Abstract

La invención se relaciona con mamíferos trasgénicos no humanos que son útiles en la producción de una proteína de interés que puede ser tóxica para los mamíferos. El mamífero se caracteriza por el hecho de que es transgénico para la producción en su leche de una forma inactiva de la proteína de interés, preferiblemente insulina humana recombinante. No es posible producir insulina humana recombinante en mamíferos transgénicos en razón a que esta molécula tiene un cierto grado de actividad biológica en Los mamíferos y podría ser tóxica para el mamífero. Así, la invención involucra donar una construcción genética que comprende una secuencia que codifica un precursor modificado de insulina humana bajo el control de un promotor de caseína beta en un vector de expresión. Esto también involucra transfectar el plásmido de expresión en las células somáticas bovinas fetales, tal como los fibroblastos, y enuclear los oocitos bovinos mediante transferencia nuclear para generar embriones transgénicos. La invención da origen a un bovino transgénico que será capaz de producir un precursor modificado de insulina humana en sus glándulas mamarias. Posteriormente, la leche de estos mamíferos transgénicos se puede recolectar, el precursor modificado de insulina humana se puede convertir in vitro en insulina humana recombinante, y la insulina humana recombinante se puede purificar para homogenizarla como un producto biofarmacéutico puro.
