发明背景
营养研究中最新和最关心的领域之一包括所谓植物化学品的食物成分。植物化学品,如异黄酮和其衍生物,仅在植物中发现,它们虽不归类为营养品,但深深影响我们的健康。参见如W.R.Bidlack等编著:植物化学品,一种新的典范,Technomic出版社出版,1998。
植物化学品的发现引起营养学新时代的曙光。见于大豆等豆类的植物化学品特别重要。豆类和其制品,例如大豆和大豆制品提供降低如癌等许多慢性疾病发生的特殊渠道。参见农业食品化学杂志,42,1666(1994);和43,1184(1995)。近年来研究表明,每日一餐大豆食品足于获得这些食物化学品之益处(参见M.Messina等著:单纯大豆和我们的健康,NY,花园城市公园,Avery出版社出版,1994)。
但大豆食物具有臭味、粗质等原因,因而多数人不愿食用。为避免不喜欢大豆食物,一种解决方式为以大豆异黄酮为饮食补助品,但不幸因其水溶性低,故大豆异黄酮和其衍生物仍未被利用。参见R.Elkinsa著:染料木苷素,犹它州,快乐谷,Woodland出版社出版,1998。
发明概述
式中R1为氢或羟基,R2为氢或甲氧基,R3为氢或CO-R4,R4为甲基或羧甲基。此水溶性豆类萃取物为干态时包括约5-约50重量份的异黄酮衍生物,于约25℃水中的溶解度为约10-约1000mg/ml。存在于水溶性豆类萃取物的异黄酮例子包括大豆异黄酮苷(daidzin)、染料木苷(genistin)、黄豆苷(glycitin)、6″-O-乙酰基大豆异黄酮苷、6″-O-乙酰基染料木苷、6″-O-乙酰基黄豆苷、6″-O-丙二酰基大豆异黄酮苷、6″-O-丙二酰基染料木苷或6″-O-丙二酰基黄豆苷。
本发明的水溶性豆类萃取物包括一种或多种异黄酮衍生物,例如一种异黄酮糖苷,其与豆类中的其它成分复合而意外地提高了水溶性。
另一方面,本发明还表征了水溶性豆类萃取物的制备方法,其包括将粉化的粗制豆类萃取物与水混合,加热该混合物,添加凝结剂到混合物中而形成悬浮液后,加热该悬浮液,并收集上清液,则得到水溶性豆类萃取物。该水溶性豆类萃取物具有风味佳的特点,且当作任何食品之成分时,都成为适宜的组成物,并有上好表现。其可与如牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤等食品配合。

发明的详细描述
本发明涉及一种水溶性豆类萃取物,其包括一种或多种如下式所示的异黄酮衍生物
式中R1为氢或羟基,R2为氢或甲氧基,R3为氢或CO-R4,R4为甲基或羧甲基。
存在于本发明水溶性豆类萃取物的异黄酮的例子包括大豆异黄酮苷(daidzin)、染料木苷(genistin)、黄豆苷(glycitin)、6″-O-乙酰基大豆异黄酮苷、6″-O-乙酰基染料木苷、6″-O-乙酰基黄豆苷、6″-O-丙二酰基大豆异黄酮苷、6″-O-丙二酰基染料木苷或6″-O-丙二酰基黄豆苷。如大豆异黄酮苷、染料木苷素等异黄酮配糖于胃或小肠中被吸收而由人小肠内产生鼠李糖、外β葡萄糖苷、内β葡萄糖苷的微生物经糖苷配基代谢为异黄酮。参见如Biol.Pharm.Bull,22(1998);Biol.Pharm.Bull,17,1369(1994);Arch.Pharm.Res.,21,17(1998)。
本发明的水溶性豆类萃取物增进水溶性,这提高了用于食品时异黄酮衍生物的生理吸收。例如,金雀异酮苷于25℃甲醇中的溶解度约5mg/ml,于25℃水中约1mg/ml,而包括约10%异黄酮衍生物的本发明水溶性豆类萃取物在水中的溶解度约为300mg/ml。代表性地,本发明的水溶性豆类萃取物使异黄酮衍生物的溶解度增加了约30倍。在干态时,包括约10到约50重量份的异黄酮衍生物,于约25℃水中的溶解度为约10到约1000mg/ml;优选地,含有约10到约20重量份的异黄酮衍生物,水中的溶解度为约30到约300mg/ml;尤特别优选地,含有约10-约20重量份的异黄酮衍生物,水中的溶解度为约50到约200mg/ml。当然,异黄酮衍生物的重量百分比影响水溶性豆类萃取物的总溶解度。例如,含异黄酮衍生物较高重量百分比的萃取物的总溶解度低于含异黄酮衍生物较低重量百分比的萃取物。
