Description
O-Phenyldithlophosphonsäureester, Verfahren zu
ihrer Herstellung und ihre Verwendung als Insektizide. Akarlzide und Nematozide
Die vorliegende Erfindung betrifft neue 0-Phenyldithiophosphonsäureester,
mehrere Verfahren zu ihrer Herstellung und ihre Verwendung als Insektizide, Akarizide und Nematizide.
Es ist bereits bekannt, daß O-Halogenphenyldithiophosphonsäureester,
z.B. 0-(2,4-Dichlor-phenyl)-S-n-propyl-thionothiolmethan-
bzw. -äthanphosphonsäureester und 0-(3-Chlor-phenyl)-S-cyanomethylthionothiol-äthanphosphonsäureester,
insektizide und akarizide Eigenschaften haben (vergleiche Japanische Patentschrift 48-98 o37 und Deutsche Offenlegungsschrift
1 954 894).
Es wurden nun die neuen O-Phenyldithiophosphonsäureester
der Formel

R für Alkyl, Halogenalkyl, Alkenyl oder Alkinyl, R1 für Alkyl,
R2, R3
und R gleich oder verschieden sind und für Wasserstoff,
Halogen, Cyan, Alkyl, Alkylthio, Alkylsulfonyl, Alkoxy,
Nitro, Carbalkoxy oder Phenyl
stehen.
Diese neuen Verbindungen zeichnen sich durch starke insektizide, akarizide und nematozide Eigenschaften aus.
Weiterhin wurde gefunden, daß die 0-Phenyldithiophosphonsäureester
(I) erhalten werden, wenn man
a) Dithioalkanphosphonsäuremonoesterhalogenide der Formel

Überraschenderweise besitzen die erfindungsgemäßen O-Phenyldithiophosphonsäureester
eine bessere insektizide, akarizide und nematizide Wirkung als die aus der Literatur vorbekannten
O-Halogenphenyl-dithiophosphonsäureester analoger Konstitution
und gleicher Wirkungsrichtung. Die Stoffe gemäß vorliegender Erfindung stellen somit eine echte Bereüierung der Technik dar.
Verwendet man beispielsweise nach Verfahrensvariante a) S-Methylthionothiol-äthanphosphonsäuremonoesterchlorid
und 4-Äthylsulfonyl-phenol
bzw. nach Verfahrensvariante b) das Kaliumsalz des 0-(2,4-Oibromphenyl)-thionothioläthanphosphonsaureesters
und iso-Propylbromid und nach Verfahrensvariante c) 0-(3-Cyano-

Die weiterhin als Ausgangsstoffe zu verwendenden O-Aryldithioalkanphosphonsäuremonoester

Die Verfahrensvarianten a), b) und c) der erfindungsgemäßen
Verbindungen werden bevorzugt unter Mitverwendung geeigneter Lösungs- oder Verdünnungsmittel durchgeführt. Als solche
Kommen praktisch alle inerten organischen Solventien infrage.
Hierzu gehören insbesondere aliphatische und aromatische, gegebenenfalls chlorierte Kohlenwasserstoffe, wie Benzol, Toluol,
Xylol, Benzin, Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Chlorbenzol, oder Äther, z.B. Diäthyl- und Dibutyläther,
Dioxan, ferner Ketone, beispielsweise Aceton, Methyläthyl-, Methylisopropyl- und Methylisobutylketon, außerdem Nitrile,
wie Aceto- und Propionitril.
Ais Säureakzeptoren können alle üblichen Säurebindemittel
Verwendung finden. Besonders bewährt haben sich Alkalicarbonate und -alkoholate, wie Natrium- und Kaliumcarbonat, Natrium-
und Kaliummethylat bzw. -äthylat, ferner aliphatische, aromatische
oder heterocyclische Amine, beispielsweise Triethylamin, Trimethylamin, Dimethylanilin, Dimethylbenzylamin und Pyridin.
Die Reaktionstemperatur kann innerhalb eines größeren Bereichs variiert werden. Im allgemeinen arbeitet man zwischen 0 und
12o°C, vorzugsweise bei den Verfahrensvarianten a) und b) bei 35 bis 7o°C und bei der Verfahrensvariante c) bei 0 bis 3o°C.
Die Umsetzung läßt man im allgemeinen bei Normaldruck ablaufen.
Zur Durchführung der Verfahrensvariante a) setzt man die Ausgangskomponenten
vorzugsweise im äquimolaren Verhältnis ein. Ein Überschuß der einen oder anderen Komponente bringt keine
wesentlichen Vorteile. Die Reaktionskomponenten werden meist in einem Lösungsmittel in Gegenwart eines Säureakzeptors bei
erhöhter Temperatur zusammengegeben und nach beendeter Reaktion

