aktivität hat, mangelt es an der gewünschten Aktivität gegen Pseudomonas aeruginosa, der Mikroorganismus, der in Verbindung mit Infektionen auftritt, die oft auf schwere Hautverbrennungen folgen.
- Eine Aufgabe der vorliegenden Erfindung ist es, eine antimikrobielle Zusammensetzung zur Verfügung zu stellen, in der eine verstärkte antimikrobielle Wirkung des Magnesiumsulfataddukts gegen bestimmte Typen von Mikroorganismen erzeugt wird.
- Eine andere Aufgabe der vorliegenden Erfindung ist es, therapeutische Zusammensetzungen und Reinigungszusammensetzungen zur Verfügung zu stellen, die diese antimikrobielle Zusammensetzung beinhalten.
- Die vorliegende Erfindung stellt eine antimikrobielle Zusammensetzung zur Verfügung, die dadurch gekennzeichnet ist, daß sie das Magnesiumsulfataddukt des 2,2'-Dithiobis-pyridin- 1,1'-dioxid und ein wasserlösliches Zinksalz in einem Anteil, der einen synergistischen Effekt gegen Pseudomonas aeruginosa zeigt, beinhaltet.
- Das bevorzugte Zinksalz, gebraucht in Übereinstimmung mit der vorliegenden Erfindung ist Zinkchlorid (ZnCl&sub2;).
- Andere wasserlösliche Zinksalze, die gebraucht werden können, sind Zinkacetat, Zinksulfat, Zinknitrat und Zinkphenylsulfonat.
- Im allgemeinen ist gefunden worden, daß, um die gewünschte Steigerung der antimikrobiellen Aktivität gemäß vorliegender Erfindung zu erhalten, das Zinksalz in einer Menge von 1 bis 10 Gewichtsteilen auf 1 Gewichtsteil des Magnesiumsulfataddukts, bevorzugt 1 Gewichtsteil auf 1 Gewichtsteil des Magnesiumsulfataddukts vorliegen sollte.
- Wenn die vorliegende Zusammensetzung angewendet wird für therapeutische- und/oder für Reinigungszusammensetzungen, hat das Magnesiumsulfataddukt im allgemeinen einen Anteil von 0,1 bis 1,5 Gewichtsprozent und das Zinksalz einen Anteil von 0,1 bis 1 Gewichtsprozent, bezogen auf die ganze Zusammensetzung.
- Es ist gefunden worden, daß eine sehr nützliche antimikrobielle Zusammensetzung, die bei Anwendung der vorliegenden Erfindung erhalten werden kann, eine chirugische Einreibungszusammensetzung ist, die die vorliegende antimikrobielle Zusammensetzung in einer wasserfreien, schaumfähigen Grundzusammensetzung verwendet. Eine solche chirurgische Einreibungszusammensetzung zeigte sich nützlich beim Abtöten des Pseudomonas aeruginosa Bakteriums.
- Die wasserfreie, schaumfähige Grundzusammensetzung enthält Petrojelly, Mineralöl und ein mildes Detergens (Natriumcocoyli-sethionat).
- Um die vorliegende Erfindung durch spezifische Beispiele zu verdeutlichen, sind eine Reihe von Zusammensetzungen, die das Magnesiumsulfataddukt und Zinkchlorid enthalten, in Übereinstimmung mit der vorliegenden Erfindung bezüglich ihrer antimikrobiellen Aktivität untersucht worden und verglichen worden mit Kontrollzusammensetzungen, die zum einen das Magnesiumsulfataddukt und/oder Zinkchlorid enthalten.
- Diese Zusammensetzungen sind gezeigt in Tabelle 1 mit der anti-mikrobiellen Aktivität bestimmt durch den "Zone of inhibition test".
- Der "Zone of inhibition test" ist die Beziehung zwischen einer Standardanwendung einer Testmischung auf einer festen Agaroberfläche und der aus dem inhibitierten Wachstum resultierenden Zone eines Testorganismus, aufgetragen auf die Agaroberfläche. Je größer die Zone der Wachtumsinhibierung, um so größer ist die antimikrobielle Aktivität. Diese Testmethode wird gebraucht, um die antimikrobielle Aktivität in Flüssigkeiten und Feststoffen zu bestimmen.
