Beispiele für pharmakologisch verträgliche Säuren sind anorganische
Säuren, wie Chlorwasserstoff säure, Schwefelsäure, Salpetersäure und Phosphorsäure, sowie organische Säuren,
wie Essigsäure, Fumarsäure, Maleinsäure, Weinsäure, Citronensäure , Bernsteinsäure und Methansulfonsäure» Die Verbindungen
der oben angegebenen Formel I und ihre pharmakologisch verträglichen Säureadditionssalze weisen verschiedene pharmakologische
Effekte, insbesondere einen ausgezeichneten hypotensiven Effekt auf, . '
Die erfindungsgemäßen Verbindungen der Formel I werden nach
dem folgenden Verfahren hergestellt:

worin A die oben angegebenen Bedeutungen besitzt und X
eine funktioneile Gruppe darstellt, die eine Stickstoff-Kohlenstoff
-Bindung bilden kann, d.h., die Verbindungen der Formel I werden hergestellt durch Umsetzung von 2-Homopiperazino-4-amino-6,7-aimethoxy--chinazölinen
der allgemeinen Formel II mit Reaktanzen der allgemeinen Formel III.
Die Reaktanten der Formel III-"werden, in die den Bedeutungen
von A und den damit ablaufenden Reaktionen entsprechenden
beiden ,Gruppen eingeteilt:
(i) wenn A den Rest R -CO-bedeütet:

Derivate der Säuren sind Säurehalogenide, Säureanhydride und reaktionsfähige Ester. Die Reaktionen können
in Abwesenheit eines Lösungsmittels oder in ,Gegenwart eines ■
organischen Reaktionslösungsmittels, das gegenüber der Reaktion inert ist, wie z.B. in Benzol, Toluol oder Xylol, durchgeführt
werden. Wenn die Reaktion bei Raumtemperatur durchgeführt wird, können Ketonlö sungsmitt el, wie Aceton oder
Methylethylketon, Verwendet werden, ohne daß Nebenraktionen

auftreten. Die Reaktion kann glatter, durchgeführt werden,
,wenn man ein Säureentfernungsmittel, wie Triäthylamin, ein
Alkalibicarbonat oder Pyridin in dem Reaktionssystem verwendet; ". "

Verbindungen der oben angegebenen allgemeinen Formel II werden
mit Halogeniden der allgemeinen Formel umgesetzt

worin X ein Halogenatom bedeutet. Die Reaktionen können in Abwesenheit eines Lösungsmittels oder in Gegenwart eines organischen
Lösungsmittels, das gegenüber der Reaktion inert ist, z.B. in Gegenwart von niederen Alkoholen, wie Methanol,
Äthanol, Propanol oder Isopropanol, einer Benzolverbindung,
wie Benzol selbst, Toluol oder Xylol, eines Äthers, wie Diäthyläther, Diisopropyläther oder Tetrahydrofuran, durchgeführt
werden. Die Reaktionstemperatur kann zweckmäßigerwe^Lse innerhalb des Bereiches von Raumtemperatur bis zum Siedepunkt des
verwendeten Lösungsmittels ausgewählt werden. Die Reaktion kann glatter durchgeführt werden, wenn man in dem Reaktionssystem ein Säureentfernungsmittel, wie Triäthylamin, ein. Alkalibicarbonat
oder Pyridin, verwendet.
Die als Ausgangsmaterialien in dem erfindungsgemäßen Verfahren verwendeten Verbindungen der Formel II stellen neue Verbindungen
dar, die nach dem folgenden Verfahren synthetisiert werden können:

