Description
Die Erfindung betrifft den Gegenstand der Ansprüche.
Bevorzugt sind die Verbindungen der allgemeinen Formel I, in
der R eine Hydroxylgruppe, R₁ ein Wasserstoffatom oder einen
Niederalkanoylrest, R₁ insbesondere ein Wasserstoffatom oder
eine Acetylgruppe, R₂ ein Wasserstoffatom oder einen Nie
deralkylrest, insbesondere ein Wasserstoffatom oder eine
Methylgruppe bedeutet, wobei R₂ mit besonderer Bevorzugung
ein Wasserstoffatom darstellt, R₃ ein Wasserstoffatom oder
einen Niederalkanoylrest, insbesondere R₃ ein Wasserstoffatom
oder eine Acetylgruppe bedeutet.
Der Ausdruck "Niederalkylrest" bedeutet einen unverzweigten oder
verzweigten Kohlenwasserstoffrest mit 1 bis 7 Kohlenstoffatomen,
beispielsweise die Methyl-, Ethyl-, Propyl-, Isopropyl-,
Butyl-, Isobutyl-, tert.-Butyl-, Pentyl- und Isopentylgruppe.
Der Ausdruck "Niederalkoxyrest" bedeutet einen entsprechenden
Rest mit 1 bis 7 Kohlenstoffatomen, beispielsweise die
Methoxy-, Ethoxy-, Propoxy-, Isopropoxy-, Butoxy-, Isobutoxy-
und tert.-Butoxygruppe.
Bei den vorgenannten Definitionen sind die Reste mit 1 bis 4
Kohlenstoffatomen bevorzugt. Besonders bevorzugt sind die Reste
mit 1 und 2 Kohlenstoffatomen. Als Niederalkoxyrest ist die
Methoxy- und die tert.-Butoxygruppe bevorzugt.
Der Ausdruck "Niederalkanoylrest" bedeutet Acylreste von Fettsäuren
mit bis zu 7 Kohlenstoffatomen, beispielsweise die
Acetyl-, Propionyl- und Butyrylgruppe, wobei die Acetylgruppe
bevorzugt ist.
Die Verbindungen der allgemeinen Formel I können auf verschiedene
Weise hergestellt werden. Gemäß einer bevorzugten Methode
wird ein Aminosäurederivat der allgemeinen Formel III mit einer
Säure der allgemeinen Formel IV in an sich bekannter Weise
acyliert, wobei die Säure der allgemeinen Formel IV vor ihrer
Umsetzung mit dem Aminosäurederivat der allgemeinen Formel III
in ein aktiviertes Carbonsäurederivat überführt wird. Beispiele
für derartige aktivierte Derivate sind ein gemischtes Anhydrid,
symmetrisches Anhydrid, Säurechlorid, aktivierter Ester, Wood
ward-Reagens K, N,N'-Carbonylbisimidazol und N-Ethoxycarbonyl-
2-ethoxy-1,2-dihydrochinolin (EEDQ).
Falls R einen Niederalkoxyrest bedeutet, kann das vorgenannte
Verfahren oder ein entsprechendes anderes bekanntes Verfahren
(vgl. beispielsweise Houben-Weyl, Methoden der organischen Chemie,
Bd. XV, Teile 1 und 2 (1974) angewandt werden.
Ist die erhaltene Verbindung ein Ester, beispielsweise wenn R
die tert.-Butoxygruppe bedeutet, so kann dieser durch Behandeln
mit einer Säure, wie Trifluoressigsäure, in die entsprechende
freie Säure (R bedeutet eine Hydroxylgruppe) überführt werden.
Umgekehrt kann auch die freie Säure in üblicher Weise verestert
werden.
Die Ausgangsverbindungen der allgemeinen Formel IV,
sind von der Aminosäure Cystein abgeleitet, die in üblicher
Weise hergestellt werden kann.
Eine bevorzugte Methode zur Herstellung von Verbindungen der
allgemeinen Formel I erfolgt durch Austausch des Halogenatoms
in Verbindungen der allgemeinen Formel V,
in der X ein Halogenatom, vorzugsweise ein Chlor- oder Bromatom,
bedeutet, mit einer Thiolsäure der allgemeinen Formel
R₃-COSH.
