Die hier gewählte Definition für R bedeutet sowohl geradkettige als auch verzweigtkettige aliphatische Cibis
Ce-Kohlenwasserstoffreste wie z. B. Methyl-, Äthyl-,
Propyl-, Isopropyl-, Butyl-, Isobutyl-, t-Butyl-, Amyl-
oder Hexyl-Gruppen.
Die Verbindungen (I) der vorliegenden Erfindung können nach an sich üblichen Methoden hergestellt

Gemäß einer Ausführungsform des Verfahrens dieser Erfindung wird (J) durch Kondensieren eines 7-Hydroxyindens,

Die Reaktion wird vorzugsweise in Gegenwart eines säurebindenden Mittels, vorzugsweise von Alkalimetallen
oder deren Derivaten, z. B. Hydroxyd, Hydrid, Amid, Alkoholat und Metall-Alkylverbindungen, durchgeführt.
Ferner wird die Reaktion bevorzugt in Gegenwart von Lösungsmitteln wie Wasser, Methanol oder
Äthanol bei Raumtemperatur oder bei erhöhter Temperatur durchgeführt
Die Verbindungen (I) dieser Erfindung können auch durch Kondensation von 7-(2,3-Epoxypropyl)-inden der
Strukturformel (VI)

mit einem Alkylamin der allgemeinen Formel (VlI)
H2N-R

in der R die genannte Bedeutung hat, hergestellt werden.
Die Reaktion wird in Gegenwart oder Abwesenheit eines organischen Lösungsmittels durchgeführt, beispielsweise
Methanol, Äthanol, Äther oder Äthylacetat bei Raumtemperatur oder erhöhter Temperatur.
Das 7-(2,3-Epoxypropyl)-inden (VI), welches als Ausgangsmaterial benutzt wird, kann durch Umsetzung
von 7-Hydroxyinden (IV) mit l^-Epoxy-3-halogenopropan
erhalten werden.
7-Hydroxyinden (IV) wird in Lösungsmitteln wie Wasser oder organischen Verdünnungsmitteln durch
basische Stoffe isomerisiert und bildet 4-Hydroxyinden.
Aus diesem Grunde enthält die Verbindung (I) der vorliegenden Erfindung ein Isomeres, welches das
4-(3-AIkylamino-2-hydroxy-propoxy)-inden oder auch l-(IndenyI-4-oxy)-3-alkylamino-2-propanol der allgemeinen
Formel

darstellt
Die Verbindungen (I) dieser Erfindung enthalten ein asymmetrisches Kohlenstoffatom in 2-Stellung der
Seitenkette und können daher als d- oder 1-Isomere oder als Racemat vorliegen.
Von den Verbindungen (I) können die im wesentlichen nicht toxischen Säureadditionssalze in der üblichen
Weise mit pharmazeutisch geeigneten anorganischen oder organischen Säuren wie Chlorwasserstoff-, Schwefel-,
Salpeter-, Phosphor-, Ameisen- oder Essigsäure hergestellt werden.
Die erfindungsgemäßen Verbindungen (I) besitzen wertvolle therapeutische Eigenschaften, welche sie
verwendbar als adrenerge ß-Rezeptorblocker (vgl. Versuch 1) machen.
Als eine ausgesprochen wertvolle Eigenschaft dieser Verbindungen sie hervorgehoben, daß sie die Herzfunktion
ohne schädliche Nebenwirkungen regulieren, während bekannte 0-adrenerge Blocker wie 7-(3-Isopropylamino-2-hydroxypropoxy)-indan
gesundheitsschädliche Effekte zur Folge haben (vgl. Versuch 2).
Demgemäß können die Verbindungen (I) dieser Erfindung als ^-Blocker dem Patienten verabreicht
werden, ohne befürchten zu müssen, das Herz zu schädigen.
Eine Charakteristik der Verbindungen (I) der vorliegenden Erfindung wird sichtbar, wenn sie oral
verabreicht werden. Sie können klinisch oral in einer Tagesdosis von etwa 5 bis 15 mg, vorzugsweise in
unterteilten Dosen, gegeben werden. Sie können aber auch intramuskulär oder intravenös verabreicht werden
in einer Totaltagedosis von etwa 2 bis 5 mg, vorzugsweise in unterteilten Dosen.
Die Erfindung wird in den folgenden Beispielen näher beschrieben.
a) Ein Gemisch aus 0,9 g 7-Hydroxyinden, 2,0 g l^-Epoxy-3-chlor-propan, 2,7 g Kaliumcarbonat
und 15 ml Aceton wurde auf etwa 570C 24 Stunden unter Rückfluß gehalten. Das Aceton wurde durch
Vakuumdestillation entfernt, der Rückstand mit 10 ml Wasser gewaschen und dann mit 20 ml Äther
dreimal extrahiert. Der Ätherextrakt wurde mit Magnesiumsulfat getrocknet und filtriert. Dann
wurde der Ätherextrakt in eine Chromatographiesäule (Innendurchmesser etwa 3 cm, Höhe etwa
50 cm), die mit einer Füllung aus Silikagel versehen
war, gegeben. Die fünften bis siebenten Fraktionen (Volumen einer Fraktion beträgt 50 ml), die aus der
Säule unter Verwendung von Chloroform als Eluiermittel erhalten wurden, wurden vereinigt und
eingeengt, wodurch 0,6 g 7-(2,3-Epoxypropoxy)-inden erhalten wurden.
b) Ein Gemisch aus 0,42 g 7-(23-Epoxypropoxy)-inden,
1,20 g Isopropylamin und 20 ml Methanol wurden in einer Flasche bei Raumtemperatur 2
Stunden geschüttelt. Methanol und nicht umgesetztes Isopropylamin wurden durch Vakuumdestillation
abgetrennt Der Rückstand wurde aus einer Mischung aus η-Hexan und Äther umkristallisiert,
lu wodurch 0,41 g 7-(3-Isopropylamino-2-hydroxypropoxy)-inden
vom Schmelzpunkt 88—89°C erhalten wurden.
c) Zu einer Lösung von 0,41 g 7-(3-Isopropylamino-2-hydroxypropoxy)-inden
in 80 ml absolutem Äther
wurde tropfenweise ein Chlorwasserstoffsäure-Äther-Gemisch
bei 0° C unter Rühren zugefügt Die so gebildeten Niederschläge wurden durch Filtration
abgetrennt und aus einem Gemisch von Äthanol-Äther umkristallisiert Es wurden 0,44 g
Hydrochlorid des 7-(3-Isopropylamino-2-hydroxypropoxy)-indens vom Schmelzpunkt 147—148°C
erhalten.