- Die Zusammensetzungen von Tabelle 1, die die vorliegende Erfindung veranschaulichen sind: TABELLE 1 SYNERGISTISCHE EFFEKTE VON ZnCL&sub2; AUF DIE AKTIVITÄT VON DEM MAGNESIUMSULFATADUKT* vs. PSEUDOMONAS AKTIVE INGREDIENTS UND PROZENT PHYSIKALISCHE FORM ZONEN (mm) VS. PSEUDOMONAS Beispiel A Lösung Omadine MDS @ 0.135% Aluminiumchlorohydrat (ACH) @ 17.5% Beispiel 4 Lösung Lotion Puder Omadine MDS @ 0.135% ACH @ 17.5% ZnCl&sub2; @ 0.10% nur Grundsatz * Das Magnesiumsulfataddukt wird von Olin Chemicals vertrieben unter dem Handelsnamen "OMADINE-MDS".
- Eine weitere nützliche Bestimmungsmethode zur Ermittlung der antimikrobiellen Aktivität, ist der Wert der minimalen inhibierenden Konzentration (M.I.C.). Die minimale inhibierende Konzentration ist eine serielle zweifach Verdünnung des Testansatzes in einem Brühenkulturmedium, welches angeimpft wurde mit einer standardtisierten Kultur von Mikroorganismen. Die Menge des Testagens, das das sichtbare Mikrobenwachstum inhibiert, wird bezeichnet als der minimale inhibierende Konzentrationswert (MIC). Je geringer die Menge an Testagens, um so größer die antimikrobielle Aktivität.
- Die Ergebnisse der experimentellen Analyse zeigen einen achtfachen Anstieg in der Aktivität gegen Pseudomonas aeruginosa für die Kombination von ZnCl&sub2; mit Omadine MDS verglichen mit der Aktivität von ZnCl&sub2; für sich, oder von Omadine MDS für sich.
- In weiteren Vergleichsstudien wurden Zusammensetzungen hergestellt und bewertet durch die minimale inhibierende Konzentrationsmethode und die Zone der Inhibierung. Die Ergebnisse sind ähnlich für alle Salze, untersucht bei einer 1%igen Konzentration mit Omadine MDS bei 0,5%.
- Wieder, die Zone der Inhibierung war fast nicht existent für die Kontrollen 0-2 mm und 10-14 mm für die Kombination des Magnesiumsulfataddukt und ZnCl&sub2;, Kombination angewendet gemäß vorliegender Erfindung.
- In der Tabelle 2 ist berichtet über eine chirurgische Einreibungszusammensetzung, Zusammensetzung I, unter Anwendung der obengenannte wasserfreien, schäumenden Grundzusammensetzung, beinhaltent Petrojelly, Mineralöl, Glyzerin, TiO&sub2; und Natriumcocoylisethionat und die Magnesiumsulfataddukt und ZnCl&sub2; Kombination in Übereinstimmung mit der vorliegenden Erfindung und die Kontroll-Grundzusammensetzung II. TABELLE 2 FORMEL ZUSAMMENSETZUNG I ZUSAMMENSETZUNG II Petrojelly Mineralöl Glyzerin TiO&sub2; Na Cocoylisethionat Omadine MDS ZnCl&sub2; (50%ige Lösung)
- Bei der Untersuchung der antimikrobiellen Aktivität, wurde die minimale inhibierende Konzentrationsaktivität gesteigert für Omadine MDS 2%, ZnCl&sub2; 1% in Zusammensetzung I, und ergab mit einen substantiellen Anstieg in der Aktivität im Vergleich zu Zusammensetzung II.
Claims (5)
1. Antimikrobielle Zusammensetzung, dadurch gekennzeichnet, daß
sie das Magnesiumsulfataddukt von 2'2'-Dithiobispyridin-
1,1'-dioxid und ein wasserlösliches Zinksalz in einer Menge
umfaßt, die einen synergistischen Effekt gegen Pseudomonas
areuginosa aufweist.
2. Zusammensetzung nach Anspruch 1, worin das Zinksalz
ausgewählt ist unter Zinkchlorid, Zinkacetat, Zinksulfat,
Zinknitrat und Zinkphenylsulfonat.
3. Zusammensetzung nach Anspruch 1 oder Anspruch 2, worin das
Zinksalz Zinkchlorid ist.
4. Zusammensetzung nach einem der Ansprüche 1 bis 3, worin das
Zinksalz in einer Menge von 1 bis 10 Gewichtsteilen pro Teil
des antimikrobiellen Addukts vorliegt.
5. Zusammensetzung nach einem der Ansprüche 1 bis 4, worin das
Magnesiumsulfataddukt in einer Menge von 0,1 bis 1,5 Gew.-%
und das Zinksalz in einer Menge von 0,1 bis 1 Gew.-% der
Gesamtzusammensetzung vorliegt.
