Description
Die Erfindung betrifft das 1α,25-Dihydroxycholecalciferol-mono
hydrat.
Die Erfindung betrifft insbesondere das 1α,25-Dihydroxycholecal
ciferol-monohydrat mit dem Drehwert [α]= +47° (0,5%, Äthanol).
Das 1α-25-Dihydroxycholecalciferol ist ein für seine interessan
ten pharmakologischen Eigenschaften und insbesondere für seine
bemerkenswerte vitaminische Aktivität bekanntes Produkt (siehe
diesbezüglich die US-PS 36 97 559 und die FR-PS 22 13 053).
Das 1α,25-Dihydroxycholecalciferol ist ein Produkt mit einer
ziemlich erarbeiteten chemischen Struktur, und die bis heute
bekannten Synthesen sind relativ lang und kostspielig und er
geben ein Produkt, das schwierig in pharmazeutisch annehmbarer
Form zu kristallisieren ist.
So beschreibt die FR-PS 22 13 053 das Chloroformsolvat des
1α,25-Dihydroxycholecalciferols, jedoch versucht man bekann
termaßen, die Verwendung von Chloroform in pharmazeutischen
Spezialitäten weitgehendst zu vermeiden.
Das erfindungsgemäße Produkt stellt ein neues Produkt mit sehr
großem industriellem Interesse dar. Es ist vollständig kristal
lisiert, es ist sehr stabil und kann leicht in reiner Form er
halten werden und somit den Anforderungen einer modernen Thera
peutik entsprechen.
Die Anwesenheit von Wasser in dem Molekül kann natürlich nicht
zu hinderlichen Nebenwirkungen führen, wenn das Produkt an den
lebenden Organismus verabreicht wird.
Das erfindungsgemäße Produkt besitzt außerdem den Vorteil, aus
gehend von amorphem 1α,25-Dihydroxycholecalciferol oder einem
seiner Solvate, erhalten nach im Stand der Technik beschriebe
nen Verfahren und insbesondere nach dem in der FR-PS 22 13 053
beschriebenen Verfahren, mit ausgezeichneter Ausbeute, wie es
nachstehend im experimentellen Teil angegeben wird, hergestellt
werden zu können.
Wie das 1α,25-Dihydroxycholecalciferol besitzt das erfindungs
gemäße Produkt bemerkenswerte vitaminische Eigenschaften: es
begünstigt die intestinale Absorption von Calcium und die Fi
xierung des Calciums an dem Knochengewebe. Es gestattet so
mit insbesondere, die Rachitis, die Hypocalcämie, die Osteo
malazie und die Spasmophilie zu bekämpfen.
Die Erfindung betrifft daher auch pharmazeutische Zusammen
setzungen, die als Wirkstoff das 1α,25-Dihydroxycholecalciferol-
monohydrat enthalten.
Die Zusammensetzungen können insbesondere auf buccalem, rekta
lem und parenteralem Weg verabreicht werden.
Sie können fest oder flüssig sein und in üblicher Weise in der
Humanmedizin verwendeten pharmazeutischen Formen vorliegen,
wie z.B. einfache Tabletten oder dragierte Tabletten,
Gelkapseln, Granulate, Suppositorien und injizierbare Präparate.
Sie werden nach üblichen Methoden hergestellt.
Der Wirkstoff kann hierbei in üblicherweise in derartigen phar
mazeutischen Zusammensetzungen verwendete Excipienten eingear
beitet werden, wie Talk, Gummi arabicum, Lactose, Stärke, Ma
gnesiumstearat, Kakaobutter, wäßrige oder nicht-wäßrige Träger,
Fettkörper tierischen oder pflanzlichen Ursprungs, Paraffin
derivate, Glykole, verschiedene Netz-, Dispergier- oder Emul
giermittel und Konservierungsmittel.
Die Verabreichungsdosis variiert entsprechend der zu behandeln
den Erkrankung, dem Patienten, dem Verabreichungsweg und dem in
Betracht gezogenen Produkt. Sie kann z.B. zwischen 0,1 und
200 µg je Tag bei oraler Verabreichung an den Erwachsenen be
tragen.
