Description
Die vorliegende Erfindung betrifft 2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran
(oder benzothiopyran)-6-yl)propenyl- und 2-(4,4-Dimethyl-1,2,3,4-
tetrahydro-6-chinolinyl)propenyl-Verbindungen der Formel
in der X -O-, -S-, =SO, =SO₂ oder =NR²; R¹ Methyl,
-CH(R³)OR⁴ oder -C(O)R⁵; R² Wasserstoff oder C₁- bis C₆-
Alkyl; R³ Wasserstoff oder Methyl; R⁴ Wasserstoff,
C₁- bis C₆-Alkyl oder Alkanoyl; R⁵ Wasserstoff,
C₁- bis C₆-Alkyl, Amino, C₁- bis C₆-Alkylamino, Di-(C₁- bis C₆-
alkyl)amino oder OR⁶; und R⁶ Wasserstoff oder C₁- bis C₆-
Alkyl bedeuten;
und Salze von Verbindungen der Formel I.
Die Erfindung betrifft weiterhin die Verwendung dieser Verbindungen
zur Behandlung von Neoplasien, Akne oder Psoriasis sowie ein
Verfahren zur Herstellung der Verbindung I und deren Salzen.
Der hier verwendete Ausdruck C₁ bis C₆ bezeichnet
Gruppen mit bis zu 6 Kohlenstoffatomen. Solche
Gruppen enthalten bevorzugt 1 bis 4 Kohlenstoffatome.
Alkylgruppen können verzweigt oder unverzweigt sein,
wie beispielsweise Methyl, Äthyl, Isopropyl oder 2-Methylpropyl.
Alkanoylgruppen leiten sich z. B. von der Essig-,
Propion- oder Pivalinsäure oder auch von den höheren Carbonsäuren
mit bis zu 20 Kohlenstoffatomen, z. B. von der
Palmitin- oder Stearinsäure ab. Bevorzugte Alkanoylgruppen
sind C₁- bis C₆-Alkanoylgruppen.
Eine bevorzugte Gruppe von Verbindungen der Formel I
sind diejenigen, in denen R¹ Carboxyl, C₁- bis C₆-Alkoxycarbonyl,
Carbamoyl oder mono- oder di-Alkylcarbamoyl ist.
Die Verbindungen der Formel I können erfindungsgemäß
dadurch erhalten werden, daß man eine Verbindung der allgemeinen
Formel
in an sich bekannter Weise
mit einer Verbindung der allgemeinen Formel
in denen entweder A eine Triarylphosphoniumäthylgruppe
der Formel
darstellt, worin
Q Phenyl und Y das Anion einer organischen oder
anorganischen Säure bezeichnet, und B Formyl ist;
oder A Acetyl ist, und B eine Dialkoxyphosphinylmethylgruppe;
und R¹¹ einen Rest R¹ mit Ausnahme des
Carboxyl- und Formylrests bedeutet,
zu einer Verbindung der allgemeinen Formel I umsetzt, und
erwünschtenfalls den Rest R¹¹ funktionell abwandelt und/oder
das Schwefelatom in einer Verbindung, in der X -S-
ist, zu einer Sulfoxyl- oder Sulfonylgruppe oxidiert.
Von den anorganischen Säureanionen Y ist das Chlor-
und Brom-ion oder das Hydrosulfat-ion, von den organischen
Säureanionen ist das Tosyloxy-ion bevorzugt.
Die Alkoxyreste in B sind vornehmlich
Alkoxyreste mit 1-6 Kohlenstoffatomen, wie Methoxy
oder Äthoxy.
Die Ausgangsverbindungen der Formel II können, soweit
ihre Herstellung nicht bekannt oder nachstehend beschrieben
ist, in Analogie zu bekannten oder den nachstehend beschriebenen
Methoden hergestellt werden.
Die Umsetzung der Verbindungen der Formeln II und III
kann nach den bekannten Methoden der Wittig- bzw. Horner-
Reaktion durchgeführt werden.
Bei der Wittig-Reaktion, d. h., bei Verwendung einer
Verbindung der Formel II mit A=Triarylphosphoniumäthyl,
werden die Komponenten in Gegenwart eines säurebindenden
Mittels, z. B. in Gegenwart einer starken Base, wie z. B.
Butyllithium, Natriumhydrid oder dem Natriumsalz von
Dimethylsulfoxyd, vornehmlich aber in Gegenwart eines gegebenenfalls
durch C₁- bis C₆-Alkyl substituierten Äthylenoxyds
wie 1,2-Butylenoxyd, gegebenenfalls in einem Lösungsmittel,
z. B. in einem Äther, wie Diäthyläther oder Tetrahydrofuran
oder in einem aromatischen Kohlenwasserstoff,
wie Benzol in einem zwischen der Raumtemperatur und dem
Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich
miteinander umgesetzt.
