tX-Halogenmethylamin-Derivate und Verfahren zu deren Herstellung
Die vorliegende Erfindung betrifft neue pharmazeutisch brauchbare^-Halogenmethylamin-Derivate sowie Verfahren
zu deren Herstellung.
Die Aminosäuren Tryptophan, 5-Hydroxytryptophan3 3>4~Di~
hydroxyphenylalanin (DOPA), Tyrosin und Phenylalanin werden metabolytisch durch eine Decarboxylase für aromatische
Aminosäuren in Tryptamin, 5-Hydroxytryptamin, 3,^-Dihydroxyphenethylamin
oder Dopamin, Tyramin bzw. Phenethylamin
überführt. Es wird angenommen, daß das Decarboxylase-Enzym für aromatische Aminosäuren nicht-spezifisch ist,
insbesondere soweit es sich um periphere Katalyse handelt. Es liegen jedoch Hinweise dafür vor, daß im Gehirn sowohl
für DOPA als auch für 5-Hydroxytryptophan spezifische Decarboxylierungsenzyme vorliegen.

Die vorstehend erwähnten aromatischen Amine sind bekanntlich
. an verschiedenen pathophysiologischen Vorgängen beteiligt. Beispielsweise wurde gefunden, dass Tryptamin, das Decarboxylierungsprodukt
von Tryptophan, enzymatisch zum Monoinethyltryptamin
methyliert wird, das seinerseits in roten Blutzellen,
Plasma und Blutplättehen enzymatisch zum Dimethyltryρtarnin
(DMT) methyliert wird. Das methylierende Enzym liegt bei zahlreichen
Säugetierarten vor und es konnte gezeigt werden, dass es im Hirngewebe mehrerer Arten wie auch beim Menschen erzeugt
wird. Dimethyltryptamin, das starke llaluzinogene oder psycho-,
mimetische Eigenschaften besitzt, kann bei der Entstehung von
Schizophrenie und anderen psychotischen Zuständen eine Rolle spielen. Ein.Mittel, das die Bildung von Dimethyltryptamin ■
blockiert, könnte daher als antipsychotisches Mittel nützlich , sein. Die Blockierung der Decarboxylierung von Tryptophan führt
zu erniedrigten Tryptaminspiegeln, womit das Substrat, zur Dimethyl
tryptamin-Bildung beseitigt v/ird. Ein Inhibitor der aromatischen
Arninosauredecarboxylase, der die' Umwandlung von Tryptophan
in Tryptamin blockiert, könnte daher als antipsychotisches

Mittel brauchbar sein. " ■ ·
Sowohl 5-Hydroxytryptamin (5-HT), das Decarboxylierungsprodukt
von 5-Hydroxytryptophan, als auch 3,4-Dihydroxyphenethylamin
(Dopamin), das Decarboxylierungsprodukt von DOPA.sind an perizentralen.
. . · pheren und physiologischen Vorgängen beteiligt, und Stoffe,
die die Spiegel dieser Amine X'jirksam kontrollieren, erwiesen
sich als bräuchbare pharmakologische Mittel. Es konnte gezeigt
werden, dass die zentralehl §pfegeirian 5-HT und Norepinephrin, ■'
>ytdas metaboliisch durch Hydroxylierung von Dopamin gebildet
wird, bei Patienten mit manischen krankheiten höher sind als bei Menschen ohne derartige Krankheiten. Es· konnte ferner
gezeigt werden, dass Stoffe, die die zentralen Spiegel der Monoamine herabsetzen, zum Beispiel 5-HT und insbesondere Norepinephrin,
bei Verabreichung an Menschen antimanische Eigenschaften erzeugen, während Stoffe, die den ivionoaminspiegel
erhöhen, bei dafür zugänglichen Menschen Geisteskrankheiten ·
auslösen können. Daher können Stoffe, die die Bildung von 5-HT und D, op am in blockieren, zum Beispiel durch Inhibierungdes
aromatischen Aminosäuredecarboxylase-Enzyms, das 5-Hydroxy- tryptophan
und DOPA in 5-HT bzw. Dopamin überführt, als antipsychotische Mittel oder Tranquilizer bei der Behandlung von
Geisteskrankheiten nützlich sein. ■ ■

Es konnte ferner gezeigt werden, dass Stoffe, die die Decarb- .
oxylierung von. DOPA zu Dopamin inhibieren, brauchbar sind zur Behandlung der Parkinson-Krankheit bei gleichzeitiger Verab- ■"_"·
reichung von exogenem DOPA oder L-DOPA. Es wird angenommen, dass die Parkinson-Krankheit zumindest zum Teil auf verminderte
zentrale Dppaminspiegel zurückgeht, da die exogene Verabreichung von DOPA oder L-DOPA ein xvirksames Mittel zur Behandlung dieser
Krankheit darstellt. Da exogen verabreichtes DOPA jedoch rasch enzymatisch peripher in .Dopamin umgewandelt wird, muss man grosse·

