Abstract

本发明通常涉及在植物材料中生产蛋白物质的领域。特别地,本发明涉及一种在藓类植物中生产所需的蛋白物质的新的方法。
用于生产目的的生物技术方法的开发为人类生产物质提供了重要的可能性,其中所述物质通过其它途径(例如通过化学合成)根本无法经济地制备并且在自然界中无法获得足够量的原材料。尽管超过一万种的植物次级代谢物是已知的,但是仅有极少数的所述化合物借助植物细胞培养的方式以工业化的规模加以生产。这些物质主要是具有药物活性的次级代谢物。可以提及的例子有a)黄连素(以4000升的规模生产),它具有抑菌和杀真菌的作用(Y.Fujita与M.Tabata,植物组织和细胞培养,植物科学(Plant tissue and cell culture,Plant Science);第3卷,第169页,C.E.Green等人(编辑)A.R.Liss Inc.纽约(1987));b)紫草宁(750升的规模),它具有抗菌和抗炎作用(M.Tabata与Y.Fujita,植物科学中的生物技(Biotechnology in plant science);第207-218页,P.Day等人(编辑),Aeademic Press,Orlando(1985)),以及c)paclitaxel(75000升的规模),它更为人知的名称为紫杉醇,具有抗肿瘤的作用(M.Jaziri等人,紫杉属细胞、组织和器官的培养作为生产紫杉醇类物质的另一种来源:文献调查,植物细胞组织器官培养(Plant Cell Tiss.Org.Cult.),46,第59-75页(1996))。
采用了植物细胞培养的另一种重要的生物技术方法为将毛地黄毒苷生物转化为一种心脏和循环药物-地高辛。在生物反应器中培养狭叶毛地黄以高产率成功地实现了这一立体特异性的水解反应(E.Reinhard与W.Kreis,Kultivierung von pflanzlichen Zellen im Bioreaktor[生物反应器中的植物细胞培养]Bio.Engin.,5,第135-136页(1989))。在H.-P.Muhlbach,生物技术中植物细胞培养的应用,生物技术年度综述(Biotechnol.Annu.Rev.),4,第113-176页(1998)中可以发现对生物技术中植物细胞培养应用的最新和最全面的论述。
在八十年代初期,高等植物的遗传转化方法的开发使得通过转化讨论中的代谢途径之特异性关键酶的基因来很大地提高植物生产特异性次级组分的生产率成为可能。不仅开发了转基因的完整植物、而且开发了植物细胞培养物。可以提到的例子为在烟草的转基因根毛培养物中细菌之赖氨酸脱羧酶的超表达,它使得生物胺、即尸胺和新烟碱的产率提高到了高达14倍(J.Berlin等人,植物次级代谢的遗传修饰:通过氨基酸脱羧酶的超表达改变产物的水平,植物生物学进展,植物科学研究(Advances in Plant Biology,Studies in Plant Science),第4卷,第57-81页,D.D.Y.Ryu与S.Furasaki(编辑)Elsevier,Amsterdam(1994))。
但是,将DNA转移到植物中的可能性不仅开辟了定量和定性地改变植物组分的途径;而且,植物和植物细胞的培养使得生产异源蛋白更为人们所感兴趣(A.S.Moffat,高技术植物预示着新产物作物的丰富,科学(Science)256,第770-771页(1992)),原则上可以选择两种不同的途径。
一个途径包括在转基因的完整植物中生产异源蛋白。除了在转基因烟草植物中生产抗体之外(J.K.-C.Ma等人,植物中分泌抗体的产生和组装,科学268,第716-719(1995)),已经描述了人血清白蛋白在转基因烟草植物和转基因马铃薯植物中的表达和校正过程(P.C.Sijmons等人,转基因植物中正确加工的人血清白蛋白的生产,生物/技术(Bio/Techn.),8,第217-221页(1990))。在转基因烟草植物中也表达出了人表皮生长因子(hEGF)(A.-H.Salmanian等人,在转基因马铃薯植物中人表皮生长因子基因的合成和表达,生物技术通讯(Biotechnol.Lett.),18,第1095-1098页(1996))。但是,也使用了其它的植物。采用鼠耳芥和欧洲油菜成功地生产出了亮氨酸-脑磷脂(E.Krebbers与J.Vandekerckhove,在植物种子中生产肽,Tibtech.,8,第1-3页(1990))。而且,使用了转基因的豇豆植物来表达起到疫苗作用的嵌合病毒颗粒(K.Dalsgaard等人,植物来源的疫苗保护目标动物抵御病毒疾病,Nat.Biotech.,15,第248-252页(1997))。
