A'thylazobiscarboxylat in 10 ml Benzol zugetropft. Nachdem die Säure vollständig gelost war,wurde die Mischung mit 0,1 η
Schwefelsäure, Wasser, einer 5%igen Natriumbicarbonatlösung
und erneut mit Wasser gewaschen, dann zur Trockne eingedampft. Der Rückstand wurde einer Dünnschichtchromatographie unterworfen und mit Methylenchlorid/Äthyläther (90/10) eluiert, wobei man 0,59 g 5c-9ß ^S-Dihydroxy-iSR-methyl-prost-^
insäureäthylester-15-THP-äther-9-p-phenylbenzoat erhielt.
in wasserfreiem THF zugegeben. Es wurde auf O0C abgekühlt und eine 348 mg Äthylazobiscarboxylat enthaltende THP-Lösung wurde zugegeben· Diese wurde 15 Minuten lang gerührt, dann zur
Trockne eingedampft. Der Rückstand wurde zwischen Äthyläther/- Pentan (1/1) und Wasser verteilt und die organische Phase
wurde mit verdünnter Säure, Wasser, Natriumbicarbonat und er neut mit Wasser gewaschen. Sie wurde getrocknet und zur
Trockne eingedampft. Der Rückstand wurde an 20 g Silicagel
chromatographiert, mit Cyclohexan/Äther (75/25) eluiert, wo bei man 452 mg 5c-9o.,15S-Dihydroxy-16S ,20-dimethyl-8,12-di- ieo-pro st-5-en-13-insäure-15-THP-äther-9-propionatmethyle st er erhielt«
5c-9 d-,15S-Dihydroxy-8,12-diiso-prost-5-en-13-insäure,
Chloroform extrahiert. Die vereinigten organischen Phasen
wurden wiederholt mit einer gesättigten Natriumbicarbonatlösung und dann mit Wasser gewaschen, über lls^SO^ getrocknet
und zur Trockne eingedampft, wobei man 6,18 g 5 OL-Hydroxymethyl-2/3 -hydroxy-cyclopentan-i/S -essigsäure-^--lacton, F.
52 bis 53°C, LcL]D = +27,3°, Td]365O = +110° (CHCl3), erhielt.
1,15 Ko!-Äquivalenten p-Toiuolsulfonylchlorid umgesetzt unter Bildung von 2 ex , 4-d.-Edhydr 02^-5/3 -(hydroxymethyl-DIOX-äther)-cyclopentan-1 o- -essigsäure- y -lacton-^—p-toluolsulf onat. Eine Lösung dieses Produkts in wasserfreiem Äthyläther wurde auf 0 bis 20C abgekühlt und es wurden 10 Moläquivalente Mg^ ija
wasserfreiem Äthyläther zugegeben. Die Mischung wurde 1 Stunde lang gerührt und dann mit Wasser und Eis hydrolysiert
fluor-prost-5-en-13-insäure-15-THP-äther (1,4 g) in 28 ml Aceton, die auf -18°C abgekühlt war, wurde mit 2,8 ml Jones-Reagens (hergestellt durch Zugabe von 61 ml konzentrierter Schwefelsäure zu 70 g Chromsäureanhydrid in 500 ml Wasser) behandelt. Die Mischung wurde 20 Minuten lang bei -10 bis -120C gerührt, mit 80 ml Benzol verdünnt, mit einer gesättigten Ammoniumsulfatlösung bis zur Neutralität gewaschen, über Na-SO^ getrocknet und dann im Vakuum zur Trockne eingedampft, wobei man in Form eines Öls 5c-9-0xo-15S-hvdroxy-16S~methyl-16R-fluor-prost-5-en-13-insäure-15-THP-äther erhielt, der in 80 ml Aceton gelöst wurde. Diese Lösung wurde 4 Stunden lang mit 50 ml einer wässrigen 0,2 η Oxalsäure bei 45°C behandelt, dann wurde die wässrige Phase nach der Entfernung des Acetons im Vakuum wiederholt mit Äther extrahiert. Die vereinigten Extrakte wurden mit einer gesättigten Ammoniumsulfatlösung gewaschen, über NanSO^ getrocknet und zur Trockne im Vakuum eingedampft. Der Rückstand (1,1 g) wurde an mit Säure gewaschenem Silicagel adsorbiert und nach der Elution mit Cyclohexan/Methylenchlorid (70/30) erhielt man 0,72 g 5c-9-Oxo-15S-hydroxy-16S-methyl-16R-fluor-prost-5-en-13-insäure.