任何豆类皆可用以制造本发明的水溶性豆类萃取物。用以制造本发明的水溶性豆类萃取物的豆类包括但不限于大豆、绿豆、黑豆、云豆。优选的豆类为大豆。通常可用二种不同型大豆,例如食用豆和制油用豆,来制造本发明的水溶性豆类萃取物。相对于食用豆来说,制油用豆的蛋白质含有量更少,因而制油用豆更为优选。代表性地,先除去制油用豆中的油份以制成加工豆。除油法可参见K.S.Liu著:大豆化学、技术和利用,马利兰州,盖滋堡,Aspen出版社出版,1999。
本发明水溶性豆类萃取物的制备方法包括混合以重量计的约1份到约50份的豆粉粗萃取物和约100份热或冷水。豆粉粗萃取物可通过用约50-90%醇水溶液或100%有机溶剂,如乙醇、甲醇或乙腈萃取来制造。豆粉粗萃取物的一个实例为大豆酱。大豆酱代表约1%重量的大豆,然而包括了相对剩余的大豆部分更高浓度的异黄酮衍生物。例如,大豆酱中的异黄酮衍生物比剩余的大豆部分高出约30倍。参见K.S.Liu著:大豆化学、技术和利用,马利兰州,盖滋堡,Aspen出版社出版,1999。豆粉粗萃取物如大豆酱可以购自美国Schousten公司,如SoyLifel50和SoyLife25。或者,豆粉粗萃取物也可以购自美国ArcharDaniels Midland公司,如NOVASOY。
然后将此水和豆粉粗萃取物的混合物加热约60至100℃之间约10到120分钟后,且每100重量份的水中添加约2.0重量份的凝结剂而形成悬浮液,再加热至约30到100℃之间约1到120分钟。以过滤或离心来收集上清液,以得到增加了溶解度的水溶性豆类萃取物。另外,可添加酸或碱以调节悬浮液或上清液的Ph值,从而避免异黄酮衍生物沉淀。代表性地,Ph值调为约6.0。
所得上清液可通过脱色以脱去任何不希望的颜色。脱色方法包括但不限于镍Ni触媒的氢化和用于制造大豆油的漂白法。脱色法可参见K.S.Liu著:大豆化学、技术和利用,马利兰州,盖滋堡,Aspen出版社出版,1999,和P.J.Wan著:油脂技术,伊利诺州,原野,AOCS出版社出版。此上清液也可用包括但不限于用分子蒸馏或蒸发等公知的蒸馏系统来浓缩。分子蒸馏系统的一个实例为购自美国351N.Dekora Woods Blv.Saukville,WI53080,POPESCIENTIFIC,INC公司的Wiped-Film Still。此外,此上清液可用如冻干法或喷干法等任何公知的干燥方法干燥(UPS5,882,717)。
本发明的水溶性豆类萃取物可由分批法或流动法,即连续萃取和过滤法来制备。代表性地,用流动法以维持合理的制造成本。
为提高异黄酮苷的溶解度,于100重量份的水中添加约0.001到3.0重量份的凝结剂,如CaCl2,CaSO4,MgCl2,MgSO4,任何有机或无机酸(例如柠檬酸、富马酸、乳酸、苹果酸、酒石酸、H2SO4、H3PO4或HCl)、糖精、葡糖酸-D-内酯,和木瓜蛋白酶。制造此萃取物时,添加凝结剂通过单纯交连或改质来形成豆蛋白或多醣的不溶性聚集物。凝结剂的浓度直接影响萃取物中豆蛋白或多糖的量。例如,凝结剂量越多,导致形成更多的不溶聚集物。此效果允许控制萃取物中水溶性蛋白和多醣的总量和异黄酮衍生物的溶解度。添加凝结剂的量通过测定所得豆类萃取物的脂肪、蛋白质和无机酸含量来决定。一般来说,添加足量凝结剂来生成约1%以下的脂肪、约1到5%的蛋白质、约0.01到3%的Na、约0.01到3%的Ca和约0.01到3%的任何酸。水溶性豆类萃取物中诸如脂肪、蛋白质、Na、Ca、酸的量可由任何公知的分析法确定,例如遵从食物药品管理局(FDA)推荐的分析法。代表性地,于萃取物添加约0.3重量份的柠檬酸。一般来说,若凝结剂添加太多,水溶性豆蛋白质、多醣或异黄酮衍生物会从萃取物中共析出来,因而降低了萃取物中异黄酮衍生物的浓度或溶解度。而若凝结剂添加太少,则豆混合物形成胶状,因而难由此分离出水溶性豆类萃取物。
