Verfahren zur Herstellung optisch aktiver Kohlenstoffverbindungen
Das Verfahren betrifft die Herstellung optisch aktiver Kohlenstoffverbindungen aus optisch inaktiven Kohlenwasserstoffen
unter Ausbildung der chiralen Zentren im Zuge von C-C-Verknüpfungen.
Aus der Literatur sind nur drei Beispiele nicht enzymatischem asymmetrischer Katalyse unter C-C-Verknüpfung bekannt. J. Furukawa et al (Bulletin ο:' the chemical Soc, of
Japan, VoI 4l, 155 (1968)) beschreibt die Synthese optisch
aktiver Trimethylcyclododecatriene aus Pentadien-(l,3) unter Verwendung eines Katalysators aus Titan-tetra-menthoxyl
und Diäthylaluminiummonochlorid bei +4o°C. Über die Ausbeute
an optisch aktiven Produkten und deren optische Reinheit werden nur unzureichende Angaben gemacht. Nach den
angegebenen Daten liegen die Ausbeuten nicht höer als Io %
bzw. 4 %. Die Autoren weisen selbst daraufhin, daß die optischen
Reinheiten nicht hoch sein können, da die Drehungen gering sind.
H. Nozaki et al (Tetrahedron VoI 24, 3655 (1968) erhalten
mit einer optischen Ausbeute von weniger als 8 % trans-1-Methyl-2-phenylcyclopropan
aus Diazomethan und l-Methyl-2-phenyl-äthylen
bei 600C mit Hilfe eines Katalysators, bestehend
aus einem optisch aktiven Kupfer-II-Komplex. Kupfer-I-Chlorid-Katalysatoren
mit komplexgebundenen Trialkyl- oder Triaryl-phosphitliganden wurden von W.R. Moser (J,
Ameri. Soc. 91:5, 1135 (19o9))zur Synthese optisch aktiver
Cyclopropanderivate aus Ä'thyldiazoacetat und Styrol eingesetzt.
Das ( (-)-Tribornylphosphlt)-Kupfer-I-Chlorid lieferte bei Jo - 600C das entsprechende Cyclopropanderivat
mit einer optischen Ausbeute von ca. 3 %>
Die nach den genannten Methoden zu erzielenden optischen Reinheiten sind unbefriedigend. Dies beruht darauf, daß
einerseits die induzierenden Gruppen am Katalysator während der Reaktion gegen inaktive Gruppen ausgetauscht werden
und daß andererseits die in den Beispielen erwähnten Reaktionen nur bei relativ hohen Temperaturen ablaufen.
Es wurde nun gefunden, daß man optisch aktive Kohlenstoffverbindungen
aus optisch inaktiven Kohlenwasserstoffen unter C-C-Verknüpfung mit bisher durch kein katalytisches
Verfahren erzielbaren optischen Reinheiten dadurch herstellen kann, daß man auf Olefingemisehe, bevorzugt bei
tiefen Temperaturen, Katalysatoren einwirken lässt, die durch Kombination von Nickelverbindungen mit Lewissäuren
und optisch aktiven Phosphinen PR, erhalten werden, wobei die Phosphine entweder das Phosphoratom als chirales Zentrum
oder aber optisch aktive Kohlenwasserstoffreste R enthalten. Die aus der Literatur bekannten und eingangs beschriebenen
Katalysatoren sind nicht in der Lage, zwei verschiedene ungesättigte Kohlenwasserstoffe unter Ausbildung
optisch aktiver Zentren zu verbinden.
Die für die Synthese geeigneten optisch inaktiven Ausgangsverbindungen
sind ungesättigte Kohlenwasserstoffe wie offenkettige bzw. cyclische Einfach- bzw. Mehrfacholefine,
bei deren C-C-Verknüpfung chirale Zentren entstehen können.
Für die Herstellung der Katalysatoren werden Verbindungen des Nickels, wie ΐΓ-Allyl-Ni-X (X=Cl, Br, J), der allgemeinen

