2. 2,2-Dimethyl-7-{3-methyI-2-octyl)-4-{4-pyridyl)-2
H-chromen-5-oI und dessen pharmakologisch zulässige Säureadditionssalze.
3. Verfahren zur Herstellung von Verbindungen nach Anspruch i, Formei i, dadurch gekennzeichnet,
daß man ein 5-Hydroxy-7-alkyl-4-(4-pyridyl)-cumarin der nachstehenden allgemeinen Formel II

ι ι in der Ri die angegebene Bedeutung hat, bei etwa 50°C
mit einem Methylmagnesiumhalogenid umsetzt und gegebenenfalls das erhaltene Chromenol in ein Salz
überführt.
Die Grignardierung bei etwa 500C wJ'J in einem
-1D unter den Reaktionsbedingungen inerten organischen
Lösungsmittel, wie Diäthyläther, Dibutyläther, Tetrahydrofuran, Anisol oder Pyridin, durchgeführt. Vorzugsweise
gibt man die Losung des 5-Hydroxy-/-aikyl-4-(4-pyridyl)-cumarins
der Formel II gelöst in Anisol zu
-'Ί einer Lösung des Methylmagnesiumhalogenids in Anisol.
Die Umsetzung wird mindestens 24 Stunden bei dieser Temperatur durchgeführt.
Zur Herstellung der Ausgangsverbindung der For-(111
mel II wird ein /?-Oxo-4-pyridinpropionsäure-alkylester
;<> der allgemeinen Formel III
in der Ri die angegebene Bedeutung hat, bei etwa
50=C mit einem Methylmagnesiumhalogenid umsetzt
und gegebenenfalls das erhaltene Chromenol in ein Salz überführt.
4. Arzneipräparat, enthaltend eine Verbindung nach Anspruch 1, Formel I, als Wirkstoff, gegebenenfalls
in Kombination mit pharmakologisch zulässigen Träger- und/oder HilfsStoffen.
5. Ausführungsform nach Anspruch 5 in Form eines oral verabreichbaren Präparates zur Stimmungsbeeinflussung,
enthaltend die Verbindung
2,2-Dimethyl-7-(3-methyl-2-octyl)-4-(4pyridyl)-2 Hchromen-5-ol oder ein davon abgeleitetes Salz
als Wirkstoff.
Die Erfindung betrifft 2,2-Dimethyl-7-alkyl-4-(4-pyndyl)-2
H-chromen-5-ole der nachstehenden allgemeinen Formel und deren pharmakologisch zulässige Salze