.Mengen verabreichen, um zentral eine erhöhte Absorption zu
erzielen. DOPA überschreitet die Blut/Hirn-Schranke rasch,
im Gegensatz zu Dopamin. Die Verabreichung von DOPA oder L-DOPA in Verbindung mit einem peripher aktiven Inhibitor
des Enzyms,-, das DOPA in Dopamin umwandelt, vermindert die.
Menge an L-DOPA, die verabreicht werden muss, damit ange-" .· messene Mengen zur zentralen Absorption zirkulieren. V7eitere
Vorteile werden erzielt, wenn man einen aromatischen Aminosäuredecarboxylase-Inhibitor
zusammen mit L-DOPA verabreicht. Indem man die periphere Bildung von Dopamin verhindert, '
können dem Dopamin zugeschriebene Ne benwir klingen^ wie Herz-'a*!L;fcythmie,
Übelkeit und Erbrechen vermieden v/erden. · '.'
Untersuchungen haben gezeigt, dass die 5-Hydroxytryptaminspiegel
bei Patienten mit depressivem Syndrorn niedriger sind
als bei Menschen ohne solche Syndrome. Auch ist die Verabreichung.
von exogenem L-5-Hydroxytryptophan (L-5-HTP) bei
der Behandlung bestimmter depressiver Patienten erfolgreich. Wie beim DOPA- muss man' jedoch auch, da L-5-HTP peripher

leicht zum 5.-HT metabolisiert wird, grosse Mengen an L-5-HTP
verabreichen, um erhöhte zentrale Spiegel der Aminosäure zu "
erzielen. Es wurde festgestellt, dass bei Verabreichung eines • Inhibitors der aromatischen Aminosäuredecarboxylase, die die
periphere Bildung von 5-HT aus 5-HTP. katalysiert, die Menge
an zur Erzielung erhöhter zentraler Spiegel benötigtem exo--.genem
5-HTP spürbar herabgesetzt wird. Somit erwiesen sich die Inhibitoren aromatischer Aminosäuredecarboxylase bei
Verwendung in Verbindung mit exogenem 5-HTP. als brauchbar zur Behandlung von Depressionen. . / , . "
Stoffe, die die periphere Umwandlung von 5-HTP in 5-HT · . .-blockieren,
können brauchbar sein zur Behandlung anderer Zustände,, die auf erhöhte zentrale Spiegel von 5-HTP ansprechen,
als Ergebnis exogener Verabreichung von 5-HTP.

Es wurde festgestellt, dass exogenes L-5-HTP. zur Behandlung
von Myoklonien geeignet ist- Untersuchungen haben auch ergeben, dass die Verabreichung von exogenem 5-HTP gegen Schlaflosigkeit
nützt. Gleichzeitige Verabreichung von 5-HTP und einem Inhibitor aromatischer Aminosäuredecafboxylase kann
daher zur Behandlung dieser Krankheiten nützlich sein.
Die Blockierung der peripheren Bildung von 5-Hydroxytryptamin ■■.·,-kann
weitere Vorteile bringen,· da bekanntlich 5-HT.z.B.eine Rolle
spielt bei der Entstehung von rheumatoider Arthritis und dem karzinoiden.Syndrom durch erhöhten Kollagenspiegel. Auch wurde
berichtet, dass. 5-HT das primäre Autocoid ist, das für anaphylactoide
Reaktionen beim Menschen und Bronchienverengung bei Asthmatikern verantwortlich ist, und Stoffe, die die
Bildung von 5-HT antagonisieren oder inhibieren sind brauchbar
zur Behandlung dieser Krankheiten. 5-HT verursacht bekanntlich Blutplättchenaggregat ion und wird als verursachender-Paktor
bei dem Dumping-Syndrom nach Gastrectomie und Migräne angesehen. Methylsergid, ein 5-Hydroxytryptarnin-Antagonist,
.·erwies sich als wirksam bei der Behandlung des Dumping-Syndroms
nach Gastrectomie. .
Es wurde vermutet, dass Phenethylamin, das Decarboxylierungsprodukt
von Phenylalanin, als endogene Verbindung zu schizophrenen Symp-fc.omen beiträgt und Migräne auslöst. Ferner wurde
vermutet, dass endogenes Tyramln, welches das Decarboxylierungs-.
produkt von Tyrosin ist, zu Anfallkrankheiten beiträgt.
Ss ist somit ersichtlich,"dass Stoffe, die zur Regulierung '
der Spiegel aromatischer Aminosäuren und Amine brauchbar sind, in zahlreichen pharmakologischen" Situationen Verwendung finden.
Die erfindungsgemässen Verbindungen sind Inhibitoren der aromatischen
Aminosäurendecarboxylase, die Tryptophan, 5-Hydroxytryptophan, 3,4-Dihydroxyphenylalanin, Tyrosin und Phenylalanin

