Description
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung von brauchbaren kleines Alpha-Halogenmethylamin-Derivaten.
Die Aminosäuren Tryptophan, 5-Hydroxytryptophan, 3,4-Dihydroxyphenylalanin (DOPA), Tyrosin und Phenylalanin werden metabolytisch durch eine Decarboxylase für aromatische Aminosäuren in Tryptamin, 5-Hydroxytryptamin, 3,4-Dihydroxyphenethylamin oder Dopamin, Tyramin bzw. Phenethylamin überführt. Es wird angenommen, daß das Decarboxylase-Enzym für aromatische Aminosäuren nichtspezifisch ist, insbesondere soweit es sich um periphere Katalyse handelt. Es liegen jedoch Hinweise dafür vor, daß im Gehirn sowohl für DOPA als auch für 5-Hydroxytryptophan spezifische Decarboxylierungsenzyme vorliegen.
Die vorstehend erwähnten aromatischen Amine sind bekanntlich an verschiedenen pathophysiologischen Vorgängen beteiligt. Beispielsweise wurde gefunden, dass Tryptamin, das Decarboxylierungsprodukt von Tryptophan, enzymatisch zum Monomethyltryptamin methyliert wird, das seinerseits in roten Blutzellen, Plasma und Blutplättchen enzymatisch zum Dimethyltryptamin (DMT) methyliert wird. Das methylierende Enzym liegt bei zahlreichen Säugetierarten vor und es konnte gezeigt werden, dass es im Hirngewebe mehrere Arten wie auch beim Menschen erzeugt wird. Dimethyltryptamin, das starke haluzinogene oder psychomimetische Eigenschaften besitzt, kann bei der Entstehung von Schizophrenie und anderen psychotischen Zuständen eine Rolle spielen. Ein Mittel, das die Bildung von Dimethyltryptamin blockiert, könnte daher als antipsychotisches Mittel nützlich sein. Die Blockierung der Decarboxylierung von Tryptophan führt zu erniedrigten Tryptaminspiegeln, womit das Substrat zur Dimethyltryptamin-Bildung beseitigt wird. Ein Inhibitor der aromatischen Aminosäuredecarboxylase, der die Umwandlung von Tryptophan in Tryptamin blockiert, könnte daher als antipsychotisches
Mittel brauchbar sein.
Sowohl 5-Hydroxytryptamin (5-HT), das Decarboxylierungsprodukt von 5-Hydroxytryptophan, als auch 3,4-Dihydroxyphenethylamin (Dopamin), das Decarboxylierungsprodukt von DOPA, sind an peripheren und zentralen physiologischen Vorgängen beteiligt, und Stoffe, die die Spiegel dieser Amine wirksam kontrollieren, erwiesen sich als brauchbare pharmakologische Mittel. Es konnte gezeigt werden, dass die zentralen oder Hirn-Spiegel an 5-HT und Norepinephrin, das metabolytisch durch Hydroxylierung von Dopamin gebildet wird, bei Patienten mit manischen Krankheiten höher sind als bei Menschen ohne derartige Krankheiten. Es konnte ferner gezeigt werden, dass Stoffe, die die zentralen Spiegel der Monoamine herabsetzen, zum Beispiel 5-HT und insbesondere Norepinephrin, bei Verabreichung an Menschen antimanische Eigenschaften erzeugen, während Stoffe, die den Monoaminspiegel erhöhen, bei dafür zugänglichen Menschen Geisteskrankheiten auslösen können. Daher können Stoffe, die die Bildung von 5-HT und Dopamin blockieren, zum Beispiel durch Inhibierung des aromatischen Aminosäuredecarboxylase-Enzyms, das 5-Hydroxytryptophan und DOPA in 5-HT bzw. Dopamin überführt, als antipsychotische Mittel oder Tranquilizer bei der Behandlung von Geisteskrankheiten nützlich sein.
Es konnte ferner gezeigt werden, dass Stoffe, die die Decarboxylierung von DOPA zu Dopamin inhibieren, brauchbar sind zur Behandlung der Parkinson-Krankheit bei gleichzeitiger Verabreichung von exogenem DOPA oder L-DOPA. Es wird angenommen, dass die Parkinson-Krankheit zumindest zum Teil auf verminderte zentrale Dopaminspiegel zurückgeht, da die exogene Verabreichung von DOPA oder L-DOPA ein wirksames Mittel zur Behandlung dieser Krankheit darstellt. Da exogen verabreichtes DOPA jedoch rasch enzymatisch peripher in Dopamin umgewandelt wird, muss man große
Mengen verabreichen, um zentral eine erhöhte Absorption zu erzielen. DOPA überschreitet die Blut/Hirn-Schranke rasch, im Gegensatz zu Dopamin. Die Verabreichung von DOPA oder L-DOPA in Verbindung mit einem peripher aktiven Inhibitor des Enzyms, das DOPA in Dopamin umwandelt, vermindert die Menge an L-DOPA, die verabreicht werden muss, damit angemessene Mengen zur zentralen Absorption zirkulieren. Weitere Vorteile werden erzielt, wenn man einen aromatischen Aminosäuredecarboxylase-Inhibitor zusammen mit L-DOPA verabreicht. Indem man die periphere Bildung von Dopamin verhindert, können dem Dopamin zugeschriebene Nebenwirkungen, wie Herzarhythmie, Übelkeit und Erbrechen vermieden werden.
Untersuchungen haben gezeigt, dass die 5-Hydroxytryptaminspiegel bei Patienten mit depressivem Syndrom niedriger sind als bei Menschen ohne solche Syndrome. Auch ist die Verabreichung von exogenem L-5-Hydroxytryptophan (L-5-HTP) bei der Behandlung bestimmter depressiver Patienten erfolgreich. Wie bei DOPA muss man jedoch auch, da L-5-HTP peripher leicht zum 5-HT metabolisiert wird, große Mengen an L-5-HTP verabreichen, um erhöhte zentrale Spiegel der Aminosäure zu erzielen. Es wurde festgestellt, dass bei Verabreichung eines Inhibitors der aromatischen Aminosäuredecarboxylase, die die periphere Bildung von 5-HT aus 5-HTP katalysiert, die Menge an zur Erzielung erhöhter zentraler Spiegel benötigtem exogenem 5-HTP spürbar herabgesetzt wird. Somit erwiesen sich die Inhibitoren aromatischer Aminosäuredecarboxylase bei Verwendung in Verbindung mit exogenem 5-HTP als brauchbar zur Behandlung von Depressionen.
Stoffe, die die periphere Umwandlung von 5-HTP in 5-HT blockieren, können brauchbar sein zur Behandlung anderer Zustände, die auf erhöhte zentrale Spiegel von 5-HTP ansprechen, als Ergebnis exogener Verabreichung von 5-HTP.
Es wurde festgestellt, daß exogenes L-5-HTP zur Behandlung von Myoklonien geeignet ist. Untersuchungen haben auch ergeben, dass die Verabreichung von exogenem 5-HTP gegen Schlaflosigkeit nützt. Gleichzeitige Verabreichung von 5-HTP und einem Inhibitor aromatischer Aminosäuredecarboxylase kann daher zur Behandlung dieser Krankheiten nützlich sein.
Die Blockierung der peripheren Bildung von 5-Hydroxytryptamin kann weitere Vorteile bringen, da bekanntlich 5-HT z.B. eine Rolle spielt bei der Entstehung von rheumatoider Arthritis und dem karzinoiden Syndrom durch erhöhten Kollagenspiegel. Auch wurde berichtet, dass 5-HT das primäre Autocoid ist, das für anaphylactoide Reaktionen beim Menschen und Bronchienverengung bei Asthmatikern verantwortlich ist, und Stoffe, die die Bildung von 5-HT antagonisieren oder inhibieren sind brauchbar zur Behandlung dieser Krankheiten. 5-HT verursacht bekanntlich Blutplättchenaggregation und wird als verursachender Faktor bei dem Dumping-Syndrom nach Gastrectomie und Migräne angesehen. Methylsergid, ein 5-Hydroxytryptamin-Antagonist, erwies sich als wirksam bei der Behanldung des Dumping-Syndroms nach Gastrectomie.
Es wurde vermutet, dass Phenethylamin, das Decarboxylierungsprodukt von Phenylalanin, als endogene Verbindung zu schizophrenen Symptomen beiträgt und Migräne auslöst. Ferner wurde vermutet, dass endogenes Tyramin, welches das Decarboxylierungsprodukt von Tyrosin ist, zu Anfallkrankheiten beiträgt.
Es ist somit ersichtlich, dass Stoffe, die zur Regulierung der Spiegel aromatischer Aminosäuren und Amine brauchbar sind, in zahlreichen pharmakologischen Situationen Verwendung finden. Die erfindungsgemäßen Verbindungen sind Inhibitoren der aromatischen Aminosäurendecarboxylase, die Tryptophan, 5-Hydroxytryptophan, 3,4-Dihydroxyphenylalanin, Tyrosin und Phenylalanin
in die entsprechenden Amine überführt, und sie stellen damit brauchbare pharmakologische Mittel dar.
Die Monoamine Adrenalin, Noradrenalin, Dopamin und 5-Hydroxytryptamin sind an der chemischen Übertragung von Nervenimpulsen sowohl im Zentralnervensystem als auch im Nervensystem der Peripherie beteiligt, ferner haben sie hormonale Aufgaben. Es besteht hinreichender Verdacht, dass die vorstehend genannten Monoamine an zahlreichen pathophysiologischen Vorgängen beteiligt sind. Beispielsweise wurde gefunden, dass im Hirn von Patienten mit depressiven Zuständen Defizit an Noradrenalin und/oder 5-Hydroxytryptamin herrscht. Bekanntlich wird die effektive Konzentration dieser Monoamine gesteuert durch die Geschwindigkeit ihrer Herstellung, durch die Geschwindigkeit des enzymatischen Abbaus (Monoaminoxydase) und durch ihre Wiederaufnahme durch die Nervenenden nach der Freigabe durch den Nervenimpuls. Jedes Mittel, das den enzymatischen Abbau oder den Wiederaufnahmevorgang inhibiert, erhöht daher die effektive Konzentration dieser Amine an der gewünschten Wirkungsstelle. Ein Mittel, das die Abgabe dieser Monoamine stimuliert (sympathomimetischer Effekt), würde ebenfalls die effektive Konzentration an der erforderlichen Wirkungsstelle erhöhen. Jedes Mittel, das daher eine oder mehrere dieser Effekte besitzt, kann als Antidepressivum brauchbar sein. So erwiesen sich zum Beispiel Pargylin (Monoaminoxydase-Inhibitor) und Imipramin (Inhibitor der Monoamin-Aufnahme) als wirksame Antidepressiva.
Es wurde gefunden, dass die Parkinson-Krankheit verknüpft ist mit einem Defizit des Dopamin-Gehalts in den Basalganglien des Gehirns. Jeder Stoff, der die dopaminergische Aktivität der Basal-Ganglien direkt oder indirekt erhöht, müsste daher zur Behandlung der Parkinson-Krankheit geeignet sein. So erwiesen sich zum Beispiel Deprenyl (ein Monoaminoxydase-Inhibitor) und
Benzotropin (ein Inhibitor der Dopamin-Aufnahme) als geeignet zur Behandlung von Parkinsonismus.
Die Entstehung der Fettleibigkeit ist zwar unbekannt, doch gibt es erhebliche Hinweise darauf, dass 5-Hydroxytryptamin bei der Zügelung des Appetits eine wichtige Rolle spielt. Außerdem erwiesen sich Stoffe, die die 5-Hydroxytryptaminkonzentration im Hirn verändern, geeignet zur Verwendung als anorexogene Mittel. Dopamin wurde ferner als wichtiger Faktor bei der Regulierung des Essverhaltens erkannt, woraus pharmazeutisch brauchbare Mittel resultierten. So erwiesen sich zum Beispiel Fenfluramin (das die 5-Hydroxytryptamin-Konzentration im Hirn verändert) und Amphetamin (das Dopamin im Hirn freisetzt) als wirksame Appetitzügler.
Ferner ist bekannt, dass Veränderungen der Aktivität des dopaminergischen Systems mit einer analgetischen Wirkung verknüpft sind, und Stoffe, die die Aktivität des dopaminergischen Systems erhöhen, sind entweder wirksam bei der Behebung von Schmerzen oder sie verstärken die schmerzlindernden Eigenschaften anderer Mittel erheblich. So wurde zum Beispiel Amphetamin allein oder in Kombination mit anderen Substanzen gegen Schmerzen eingesetzt.
Das periphere Nervensystem ist wichtig bei der Regulierung der Aktivität des cardiovaskulären Systems, und insbesondere Nerven, die Noradrenalin verwenden, spielen eine fundamentale Rolle bei der Steuerung von Gefäßdruck und Herzgeschwindigkeit. Stoffe, die die Aktivität dieser Nerven durch eine sympathomimetische Wirkung erhöhen, erhöhen daher Blutdruck und Herzgeschwindigkeit. Eine derartige Wirkung ist erwünscht bei der Behandlung hypotensiver Krisen, beispielsweise nach schwerem Blutverlust. Sowohl Adrenalin als auch Noradrenalin
erwiesen sich in dieser Hinsicht als brauchbar. Umgekehrt ist bekannt, dass Noradrenalin im Hirn eine Drepressorwirkung auf das cardiovaskuläre System und insbesondere auf den Blutdruck ausübt. Stoffe, die primär im Hirn das Noradrenalinsystem aktivieren, sollten daher brauchbare antihypertensive Mittel sein.
Sämtliche vorstehend erwähnten Monoamine müssen zunächst von Lagerstellen in Nerven oder in endokrinem Gewebe freigesetzt werden, worauf ihre Wirkung beendigt wird entweder durch Wiederaufnahme von den Nerven oder durch enzymatischen Abbau zu inaktiven Metaboliten. Stoffe, die auf ein oder mehrere dieser Systeme wirken, das heißt entweder Monoamine aus Nerven freisetzen, die Monoaminrezeptoren direkt aktivieren, die Wiederaufnahme blockieren und/oder den enzymatischen Abbau der Monoamine inhibieren, oder die mehr als eine dieser Wirkungen zeigen, könnten zu Mitteln führen, die für die oben erwähnten Zwecke brauchbar wären. Jeder pathophysiologische Zustand sowohl im Hirn als auch in der Peripherie, der mit einem Defizit der Monoaminfunktion verbunden ist, könnte durch Behandlung mit derartigen Mitteln erleichtert werden.
Die erfindungsgemäßen Verbindungen besitzen die folgende allgemeine Formel