- Patentansprücheworin bedeuten:R ein Wasserstoff atom, eine C^-C^-Alkylgruppe oder ein Kation einer pharmazeutisch verträglichen Base ^einer der Reste Rx, und Rp Wasserstoff und der andere Hydroxy oder Acyloxy oder beide Reste gemeinsam eine Oxogruppe,einer der Reste R^ und R^ Wasserstoff und der andere Hydroxy,Br und Rx- einzeln jeweils Wasserstoff, Fluor oder Cx,-C^.-Alkyl, mit der Liaßgabe, daß dann, wenn einer von ihnen G^-G1,-Alkyl bedeutet, der andere Wasserstoff oder Fluor bedeutet, und daß dann, wenn einer von ihnen Fluor bedeutet, der andere G^-G1,-Alkyl bedeutet,E -(CH^) -, worin η eine ganze Zahl von 1 bis 6 bedeutet, oder -(CH,,) -0-(CH0) -, worin n^ und no unabhängig voneinander jeweils die" Zahl 0, 1, 2 oder 3 bedeuten,Methyl, Cycloalkyl mit 3 "bis 7 Ringkohlenstoff atomen, das gegebenenfalls ein oder mehrere Ringsauerstoff- oder -schwefelatome enthält, oder Phenyl, das unsubstitu-609840/1.U7iert ist oder gegebenenfalls substituiert ist durch einen oder mehrere Substituenten aus der Gruppe Halogen, Phenyl und Trihalogenmethyl,und worin die an die Kohlenstoffatome in den 8- und 12-Stellungen gebundenen Ketten eine trans-Konfiguration aufweisen.

Verbindungen nach Anspruch 1, gekennzeichnet durch die Formel5c-9-Oxo-15S-hydroxy-16S-methyl-prost-5-en-13-insäure, 5c-9-0x0-15S-hydroxy-16S-methyl-8,12-diiso-proRt-5-en-13-insäure, 5c-9-Oxo-15S-hydroxy-16R-methyl-prοst-p-en-13-insäure, 5c-9-0xo-i5S-hydroxy-:16R-methyl-8,12-diiso-prost-5-en-13-insäure, 5c-9-0xo-153-hydroxy-16S,20-diinethyl-pro5t-5-en-13-insäure, 5c-9-Oxo-15S-hydroxy-16R,20-dimethyl-prost-5-en-13-insäure t 5c-9-Oxo-15S-h7^droxy-16S-methyl-16R-fluor-prost-5-en-13-insäure, 5c-9-0xo-15S~hydroxy-16S-fluor-16R-methyl-prost-5-en-13-insäure sowie ihre 15R-Epimeren.
- 3. Verfahren zur Herstellung der optisch aktiven 13,14—Dehydro-11-desoxy-prostaglandine der in Anspruch 1 angegebenen allgemeinen Formel I, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen FormelCOOR(H)vrorin R, R^, Rp, Rr, Rg, Ξ und<£ die in Anspruch 1 angegebenen Bedeutungen haben und einer der Reste R'7 und R1^ eine bekannte Schutzgruppe, die über ein Äthersauerstoffatom an die Kette gebunden ist, und der andere Wasserstoff bedeuten und worin609840/1U7Vfdie an die Kohlenstoffatome in den 8- und 12-Stellungen gebundenen Ketten eine trans-Konfiguration aufweisen,entäthert und gewünschtenfalls nach an sich bekannten Verfahren eine Verbindung der in Anspruch 1 angegebenen allgemeinen Formel I in eine andere Verbindung der allgemeinen Formel I überführt.
- 4·. Pharmazeutisches Mittel, dadurch gekennzeichnet, daß es als Wirkstoff mindestens ein optisch aktives 13»14— Dehydro-11-desoxy-prostaglandin der in Anspruch 1 angegebenen allgemeinen Formel Iv gegebenenfalls in Kombination mit einem geeigneten Träger und/oder Verdünnungsmittel5enthält.