in die entsprechenden Amine überführt, und sie stellen damit
brauchbare pharmakologische Mittel dar.
£>ie Monoamine Adrenalin, Noradrenalin, Dopamin und 5-Hydroxytryptamin
sind an der chemischen Übertragung Von Nervenimpulsen sowohl im Zentralnervensystem als auch im Nervensystem der
Peripherie beteiligt, ferner haben sie hormonale Aufgaben» Es besteht hinreichender Verdacht, dass die vorstehend genannten
Monoamine an zahlreichen pathophysiologischen Vorgängen beteiligt sind. Beispielsweise wurde gefunden., dass
im Hirn von Patienten mit depressiven Zuständen Defizit an Noradrenalin und/oder 5-Hydroxytryptamin herrscht« Bekanntlich
wird die effektive Konzentration dieser Monoamine ge- ' :
steuert durch die.Geschwindigkeit ihrer Herstellung,, durch die
Geschwindigkeit des enzymatischen Abbaus (Monoaminoxydase)
und durch ihre Wiederaufnahme durch die Nervenenden nach der Freigabe durch den Nervenimpuls. Jedes Mittel, das den
enzymatischen Abbau oder den Wiederaufnahrnevorgang inhibiert«,
erhöht daher die effektive Konzentration dieser Amine an der ■ gewünschten Wirkungsstelle» Ein Mittel<>
das die Abgabe dieser Monoamine stimuliert (sympathomimetischer Effekt)s würde ebenfalls
die effektive" Konzentration an der erforderlichen Hirkungsstelle
erhöhen. Jedes Mittel, das daher eine oder mehrere · dieser Effekte besitzt,,, kann als Antidepressivum brauchbar
sein. So erwiesen sieh'zum Beispiel Pargylin (Monoaminoxydase.-Ihhibitor)
und Imipramin (Inhibitor der" Monosmin-Aufnahme)
als wirksame Äntldepressiva.. ·
Es wurde gefunden, dass die Parkinson-Krankheit verknüpft ist mit einem Defizit des Dopamin-Gehalts in den Basalganglien des
Gehirnsi Jeder Stoff, der" die «äopaminergische Aktivität der
Easal-Ganglien direkt oder indirekt erhöht, müsste daher zur
■ Behandlung der Parkinson-Krankheit geeignet sein» So erwiesen
sich zum Beispiel Deprenyl (ein Monoaminoxydase-Inhibitor) und

;otropin (ein Inhibitor der Dopamin-Äufnähme) als geeignet
zur Behandlung von Parkinsonismus.
Die Entstehung der Fettleibigkeit ist zwar unbekannt, doch gibt es erhebliche Hinweise darauf, dass 5-Hydroxytryptamin·
bei der Zügelung des Appetits eine wichtige Rolle spielt. Ausserdem erwiesen sich Stoffe, die die 5-Hydroxytryptaminkonzentration
im Hirn verändern, geeignet zur Verwendung als anprexaSeIiß.ttel. Dopamin wurde ferner als wichtiger Faktor
bei der Regulierung des Essverhaltens erkannt, woraus pharmazeutisch brauchbare Mittel resultierten. So erwiesen sich zum
Beispiel Fenflu_ramin (das die 5-Hydroxytryptamin-Konzentration
im Hirn verändert) und Amphetarnin (das Dopamin im Hirn freisetzt)
als wirksame Appetitzügler. ·
Ferner ist bekannt, dass Veränderungen der Aktivität des ■ dopaminergischen Systems mit einer aäalgetischen Wirkung . '

Das periphere Nervensystem ist wichtig bei der Regulierung.
der Aktivität des cardiovaskulären Systems, und insbesondere Nerven, die Noradrenalin verwenden, spielen eine fundamentale
Rolle bei der Steuerung von Gefässdruck und Kerzgeschwindigkeit.. Stoffe, die die Aktivität dieser Nerven durch eine
syiupathomimetische Wirkung erhöhen, erhöhen daher Blutdruck
und Herzgeschwindigkeit. Eine derartige Wirkung ist erwünscht bei der Behandlung hypotensiver Krisen, beispielsweise nach
schiverem Blutverlust. Sowohl Adrenalin als auch Noradrenalin

erwiesen sich in dieser Hinsicht als brauchbar. Umgekehrt '
ist bekannt, dass Noradrenalin im Hirn eine Depress orwirkung auf das cardiovaskuläre System und insbesondere auf den Blutdruck
ausübt. Stoffe, die primär im Hirn das Noradrenalinsystera aktivieren, sollten daher brauchbare antihypertensive.
Mittel sein.
Sämtliche vorstehend erwähnten Monoamine müssen zunächst von Lagerstellen in Nerven oder in endokrinem Gewebe. freigesetzt
v/erden, worauf ihre Wirkung beendigt wird entweder durch Wiederaufnahme von den Nerven oder durch enzymatischen
Abbau zu inaktiven Metaboliten. Stoffe, die auf ein oder mehrere dieser Systeme wirken, das heisst entweder Monoamine
aus Nerven freisetzen, die Monoaminrezeptoren direkt aktivieren, die Wie der aufnähme blockieren und/oder den enzyraatischen.
Abbau der Monoamine inhibieren, oder die mehr . als eine dieser V7irkungen zeigen, könnten zu Mitteln führen,