Nickel-monocarbonyl-tri-methyl- ■ -■ ■' lsonitril oder Nickel-bis-acrolein, Nickel-bis-acrylnitril, Nickelbis-acrylnitril-dipyridyl,
Nickel-bis-fumaronitril, Nickel-biszimtsäurenitril oder Bis-"7"-AlIy 1-Nickel, Bis-cyclooctadien-nickel
eingesetzt.
Als Lewissäuren kommen vor allem AluminiumalkyIe, Aluminiumalkylhalogenide
oder Aluminiumhalogenide, wie Aluminiumtrialkyle, Dialkylaluminiumhydride,
Dialkylaluminiumhalogenide, Alkylaluminiumsesquihalogenide, Alkylaluminium-dihalogenide, Dialkylaluminiumalkoxyle
zum Einsatz. Werden Nickel-II-Verbindungen eingesetzt,
müssen die Lewissäuren reduzierende Gruppen enthalten. j
Als induzierende Liganden werden erfindungsgemäß Phosphine PR.,
eingesetzt, die entweder das Phosphoratom als chirales Zentrum enthalten, wobei am Phosphor drei verschiedene Reste R gebunden
sind und eines der Enantiomeren (^PR.RpR^) durch Racematspaltung
nach bekannten Methoden gewonnen wurde, oder in denen optisch aktive Reste R* an das Phosphoratom (pR^_n Rn^ gebunden sind.
Auch die Kombination beider Typen ist möglich, so daß Phosphine
mit chiralem Phosphor und optisch aktiven Resten, z.B. ( PR.RpR*)
eingesetzt werden. ' \
Charakteristische Beispiele für die genannten drei Typen sind:

Besonders leicht herstellbar und trotzdem hochwirksam sind die Phosphine des 2. Typs, da man zu ihrer Herstellung auf optisch
aktive Naturprodukte zurückgreifen kann, ohne daß Racematspaltungen
durchgeführt werden müssen. Die besten Ergebnisse liefert das erfindungsgemäße Verfahren, wenn Phosphine eingesetzt werden,
deren Reste R hohe Raumbeanspruchung aufweisen.
Das neue Verfahren läßt sich innerhalb eines weiten Temperaturbereiches,
nämlich etwa zwischen -120° bis +1000C durchführen. Die bevorzugten Reaktionstemperaturen liegen zwischen -78 und
00C.
Die Umsetzung kann bei Atmosphärendruck durchgeführt werden, aber ebenso bei erhöhtem Druck, der durch den Dampfdruck der Olefine
und des verwendeten Lösungsmittels bei der Reaktionstemperatur bestimmt wird.
Das neue Verfahren kann in einem inerten Lösungsmittel durchgeführt
werden. Geeignete Lösungsmittel sind beispielsweise 'chlorierte Kohlenwasserstoffe, wie Chlorbenzol oder Methylchlorid. Es empfieHt
sich, sorgfältig gereinigte und wasserfreie Lösungsmittel zu verwenden.
Mit Hilfe des erfindungsgemäßen Verfahrens lassen sich erstmals optisch aktive Kohlenwasserstoffe auf katalytischem Wege herstellen,
die außer den chiralen C-Atomen funktioneile Gruppen enthalten, so daß weitere Umwandlungen möglich sind. So liefert z.B. die
Polymerisation des verfahrensgemäß herstellbaren, optisch aktiven Vinylcycloocten optisch aktive Polymere. Die Umsetzung des Vinylcyclooctens
mit Borwasserstoffen bzw. Aluminiumwasserstoffen, anschließende Oxydation und Hydrolyse liefern optisch aktiven
Cyclooctenyläthylalkohol. Aufgrund der weitgehenden Zugehörigkeit der Verfahrensprodukte zu jeweils einer der möglichen optischen
Reihen (d bzw. 1 oder R bzw. S) können physiologisch interessante Produkte abgeleitet werden^ die als Arzneimittel verwendet werden
können.