当采用如上所述举例的完整植物时,一个主要的缺点为必须培养它们,这既耗时又昂贵,而且需要很大的培养面积来进行工业化规模的生产。而且,从完整植物中分离所需的目标物质通常需要繁琐的处理步骤,尤其是当产物的一致性和质量必须满足较高的要求时,例如在某些情况下所述的物质用于药物或营养目的的时候。
在第二个途径中,开发了转基因烟草细胞的培养来生产抗体。例如,有所描述的是抗体的表达及其分泌至培养基中(N.S.Magnuson等人,从遗传修饰的烟草细胞悬浮培养物中分泌的哺乳动物蛋白回收的提高,Prot.Expr.Pur.,7,第220-228(1996))。由于从细胞中纯化异源蛋白是很复杂的,因此将目标蛋白分泌到培养基中标志着一个显著的进步。而且从安全的观点来看,在细胞培养物中生产与药物有关的重组蛋白也是有益的,这是因为转基因植物细胞能够仅仅在生物反应器中培养并且无需释放。通过开发生物反应器以便以大规模进行异源植物细胞的培养,已经使必须物质的培养成为可能(M.L.Shuler等人,作为能够使技术触及生物多样性的生物反应器工程:紫杉醇实例,Ann.N.Y.Acad.Sci.,745,第455-461(1994))。
其中采用了植物悬浮培养的第二种途径的主要缺点为生长速率低、次级代谢物的产生相对缓慢、在高细胞密度下产物形成的抑制,并且结果为单位体积的生产率低、聚集体和细胞壁组分的形成以及细胞对剪切力敏感度的增加。还必须考虑的是当采用异源细胞培养时必须总要提供具有多种组分的复合培养基,其中一些组分是昂贵的。但是,需提到的最严重的缺点为在植物的体外细胞培养中体细胞克隆变异的发生,它导致在异源蛋白的生产中定量和定性的改变(参见例如M.G.K.Jones与K.Lindsey,植物生物技术,分子生物学和生物技术,J.M.Walker与E.W.Gingold(编辑),第二版,Royal Soc.of Chem.,Burlington House,伦敦(1988))。所生成产物及其功能特性的不均一性是令人无法接受的,尤其与生产那些官方批准需要可靠的质量保障和标准化生产方法的药物和其它所需物质有关的情况更是如此。
因此,本发明的目的在于提供一种在植物材料中标准化地生产异源蛋白物质的方法,该方法基本上消除了上述采用完整植物的缺点,而且还消除了采用细胞培养系统的缺点。
根据本发明,通过提供一种新的方法来实现这一目的,该方法用于在植物材料中生产异源蛋白物质,其中在标准的条件下培养充分分化的藓类植物,并且基本上无需破碎产生的组织或细胞就可以从培养基中获得所产生的蛋白物质。
本文中使用的术语“蛋白物质”包括特别适合于诊断、临床、药物和营养目的的肽、多肽、蛋白以及所述物质的片段。还包括有具有肽键以及由植物材料翻译的那些分子。
在本发明一个优选的实施方案中,所需的异源蛋白物质以其生物活性形式被释放到培养基中。
本申请说明书中使用的术语“生物活性”是指提供有这种特性的目标物质具有希望的或者需要的用于相应目的的功能特性。例如如果希望生产抗体的话,那么当所产生的蛋白或其功能性片段能够与抗原建立起预期的特异性结合时,所述的物质便是具有生物活性的。对于本领域的普通技术人员来说,显而易见的是针对此目的并不总是需要完全蛋白,而是有可能去寻找能够确保所需生物活性或功能的抗原决定基或低分子量的结构。例如,当一种酶能够转化其目标底物时,它便是有生物活性的。
在本发明另一个优选的实施方案中,在基本上不含糖、维生素和植物激素或其功能性片段的培养基中以完整藓类植物的形式培养植物材料。
本发明的方法使得可能可以在能够标准化的光自养条件下培养完整的和充分分化的植物,即无需添加现有技术中异养悬浮细胞培养系统所需的糖、维生素和植物激素等。因为使用了廉价且简单的培养基,所以可以在很大程度上加速获得和纯化所需目标物质的步骤。
在本发明的方法中采用的植物材料优选为完整的藓类植物,所述植物选自包括苔类植物在内的藓类植物的小组,它们属于剑叶藓属、葫芦藓属、泥炭藓属和角齿藓属的种,其中特别优选采用地钱属和Sphaerocarpos。最优选使用展叶剑叶藓来实现本发明的方法。
在另一个优选的实施方案中,用于转化的核酸构建体不仅编码所需的蛋白物质,而且编码用于将所述物质从宿主细胞分泌到培养基中的转运肽。在本发明中可以使用本领域普通技术人员公知的任何自体和异源的核酸序列并且可以用来产生用于转化生产组织的表达盒。特别优选使用内质网或细胞转运的信号肽。
实施本发明的工作表明在细胞培养中遇到的上述体细胞克隆变异的问题在藓类植物的光自养液体培养中并不存在。而且,在本发明中使用的藓类植物具有优于其它准确限定的分化步骤之确切序列的系统(chloronema、茎原丝、芽、配子托)的优点,而这些系统可以通过添加植物激素(吲哚-3-乙酸诱导茎原丝的发育,异戊烯基腺嘌呤诱导芽的发育)来影响(例如参见N.W.Ashton等人,使用生长素和细胞分裂素抗性突变体分析展叶剑叶藓中配子体的发育,Planta 144,第427-435页(1979))。于是使得可能在生物反应器的培养过程中进行异源蛋白定向的分化特异性表达,根据本发明,由于在生物反应器中可以控制的统一的蛋白生产及其适合于使用激素依赖型或分化特异性启动子,因此同步分裂、纯的并因而均一的chloronema的培养是特别优选适合的。