Die vorliegende Erfindung betrifft auch pharmazeutisch akzeptable Salze und einzelne optische Isomere der Verbindungen der Formel I.
Die Verbindungen der Formel I sind als pharmakologische Mittel brauchbar; sie stellen Inhibitoren für aromatische Aminosäuredecarboxylase dar. Sie sind ferner als Zwischenprodukte zur Herstellung brauchbarer pharmakologischer Mittel geeignet.
Unter einem Alkylcarbonylrest wird ein Rest der Formel
verstanden, wobei der Alkylanteil geradkettig oder verzweigt ist und 1 bis 6 Kohlenstoffatome aufweist.
Unter dem Benzoylrest wird der Rest der Formel
verstanden.
Unter einem Phenylalkylencarbonylrest wird ein Rest der Formel
verstanden, dessen Alkylenanteil geradkettig oder verzweigt sein kann und 1 bis 6 Kohlenstoffatome besitzt, wie zum Beispiel der Methylen-, Ethylen-, Isopropylen- oder Butylenrest.
Beispiele für geradkettige oder verzweigte Alkylreste mit 1 bis 8 Kohlenstoffatomen sind der Methyl-, Ethyl-, n-Propyl-, Isopropyl-, n-Butyl-, tert.-Butyl-, n-Pentyl- und n-Octylrest.
Beispiele für niedere Alkylreste Y[tief]2 und Y[tief]3 mit 1 bis 4 Kohlenstoffatomen sind der Methyl-, Ethyl-, n-Propyl-, Isopropyl- und n-Butylrest.
Beispiele für pharmazeutisch akzeptable Salze erfindungsgemäßer Verbindungen sind nicht-toxische Säureadditionssalze mit anorganischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure und Phosphorsäure, oder mit organischen Säuren, wie Methansulfonsäure, Salicylsäure, Maleinsäure, Malonsäure, Weinsäure, Zitronensäure und Ascorbinsäure. Die Salze werden in konventioneller Weise hergestellt.
Bevorzugte erfindungsgemäße Verbindungen sind solche der Formel I, worin R[tief]1 Wasserstoff oder einen Alkylcarbonylrest mit einem geradkettigen oder verzweigten Alkylanteil mit 1 bis 4 Kohlenstoffatomen bedeutet, wobei Verbindungen, bei denen R[tief]1 Wasserstoff bedeutet, besonders bevorzugt sind. Eine weitere bevorzugte Ausführungsform der Erfindung sind Verbindungen der Formel I, worin R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 Wasserstoff oder den Rest -OR[tief]8 bedeuten, wobei R[tief]8 Wasserstoff darstellt. Ferner sind solche Verbindungen bevorzugt, bei denen Y den Rest FCH[tief]2- oder F[tief]2CH- bedeutet.