Bei der Horner-Reaktion, d. h. bei Verwendung einer
Verbindung der Formel III mit B=Dialkoxyphosphinylmethyl,
werden die Komponenten mit Hilfe einer Base und vorzugsweise
in Gegenwart eines inerten organischen Lösungsmittels,
z. B. mit Hilfe von Natriumhydrid in Benzol, Toluol, Dimethylformamid,
Tetrahydrofuran, Dioxan oder 1,2-Dimethoxyalkan,
oder auch mit Hilfe eines Natriumalkoholates in
einem Alkanol, z. B. Natriummethylat in Methanol, in einem
zwischen 0° und dem Siedepunkt des Reaktionsgemisches
liegenden Temperaturbereich kondensiert.
Als funktionelle Abwandlung eines Substituenten R¹
bzw. R¹¹ in einer erhaltenen Verbindung der Formel I kommen
z. B. die Verseifung eines Carbonsäureesters oder dessen
Reduktion zur Hydroxymethylgruppe in Betracht. Die Hydroxymethylgruppe
kann auch zur Formylgruppe aufoxidiert oder
verestert oder veräthert werden. Eine Carboxylgruppe kann
dann weiterhin in ein Salz, einen Ester, ein Amid oder die
Hydroxymethylgruppe umgewandelt werden.
Weiterhin kann ein Carbonsäureester durch Umsetzung
mit einer Methyl-magnesiumhalogenidverbindung bei tiefer
Temperatur bzw. eine Carboxylgruppe durch Reaktion mit
Methyl-lithium in eine Acetylgruppe überführt werden. Die
Acetylgruppe läßt sich durch Reduktion mit einem komplexen Metallhydrid,
wie z. B. Natriumborhydrid, in den
sekundären Alkohol überführen, der nach bekannten Methoden
alkyliert bzw. acyliert werden kann.
Alle diese Abwandlungen können nach an sich bekannten
Methoden vorgenommen werden.
Ein Carbonsäureester der Formel I kann in an sich bekannter
Weise, z. B. durch Behandeln mit Alkalien, insbesondere
durch Behandeln mit wäßriger alkoholischer Natron-
oder Kalilauge in einem zwischen Raumtemperatur und dem
Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich
hydrolysiert und entweder über ein Säurehalogenid
oder, wie nachstehend beschrieben, unmittelbar amidiert
werden.
Eine Carbonsäure der Formel I kann in an sich bekannter
Weise, z. B. durch Behandeln mit Thionylchlorid, vorzugsweise
in Pyridin, oder Phosphortrichlorid in Toluol in das Säurechlorid
übergeführt werden, das durch Umsetzen mit Alkoholen
in Ester, mit Aminen in das entsprechende Amid umgewandelt
werden kann.
Ein Carbonsäureester der Formel I kann z. B. durch Behandeln
mit Lithiumamid direkt in das entsprechende Amid
umgewandelt werden. Das Lithiumamid wird vorteilhaft bei
Raumtemperatur mit dem betreffenden Ester zur Reaktion
gebracht.
Eine Carbonsäure oder ein Carbonsäureester der Formel
I kann in an sich bekannter Weise zu dem entsprechenden
Alkohol der Formel I reduziert werden. Die Reduktion wird
vorteilhaft mit Hilfe eines Metallhydrids oder Alkylmetallhydrids
in einem inerten Lösungsmittel durchgeführt. Als
Hydride haben sich vor allem gemischte Metallhydride, wie
Lithiumaluminiumhydrid oder bis-[Methoxy-äthylenoxy]-
natriumaluminiumhydrid als geeignet erwiesen. Als Lösungsmittel
verwendbar sind u. a. Äther, Tetrahydrofuran oder
Dioxan, wenn Lithiumaluminiumhydrid verwendet wird; und
Äther, Hexan, Benzol oder Toluol, wenn Diisobutylaluminiumhydrid
oder bis-[Methoxy-äthylenoxy]-natriumaluminiumhydrid
eingesetzt werden.
Ein Alkohol der Formel I kann z. B. in Gegenwart einer
Base, vorzugsweise in Gegenwart von Natriumhydrid, in einem
organischen Lösungsmittel wie Dioxan, Tetrahydrofuran,
1,2-Dimethoxyäthan, Dimethylformamid, oder auch in Gegenwart
eines Alkalimetallalkoholates in einem Alkanol, in
einem zwischen 0° und Raumtemperatur liegenden Temperaturbereich
mit einem Alkylhalogenid, z. B. mit Methyliodid,
veräthert werden.
Ein Alkohol der Formel I kann auch durch Behandeln mit
einem Alkanoylhalogenid oder Anhydrid, zweckmäßig in
Gegenwart einer Base, beispielsweise in Gegenwart von
Pyridin oder Triäthylamin in einem zwischen Raumtemperatur
und dem Siedepunkt des Reaktionsgemisches liegenden Temperaturbereich
verestert werden.
Die Carbonsäuren der Formel I bilden mit Basen, insbesondere
mit den Alkalimetallhydroxyden, vorzugsweise
mit Natrium- oder Kaliumhydroxyd, Salze.