除了这类表达系统之外,根据本发明还可以使用诱导型启动子系统,尤其对于生产具有短的半衰期或具有细胞毒性的蛋白更是如此,特别优选使用根癌土壤杆菌1’-启动子。
例如按体积数量级为20毫升至6升以上、乃至高达10升的规模,通过在摇瓶培养或者通气的玻璃容器中使用剑叶藓属的植物可以实现本发明所提出的藓类植物的培养,该培养从经济的角度考虑可以用于生产异源蛋白(参见例如R.Reski,Zell-und molekularbiologischeUntersuchungen der cytokinin-induzierbaren Gewebedifferenzierungund Chloroplastenteilung bei Physcomitrella patens(Hedw.)B.S.G.,[展叶剑叶藓中细胞分裂素诱导的组织分化和叶绿体分裂的细胞和分子生物学的研究(Hedw.)B.S.G.],博士论文,汉堡大学(1990))。由于这属于分化的光自养植物的培养,因此培养基既不需要补充植物激素、又不需要补充维生素或糖。与例如用于动物细胞培养所需的复合培养基相比,成本要低100倍。根据本发明所呈现出的情况是在存在PVP的情况下在培养基中具有生物活性的异源蛋白的产率可以提高至35倍,这也正是为何在本发明的方法中优选在培养基中使用PVP。
在现有技术中描述了在生物反应器中进一步培养在本发明中合适的藓类植物的详细信息,所述的藓类植物例如为薄囊藓和中位泥炭藓(参见例如E.Wilbert,Biotechnologische Studien zurMassenkultur von Moosen unter besonderer Berücksichtigung desArachidonsurestoffwechsels[在特别考虑到花生四烯酸代谢的情况下有关藓类植物大量培养的生物技术的研究]博士论文,Mainz大学(1991);H.Rudolph与S.Rasmussen,关于在生物反应器中培养的泥炭藓属的次级代谢的研究,Crypt.Bot.,3,第67-73页(1992))。针对本发明特别优选使用剑叶藓属的植物,而这是由于针对这种生物体已经确立了所有常规的分子生物学技术(为了回顾参见R.Reski,藓类植物的发育、遗传学和分子生物学,Bot.Acta.,111,第1-15页(1998))。
为了生产异源蛋白,开发了合适的转化系统以便通过生物技术使用剑叶藓属的植物。例如,通过使用基因枪将DNA直接转移到原丝体中便可以实现成功的转化。通过PEG-介导的将DNA转移到藓类植物原生质体中也是成功的。这种针对剑叶藓属植物的转化方法已经重复地加以描述并且导致生成了瞬间的和稳定的转化体(例如参见K.Reutter与R.Reski,在生物反应器中完全分化的藓类植物的培养中异源蛋白的生产,植物组织培养与生物技术(Pl.Tissue culture,@Biotech.),2,第142-147页(1996))。
尽管本发明基本上适合于生产任何的蛋白物质,但在下文中将会以人血管内皮生长因子(VEGF)为例说明与药物有关的蛋白的生产。
VEGF首先是由N.Ferrara和W.J.Henzel分离出来的(垂体滤泡细胞分泌一种对血管内皮细胞具有特异性的新的肝素-结合生长因子,Biochem.Biophy.Res.Commun.,161,第851-858页(1989))并且其特征在于在正常的生理条件下作为受控的血管发生和内皮细胞分裂的调节因子(N,Ferrare等人,多肽血管内皮生长因子家族,细胞生物化学杂志(J.Cell.Biochem.),47,第211-218页(1991))。作者同时还说明该生长因子对血管内皮细胞具有非常高的特异性并且对其它的细胞类型没有活性。VEGF是一种由二硫键连接的均一二聚糖蛋白。已知四种不同形式的人VEGF。四种同种型为长度为121,165,189和206氨基酸并且它们是由VEGF RNA不同的拼接形成的。VEGF206仅在胎儿肝脏cDNA中得到了印证,而VEGF121、VEGF165和VEGF189的转录则在许多肿瘤细胞和肿瘤组织中得到了印证。所有VEGF的同种型都具有用于分泌的前导序列,但是仅仅有效地分泌出两种最小的形式(参见例如G.Martiny-Baron与D.Marmé,VEGF介导的肿瘤的血管发生:癌症疗法的新目标,Curr.Opin.Biotechnol.,6,第675-680页(1995))。