Die Verbindung der Formel II kann hergestellt werden durch
■umsetzung von 2-Chlor-4—amino-e^-dimethoxy-chinazolin
(IV) mit IT-iOrmyl-homop.iperazin (V) und anschließende
Deformylierung des dabei erhaltenen 2-(li'-Jlormyl-homopiperazino)—il--amino-6i7-ä.imethoxy-chinazolins(VI)
auf an sich bekannte Weise. ■-.".. .
Die-biologischen Aktivitäten der erfindungsgemäßen Verbindungen,
insbesondere im Hinblick auf ihre Wirksamkeit zur Herabsetzung des Blutdruckes bei Hypertension,werden durch
die-nachfolgend beschriebenen pharmakologischen Versuche
erläutert. Einige der erfindungsgemäßen Verbindungen der Formel I weisen eine noch bessere biologische Aktivität
auf als Prazosin (US-Patentschrift 3 511 836), das hinsichtlich
seiner chemischen Formel und seiner biologischen Aktivität mit den Verbindungen der Formel I vergleichbar ist.
Nachfolgend sind die chemischen Formeln von zwei repräsentativen
Vertretern der erfindungsgemäßen Verbindungen und der
Standardverbindung Prazosin angegeben:

Die Erfindung wird nachfolgend unter Bezugnahme auf die beiliegenden Zeichnungen näher erläutert. Die Figuren 1 bis
3 der beiliegenden Zeichnungen sseigen Diagramme, auf denen die Ergebnisse von pharmakologischen Versuchen dargestellt
sind, die mit den beiden erfindungsgemäßen Verbindungen und Erazosin durchgeführt wurden.
Die hypotensive Wirkung wurde durch die folgenden Tierversuche
gezeigt:
Versuch 1
Die Verbindungen wurden in einer Kochsalzlösung gelöst und intraperitoneal an normotensive Ratten (mit einem normalen
Blutdruck) unter Pentobarbitalnatrium-Anasthesie verabreicht. Die Verbindungen A und B sowie Prazosin verminderten den
Blutdruck um 20 bis 40 mm Hg und verhinderten den blutdruckerhöhenden
Effekt von Adrenalin (3 jug/kg, intravenös)» Während
bezüglich des Grades der nypotensiven Wirkung zwischen den
drei Verbindungen keine feststellbare Differenz bestand,
war der antiadrenergische Effekt der Verbindung A stärker
ausgeprägt als derjenige der Verbindung B und von Prazosin.
Versuch 2
Durch chronische Behandlung mit Desoxycorticosteronacetat
und einer Kochsalzlösung wurde bei Hatten eine experimentelle Hypertension induziert. Bei einer oralen Dosis von 1 mg/kg
der jeweiligen Verbindungen wurde der Blutdruck um 40 bis

60 mm Hg gesenkt und der hypotensive Effekt dauerte mehr
als 6 Stunden an. ·

Versuch 3
Die Verbindungen senkten den Blutdruck eines durch Inzucht
erzeugten Stammes einer Ratte mit einer spontanen Hypertension (SHR), der von Okamoto und Äoki gezüchtet wurde. Die
Pig. 1 der heiliegenden Zeichnungen erläutert in vergleichender Weise den hypotensiven Effekt der jeweiligen Verbindungen
bei einer oralen Dosis von 1 mg/kg bei SHR. Die
Verbindung A wirkte bei dieser Modellhypertension am stärksten und der Effekt der Verbindungen A und B hielt mehr- als 12
Stunden an. ' ■
Durch einige experimentelle Daten in bezug auf ihren Mechanismus
der hypotensiven Wirkung und die allgemeinen pharmakologischen Eigenschaften wurde das folgende spezifische Merkmal
der Verbindungen A und B erläutert: ·
Versuch 4-
Die Kontraktion, eines isolierten Vas deferens durch Hör—
adrenalin wurde durch.Behandlung mit der Verbindung Ä und mit der· Verbindung B sowie mit Prazosin verhindert. Die'
Figuren 2-und 3 der beiliegenden Zeichnungen erläutern die Wechselwirkung zwischen Horadfenalin und Prazosin bzw.
zwischen Horadrenalin und der Verbindung A. Die Sigmoid-Kurve der Dosis (Konzentration an Nor adrenalin (in Mol/ml ))-Ansprechempfindlichkeit (Kontraktion des Vas deferens)
wurde unter dem Einfluß von Prazosin (1 χ 10 bis 1 χ ΛΟ Μοί/ml) parallel nach rechts verschoben, d.h. es wurde ein Konkurrenzantagönismus zwischen Noradrenalin und Prazosin festgestellt (Fig. 2). Durch die Verbindung A wurde die Dosis—
adrenalin wurde durch.Behandlung mit der Verbindung Ä und mit der· Verbindung B sowie mit Prazosin verhindert. Die'