Die vorliegende Erfindung betrifft auch pharmazeutisch akzeptable Salze und einzelne optische Isomere
der Verbindungen der Formel I.
Die Verbindungen der Formel I sind als pharmakoiogische
Mittel brauchbar; sie stellen Inhibitoren für aromatische Aminosäuredecarboxyiase dar. Sie sind
ferner als Zwischenprodukte zur Herstellung brauchbarer pharmakoiogischer Mittel geeignet.
Unter einem Alkylcarbonylrest wird ein Rest derjEOrinel Alkyl-C-verstanden,'
wobei der Alkylanteil geradkettig oder verzweigt ist und 1 bis "6 Kohlenstoffatome aufweist.
Unter dem Benzoylrest wird der Rest der Formel
verstanden.
Unter «inem Phenylaxkylencarbonylrest wird ein Rest der
Formel
verstanden, dessen Alkyienanteil geradkettig oder verzweigt sein kann und 1 bis 6 Kohlenstoffatome besitzt,
wie zum Beispiel der Methylen-, Ethylen-,Isopropylen- oder Butylenrest.

Beispiele für geradkettige oder verzweigte Alkylreste mit .
Ibis 8 Kohlenstoffatomen sind der Methyl-, Ethyl-, n-Propylr,
Isopropyl-, η-Butyl-, tert.-Butyl-, n-Pentyl- Und n-Octylrest.
Beispiele für niedere Alkylreste Y2 und Y-, mit 1 bis % Kohlenstoffatomen
sind der Methyl-, Ethyl-, n-Propyl-, Isopropyl- und " n-Butylrest.
Beispiele für pharmazeutisch .akzeptable Salze, erfinduhgsgemässer
Verbindungen sind nicht-toxische Säureadditionssalze mit anorganischen Säuren^wie Salzsäure, Bromwasserstoffsäure,
Schwefelsäure und Phosphorsäure, oder mit organischen Säuren, wie Methansulfonsäure, Salicylsäure, Maleinsäure, Malonsäure,
weinsäure, Zitronensäure und Ascorbinsäure. Die. Salze werden in konventioneller Weise hergestellt. .
Bevorzugte erfindungsgemässe Verbindungen sind solche der
Formel I, worin R, Wasserstoff oder einen Alkylcarbonylrest . mit einem geradkettigen oder verzweigten Alkylanteil mit
1 bis 4 Kohlenstoffatomen bedeutet, wobei Verbindungen, bei . denen R, Wasserstoff bedeutet, besonders bevorzugt sind.
Sine weitere bevorzugte Ausführungsform der Erfindung sind Verbindungen der Formel I, worin R.,,. R^, R_, R'h und Rg - ·- '. '

Die Verbindungen der Formel I sind irreversible Inhibitoren
des Enzyms, das metabolytisch die Umwandlung von Tryptophan,
5-Hydroxytryptophan, 5,4-Dihydroxyphenylalanin, Tyrosin und ·
Phenylalanin zu Tryptamin, 5-Hydroxytryptarain, 3,^-Dihydroxyphenylethylatnin,
Tyramin bzw. Phenethylamin katalysiert. Wie £.-bereits
erwähnt, lassen Untersuchungen annehmen,, dass das für die periphere Umwandlung obiger Aminosäuren zu den Aminen
verantwortliche Enzym eine nicht-spezifische Decarboxylase für aromatische Aminosäuren ist. Bei der zentralen Unn*andlung
sind spezifische Becarboxylasen für die Umwandlung, von 5-ifydroxytryptophan
bzw. 3,4-Dihydroxyphenylalanin verantwortlich, Viährend