为了开发和改进现存的肿瘤治疗的方法,同时也是为了表征VEGF,在过去和目前都仍然需要适量的VEGF。在实践本发明工作的早些时间,所描述的都是通过杆状病毒表达系统在昆虫细胞中重组生产VEGF(例如B.L.Fiebich等人,在昆虫细胞中合成和组装具有功能活性的人血管内皮生长因子均二聚体,欧洲生物化学杂志(Eur.J.Biochem.),211,第19-26页(1993))。酿酒酵母(S.Kondo等人,由酿酒酵母产生的人血管内皮生长因子/血管通透因子最短的同种型(VEGF/VPF121)既促进了血管发生,又促进了血管的通透性,Biochim.Biophys.Acta,1243,第195-202页(1995))、巴斯德毕赤氏酵母(D.Nohanraj等人,在酵母中具有生物活性的人血管内皮生长因子的表达,生长因子(Growthfactors),12,第17-27页(1995))以及大肠杆菌(G.Siemeister等人,大肠杆菌中肿瘤血管发生因子VEGF的具有生物活性同种型的表达,Biochem.Biophys.Res.Commun.,222,第249-255页(1996))紧接着作为进一步用于生产的生物体。通过所有这些重组系统生产出了具有生物活性的VEGF。但是,就蛋白的纯化和重构而言大肠杆菌的表达系统是很复杂的,这是因为后者要被包装进入到内含体中。
实施例
概述
可控的展叶剑叶藓大量培养的确立(Reutter与Reski,loc.cit.)以及将DNA转移到展叶剑叶藓的方法(K.Reutter,在展叶剑叶藓中异源基因的表达(Hedw.)B.S.G.[在展叶剑叶藓中异源基因的表达(Hedw.)B.S.G.],博士论文,汉堡大学(1994))为通过生物技术利用这种植物创造了基本的先决条件。
在初期进行的工作表明使用来源于Reutter(loc.cit.,1994)的转基因展叶剑叶藓的整合具有长期的稳定性。举例来说,针对此目的而使用的异源npt II和gus基因的表达在四年后仍然可以检测得到。
剑叶藓属植物在生物反应器中的培养得到了优化。开发了一种搅拌器使得protonemata粉碎,并从而确保了在300-500rpm的连续速度下培养物达到所需的均匀性。因此使得标准化的取样成为可能。与此同时,进入的空气被更均匀地分散到液体培养基中。在这些条件下证明了在没有外部pH调节下,生物质和蛋白的形成以与采用pH调节相同的方式进行;因此令人吃惊的是后者并不是必须的。在半分批的条件下,达到了一周生产的生物质为每升500毫克干重、或22毫克的总蛋白。这意味着生物质的生产比起常规5升玻璃烧瓶的培养增加到了5倍。将Knop培养基的盐浓度减少到十分之一获得了类似的数据并因而降低了成本。
通过减少延滞期,5mM酒石酸铵的加入加速了生物质的形成。与此同时,酒石酸铵的加入使得培养物中实质上仅仅包含chloronema细胞。借助流式细胞仪证明了实质上这些培养物的百分之百的细胞都处于细胞周期的G2/M期。通过进一步采用生长素进行的生理研究以及通过采用分化特异性突变体cal112和cal113进行的研究证实了这一结果,可以断定茎原丝细胞在大部分时间里处于G1/G0期,而chloronema细胞则主要处于G2/M期。
通过使用土壤杆菌1’-启动子,研究了藓类植物中启动子可能的诱导性。β-葡糖醛酸糖苷酶(gus)基因被用作标记基因。在瞬间转化的藓类植物原生质体中(转化率=3×10-4),随着采用5μM吲哚-3-乙酸的诱导,观察到了gus基因的表达。在任何对照组中都没有观察到表达。
把人血管内皮生长因子之121个氨基酸拼接形式(VEGF121)的基因以0.5×10-5和3.3×10-6的转化率转移到藓类植物中。为此,将该基因克隆到组成型35S启动子的后面并继而克隆到适合于植物的转化载体pRT99中。在第二种方法中,额外地克隆了编码相应的人ER转运肽的序列。通过使所得的稳定转化体进行Southern分析,证实了异源DNA的整合以及描述了整合的类型。Northern分析证明了在这些转化体中存在着nptII和两种VEGF转录物。通过间接的免疫荧光测定证实了VEGF121在藓类植物细胞中的表达。借助同焦激光扫描显微镜证实了蛋白明确地位于细胞之中。对于不含转运肽的转化体而言,这些研究表明蛋白特别位于细胞质中。在另外还含有ER转运肽的转化体中,在核区以及顶端细胞的顶端区域(该区域具有非常高含量的ER)可以发现所述的蛋白。通过对从培养基中得到的VEGF蛋白进行ELISA测定和两种功能测定证实了在本发明中产生的异源蛋白的生物活性。
材料和方法
除了在本文中另作具体描述的以外,所使用的化学物质质量均是分析级的并且从Fluka(Neu-Ulm)、Merck(Darmstadt)、Roth(Karlsruhe)、Serva(Heidelberg)和Sigma(Deisenhofen)。