Die Verbindungen der allgemeinen Formel I weisen zwei Asymmetrie
zentren auf. Dementsprechend existieren diese Verbindungen
in diastereoisomeren Formen oder in racemischen Gemischen. Alle
derartigen Erscheinungsformen dieser Verbindungen sind Gegenstand
der Erfindung. Für die vorgenannten Synthesen können als
Ausgangsstoffe die entsprechenden Racemate oder eines der
Enantiomeren eingesetzt werden. Bei Verwendung eines racemischen
Ausgangsstoffs können die im Endprodukt vorliegenden
Stereoisomeren durch übliche Methoden der Chromatographie oder
fraktionierten Kristallisation getrennt werden. Im allgemeinen
ist das L-Isomere (hinsichtlich des Kohlenstoffatoms der Amino
säure) die bevorzugte Isomerform. Die in der allgemeinen
Formel I angegebenen Sterne bezeichnen asymmetrische Kohlenstoffatome.
Die Verbindungen der allgemeinen Formel I können mit Säuren oder
Basen Salze bilden. Als Säuren können sehr verschiedene anorganische
und organische Säuren eingesetzt werden. Dabei werden
beispielsweise Hydrohalogenide, vorzugsweise Hydrochloride und
Hydrobromide, Sulfate, Nitrate, Borate, Phosphate, Oxalate,
Tartrate, Maleate, Citrate, Acetate, Ascorbate, Succinate,
Benzolsulfonate, Methansulfonate, Cyclohexansulfamate und
Toluolsulfonate hergestellt.
Die Salze werden in üblicher Weise durch Umsetzen der entsprechenden
freien Säuren mit einem oder mehreren Äquivalenten der
entsprechenden Säure oder Base erhalten, wobei das gewünschte
Anion oder Kation in einem Medium zugeführt wird, in dem das
Salz unlöslich ist. Die Salzbildung kann auch im Wasser erfolgen,
wobei anschließend das Wasser durch Gefriertrocknen abgetrennt
wird. Durch Behandeln eines derartigen Salzes mit einer
unlöslichen Säure, wie einem Kationenaustauscherharz in der H-Form
(beispielsweise einem Polystyrolsulfonsäureharz, wie
Dowex 50, Mikes, Laboratory Handbook of Chromatographic
Methods, Van Nostrand, 1961, S. 256), Eluieren mit einer
flüchtigen Puffersubstanz, wie einem Gemisch aus Pyridin und
Essigsäure, und Extrahieren mit einem organischen Lösungsmittel
kann die entsprechende freie Säure erhalten und gegebenenfalls
in ein anderes Salz überführt werden.
Die erfindungsgemäßen Verbindungen hemmen die Umwandlung des
Decapeptids Angiotensin I in Angiotensin II und sind deshalb
wertvoll zur Verminderung oder Beseitigung von durch Angiotensin
bedingtem Hochdruck. Die Wirkung des Enzyms Renin auf
Angiotensinogen, ein Pseudoglobulin im Blutplasma, führt zur
Bildung von Angiotensin I. Dieses wird durch das Angiotensin-
umwandelnde Enzym (ACE) in Angiotensin II überführt. Letzteres ist eine wirksame blutdruckerhöhende Substanz, die mit als ur
sächlich für mehrere Formen von Hochdruck bei verschiedenen
Säugetierarten, wie Ratten und Hunden, angesehen worden ist.
Die erfindungsgemäßen Verbindungen greifen in die Reaktions
folge Angiotensinogen → Angiotensin I → Angiotensin II durch
Hemmen des Angiotensin-umwandelnden Enzyms ein und vermindern oder
verhindern die Bildung der blutdruckerhöhenden Substanz Angio
tensin II.