In eine Lösung von 1,845 g ( = 10,25 mMol) X\ -Allylnickelbromid,
5,429 g ( = 12,30 mMol) (-) Tris(trans-myrtanyl)-phosphin (£<]|2 = -12,9°, c = 1,710, C6H6; Schmp. 51 -520C)
und 155,5 g ( = 1,44 Mol) Cyclooctadien-1,3 in 300 ml Chlorbenzol
leitet man "bei O0C unter Rühren Äthylen von 1 atm bis
zur Sättigung ein und startet die katalytisch^ Reaktion durch
Zugabe einer Lösung von 6,51 g ( =26,30 mMol) Äthylaluminiumsesquichlorid
in 85 ml Chlorbenzol (innerhalb von 10 Min.). Die Äthylenzufuhr bei -5° bis O0C wird unter Rühren 6 Stdn. fortgesetzt;
danach bricht man die Reaktion durch Einleiten gasförmigen Ammoniaks in die Lösung bei -3O0C ab. Die Reaktionsmischung
wird auf 200C erwärmt, wobei der Hauptanteil der Äthylendimeren
entweicht; durch Destillation im Vakuum (0,2 bzw. 10 Torr) wird das restliche Reaktionsprodukt vom Katalysator getrennt.
Äthylentrimere (Sdp. 52-70°C/760 Torr) sowie Chlorbenzol
(Sdp. 51-52°C/3O Torr) und nicht umgesetztes Cyclooctadien-1,3
(Sdp. 51-52 u/30 Torr) werden zunächst an einer Vigreux-Kolonne
grob abgetrennt; anschließend destilliert man an einer 1 m-Drehband-Kolonne
und erhält 17,7 g einer Fraktion vom Sdp. 78-83,5°C/
30 Torr\[r\]jp'5 = -12,1° (c = 1,88 ■, CgH6), bestehend aus 1,1 $>
Cyclooctadien-I,3 , 0,2 $ Chlorbenzol, 89,0 ^ 3-Vinylcycloocten,
8,0 io 3-Äthylidencycloocten eis und trans, sowie 39»9 g einer
Fraktion vom Sdp. 83,5-111°C/30 Torr.pnjp5 = -0,9C(e = 2,4 ,
C6H6) mit der Zusammensetzung: 3,4 $>
Vinylcycloocten, 83,5 # eis und trans 3-Äthylidencycloocten; der Rest besteht aus Olefinen
im C12-Bereich. Der Destillationsrückstand (29t3 g)

Oligoniere.

Das durch preparative Gaschromatographie in reiner Form iso-

lierte 3-Vinylcycloocten zeigt einen Drehwert νοη^'ϊ/ί; ' = —13,6
(c = 3,82 , C^Hg), entsprechend einer optischen Ausbeute von
8-9 rfo. Der Drehwert des durch unabhängige Synthesen und Racematspaltung
erhaltenen optisch reinen (-) 3-Vinylcyclooctens beträgt
\k]l5 =-162° (c = 4,8 , G6H6).
Analog dem Beispiel 1 wird zu einer Lösung von 0,310 g ( = 1,73 mMol) T -Allylnickelbromid, 0,879 g ( = 1,99 mMol)
(-) Tris(trans-myrtanyl)phosphin und 26,0 g ( = 241 mMol) Cyclooctadien-1,3 in 100 ml Chlorbenzol in Gegenwart von
Äthylen (1 atm) bei -4O0G eine Lösung von 1,062 g ( = 4,30 mMol)
Äthylaluminiumsesquichlorid in 30 ml Chlcrbenzol über einen Zeitraum von 1 Stde. zugegeben. Äthylen wird noch eine weitere
Stunde bei -45 bis-40 C eingeleitet, dann arbeitet man gemäß
Beispiel 1 auf. Man erhält 3,5 g einer Fraktion mit Sdp. 78-83,5°C/
30 Torr.jt^)^1'5 = -27,9° (c = 8,28 , CgH6), bestehend aus 4,0 <$>
Cyclooctadien-1,3 , 86,4 $ 3-Vinylcycloocten und 6,4 $ eis und
trans-3-Äthylidencycloocten. Der Rückstand (4,9 g) ,Joe] ^8 = -7,7°
(c = 5,43 , G6Hg), besteht zu 44,5 i° aus eis und trans-3-Äthylidencycloocten
und zu 55 Ί° aus C^p-Kohlenwasserstoffen und
höheren Olefinen. Das in reiner Form isolierte 3-Vinylcycloocten zeigt einen Drehwert vonjqjj"5 = -33,3° (c = 8,44 , C5H6),