Zur Erzielung der gewünschten Effekte können die erfindungsgemässen
Verbindungen auf verschiedene Arten verabreicht werden. Die Verbindungen können allein oder in Form pharmazeutischer
Fräparate oral oder parenteral, zum Beispiel subkutan,
intravenös oder intraperitoneal gegeben werden. Die ' . .'
Verbindungen können auch intranasal verabreicht -werden oder auf Schleimhäute, zum Beispiel der Nase, des Rachens und
der Bronchien appliziert werden, beispielsweise in einem . Aerosol-Spray, der kleine Teilchen einer erfindungsgemässen
Verbindung in Sprühlösung oder trockener Pulverform enthält.
Als zu verabreichende Menge eignet sich jede wirksame Menge. Je nach dem Patienten, dem zu behandelnden Zustand und der
Art der Verabreichung, schwankt die zu verabreichende Menge innerhalb eines breiten Bereiches. Werden die Verbindungen
der Formel I verabreicht'zur Bewirkung einer peripherenirreversiblen
Inhibierung aromatischer Aminosäuredecarboxylase oder zur zentralen Inhibierung von Moncaminoxydase, so liegt
öle wirksame Menge bei etwa 0,1 mg bis 100 mg/kg Körpergenricht/
Dosis, und vorzugsweise bei etwa 5 bis 25 mg/kg Körpergewicht.
Zum Beispiel kann der gewünschte Effekt erzielt werden mit
einer Dosiseinheit-wie Tabletten, die 10 bis 250 mg einer
erfindungsgemässen Verbindung enthalten und 1- bis 4-mal / täglich
genommen werden. Werden die Verbindungen der Formel I verabreicht zur Erzielung einer zentralen irreversiblen ·
Inhibierurig von aromatischer Aminosäuredecarboxylase oder ' ^,^-Dihydroxyphenylalanin-Decarboxylase, so liegt die wirk- .·".'■"
same Menge bei etwa 100 bis 500 mg/kg Körpergewicht/Tag, und ! ·
vorzugsweise bei etwa/(i'50)bis 300 mg/kg Körpergewicht. Zum
Beispiel kann der gewünschte zentrale inhibierende Effekt erzielt werden, indem man eine'Dosiseinheit, zum Beispiel
Tabletten mit etwa 550 bis 500 mg einer erfindungsgemässen
Verbindung, ein- bis viermal täglich einnimmt. ·

Die feste Dosiseinheit kann konventioneller Art sein» Beispielsweise
kann eine übliche Gelantinekapsel vorliegen^ die die erfindungsgeraäße Verbindung und einen Trägers zum
Beispiel Gleitmittel und inerte Füllstoffe, wie Lactose s
Rohrzucker und Maisstärke enthält. Gemäß einer weiteren Ausführungsform werden die erfindungsgemäßen Verbindungen
mit konventionellen Tablettenbasen wie Lactose3Rohrzucker
oder Maisstärke und Bindemittel^ wie Gummi acacia,, Maisstärke
oder Gelatines Sprengmitteln.,wie Maisstärke„
Kartoffelstärke oder Alginsäure und einem Gleitmittel wie Stearinsäure oder Magnesiumstearat tablettiert.
Zur parenteralen Verabreichung können die Verbindungen verabreicht
werden in Form injizierbarer Dosen einer Lösung oder Suspension der betreffenden Verbindung in einem physiologisch
zulässigen Verdünnungsmittel mit einem pharmazeutischen Träger.,, der eine" sterile Flüssigkeit^wie V/asser '
oder ein Öl sein kann.,, gegebenenfalls in Verbindung mit
einem oberflächenaktiven Mittel und anderen pharmazeutisch
zulässigen Hilfsstoffen» Beispiele für zur Herstellung derartiger Präparate geeignete Öle sind solche aus Erdöl- Ölen
tierischen^, pflanzlichen oder synthetischen Ursprungs^ beispielsweise

Die Reduktion der Ketone der Formel IV zum entsprechenden Alkohol erfolgt chemisch, wobei man zum Beispiel 1 bis 10
Äquivalente eines Metallhydrids, wie Lithiumborhydrid,
Natriumborhydrid, Natriumcyanborhydrid oder Lithiumaluminiumhydrid,
Boran oder Dimethylthioboran als Reduktionsmittel verwendet, oder katalytisch, zum Beispiel mit Raney-Nickel,
Rhodium, Palladium auf Kohle oder Platinoxid. Die gesamte Reaktionszeit beträgt etwa 10 Min. bis 24 Std. und die
Temperatur liegt bei etwa -40 bis 100 0C, je nach dem verwendeten
Reduktionsmittel. Bei chemischer Reduktion beträgt die Reduktionszeit im allgemeinen etwa 10 Min. bis 24 Std.,
wobei die Temperaturen etwa -40 bis 65 0C betragen. Geeignete
Lösungsmittel zur chemischen Reduktion von Verbindungen der allgemeinen Formel IV sind niedere Alkohole, wie
Methanol oder Ethanol oder Ether, wie Diethylether oder Tetrahydrofuran. Bei katalytischer Reduktion beträgt die
Reaktionszeit etwa 1 bis 24 Std., die Reaktionstemperatur liegt bei etwa 25 bis 100 0C und der Druck bei 1 bis
120 bar. Geeignete Lösungsmittel zur katalytischem Reduktion der Verbindungen der Formel IV sind niedere Alkohole, wie
Methanol oder Ethanol, Essigsäure oder Ethylacetat. Die chemische Reduktion wird bevorzugt.
Die Hydrolyse zum Amin und Entfernung sämtlicher Schutzgruppen
am Ring erfolgt mit einer starken Mineralsäure, wie Salzsäure, Bromwasserstoffsäure oder Schwefelsäure, oder
mit einer organischen Säure, wie Toluolsulfonsäure oder TrI-fluoressigsäure,
in Wasser bei Rückflusstemperatur während etwa 4 bis 48 Std., oder unter Verwendung von beispielsweise
1 bis 3 Äquivalenten Hydrazin, Methylhydrazin oder Methylamin'bei einer Temperatur von etwa 25 0G bis Rückflusstemperatur
während etwa 1 bis 12 Std.; gegebenenfalls erfolgt dann Behandlung mit einer starken Mineralsäure oder
organischen Säure, wie vorstehend beschrieben, oder Entfernung der Schutzgruppen durch Hydrogenolyse.