溶液均是用纯化的、不含热原的水制备的,这种水在下文中称作为H2O,它来自Milli-Q水纯化系统(Millipore,Eschborn)。
限制性内切核酸酶、DNA-修饰酶以及分子生物学试剂盒从AGS(Heidelberg)、Amersham(Braunschweig)、Applied Biosystems(Weiterstadt)、Biometra(Gttingen)、Boehringer Mannheim GmbH(Mannheim)、Genomed(Bad Oeynhausen)、New England Biolabs(Schwalbach/Taunus)、Novagen(Madison,威斯康星,美国)、Pharmacia(Freiburg)、Qiagen(Hilden)和Stratagene(Heidelberg)获得。除非另作具体说明的以外,根据生产说明书来使用这些物质。
载体和构建体
质粒pCYTEXP-VEGF121是pCYTEXPl(T.N.Belev等人,用于优化大肠杆菌中的基因表达的完全模块载体系统,质粒(Plasmid),26,第147-150页(1991))的衍生物,其中为了在大肠杆菌中表达,整合了人VEGF121的cDNA。使用限制性内切核酸酶Nde I和Sal I从pCYTEXP-VEGF121切下VEGF121cDNA,将其纯化、使其成为平端并克隆到介于CaMV的35S启动子和聚腺苷酸化序列之间的pRT101的SmaI切割位点处(R.Tpfer等人,用于转录和翻译的融合的一套植物表达载体,核酸研究(NucleicAcids Res.),15,第5890页(1987))。使用Hin dIII,再次切除如此得到的盒并将其克隆到转化载体pRT99的Hin dIII限制切割位点。在CaMV的35S启动子和相应的聚腺苷酸化序列的调节下,pRT99不仅含有多克隆位点,而且还含有新霉素磷酸转移酶基因(R.Tpfer等人,植物细胞中用于瞬时基因表达和直接基因转移的多用途克隆载体,核酸研究(Nucleic Acids Res.),16,第8725页(1988))。在稳定转化的植物中,该基因赋予了对抗生素G418的抗性。将该质粒复制到大肠杆菌菌株DH5α中(J.Sambrook等人,分子克隆:实验手册,冷泉港实验室出版社(Cold Spring Harbor Laboratory Press),纽约(1989))。
使用限制酶BamHI和BglII,将cDNA从载体pVE-121中切下,最初构建所述载体以用于在昆虫细胞中表达VEGF121并且除了VEGF121序列之外该载体还包含编码天然转运肽的DNA,其中在动物细胞系统中,该转运肽通过内质网介导分泌到培养基中(Fiebich等人,在昆虫细胞中具有功能活性的人血管内皮生长因子均二聚体的合成和装配,欧洲生物化学杂志(Eur.J.Biochem.),211,第19-26页(1993)),并且使用pRT101将cDNA克隆到pRT99中并加以验证。
质粒pNA201是二元载体pBI101的衍生物(A.R.Jefferson等人,测定植物中的嵌合基因:GUS基因融合系统,Plant Mol.Rep.,5,第387-405页(1987))。在胭脂氨酸合酶启动子之下,它含有作为植物选择标记的nptII基因。同样存在的gus基因受到来自根癌土壤杆菌的1’-启动子的调节。pNA101适合于直接转化展叶剑叶藓。
抗体
使用了抗VEGF蛋白的两种不同抗体。第一抗体为兔-抗-VEGF抗体并且对合成肽具有抗性,所述的合成肽对应于天然的人VEGF之第1至第20个氨基酸(Fiebich等人,loc.cit.(1993))。第二抗体为抗人VEGF121蛋白的单克隆小鼠抗体(R&D Systems,Wiesbaden)。
植物材料
使用了展叶剑叶藓的野生型株系(Hedw.)B.S.G.,该株系来源于汉堡大学普通植物学系遗传学小组的收集品。其来源为16/14株系,该株系已经由Huntingdonshire(英格兰)Gransden Wood的H.L.K.Whitehouse收集并且已经从孢子进行了传代培养。
在含有Knop培养基(R.Reski与W.O.Abel,采用异戊烯基腺嘌呤诱导展叶剑叶藓的chloronemata和茎原丝的发芽,Planta,165,第354-358页(1985))的液体培养物中或者在含有1%oxoid琼脂(Unipath,Basingstoke,英格兰)的固体Knop培养基上培育野生型株系。如Reski所述(loc.cit.,1990)进行液体培养。
生物反应器中的培养