Eine Verbindung der Formel I, in der X für -S- steht,
kann mit an sich bekannten Methoden zu einer Verbindung
der Formel I, in der X für =SO oder =SO₂ steht, oxidiert
werden. Die Oxidation zur Sulfoxidgruppe kann mit Oxidationsmitteln
wie Periodaten, z. B. NaIO₄ oder mit organischen
Persäuren, wie m-Chlorperbenzoesäure vorgenommen
werden. Bei der Oxidation mit organischen Persäuren setzt
man etwa ein Äquivalent Persäure ein, um eine Sulfoxidverbindung
(X=SO) zu erhalten, wogegen die Verwendung
von zwei Äquivalenten Persäure zu Sulfonen (X=SO₂) führt.
Die Verbindungen der Formel I können in trans- oder
cis-Form vorliegen. Bei der Herstellung fallen sie mehrheitlich
in der trans-Form an. Gegebenenfalls anfallende
cis-Anteile können in an sich bekannter Weise, falls erwünscht,
abgetrennt werden.
Die Verfahrensprodukte der Formel I und ihre physiologisch
verträglichen Salze stellen pharmakodynamisch wertvolle
Verbindungen dar. Sie können zur topischen und
systemischen Therapie von benignen und malignen Neoplasien,
von prämalignen Läsionen, sowie ferner auch zur systemischen
und topischen Prophylaxe der genanntne Affektion
verwendt werden.
Sie sind des weiteren für die topische und systemische
Therapie von Akne, Psoriasis und anderen mit einer
verstärkten oder pathologisch veränderten Verhornung einhergehenden
Dermatosen, wie auch von entzündlichen und
allergischen dermatologischen Affektionen geeignet. Die
Verfahrensprodukte der Formel I können ferner auch zur
Bekämpfung von Schleimhauterkrankungen mit entzündlichen
oder degenerativen bzw. metaplastischen Veränderungen eingesetzt
werden. Im Papillomtest (Europ. J. Cancer Vol. 10,
pp. 731-737, 1974) zeigt die in Beispiel 1 hergestellte
Verbindung (I, X=O, R¹=Äthoxycarbonyl) mit 12,5 mg/kg 43%
Regression der Papillome und die in Beispiel 14 hergestellte
Verbindung (I, X=NHCH₃ R¹=Äthoxycarbonyl) mit 3 mg/kg
46% Regression der Papillome.
Zur Behandlung der obenerwähnten Erkrankungen werden
die erfindungsgemäßen Verbindungen oral, zweckmäßig in
einer Dosierung beim Erwachsenen von etwa 1-100 mg pro
Tag, vorzugsweise 5-30 mg/Tag, verabreicht. Eine etwaige
Überdosierung kann sich in Form einer Vit-A-Hypervitaminose
äußern, und ist an deren Symptomen (Hautschuppung,
Haarausfall) leicht zu erkennen.
Die Dosis kann als Einzeldosis oder auf mehrere Teildosen
verteilt verabreicht werden.
Die Verbindungen der Formel I und deren Salze können
deshalb als Heilmittel, z. B. in Form pharmazeutischer
Präparate, Anwendung finden.
Die zur systemischen Anwendung dienenden Präparate
können z. B. dadurch hergestellt werden, daß man eine Verbindung
der Formel I oder ein Salz davon als wirksamen
Bestandteil nichttoxischen, inerten, an sich in solchen
Präparaten üblichen festen oder flüssigen Trägern zufügt.
Die Mittel können enteral, parenteral oder topisch
verabreicht werden. Für die enterale Applikation eignen
sich z. B. Mittel in Form von Tabletten, Kapseln, Drag´es,
Sirupen, Suspensionen, Lösungen und Suppositorien. Für
die parenterale Applikation sind Mittel in Form von Infusions-
oder Injektionslösungen geeignet.
Die Dosierungen, in denen die Präparate verabreicht
werden, können je nach Anwendungsart und Anwendungsweg
sowie nach den Bedürfnissen der Patienten variieren.
Die Präparate können in
einer oder mehreren Dosierungen verabreicht werden. Eine
bevorzugte Darreichungsform sind Kapseln mit einem Gehalt
von ca. 0,5-20 mg Wirkstoff.
Die Präparate können inerte oder auch pharmakodynamisch
aktive Zusätze enthalten. Tabletten oder Granula z. B.
können eine Reihe von Bindemitteln, Füllstoffen, Trägersubstanzen
oder Verdünnungsmitteln enthalten. Flüssige
Präparate können beispielsweise in Form einer sterilen, mit
Wasser mischbaren Lösung vorliegen. Kapseln können neben
dem Wirkstoff zusätzlich ein Füllmaterial oder Verdickungsmittel
enthalten. Des weiteren können geschmacksverbessernde
Zusätze, sowie die üblicherweise als Konservierungs-,
Stabilisierungs-, Feuchthalte- und Emulgiermittel verwendeten
Stoffe, ferner auch Salze zur Veränderung des osmotischen
Druckes, Puffer und andere Zusätze vorhanden sein.