Die durch die Verbindungen der allgemeinen Formel I hervorgerufene
Hemmung des Angiotensin-umwandelnden Enzyms kann in vitro
mit aus Kaninchenlungen isoliertem Angiotensin-umwandelndem
Enzym (vgl. Biochem. Pharmacol., Bd. 20 (1971), S. 1637) und
an exzitierter glatter Muskulatur (vgl. Federation Proc., Bd.
31 (1972), S. 511) gemessen werden. Dabei erweisen sich diese
Verbindungen als starke Inhibitoren der kontraktilen Wirksamkeit
von Angiotension I und als Verstärker der kontraktilen Wirksamkeit
von Bradykinin.
Aus Chemical Abstracts, Bd. 83 (1975), 143 573p ist ein Hemmstoff
des Angiotensin-umwandelnden Enzyms bekannt. Dieser
Hemmstoff ist das Nonapeptid SQ 20,881. Wie sich jedoch auch
aus Science, Bd. 196 (1977), S. 441 linke Spalte 1. Absatz
ergibt, ist dieses bisher bekannte ACE-hemmende Nonapeptid
bei oraler Verabreichung in keiner toxisch annehmbaren Menge
und Form an Warmblütlern wirksam. Dagegen weist die erfindungsgemäße
Verbindung SQ 14,735 (Verbindung von Beispiel 2)
bei oraler Verabreichung eine ausgeprägte ACE-Hemmwirkung an
normotensiven Ratten auf (vgl. Diagramm).
Die Verbindungen der Erfindung werden einzeln oder in Kombination,
gegebenenfalls in Form entsprechender Salze, in üblichen
Dosen verabreicht. Modellversuche an Tieren sind in Proc. Soc.
Exp. Biol. Med., Bd. 143 (1973), S. 483 beschrieben.
Die Verbindungen der allgemeinen Formel I lassen bei kontrollierter
Verwendung und Dosierung keine toxischen
Symptome erwarten.
Die Verabreichung der erfindungsgemäßen Verbindungen erfolgt
vorzugsweise oral, kann jedoch auch parenteral, wie subcutan,
intramuskulär, intravenös oder intraperetoneal, vorgenommen werden.
Die erfindungsgemäßen Verbindungen können in üblicher Form,
beispielsweise in Form von Tabletten, Kapseln, Elexieren zur
oralen Verabfolgung oder in Form steriler Lösungen oder Suspensionen
zur parenteralen Verfolgung angewandt werden. Dabei
können übliche Mengen, z. B. 10 bis 500 mg einer dieser Verbindungen
oder ein Gemisch dieser Verbindungen, gegebenenfalls
auch in Form entsprechender Salze, mit üblichen Trägerstoffen
und/oder Verdünnungsmitteln und/oder Hilfsstoffen zu Arzneimitteln
formuliert werden.
Die folgenden Beispiele erläutern die Herstellung der erfindungsgemäßen Verbindungen.
Eine in einem Eisbad gekühlte Lösung von 0,85 g L-Prolin-
tert.-butylester und 0,67 g Hydroxybenzotriazol in 10 ml Me
thylenchlorid wird zuerst mit 1,03 g Dicyclohexylcarbodiimid
und dann mit 1,7 g N-tert.-Butyloxycarbonyl-S-p-methoxybenzyl-
D-cystein versetzt. Nach 15 Minuten wird das Eisbad weggenommen,
und das Gemisch wird 16 bis 18 Stunden bei Raumtemperatur
gerührt. Der gebildete Niederschlag wird abfiltriert. Das Filtrat
wird mit 10prozentiger Kaliumbisulfatlösung, Wasser, gesättigter
Natriumbicarbonatlösung und wieder Wasser gewaschen.
Die organische Phase wird dann getrocknet und unter vermindertem
Druck eingedampft. Man erhält die Titelverbindung als
Öl. Rf = 0,2 (Kieselgel, Chloroform).
Eine in einem Eisbad gekühlte Lösung von 1,8 g der gemäß Bei
spiel 1 erhaltenen Verbindung und 4,4 ml Anisol in 8 ml Di
chlormethan wird mit 6,0 g Trifluormethansulfonsäure versetzt.