对于大量培养来说,将植物材料引入到一个装配有双层夹套的7升圆底玻璃烧瓶生物反应器(Applikon Biotek,Knüllwald)中。在该生物反应器系统中,将废气冷凝器、通气管、pH电极(Conducta,Gerlingen)、温度传感器、搅拌装置、取样管以及酸、碱和培养基的入口通过盖子的孔洞从上部引入到培养容器中。在25℃下进行培养,并且通过与双层夹套相连的适当调节的水浴来控制。在通过pH调节进行的实验中,通过滴定装置保持pH恒定在5.8。通过生物控制器ADI 1030(Applikon Biotek,Knüllwald)实现温度的测量和pH的调节。通过电机控制器ADI 1012(Applikon Biotek,Knüllwald)来改变搅拌器的速度。通过多孔喷射部件用1巴的空气给培养基恒定地通气。为了确保培养容器无菌,所有的入口和排出管线都配备有过滤灭菌装置(Midisart,0.2μm;Sartorius,Gttingen)。生物反应器中的培养是在具有侧面光照(白光;Osram L40W/20;最大100μmols-1m-2)的环境受控的小室中进行的。在无菌条件下,以生物反应器中每升培养物0.5-1克湿重的植物材料接种培养物。取样管与搅拌器处于同一水平,从而确保了在搅拌时均匀地取样。采用具有路厄氏锁式接头的无菌注射器对小量样品(<100毫升)取样,而大量样品的取出例如使用配备有501 RI头的302/3A型蠕动泵(Sartorius,Gttingen)。
通过用5mM酒石酸铵补充Knop培养基来生产野生型的chloronema的培养物。
当培养基中存在灭过菌的聚乙烯吡咯烷酮(PVP)时,可以显著地增加所分泌的具生物活性的异源蛋白的产率。
通过外源添加生理量的生长素(合适的浓度例如为5μmol/l的吲哚-3-乙酸(IAA)),可以诱导并提高在原丝体发育过程中发生的茎原丝的分化。
为了在选择压力下进行转化体的液体培养,将50毫克/升的抗生素G418(Calbiochem,Bad Soden)添加到Knop培养基中。为此,每隔十天在粉碎之前立即通过在无菌的100μm筛网(Wilson Sieves,Nottingham,英格兰)过滤来取出培养物,并将其转移到装有选择培养基的锥形瓶中。
为了进行营养元素实验,用水以1∶10的比例稀释Knop培养基。
转化
所选择的转化方法是如Reutter和Reski所述的PEG-介导的直接将DNA转移到原生质体中的方法(loc.cit.,1996)。在每次转化中,针对3×105的原生质体使用了50μg的质粒DNA。除非另作具体说明的以外,如Reutter和Reski所述(loc.cit.,1996)进行原生质体的再生和对稳定转化体的选择。
间接免疫荧光测定
缓冲液:MSB:100mM PIPES;5-10mM EGTA,5mM MgSO4,pH6.8
F-MSB:MSB+5%DMSO
E-MSB:MSB+5%DMSO+5%Nonidet
W-MSB:用水以1∶2的比例稀释的MSB(洗涤缓冲液)
酶溶液:1%纤维素酶、1%果胶酶、2%Driselase的MSB溶液,pH5.6(所有均来自Deisenhofen的Sigma公司)
通过培养不超过10分钟的时间,将藓类植物的protonemata固定在1.25%戊二醛的F-MSB(v/v)溶液中,并且在W-MSB中简短地洗涤一下。然后,用2%低聚甲醛的MSB溶液(v/v)培养protonemata 40分钟,并用W-MSB洗涤3次(漂洗1次,洗涤2次5分钟)。
通过加入MSB和一刮刀尖的固体硼烷并且培养10分钟,降低了通过洗涤无法除去的游离醛。通过用W-MSB洗涤三次来除去硼烷。
在接下来的步骤中,通过加入酶溶液处理10分钟使得细胞壁可以透过。通过改变pH(MSB,pH6.8)来终止酶促反应。再次用W-MSB洗涤混合物3次。
通过用洗涤剂溶液培养120分钟的一段时间来提取叶绿素。然后通过用W-MSB洗涤三次来除去溶液。
在上述制备之后,可以用初级抗体(抗-VEGF;稀释比例为1/50)培养藓类植物的protonemata。这需要在37℃下处理45分钟。在用W-MSB洗涤三次后,在37℃下加入标记的次级抗体(抗-兔或抗-小鼠;稀释比例为1/30;用异硫氰酸盐荧光素(FITC)(Molecular Probes,Leiden,荷兰)标记)处理45分钟。除了进行如上的三次洗涤步骤之外,用W-MSB+0.1%Triton洗涤混合物一次。接下来将protonemata提取到W-MSB中并且在4℃下至少保存过夜。
借助TCS 4D型同焦激光扫描显微镜(CLSM)(Leica Lasertechnik,Heidelberg)和Scanware5.0软件(Leica lasertechnik,Heidelberg)来评价所述的物质。
为了通过CLSM分析样品,将它们放置到载玻片上的封固溶液(Dabco,Sigma,Deisenhofen)中。借助氩-氪激光仪,在488nm的波长下进行偶联到次级抗体上的荧光染料FITC的激发。FITC发出波长为528nm的光。