Die vorstehend erwähnten Trägersubstanzen und Verdünnungsmittel
können aus organischen oder anorganischen
Stoffen, z. B. aus Wasser, Gelatine, Milchzucker, Stärke,
Magnesiumstearat, Talkum, Gummi arabicum und Polyalkylenglykolen
bestehen. Voraussetzung ist, daß alle bei
der Herstellung der Präparate verwendeten Hilfsstoffe
untoxisch sind.
Zur topischen Anwendung werden die Wirkstoffe zweckmäßig
in Form von Salben, Tinkturen, Cremes, Lösungen,
Lotionen, Sprays und Suspensionen verwendet. Bevorzugt
sind Salben und Cremes sowie Lösungen. Diese zur
topischen Anwendung bestimmten Präparate können dadurch
hergestellt werden, daß man die Verfahrensprodukte als
wirksamen Bestandteil nichttoxischen, inerten, für topische
Behandlung geeigneten, an sich in solchen Präparaten üblichen
festen oder flüssigen Trägern zumischt.
Für die topische Anwendung sind zweckmäßig ca. 0,1-
5%ige, vorzugsweise 0,3-2%ige Lösungen, sowie ca. 0,1-5%ige,
vorzugsweise ca. 0,3-2%ige Salben oder Cremes geeignet.
Den Präparaten kann gegebenenfalls ein Antioxydationsmittel,
z. B. Tocopherol, N-Methyl-γ-tocopheramin sowie
butyliertes Hydroxyanisol oder butyliertes Hydroxytoluol
beigemischt sein.
15,9 g [1-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-6-yl-äthyl]triphenyl
phosphoniumbromid werden mit 5,5 g 4-Äthoxycarbonylbenzaldehyd
in 150 ml Butylenoxid über Nacht am Rückfluß
erhitzt. Man kühlt ab, gießt auf 500 ml Methanol/Wasser
(6 : 4), extrahiert dreimal mit Hexan, wäscht die organische
Phase mit Wasser, trocknet über Natriumsulfat und dampft
ein. Man erhält 8,1 g eines gelblichen Öls, das durch
Filtration über Kieselgel (Elutionsmittel Hexan/Äther
9 : 1) gereinigt wird. Nach dem Umkristallisieren aus
Hexan/Äther erhält man 5,7 g p-[(E)-2-[3,4-Dihydro-4,4-
dimethyl-2H-1-benzopyran-6-yl]propenyl]benzosäureäthylester in
farblosen Kristallen, Schmelzpunkt 86-87°.
Das als Ausgangsmaterial verwendete Phosphoniumsalz
läßt sich wie folgt herstellen:
3,4 g Acetylchlorid werden in 30 ml Nitrobenzol gelöst und bei 0-5°C portionsweise mit 5,7 g Aluminiumtrichlorid versetzt. Zu diesem Gemisch tropft man bei 0-5°C eine Lösung von 6,9 g 4,4-Dimethylchroman in 15 ml Nitrobenzol. Nach 1stündigem Rühren gießt man auf Eiswasser, extrahiert mit Äther, wäscht die organische Phase mit 1N NaOH und einer gesättigten Kochsalzlösung, trocknet über Natriumsulfat und dampft ein. Das zurückbleibende schwarze Öl wird im Hochvakuum destilliert. Man erhält 5,1 g 4,4-Dimethyl-6-acetylchroman als bläuliches Öl, Siedepunkt 110°C/2,66 Pa.

20,4 g 4,4-Dimethyl-6-acetyl-chroman werden in 400 ml
Methanol gelöst und unter Eiskühlung portionsweise mit
3,3 g Natriumborhydrid versetzt. Man rührt über Nacht bei
Raumtemperatur, gießt auf Eis/Wasser und extrahiert mit
Äther. Nach dem Waschen mit Wasser, Trocknen der organischen
Phase mit Natriumsulfat und Abdampfen des Lösungsmittels
erhält man 19,4 g eines braunen Öls, das durch
Filtration über Kieselgel (Elutionsmittel Hexan/20%
Äther) weiter gereinigt werden kann. Man erhält 18,2 g
4,4-Dimethyl-6-(1-hydroxyäthyl)chroman als farbloses Öl.
6,2 g 4,4-Dimethyl-6-(1-hydroxyäthyl)chroman werden
in 3 ml abs. Äther und 20 ml Hexan gelöst und mit 3 Tropfen
Pyridin versetzt. Bei einer Temperatur von 0-5°C
tropft man eine Lösung von 5,4 g Phosphortribromid in
20 ml Hexan langsam hinzu. Man rührt noch 3 Stunden bei
0°C, gießt auf Eis und extrahiert mit Äther. Die organische
Phase wird mit verdünnter Natriumbicarbonat-Lösung
und Wasser gewaschen, über Natriumsulfat getrocknet und
eingedampft. Man erhält 4,5 g 4,4-Dimethyl-6-(1-bromäthyl)chroman
als rötliches Öl. Es wird ohne weitere
Reinigung in das Phosphoniumbromid übergeführt.