Zur Herstellung der Salze der Chromenolverbindungen der Formel I werden die freien Basen in einer wäßrigen
Lösung gelöst, die die einsprechende Säure enthält, und durch Eindampfen der Lösung isoliert. Beispiele
für zur Salzbildung verwendbare Säuren sind Salzsäure, Bromwasserstoffsäure, |odwasserstoffsäure
und Picrinsäure. Sowohl die freien Basen als auch die Salze mit Säuren haben pharmakologischc Aktivität.
Zur Herstellung von Arzneipräparaten werden zur
Salzbildung pharmakologisch verträgliche Säuren verwendet.
Die Verbindungen der Formel I haben cardiovaskuläre und Zentralnervensystemaktivität. Bei oraler Verabreichung
an Mäuse und Ratten im Standardtest, bei denen die psychomotorische Aktivität, die Reaktionsfähigkeit
gegenüber Reizen und die Fähigkeil, normale, nicht bedingte motorische Aufgaben durchzuführen,
beobachtet wurde, konnte diese Aktivität festgestellt werden. Die Verbindungen können daher zur Behandlung
von Hochdruck und als Analgetika sowie als Antidepressiva und Ataraktika verwendet werden. Die Verbindungen
zeigen auch Aktivität bei einer Anzahl von bedingten Verhaltenstests bei Affen. Diese Aktivität ist
ein Anzeichen für ihre Brauchbarkeit als psychotrope Verbindungen. Außerdem erzeugen bestimmte Verbindungen
Vagusreizungen bei Blutdruckuntersuchungen an der Katze.
Ein wesentlicher Vorteil der erfindungsgemäßen Verbindungen ist ihr ungewöhnlich breites Wirkungsspektrum
in bezug auf die Beeinflussung der Psyche und die Tatsache, daß sie sich von bekannten Neuroleptika und
Tranquillisern unterscheiden. So tritt mit den erfindungsgemäßen
Verbindungen bei niedriger Dosierung eine deutliche Beruhigung und Narkose- und Hypnotikapotenzierung
ein, während die Motorkoordination und die Lernfähigkeit noch nicht beeinträchtigt sind.
Die Erfindung betrifft daher Arzneipräparate. die
eine Verbindung der Forme! I als Wirkstoff, gegebenenfalls in Kombination mit pharmakologisch zulässigen
Träger- und/oder Hilfsstoffen, enthalten.
In einer bevorzugten Ausführungsform enthält das oral verabreichbaic Präparat zur Slimmungsbeeinflussung
die Verbindung 2,2- Dimethyl-"■ (3-methyl-2-octyl)-4-(4-pyridyl)-2
H-chromen-5-pl oder ein davon abgeleitetes Salz als Wirkstoff.
Die erfindungsgemäßen Verbindungen können oral oder parenteral verabreicht werden. Zur enteralen Verabreichung
kommen Tabletten, Pillen, Pulver, Kapseln, Granulate, Suspensionen, Dispersionen, Lösungen oder
Emulsionen in Frage, die Verdünnungsmittel, Bindemittel, Dispergiermittel, Netzmittel, Geschmacksverbesserer,
Gleitmittel, Farbstoffe, Überzugsmittel, Lösungsmittel oder Eindickmitlei enthalten können. Einzeldosen
in Form von Tabletten oder Kapseln zur oralen Verabreichung enthalten den Wirkstoff entweder
allein oder in Kombination mit geeigneten Hilfsstoffen, wie Calciumcarbonat, Stärke. Milchzucker, Talkum, Ma
gnesiumstearat oder Akazienschleim in wäßrigem Alkohol, Glykol oder öllösungen oder Öl-in-Wasser-Emulsionen.
Die Injektionspräparate können als wäßrige
oder nicht wäßrige Lösung, Suspension oder Emulsion in einer pharmakclogisch verträglichen Flüssigkeit
oder einem Flüssigkeitsgemisch hergestellt werden und können Bakteriostatika. Antioxydationsmittel. Puffer
oder andere Zusätze enthalten, um die Präparate isoton einzustellen. Weiter können diese Präparate Eindickmittel,
Suspendiermittel oder andere pharmakologisch verträgliche Zusätze enthalten. Die injizierbaren Präparate
werden sterilisiert und in Einzeldosen, Ampullen oder Flaschen verpackt. Präparate zur intramuskulären
Injektion können durch Auflösen eines Salzes der erfindungsgemäßen Verbindungen in Wasser oder einer
äquivalenten Menge einer nichttoxischen Säure oder in einem physiologisch verträglichen wäßrigen Medium,
wie physiologischer Kochsalzlösung, gelöst und in Ampullen abgefüllt werden.
Die Beispiele erläutern die Erfindung.
Beispiel 1
5-Hydroxy-7-(3-methyl-2-octyl)-4-(4-pyridyl)-cumarin
Ein Gemisch aus 2,1 g (11,7 mMol) /J-Oxo-4-pyridinpropionsäuremethylester
und 2,8 g (11,8 mMol) 5-(3-Methy!-2-octyl)-resorcin wird tropfenweise unter Kühlung mit 5 ml konzentrierter Schwefelsäure versetzt.
Danach werden in das Gemisch rasch 3 ml Phosphoroxychlorid eingetragen. Die viskose Lösung wird
bei Raumtemperatur 48 Stunden gerührt. Anschließend wird das Reaktionsgemisch in kalte Kaliumbicarbonatlösung
eingegossen und mehrmals mit Äther extrahiert. Die vereinigten Ätherextrakte werden gewaschen, getrocknet
und eingedampf„ Es hinterbleibt ein gelber Feststoff, der aus verdünntem Äthanol umkristallisiert
wird.
Ausbeute: 3,4 g, Fp. 191 bis 194 C.