durch Eingießen in Wasser und Ausschütteln mit einem organischen Lösungsmittel, z.B. Toluol, wie üblich durch Abtrennen
der organischen Phase, Waschen, Trocknen und Abdestillieren des Lösungsmittels aufgearbeitet.
Bei der Verfahrensvariante b) setzt man die Phosphonsäurekomponente
vorzugsweise in ihrer Salzform in einem bis zu j oftigen molaren Überschuß gegebenüber dem Halogenid ein. Auch
hier erfolgt die Umsetzung in einem organischen Lösungsmittel und nach beendeter Reaktion wird die Lösung durch Ausschütteln
mit einem organischen Lösungsmittel, wie z.B. Toluol, wie üblich aufgearbeitet.
Bei der Durchführung der Verfahrensvariante c) wird vorzugsweise das Mercaptan im geringen Überschuß eingesetzt. Die
Umsetzung erfolgt vorzugsweise in einem Lösungsmittel in Gegenwart eines Säureakzeptors bei den angegebenen Temperaturen.
Die Aufarbeitung erfolgt in der bei den anderen Methoden beschriebenen Weise.
Die neuen Verbindungen fallen in Form von ölen an, die sich
zum Teil nicht unzersetzt destillieren lassen, jedoch durch sogenanntes "Andestillieren", d.h. durch längeres Erhitzen
unter vermindertem Druck auf mäßig erhöhte Temperaturen von den letzten flüchtigen Anteilen befreit und auf diese Weise
gereinigt werden. Zu ihrer Charakterisierung dient der Brechungsindex.
Wie bereits mehrfach erwähnt, zeichnen sich die erfindungsgemäßen O-Phenyl-dithiophosphonsäureester durch eine hervorragende
insektizide, akarizide und nematozide Wirksamkeit aus. Sie wirken nicht nur gegen Pflanzen-, Hygiene- und Vorratsschädlinge,
sondern auch auf dem veterinär-medizinischen Sektor gegen tierische Parasiten (Ektoparasiten), wie parasi-

tierende Fliegenlarven und Milben. Sie besitzen bei geringer Phytotoxizität sowohl eine gute wirkung gegen saugende als
auch fressende Insekten und Milben.
Aus diesem Grunde können die erfindungsgemäßen Verbindungen mit Erfolg im Pflanzenschutz sowie auf dem Hygiene-, Vorratsschutz- und Veterinärsektor als Schädlingsbekämpfungsmittel
eingesetzt werden.
Die Wirkstoffe eignen sich bei guter Pflanzenverträglichkeit und günstiger Warmblütertoxizität zur Bekämpfung von tierischen
Schädlingen, Insbesondere Insekten, Spinnentieren und Nematoden,
die in der Landwirtschaft, in Forsten, im Vorrats- und Materialschutz sowie auf dem Hygienesektor vorkommen. Sie sind gegen
normal sensible und resistente Arten sowie gegen alle oder einzelne Entwicklungsstadien wirksam. Zu den oben erwähnten
Schädlingen gehören:

Saatgut, ferner in Formulierungen mit Brennsätzen, wie Ra'ucherpatronen,
-dosen, -spiralen u.a. sowie ULV-KaIt- und Wannnebel-Formulierungen