4,6 g 4,4-Dimethyl-6-(1-bromäthyl)chroman werden in
40 ml Xylol gelöst und mit 5,3 g Triphenylphosphin versetzt.
Die Mischung wird über Nacht auf 100°C erhitzt, abgekühlt,
die ausgefallenen Kristalle abgenutscht und mit
Hexan gewaschen. Zur weiteren Reinigung kann das Phosphoniumsalz
in Methylenchlorid gelöst werden und durch Zugabe
von Essigester erneut ausgefällt werden. Man erhält
3,5 g [1-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-6-yl-äthyl]
triphenylphosphoniumbromid in farblosen Kristallen, Schmelzpunkt
142-148°.
4,0 g p-[(E)-2-[4,4-Dimethyl-6-chromanyl]propenyl]benzoesäureäthylester
werden in 80 ml Äthanol gelöst und mit einer
Lösung von 5,6 g Kaliumhydroxid in 20 ml Wasser und 20 ml
Äthanol versetzt. Nach 3stündigem Rühren bei 50°C kühlt
man ab, gießt auf Eiswasser, säuert mit 2N Schwefelsäure
an und extrahiert mehrfach mit Essigester. Die organische
Phase wird mit Wasser gewaschen, über Natriumsulfat getrocknet
und eingedampft. Nach dem Umkristallisieren des
Rückstandes aus Methylenchlorid/Hexan erhält man 3,4 g
p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-6-yl)
propenyl]benzoesäure in farblosen Kristallen, Schmelzpunkt
196-197°C.
4,0 g p-[(E)-2-[3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-6-yl]propenyl]
benzoesäureäthylester werden in 50 ml Äther
gelöst und bei 0°C
langsam mit 25,2 ml einer Diisobutylaluminiumhydrid-Lösung
(20% in Toluol) versetzt. Nach einstündigem Rühren bei 0°C
tropft man vorsichtig 25 ml eines Methanol/Wasser-Gemisches
(1 : 1) hinzu, rührt 30 Minuten bei Raumtemperatur und filtriert
den Niederschlag ab. Das Filtrat wird getrocknet,
über eine kurze Säule mit Kieselgel (Elutionsmittel
Hexan/Äther=1 : 1) filtriert und eingedampft. Nach dem
Umkristallisieren aus Hexan/Äther erhält man 2,9 g p-[(E)-
2-(3,4-Dihydro-4,4-dimethyl-1-benzopyran-6-yl)propenyl]benzylalkohol-
in farblosen Kristallen, Schmelzpunkt 80-81°C.
105 mg einer 50%igen Dispersion von Natriumhydrid in
Mineralöl werden dreimal mit Pentan gewaschen, im Vakuum
getrocknet und in 10 ml Dimethylformamid suspendiert. Bei
0°C tropft man eine Lösung von 500 mg p-[(E)-2-(3,4-Dihydro-
4,4-dimethyl-1-benzopyran-6-yl)propenyl]benzylalkohol in
5 ml Dimethylformamid hinzu. Nach 30 Minuten gibt man 400 mg
Äthyliodid zu und rührt 2 Stunden bei Raumtemperatur. Man
gießt auf Eiswasser, extrahiert mit Äther, trocknet und
dampft ein. Umkristallisation des Rohproduktes aus Hexan
ergibt 150 mg 6-[(E)-p-(Äthoxymethyl]-α-methylstyryl]-3,4-dihydro-4,4-dimethyl-2H-1-benzopyran,
Schmelzpunkt 59-60°C.
0,5 g p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-1-benzopyran-
6-yl)propenyl]benzylalkohol werden in 20 ml Methylenchlorid
gelöst und mit 10 g Mangandioxid versetzt. Nach 6stündigem
Rühren bei Raumtemperatur filtriert man ab, wäscht gut mit
Methylenchlorid nach und dampft das Filtrat ein. Nach Umkristallisation
des Rückstandes aus Hexan/Äther erhält man
350 mg p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-
6-yl)propenyl]benzaldehyd, Schmelzpunkt 102-104°C.
0,5 g p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-
6-yl)propenyl]benzoesäure werden in 10 ml Tetrahydrofuran
gelöst und mit 0,29 g 1,1′-Carbonyldiimidazol versetzt.
Nach 2stündigem Rühren bei Raumtemperatur erhält
man eine klare Lösung, in welche unter Eiskühlung während
1 Stunde trockenes Äthylamin eingeleitet wird. Man gießt
anschließend auf Eis, säuert mit 2N Schwefelsäure an, extrahiert
mit Essigester, trocknet und dampft ein. Das Rohprodukt
wird über eine Säule mit Kieselgel filtriert (Elutionsmittel
Hexan/Essigester=3 : 1) und aus Essigester/Hexan
umkristallisiert. Man erhält 0,4 g p-[2-(3,4-Dihydro-
4,4-dimethyl-2H-1-benzopyran-6-yl)propenyl]-N-äthyl-benzamid,
Schmelzpunkt 161-163°C.
