Description
N-CH2-X-R3
in der X Sauerstoff oder Schwefel, Ri, R2 und R3
gleiche oder verschiedene substituierte oder unsubstituierte Aryl- oder Aralkylreste, substituierte oder
unsubstituierte gesättigte oder ungesättigte, geradkettige, verzweigte oder cyclische aliphatische Reste
mit bis 12 Kohlenstoffatomen darstellen, deren Kohlenstoffkette auch durch Sauerstoff-, Schwefel-
und Stickstoffatome unterbrochen sein kann, wobei Ri und R2 zusammen mit dem Stickstoffatom auch
cyclische oder polycyclische Gruppen mit bis 8 Kohlenstoffatomen bilden können, die Sauerstoff,
Schwefel und Stickstoff als weitere Heteroatome enthalten und Substituenten der für Ri, R2 und R3
angegebenen Art aufweisen können und/oder deren Salze mit anorganischen und/oder organischen
Säuren.
2. Mittel nach Anspruch 1, dadurch gekennzeichnet, daß sie außerdem oberflächenaktive Verbindungen,
saure, neutrale oder alkalische Salze oder Puffersubstanzen enthalten.
Die Konservierung von Kosmetika, z. B. von Cremes,
Lotionen und Shampoos, sowie von technischen Lösungen, Emulsionen, Suspensionen und Dispersionen,
wie zum Beispiel Kühlschmierstoffen mit Mischungen auf Formaldehydbasis, ist bekannt, führt aber aufgrund
des stechenden Geruchs, der Schleimhautreizungen und der relativ hohen Toxizität zu Problemen. Auch andere
bekannte Substanzen, zum Beispiel Addukte aus Formalin und primären Alkanolamine^ wie Hexahydrotriazine,
lösen die genannten Probleme nicht, da ihre antimikrobiellen bzw. konservierenden Eigenschaften
auf der Abspaltung des angelagerten Formaldehyds beruhen (vgl. DE-PS 11 48 706). Die Wirkungsweise der
in der US-PS 21 59/43 beschriebenen Kondensationsprodukte aus Formaldehyd und Aminen beruht
ebenfalls auf der Abspaltung von Formaldehyd.
Der Erfindung lag deshalb die Aufgabe zugrunde, Konservierungs- und Desinfektionsmittel mit einem
Gehalt an antimikrobiellen bzw. konservierend wirksamen Substanzen bereitzustellen, die die Wirksamkeit
des Formaldehyds gegenüber Protisten aufweisen oder übertreffen, ohne dessen Nachteile aufzuweisen.
Die erfindungsgemäß als Wirkstoffe eingesetzten Kondensationsprodukte aus sekundären Aminen, Formaldehyd
und Alkoholen erfüllen diese Forderung. Sie besitzen ausgezeichnete antimikrobielle Wirksamkeit,
ohne Geruchsbelästigungen und Schleimhautreizungen zu verursachen. Ihr antimikrobielles Spektrum ist sehr
breit, und die statische Wirkung selbst bei Konzentrationen von c<0,01% noch nachweisbar gut In Konzentrationen
von <0,l% besitzen sie bakterizide, fungizide und algizide Wirkung. Sie eignen sich daher besonders
zur Wachstumshemmung von Keimen in kosmetischen Zubereitungen und auch in technischen Lösungen, wie
Bohrölemulsionen.
Bei ihrer Anwendung können sie auch mit bekannten antimikrobiell wirksamen Substanzen kombiniert werden.
Hierfür eignen sich Aminale, Oxazine, Oxazolidine, Schwermetallverbindungen, Peroxide, N-Oxide, Halogene
und Halogenverbindungen, Epoxide, Alkohole, Aldehyde, Acetale, Carbonsäuren, Phenole und heterocyclische
Verbindungen, wie 2-Methyl-5-chlor-thiazor> lon-3 und 2-Mercaptopyridin. Zur Erleichterung der
Anwendbarkeit und zur Verstärkung der desinfizierenden und konservierenden Wirkung können auch
oberflächenaktive Verbindungen sowie Alkalien, saure, neutrale oder alkalische Salze oder Puffersubstanzen

Die in den erfindungsgemäßen Desinfektions- und
Konservierungsmitteln verwendeten Aminomethyläther und Aminomethylthioäther werden nach den
Angaben von G. M. Robinson und R. Robinson in Soc.

Bevorzugte Aminomethyläther und Aminomethylthioäther sind solche der oben angegebenen Formel I, in
der Ri, R2 und R3 gleiche geradkettige oder verzweigte
Alkylreste mit 1 bis 12, vorzugsweise 1 bis 8 und so insbesondere 1 bis 6 Kohlenstoffatomen sind.