Diese Formulierungen werden in bekannter Weise hergestellt, z.3. durch Vermischen der Wirkstoffe mit Streckmitteln, also
flüssigen Lösungsmitteln, unter Druck stehenden verflüssigten Gasen und/oder festen Trägerstoffen, gegebenenfalls unter Verwendung

Es können Farbstoffe wie anorganische Pigmente, z.B. Eisenoxid, Titanoxid, Ferrocyanblau und organische Farbstoffe,
wie Alizarin-, Azo-Metallphthalocyaninfarbstoffe und
Spurennährstoffe wie Salze von Eisen, Mangan, Bor, Kupfer, Kobalt, Molybdän und Zink verwendet werden.
Die Formulierungen enthalten im allgemeinen zwischen 0,1
und 95 Gewichtsprozent Wirkstoff, vorzugsweise zwischen 0,5 und 90 %.

Die Anwendung der erfindungsgemäßen Wirkstoffe erfolgt in Form ihrer handelsüblichen Formulierungen und/oder den aus
diesen Formulierungen bereiteten Anwendungsformen.
Der Wirkstoffgehalt der aus den handelsüblichen Formulierungen
bereiteten Anwendungsformen kann in weiten Bereichen variieren. Die Wirkstoffkonzentration der Anwendungsformen kann von
0,0000001 bis zu 100 Gew.-# Wirkstoff, vorzugsweise zwischen
0,01 und 10 Gew.-96 liegen.
Die Anwendung geschieht in einer den Anwendungsformen angepaßten
üblichen Weise.
Bei der Anwendung gegen Hygiene- und Vorratsschädlinge zeichnen sich die Wirkstoffe durch eine hervorragende Residual
wirkung auf Holz und Ton sowie durch eine gute Alkali stabilität auf gekalkten Unterlagen aus.
Die Anwendung der erfindungsgemäßen Wirkstoffe geschieht
im Veterinärsektor in bekannter Weise, wie durch orale Anwendung in Form von beispielsweise Tabletten, Kapseln,
Tränken, Granulaten, durch dermale Anwendung in Form beispielsweise des Tauchens (Dippen), Sprühens (Sprayen),
Aufgießens (pour-on and spot-on) und des Einpuderns sowie durch parenterale Anwendung in Form beispielsweise der
Injektion.

Grenzkonzentrations-Test
Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung
vermischt man 1 Gewichtsteil Wirkstoff mit der angegebenen Menge Lösungsmittel, gibt die angegebene Menge Emulgator zu
und verdünnt das Konzentrat mit Wasser auf die gewünschte Konzentration.
Die Wirkstoffzubereitung wird innig mit Boden vermischt, der
mit den Testnematoden stark verseucht ist. Dabei spielt die Konzentration des Wirkstoffs in der Zubereitung praktisch
keine Rolle, entscheidend ist allein die Wirkstoffmenge pro Volumeneinheit Boden, welche in ppm angegeben wird. Man
füllt den behandelten Boden in Töpfe, sät Salat ein und hält die Töpfe bei einer Gewächshaus-Temperatur von 270C.
Nach vier Wochen werden die Salatwurzeln auf Nematodenbefall (Wurzelgallen) untersucht und der Wirkungegrad des Wirkstoffs in % bestimmt. Der Wirkungsgrad ist 100 %, wenn der
Befall vollständig vermieden wird, er ist 0 %, wenn der Befall genau so hoch ist wie bei den Kontrollpflanzen in unbehandeltem, aber in gleicher Weise verseuchtem Boden.

Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung vermischt
man 1 Gewichtsteil Wirkstoff mit der angegebenen Menge Lösungsmittel, gibt die angegebene Menge Emulgator zu und verdünnt
das Konzentrat mit Wasser auf die gewünschte Konzentration.
Die Wirkstoffzubereitung wird innig mit dem Boden vermischt.
Dabei spielt die Konzentration des Wirkstoffs in der Zubereitung praktisch keine Rolle, entscheidend ist allein die Wirkstoff
gewichtsmenge pro Volumeneinheit Boden, welche in ppm (s mg/1) angegeben wird. Man füllt den Boden in Töpfe und
läßt diese bei Raumtemperatur stehen.
Nach 24 Stunden werden die Testtiere in den behandelten Boden gegeben und nach weiteren 2 bis 7 Tagen wird der Wirkungegrad des Wirkstoffs durch Auszählen der toten und lebenden
Testinsekten In % bestimmt. Der Wirkungsgrad ist 100 X, wenn
alle Testinsekten abgetütet worden sind, er ist 0 #, wenn
noch genau so viele Testinsekten leben wie bei der unbehandelten Kontrolle.

Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung vermischt
man 1 Gewichtsteil Wirkstoff mit der angegebenen Menge Lösungsmittel, gibt die angegebene Menge Emulgator zu und verdünnt
das Konzentrat mit Wasser auf die gewünschte Konzentration.
Die Wirkstoffzubereitung wird innig mit dem Boden vermischt.
Dabei spielt die Konzentration des Wirkstoffs in der Zubereitung
praktisch keine Rolle, entscheidend ist allein die Wirkstoffgewichtsmenge pro Volumeneinheit Baden, welche in ppm
(= mg/1) angegeben wird. Man füllt den Boden in Töpfe und läßt diese bei Raumtemperatur stehen.
Nach 24 Stunden werden die Testtiere in den behandelten Boden gegeben und nach weiteren 2 bis 7 Tagen wird der Wirkungsgrad
des Wirkstoffs durch Auszählen der toten und lebenden Testinsekten in % bestimmt. Der Wirkungsgrad ist 100 %, wenn
alle Testinsekten abgetötet worden sind, er ist 0 %, wenn noch genau so viele Testinsekten leben wie bei der unbehandelten
Kontrolle.

Mit der Wirkstoffzubereitung werden Bohnenpflanzen (Phaseolus vulgaris), die stark von allen Entwicklungsstadien der gemeinen
Spinnmilbe oder Bohnenspinnmilbe (Tetranychus urticae) befallen sind, tropfnaß besprüht.
Nach den angegebenen Zeiten wird die Abtötung in % bestimmt.
Dabei bedeutet 100 %, daß alle Spinnmilben abgetötet wurden;
0 % bedeutet, daß keine Spinnmilben abgetötet wurden.

Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung
vermischt man 20 Gewichtsteile der betreffenden aktiven Substanz mit der angegebenen Menge des Emulgators and
verdünnt das so erhaltene Gemisch mit Wasser auf die gewünschte Konzentration.
Etwa 20 Fliegenlarven (Lucilia cuprina) werden in ein mit Wattestopfen entsprechender Größe beschicktes Teströhrchen
gebracht, welches ca. 3 ml einer 20%igen Eigelbpulver-Suspension in Wasser enthält. Auf diese Eigelbpulver-Suspension
werden 0,5 ml der Wirkstoffzubereitung gebracht. Nach 24 Std.
wird der Abtötungsgrad in % bestimmt. Dabei bedeuten 100 % daß alle und 0%, daß keine Larven abgetötet worden sind.

Zur Herstellung einer zweckmäßigen Wirkstoffzubereitung vermischt
man die betreffende aktive Substanz mit dem angegebenen Lösungsmittel im Verhältnis 1:2 und verdünnt das so erhaltene
Konzentrat mit Wasser auf die gewünschte Konzentration.
Etwa 10-25 Räudemilben (Psoroptes cuniculi) werden in 1 ml der zu testenden Wirkstoffzubereitung gebracht, die in Tablettennester
einer TiefZiehverpackung pipettiert wurden. Nach 24 Stunden wird der Abtötungsgrad in Prozent bestimmt. Dabei
bedeuten 100 %, daß alle und 0 %, daß keine Milben abgetötet worden sind.