0,5 g p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-
6-yl)propenyl]benzoesäure werden in 10 ml Tetrahydrofuran
gelöst. Unter Eiskühlung tropft man langsam 2,3 ml
einer 1,6molaren Lösung von Methyllithium in Äther hinzu.
Nach 30minütigem Rühren bei Raumtemperatur gießt man auf
Eis, extrahiert mit Äther, trocknet und dampft ein. Nach
Umkristallisation des Rückstandes aus Hexan/Äther erhält
man 370 mg 4′-[2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran-
6-yl)propenyl]acetophenon, Schmelzpunkt 109-110°C.
37 g Aluminiumchlorid werden bei 0°C in 200 ml Methylenchlorid
suspendiert und unter kräftigem Rühren langsam
mit 22 g Acetylchlorid versetzt. Nach 30 Minuten tropft man
eine Lösung von 49,4 g 3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran
in 150 ml Methylenchlorid hinzu. Nach 1stündigem
Rühren bei 0°C gießt man auf Eiswasser und extrahiert mit
Methylenchlorid. Die organische Phase wird mit 1N Natriumhydroxid
und Wasser neutral gewaschen, getrocknet und eingedampft.
Das ölige Rohprodukt wird über eine kurze Säule
mit Kieselgel filtriert (Elutionsmittel Hexan/Äther=
9 : 1). Man erhält 51,5 g 3,4-Dihydro-4,4-dimethyl-2H-1-
benzothiopyran-6-yl-methylketon als farbloses Öl, das
beim Stehen im Kühlschrank erstarrt.
2,6 g einer 50%igen Dispersion von Natriumhydrid in
Mineralöl werden dreimal mit Pentan gewaschen, im Vakuum
getrocknet und in 100 ml Dimethylformamid suspendiert. Bei
Raumtemperatur tropft man eine Lösung von 16,4 g Diäthyl-
4-äthoxycarbonyl-benzylphosphonat in 100 ml Dimethylformamid
hinzu. Nach 30minütigem Rühren bei Raumtemperatur
tropft man eine Lösung von 10 g 3,4-Dihydro-4,4-dimethyl-
2H-1-benzothiopyran-6-yl-methylketon in 80 ml Dimethylformamid
hinzu und erwärmt anschließend 2,5 Stunden auf
70°C. Nach dem Abkühlen gießt man auf Eiswasser, säuert
mit 2N Salzsäure an und extrahiert mit Äther. Die organische
Phase wird mehrfach mit Wasser gewaschen, getrocknet
und eingedampft. Man erhält ein gelbes Öl, das nach Chromatographie
an Kieselgel (Elutionsmittel Hexan/Essigester=
4 : 1) und Kristallisation aus Hexan 9,8 g p-[(E)-2-
(3,4-Dihydro-4,4-dimethyl-2H-1-benzothiopyran-6-yl)propenyl]benzoesä-ureäthylester
als schwach gelbliche Kristalle ergibt,
Schmelzpunkt 91-92°C.
4,0 g p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-
benzothiopyran-6-yl)propenyl]-benzoesäureäthylester werden in 100 ml
Chloroform gelöst. Unter Rühren tropft man bei 0-5°C eine
Lösung von 5 g m-Chlorperbenzoesäure (ca. 90%) in 100 ml
Chloroform langsam hinzu und rührt über Nacht bei 0-5°C
nach. Das Reaktionsgemisch wird mit Chloroform verdünnt,
zweimal mit verdünnter Sodalösung und Wasser gewaschen,
getrocknet und eingedampft. Man erhält nach Filtration des
Rohproduktes über Kieselgel (Elutionsmittel Hexan/Essigester
=3 : 1) und Kristallisation aus Hexan/Essigester 2,3 g
p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzo
thiopyran-6-yl)propenyl]benzoesäureäthylester-1,1-dioxid in
farblosen Kristallen, Schmelzpunkt 142-144°C.
In Analogie zu Beispiel 10 erhält man aus 4,0 g
p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzothiopyran-6-
yl)propenyl]benzoesäureäthylester und 2,5 g m-Chlorperbenzoesäure nach
Filtration über Kieselgel (Elutionsmittel Hexan/Essigester
=1 : 2) und Kristallisation aus Hexan/Essigester
2,8 g p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-
1-benzothiopyran-6-yl)propenyl]benzoesäureäthylester-1-oxid,
Schmelzpunkt 97-99°C.
In Analogie zu Beispiel 1 erhält man aus [1-(3,4-Dihydro-
4,4-dimethyl-2H-1-benzothiopyran-6-yl-äthyl]-triphenylphosphoniumbro-mid
und 4-Äthoxycarbonylbenzaldehyd den p-[(E)-
2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzothiopyran-6-yl-
propenyl]benzoesäureäthylester.