Base behandelt, zum Beispiel einem Metallhydrid, wie Natriumhydrid oder Kaliumhydrid, Natriumamid oder Lithiumamid,
Lithiumdiisopropylamid, Lithium- oder Natriumhexamethyldisilazan,
Lithiumacetylid oder Lithiumcarbid, und
zwar in einem aprotischen Lösungsmitteln wie Tetrahydrofuran,
Diethylether, Dimethoxyethan, Glyme, BenzoI^ HexamethyI-phosphoramid
oder Dimethylsulfoxid. Dann erfolgt Umsetzung mit einem geeigneten Difluormethylhalogenalkylierungsmittel,
zum Beispiel mit Chlordifluormethan, Bromdifluormethan
oder Difluoriodmethan. Die Umsetzung erfolgt bei Temperaturen von etwa -20 bis 60 0C, und vorzugsweise etwa
25 0C, die Reaktionszeit beträgt etwa 15 Min. bis 24 Stunden.
Das Difluormethylderivat wird mit einer starken Mineralsäure behandelt, zum Beispiel mit Salzsäure oder Schwefelsäure in
Wasser, oder mit einer starken organischen Säure, zum Beispiel p-Toluolsulfonsäure oder Trifluormethylessigsäure
in Wasser bei Temperaturen von etwa 25 bis 180 0C. während
etwa 1/2 bis 48 Std., wobei man die entsprechende 2-Difluormethyl-j5-(substituiert-phenyl)-propionsäure
erhält. Ist eine der niederen Alkylestergruppen ein tertiärer Butylrest, so wird das Difluormethylderivat vorzugsweise mit Trifluormethylessigsäure

decarbojjcyliert, zum Beispiel mit Essigsäure oder Propionsäure
bei Temperaturen von etwa 100 bis l60°C und Reaktionszeiten von etwa 1 bis 2k Stunden. Vorzugsv/eise wird das
Monoesterderivat decarboxyliert durch Behandlung mit
Essigsäure bei etwa 130 0C während 12 Std. Beim Einengen
bei vermindertem Druck erhält man den entsprechenden niederen Alkylester der 2-Difluormethyl-3-(substituiert-phenyl)-propionsäure,
der mit einer starken Mineralsäure, wie Salzsäure oder Schwefelsäure, oder einer organischen Saure,
wie p-Toluolsulfonsäure oder Trifluormethylessigsäure in
Wasser bei Temperaturen bislzur Rückflußtemperatur des
Lösungsmittels während etwa 1 bis 24 Std. hydrolysiert wird. Die freie Säure wird unter den Bedingungen der Schmidt'sehen
Reaktion oder der Curtius-Reaktion in das Aminderivat überführt , siehe zum Beispiel Organic Reactions, Bu.Ill,
S0 308 bzw. 338. Unter den Bedingungen der Schmidt'sehen
Reaktion wird die freie Säure etwa 1 bis 2k Std. bei Temperaturen von etwa O bis 60 0C mit Stickstoffwasserstoffsäure
umgesetzt. Unter den Bedingungen der Curtius-Reaktion wird die freie Säure

mit einem Thiony!halogenid, zum Beispiel Thinylchlorid bei
Rückflusstemperatur etwa 1 bis 4 Std. umgesetzt, oder das Natriumsalz der Säure wird mit Oxalylchlorid zum entsprechenden

