3. Arzneimittel, enthaltend die Verbindung gemäß Anspruch 1 als Wirkstoff.

Die Erfindung betrifft ein neues Phenylacetonitrilderivat und dessen Salze sowie Verfahren zu deren
Herstellung und diese Verbindungen enthaltende Arzneimittel entsprechend den vorstehenden Patentansprüchen.
Die deutschen Patente 1154 810 und 1158 083
betreffen Verfahren zur Herstellung basisch substituierter Phenylacetonitrile, welche infolge ihrer pharmakologischen
Wirksamkeit wertvolle Arzneimittel darstellen und als Herzsympaticolytica und als Coronardilatoren
Verwendung finden können.
Es wurde nun gefunden, daß auch das a-(3,4,5-Trimethoxybenzyl)
-«- [(N - methyl - N - homoveratryl)-5'
- aminopropyl] - 3,4,5 - trimethoxyphenylacetonitril
der Formel

basisch substituierten Phenylacetonitrile Verwendung fanden, indem man

durch Methylierung in das entsprechende tertiäre Amin überführt.
Die Alkylierungsreaktionen erfolgen bei erhöhter Temperatur, wobei man die Reaktionspartner in verschiedener
Weise aufeinander einwirken lassen kann. Man kann beispielsweise die Verbindung der Formel
II mit dem basischen Kondensationsmittel, vorzugsweise Natriumamid, so lange in einem organischen
Lösungsmittel zum Sieden erhitzen, bis die Ammoniakentwicklung abklingt, und dann die berechnete
Menge der Verbindung III hinzugeben. Hierbei kann die Verbindung III im gleichen Lösungsmittel gelöst
sein. Man kann aber auch die Verbindung III und ein Kondensationsmittel gleichzeitig auf das Nitril einwirken
lassen, hierbei wird die Bildung der Nebenprodukte, wie beispielsweise Amidinsalze und Polymerisationsprodukte,
zurückgedrängt. Weiterhin kann man auch in der Weise vorgehen, daß man der siedenden
Lösung beider Reaktionspartner langsam die Suspension des basischen Kondensationsmittcls zugibt.
Bei kleineren und mittleren Ansätzen besteht eine weitere Möglichkeit zur Durchführung der Umsetzung
darin, die drei Reaktionspartner in der Kälte mit dem Lösungsmittel zusammenzugeben und allmählich
zu erwärmen. In vielen Fällen läuft die Reaktion bereits unterhalb des Siedepunkts des Lösungsmittels
ab.
Als basisches Reaktionsmittel kommen bei Durchführung der Reaktion neben Natriumamid auch Natrium,
Natriumhydrid, Natriumhydroxyd, Kaliumamid, Lithiumamid oder Lithium- bzw. Magnesiumverbindungen
sekundärer Amine in Betracht.
Als Lösungsmittel für die Reaktion sind aromatische Kohlenwasserstoffe wie Benzol, Toluol oder Xylole
besonders gut geeignet. Man kann jedoch auch höhersiedende aliphatische Äther verwenden.
Verwendet man als Ausgangsmaterial Verbindungen der Formel IV mit einem Wasserstoffatom in
«-Stellung zur Nitrilgruppe, so verfährt man in entsprechender Weise unter Anwendung der obengenannten
basischen Kondensationsmittel und Lösungsmittel im gleichen Temperaturbereich. Auch bei
dieser Ausführungsform des Verfahrens erfolgt die Zugabe der Reaktionspartner und des pasischen Kondensationsmittels
in der oben beschriebenen Reihenfolge.
Eine andere Ausführungsform des Verfahrens besteht in der Umsetzung der Verbindungen V mit ter-

tiären Aminen der allgemeinen Formel UI und Umsetzung einer Verbindung der Formel