2,2-Dimethyl-7-(3-methyl-2-octyl)-4-(4-pyridyl)-2 H-chromen-5-ol
Eine Lösung von 5,0 g (13,7 mMol) 5-Hydroxy-7-(3-methyI-2-octyl)-4-(4-pyridyl)-cumarin,
hergestellt gemäß Beispiel I, in Anisol wird tropfenweise zu einer Lösung von Methylmagnesiumbromid in Anisol gegeben,
die aus 3,3 g (138 mMol) Magnesium hergestellt worden ist. Nach beendeter Zugabe wird das Gemisch
gerührt und 16 Stunden auf 50°C erwärmt. Nach dem Abkühlen wird das Gemisch in überschüssige 4 n-Schwefelsäure
eingegossen und zur Abtrennung von Anisol dampfdestilliert. Der Rückstand wird mit Äther
extrahiert, die vereinigten Ätherextrakte werden gewaschen, getrocknet und eingedampft. Es hinterbleibt
ein rötliches öl, das in 250 ml Heptan aufgenommen, mit 5 ml 48prozentiger Bromwasserstoffsäure versetzt
und 6 Stunden unter Rückfluß erhitzt wird. Nach dein Abkühlen wird das Heptan im Rotationsverdampfer
abdestilliert und der Rückstand mit Äther extrahiert. Der Ätherextrakt wird mit wäßriger Natriumbicarbonatlösung
und dann mit Wasser gewaschen und über Natriumsulfat getrocknet. Nach dem Abdampfen des
Äthers hinterbleibt ein etwas schmieriger Rückstand, der beim Anreiben mit Acetonitril kristallisiert. Das
feste Reaktionsprodukt wird aus Acetonitril umkristallisiert.

Pharmakologischc Versuche

Die Verbindung des Beispiels 2, 2,2-Dimeihyl-7-(3-methyl-2-octyI)-4-(4-pyridyl)-2
H-chronien-5-ol, ist ungiftig; sowohl bei der Ratte als auch bei der Maus
liegt der LDso-Wert nach oraler Verabreichung über 5000 mg/kg. Ein exakter Wert kann wegen des Mangels
an Toxizität nicht ermittelt werden.
Entsprechendes gilt für die Verbindung der Formel I, in der Ri der 2-Octylrest ist.
b) Vermeidungs-Konditionierungstest
Im Prinzip müssen bei diesem Test Tiere eine bestimmte Aufgabe durchführen, d. h. einen Hebel drükken
oder ein Gitter (Barriere) zu einem anderen Teil des Käfigs passieren, um dadurch einem schwachen
Stromschlag auf die Pfoten zu entgehen. Das Tier hat gelernt, daß ein Licht oder ein Signal den bevorstehenden
Schock anzeigt und daß es innerhalb einer bestimmten Zeit die Aufgabe durchführen muß (>/konditiuniener
Stimulus«), um dem Schock zu entgehen (»nicht konditionierter Stimulus«).
Die Wirkung von Chlorpromazin und ähnlichen Neuroleptika oder antipsychotischen Mitteln in diesem
Test ist von M. Ourdtin in »Psychopharmacrilogical
Agents«, Band 2, Academic Press, Ixindun, 1%7, beschrieben
worden. Chlorpromazin-ähnliehe MitIeI unterbinden
in typischer Weise dieses KonditionierLngsverhalten.
Mil erfindungsgemäben Verbindungen ist die Ratte bei einer Dosis von 5 mg/kg noch aktiv.
c) Andere Tests
Bei anderen bekannten Tests erhält man mit der Verbindung des Beispiels 2 in Form des HCI-Additionssalzes

Das Aktivitätsspektrum der Verbindung der Formel I, in der Ri der 2-Octylrest ist, ist ähnlich breit, die Verbindung
ist jedoch insgesamt nur etwa ein Drittel so aktiv.
Bei beiden Verbindungen tritt son it bereits bei niedriger
Dosierung eine deutliche Beruhigung ein, die Motorkoordination und die Lernfähigkeit sird jedoch
noch nicht beeinträchtigt.
Claims (1)
1. 2,2-Dimethyl-7-alkyl-4-(4-pyridyl)-2 H-chromen-5-ole
dar nachstehenden allgemeinen Formel und deren pharmakologisch zulässige Salze
Die Erfindung betrifft ferner ein Verfahren zur Herstellung der Verbindungen der Formel I, das dadurch
gekennzeichnet ist, daß man ein 5-Hydroxy-7-alkyl-4-(4-pyridyl)-cumarin der nachstehenden allgemeiner