Die Verbindungen der Formel I sind irreversible Inhibitoren des Enzyms, das metabolytisch die Umwandlung von Tryptophan, 5-Hydroxytryptophan, 3,4-Dihydroxyphenylalanin, Tyrosin und Phenylalanin zu Tryptamin, 5-Hydroxytryptamin, 3,4-Dihydroxyphenylethylamin, Tyramin bzw. Phenethylamin katalysiert. Wie bereits erwähnt, lassen Untersuchungen annehmen, dass das für die periphere Umwandlung obiger Aminosäuren zu den Aminen verantwortliche Enzym eine nicht-spezifische Decarboxylase für aromatische Aminosäuren ist. Bei der zentralen Umwandlung sind spezifische Decarboxylasen für die Umwandlung von 5-Hydroxytryptophan bzw. 3,4-Dihydroxyphenylalanin verantwortlich, während die restlichen der oben genannten Aminosäuren wiederum durch eine
nicht-spezifische aromatische Aminosäuredecarboxylase in die betreffenden Amine überführt werden. Die erfindungsgemäßen Verbindungen bewirken eine irreversible Inhibierung der zentralen und peripheren Aktivität nicht-spezifischer aromatischer Aminosäuredecarboxylase und der Aktivität von 3,4-Dihydroxyphenylalanin (DOPA)-Decarboxylase. Unter "zentral" wird das Zentralnervensystem, insbesondere das Gehirn verstanden, während unter der Bezeichnung "peripher" andere Körpergewebe verstanden werden, in denen das Decarboxylaseenzym vorliegt. Die Selektivität von zentraler oder peripherer Inhibierung der Aminosäuredecarboxylase ist bei Verabreichung der Verbindungen der Formel I dosisabhängig.
Als irreversible Inhibitoren aromatischer Aminosäuredecarboxylase und DOPA-Decorboxylase besitzen die erfindungsgemäßen Verbindungen zahlreiche pharmakologische Brauchbarkeiten. Als peripher irreversible Inhibitoren aromatischer Aminosäuredecarboxylase sind die Verbindungen der Formel I brauchbar zur Behandlung der Parkinson-Krankheit bei Verabreichung zusammen mit 3,4-Dihydroxyphenylalanin (DOPA) oder L-3,4-Dihydroxyphenylalanin (L-DOPA). DOPA, und insbesondere das aktive Isomere L-DOPA, sind bekanntlich wirksam bei der Behandlung von Parkinsonismus bei systemischer Verabreichung, gewöhnlich in Mengen von 0,5 bis 1 g/Tag zu Beginn, worauf die zu verabreichende Menge allmählich im Verlauf von 3 bis 7 Tagen auf die größte verträgliche Tagesdosis von etwa 8 g gesteigert wird. Die gleichzeitige Verabreichung von einer Verbindung der Formel I und L-DOPA stellt ein verbessertes Verfahren zur Behandlung des Parkinsonismus dar, da die Verbindung der Formel I die periphere Decarboxylierung von L-DOPA zu L-3,4-Dihydroxyphenethylamin (L-Dopamin) blockiert durch Inhibierung der aromatischen Aminosäuredecarboxylase, wodurch hohe zirkulierende L-DOPA-Spiegel zur zentralen Absorption zur Verfügung stehen und die periphere Bildung erhöhter Dopamin-Spiegel verhütet wird,
die bekanntlich zu bestimmten unerwünschten Nebeneffekten, wie Herzarhythmien führt. Bei gleichzeitiger Verabreichung einer Verbindung der Formel I mit L-DOPA kann die zu verabreichende Menge an L-DOPA um das 2- bis 10-fache reduziert werden, verglichen mit den bei alleiniger Verabreichung von L-DOPA benötigten Mengen. Vorzugsweise werden die erfindungsgemäßen Verbindungen vor der Verabreichung von L-DOPA gegeben. Beispielsweise kann man eine Verbindung der Formel I 30 Min. bis 4 Std. vor der Verabreichung von L-DOPA geben, je nach der Art der Verabreichung und dem Zustand des zu behandelnden Patienten.
Die Verbindungen der Formel I sind auch brauchbar zur Behandlung depressiver Syndrome bei Verwendung zusammen mit 5-Hydroxytryptophan oder insbesondere dem aktiven linksdrehenden Isomer, das bekanntlich bei systemischer Verabreichung zur Behandlung von Depressionen geeignet ist. Die Verbindungen der Formel I blockieren über die periphere Inhibierung der aromatischen Aminosäuredecarboxylase die Umwandlung von 5-Hydroxytryptophan zu 5-Hydroxytryptamin, so dass höhere zirkulierende Spiegel an 5-HTP zur zentralen Absorption zur Verfügung stehen. Die Verbindungen der Formel I sind bei gleichzeitiger Verabreichung mit exogenem 5-HTP auch brauchbar zur Behandlung von Myoklonien, die bekanntlich mit Erfolg mit steigenden zentralen Spiegeln von 5-HTP behandelt werden können.
Aufgrund ihrer peripheren inhibierenden Wirkung auf die Decarboxylase für aromatische Aminosäuren sind die Verbindungen der Formel I auch brauchbar zur Behandlung von rheumatoider Arthritis, karzinoidem Syndrom, anaphylaktoiden Reaktionen beim Menschen, Bronchienverengung bei Asthmatikern und anderen Zuständen, die durch hohe periphere Mengen an 5-Hydroxytryptamin verursacht werden.
Wie bereits erwähnt, können Mittel, die erhöhte Spiegel an 5-HT und Norepinephrin, dem Hydroxylierungsprodukt von Dopamin, senken, zur Behandlung von Patienten mit Geisteskrankheiten verwendet werden. Als zentral wirksame irreversible Inhibitoren aromatischer Aminosäuredecarboxylase und DOPA-Decarboxylase sind die Verbindungen der Formel I somit brauchbar bei der Behandlung von Geisteskrankheiten. Aufgrund der zentralen inhibierenden Wirkung auf aromatische Aminosäuredecarboxylase sind die Verbindungen der Formel I auch als antipsychotische Mittel verwendbar, da die zentralen Tryptaminspiegel gesenkt werden, sie eignen sich ferner zur Behandlung von Schizophrenie und Anfallkrankheiten, da die zentralen Spiegel an Phenethylamin und Tyramin ebenfalls gesenkt werden.
Die Verbindungen der Formel I sind auch zentrale Inhibitoren der Monoaminoxydase, das heißt sie inhibieren die enzymatische Wirkung von Monoaminoxydase im Hirn. Die Verbindungen sind daher als Antidepressiva, Appetitzügler, Analgetika, Hypotensiva und antihypertensive Mittel verwendbar.
Die Brauchbarkeit der Verbindungen der Formel I als irreversible Inhibitoren aromatischer Aminosäuredecarboxylase wird wie folgt demonstriert:
Eine Verbindung der Formel I wird in wässriger Lösung oder Suspension an Ratten oder Mäuse verabreicht. In verschiedenen zeitlichen Abständen 1 bis 48 Std. nach der Verabreichung werden die Tiere enthauptet und die Aktivität der aromatischen Aminosäuredecarboxylase wird radiometrisch nach Christenson et al., Arch. Zur Erzielung der gewünschten Effekte können die erfindungsgemäßen Verbindungen auf verschiedene Arten verabreicht werden. Die Verbindungen können allein oder in Form pharmazeutischer Präparate oral oder parenteral, zum Beispiel subkutan, intravenös oder intraperitoneal gegeben werden. Die Verbindungen können auch intranasal verabreicht werden oder auf Schleimhäute, zum Beispiel der Nase, des Rachens und der Bronchien appliziert werden, beispielsweise in einem Aerosol-Spray, der kleine Teilchen einer erfindungsgemäßen Verbindung in Sprühlösung oder trockener Pulverform enthält.
Als zu verabreichende Menge eignet sich jede wirksame Menge. Je nach dem Patienten, dem zu behandelnden Zustand und der Art der Verabreichung, schwankt die zu verabreichende Menge innerhalb eines breiten Bereiches. Werden die Verbindungen der Formel I verabreicht zur Bewirkung einer peripheren irreversiblen Inhibierung aromatischer Aminosäuredecarboxylase oder zur zentralen Inhibierung von Monoaminoxydase, so liegt die wirksame Menge bei etwa 0,1 mg bis 100 mg/kg Körpergewicht/Dosis, und vorzugsweise bei etwa 5 bis 25 mg/kg Körpergewicht. Zum Beispiel kann der gewünschte Effekt erzielt werden mit einer Dosiseinheit wie Tabletten, die 10 bis 250 mg einer erfindungsgemäßen Verbindung enthalten und 1- bis 4-mal täglich genommen werden. Werden die Verbindungen der Formel I verabreicht zur Erzielung einer zentralen irreversiblen Inhibierung von aromatischer Aminosäuredecarboxylase oder 3,4-Dihydroxyphenylalanin-Decarboxylase, so liegt die wirksame Menge bei etwa 100 bis 500 mg/kg Körpergewicht/Tag, und vorzugsweise bei etwa 50 (150) bis 300 mg/kg Körpergewicht. Zum Beispiel kann der gewünschte zentrale inhibierende Effekt erzielt werden, indem man eine Dosiseinheit, zum Beispiel Tabletten mit etwa 350 bis 500 mg einer erfindungsgemäßen Verbindung, ein- bis viermal täglich einnimmt.
Unter dem "Patienten" werden warmblütige Tiere wie Säugetiere, zum Beispiel Katzen, Hunde, Ratten, Mäuse, Meerschweinchen, Schafe, Pferde, Rinder und Menschen verstanden.
Die feste Dosiseinheit kann konventioneller Art sein. Beispielsweise kann eine übliche Gelatinekapsel vorliegen, die die erfindungsgemäße Verbindung und einen Träger, zum Beispiel Gleitmittel und inerte Füllstoffe, wie Lactose, Rohrzucker und Maisstärke enthält. Gemäß einer weiteren Ausführungsform werden die erfindungsgemäßen Verbindungen mit konventionellen Tablettenbasen wie Lactose, Rohrzucker oder Maisstärke und Bindemitteln, wie Gummi acacia, Maisstärke oder Gelatine, Sprengmitteln, wie Maisstärke, Kartoffelstärke oder Alginsäure und einem Gleitmittel wie Stearinsäure oder Magnesiumstearat tablettiert.
Zur parenteralen Verabreichung können die Verbindungen verabreicht werden in Form injizierbarer Dosen einer Lösung oder Suspension der betreffenden Verbindung in einem physiologisch zulässigen Verdünnungsmittel mit einem pharmazeutischen Träger, der eine sterile Flüssigkeit, wie Wasser oder ein Öl sein kann, gegebenenfalls in Verbindung mit einem oberflächenaktiven Mittel und anderen pharmazeutisch zulässigen Hilfsstoffen. Beispiele für zur Herstellung derartiger Präparate geeignete Öle sind solche aus Erdöl, Ölen tierischen, pflanzlichen oder synthetischen Ursprungs, beispielsweise Erdnussöl, Sojabohnenöl und Mineralöl. Im allgemeinen sind Wasser, Kochsalzlösung, wässrige Dextroselösung und ähnliche Zuckerlösungen, Ethanole und Glycole, wie Propylenglycol oder Polyethylenglycol bevorzugte flüssige Träger, insbesondere für injizierbare Lösungen.
Die Verbindungen können auch in Form einer Depot-Injektion oder eines Implantats verabreicht werden, das in solcher Weise
hergestellt ist, dass eine verzögerte Abgabe des Wirkstoffs erfolgt. Der Wirkstoff kann zu Pellets oder kleinen Zylindern verpresst und subkutan oder intramuskulär als Depot-Injektion oder Implantat gegeben werden. In Implantaten können inerte Materialien wie biologisch abbaubare Polymere oder synthetische Silikone, zum Beispiel "Silastic" (Silikonkautschuk der DOW-Corning Corporation) verwendet werden.
Zur Verwendung in Aerosolen werden die Verbindungen in Lösung oder Suspension in einem unter Druck stehenden Aerosolbehälter verpackt, zusammen mit geeigneten Treibmitteln, zum Beispiel Kohlenwasserstoffen wie Propan, Butan oder Isobutan, oder Kohlendioxid oder Stickstoff, oder anderen, zulässigen Treibmitteln, wobei übliche Hilfsstoffe, wie Co-Lösungsmittel und Netzmittel je nach Bedarf oder Wunsch eingesetzt werden. Die Verbindungen können auch aus nicht unter Druck stehenden Behältern, wie Vernebelungsgeräten verabreicht werden.
Wie bereits erwähnt, finden die erfindungsgemäßen Verbindungen der Formel I insbesondere Anwendung durch Verabreichung zusammen mit exogenem L-DOPA, wobei man in diesem Fall einzelne Formulierungen mit einer Verbindung der Formel I sowie mit L-DOPA geben oder beide Wirkstoffe zu einer kombinierten pharmazeutischen Formulierung vereinigen kann. In jedem Fall beträgt die Menge an Verbindung der Formel I zur Menge an L-DOPA etwa 1:1 bis 1:10. Ein Kombinationspräparat kann einen inneren Teil, enthaltend das L-DOPA und einen äußeren Teil, enthaltend eine Verbindung der Formel I, aufweisen, wobei jeder Wirkstoff geeignet formuliert ist. Eine besonders zweckmäßige Kombination kann hergestellt werden, indem man L-DOPA, gegebenenfalls mit geeigneten Trägern, zu einem Kern verpresst, den Kern mit einer Magensaft-resistenten Laminatschicht versieht und über den beschichteten Kern eine Außenschicht anbringt, die die Ver-
bindung der Formel I in geeigneter Formulierung enthält. Bei Verwendung einer derartigen Kombination wird der Decarboxylase-Inhibitor, das heißt die Verbindung der Formel I, vorzugsweise 30 bis 60 Min. vor dem L-DOPA freigesetzt. Die Laminatschicht kann unter Verwendung einer nicht-wässrigen Glyceridlösung oder eines wasserunlöslichen Polymeren wie Ethylcellulose oder Celluloseacetatphthalat hergestellt werden. Auch Formulierungen, in welchen das L-DOPA enterisch beschichtet ist unter Verwendung von Gemischen aus Schellack und Schellack-Derivaten und Celluloseacetatphthalaten können verwendet werden.
Die Verbindungen der Formel I, worin R[tief]1 Wasserstoff bedeutet, sind nicht nur brauchbare pharmakologische Mittel, sondern sie sind auch geeignet als Zwischenprodukte zur Herstellung von Cephalosporin-Antibiotika der Formel II:

In obiger Formel II besitzen Y, R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 die für Formel I angegebene Bedeutung. M bedeutet Wasserstoff oder eine negative Ladung, R[tief]2 bedeutet Wasserstoff oder die Acetoxygruppe.
Die Verbindungen der Formel II und ihre pharmazeutisch zulässigen Salze und einzelnen optischen Isomeren sind neue Verbindungen, die als Antibiotika brauchbar sind. Sie können ähnlich wie die bekannten Cephalosporinderivate wie Cephalexin, Cephalothin oder Cephaloglycin verabreicht werden. Die Verbindungen der Formel II, ihre pharmazeutische zulässigen Salze
und Isomeren können allein oder in Form pharmazeutischer Präparate oral, parenteral oder topisch an warmblütige Tiere verabreicht werden, das heißt an Vögel und Säugetiere wie zum Beispiel Katzen, Hunde, Rinder, Schafe, Pferde und Menschen. Zur oralen Verabreichung können die Verbindungen in Form von Tabletten, Kapseln oder Pillen oder in Form von Elixieren oder Suspensionen gegeben werden. Zur parenteralen Verabreichung werden die Verbindungen am besten in Form einer sterilen wässrigen Lösung eingesetzt, die andere gelöste Stoffe enthalten kann, zum Beispiel Kochsalz oder Glucose in zur Herstellung einer isotonischen Lösung ausreichender Menge. Zur topischen Verabreichung werden die Verbindungen der Formel II, ihre Salze und Isomere in Cremes oder Salben eingearbeitet.
Beispiele für Bakterien, gegen die die Verbindungen der Formel II, ihre pharmazeutisch zulässigen Salze und optischen Isomeren wirksam sind, sind Staphylococcus aureus, Salmonella schottmuehleri, Klebsiella pneumoniae, Diplococcus pneumoniae und Streptococcus pyogenes.
Beispiele für pharmazeutisch zulässige nicht-toxische, anorganische Säureadditionssalze der Verbindungen der Formel II sind Additionssalze mit Mineralsäuren, wie zum Beispiel Hydrochloride, Hydrobromide, Sulfate, Sulfamate, Phosphate und Additionssalze mit organischen Säuren, wie das Maleat, Acetat, Citrat, Oxalat, Succinat, Benzoat, Tartrat, Fumarat, Malat und Ascorbat. Die Salze werden in konventioneller Weise hergestellt.
Beispiele für Cephalosporinderivate der Formel II sind die 7-((4-((1-Difluormethyl-2-(3-hydroxyphenyl)ethyl)aminomethyl)phenyl)acetylamino)-3-acetyloxymethyl-8-oxo-5-thia-1-azabicyclo[4.2.0.]oct-2-en-2-carbonsäure und 7-((4-((1-Difluormethyl-2-
(3,4-dihydroxyphenyl)ethyl)aminomethyl)phenyl)acetylamino)-3-acetyloxymethyl-8-oxo-5-thia-1-azabicyclo[4.2.0.]oct-2-en-2-carbonsäure.
Die Verbindungen der Formel II werden hergestellt, indem man eine Verbindung der Formel III

worin R[tief]2 und M die für Formel II angegebene Bedeutung besitzen (Herstellung siehe US-PS 3 919 206), mit einer Verbindung der Formel I, worin R[tief]1 Wasserstoff bedeutet, umsetzt. Die Umsetzung erfolgt gewöhnlich in einem Lösungsmittel, zum Beispiel einem niederen Alkohol, wie Methanol, Ethanol oder Isopropylalkohol, oder in Dimethylsulfoxid, Dimethylformamid oder wässrigen Gemischen dieser Lösungsmittel. Die Reaktionstemperatur beträgt etwa 0 bis 125°C, die Reaktionszeit etwa 1/2 bis 24 Stunden. Nach der Solvolyse wird die Amin-Schutzgruppe durch saure Hydrolyse entfernt, dann werden die Cephalosporinprodukte in konventioneller Weise isoliert.
Die Verbindungen der Formel I, worin Y einen der Reste -CH[tief]2F oder -CHF[tief]2 und R[tief]1 Wasserstoff bedeuten, während R[tief]3 und R[tief]4 einen Rest -OR[tief]8, worin R[tief]8 Wasserstoff ist, oder R[tief]4 und R[tief]5 einen Rest -OR[tief]8, worin R[tief]8 Wasserstoff ist, oder R[tief]4 und R[tief]5 zusammen einen Rest -O-CH[tief]2-O- darstellen, oder worin R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 eine sonstige Bedeutung gemäß Tabelle IA besitzen, ausgenommen R[tief]8 = Methyl, und wobei R[tief]5 von -NY[tief]2Y[tief]3 verschieden ist, werden hergestellt, indem man ein entsprechend substituiertes Phenylhalogenketon der nachstehenden Formel IV zum entsprechenden Alkohol reduziert, diesen mit 1 Äquivalent eines Imids, wie
Phthalimid, Succinimid oder Maleimid, 1,1 Äquivalenten eines Phosphins, wie Triphenylphosphin oder eines Trialkylphosphins wie Tri-n-butylphosphin und 1,1 Äquivalenten Diethyl-azodicarboxylat in einem Lösungsmittel, zum Beispiel einem Äther, wie Diethyläther, Tetrahydrofuran oder p-Dioxan, Benzol oder Dimethoxyethan bei etwa 0 bis 100°C, vorzugsweise bei etwa 25°C etwa 1/2 bis 24 Std. in Inertgasatmosphäre, wie Stickstoff oder Argon umsetzt, und das so erhaltene Imidoderivat zum freien Amin hydrolysiert.
In obiger Formel IV bedeutet Y' den -CHF[tief]2- oder -CH[tief]2F-Rest, während R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die aus nachstehender Tabelle IIA ersichtliche Bedeutung haben:

Die Reduktion der Ketone der Formel IV zum entsprechenden Alkohol erfolgt chemisch, wobei man zum Beispiel 1 bis 10 Äquivalente eines Metallhydrids, wie Lithiumborhydrid, Natriumborhydrid, Natriumcyanborhydrid oder Lithiumaluminiumhydrid, Boran oder Dimethylthioboran als Reduktionsmittel verwendet, oder katalytisch, zum Beispiel mit Raney-Nickel, Rhodium, Palladium auf Kohle oder Platinoxid. Die gesamte Reaktionszeit beträgt etwa 10 Min. bis 24 Std. und die Temperatur liegt bei etwa -40 bis 100°C, je nach dem verwendeten Reduktionsmittel. Bei chemischer Reduktion beträgt die Reduktionszeit im allgemeinen etwa 10 Min. bis 24 Std., wobei die Temperaturen etwa -40 bis 65°C betragen. Geeignete Lösungsmittel zur chemischen Reduktion von Verbindungen der allgemeinen Formel IV sind niedere Alkohole, wie Methanol oder Ethanol oder Ether, wie Diethylether oder Tetrahydrofuran. Bei katalytischer Reduktion beträgt die Reaktionszeit etwa 1 bis 24 Std., die Reaktionstemperatur liegt bei etwa 25 bis 100°C und der Druck bei 1 bis 120 bar. Geeignete Lösungsmittel zur katalytischen Reduktion der Verbindungen der Formel IV sind niedere Alkohole, wie Methanol oder Ethanol, Essigsäure oder Ethylacetat. Die chemische Reduktion wird bevorzugt.
Die Hydrolyse zum Amin und Entfernung sämtlicher Schutzgruppen am Ring erfolgt mit einer starken Mineralsäure, wie Salzsäure, Bromwasserstoffsäure oder Schwefelsäure, oder mit einer organischen Säure, wie Toluolsulfonsäure oder Trifluoressigsäure, in Wasser bei Rückflusstemperatur während etwa 4 bis 48 Std., oder unter Verwendung von beispielsweise 1 bis 3 Äquivalenten Hydrazin, Methylhydrazin oder Methylamin bei einer Temperatur von etwa 25°C bis Rückflusstemperatur während etwa 1 bis 12 Std.; gegebenenfalls erfolgt dann Behandlung mit einer starken Mineralsäure oder organischen Säure, wie vorstehend beschrieben, oder Entfernung der Schutzgruppen durch Hydrogenolyse.
Wie erwähnt, kann ein Tri-alkylphosphin, wie zum Beispiel Tri-n-butylphosphin, bei der Reaktion verwendet werden. Der Alkylrest besitzt in diesem Fall 1 bis 10 Kohlenstoffatome. Die Trialkylphosphine sind bekannt oder können nach bekannten Methoden hergestellt werden.
Verbindungen der Formel I, worin Y den -CHF[tief]2-Rest und R[tief]1 Wasserstoff bedeuten, während R[tief]3 und R[tief]4 beide den Rest -OR[tief]8, worin R[tief]8 Wasserstoff ist, oder R[tief]4 und R[tief]5 beide den Rest -OR[tief]8, worin R[tief]8 Wasserstoff ist, oder R[tief]4 und R[tief]5 zusammen den Rest -O-CH[tief]2-O- darstellen, oder worin R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 eine der anderen Bedeutungen gemäß Tabelle IA und IB haben, ausgenommen, daß R[tief]8 Methyl bedeutet und R[tief]5 von -NY[tief]2Y[tief]3 verschieden ist, können hergestellt werden, indem man einen nied.-Alkyl-malonsäurediester, das heißt eine Verbindung der Formel