ELISA测定
通过采用ELISA测定这样的常规方法以及上述的抗体来定性和定量地测定培养基中的VEGF蛋白,其中所述的蛋白是在适当转化的藓类植物中形成的。在ELISA测定中直接使用了200μl的培养基。
功能测定
通过促有丝分裂测定(Miyazono等人,来自人血小板的内皮细胞生长因子的纯化和特性,生物化学杂志(J.Biol.Chem.),262,第4098-4103页(1987))和“13-天尿囊绒膜血管发生测定”(Wilting等人,兔子角膜测定的形态学研究,Anat.Embryol.,183,第1167-1174页(1991))来检测从培养基中获得的重组生成的VEGF之生物活性。事先使培养基进行超滤,然后进行冷冻干燥并随后将其重新悬浮在缓冲液中。如果需要的话,可以采用阳离子柱进行进一步纯化的步骤。
1’-启动子的诱导
用5μmol/l的吲哚-3-乙酸(IAA)测定由生长素对1’-启动子的诱导性。在转化后的第五天,将与pNA201进行了转化反应的100μl原生质体的等分试样转移到96-孔微滴板(Nunc,Wiesbaden)的孔洞中。采用IAA(最终浓度为5μM)使原生质体悬浮液培养5小时。在培养过后,通过β-葡糖醛酸糖苷酶的定量测定直接评价诱导实验。
β-葡糖醛酸糖苷酶的定性测定
通过定量测定来确定β-葡糖醛酸糖苷酶的活性(A.R.Jefferson,测定植物中的嵌合基因:GUS基因融合系统,Plant Mol.Rep.,5,第387-405页(1987))。

在37℃下,将处于Knop或者再生培养基中的藓类植物的protonemata以及藓类植物的原生质体在等体积染色溶液中培养达72小时,之后用显微镜立即进行评价。
结果
生物反应器中培养物的均匀性;取样
只有均匀的培养才能确保从细胞培养物中的取样是标准的。在延长的培养中,原丝体生长成为长的细胞丝状体会频繁地导致细胞聚集在一起,并因而导致在液体培养物中植物材料分布得不均匀。为了避免这种聚集,在一定的时间间隔内粉碎这些protonemata:通过使用高速搅拌器/匀浆器,在生物反应器中自第10天开始每隔1天就进行粉碎,而在摇瓶培养中每12天就进行粉碎。为了即便在延长的培养时段的情况下仍然可能在生物反应器中保持连续的条件并且与此同时使取样可以标准化,可取的是通过研磨搅拌桨的边缘、从而将它们转变成为剪切桨叶来改进具有三个搅拌桨的涡轮式搅拌器。因此,在300-500rpm下的恒定的“搅拌”使得在生物反应器中进行均匀的培养成为可能。
在采用涡轮式搅拌器的对照培养中于500rpm下35天(840小时)所形成的生物质(以DW[毫克/升]为单位)与采用剪切桨式搅拌器搅拌所进行的生物反应器的培养是相同的。
通过比较每种情形下的六个平行样品来评价培养的均匀性。确定来自100毫升体积样品的植物材料的干重作为比较参数。当使用未经改进的涡轮式搅拌器时,随着生物质浓度的增加,标准偏差也增加。由于采用剪切桨式搅拌器进行搅拌,产生的样品标准偏差保持在很低的水平。这就使得可以断定在35天的培养中采用经改进的搅拌器可以获得均匀的培养物。
对1’-启动子诱导性的研究
在质粒pNA201中,1’-启动子位于gus基因的上游并且起到了控制元件的作用。公知的β-葡糖醛酸糖苷酶测定适合于作为藓类植物的诱导实验的检测方法。采用转基因烟草的实验表明1’-启动子导致在高生长素含量下β-葡糖醛酸糖苷酶在组织中的表达,并因而假定该启动子是依赖于生长素的。在瞬时转化的原生质体中研究了展叶剑叶藓中生长素对1’-启动子的诱导性。
在5μM吲哚-3-乙酸(IAA)存在下培养(5小时)以及不培养的情况下使转化反应进行β-葡糖醛酸糖苷酶的测定。在不添加IAA的对照组中,显微镜下的评价揭示出在任何反应中均不存在蓝色的原生质体。与此相反,用生长素培养的原生质体的评价证实了gus基因的表达。在蓝色原生质体的基础上,实现了3×10-4的转化率。这清楚地揭示出在瞬时转化的藓类植物原生质体中,1’-启动子受到了植物激素-生长素的诱导。
用于VEGF转化的载体的产生
为了将不含前导序列的VEGF121的cDNA(在下文中称作VEGFC)以及含前导序列的VEGF121的cDNA(在下文中称作VEGFP)转化到剑叶藓属植物中,必须在启动子和终止子单元之间克隆适合于植物的序列。针对这一目的,选择了35S CaMV启动子和相应的聚腺苷酸化信号。将适当制得的VEGF cDNA序列克隆到载体pRT101多克隆位点的Sma I限制切割位点处。
利用来源于35S启动子的终止区的引物来测定所得载体(pRT101VEGFC3和VEGFP21)的序列,并且验证了位于启动子和聚腺苷酸化位点之间整合的正确性。