Reduktionsmittel. Das Keton der Formel VIII kann auch durch reduktive Alkylierung oder Aminierung mit überschüssigem
Ammoniak und einem Katalysator, wie Platin, Raney-Nickel, oder Palladium in niederen Alkoholen, wie Methanol, Ethanol
oder tert.-Butanol, oder Ethern, wie Diethylether oder
Tetrahydrofuran oder deren Gemischen mit Wasser bei einer Temperatur von etwa 25 bis 200 0C, und vorzugsweise etwa
100 bis I50 0C unter hohen Drucken von beispielsweise etwa
52 bis l40 bar in das entsprechende Amin umgewandelt werden.

mit Thionylchlorid in einem aprotischen Lösungsmittel, wie Diethylether, Tetrahydrofuran . Benzol oder I-lethylenchlorid
bei einer Temperatur von etwa 0 0C bis zur Rückflusstemperatur
des Lösungsmittels während etwa 1 bis 24 Stunden. Man kann auch 1 Äquivalent der entsprechenden Säure mit
1 Äquivalent Oxalylchlorid in einem der obigen aprotischen Lösungsmittel bei einer Temperatur von etwa 0 bis 40 0C
etwa 1 bis 24 Std. umsetzen. Die angewandten entsprechend substituierten 2-Phenyiessigsäurederivate sind bekannt oder
können durch saure oder basische Hydrolyse eines geeignet substituierten Benzylnitrils in bekannter Weise hergestellt
werden. Die substituierten Benzylnitriie sind bekannt oder können hergestellt werden, indem man das betreffende Benzylhalogenid,
beispielsweise das Chlorid, mit Kaliumcyanid oder Natriumcyanid in einem protischen Lösungsmittel, wie
Ethanol, Dimethoxyethan, Wasser oder Glycol, oder in einem aprotischen Lösungsmittel, wie Diethylether, Benzol, Hexamethyiphosphortriamid,
Dimethylsulfoxid, Dimethylformamid, Acetamid oder Dimethy!acetamid etwa 1 bis 48 Std. bei etwa
0 bis 150 0C umsetzt. Die geeignet substituierten Benzylhalogenide

wobei die vorstehend erwähnten Reaktionsbedingungen angewandt werden. Das geeignet substituierte Benzyltriphenylphosphoniumsalz
wird zu einem Überschuß ( bis zu 25 /ümetallischem
Natrium oder Lithium,gelöst in flüssigem Ammoniak, zugegeben, wozu eine katalytische Menge Perrinitrat zugesetzt
wird, wobei man etwa 10 Min,, bis 3 Std. rührt, dann wird
der Ammoniak unter inerter Atmosphäre, wie Stickstoff oder Argon, abgedampft. Nach Zusatz eines geeigneten Lösungsmittels,
wie Benzol, Toluol, Diethylether, Tetrahydrofuran oder Dimethoxyethan wird das resultierende Benzylidenphosphoran
gesammelt. Es wird mit einem Ester, zum Beispiel einem niederen Alkylester, wie dem Methyl-,Ethyl-,n-Propyl-, Isopropyl-
oder n-Butylester der Trifluoressigsäure in einem Lösungsmittel, wie Benzol, Toluol, Diethylether, Tetrahydrofuran
oder Dimethoxyethan unter inerter Atmosphäre, ,wie Stickstoff oder Argon, bei einer Temperatur von etwa 0 0C bis zur
Rückflußtemperatur des Lösungsmittels etwa 30 Min. bis
2k Std. umgesetzt, dann wird das Reaktionsgemisch eingeengt
und destilliert, wobei man das Olefin erhält, das mit wässriger Mineralsäure, wie Salzsäure oder Bromwasserstoffsäure
oder einer organischen Säure, wie Trifluoressigsäure oder p-Toluolsulfonsäure, unter Verwendung eines Co-Lösungsmittels,
wie Tetrahydrofuran, Diethylether oder Benzol etwa 30 Min. bis 2k Std. bei etwa 0 0C bis zur Rückflußtemperatur
des Lösungsmittels behandelt wird. Die angewandte Säuremenge kann von katalytischen Mengen bis zur konzentrierten
Säure variieren. Die Herstellung der Verbindungen der Formel VIII ist in allgemeiner Form von P.H. Betmann,
Ang. Chem.Int. Ed* 1966, S. 308, beschrieben worden.