"bei O0C eine Lösung von 0,95 g ( =3,84 mMol) Äthylaluminiumsesquichlorid
in 40 ml Chlorbenzol über einen Zeitraum von 15 Min. zugegeben. Äthylen wird noch weitere 6 Stdn. bei O0G
in die Lösung eingeleitet und dann das Reaktionsgemisch aufgearbeitet:
Man erhält 2,76 g einer Fraktion mit Sdp. 78-83,50C/
30 Torr mit[Vj ß5 = +7,8° (c = 6,43 , CgH6), bestehend zu 76,1 #
aus ^-Vinylcycloooten und zu 23,0 $ aus eis und trans-3-Äthylidencycloocten.
Daraus errechnet sich für reines 3-Vinylcycloocten
(2,10 g) ein Drehwert vonMJ ^ = +10,2°, entsprechend einer μ
optischen Ausbeute von 6-7 $>*
Lösungen von 0,206 g ( = 1 ,52 mMol) ~u -Allylnickelchlorid,
0,593 g ( = 1,83 mMol) Dimenthylmethylphosphin (ff] p2 = -216°,
c = 3,72 , C6H6; Sdp. 138°C/0,1-0,2 Torr) und 29,6 g ( = 274 mMol)
Cyclooetadien-1,3 in 120 ml Lösungsmittel werden bei .verschiedenen
Reaktionstemperaturen (s. Tabelle I) in Gegenwart von Äthylen mit Lösungen von 0,94 g ( = 3,81 mMol) Äthylaluminiumsesquichlorid Λ
in 20 ml Lösungsmittel versetzt. Nach 6 Stdn. wird gemäß Beispiel 1
aufgearbeitet. Die Cg-Fraktion (Äthylentrimere) enthält u.a. das
optisch aktive 3-Methylpenten-1 (Sdp, 54,3°C/76O Torr), das durch
Feinfraktionierung isoliert wird.
Die Reaktionsbedingungen, die verwendeten Lösungsmittel, die
Ausbeuten an 3-Methylpenten-l bzw. 3-Vinylcycloocten sowie
die optischen Ausbeuten sind in der Tabelle I zusammengestellt:

optisch aktives Phenylmethyl-n-propylphosphin (K. Naumann,
G. Zon, K. Mislow, J. Am. Chem. Soc, £1, 7ol2 (1969)) (charakterisiert
als Phenylmethyl-benzyl-n-propylphosphoniumbromid;
Sdp . 196 - 2o2° C CQq2= -35,o°, c = l,o, CH OH) und 28,3 g
(= 265 mMol) Cyelooctadien-1,3 in loo ml Chlorbenzol in Gegenwart von Äthylen (1 atm) bei -4o°C eine Lösung von I,o9 g
(= 4,38 mMol) Ä'thylaluminiumsesquichlorid in Io ml Chlorbenzol
innerhalb von 5 Minuten zugegeben und anschliessend Äthylen
innerhalb von 3 Stunden bei -4o°C in die Lösung eingeleitet.
Die Aufarbeitung liefert o,45 g einer Fraktion Sdp. 78 - 820C/
3.0 Torr, QC]^= +17,3° (c = 2,5, CgHg), bestehend aus 8,8 %
Cyclooctad'ien-1,3 und 89,9 % 3-Vinylcycloocten. Für reines
p- -.25 ^-Vinylcyeloocten errechnet sich /σΟ™ = +19,25 entsprechend
optischen . D
einer/Ausbeute von 12 %,
In eine Lösung von o,48o g (= 1,86 mMol) Nickelacetylacetonat,
o,945 g (= 2,14 mMol) (-) Tris(trans-myrtanyl)phosphin und 26,3 g (=244 mMol) Cyelooctadien-1,3 in loo ml Chlorbeni'.ol
tropft man unter Rühren bei -4o°C innerhalb von 15 Minuten
eine Lösung von 1,15 S (~ ^j 65 mMol) Äthylaluminiumsesquichlorid
in Io ml Chlorbenzol ein. Man sättigt bei -4o°C mit Propylen
von 1 atm., wärmt auf O0C auf und leitet weitere 2o Stunden
Propylen unter Rühren ein. Durch Zugabe von Io ml konzentriertem Ammoniak wird die Reaktion abgebrochen, die organische
Schicht unter Luftzutritt mit verdünnter Salzsäure geschüttelt,
neutral gewaschen und mit CaCIp getrocknet. Propylendimere (15 g, Sdp, 54 - 72°C/76o Torr) und Chlorbenzol werden an einer
0,7 m-Vigreux-Kolonne grob abdestilliert, der Destillationsrückstand im Hochvakuum umkondensiert (Badtemperatur bis 5o°C),
anschliessend destilliert man an einer o,7 m-Drehband-Kolonne
weiter. Man erhält 24,5 g C1,-Olefine, (Sdp. 98 - lo4°C/3o Torr;