worin die niederen Alkylreste, die gleich oder verschieden sein können, geradkettig oder verzweigt sind und 1 bis 4 Kohlenstoffatome besitzen, wie der Methyl-, Ethyl-, n-Propyl-, Isopropyl-, tert.-Butyl- oder n-Butylrest, mit einer starken Base, wie Natriumhydrid, Kaliumhydrid, Kalium-tert.-butylat, Lithiumacetylid, Lithiumcarbid, Natriumamid, Lithiumamid, Lithium- oder Natriumhexamethyldisilazan, einem Natrium- oder Kaliumalkylat, wie dem Methylat oder Ethylat, oder Natrium- oder Kaliumhydroxid in einem protischen Lösungsmittel, wie einem niederen Alkohol, zum Beispiel Methanol oder Ethanol, oder in einem aprotischen Lösungsmittel, wie Diethylether, Tetrahydrofuran, Benzol, Ethylenglycol, Diethylether, Dimethylsulfoxid oder Hexamethylphosphortriamid umsetzt unter Bildung eines Carbanions. Dieses wird mit einem Alkylierungsmittel der Formel
umgesetzt, worin R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die in Tabelle IIA und IIB angegebene Bedeutung besitzen und X eine leicht abgehende Gruppe ist, zum Beispiel Chlor, Brom, Iod, der Tosylat-, Acetat- oder Mesylatrest. Die Alkylierungsreaktion erfolgt bei Temperaturen von etwa -30°C bis zur Rückflusstemperatur des Lösungsmittels, die Reaktionszeit beträgt etwa 1/2 bis 24 Stunden. Der so erhaltene alkylierte Malonsäurediester wird mit einer Base behandelt, zum Beispiel einem Metallhydrid, wie Natriumhydrid oder Kaliumhydrid, Natriumamid oder Lithiumamid, Lithiumdiisopropylamid, Lithium- oder Natriumhexamethyldisilazan, Lithiumacetylid oder Lithiumcarbid, und zwar in einem aprotischen Lösungsmittel, wie Tetrahydrofuran, Diethylether, Dimethoxyethan, Glyme, Benzol, Hexamethylphosphoramid oder Dimethylsulfoxid. Dann erfolgt Umsetzung mit einem geeigneten Difluormethylhalogenalkylierungsmittel, zum Beispiel mit Chlordifluormethan, Bromdifluormethan oder Difluoriodmethan. Die Umsetzung erfolgt bei Temperaturen von etwa -20 bis 60°C, und vorzugsweise etwa 25°C, die Reaktionszeit beträgt etwa 15 Min. bis 24 Stunden. Das Difluormethylderivat wird mit einer starken Mineralsäure behandelt, zum Beispiel mit Salzsäure oder Schwefelsäure in Wasser, oder mit einer starken organischen Säure, zum Beispiel p-Toluolsulfonsäure oder Trifluormethylessigsäure in Wasser bei Temperaturen von etwa 25 bis 180°C während etwa 1/2 bis 48 Std., wobei man die entsprechende 2-Difluormethyl-3-(substituiert-phenyl)-propionsäure erhält. Ist eine der niederen Alkylestergruppen ein tertiärer Butylrest, so wird das Difluormethylderivat vorzugsweise mit Trifluormethylessigsäure bei etwa
25°C 1 Std. behandelt, dann wird bei vermindertem Druck eingeengt, wobei man das Difluormethyl-malonsäuremonoesterderivat der Formel

erhält, worin R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die in Tabelle IIA und IIB angegebene Bedeutung besitzen, während der niedere Alkylrest einem der obigen Reste, mit Ausnahme des tertiären Butylrests, entspricht. Das Difluormethyl-malonsäuremonoesterderivat wird durch Behandlung mit einer organischen Säure decarboxyliert, zum Beispiel mit Essigsäure oder Propionsäure bei Temperaturen von etwa 100 bis 160°C und Reaktionszeiten von etwa 1 bis 24 Stunden. Vorzugsweise wird das Monoesterderivat decarboxyliert durch Behandlung mit Essigsäure bei etwa 130°C während 12 Std. Beim Einengen bei vermindertem Druck erhält man den entsprechenden niederen Alkylester der 2-Difluormethyl-3-(substitiert-phenyl)-propionsäure, der mit einer starken Mineralsäure, wie Salzsäure oder Schwefelsäure, oder einer organischen Säure, wie p-Toluolsulfonsäure oder Trifluormethylessigsäure in Wasser bei Temperaturen bis zur Rückflußtemperatur des Lösungsmittels während etwa 1 bis 24 Std. hydrolysiert wird. Die freie Säure wird unter den Bedingungen der Schmidt'schen Reaktion oder der Curtius-Reaktion in das Aminderivat überführt, siehe zum Beispiel Organic Reactions, Bd. III, S. 308 bzw. 338. Unter den Bedingungen der Schmidt'schen Reaktion wird die freie Säure etwa 1 bis 24 Std. bei Temperaturen von etwa 0 bis 60°C mit Stickstoffwasserstoffsäure umgesetzt. Unter den Bedingungen der Curtius-Reaktion wird die freie Säure
mit einem Thionylhalogenid, zum Beispiel Thionylchlorid bei Rückflusstemperatur etwa 1 bis 4 Std. umgesetzt, oder das Natriumsalz der Säure wird mit Oxalylchlorid zum entsprechenden Säurechlorid umgesetzt, das dann mit Natriumazid bei Rückflusstemperatur etwa 40 bis 100 Std. behandelt wird. Dann erfolgt saure Hydrolyse mit einer starken Mineralsäure, wie Salzsäure oder Schwefelsäure, oder mit einer organischen Säure, wie p-Toluolsulfonsäure, während etwa 1 bis 24 Std. bei Rückflusstemperatur.
Die Verbindungen der Formel I, worin Y den Trifluormethylrest und R[tief]1 Wasserstoff bedeuten, während R[tief]3 und R[tief]4 beide den Rest -OR[tief]8, worin R[tief]8 Wasserstoff bedeutet, oder R[tief]4 und R[tief]5 beide den Rest -OR[tief]8, worin R[tief]8 Wasserstoff bedeutet, oder R[tief]4 und R[tief]5 zusammen den Rest -O-CH[tief]2-O- darstellen, oder worin R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 eine sonstige Bedeutung gemäß Tabelle IA und IB besitzen, mit den Ausnahmen, daß R[tief]8 Methyl ist und R[tief]5 vom Rest -NY[tief]2Y[tief]3 verschieden ist, werden hergestellt, indem man ein Keton der Formel

worin Y[tief]4 den Trifluormethylrest bedeutet und R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die in Tabelle IIA und IIB angegebene Bedeutung besitzen, mit einem Hydroxylaminsalz, wie zum Beispiel dem Hydrochlorid oder Sulfat, in einem niederen alkoholischen Lösungsmittel oder Wasser, oder einer Kombination aus niederem Alkohol und Wasser in Gegenwart einer mineralischen Base, wie Natrium- oder Kaliumhydroxid oder Natriumacetat oder einer organischen Base, wie Pyridin, etwa 1 bis 8 Std. bei etwa 20 bis 100°C umsetzt, wobei man das entsprechende Oxim erhält, das zum
Amin reduziert wird, zum Beispiel mit Lithiumaluminiumhydrid, beispielsweise in Ethern, wie Diethylether, Tetrahydrofuran oder Dioxan, mit Natriumborhydrid, zum Beispiel in niederen Alkoholen, wie Methanol, Ethanol oder tert.-Butanol, mit Zink und Essigsäure, Natrium und Methanol in flüssigem Ammoniak oder durch katalytische Reduktion, zum Beispiel mit Palladium auf Kohle, mit Raney-Nickel oder Platinoxid-Katalysator in einem Lösungsmittel, wie Alkoholen, zum Beispiel Methanol, Ethanol, Isopropylalkohol oder tert.-Butanol oder deren Gemischen mit Wasser oder in Ethylacetat. Die Reduktion erfolgt bei Temperaturen von etwa 0°C bis zur Rückflusstemperatur des Lösungsmittels, die Reaktionszeit beträgt etwa 1/2 bis 8 Std., je nach dem angewandten Reduktionsmittel. Das Keton der Formel VIII kann auch durch reduktive Alkylierung oder Aminierung mit überschüssigem Ammoniak und einem Katalysator, wie Platin, Raney-Nickel, oder Palladium in niederen Alkoholen, wie Methanol, Ethanol oder tert.-Butanol, oder Ethern, wie Diethylether oder Tetrahydrofuran oder deren Gemischen mit Wasser bei einer Temperatur von etwa 25 bis 200°C, und vorzugsweise etwa 100 bis 150°C unter hohen Drucken von beispielsweise etwa 52 bis 140 bar in das entsprechende Amin umgewandelt werden. Diese Verfahren sind an sich bekannt, vergleiche zum Beispiel W.R. Nes und A. Burger, J.Am.Chem.Soc. 72, 5409 (1960), und R.M. Pinder und A. Burger, J.Pharm.Sce., 56, 970 (1976).
Die Verbindungen der allgemeinen Formel I, worin Y einen Rest der Formel -CH[tief]2F bedeutet, R[tief]1 ein Wasserstoffatom darstellt, beide Reste R[tief]3 und R[tief]4 den -OR[tief]8-Rest bedeuten, worin R[tief]8 ein Wasserstoffatom ist, oder beide Reste R[tief]4 und R[tief]5 den -OR[tief]8-Rest bedeuten, worin R[tief]8 ein Wasserstoffatom ist, oder beide Reste R[tief]4 und R[tief]5 zusammen einen Rest der Formel -O-CH[tief]2-O- bedeuten, oder worin jeder der Reste R[tief]3, R[tief]4, R[tief]5, R'[tief]4 und R[tief]6 die anderweitig in den Tabellen IA und IB angegebene Bedeutung besitzt, ausgenommen, daß R[tief]8 einen Methylrest bedeutet und R[tief]5 eine andere Bedeutung als NO[tief]2 oder NY[tief]2Y[tief]3 besitzt, können dadurch hergestellt werden, daß man sehr langsam ein Benzylhalogenid der Formel

worin R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die in Tabelle IIA und IIB angegebene Bedeutung besitzen, ausgenommen die Verbindungen, bei denen R[tief]11 NO[tief]2 ist und Xa ein Chloratom oder ein Bromatom bedeuten, Magnesiumspänen in einem geeigneten Ether als Lösungsmittel, wie Tetrahydrofuran, Diethylether oder Gemischen derselben, zusetzt und die Reaktion bei einer Temperatur von etwa -20 bis 70°C, vorzugsweise von etwa 25°C bis zum Siedepunkt des Lösungsmittels, 30 Minuten bis 24 Stunden ablaufen läßt. Bei Beginn der Reaktion setzt man eine Spur Methyljodid zu. Falls entweder R[tief]9 oder R[tief]11 einen Methoxy-Rest bedeuten, wird die Reaktion ebenfalls in Tetrahydrofuran initiiert.
Das so erhaltene Grignard-Derivat wird in einem Verhältnis, das von 0,5 bis 3,0 variieren kann, in einem aprotischen Lösungsmittel, wie Tetrahydrofuran, Diethylether, Dioxan, Benzol, Dimethoxyethan oder Dimethoxymethan oder Gemischen derselben, mit Fluoracetonitril versetzt. Die Reaktionstemperatur während der Zugabe von Fluoracetonitril kann von etwa -20 bis -70°C, vorzugsweise von etwa -20 bis -25°C variieren und die Reaktionszeit variiert von etwa 10 Minuten bis 12 Stunden, vorzugsweise von 10 Minuten bis 1 Stunde, wobei das Ketiminsalz-Derivat der Formel

