Description
Die Erfindung bezieht sich auf ein Verfahren zur mikrobiellen Herstellung von Alkoholoxidase, bei dem
fakultativ Methanol verwertender Hefe der Bezeichnung Hansenula polymorpha, die sich in einer
anorganische Nährstoffe, Vitamine sowie als Kohlenstoff- und Energiequelle Methanol enthaltenden Nährlösung befindet, kontinuierlich weitere Nährlösung
zugeführt wird, und bei dem aus dem so gebildeten Kulturmedium Zellmasse und somit in der Zellmasse
enthaltene Alkoholoxidase in einer der zugeführten Menge der Nährlösung entsprechenden Menge entnommen wird, wobei die die Hefezellen enthaltende, in
einem Reaktionsgefäß befindliche und einen pH-Wert von 4,0 bis 6,0 aufweisende Nährlösung auf einer
Reaktionstemperatur von 25 bis 45°C gehalten und mit
Luft oder einem Luft-Sauerstoffgemisch begast wird.
Das Enzym Alkoholoxidase katalysiert folgende Reaktion:
Dieses Enzym kann daher zur Gewinnung von Formaldehyd sowie von Wasserstoffperoxid aus Methanol verwendet werden. Es kann ferner zur analytischen
Bestimmung der Alkohole Methanol, Äthanol. N-Propanol und N-Butanol eingesetzt werden, wobei als
Meßeinrichtung beispielsweise Enzymelektroden Verwendung finden.
Es ist bekannt, daß in Methanol verwertenden Hefen, wie Candida, Hansenula, Pichia und Torulopsis-Stämmen, beim Wachstum auf Methanol eine Flavin-Adenosin-Dinucleotid (FAD) enthaltende Alkoholoxidase
induziert wird, welche die Oxidation des Methanols zu Formaldehyd im Stoffwechsel katalysiert.
Es ist ferner bekannt, zur Gewinnung des Enzyms Alkoholoxidase die Hefen Candida boidinü und
Hansenula polymorpha zu verwenden. Dabei wird die Hefe Candida boidinü in batch-Kulturen auf Mineral
salzmedium mit Methanol als Kohlenstoff-Energiequel
le und den Vitaminen Biotin und Thiamin angezogen. Diese Methode ist jedoch wenig effektiv. Man hat daher
auch, um eine höhere Zelldichte zu erzielen, im Laufe des Wachstums der Zellen dem Kulturmedium weiteres
ίο Methanol zugeführt (vergleiche hierzu Reuß M. et al.
»Chemie-Ingenieur-Technik, 46, 6f 9 [74]«). Nach Aufschluß der auf diese Weise herangezogenen Hefezellen
erhält man eine spezifische Aktivität der Alkoholoxidase im Rohextrakt von 0,3 bis 1,0 Enzymeinheit pro mg
Protein.
Bei der Gewinnung von Alkoholoxkj-se aus der
Methanol verwertenden Hefe Hansenula polymorpha wurde das Kulturmedium unter kontinuierlicher Zuführung einer Nährlösung herangezogen, welche Methanol
und die Vitamine Biotin und Thiamin enthält. Die Menge an Alkoholoxidase beträgt bei diesem bekannten
Verfahren bei einer Wachstumslimitierung durch die Kohlenstoffenergiequelle Methanol und bei einer
Wachstumsrate von 0,16 h-' (dies entspricht
VoIx Voh1 χh~') etwa 7% des Gesamtproteins, was
einer spezifischen Aktivität im Rohextrakt von 2,5 bis 3,0 Enzymeinheiten/mg Protein entspricht. Bei einer
Wachstumsrate von 0,03 h-' unter gleichen Kulturbedingungen lag die Alkoholoxidasemenge bei etwa 20%
bezogen auf das Gesamtprotein, so daß die spezifische
MicrobioUll,137[76]«).
Reindarstellung rforderliche Isolierung des Enzyms geschieht üblicherweise mittels Säulenchromatographie
und Enzymfällung.
Es ist Aufgabe der Erfindung, ausgehend von dem bekannten Verfahren, bei dem die Hefe Hansenula
polymorpha eingesetzt wird, ein Verfahren zur Herstellung von Alkoholoxidase zu schaffen, das eine höhere
Ausbeute an Alkoholoxidase pro gebildetem Protein ergibt als dies nach den bekannten Verfahren möglich
ist.
Die der Erfindung zugrunde liegende Aufgabe wird gemäß der Erfindung dadurch gelöst, daß die zusätzlich
als Kohlenstoff- und Energiequelle wenigstens ein auf die Alkoholoxidase nicht katabolitrerrimierend wirkendes Substrat, wie Glyzerin, Sorbit oder Xylose, in einer
Konzentration von 0,1 bis 1.0% enthaltende Nährlösung
mit einer Durrhflußrate von 0,03 bis 0.15 h -' zugegeben wird. Die während der kontinuierlichen Prozeßführung,
welche unter chemostatischen Bedingungen erfolgt, dem Kulturmedium entnommene Zellmasse wird
sodann in an sich bekannter Weise aufgeschlossen und das Enzym Alkoholoxidase isoliert.
Die erfindungsgemäße Verwendung eines Mischsubstrats, das aus Methanol und aus wenigstens einem auf
die Alkoholoxidase nicht katabolitreprimierend wirken
den Substrat besteht, führt in überraschender Weise zu
wesentlich höheren Enzymeinheiten. Dabei werden bei der Durchführung des Verfahrens gemäß der Erfindung
Zellen produziert, die eine höhere Alkoholoxidase-Aktivität enthalten als Zellen, die nach den bisher bekannten
Verfahren kultiviert werden. Dabei hat sich als vorteilhaft erwiesen, eine Nährlösung zu verwenden, die
Phosphat, Nitrat oder Kalium in einer das Wachstum der Zellen limitierenden Dosierung enthält. Bei dieser
Verfahrensvariante werden noch höhere Ausbeuten an Alkoholoxidase erzielt
AusfGhrungsbeispiel 1
In ein 12-Liter-Gefäß, das ein Arbeitsvolumen von 7 Liter umfaßt, wurde eine Nährlösung folgender
Zusammensetzung gegeben:
5,0 g/l Xylose
4,0 g/l Methanol
0,047 g/l KH2PO4
0,013 g/l NaH2PO4
0,125 g/l Na2SO4
0,4 g/I K2SO4
04 g/l (NH4J2SO4