in Gegenwart der genannten basischen Kondensationsmittel. Hierbei kann die Reihenfolge der Zugabe
der Verbindungen beliebig gewählt und auf eine Isolierung des Zwischenproduktes der Formel III
verzichtet werden.
Verwendet man zur Herstellung des neuen (L - (3,4,5 - Trimethoxy benzyl) - « - [(N - methyl - N - homoveratryl)
-γ- aminopropyl] - 3,4,5 - trimethoxyphenylacetonitrils als Ausgangsmaterial Verbindungen der
Formel VI und das Amin VlI, so erfolgt die Umsetzung durch einfaches Erhitzen der beiden Reaktionskomponenten.
Bei nachträglicher Einführung der Methylgruppe in die erhaltenen sekundären Amine kann man beispielsweise
so vorgehen, daß man dieselben mit Formaldehyd und einem Reduktionsmittel oder mit Methylhalogeniden
bzw. Dimethylsulfat in bekannter Weise behandelt.
Die Ausgangsverbindung für das Verfahren e) ist beispielsweise erhältlich durch Umsetzen von Homoveratrylamin
mit 4,5-(3,4,5-Trimethoxyphenyl)-4-cyan-1-chlorpentan
oder reduktive Kondensation von Homoveratrylamin mit 4,5-(3,4,5-Trimethoxyphenyl)-4-cyan-pentanal-l
oder reduktive Kondensation von 4,5-(3,4,5-Trimethoxyphenyl)-4-cyan-1 -aminopentan
mit 3,4-Dimethoxyphenylacetaldehyd.
Untersucht man die Veränderung der funktioneilen Refraktärzeit, welche die neue Verbindung am linken
Meerschweinchen-Vorhof bewirkt (Methode der gepaarten elektrischen Stimulation in Anlehnung an
W. C. Go vier, J. Pharmokol. Exp. Therap. 148, 100 [1965]) und bestimmt zusätzlich die Kontraktionskraft
des Herzmuskels, so zeigt sich, daß die neue Verbindung im Vergleich mit Verapamil in
äquieffektiv refraktärzeitverlängernden Dosen die Kontraktionskraft des Herzens wesentlich weniger
herabsetzt. So verlängert sich die funktionell Refraktärzeit bei der Applikation von 0,01 mMol/1 Verapamil
und von 0,05 mMol/1 a-(3,4,5-Trimethoxybenzyl) -<L- [(N - methyl - N - homoveratryl) -γ- aminopropyl]-3,4,5-trimethoxyphenylacetonitril
um etwa denselben Betrag (+ 77% bzw. + 74%). Nach der Verapamilgabe sinkt die Kontraktionskraft des isolierten
linken Meerschweinchen-Vorhofs um 53% bei der neuen Verbindung jedoch nur um 31%. Trotz höherer
Dosierung ist die neue Verbindung daher dem Verapamil deutlich im Hinblick auf den Sicherheitsabstand
zwischen erwünschter und unerwünschter Wirkung überlegen.
Die Erfindung wird nachstehend an Hand von Ausführungsbeispielen beschrieben. (Die Schmelzpunktangaben
sind korrigierte Werte.)
Zu einer Lösung von 193,7 g (0,5 Mol) 2,3-Bis-(3,4,5 -trimethoxyphenyl)-propionitril und von aus
154,2 g (0,5 Mol) ihres Hydrochlorids frisch bereitetem
N - Methyl - N - 3 - chlorpropyl - homoveratrylamin in 2 1 trockenem Toluol werden bei 105 bis 110 C
unter Rühren innerhalb von 45 Minuten 84,5 μ (1,3 χ 0,5 Mol) einer 30%igen Natriumamidlösung
getropft. Nach 4stündigem Erhitzen unter Rückfluß wird das Reaktionsgemisch in Eiswasser eingetragen
und mit Salzsäure angesäuert, wobei sich ein zähflüssiges öl abscheidet. Man trennt die Toluolschicht
ίο ab, versetzt die wäßrige Phase zusammen mit dem öl
mit Natronlauge und nimmt die Base in Toluol auf. Nach Trocknen der Toluollösung über Kaliumcarbonat
und Eindampfen erhält man 309 g «-(3,4,5-Trimethoxybenzyl) - a - [(N - methyl - N - homoveratryl)-v-aminopropyl]-3,4,5-trimethoxyphenylacetonitril.
Durch Umsetzen dieser Base mit Chlorwasserstoff in Isopropanol und Essigsäureäthylester erhält man
das Hydrochlorid, Fp. = 194 bis 195,5° C, Ausbeute 270 g = 81% der Theorie.
Das Phosphat scheidet sich aus der Lösung der Base in Isopropanol/Essigsäureäthylester 1:1 nach
Zugabe von Phosphorsäure aus, Fp. = 162 bis 1650C
(Äthanol). Das Salz enthält pro Mol Base 2 Mol Phosphorsäure.
Das als Ausgangsmaterial benötigte 2,3-Bis-(3,4,5 - trimethoxyphenyl) - propionitril erhält man
durch Umsetzen von 3,4,5-Trimethoxybenzylcyanid und 3,4,5-Trimethoxybenzylchlorid mit Natriumamid
in Toluol bei etwa 1000C, Extraktion der Toluollösung mit Eiswasser und fraktionierter Destillation
der organischen Phase, Kp. = 216 bis 22 Γ C. Fp. = 118 bis 122°C (Isopropanol).
a) 387,4 g (1 Mol) 2,3 - Bis - (3,4,5 - trimethoxyphenyl)-propionitril
(vgl. Beispiel 1) werden in 1,5 1 trockenem Toluol gelöst und mit 46,8 g (1,2 Mol)
Natriumamid 2 Stunden unter Rückfluß erwärmt. Nach Abkühlen auf Raumtemperatur werden 314,9 g
(2MoI) l-Brom-3-chlor-propan zugegeben. Sobald
die leicht exotherme Reaktion abgeklungen ist, erwärmt man das Gemisch 2 Stunden auf 9O0C und
anschließend 3 Stunden unter Rückfluß. Die Toluolphase wird mit Eiswasser extrahiert und bis 1500C
(Badtemperatur)/10 Torr eingeengt. Der Rückstand enthält 301 g (0,648 Mol) l,2-Bis-(3,4,5-trimethoxyphenyl)-2-cyan-5-chlorpropan,
Ausbeute 65% der Theorie.
b) Der gemäß a) erhaltene Rückstand wird mit 254,4 g (2 χ 0,648 Mol) Methylhomoveratrylamin vermischt
und unter Rühren unter Stickstoff 4 Stunden auf 110 bis 1500C gehalten. Anschließend fügt man
2 1 Toluol zu. Der entstandene Niederschlag wird abfiltriert und das Filtrat mit 250 ml konzentrierter
Salzsäure in 1,5 1 Wasser versetzt. Das abgeschiedene ölige Hydrochlorid wird mit Wasser verrührt, nach
Zugabe von Natronlauge mit Toluol extrahiert, getrocknet und eingedampft. Das ölige «-(3,4,5-Trimethoxybenzyl)-«-[(N-methyl-N-homoveratryl)-y-aminp-
propyl]-3,4,5-trimethoxyphenylacetonitril wird analog Beispiel 1 in das Hydrochlorid übergeführt. Man
erhält 333 g(78% der Theorie), Fp. = 194 bis 195,5°C.
Ein nach Beispiel 2 a) erhaltenes Gemisch, enthaltend 301 g (0,648 Mol) l,2-Bis-(3,4,5-trimethoxyphenyl)-2-cyan-5-chlorpentan
wird in gleicher Weise wk im Beispiel 2b) beschrieben mit 234,9 g (2 χ 0,648 Mol
Homoveratrylamin kondensiert und aufgearbeitet. Dabei fallen 385 g « - (3,4,5 - Trimcthoxybenzyl)-
Ii - [(N - homoveratryl) - ;· - aminopropyl] - 3,4,5 - trimcthoxyphenylacetonitril
an. Man löst diese Substanz in 1,2 1 Äthanol, fügt 55,7 g 35gewichtsprozentige
wäßrige Formalinlösung sowie 25 g 10%igc Palladium-Kohle hinzu und hydriert bei 760 Torr/
45'' C. Nach 14 Stunden filtriert man vom Katalysator ab, verdampft das Lösungsmittel, löst den öligen
Rückstand in 400 ml Isopropanol und säuert mit konzentrierter Salzsäure an. Das auskristallisierte
Hydrochlorid ist nach zweimaligem Umlösen aus Isopropanol rein. Die Ausbeute beträgt 302 g = 71%
der Theorie. Fp. = 194 bis 195° C.
Man kann das in der ersten Stufe anfallende sekundäre Amin auch in nachstehender Weise am basischen
Stickstoff methylieren: 385 g «-(3,4,5-Trimethoxybenzyl)-u-[(N-homoveratryl)-;'-aminopropyl]-3,4,5
-trimethoxyphenylacetonitril werden in 1,5 1 Äthanol gelöst, mit etwa 33 g 99%iger Ameisensäure neutralisiert,
mit 77,2 ii 35%iger wäßriger Formalinlösung (= 25,3 g CH2O = 1,3 χ 0,648 Mol) versetzt und
2 Stunden unter Rückfluß zum Sieden erhitzt. Während der ersten Stunde tropft man weitere 39,1 g
(1,3 χ 0,648 Mol) 99%ige Ameisensäure zu. Man dampft das Lösungsmittel ab, versetzt den Rückstand
mit Natronlauge und nimmt die entstandene tertiäre Base in Toluol auf. Nach Abdampfen, Lösen des
Rückstands in Isopropanol und Ansäuern mit konzentrierter Salzsäure erhält man das kristallisierte
«-(3,4,5 -Trimethoxybenzyl)- a- [(N -methyl -N- homoveratryl)
- γ - aminopropyl] - 3,4,5 - trimethoxyphenylacetonitril-Hydrochlorid.
Nach zweimaligem Umlösen aus Isopropanol beträgt die Ausbeute 285 g = 67% der Theorie.