Die Erfindung betrifft auch ein Verfahren zur Herstellung des
1α,25-Dihydroxycholecalciferol-monohydrats, das dadurch ge
kennzeichnet ist, daß man das Chloroformsolvat von
1α,25-Dihydroxycholecalciferol mit Ethylether desolva
tisiert, die erhaltene Lösung zur Trockne bringt, den Rückstand in Aceton löst, zu der so erhaltenen
Lösung Wasser zugibt, das ausgefallene kristalline Produkt abfiltriert und vollständig hydrolysieren läßt.
Die folgenden Beispiele erläutern die Erfindung.
Man löst bei 20°C 1,585 g 1α,25-Dihydroxycholecalciferol (Chlo
roformsolvat, hergestellt wie in Beispiel 7 der FR-PS 22 13 053
angegeben) in 16 Volumenteilen Äthyläther. Man filtriert die er
haltene Suspension (leichte Unlöslichkeiten) und bringt die er
haltene Lösung unter Vakuum bei niedriger Temperatur zur Trockne.
Man erhält ein weißes Pulver, das man bei 20°C in 18 Volumen
teilen Aceton löst. Man bringt darauf bei 20°C 27 Volumenteile
Wasser ein. Man beläßt das Reaktionsgemisch unter Rühren eine
Stunde bei 18/20°C und beläßt danach 2 Stunden. Man saugt ab
und wäscht das erhaltene Produkt mit Wasser. Man nimmt das Pro
dukt erneut bei Raumtemperatur 1/2 Stunde in Wasser auf. Man
saugt ab und erhält 3,405 g eines Produkts, das man in Gegen
wart von Kaliumhydroxid trocknet. Man erhält so 1,252 g eines
Produkts, das man sich während etwa 1 1/2 Stunden vollständig
hydratisieren läßt. Man gewinnt so das gewünschte Produkt in
Form eines weißen Pulvers.
Wassergehalt: 4,1%
Chloroformgehalt: keiner
[α]= +47° (0,5%, Äthanol)
UV-Spektrum:E bei 265 nm (in Äthanol 95°) = 412
NMR-Spektrum (CDCl3):
18 CH3 bei 0,55 ppm
26 CH3, 27 CH3 bei 1,22 ppm
CH2 in 19-Stellung bei 5 und 5,3 ppm
H6 und H7 bei 6,04 und 6,45 ppm
Anwesenheit von Wasser bei 1,5 ppm
Chloroformgehalt: keiner
[α]= +47° (0,5%, Äthanol)
UV-Spektrum:E bei 265 nm (in Äthanol 95°) = 412
NMR-Spektrum (CDCl3):
18 CH3 bei 0,55 ppm
26 CH3, 27 CH3 bei 1,22 ppm
CH2 in 19-Stellung bei 5 und 5,3 ppm
H6 und H7 bei 6,04 und 6,45 ppm
Anwesenheit von Wasser bei 1,5 ppm
Man stellte Kapseln her, die 0,25 µg, 0,5 µg und 1 µg Wirk
stoff enthielten.
Wirkstoff: 1α,25-Dihydroxycholecalciferol-monohydrat
Öliger steriler Excipient: quantum satis für 1 Kapsel.
Claims (4)
1. 1α,25-Dihydroxycholecalciferol-monohydrat.
2. 1α,25-Dihydroxycholecalciferol-monohydrat mit dem
Drehwert [α]= +47° (0,5 % Äthanol).
3. Verfahren zur Herstellung von 1α,25-Dihydroxy
cholecalciferol-monohydrat gemäß Anspruch 1 oder 2,
dadurch gekennzeichnet, daß man das Chloroformsolvat von
1a,25-Dihydroxycholecalciferol mit Ethylether desolva
tisiert, die erhaltene Lösung zur Trockne bringt, den Rückstand in Aceton löst, zu der so erhaltenen
Lösung Wasser zugibt, das ausgefallene kristalline Produkt abfiltriert und vollständig hydratisieren läßt.
4. Pharmazeutische Zusammensetzungen, enthaltend als
Wirkstoff 1α,25-Dihydroxycholecalciferol-monohydrat gemäß
Anspruch 1 oder 2.