Das als Ausgangsmaterial verwendete Phosphoniumsalz
kann ausgehend von 4,4-Dimethylthiochroman in Analogie
zu der in Absatz 2-4 von Beispiel 1 beschriebenen Weise
hergestellt werden.
In Analogie zu Beispiel 1 erhält man aus 1-(4,4-
Dimethyl-1,2,3,4-tetrahydro-6-chinolinyl]-äthyl]-triphenyl-
phosphoniumbromid und 4-Äthoxycarbonylbenzaldehyd den
p-[(E)-2-(4,4-Dimethyl-1,2,3,4-tetrahydro-6-chinolinyl]
propenyl]benzoesäureäthylester.
Das als Ausgangsmaterial verwendete Phosphoniumsalz
kann ausgehend von 4,4-Dimethyl-1,2,3,4-tetrahydrochinolin
in Analogie zu der in Absatz 2-4 von Beispiel beschriebenen
Weise hergestellt werden.
In Analogie zu Beispiel 1 erhält man aus [1-(1,4,4-
Trimethyl-1,2,3,4-tetrahydro-6-chinolinyl)äthyl]triphenyl
phosphoniumbromid und 4-Äthoxycarbonylbenzaldehyd den
p-[(E)-2-(1,4,4-Trimethyl-1,2,3,4-tetrahydro-6-
chinolinyl)propenyl]benzoesäureäthylester.
Das als Ausgangsmaterial verwendete Phosphoniumsalz
kann ausgehend von 1,4,4-Trimethylchinolin in Analogie
zu der in Absatz 2-4 von Beispiel 1 beschriebenen Weise
hergestellt werden.
Durch Behandlung des p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-
2H-1-benzopyran-6-yl)propenyl]benzylalkohols mit Acetylchlorid und
Pyridin erhält man das p-[(E)-2-(3,4-Dihydro-4,4-dimethyl-2H-
1-benzopyran-6-yl)propenyl]benzylacetat.
74,5 mg Natriumhydrid (50% in Mineralöl) werden mit
absolutem Pentan gewaschen, am Wasserstrahlvakuum getrocknet
und in 5 ml absolutem Dimethylformamid suspendiert. Bei
Raumtemperatur tropft man eine Lösung von 465 mg Diäthyl-4-
äthoxycarbonyl-benzylphosphonat in 5 ml absolutem Dimethylformamid
hinzu. Nach 10minütigem Rühren gibt man eine
Lösung von 410 mg 6-Acetyl-1,2,3,4-tetrahydro-1,4,4-
trimethyl-chinolin in 5 ml Dimethylformamid hinzu und erwärmt
1 Stunde auf 70°C, anschließend weitere 2 Stunden auf
90°C. Nach dem Abkühlen gießt man das Reaktionsgemisch auf
Eis, extrahiert mehrfach mit Äther, wäscht mit Wasser,
trocknet über Natriumsulfat und dampft ein. Nach Chromatographie
des Rohproduktes an Kieselgel (Elutionsmittel
Hexan/Äther=4 : 1) erhält man p-[(E)-2-(1,2,3,4-
Tetrahydro-1,4,4-trimethyl-6-chinolinyl)propenyl]benzoesäureäthylest-er
in blaßgelben Kristallen, Schmelzpunkt 100-101°C.
Das als Ausgangsmaterial verwendete 6-Acetyl-1,2,3,4-
tetrahydro-1,4,4-trimethyl-chinolin läßt sich wie folgt
herstellen:
21,5 g 3,4-Dihydro-1,4,4-trimethyl-2(1H)-chinolinon werden in 43 g Schwefelkohlenstoff gelöst und mit 85 g Aluminiumchlorid versetzt. Unter Eiskühlung und kräftigem Rühren tropft man 17,8 g Acetylchlorid langsam hinzu. Anschließend erwärmt man 1,5 Stunden am Rückfluß, kühlt ab und versetzt vorsichtig mit Eiswasser. Man extrahiert 3mal mit Essigester, wäscht mit Wasser, trocknet und dampft ein. Das so erhaltene bräunliche Öl wird nach Filtration über Kieselgel (Elutionsmittel Hexan/Essigester=2 : 1) aus Hexan/Essigester umkristallisiert. Man erhält 19,5 g 6-Acetyl-3,4-dihydro-1,4,4-trimethyl-2(1H)-chinolinon in farblosen Kristallen, Schmelzpunkt 75-77°C.