Ansprechempfindlichkeitskurve von Noradrenalin in ausgeprägterem
Maße als durch Prazosin nach, rechts verschoben. Außer dieser Verschiebung nach rechts wurde die maximale
Kontraktion durch Noradrenalin unter dem Einfluß der Verbindung
A merklich vermindert. Dies bedeutet, daß die Verbindung A nicht nur eine Konkurrenzkömponente, sondern auch
eine Komponente für die Mcht-Konkurrenzinhibierung gegen den Antagonist en aufweist (Fig. j5). Die Wechselwirkung zwischen der Verbindung B und Boradrenalin war, wie gefunden
wurde, analog zu derjenigen mit der Verbindung A. In der folgenden Tabelle I ist die 5° %-Inhibierungskonzentration'
(IDj-q) der Verbindungen A, B und von Prazosin gegen Noradrenalin,
d.h. die errechnete Konzentration der geweiligen Verbindung angegeben, welche die Hälfte der Kontraktion
des Vas deferens unter dem Einfluß von Foradrenalin (1 χ 10"~
Mol/ml) verhindern kann. Die dabei erhaltenen relativen
Wirksamkeitswerte betrugen danach bei der Verbindung A:10,
bei der Verbindung B:4- und bei Prazosin:1 (Tabelle I).

Es wurde gefunden, daß die Verbindungen A und B einen starken
Inhibierungseffekt auf die Blutplättchenaggregation aufweisen.
Die Aggregation von Blutplattchen wurde durch Zugabe von Adenosindiphosphat (1 χ 10 Mol/ml) zu einem an Blutplattchen
reichen Plasma induziert, das durch 20-minütiges Zentrifugieren bei 150 g aus Kaninchenplasma hergestellt wurde. Die
Aggregation der Blutplattchen wurde in Gegenwart von 1,25 x 10"

g/ml der Verbindung A sowie der Verbindung B eindeutig
verhindert, während sie durch· Prazosin nicht beeinflußt wurde (Tabelle II).

Aus den vorstehend angegebenen Ergebnissen wurden die folgenden
Schlüsse gezogen: . . .
1.) -Die Verbindung A und die Verbindung B waren in bezug auf
ihre hypotensive Wirkung gleich wirksam oder etwas wirksamer
als Prazosin. Es ist insbesondere bemerkenswert, daß die hypotensiven
Wirkungen der Verbindungen A und B bei Ratten mit einer spontanen Hypertension ausgeprägter und langer anhaltend
waren als diejenigen von Prazosin, weil die Tiere mit einer angeborenen Hypertension manchmal als'stärker bevorzugtes
Modell für die genuine Hypertension bezeichnet werden.
2.) Die Ergebnisse des. Versuchs Nr. 4 in bezug auf. die Wechselwirkung
mit Noradrenalin zeigen, daß die' Verbindungen A und B sich hinsichtlich des Wirkungsmechanismus von Prazosin
unterscheiden können; die Verbindungen A und B weisen zusätzlich zu ihrer starken a-Blockierungswirkung einen direkten
relaxierenden Effekt auf die glatte Muskulatur auf.
3.) Zusätzlich zu der hypofcensiven Wirkung verhindern die
Verbindungen A und B in wirksamer Weise eine experimentell