本发明的水溶性豆类萃取物可以干态或湿态加入食品。食品可为固体、膏状或液体,如牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤。
本发明的水溶性豆类萃取物也包括大麦、米、麦芽等其它谷物的共萃取物。例如,一种或多种的其它谷物与豆类可由上述方法制成本发明的水溶性豆类萃取物。此外,水溶性豆类萃取物可用电解质、香料、防腐剂和其它添加剂(例如维生素补充剂和麦芽糊精)来增强。防腐剂的实例包括但不限于抗坏血酸和五倍子酸丙脂,电解质的实例包括但不限于硫酸镁和氯化钾。
不用进一步的阐述,上面的描述已经充分公开了木发明。以下的举例将详述本发明,但本发明不限于此。所有的出版物,包括专利公报引用于此,以作参考。实施例1
用约90%乙醇水溶液萃取的豆粉粗萃取物1公斤中,如购自美围Schousten公司的SoyLife150中加入3公升70℃热水。于90℃搅拌30分钟和50℃搅拌30分钟之后,于50℃,10,000×g离心粗分离残滓和溶液。倾析后,于上清液中添加食品添加剂级的氯化钙6克(约0.02M),并于30℃继续搅拌30分钟后,上清液经1μm微滤器过滤,得到黄色溶液(2.5公升)。将此滤液移入5-L玻璃烧瓶中,在有催化量(约3克)的5%pd/c存在下,在10个大气压下氢化5小时。将此脱色后的混合物经0.2μm微滤器过滤,得到澄清的水溶性豆类萃取物溶液(2.3公升)。
此水溶性豆类萃取物溶液用高压液相色谱柱(HPLC)表征。HPLC是购自Waters公司的C18逆相柱150×4.6mm对称掩蔽RP18,依照梯度溶析法,于30℃下用15-40%乙腈水溶液和0.1%三氟乙酸以1ml/min溶出20分钟。HPLC测定器设定为254nm。图1表示实例1水溶性豆类萃取物中主成分的HPLC图。其各峰、滞留时间、峰面积和面积百分比都列入表1中。通过比较未知成分的滞留时间和于同一实验条件下HPLC分析的已知异黄酮苷的滞留时间来鉴定萃取物中的若干成分。
水溶性豆类萃取物溶液也测定萃取物成分的相对量来表征。萃取物的样品送至位于美国奥列冈州,波多兰市的食品实验公司并依FDA推荐的方法分析,结果含有<0.1%脂肪、5%蛋白质、65%碳水化物(多醣)、20%异黄酮、8%矿物质和1%水分。

用约90%乙醇水溶液萃取的豆粉粗萃取物(1公斤),如购自美国Schousten公司的SoyLife150中加入27公升70℃热水,于90℃搅拌15分钟后,注入90克柠檬酸水溶液3升。于90℃搅拌15分钟后,真空下经硅藻土床滤纸过滤。滤液为淡黄色澄清溶液(27公升)。加入碳酸钾(50克)调节Ph值至6.0。真空下经0.45μm穴滤器(毫孔公司)过滤除菌纯化后,于70℃,20mm Torr下用2英寸的Wiped-Film Still(HOPE科学公司)浓缩,得到淡黄色水溶性豆类萃取物溶液(18公升)。实施例3
用实施例1的方法制得水溶性豆类萃取物溶液2公升后,用Bertis公司制造的冻干机于-85℃,10mm Torr下冻干2日,得到350克淡黄色于饼。此干饼包括70克异黄酮衍生物,溶解度约200mg/ml。溶解度是通过首先将1克干燥萃取物于25℃的1ml纯水中搅拌5分钟,然后,观察混合物是否完全溶解,若未完全溶解,追加1ml水并于25℃搅拌5分钟。再观察是否完全溶解,若不,再追加1ml水且于25℃搅拌5分钟,如此至观察到完全溶解为止。其它实施例

Claims (37)
2.如权利要求1所述的水溶性豆类萃取物,其特征在于,所述豆类萃取物基于大豆、绿豆、黑豆、或云豆。
3.如权利要求1所述的水溶性豆类萃取物,其特征在于,所述豆类萃取物基于大豆。
4.如权利要求1所述的水溶性豆类萃取物,其特征在于,所述水溶性豆类萃取物在干态时包括10到20重量份的异黄酮衍生物,于25℃水中的溶解度为约30到300mg/ml。
5.如权利要求1所述的水溶性豆类萃取物,其特征在于,所述水溶性豆类萃取物在干态时包括10到20重量份的异黄酮衍生物,于25℃水中之溶解度为50-200mg/ml。