Zum Nachweis der antimikrobiellen Wirksamkeit der erfindungsgemäß eingesetzten Aminomethyläther und
Aminomethylthioäther wurden diese gemäß den Riehlinien
der Deutschen Gesellschaft für Hygiene und Mikrobiologie (DGHM) auf ihre minimale Hemmkonzentration
(MHK) und ihre Wirksamkeit im Suspensions- und Flächenversuch untersucht. Ferner wurden
ihre konservierenden Eigenschaften in praxisnahen Kosmetik- und Bohrölemulsionen festgestellt
1. Minimale Hemmkonzentration (MHK)
Die minimale Hemmkonzentration wurde durch Reihenverdünnung wäßriger Lösungen gegenüber Staphylococcus
aureus, Escherichia coli, Pseudomonas aeruginosa, Bacillus subtilis, Aspergillus niger, Penicillium
glaucum und Candida albicans festgestellt Die erhaltenen Ergebnisse sind in der Tabelle 1 zusammengestellt:

2. Suspensionsversuch
Die Suspensionsversuche wurden gemäß den Richtlinien der DGHM mit folgenden Keimarten durchgeführt;
Staphylococcus aureus, Klebsieila pneumoniae, Pseudomonas aeruginosa, Proteus vulgaris, Trichophyton
mentagrophytes, Aspergillus niger und Candida
albicans. Wegen ihrer unterschiedlichen Wasserlöslic
keit wurden die Aminomethyläther in einer Standar formulierung, die stets die gleiche Menge ein
handelsüblichen Emulgators enthielt, untersucht. D erhaltenen Ergebnisse sind in der Tabelle 2 zusamme
gestellt. Die Abtötungszeiten sind in Minuten angeg ben.

4. Konservierungsversuche
4.1. Kosmetik
4.1. Kosmetik
Die Konservierungseigenschaften der erfindungsgemäß eingesetzten Wirkstoffe wurden in einer als
cremige Lotion (Öl/Wasser) vorliegenden kosmetischen Zubereitung untersucht. Diese Formulierung wurde mit
Mikroorganismen beimpft, und zwar je Prüfung
a) mit vegetativen Keimen der Arten
Staphylococcus aureus, Pseudomonas aeruginosa und Escherichia coli,
Staphylococcus aureus, Pseudomonas aeruginosa und Escherichia coli,
b) mit Hefe- und Schimmelpilzen der Arten
Candida albicans und Aspergillus niger und
Candida albicans und Aspergillus niger und
c) mit einer Mischung aus a) und b),
um eine antagonistische oder synergistische Beeinflussung der Keimarten zu prüfen.
Die von festen Nähragarplatten gewonnenen Kulturen wurden in steriler Pufferlösung suspendiert und auf
eine Keimzahl von lOVml und eine Sporendichte von 10VmI verdünnt Die Impfstoffmenge betrug jeweils
0,2 ml/50 ml Prüfgut. Die Keimbelastung wurde durch periodisches Beimpfen der Versuchsansätze erreicht.
Vor jeder Neubeimpfung wurden Ausstriche der einzelnen Probenansätze vorgenommen, aus denen sich
der Grad der konservierenden Wirkung ermitteln läßt. Keimfreiheit wurde mit »negativ« bewertet. Die
Konservierungseigenschaften sind um so ausgeprägter, je mehr Beimpfungs-/Ausstrichcyclen mit negativ
bewertet werden können. In der Tabelle 4 sind die Konservierungsergebnisse einer repräsentativen Auswahl
von erfindungsgemäß eingesetzten Aminomethyläthern aufgeführt.

4.2. Kühlschmierstoffe
Zur Prüfung der konservierenden Eigenschaften in einem Kühlschmierstoff wurden jeweils 50 ml handelsüblicher
synthetischer Schmierstoffe mit einer geeigneten Menge des zu prüfenden Wirkstoffes versetzt
2 χ wöchentlich wurden die Proben mit Mischungen verschiedener Mikroorganismen beimpft Die Mischkultüren
der Teststämme wurden aus einer belüfteten, erwärmten, flüssigen Dauerkultur entnommen, der
periodisch die Einzelstämme in Reinkultur zugesetzt wurden. Die Impfdichte betrug 0,5 ml/50 ml, was,
bezogen auf die Ausgangskultur, 1O5 bis 10* koloniebildende
Keime je ml bedeutet Folgende Mikroorganismen gelangten zur Anwendung:
a) vegetative Keime
Escherichia coli, Pseudomonas aeruginosa, Proteus
vulgaris, Aerobacter aerogenes
b) Hefe- und Schimmelpilze
Candida spec, Rhodotomla speo, Aspergillus spec,
Fusarium oxysporum, Cephalosporum spec.
Parallel zur Beimpfung wurden auf Nähragarplatten Ausstriche der einzelnen Probenansätze vorgenommen.
Die Keimdichte der Ausstriche wurde mit »negativ« oder »positiv« bewertet Die Versuchsansätze wurden,
um praktischen Bedingungen möglichst nahe zu
kommen, auf einer Schüttelmaschine täglich 8 Stunden bewegt, und als physikalische Belastung wurden je 1 g
Gußspäne zugesetzt. Die Anzahl der negativen Beimpf- und Ausstrichcyclen (mindestens 10) und die physikalische
Stabilität sind ein Maß für die konservierenden Eigenschaften der eingesetzten Wirkstoffe. Eine Auswahl
der erhaltenen Ergebnisse ist in Tabelle 5 zusammengefaßt.

aus sekundären Aminen, Formaldehyd und Alkoholen als Wirkstoffen sind die folgenden:
