Abstract
Το γενωμικό DNA του Streptoverticillium sp. 3-7, το οποίο παράγει το UK-2, αναλύθηκε για την ταυτοποίηση μιας περιοχής αναμενόμενης να είναι ένα σύμπλεγμα UK-2 βιοσυνθετικών γονιδίων. Επιπλέον, με υβριδοποίηση αποικίας, απομονώθηκαν επιτυχώς τα DNA στην περιοχή. Περαιτέρω, τα DNA χρησιμοποιήθηκαν για την παρασκευή ενός στελέχους στο οποίο είχαν διασπαστεί τα γονίδια που υπήρχαν στην περιοχή. Το στέλεχος βρέθηκε ότι δεν παράγει UK-2. Επιβεβαιώθηκε ότι η γενωμική περιοχή ήταν το σύμπλεγμα UK-2 βιοσυνθετικών γονιδίων. Περαιτέρω, το Streptoverticillium sp. 3-7 μετασχηματίστηκε με εισαγωγή ενός φορέα στον οποίο είχε εισαχθεί το απομονωμένο σύμπλεγμα UK-2 βιοσυνθετικών γονιδίων. Βρέθηκε επίσης ότι η παραγωγικότητα σε UK-2 από το μετασχηματισμένο κύτταρο ήταν βελτιωμένη περίπου 10 έως 60 φορές ή περισσότερο σε σύγκριση με εκείνη του γονικού στελέχους. Επιπλέον, αποκαλύφθηκε ότι υπήρχαν 2 αντίγραφα του συμπλέγματος UK-2 βιοσυνθετικών γονιδίων ανά κύτταρο σε αυτά τα μετασχηματισμένα κύτταρα, αντιστοίχως.
