Abstract
Παρασκευή των βιοφαρμακευτικών από CHO κύτταρα απαιτεί δημιουργία κύριων-, εργασίας- και μετά την παραγωγή τραπεζών κυττάρου υψηλής ποιότητας, μερικώς υπό GMP συνθήκες. Απαιτείται μία βέλτιστη στρατηγική κρυοσυντήρησης για κάθε νέα παραγωγή κυτταρικής γραμμή, ιδιαιτέρως σε σχέση προς τις επιθυμητές για να θεμελιώνονται μέθοδοι παραγωγής που αποφεύγουν πλήρως ορό ή ακόμη οποιαδήποτε ζωικά συστατικά και για να διασφαλίζεται εύρωστη συμπεριφορά επανατήξης για αξιόπιστη παραγωγή. Εδώ περιγράφουμε μία νέα στρατηγική που χρησιμοποιεί κυτομετρική ανάλυση ροής (FC) των κυττάρων που έχουν υποστεί χρώση με Annexin V για υψηλής απόδοσης χαρακτηρισμό των τραπεζών κυττάρου CHO. Τα δεδομένα μας δείχνουν ότι αυτή η μέθοδος επιτρέπει αξιολόγηση μιας μεθόδου κρυοσυντήρησης μόλις 6 ώρες μετά την επανατήξη.