worin R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die in Formel XI angegebene Bedeutung besitzen, erhalten wird. Das Ketiminsalz-Derivat wird reduziert, indem man es bei einer Temperatur von etwa -20 bis 25°C in eine Lösung von Natriumborhydrid, Kaliumborhydrid, Natriumcyanoborhydrid oder Lithiumborhydrid in protischen Lösungsmitteln, wie niederen Alkoholen, z.B. Methanol und Ethanol oder Gemischen derselben mit Wasser, oder Lithiumaluminiumhydrid in aprotischen Lösungsmitteln, wie Diethylether, Tetrahydrofuran, Benzol, Pentan, Hexan, Dimethoxyethan oder Dimethylsulfoxid, gießt und die Reaktion etwa 1 Stunde bis 20 Stunden ablaufen läßt, wobei die entsprechenden kleines Alpha-CH[tief]2F-Derivate der Formel I erhalten werden. Die bevorzugten Reduktionsmittel sind die Borhydride.
Die Verbindungen der Formel I, worin R[tief]1 Wasserstoff bedeutet und eines der Symbole R[tief]3, R[tief]4, R'[tief]4 oder R[tief]5 den Rest -OR[tief]8 darstellt, worin R[tief]8 Wasserstoff bedeutet, werden aus dem entsprechenden Derivat, worin R[tief]3, R[tief]4, R'[tief]4 oder R[tief]5 den Methoxyrest bedeutet, hergestellt durch Behandlung dieses Derivats mit Bromwasserstoff in Wasser oder Essigsäure bei einer Temperatur von etwa 25 bis 125°C während etwa 4 bis 24 Stunden.

Die bei diesem Verfahren verwendeten Alkylhalogenide sind bekannt oder können nach bekannten Verfahren hergestellt werden.
Die Verbindungen der Formel I, worin R[tief]1 Wasserstoff bedeutet und eines der Symbole R[tief]3, R[tief]4, R'[tief]4 oder R[tief]5 den Rest -OR[tief]8 darstellt, worin R[tief]8 ein Alkylcarbonylrest mit einem geradkettigen oder verzweigten Alkylanteil mit 1 bis 6 Kohlenstoffatomen, der Benzoylrest oder ein Phenylalkylencarbonylrest mit einem geradkettigen oder verzweigten Alkylenanteil mit 1 bis 6 Kohlenstoffatomen ist, werden hergestellt, indem man die entsprechenden Derivate mit R[tief]8 = Wasserstoff mit einem

umsetzt, worin das Halogen aus Chlor oder Brom besteht und R[tief]14 einen geradkettigen oder verzweigten Alkylrest mit 1 bis 6 Kohlenstoffatomen, den Phenylrest oder einen Phenylalkylenrest mit einem geradkettigen oder verzweigten Alkylenanteil mit 1 bis 6 Kohlenstoffatomen bedeutet. Die Umsetzung erfolgt in Gegenwart einer organischen Base, wie Pyridin, Chinolin oder Triethylamin, die als Lösungsmittel dient, während etwa 1 bis 24 Std. bei etwa 25 bis 100°C, unter der Maßgabe, dass vor der Reaktion die kleines Alpha-Aminogruppe des Hydroxy-substituierten Ausgangsmaterials durch eine geeignete Schutzgruppe geschützt wird, wie zum Beispiel den tert.-Butoxycarbonylrest oder den Benzyloxycarbonylrest, der anschließend durch Behandlung mit Säure, zum Beispiel Trifluoressigsäure, und Hydrogenolyse entfernt wird. Die bei diesem Verfahren verwendeten Säureanhydride und Säurehalogenide sind bekannt oder können aus entsprechenden Säuren nach bekannten Methoden hergestellt werden.
Die Verbindungen der Formel I, worin R[tief]1 Wasserstoff und R[tief]5 einen Rest der Formel -NY[tief]2Y[tief]3 darstellen, werden hergestellt durch Reduktion einer Verbindung, bei welcher R[tief]5 die Nitrogruppe bedeutet und die freie Aminogruppe geeignet geschützt ist. Der Schutz der freien Aminogruppe im Ausgangsmaterial erfolgt zum Beispiel durch Behandlung mit einem niederen Halogenameisensäurealkylester, wie Chlorameisensäuremethylester, einem Halogenameisensäurearylester, wie Chlorameisensäurebenzylester, oder einem niederen Alkancarbonsäurehalogenid, wie Acetylchlorid oder Propionylchlorid, oder mit einem aromatischen Säurehalogenid, wie Benzoylchlorid, auf an sich bekannte Weise. Die Reduktion wird mit Metallen, wie Zink oder
Zinn, und Säure, zum Beispiel Zink und Essigsäure oder Zink und einem niederen Alkohol, wie Ethanol ausgeführt. Die Reaktionszeit beträgt etwa 1 bis 24 Std. bei Temperaturen von etwa 25 bis 100°C unter Bildung von Verbindungen, worin Y[tief]2 und Y[tief]3 Wasserstoff bedeuten. Diese Verbindungen können alkyliert werden zu Verbindungen, worin Y[tief]2 und Y[tief]3 niedere Alkylreste sind, beispielsweise durch Umsetzung mit Boran/Natriumcyanid und Formaldehyd in einem niederen Alkohol, wie Methanol oder Ethanol während etwa 2 bis 48 Std. bei Temperaturen von etwa 25 bis 80°C. Die Aminschutzgruppe wird durch saure Hydrolyse entfernt, zum Beispiel durch Behandeln mit Salzsäure, Bromwasserstoffsäure oder Schwefelsäure bei Rückflusstemperatur während etwa 4 bis 24 Stunden.
Die Verbindungen der Formel I, worin R[tief]1 einen Alkylcarbonylrest mit einem geradkettigen oder verzweigten Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeutet, werden hergestellt, indem man die entsprechenden Derivate, worin R[tief]1 Wasserstoff bedeutet, mit einem Säurehalogenid der Formel
worin das Halogen zum Beispiel Chlor oder Brom ist und R[tief]15 einen geradkettigen oder verzweigten Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeutet, in Wasser in Gegenwart einer Base, wie Natriumhydroxid oder Natriumborat bei einer Temperatur von 0 bis 25°C 1/2 bis 6 Std. umsetzt.

atomen bedeutet, in Wasser in Gegenwart einer Base, wie Natriumhydroxid oder Natriumborat bei einer Temperatur von etwa 0 bis 25°C etwa 1/2 bis 6 Std. umsetzt.
Die Verbindungen der Formel I, worin R[tief]1 einen Rest der Formel
darstellt, worin R[tief]7 Wasserstoff, einen geradkettigen oder verzweigten niederen Alkylrest mit 1 bis 4 Kohlenstoffatomen, den Benzyl- oder p-Hydroxybenzylrest bedeutet, werden hergestellt, indem man das entsprechende Derivat mit R[tief]1 = Wasserstoff mit einer Säure der Formel
oder deren Anhydrid, deren Aminogruppe geeignet geschützt ist, zum Beispiel durch den Benzyloxycarbonylrest oder den tert.-Butoxycarbonylrest, und in der R[tief]7 die vorstehend angegebene Bedeutung besitzt, in einem Ether, wie Tetrahydrofuran oder Dioxan, in Methylenchlorid oder Chloroform und in Gegenwart eines Dehydratisierungsmittels bei Verwendung der freien Säure bei einer Temperatur von etwa 0 bis 35°C etwa 1 bis 12 Std. umsetzt, worauf die Schutzgruppen durch saure Hydrolyse entfernt werden.
Die einzelnen optischen Isomeren der Verbindungen der Formel I, worin R[tief]1 Wasserstoff bedeutet, können unter Verwendung eines (+) oder (-) Binaphthylphosphorsäuresalzes nach der Methode von R. Viterbo et al., Tetrahedron Letters 48, 4617 (1971) hergestellt werden. Auch andere Zerlegungsmittel wie (+)Campher-10-sulfonsäure können verwendet werden. Die einzelnen optischen Isomeren der Verbindungen der Formel I, worin R[tief]1 von Wasserstoff verschieden ist, werden dann wie vorstehend für das Racemat beschrieben hergestellt, wobei man lediglich von dem zerlegten Amin ausgeht.
Verbindungen der Formel IV, worin Y den FCH[tief]2-Rest bedeutet, werden hergestellt, indem man ein entsprechendes Benzyl-substituiertes Methylketon der Formel