hervorgerufene Blutplättchenaggregation. Die Aggregation von Blutplättchen ist einer der Faktoren für die Bildung
von Thromben, die manchmal die· Todesursache bei Störungen der Coronar- und Cerebralzirkulation in Verbindung mit einer
,hochfrequenten Hypertension sind. Deshalb wird der Inhibierungseffekt
auf die Aggregation von Blutplättchen als vorteilhaft
angesehen. Prazosin weist eine solche Wirkung nicht auf oder seine Wirkung ist vernachlässigbar gering.
Wie oben angegeben, sind die erfindungsgemäßen !/"erbindungen,
wirksam für die Verhinderung und Behandlung von verschiedenen Typen der Hypertension, beispielsweise der genuinen, der
renalen und bösartigen Hypertension. Die Verbindungen können sowohl oral als auch parenteral in geeigneten Dosierungsformen, beispielsweise in Form von Tabletten, Pillen, Körnchen,
Kapseln^ Pulvern, Lösungen und Suppositorien, verwendet werden. Bei einer mäßigen oder schweren Hypertension kann
die für einen Erwachsenen empfohlene Dosis innerhalb des Bereiches
yon 1 bis 200 mg pro Tag, vorzugsweise von 1 bis
mg pro Tagjbei oraler Verabreichung liegen.
Die Erfindung wird durch die folgenden Beispiele näher erläutert, ohne jedoch darauf beschränkt zu sein.
Synthese von 2-[N ~(2-Furoyl)homopiperazino3-4-amino-6,7-dimethoxy-chinazolin
(a) Synthese von 2-Homopiperazino-4~ainino-6,7-dimethoxy-
£hinazo_lin r
17 g 2-Ghlor-4-amino-6,7-dimethoxy-chinazolin und 18,2 g
N-Formylhomopiperazin wurden zu 170 ml Butanol zugegeben
und das Ganze wurde unter Rühren 3 Stunden lang unter Rückfluß gekocht. Nach. Beendigung der Reaktion wurde die Mischung
abgekühlt und die ausgefallenen Kristalle wurden

abfiltriert, mit einer geringen Menge Äthanol gewaschen
und an der Luft getrocknet. Es wurden 25 g rohe Kristalle
erhalten. 13 g der" so erhaltenen. Kristalle'wurden mit 80 ml
einer 9 %igen Chlorwasserstoff säure versetzt. Die Mischung wurde unter Rühren 60 Minuten lang unter Rückfluß gekocht·
Nach Beendigung der Reaktion wurde die Mischung abkühlen gelassen. Die auf diese Weise ausgefallenen Kristalle wurden
abfiltriert und dann aus einer Methanol/Äthanol-Mischung
umkristallisiert, Ausbeute 10,7 g (80,4 %)v F. 24-6 bis 247OC.

(b) Synthese von 2-[IP-(2-Furöyl)-homopiperazino3-4-amino-
Eine Lösung von 3 g 2-Homopiperazino-4-amino-6,7-dimethoxy—
chinazolin in 60'ml Aceton wurde zu einer Lösung von 1,3 g
2-Eurancarbonsäurechlorid in 30 ml Aceton unter Rühren und
unter Eiskühlung zugetropft. Nach Beendigung der Zugabe wurde
das Rühren eine weitere Stunde lang fortgesetzt, um die
Reaktion zu vervollständigen* Die dabei ausgefallenen Kristalle
wurden abfiltriert und aus einem Methanol/lthano 1-Gemisch
umkristallisiert, Ausbeute 3,1 g (70,4 %), F. 278 bis 280°C.

Beispiel 2 . .
Synthese von 2-(N -Butyl-homopiperaz'ino)-4>-amino-6,7-dimethoxy-chinazolin

2 g Triäthylamin wurden zu einer Lösung von 1,5 S 2-Homo—
piperazino^-amino-ö^-dimethoxy-chinazolin und 1,5 g
a-Butylchlorid in 20 ml n-Butatiol zugegeben und die Mischung
wurde unter Rühren 24 Stunden lang unter Rückfluß gekocht.

Nach Beendigung der Reaktion wurde das Lösungsmittel abdestilliert und dann wurden der so erhaltene Rückstand .
durch Zugabe einer 10 %igen wäßrigen Natriumhydroxyd«
lösung .alkalisch gemacht. Die sich dabei abscheidende ölige Substanz wurde mit Chloroform extrahiert. Der Extrakt
wurde mit Wasser gewaschen, übes? Kaliumcarbonat getrocknet und filtriert· Das Filträt wurde eingeengt. Der dabei erhaltene
Rückstand wurde in 39 nil Isopropanol gelöst· Die *
Lösung wurde mit 3 ml gesättigter Isopropanol/Chlorwasserstoff
säure versetzt und die dabei erhaltenen Kristalle wurden abfiltriert und aus einem Methanol/Äthanol-G-emiscbi
umkristallisiert. Die gewünschte Verbindung wurde in Form des Dihydrochloride erhalten, Ausbeute 0,9 g (50,6 %)·