0,05 g/l MgSO4 · 7 H2O

Der pH-Wert der Nährlösung betrug 5,0; die Temperatur der Lösung lag bei etwa 37° C.

25 Die Nährlösung wurde mit einer Rate von 1 vvm (1 l/min · 1 Reaktorvolumen) belüftet und mechanisch
mit einer Rührerdrehzahl von 1000 UpM gerührt. In der
in dem Reaktionsgefäß befindlichen Nährlösung wurde die Hefe Hansenula polymorpha der Stammnummer
CBS 4732 kultiviert
Der in dem Reaktionsgefäß gebildeten Hetekultur
wurde sodann Nährlösung der vorgenannten Zusammensetzung in einer Zuflußrate von 0,05 h~' zugeführt
und dem Reaktionsmedium in entsprechender Menge Zellmasse entnommen.
Bei dieser Verfahrensweise konnten zirka 30 000 Alkoholoxidase-Einheiten pro Liter Medium entnommen
werden, was — auf Zellmasse bezogen — etwa 28 Einheiten/mg Protein entspricht
Ausführungsbeispiel 2
Wie in Ausführungsbeispiel 1 beschrieben, wurde mit einer Nährlösung die der in Ausführungsbeispiel 1
weitgehend entsprach, anstelle von 5,0 g/l Xylose jedoch 5 g/l Glyzerin enthielt, ein Kulturmedium
gebildet.
Bei dieser Verfahrensweise konnten zirka 25 000 Alkoholoxidase-Einheiten pro Liter Medium gewonnen
werden, was etwa 22 Einheiten/mg Protein entspricht.
Claims (2)
1. Verfahren zur mikrobiellen Herstellung von
Alkoholoxidase, bei dem fakultativ Methanol verwertender Hefe der Bezeichnung Hansenula polymorpha, die sich in einer anorganische Nährstoffe,
Vitamine sowie als Kohlenstoff- und Energiequelle Methanol enthaltenden Nährlösung befindet, kontinuierlich weitere Nährlösung zugeführt wird, und bei
dem aus dem so gebildeten Kulturmedium Zellmasse und somit in der Zellmasse enthaltene Alkoholoxidase in einer der zugeführten Menge der Nährlösung
entsprechenden Menge entnommen wird, wobei die die Hefezellen enthaltende, in einem Reaktionsgefäß
befindliche und einen pH-Wert von 4,0 bis 6,0 aufweisende Nährlösung auf einer Reaktionstemperatur von 25 bis 45° C gehalten und mit Luft oder
einem Luft-Sauerstoffgemisch begast wird, dadurch gekennzeichnet, daß die zusätzlich
als Kohlenstoff- und Energiequelle wenigstens ein auf die Alkoholoxidase nicht katabolitreprimierend
wirkendes Substrat, wie Glyzerin, Sorbit oder Xylose, in einer Konzentration von 0,1 bis 1,0%
enthaltende Nährlösung mit einer Durchflußrate von 0,03 bis 0,15 h~' zugegeben wird.
2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß die Nährlösung Phosphat, Nitrat oder
Kalium in einer Jas Wachstum der Zellen limitierenden Dosierung enthält.