Unter einer Stickstoffatmosphäre und unter Rühren mit
einem Magnetrührer wurde eine Lösung von 0,1 Mol p-Methoxybenzylchlorid
in Tetrahydrofuran (130 ml) langsam (20 Tropfen pro Min.) zu Ί,8 g Magnesiumspänen'
in Tetrahydrofuran (90 ml) zugesetzt.Die Bildung des
Grignar.d-Reagenzes wurde durch Zusatz von 2 Tropfen Methyliodid eingeleitet. Während der Reaktion wurde
der Kolben in ein Bad, das Wasser bei Raumtemperatur enthielt , eingetaucht. Das Rühren wurde weitere 30
Minuten fortgesetzt . Anschließend wurde das Grignard-Reagenz von dem überschüssigen Magnesium abgetrennt,
in einen zweiten Kolben gebracht und auf.-20 0C
(innere Temperatur) gekühlt. 6,35 g Fluoracetonitril in 4-5 ml Tetrahydrofuran wurden langsam in einer solchen
Geschwindigkeit zugesetzt, daß die innere Temperatur bei -20 0C gehalten wurde..
Nach Zugabe wurde das Gemisch in eine Lösung von M,l6 g
Natriumborhydrid in wässrigem Methanol (350 ml Methanol und 7 ml Wasser) gegossen und 90 Min. bei -20 0C, gehalten,·
dann wurde mit konzentrierter HCl auf etwa pH 2 angesäuert und das Meiste des Lösungsmittels unter vermindertem
Druck entfernt. 235 ml 3 N HCl wurden zugesetzt und das Gemisch wurde zweimal (2 χ 200 ml ) mit Ether
extrahiert, anschließend stark alkalisch gemacht (NaOH) und erneut mit Ether (4 χ 200 ml) extrahiert. Nach Trocknen
(Na2SO1J) und Entfernen des Lösungsmittels wurde 1-Pluormethyl-2-(4-methoxyphenyl)ethylamin
erhalten.

Behandlung des so erhaltenen Propionsäure-iberivates bei Temperaturen von etwa 0° bis 60° G mit Stickstoffwasserstoffsäure unterwirft oder das Bropionsäure-^erivat in das Säurehalogenid überführt, das: etwa 40 bis 100 Stunden bei Rückflußtemperaturen mit Natriumazid behandelt wird und anschließend das erhaltene Produkt der Säurehydrolyse unterwirft, (c) wenn Y einen Rest der Formel , F,C- und R1 ein Wasserstoffatom bedeuten, ein Keton der Formelworin R1J einen Rest der Formel -CF- bedeutet und Rq Ri0» Rll» Rl10 und R12 die in Tabelle HB angegebenen Bedeutungen besitzen, in einem niederen alkoholischen Lösungsmittel oder Wasser oder Kombinationen derselben in Gegenwart einer mineralischen oder organischen Base. bei etwa 20° bis 100° C etwa 1 bis 8 Stunden mit einem Hydroxylaminsalz behandelt und das dabei erhaltene entsprechende Oximzum Amin reduziert,(d) wenn R1 einen Alkylcarbonylrest, wobei der Alkylrest 1 bis ί Kohlenstoffatome aufweist und gerad- oder verzweigtkettig ist, bedeutet, das entsprechende Derivat,030045/0793worin R1 ein Wasserstoffatom bedeutet, in einem geeigneten Lösungsmittel in Gegenwart einer Base etwa eine halbe Stunde bis 6 Stunden bei einer Temperatur von etwa O bis 25 0C mit einem Säurehalogenid der FormelR15-C-HaIoworin Halo ein Halogenatom und R1,- einen gerad- oder verzweigt kett igen Alkylrest mit 1 bis H Kohlenstoffatomen bedeuten, behandelt,(e) wenn R1 einen Alkoxycarbonylrest, wobei der Alkoxyteil 1 bis 1J Kohlenstoffatome aufweist und gerad- oder verzweigtkettig ist, bedeutet, das entsprechende Derivat, worin R1 ein Wasserstoffatom bedeutet, in Wasser in Gegenwart einer Base etwa eine halbe Stunde bis 6 Stunden bei einer Temperatur von etwa 0 bis 25 0C mit einem Halogenalkylformiat der FormelIlHaIo-C-OR16,worin Halo ein Halogenatom und R1^ einen gerad- oder verzweigtkettigen Alkylrest mit 1 bis 4 Kohlenstoffatomen bedeuten, behandelt,(f) wenn R1 einen Rest der FormelIl-C-CH-R7 NH230 045/0793BAD OFttG»NAL .worin Ry die in Anspruch 1 angegebene Bedeutung besitzt, bedeutet, das entsprechende Derivat, worin R^ ein Wasserstoffatora bedeutet, in einem geeigneten Lösungsmittel und in Gegenwart eines Dehydratisierungsmittels, wenn die freie Säure angewandt wird, etwa 1 bis 12 Stunden bei einer Temperatur von etwa 0 bis 35 0C mit einer Säure der FormelHOOC-CH-R7-t 7 NHnworin R7 die vorstehend angegebene Bedeutung besitzt und die Aminogruppe in geeigneter Weise geschützt ist, oder einemAnhydrid derselben behandelt und das dabei erhaltene Produkt der sauren und basischen Hydrolyse unterwirft und(g) wenn ein pharmazeutisch akzeptables Salz gewünscht wird, die so erhaltene Verbindung mit einer pharmazeutisch akzeptablen Säure umsetzt.0300 4 5/0^03*BAD ORIGINAL
