s2, carrying out sand pre-storage treatment on seeds in September, cleaning the seeds, sun-drying the seeds for later use, soaking the seeds in warm water for 24 hours, lightly rubbing the seeds by hands until clear water is obtained, mixing the seeds into cleaned sand, wherein the proportion of sand to the seeds is 20:1 (V/V), adding water mixed with carbendazim, specifically, the water mixed with carbendazim is obtained by mixing 1g of 25% carbendazim wettable powder with 250g of water, stirring uniformly, and taking the sand with dryness and humidity approximately equal to that the sand is not scattered after loosening by hand;
s3, bagging seeds, placing the seeds in a refrigerated cabinet for storage, wherein the initial temperature of Sha Cangqi is 15 ℃, the temperature is reduced to 13 ℃ after two weeks, the temperature is reduced to 11 ℃ after four weeks, the temperature is reduced to 9 ℃ after six weeks, the temperature is reduced to 7 ℃ after eight weeks, the temperature is reduced to 4 ℃ after ten weeks, and the temperature is kept after the temperature is reduced to 4 ℃ and no more reduction is performed;

Example 4
The embodiment provides a method for culturing epimedium seeds, which comprises the following steps: the epimedium seeds treated by the storage methods of the example 1, the comparative example and the comparative example 2 are sown on a sand bed, the humidity of the sand bed is kept to 65 percent, and the illumination is carried out for 16 h.d -1 。
Three groups of seeds were treated with 100 seeds according to the culture method, each group was provided with 3 repeated groups, and 900 seeds were treated in total. The sand bed sowing day is calculated as 0 day, seeds start to germinate from the 10 th day, the seeds start to germinate stably from the 20 th day after sowing, the seed germination rate is counted from the 20 th day, and the calculation formula of the seed germination is as follows:

As shown in the figure 1, the germination rate of the seeds of the non-sand control group is 0 after the seeds are stored at a high temperature of 25 ℃ for 90 days; after sand storage at a constant temperature of 4 ℃ for 90%, the germination rate of seeds is 52%; the initial temperature of sand collection is 15 ℃,2 ℃ is regulated down every two weeks, the temperature is reduced and laminated step by step, the temperature is kept to 4 ℃, the total storage is carried out for 90 days, and the germination rate of seeds is 80%; the above results indicate that: the seeds are stored for 3 months at the constant temperature of 25 ℃, the initial temperature is 15 ℃, the germination rate of the seeds is highest after the step-by-step cooling lamination treatment for 90 days, the germination rate of the seeds for 20 days is up to more than 80%, and the seeds have obvious difference (P is less than 0.05) compared with the sand storage treatment at the constant temperature of 4 ℃.
Example 5
The embodiment provides a planting method of epimedium seeds, which comprises the following steps: applying 1.5 tons of organic fertilizer to each mu of land, and uniformly rotary tillage; then, the cage is made by gathering, the cage surface is flat, the width of the cage surface is 1 meter, the width of the drainage ditch is 25 cm, and the depth is 20-30 cm; then spreading earthworm soil on the carriage surface for 5-10 cm; sowing, mixing the epimedium seeds treated by the storage method, sand and carbendazim water, specifically, mixing 25% of carbendazim wettable powder 1g and 220g of water, and uniformly sowing the seeds subjected to sand storage in example 1 on a compartment surface according to 800 seeds per square; then covering earthworm soil with the thickness of about 2-3cm on the carriage surface, watering, thoroughly watering seeds and sand soil, covering a greenhouse by using a fiber rod, and maintaining the temperature in the greenhouse at 25 ℃ and the humidity at 65%.
Germination rate statistics: the sowing time is calculated from 0 day to 10 days, the seeds start to germinate, the seed germination is stable on the 10 th day after sowing, as shown in figure 2, the seed germination condition is estimated by the 20 th day, the seeds are sowed in the next place, and the seed germination rate is more than 80%.
The advantages of the invention include: the invention adopts wet sand to cool and laminate and store seeds step by step, and can effectively remove dormancy of the seeds, and shorten dormancy period of 8-9 months under natural conditions to about 90 days. The germination rate of the seeds stored by the method of the invention reaches more than 80% within 20 days. Far higher than the 45% germination rate reported in the literature. In addition, the method is simple, low in cost and convenient to operate, and the field sowing verification is carried out in the small arch shed, so that the method can directly guide the actual production and application.