Kaliumcarbonat und 25o ml Acetonitril wird 2 Stunden bei 5o C gerührt und anschließend mit 2o,25 g (o,l Mol) S-sek.-ButylmethanthionothiolphesphonsHuresterchlorid
tropfenweise versetzt. Nach zv/exstündigem Rühren bei 5o°C wird die Reaktionslösung
mix 2oo ml Wasser und 4oo ml Toluol geschüttelt und im Scheidetrichter
getrennt. Die organische Phase wird über Magnesiumsulfat getrocknet und nach der Filtration das Toluol am Rotationsverdampfer
im VaKuum abgezogen. Es verbleiben 25 g (96 % der Theorie) S-sek.-Butyi-O-phenyl-thionothiolphosphonsäureester
22 in Form eines farblosen Öls mit dem Brechungsindex n~ : l,56öi.
Beispiel 2; (Methode B)
Stufe
Zu 1,79 kg (l Mol) Äthanthionophosphonsäuredichlorid werden
bei 5-lo°C innerhalb von 2 bis 3 Stunden eine Lösung von 1,29 kg (Io Mol) 3-Chlorphenol und o,4 kg (Io Mol) Natriumhydroxid
in 1,7 1 Wasser getropft. Nach beendeter Zugabe wird die Kühlung entfernt und Io bis 12 Stunden bei Raumtemperatur
nachgerührt. Dann wird das Reaktionsgemisch mit Io 1 V/asser

verdünnt und mit Io 1 Toluol extrahiert. Die Toluollösung wird
über Natriumsulfat getrocknet und dann eingeengt. Der Rückstand wird bei loo°C/2 Torr andestilliert. Man erhält 2,3 kg (82 %
der Theorie) 0-(3-Chlor-phenyl)-thionoäthanphosphonsäureesterchiorid
in Form eines hellgelben Öles.

In eine Suspension von 2,67 kg (19,3 Mol) Pottasche in lh 1
Acetonitril werden bei -15°C 31β g (9,35 Mol) Schwefelwasserstoff eingeleitet. Dazu tropft man innerhalb von 1 bis 2 Stunden
2,3 kg (9 Mol) 0-(3-Chlor-phenyl)-thionoäthanphosphonsäureesterchlorid,
läßt die Temperatur auf 2o°C steigen und rührt 12 Stunden bei 2o°C. Dann wird noch einmal Schwefelwasserstoff
eingeleitet, bis die Reaktion vollständig ist. Anschließend wird der überschüssige Schwefelwasserstoff im Vakuum entfernt,
dj.e Reaktionsmischung wird filtriert und das Filtrat
im Vakuum eingeengt. Man erhält 2,6 kg (99 % der Theorie) des KaliumsaLzes vom 0-(3-Chlor-phenyl)-thionothioläthanphosphonsäureester
in Form beige gefärbter Kristalle.
3. Siufe
Zu einer Suspension von 1,6 kg (5,4 Mol) des Kaliumsalzes von
O-p-ChlorphenylJ-thionothioläthanphosphonsäureester in 5 1
Acetonitril werden bei 45°C 0,615 kg (5 Mol) n-Propylhromid getropft, wobei
die Temperatur auf 55°C ansteigt. Es wird 24 Stunden bei 2o-25°C nachgerührt, das Reaktionsgemisch in 6 1 Toluol gegossen, die
Toluollösung zweimal mit je 6 1 Wasser gewaschen, über Natriumsulfat
getrocknet und dann eingeengt. Der Rückstand wird bei 8o°C

und 2 Torr andestilliert. Man erhält 1,31 kg (82 % der Theorie)
S-n-Propyl-O-(3-chlor-phenyl)-thionothioläthanphosphonsäure-
25 ester in Form eines gelben Öles mit dem Brechungsindex n^ :