Zu einer Lösung von 1 g 7-(2,3-Epoxypropoxy)-inden in 8 ml absolutem Äthanol wurden 1,2 g sek.-Butylamin
hinzugegeben. Die erhaltene Lösung wurde bei Raumtemperatur über Nacht gerührt. Die Reaktionslösung
wurde unter vermindertem Druck eingeengt
Es wurde ein gelbes öliges Produkt erhalten, welches in 25 ml Diäthyläther aufgelöst wurde. Zu der
erhaltenen Lösung wurde Chlorwasserstoff, gelöst in Äther, tropfenweise hinzugefügt, woraufhin weiße
Niederschläge ausfielen. Die Niederschläge wurden durch Filtration abgetrennt und aus Isopropylalkohol
umkristallisiert. Es wurden 1,1 g 7-(3-sek.-Butylamino-2-hydroxypropoxy)-inden-Hydrochlorid
vom Schmelzpunkt 125—126° C erhalten.

Berechnet: C 64,53, H 8,12, N 4,70, Cl 11,90%;
gefunden: C 64,30, H 8,12, N 4,83, Cl 11,72%.

Zu einer Lösung von 1 g 7-(2,3-Epoxypropoxy)-inden in 6 ml absolutem Äthanol wurden 1,2 g tert-Butylamin
hinzugefügt Die erhaltene Lösung wurde über Nacht bei Raumtemperatur gerührt. Die erhaltene Lösung
wurde unter vermindertem Druck eingeengt. Es wurde ein gelbes öliges Produkt erhalten, welches einer
Säulenchromatographie unter Verwendung einer Silikagel-Füllung
(Innendurchmesser 3.5 cm, Höhe 8 cm) unterworfen wurde. Die neunten bis vierzehnten
Fraktionen (das Volumen einer Fraktion betrug 10 ml)
wurden aus der Säule unter Verwendung von Aceton als Eluiermittel gewonnen, vereinigt und eingeengt und zur
Bildung von Niederschlagen stehengelassen. Die gebildeten Niederschläge wurden aus η-Hexan umkristallisiert
Es wurden 0,2 g weißes kristallines 7-(3-tert-Butylamino-2-hydroxypropoxy)-inden
vom Schmelzpunkt 84-85°C erhalten.
Elementaranalyse (C16H23NO2):
Berechnet: C 73,53, H 8,87, N 5,36%;
gefunden: C 73,66, H 9,13, N 5,32%.

Zu einer Lösung von 0,44 g 7-Hydroxyinden in 4 ml wäßriger 10%iger Kaliumhydroxydlösung wurden
0,65 g l-Chlor-2-hydroxy-3-isopropylaminopropan zugegeben. Nach Durchrühren wurde die erhaltene
Mischung bei Raumtemperatur 15 Stunden stehen gelassen. Das Reaktionsgemisch wurde mit Äther
extrahiert der Ätherextrakt mit wasserfreiem Natriumcarbonat getrocknet und der Äther unter vermindertem
Druck abdestilliert. Der Rückstand wurde in einer Säulenchromatographie (Füllung: Silikagel) unter Verwendung
von Aceton als Eluiermittel unterzogen. 0,23 g weiße Kristalle 7-(3-Isopropylamino-2-hydroxypropoxy)-inden
wurden erhalten.
Wenn diese Kristalle mit dem 7-(3-Isopropylamino-2-hydroxypropoxy)-inden,
erhalten nach dem in Beispiel 1 beschriebenen Verfahren, gemischt wurden, konnte
keine Schmelzpunkterniedrigung festgestellt werden. In bezug auf die adrenerge 0-Rezeptorblockerwir-
kung wurden 7-(3-Isopropylamino-2 hydroxy-propoxy)-inden, erhalten nach Beispiel 1, und 7-(3-lsopropylamino-2-hydroxy-propoxy)-indan,
erhalten nach Beispiel 5 der US-PS 34 15 873, geprüft (vgl. Versuch 1).
Versuch 1
Die Striae, erhalten von einem männlichen Hartley-Meerschweinchen mit einem Gewicht von 300—500 g,
wurden präpariert und in Kammern eingesetzt; Tyrode-Lösung wurde mit 95% Sauerstoff und 5%
Kohlendioxid belüftet und bei 300C als Kammerfüllung gehalten.
Die Kontraktionskraft wird mit einem isometrischen Hebel, der mit einem Belastungsspannungsmesser und
einem Übertragungssystem verbunden war, aufgezeichnet. Die antagonistische Aktivität des zu prüfenden
Arzneimittels gegenüber Isoproterenol wurde durch die erhöhte Herzvorhof-Kontraktion bestimmt
Die Kontrollreaktion wurde durch Zugabe von Isoproterenol zu der Badflüssigkeit mit einer Endkonzentration
von 5 χ 10-8g/ml gemessen. Dann wurde
das Prüfobjekt gewaschen und die gleiche Messung 10 Minuten nach Verabreichung der zu testenden Arzneimittel
wiederholt, nämlich von 7-(3-Isopropylamino-2-hydroxy-propoxyj-inden
bzw. 7-(3-Isopropylamino-2-hydroxypropoxy)-indan (Endkonzentration von 8 χ ΙΟ-9 g/ml oder 5 χ 10~8 g/ml). Die zweite Verbin-
jo dung ist das ähnlich konstituierte Mittel gemäß Stand
der Technik. Die Ergebnisse zeigt Tabelle 1.

Geprüfte Verbindung
Das Ganze wurde zu 20 Tabletten verarbeitet. Die Tabletten wurden mit einem 7,0 mm tiefen Tablettierwerkzeug
gefertigt. Sie können gemäß einem herkömmlichen Verfahren mit einem Schutzüberzug versehen
werden, falls dies gewünscht wird.

Herstellung von Injektions-Präparaten
In 100 ml Salzlösung wurden 100 mg 7-(3-Isopropylamino-2-hydroxy-propoxy)-inden-Hydrochlorid
aufgelöst Die so gebildete Lösung wurde aseptisch in 2-ml-Abmessungen in 50 Ampullen gefüllt, wobei jede
2 mg der aktiven Verbindung enthielt.

Direkte Wirkungen der Verbindungen auf ein Herz ohne autonome nervliche Steuerung wurden gemäß
dem folgenden Versuch 2 untersucht.
Versuch 2
Die Untersuchungen wurden an Wistar-Ratten mit einem Gewicht von 250—350 g, die mit Pentobarbital
(50 mg/kg, i. p.) anästhesiert wurden, 18—24 Stunden nach Reserpingabe (8 mg/kg, s. c.) durchgeführt Beide
Nebennieren wurden entfernt und bilateral der Vagus im Nacken durchschnitten. 1 ml/kg 7-(3-Isopropylamino-2-hydroxy-propoxy)-inden
bzw. 7-(3-Isopropylamino-2-hydroxypropoxy)-indan wurden intravenös jeweils während drei Minuten in einer Einzeldosis im Bereich
von 0,01 bis 0,1 mg/kg injiziert, und der Herzschlag wurde mit einem Cardiotachometer der Firma Nihon
koden Co (RM 100) gemessen. Die Ergebnisse sind in
der Tabelle 2 wiedergegeben. In der Tabelle 2 bedeutet (+) Erhöhung des Herzschlages, (-) bedeutet Erniedrigung
des Herzschlages, und eine Standardfehlerbreite ist in Klammern ( ) angegeben.

- Patentansprüche:\. l-(lndenyl-7-oxy)-3-alkylamino-2-propanole der allgemeinen Formel (I)0-CH2CH(OH)CH3NH-R(Din der R eine gerad- oder verzweigtkettige Ci- bis Ce-Alkylgruppe darstellt, und ihre nichttoxischen Säureadditionssalze.
- 2. 1 -(Indenyl-7-oxy)-3-isopropylamino-2-propanol.
- 3. Verfahren zur Herstellung der l-(Indenyl-7-oxy)-3-aIkylamino-2-propanole der allgemeinen Formel (I) nach Anspruch 1, dadurch gekennzeichnet, daß man in an sich bekannter Weise eine Verbindung der allgemeinen Formel (II)