6.如权利要求1所述的水溶性豆类萃取物,其特征在于,所述异黄酮衍生物为大豆异黄酮苷(daidzin)、染料木苷(genistin)、黄豆苷(glycitin)、6″-O-乙酰基大豆异黄酮苷、6″-O-乙酰基染料木苷、6″-O-乙酰基黄豆苷、6″-O-丙二酰基大豆异黄酮苷、6″-O-丙二酰基染料木苷或6″-O-丙二酰基黄豆苷。
7.如权利要求6所述的水溶性豆类萃取物,其特征在于,所述豆类萃取物基于大豆。
8.如权利要求6所述的水溶性豆类萃取物,其特征在于,所述水溶性豆类萃取物在干态时包括10到20重量份的异黄酮衍生物,于25℃水中的溶解度为30到300mg/ml。
9.如权利要求6所述的水溶性豆类萃取物,其特征在于,所述水溶性豆类萃取物在干态时包括10到20重量份的异黄酮衍生物,于25℃水中的溶解度为50到200mg/ml。
10.一种水溶性豆类萃取物的制备方法,其包括:
将粉化粗制豆类萃取物与水混合;
加热此混合物;
添加凝结剂于此混合物中以形成悬浮液;
加热此悬浮液;及
收集上清液,得到水溶性豆类萃取物。
11.如权利要求10所述的制备方法,其特征在于,所述豆类萃取物基于大豆、绿豆、黑豆、或云豆。
12.如权利要求10所述的制备方法,其特征在于,所述豆类萃取物基于大豆。
13.如权利要求10所述的制备方法,其特征在于,约0.01到3重量份的所述凝结剂被添加至每100重量份的水中。
14.如权利要求13所述的制备方法,其特征在于,所述凝结剂为柠檬酸。
15.如权利要求10所述的制备方法,其特征在于,所述上清液是通过过滤悬浮液来收集的。
16.如权利要求10所述的制备方法,其进一步包括了将所述上清液干燥以得干燥的水溶性豆类萃取物。
17.一种水溶性豆类萃取物的制备方法,其包括:
将约1到50重量份的粉化粗制豆类萃取物与100重量份的水混合,
加热此混合物至约60到100℃之间,约10到120分钟;
添加凝结剂于此混合物中以形成悬浮液;
加热此悬浮液至约30到100℃之间,约1到120分钟;及
收集上清液,得到水溶性豆类萃取物。
18.如权利要求17所述的制备方法,其特征在于,所述豆类萃取物基于大豆、绿豆、黑豆、或云豆。
19.如权利要求18所述的制备方法,其特征在于,所述豆类萃取物基于大豆。
20.一种由权利要求10所述的方法制备的水溶性豆类萃取物。
21.一种由权利要求11所述的方法制备的水溶性豆类萃取物。
22.一种由权利要求12所述的方法制备的水溶性豆类萃取物。
23.一种由权利要求13所述的方法制备的水溶性豆类萃取物。
24.一种由权利要求14所述的方法制备的水溶性豆类萃取物。
25.一种由权利要求15所述的方法制备的水溶性豆类萃取物。
26.一种由权利要求16所述的方法制备的水溶性豆类萃取物。
27.一种由权利要求17所述的方法制备的水溶性豆类萃取物。
28.一种食物制品,其包括如权利要求1所述的水溶性豆类萃取物。
29.如权利要求28所述的食物制品,其特征在于,所述豆类萃取物基于大豆、绿豆、黑豆、或云豆。
30.如权利要求29所述的食物制品,其特征在于,所述豆类萃取物基于大豆。
31.一种食物制品,其包括权利要求9所述的水溶性豆类萃取物。
32.一种食物制品,其包括权利要求20所述的水溶性豆类萃取物。
33.如权利要求28所述的食物制品,其特征在于,所述食物制品为牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤。
34.如权利要求29所述的食物制品,其特征在于,所述食物制品为牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤。
35.如权利要求30所述的食物制品,其特征在于,所述食物制品为牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤。
36.如权利要求31所述的食物制品,其特征在于,所述食物制品为牛乳、茶、软饮料、果汁、咖啡、调味料、谷物、水、啤酒、脆饼、口香糖、巧克力或汤。