worin R[tief]9, R[tief]10, R[tief]11, R'[tief]10 und R[tief]12 die für Formel IV angegebene Bedeutung besitzen und R[tief]16 ein abspaltbarer Rest wie ein Halogen, zum Beispiel Chlor, Brom oder Iod, der Mesylat-, Tosylat-, Triflat- oder Trifluoracetatrest ist, mit einem geeigneten Fluorierungsmittel, wie Kaliumfluorid, Silberfluorid, Cäsiumfluorid, Thalliumfluorid oder Tetrabutylammoniumfluorid in einem geeigneten Lösungsmittel, wie Dimethoxyethan, Dimethylsulfoxid, Dimethylformamid, Ethylenglycol, Acetonitril, Aceton, Benzol oder Fluorwasserstoff bei einer Temperatur von etwa 0 bis 200°C etwa 2 bis 48 Std. umsetzt. Der abgehende Rest R[tief]16 in Formel IX kann auch eine Diazogruppe sein, in welchem Fall man als Fluorierungsmittel Fluorwasserstoff/Pyridin einsetzt. Falls R[tief]16 eine Diazogruppe ist, sind geeignete Lösungsmittel aprotische Lösungsmittel, wie Diethylether, Tetrahydrofuran und Pentan, und die Reaktionszeit beträgt etwa 30 Min. bis 24 Std. bei etwa -20 bis 65°C. Beispielsweise wird ein geeignetes Diazoketon, das heißt eine Verbindung der Formel IX, worin R[tief]16 eine Diazogruppe darstellt, in einem geeigneten aprotischen Lösungsmittel zu einer auf -10°C gekühlten Lösung von Fluorwasserstoff in Pyridin zugegeben. Das Reaktionsgemisch wird 1 Std. bei -10°C und dann 2 Std. bei etwa 25°C kräftig gerührt und danach auf Eis gegossen. Die organische Phase wird abgetrennt, mit einer Base gewaschen, zum Beispiel mit Natriumbicarbonatlösung, über Magnesiumsulfat getrocknet und im Vakuum
eingeengt, wobei man das entsprechend substituierte Benzylfluormethylketon der Formel IV erhält.
Die Diazoketone, das heißt Verbindungen der Formel IX, worin R[tief]16 eine Diazogruppe darstellt, werden aus dem betreffenden Säurehalogenid hergestellt, das heißt einem entsprechend substituierten 2-Phenylacetylhalogenid, dessen Halogen zum Beispiel aus Chlor bestehen kann, indem man langsam dieses Säurehalogenid in einem aprotischen Lösungsmittel, wie Diethylether, Tetrahydrofuran, Pentan, Hexan, Benzol, Dimethoxyethan oder Dioxan zu einer auf etwa -40 bis 20°C gekühlten Diazomethanlösung in Ether zugibt und anschließend etwa 1 bis 24 Std. bei etwa 25°C kräftig rührt. Das dabei entstandene Diazoketon kann auf konventionelle Weise isoliert werden, zum Beispiel durch Abdunsten des Lösungsmittels und Reinigen durch Umkristallisieren oder Chromatographieren, oder es kann ohne Isolierung direkt mit einem Fluorierungsmittel auf vorstehend beschriebene Weise umgesetzt werden.
Das entsprechend substituierte Diazoketon kann auch verwendet werden zur Herstellung von Verbindungen der Formel IX, worin R[tief]16 zum Beispiel ein Halogen, den Mesylat-, Tosylat-, Triflat- oder Trifluoracetatrest bedeutet, wobei man an sich bekannte Methoden anwendet. Zur Herstellung von Verbindungen der Formel IX, worin R[tief]16 Halogen, wie zum Beispiel Chlor, Brom oder Iod bedeutet, wird die entsprechende Verbindung der Formel IX, worin R[tief]16 eine Diazogruppe ist, in einem geeigneten aprotischen Lösungsmittel mit wässrigem Chlorwasserstoff, Bromwasserstoff bzw. Iodwasserstoff umgesetzt. Zur Herstellung von Verbindungen der Formel IX, worin R[tief]16 den Mesylat-, Tosylat-, Triflat- oder Trifluoracetatrest bedeutet, wird das betreffende Diazoketon IX in einem geeigneten aprotischen Lösungsmittel mit verdünnter Schwefelsäure umgesetzt, wobei man
das Benzylmethanolketon erhält, das mit dem Säurechlorid oder Säureanhydrid der Methansulfonsäure, p-Toluolsulfonsäure, Trifluormethylsulfonsäure oder Trifluoressigsäure umgesetzt wird.
Das zur Herstellung des Diazoketons der Formel IX, d.h. Verbindungen, worin R[tief]16 eine Diazogruppe bedeutet, verwendete substituierte 2-Phenylacetylhalogenid wird aus der betreffenden Säure erhalten, zum Beispiel durch Behandlung mit Thionylchlorid in einem aprotischen Lösungsmittel, wie Diethylether, Tetrahydrofuran, Benzol oder Methylenchlorid bei einer Temperatur von etwa 0°C bis zur Rückflusstemperatur des Lösungsmittels während etwa 1 bis 24 Stunden. Man kann auch 1 Äquivalent der entsprechenden Säure mit 1 Äquivalent Oxalylchlorid in einem der obigen aprotischen Lösungsmittel bei einer Temperatur von etwa 0 bis 40°C etwa 1 bis 24 Std. umsetzen. Die angewandten entsprechend substituierten 2-Phenylessigsäurederivate sind bekannt oder können durch saure oder basische Hydrolyse eines geeignet substituierten Benzylnitrils in bekannter Weise hergestellt werden. Die substituierten Benzylnitrile sind bekannt oder können hergestellt werden, indem man das betreffende Benzylhalogenid, beispielsweise das Chlorid, mit Kaliumcyanid oder Natriumcyanid in einem protischen Lösungsmittel, wie Ethanol, Dimethoxyethan, Wasser oder Glycol, oder in einem aprotischen Lösungsmittel, wie Diethylether, Benzol, Hexamethylphosphortriamid, Dimethylsulfoxid, Dimethylformamid, Acetamid oder Dimethylacetamid etwa 1 bis 48 Std. bei etwa 0 bis 150°C umsetzt. Die geeignet substituierten Benzylhalogenide sind bekannt oder können aus den betreffenden substituierten Benzoesäuren oder Benzaldehyd-Derivaten, die bekannt sind, hergestellt werden. Beispielsweise kann man die Benzylhalogenide herstellen durch Reduktion des betreffenden Benzaldehyds mit Natriumborhydrid oder Lithiumaluminiumhydrid oder durch katalytische Reduktion, oder indem man den betreffenden Benzoesäureester mit Lithiumaluminiumhydrid oder Boran oder das entsprechende Benzoesäurederivat mit Lithium-
aluminiumhydrid reduziert und die so erhaltenen Benzylalkohole beispielsweise mit Thionylchlorid, Phosphoroxychlorid, Phosphortribromid oder Phosphorpentachlorid umsetzt.
Die Verbindungen der Formel IV, worin Y' den Difluormethylrest bedeutet, werden wie folgt hergestellt:
Aminoacetonitril wird mit einer Carbonylgruppen-haltigen Verbindung in an sich bekannter Weise zu einer Schiff'schen Base der Formel

umgesetzt, worin R[tief]b Wasserstoff, den Phenylrest, einen geradkettigen oder verzweigten Alkylrest mit 1 bis 8 Kohlenstoffatomen, die Methoxy- oder Ethoxygruppe, und R[tief]c den Phenylrest oder einen geradkettigen oder verzweigten Alkylrest mit 1 bis 8 Kohlenstoffatomen darstellen, oder R[tief]b und R[tief]c zusammen einen Alkylenrest mit 5 bis 7 Kohlenstoffatomen, das heißt einen Rest der Formel -CH[tief]2-(CH[tief]2)[tief]n-CH[tief]2-, worin n eine ganze Zahl von 3 bis 5 bedeutet, bilden. Beispiele für geradkettige oder verzweigte Alkylreste R[tief]b und R[tief]c mit 1 bis 8 Kohlenstoffatomen sind der Methyl-, Ethyl-, n-Propyl-, Isopropyl-, n-Butyl-, tert.-Butyl-, n-Hexyl-, n-Octyl- und Neopentylrest.
Bedeutet (a) R[tief]b Wasserstoff, so wird Aminoacetonitril mit Benzaldehyd oder einem Alkanal mit 1 bis 9 Kohlenstoffatomen in gerader oder verzweigter Kette, zum Beispiel mit 1-Propanal, 1-Butanal, 2,2-Dimethylpropan-1-al oder 2,2-Diethylbutan-1-al umgesetzt, bedeutet (b) R[tief]b den Phenylrest, so wird Aminoacetonitril mit Benzophenon oder einem Phenylalkylketon mit einem Alkylanteil von 1 bis 8 Kohlenstoffatomen in gerader oder verzweigter Kette, zum Beispiel mit Phenylmethylketon, Phenylethyl-
keton, Phenylisopropylketon, Phenyl-n-butylketon oder Phenyl-tert.-butylketon umgesetzt, bedeutet (c) R[tief]b einen geradkettigen oder verzweigten Alkylrest mit 1 bis 8 Kohlenstoffatomen, so wird Aminoacetonitril mit einem Dialkylketon mit geradkettigen oder verzweigten Alkylresten mit 1 bis 8 Kohlenstoffatomen, zum Beispiel Methylisopropylketon, Di-n-butylketon oder Methyl-tert.-butylketon umgesetzt. Die Carbonylgruppen-haltigen Verbindungen sind bekannt oder können nach bekannten Methoden hergestellt werden.
Bedeutet R[tief]b die Methoxy- oder Ethoxygruppe, so wird Aminoacetonitril mit einem Benzoylhalogenid, zum Beispiel Benzoylchlorid, oder einem Alkancarbonsäurehalogenid einer Alkancarbonsäure mit 1 bis 9 Kohlenstoffatomen in gerader oder verzweigter Kette, beispielsweise mit Acetylchlorid, Propionylchlorid, Butyrylchlorid, tert.-Butyrylchlorid, 2,2-Diethylbuttersäurechlorid oder Valerylchlorid bei 0°C in Äther, Methylenchlorid, Methylformamid, Methylacetamid oder Chlorbenzol in Gegenwart einer organischen Base, wie Triethylamin oder Pyridin, umgesetzt, worauf man das Reaktionsgemisch sich 1 Std. auf etwa 25°C erwärmen lässt. Das resultierende Amid-Derivat wird mit einem Alkylierungsmittel, wie Methylfluorsulfonat, Dimethylsulfat, Methyliodid, Methyl-p-toluolsulfonat oder Trimethyloxonium-hexafluorophosphat, falls R[tief]b die Methoxygruppe ist, oder mit Triethyloxonium-tetrafluoroborat, falls R[tief]b die Ethoxygruppe ist, bei etwa 25°C in einem chlorierten Kohlenwasserstoff, wie Methylenchlorid, Chlorbenzol oder Chloroform umgesetzt unter etwa 12- bis 20-stündigem Erhitzen des Reaktionsgemischs am Rückfluss. Das Gemisch wird dann auf etwa 25°C abgekühlt und mit einer organischen Base, wie Triethylamin oder Pyridin versetzt, worauf die Lösung mit Kochsalzlösung extrahiert und das Produkt isoliert wird.
Bilden R[tief]b und R[tief]c zusammen einen Alkylenrest mit 5 bis 7 Kohlenstoffatomen, so wird Aminoacetonitril mit einem der cyclischen Alkanone Cyclopentanon, Cyclohexanon oder Cycloheptanon in an sich bekannter Weise zu einer Schiff'schen Base umgesetzt.
1 Äquivalent dieser Schiff'schen Base wird mit 1 Äquivalent einer starken Base, wie einer Alkyllithiumverbindung, zum Beispiel Butyllithium oder Phenyllithium, einem Lithiumdialkylamid, wie zum Beispiel Lithiumdiisopropylamid oder mit Lithiumamid, Kalium-tert.-butylat, Natriumamid, einem Metallhydrid, wie Natriumhydrid, tertiären Amin, wie Triethylamin, Lithiumacetylid oder Dilithiumacetylid, mit Natrium- oder Lithiumhexamethyldisilazan umgesetzt, worauf Behandlung mit einem Alkylierungsmittel der Formel VI folgt. Die Alkylierungsreaktion kann in einem aprotischen Lösungsmittel, wie Benzol, Ether, Tetrahydrofuran, Dimethylsulfoxid, Hexamethylphosphortriamid, Ethylenglycoldiethylether oder Dimethoxyethan erfolgen. Die Reaktionszeit beträgt etwa 1/2 bis 24 Std. bei Temperaturen von etwa -120 bis 65°C.
Die so erhaltene alkylierte Schiff'sche Base wird dann mit einer starken Base behandelt, wobei ein Carbanion entsteht, das mit einem geeigneten Halogenmethyl-Alkylierungsmittel umgesetzt und dann sauer hydrolysiert wird. Geeignete starke Basen sind solche, die ein Proton vom zur Nitrilgruppe kleines Alpha-ständigen Kohlenstoffatom abziehen und die vorstehend bereits aufgeführt wurden. Geeignete Halogenmethyl-Alkylierungsmittel sind Chlordifluormethan, Bromdifluormethan und Difluoriodmethan. Die Umsetzung kann in einem aprotischen Lösungsmittel, wie Benzol, Toluol, Ethern, wie Tetrahydrofuran oder Diethylether, Dimethylsulfoxid oder Hexamethylphosphortriamid erfolgen. Die Reaktionstemperatur beträgt etwa -120 bis
65°C, und vorzugsweise 40°C, die Reaktionsdauer etwa 1/2 bis 24 Stunden.
Das Difluormethylderivat wird mit einem geringen Überschuss eines Hydrazinderivats, wie Phenylhydrazin oder N,N-Dimethylhydrazin in einem Lösungsmittel, wie einem Ether, zum Beispiel Tetrahydrofuran oder Diethylether, in chlorierten Kohlenwasserstoffen, wie Methylenchlorid, Chloroform oder Tetrachlorkohlenstoff oder niederen Alkoholen, wie Methanol oder Ethanol umgesetzt. Die Umsetzung erfolgt bei Temperaturen von etwa 0°C bis zur Rückflusstemperatur des Lösungsmittels während etwa 1 bis 72 Std., wobei das freie Aminonitril entsteht, das zum Keton der Formel IV hydrolysiert wird. Die Hydrolyse erfolgt vorzugsweise in niederen Alkoholen, wie Methanol oder Ethanol, die Cupri- oder Ferroionen enthalten (CuSO[tief]4 . 5H[tief]2O oder FeSO[tief]4 . 7H[tief]2O), bei Rückflusstemperatur während etwa 5 Min. bis 6 Std., oder durch Behandlung mit verdünnter wässriger Säure, wie Salzsäure oder Schwefelsäure bei Temperaturen von etwa 25 bis 100°C während etwa 1 bis 24 Stunden.
Die Verbindungen der Formel VIII werden hergestellt, indem man eine Verbindung der Formel