Nach Wegnehmen des Eisbads wird das Gemisch 30 Minuten bei
Raumtemperatur gerührt. Das Dichlormethan wird unter vermindertem
Druck abdestilliert, und der Rückstand wird zweimal mit
je 200 ml Hexan digeriert. Der Rückstand wird in Wasser gelöst
und zweimal mit Diethylether extrahiert. Die wäßrige Phase
wird auf eine mit 200 ml eines Kationenaustauscherharzes in
der H-Form (Dowex 50) gegeben. Die Säule wird dann mit Wasser ge
waschen, bis keine Säure mehr eluiert wird. Mit einem Puffersystem
aus Pyridin und Essigsäure vom pH-Wert 6.5 wird die
Titelverbindung eluiert. Ausbeute 0,66 g. Rf = 0,38 (Kieselgel;
Gemisch aus Chloroform, Methanol, Essigsäure und Wasser).
Gemäß Beispiel 1, jedoch unter Einsatz von N,S-Diacetyl-DL-
cystein wird die Titelverbindung erhalten. Rf = 0,25 (Kieselgel,
Ethylacetat).
1,9 g der gemäß Beispiel 3 erhaltenen Verbindung werden in
einem Gemisch von 6 ml Anisol und 12 ml Trifluoressigsäure
gelöst. Nach 1stündigem Stehenlassen bei Raumtemperatur werden
aus dem Gemisch die Lösungsmittel unter vermindertem Druck
abdestilliert. Der Rückstand wird aus einem Gemisch aus Ethyl
acetat, Diethylether und Hexan ausgefällt. Ausbeute 1,08 g der
Titelverbindung vom F. 80 bis 140°C.
0,3 g der gemäß Beispiel 4 erhaltenen Verbindungen werden in einer
Argonatmosphäre in einem Gemisch aus 4 ml Wasser und 4 ml
konzentrierter Ammoniaklösung gelöst. Nach 30minütigem Stehen
bei Raumtemperatur wird die Lösung mit Natriumchlorid gesättigt
und dann mit Ethylacetat und Chloroform extrahiert. Die organischen
Schichten werden vereinigt und unter vermindertem Druck
eingedampft. Ausbeute 0,1 g der Titelverbindung. Rf = 0,25
(Kieselgel; Gemisch aus Benzol und Essigsäure 75 : 25).
Gemäß Beispiel 1, jedoch unter Einsatz von N-tert.-Butyloxy
carbonyl-S-p-methoxybenzyl-L-cystein und nachfolgender Be
handlung des erhaltenen Produkts gemäß Beispiel 2, werden N-
tert.-Butyloxycarbonyl-S-p-methoxybenzyl-L-cysteinyl-prolin-
tert.-butylester und die Titelverbindung erhalten.
Claims (3)
1.Prolinderivate der allgemeinen Formel I
in der R eine Hydroxylgruppe oder einen Niederalkoxyrest,
R₁ ein Wasserstoffatom oder einen Niederalkonylrest, R₂
ein Wasserstoffatom oder einen Niederalkylrest, R₃ ein
Wasserstoffatom oder einen Niederalkonylrest bedeutet,
und deren Salze mit Säuren oder Basen.
2. Verfahren zur Herstellung der Verbindungen nach Anspruch
1, dadurch gekennzeichnet, daß man in an sich bekannter
Weise ein Prolinderivat der allgemeinen Formel III
in der R die im Anspruch 1 angegebene Bedeutung hat, mit
einer Säure der allgemeinen Formel IV
acyliert, in der R₁ eine Acylgruppe darstellt und R₂ und
R₃ die in Anspruch 1 angegebene Bedeutung haben, wobei
die Säure der allgemeinen Formel IV vor ihrer Umsetzung
mit dem Prolinderivat der allgemeinen Formel III in
ein aktiviertes Carbonsäurederivat überführt worden ist,
sowie gegebenenfalls die so erhaltene Verbindung mit einer
Säure oder einer Base in ein Salz überführt.
3. Verwendung der Verbindungen der allgemeinen Formel I bei
der Bekämpfung von durch Angiotensin bedingtem Hochdruck.
