本发明涉及分子生物学领域,具体涉及一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法;所述方法为降低柑橘属植物中CsEXPA8基因的转录水平,其中降低柑橘属植物中CsEXPA8基因的转录水平是通过RNA干扰(RNAi)实现的。通过这种方法获得的转基因柑橘的叶片接种溃疡细菌部位的病菌细胞的数量降低至现有野生型对照柑橘的16.9%,显著抑制了转基因柑橘中溃疡细菌入侵部位溃疡病菌的生长,提高了柑橘对溃疡病的抗性。

技术领域
本发明涉及分子生物学领域,具体涉及一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法。
背景技术
柑橘溃疡病是由地毯黄单胞杆菌引起的、发生在柑橘的病害。柑橘溃疡病是一种世界性病害,也是中国柑橘产区重要病害之一。目前为控制柑橘溃疡病危害通常采取化学防治为主,生物防治为辅的综合防治策略。由于以上防治措施对环境不友好,且需要投入大量的人力、物力,亟待通过培育抗病新品种来减少溃疡病带来的损失(陈力等,2008;朱雪梅等,2017)。利用基因工程的手段,在柑橘中超量表达抗菌肽基因(Peng et al.2015)、CsBZIP基因(Li et al.2019)和对柑橘体内的感病基因CsLOB1(Peng et al.2017)进行CRISPR/Cas9介导的基因编辑,均获得了溃疡病抗性增强的转基因柑橘植株。
中国专利申请2022115636391公开了一种基于基因干扰提高柑橘对溃疡病抗性的方法,该方法为通过干扰RNA(RNAi)降低柑橘属植物中长链脂肪醇氧化酶CsFAO3基因的转录水平实现的。
该现有技术具有以下不足:该现有技术仅抑制了该转基因柑橘植株内受到溃疡病菌侵染的病菌,无法对抑制溃疡病菌最开始入侵部位的溃疡病菌的生长,无法从源头抑制溃疡病菌;且该现有技术中仅给出了离体后的叶片抑制溃疡病菌的生长效果,无法保证用于活体植株后是否抑制溃疡病菌生长效果还是如此。
发明内容
本发明为提高柑橘对溃疡病的抗性,提供一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法。该方法是将CsEXPA8基因中372 bp的编码序列作为干扰序列,导入干扰载体中,通过农杆菌介导的遗传转化法稳定整合到柑橘基因组中,有效提高柑橘对溃疡病的抗性。
为实现上述目的,本发明的技术方案为:
一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,所述方法为降低柑橘属植物中CsEXPA8基因的转录水平,其中降低柑橘属植物中CsEXPA8基因的转录水平是通过RNA干扰(RNAi)实现的。
进一步地,所述方法包括以下步骤:
(1)克隆柑橘CsEXPA8的干扰序列;
(2)进行包含所述干扰序列的CsEXPA8干扰表达载体构建;
(3)CsEXPA8干扰表达载体转化柑橘得到转基因柑橘植株。
进一步地,所述干扰序列的核苷酸序列如SEQ ID NO:1所示。
进一步地,步骤(1)中,所述干扰序列的克隆方法为:提取柑橘总RNA,然后反转录为cDNA,以cDNA为模板采用PCR扩增得到干扰序列。
进一步地,所述PCR扩增所采用的引物为rCsEXPA8-F和rCsEXPA8-R,其核苷酸序列分别如SEQ ID NO:2和SEQ ID NO:3所示。
进一步地,步骤(2)中,CsEXPA8干扰表达载体构建方法为:
A. 构建出正向干扰序列载体:利用AscI和SwaI分别双酶切pUC-RNAi载体和含有所述干扰序列的T-克隆载体,用T4-DNA连接酶将酶切后的pUC-RNAi载体大片段和所述干扰序列的酶切片段连接;
B.构建CsEXPA8干扰表达载体:将步骤A得到的正向干扰序列载体和含有所述干扰序列的T-克隆载体再分别利用XbaI和BamHI进行双酶切,用T4-DNA连接酶将酶切后的正向干扰序列载体大片段和干扰序列片段连接,获得同时包含正向和反向干扰序列片段的pUC-RNAi-CsEXPA8干扰载体;最后,利用KpnI和SalI分别双酶切所述pUC-RNAi-CsEXPA8干扰载体和pLGNe载体,利用T4-DNA连接酶将酶切后的所述正向和反向干扰序列片段和线性化的pLGNe载体连接。
进一步地,步骤(3)中,用所述CsEXPA8干扰表达载体转化柑橘的方法为:通过冻融法将所述CsEXPA8干扰表达载体导入农杆菌菌株EHA105中,再用根癌农杆菌介导法转化柑橘上胚轴茎段,经离体培养后、染色鉴定和嫁接得到转基因柑橘植株。
进一步地,步骤(3)得到转基因柑橘植株后,对转基因柑橘植株进行抗性评价,通过活体植株接种溃疡病菌,判定活体植株中CsEXPA8基因干扰与柑橘溃疡病抗性的相关性。
进一步地,对转基因柑橘植株进行抗性评价前,通过PCR验证转基因柑橘植株,采用的引物为GUS-F和GUS-R,其核苷酸序列分别如SEQ ID NO:4和SEQ ID NO:5所示。
进一步地,PCR验证后,用qRT-PCR验证转基因柑橘植株中CsEXPA8基因的转录水平,检测所采用的引物为qCsEXPA8-f和qCsEXPA8-r,其核苷酸序列分别如SEQ ID NO:6和SEQ ID NO:7所示;内参基因为柑橘Actin基因,采用引物为Actin-F和Actin-R,其核苷酸序列分别如SEQ ID NO:8和SEQ ID NO:9所示。
与现有技术相比,本发明的有益效果在于:
本发明提供了一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法。通过这种方法获得的转基因柑橘的活体叶片接种溃疡细菌部位的病菌细胞的数量降低至现有野生型对照柑橘的16.9%,显著抑制了转基因柑橘中溃疡细菌入侵部位溃疡病菌的生长,提高了柑橘对溃疡病的抗性。
本发明利用的CsEXPA8基因是一种扩增蛋白基因,已知的功能仅仅是一类引起细胞壁松驰和细胞壁柔韧性增加的蛋白,而对植物特别是对柑橘品质和性能的影响并无深入研究。通过干扰这个基因的表达,在大大提高柑橘对溃疡病的抗性的同时,最大程度上减少了对柑橘已知品质和性能的不良影响,开辟了柑橘抗溃疡病育种的另外一条途径,促进了柑橘抗溃疡病育种的发展。