worin Xa ein Halogen, wie Chlor oder Brom bedeutet, mit Triphenylphosphin in einem Lösungsmittel, wie Kohlenwasserstoffen, zum Beispiel Benzol oder Toluol, oder niederen Alkoholen, wie Methanol oder Ethanol, in Acetonitril, Tetrahydrofuran, Diethylether oder Dimethoxyethan etwa 10 Min. bis 48 Std. umsetzt. Beim Abkühlen entsteht ein Niederschlag,
der mit Lösungsmitteln gewaschen und umkristallisiert wird, zum Beispiel aus Ethylacetat, Acetonitril oder einem niederen Alkohol, wobei man das entsprechend substituierte Benzylphosphoniumsalz erhält. Dieses kann auch hergestellt werden, indem man den entsprechend substituierten Benzylalkohol mit einem Triphenylphosphoniumsalz, zum Beispiel Triphenylphosphoniumbromid, umsetzt, wobei die vorstehend erwähnten Reaktionsbedingungen angewandt werden. Das geeignet substituierte Benzyltriphenylphosphoniumsalz wird zu einem Überschuß (bis zu 25 %) metallischem Natrium oder Lithium, gelöst in flüssigem Ammoniak, zugegeben, wozu eine katalytische Menge Ferrinitrat zugesetzt wird, wobei man etwa 10 Min. bis 3 Std. rührt, dann wird der Ammoniak unter inerter Atmosphäre, wie Stickstoff oder Argon, abgedampft. Nach Zusatz eines geeigneten Lösungsmittels, wie Benzol, Toluol, Diethylether, Tetrahydrofuran oder Dimethoxyethan wird das resultierende Benzylidenphosphoran gesammelt. Es wird mit einem Ester, zum Beispiel einem niederen Alkylester, wie dem Methyl-, Ethyl-, n-Propyl-, Isopropyl- oder n-Butylester der Trifluoressigsäure in einem Lösungsmittel, wie Benzol, Toluol, Diethylether, Tetrahydrofuran oder Dimethoxyethan unter inerter Atmosphäre, wie Stickstoff oder Argon, bei einer Temperatur von etwa 0°C bis zur Rückflußtemperatur des Lösungsmittels etwa 30 Min. bis 24 Std. umgesetzt, dann wir das Reaktionsgemisch eingeengt und destilliert, wobei man das Olefin erhält, das mit wässriger Mineralsäure, wie Salzsäure oder Bromwasserstoffsäure oder einer organischen Säure, wie Trifluoressigsäure oder p-Toluolsulfonsäure, unter Verwendung eines Co-Lösungsmittels, wie Tetrahydrofuran, Diethylether oder Benzol etwa 30 Min. bis 24 Std. bei etwa 0°C bis zur Rückflußtemperatur des Lösungsmittels behandelt wird. Die angewandte Säuremenge kann von katalytischen Mengen bis zur konzentrierten Säure variieren. Die Herstellung der Verbindungen der Formel VIII ist in allgemeiner Form von P.H. Betmann, Ang. Chem., Int. Ed. 1966, S. 308, beschrieben worden.
Die nachfolgenden Beispiele dienen der weiteren Erläuterung der vorliegenden Erfindung.
Beispiel 1
7-((4-((1-Difluormethyl-2-(3-hydroxyphenyl)ethyl)aminomethyl)phenyl)acetylamino)-3-acetyloxymethyl-8-oxo-5-thia-1-azabicyclo[4.2.0]oct-2-en-2-carbonsäure
Ein Gemisch aus 1 g 3-Acetyloxymethyl-7-[[2-[4-(chlormethyl)phenyl]acetyl]amino]-8-oxo-5-thia-1-azabicyclo[4.2.0]oct-2-en-2-carbonsäure und 1 g 1-Difluormethyl-2-(3-hydroxyphenyl)ethylamin in 50 ml Ethanol wurde 24 Std. bei 25°C gerührt, dann wurde das Lösungsmittel entfernt, wobei ein Rückstand erhalten wurde. Dieser wurde an Silicagel chromatographiert unter Eluieren mit Benzol/Aceton; dabei erhielt man die Titelverbindung.
Beispiel 2
Gemisch für harte Gelatinekapseln:
(a) 1-Difluormethyl-2-(3-hydroxyphenyl)ethylamin-hydrochlorid 20 mg
(b) Talkum 5 mg
(c) Lactose 90 mg
Die Zubereitung wurde hergestellt, indem man die trockenen Pulver (a) und (b) durch ein feinmaschiges Sieb trieb und gut vermischte. Das Pulver wurde dann in harte Gelatinekapseln in einer Menge von 115 mg/Kapsel eingefüllt.
Beispiel 3
Gemisch für Tabletten:
(a) 1-Difluormethyl-2-(3,4-dihydroxyphenyl)ethylamin-hydrochlorid 20 mg
(b) Stärke 43 mg
(c) Lactose 45 mg
(d) Magnesiumstearat 2 mg
Das durch Vermischen der Lactose mit der Verbindung (a) und einem Teil der Stärke und Granulieren mit Stärkepaste erhaltene Granulat wurde getrocknet, gesiebt und mit dem Magnesiumstearat vermischt. Das Gemisch wurde zu Tabletten von 110 mg verpresst.
Beispiel 4
Injizierbare Suspension für 1 ml-Ampullen zur intramuskulären Injektion:

(a) 1-Difluormethyl-2-(4-hydroxyphenyl)ethylamin-hydrochlorid 1,0
(b) Polyvinylpyrrolidon 0,5
(c) Lecithin 0,25
(d) Wasser für Injektionszwecke auf 100,0
Die Komponenten (a) bis (d) wurden vermischt, homogenisiert und in 1 ml-Ampullen eingefüllt, die verschlossen und 20 Min. bei 121°C im Autoklaven behandelt wurden. Jede Ampulle enthielt pro ml 100 mg der neuen Verbindung (a).
Beispiel 5
1-Fluormethyl-2-(4-methoxyphenyl)ethylamin
Unter einer Stickstoffatmosphäre und unter Rühren mit einem Magnetrührer wurde eine Lösung von 0,1 Mol p-Methoxybenzylchlorid in Tetrahydrofuran (130 ml) langsam (20 Tropfen pro Min.) zu 4,8 g Magnesiumspänen in Tetrahydrofuran (90 ml) zugesetzt. Die Bildung des Grignard-Reagenzes wurde durch Zusatz von 2 Tropfen Methyliodid eingeleitet. Während der Reaktion wurde der Kolben in ein Bad, das Wasser bei Raumtemperatur enthielt, eingetaucht. Das Rühren wurde weitere 30 Minuten fortgesetzt. Anschließend wurde das Grignard-Reagenz von dem überschüssigen Magnesium abgetrennt, in einen zweiten Kolben gebracht und auf -20°C (innere Temperatur) gekühlt. 6,35 g Fluoracetonitril in 45 ml Tetrahydrofuran wurden langsam in einer solchen Geschwindigkeit zugesetzt, daß die innere Temperatur bei -20°C gehalten wurde.
Nach Zugabe wurde das Gemisch in eine Lösung von 4,16 g Natriumborhydrid in wässrigem Methanol (350 ml Methanol und 7 ml Wasser) gegossen und 90 Min. bei -20°C gehalten, dann wurde mit konzentrierter HCl auf etwa pH 2 angesäuert und das Meiste des Lösungsmittels unter vermindertem Druck entfernt. 235 ml 3 N HCl wurden zugesetzt und das Gemisch wurde zweimal (2 x 200 ml) mit Ether extrahiert, anschließend stark alkalisch gemacht (NaOH) und erneut mit Ether (4 x 200 ml) extrahiert. Nach Trocknen (Na[tief]2SO[tief]4) und Entfernen des Lösungsmittels wurde 1-Fluormethyl-2-(4-methoxyphenyl)ethylamin erhalten.
Beispiel 6
1-Fluormethyl-2-(3,4-dimethoxyphenyl)ethylamin
Wurde im Verfahren von Beispiel 5 das dort verwendete p-Methoxybenzylchlorid durch eine entsprechende Menge 3,4-Dimethoxybenzylchlorid ersetzt, so wurde 1-Fluormethyl-2-(3,4-dimethoxyphenyl)ethylamin erhalten.