Beispiel 3: (Methode C )
ι. stufe
Zu 74,5 g (o,5 Mol) Thionomethanphosphonsäuredichlorid wird
innerhalb von 2 Stunden bei ca. lo°C eine Lösung von 2o g (o,5 Mol) Natriumhydroxyd und 64,25 g (o,5 Mol) 3-Chlor-phenol
getropft. Dann läßt man 15 Stunden bei 2o-25°C nachreagieren und gießt anschließend das Reaktionsgemisch in o,5 1 Toluol.
Die Toxuoliösung wird zweimal mit Eiswasser gewaschen, über Natriumsulfat getrocknet und dann eingeengt. Nach dem Andestillieren
erhält man 75 g (63 % der Theorie) 0-(3-Chlor-phenyl)-thionomethanphosphonsäureesterchlorid
in Form eines gelben Öles mit dem Brechungsindex n~': I,58o8.
2. Stufe
Zu einer Lösung von 8,1 g (o,12 Mol) Natriumäthylat und 9 g
(o,115 Mol) n-Propylmercaptan in 5o ml Äthanol werden innerhalb von 3o Minuten bei o-5°C 24,1 g (o,l Mol) 0-(3-Chlorphenyl)-thionomethanphosphonsäureesterchlorid

Die als Ausgangsstoffe bei der Methode A zu verwendenden
S-Alkyl-thionothiolphosphonsäureesterchloride kennen nach
zwei Verfahren synthetisiert werden:
(Verfahren A) « ^SC, Hg-sek.

Eine Lösung von 292,4 g (2,5 Mol) Methyldichlorphosphin in
1600 ml trockenem Toluol wird bei -2o°C innerhalb von 3o Minuten unter Rühren tropfenweise mit einem Gemisch aus 225 g
(2,5 Mol) sek.-Butylmercaptan und 2oo g (2,5 Mol) Pyridin versetzt.
Man läßt anschließend noch 3o Minuten bei -2o°C nachrühren, läßt die Temperatur innerhalb von einer Stunde auf
Raumtemperatur ansteigen, trägt bei 3o°C 80 g (2,5 Mol) fein pulverisierte Schwefelblüte in die Reaktionslösung ein. Man
erwärmt die Reaktionsmischung anschließend eine Stunde auf 9o°C und kocht sie danach eine halbe Stunde unter Rückfluß.
Nach Abkühlung auf 25°C wird filtriert, das Filtrat am Rotationsverdampfer im Vakuum eingeengt und der Rückstand destilliert.
Es verbleiben 24o g (47 % der Theorie) S-sek.-Butyl-thionothiolmethanphosphonsäureesterchlorid
mit einem Siedepunkt von 950C/ 4 Torr.

Eine Lösung von 14,8 g (o,l Mol) Methanthionophosphonsäuredichlorid
in 15o ml Toluol wird bei lo°C unter Rühren tropfenweise mit einem Gemisch aus 9 g (o,l Mol) sek.-Butylmercaptan
und 18,o g (o,l Mol) Pyridin versetzt. Danach erwärmt man die Reaktionslösung noch Io Stunden auf 800C, kühlt anschließend
auf Raumtemperatur ab, filtriert, engt das Filtrat am Rotationsverdampfer im Vakuum ein und destilliert den Rückstand. Es
verbleiben 16 g (80 Ji der Theorie) S-sek.-Butyl-thionothiolmethanphosphonsäureesterchlorid
mit dem Siedepunkt von 95°C/ 4 Torr.

Erdalkalisalzes und gegebenenfalls in Gegenwart eines Lösungsmittels umsetzt. - 3. Insektizide, akarizide und nematozide Mittel, gekennzeichnet durch einen Gehalt an mindestens einer Verbindung gemäß Anspruch 1 .
- 4. Verfahren zur Bekämpfung von Insekten, Spinnentieren und Nematoden, dadurch gekennzeichnet, daß man Verbindungen gemäß Anspruch 1 auf die genannten Schädlinge oder ihren Lebensraum einwirken läßt.
- 5. Verwendung von Verbindungen gemäß Anspruch 1 zur Bekämpfung von Insekten, Spinnentieren und Nematoden.
- 6. Verfahren zur Herstellung von insektiziden, akariziden und nematiziden Mitteln, dadurch gekennzeichnet, daß man Verbindungen gemäß Anspruch 1 mit Streckmitteln und/oder oberflächenaktiven Mitteln vermischt.Le A 17 961 - 66 -809841/0193