用限制酶Hin dIII切下得到的盒并将其克隆到实际的转化载体pRT99的Hin dIII切割位点处(pRT99VEGFC3和VEGFP21)。通过用SmaI和Hinc II的限制酶切分析来确定相对于NPTII盒的上述盒的取向。在研究启动子-聚腺苷酸化信号取向的过程中,得到了5250bp(VEGFC)和5380bp(VEGFP)的片段,与此同时逆取向提供了1100(VEGFC)/1230(VEGFP)以及4150bp(VEGFC和P)的片段。限制酶切分析揭示出相对于pRT99的npt II基因的启动子-聚腺苷酸化信号的取向已经仅仅掺入了5250/5380bp的片段以及VEGFC/P盒。
VEGFC转化到剑叶藓属植物中
在反复地从Knop培养基转换到选择培养基之后,野生型原生质体中VEGFC构建体转化的绝对转化率为0.5×10-5,所述的转化体具有恒定的稳定性。
对转化质粒整合的证明
通过Southern杂交证明了向植物基因组中成功的整合。
所用的探针首先是来自pRT99的npt II基因的Nco I片段,其次是来自pCYTEXP-VEGF121的Nde I/Sal I VEGFC片段。
在转化体未切割的总DNA中检测到的信号证实了质粒DNA向植物基因组中成功的整合。甚至于在通过用Hin dIII进行的限制性分析检测了整合之后,仍然得到了所有的35S VEGFC PolyA盒。如果在整合过程中所述的盒仍保持完整的话,那么这种限制酶从总DNA中切下了1100bp的片段。
采用VEGFC探针的杂交形式揭示出对所有转化体而言都具有1100bp的片段。从而已经证实了完整的VEGFC表达单元的整合,而这正是VEGF121在藓类植物中正确转录和表达的先决条件。
异源基因转录的证明
使用VEGF和NPT II探针,通过非放射性DIG检测系统来检测转化体中异源基因VEGFC和NPTII的转录。采用编码VEGFC转录物的760个核苷酸以及编码NPT II转录物的1100个核苷酸,在荧光图谱中检测到的转录物的大小都在针对每种情况所预期的范围之内。正如所预料的那样,在WT对照组中检测不到这两种异源转录物。
采用人的转运肽分析转化体
在每个转化反应中由PEG介导的50μg pRT99P 21质粒DNA的DNA转移产生了在选择培养基中永久稳定的转化体。稳定的转化率达到了3.3×10-6。
转化质粒整合的证明
如上所述通过采用所述探针进行的Southern杂交证实了上述具有转运肽的转化体的整合,并且经Hind III切割的总DNA与VEGF探针的杂交揭示出一个1230bp的片段:这证实了整合的35S VEGFP PolyA盒的完整性。
异源基因转录的证明
通过以上所列举的方法可以检测NPTII和VEGFP转录物两者。采用同焦激光扫描显微镜检测转基因藓类植物细胞中的人VEGF121
在该方法中,将测试蛋白直接标记在封固的细胞中。在同焦激光扫描显微镜下进行评价,其中该显微镜与普通光学显微镜相比分辨能力有所提高。
重组的VEGF121蛋白-如果在藓类植物细胞中可以检测到的话-应该在VEGFC转化体的细胞质中积累。在VEGFP转化体中,如果转运肽在藓类植物中发挥信号作用的话,那么它应当在ER系统中可以被检测到。
采用间接免疫荧光方法以及借助同焦激光扫描显微镜的计算机辅助的评价,人VEGF121在转基因藓类植物细胞中的表达已经得到了成功的证实。此外,据证实在转基因的藓类植物中,在存在相应的人转运肽的情况下,VEGF121被成功地转运到内质网中。
有关培养基中存在VEGF的测定
通过ELISA测定在PVP存在的情况下200μl的培养基的等分试样,结果表明用包含转运肽编码序列的表达盒转化的藓类植物能够将VEGF释放到培养基中。阳性的结果使得可以断定存在着具有功能的VEGF蛋白。
生物活性的测定
用于验证释放到培养基中的VEGF蛋白生物活性的两种测定均给出了阳性的结果,并且证实了可以从培养基中获得本发明所产生的VEGF,而其具有所需的生物活性。
Claims (6)
1.一种用于在植物材料中生产异源蛋白物质的方法,其特征在于将原丝体藓类植物组织用作植物材料,并且所产生的蛋白物质是在不破坏生产组织或细胞的情况下从培养基中获得的。
2.根据权利要求1所述的方法,其特征在于所述的释放到培养基中的蛋白物质是具有生物活性的。
3.根据权利要求1或2所述的方法,其特征在于使用培养基,所述的培养基不含有糖、维生素和植物激素或其功能性片段。
4.根据权利要求1至3之任一的方法,其特征在于所述的藓类植物组织选自包括苔类植物在内的藓类植物。
5.根据权利要求4所述的方法,其特征在于所述的藓类植物组织选自由剑叶藓属、葫芦藓属、泥炭藓属和角齿藓属组成的组的藓类植物。
6.根据权利要求4所述的方法,其特征在于所述的藓类植物组织选自由地钱属和Sphaerocarpos组成的组的苔类植物。