21,5 g 3,4-Dihydro-1,4,4-trimethyl-2(1H)-chinolinon werden in 43 g Schwefelkohlenstoff gelöst und mit 85 g Aluminiumchlorid versetzt. Unter Eiskühlung und kräftigem Rühren tropft man 17,8 g Acetylchlorid langsam hinzu. Anschließend erwärmt man 1,5 Stunden am Rückfluß, kühlt ab und versetzt vorsichtig mit Eiswasser. Man extrahiert 3mal mit Essigester, wäscht mit Wasser, trocknet und dampft ein. Das so erhaltene bräunliche Öl wird nach Filtration über Kieselgel (Elutionsmittel Hexan/Essigester=2 : 1) aus Hexan/Essigester umkristallisiert. Man erhält 19,5 g 6-Acetyl-3,4-dihydro-1,4,4-trimethyl-2(1H)-chinolinon in farblosen Kristallen, Schmelzpunkt 75-77°C.
5,0 g 6-Acetyl-3,4-dihydro-1,4,4-trimethyl-2(1H)-
chinolinon werden in 200 ml Benzol gelöst und nach Zugabe
von 2,1 g Äthylenglycol und einigen Kristallen p-Toluolsulfonsäure
5 Stunden am Wasserabscheider gekocht. Nach dem
Abkühlen verdünnt man mit Essigester, wäscht mit verdünnter
Natriumhydrogencarbonat-Lösung, trocknet und dampft ein. Man
erhält 5,6 g 3,4-Dihydro-1,4,4-trimethyl-6-(2-methyl-1,3-
dioxolan-2-yl)-2(1H)-chinolinon als farbloses Öl, das beim
Stehen im Kühlschrank erstarrt, Schmelzpunkt 81-83°C. Die
Verbindung wird ohne weitere Reinigung in der nächsten
Stufe eingesetzt.
0,14 g Lithiumaluminiumhydrid werden in 10 ml abs.
Äther suspendiert und bei Raumtemperatur tropfenweise mit
einer Lösung von 1,0 g 3,4-Dihydro-1,4,4-trimethyl-6-(2-
methyl-1,3-dioxolan-2-yl)-2(1H)-chinolinon in 15 ml abs.
Tetrahydrofuran versetzt. Nach 2stündigem Rühren bei Raumtemperatur
tropft man unter Kühlung langsam Eiswasser zum
Reaktionsgemisch, extrahiert 3mal mit Essigester, wäscht
die organische Phase mit Wasser, trocknet und dampft ein.
Das so erhaltene bräunliche Öl wird in 10 ml Tetrahydrofuran
gelöst und nach Zugabe von 5 ml 1N Schwefelsäure
während 1,5 Stunden bei Raumtemperatur gerührt. Danach
gießt man auf Eiswasser, macht durch Zugabe von 1N Natronlauge
alkalisch und extrahiert mit Essigester. Die organische
Phase wird mit Wasser gewaschen, getrocknet und eingedampft.
Dieses Rohprodukt wird durch Chromatographie an
Kieselgel (Elutionsmittel Hexan/Essigester=9 : 1)
weiter gereinigt und ergibt 460 mg 6-Acetyl-1,2,3,4-tetrahydro-
1,4,4-trimethyl-chinolin als grünliches, viscoses Öl.
Kapseln zur oralen Verabreichung können folgende

1. 2-(3,4-Dihydro-4,4-dimethyl-2H-1-benzopyran (oder
benzothiopyran)-6-yl)propenyl- und 2-(4,4-Dimethyl-1,2,3,4-tetrahydro-6-
chinolinyl)propenyl-Verbindungen der allgemeinen Formel
in der
X -O-, -S-, =SO, =SO₂ oder =NR²;
R¹ Methyl, -CH(R³)OR⁴ oder -C(O)R⁵;
R² Wasserstoff oder C₁- bis C₆-Alkyl;
R³ Wasserstoff oder Methyl;
R⁴ Wasserstoff, C₁ bis C₆-Alkyl oder Alkanoyl;
R⁵ Wasserstoff, C₁- bis C₆-Alkyl, Amino, C₁- bis C₆-Alkylamino, Di-(C₁- bis C₆-alkyl)amino oder OR⁶; und

13. Verfahren zur Herstellung von Verbindungen der allgemeinen
Formel I, und deren Salzen, dadurch gekennzeichnet, daß
man eine Verbindung der allgemeinen Formel
in an sich bekannter Weise
mit einer Verbindung der allgemeinen Formel
in denen entweder A eine Triarylphosphoniumäthylgruppe
der allgemeinen Formel
darstellt, worin
Q Phenyl und Y das Anion einer organischen oder
anorganischen Säure bezeichnet, und B Formyl ist;
oder A Acetyl ist, und B eine Dialkoxyphosphinylmethylgruppe;
und R¹¹ einen Rest R¹ gemäß Definition
in Anspruch 1 mit Ausnahme des Carboxyl- und Formylrests
bedeutet,
zu einer Verbindung der allgemeinen Formel I umsetzt, und erwünschtenfalls den Rest R¹¹ funktionell abwandelt und/ oder das Schwefelatom in einer Verbindung, in der X -S- ist, zu einer Sulfoxyl- oder Sulfonylgruppe oxidiert.