D = 4,65 , unverd.), bestehend aus 27,ο % 3-Isopropenyl-

(in 78 $iger Reinheit) bzw. Bicyclo-f6.0.3]-undecen-3
(in 93 ^iger Reinheit) zeigen Drehwerte vontx7D =

Claims (9)
- PatentansprücheVerfahren zur Herstellung von optisch aktiven Kohlen- · stoffverbindungen aus optisch inaktiven Ausgangsstoffen unter C-C-Verknüpfung, dadurch gekennzeichnet, daß man auf Olefine Katalysatoren einwirken lässt, die durch Kombination von Nickelverbindungen, Lewissäuren und optisch aktiven Phosphinen PR, erhalten werden, die entweder das Phosphoratom als chirales Zentrum oder aber optisch aktive Reste R enthalten.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, daß man Nickelverbindungen, wie Nickelhalogenide, Nickelsalze organischer Säuren, Nickelalkoholate, Nickelenolate, Niekelphenolate bzw.)i -Allylnickelhalogenide verwendet.
- >. Verfahren nach Anspruch 1 und 2, dadurch gekennzeichnet, daß man als Lewissäuren Alumlniumalkyle, Aluminiumalkylhalogenide oder Aluminiumhalogenide einsetzt.
- 4. Verfahren nach Ansprüchen 1 - 3* dadurch gekennzeichnet, daß Nickel-II-Verbindungen und Lewissäuren mit reduzierenden Gruppen verwendet werden.
- 5. Verfahrennach Ansprüchen 1 - 4, dadurch gekennzeichnet, daß man optisch aktive Phosphine einsetzt, die drei verschiedene Reste R enthalten, wobei mindestens ein· Rest R ein Alkyl, Aralkyl oder Arylrest sein muss, und die Phosphine nach bekannten Verfahren in die optischen Antipoden getrennt wurden.
- 6. Verfahren nach Ansprüchen 1 - 5, dadurch gekennzeichnet, daß man Phosphine PR, einsetzt, die mindestens einen optisch aktiven Rest R mit mindestens einem chiralen Zentrum enthalten.
- T. Verfahren nach Ansprüchen 1 - 5, dadurch gekennzeichnet daß man Phosphine einsetzt, die sowohl das Phosphoratom als chirales Zentrum als auch optisch aktive Reste R enthalten und die nach bekannten ,Verfahren Jn JIe optischen Antipoden getrennt
- 8. Verfahren nach Ansprüchen 1 - 5> dadurch gekennzeichnet daß man offenkettige bzw. cyclische Einfach- bzw. Mehrfacholefine, bei deren C-C-Verknüpfung die chiralen Zentren entstehen , verwendet.
- 9. Verfahren nach Ansprüchen 1-8, dadurch gekennzeichnet, daß man bei Temperatur»
-78° bis O0C arbeitet.daß man bei Temperaturen von -12o bis +loo°C, bevorzugt vonIo. Verfahren nach Ansprüchen 1 bis 9, dadurch gekennzeichnet, daß man als Lösungsmittel. Halogenkohlenwasserstoffe verwendet.109887/1878