图4提供了干扰CsEXPA8表达的转基因植株活体接种溃疡病菌12d后的细菌生长情况,细菌生长单位为细胞/cm2。其中WT代表野生型植株,RNAi-5和RNAi-6分别代表含有本发明所述干扰序列的其中两株转基因植株编号。含有本发明所述干扰序列的转基因植株的细菌细胞的数量与野生型植株相比具有显著差异,而转基因植株本身之间的差异不明显。

所举实施例是为了更好地对本发明的内容进行说明,但并不是本发明的内容仅限于所举实施例。所以熟悉本领域的技术人员根据上述发明内容对实施方案进行非本质的改进和调整,仍属于本发明的保护范围。
本发明选取的柑橘晚锦橙为中国农业科学院柑桔研究所于20世纪80年代初至2010年期间,从普通锦橙(Citrus sinensis)中选育获得的晚熟锦橙优系,审定编号为“渝审柑桔2011001”。
本发明使用的引物均由北京六合华大基因科技有限公司合成。
本发明中未特别指明的实验试剂均为本申请人单位市购获得的常规试剂。
本发明中所指的“基因干扰”是指采用特定方式关闭或抑制目的基因的转录和/或表达,常用的方法包括反义RNA、三链DNA、干扰RNA(RNA干扰)等,其中本发明中特指采用RNA干扰技术进行的基因干扰,因此本发明中使用基因干扰或干扰RNA(RNA干扰)表达的意思是一样的。
本发明中所指的RNA干扰(RNA interference,简称RNAi)是指通过人工克隆目的基因中高度保守的片段并将其转基因到生物体内以RNA形式诱发目的基因的同源mRNA高效特异性降解的现象。其中“RNA干扰”也可以用“干扰RNA”进行表达,在本发明中表达一样的意思。
本发明所使用的“干扰序列”、“干扰片段”或“CsEXPA8干扰序列”是指用于本发明转基因后进行RNA干扰的克隆DNA片段,特指SEQ ID NO:1所示核苷酸序列的DNA片段;因此在本发明中“干扰序列”、“干扰片段”或“CsEXPA8干扰序列”是可以交换使用的
本发明所使用的术语“载体(Vector)”是指在基因工程重组DNA技术中将DNA片段转移至受体细胞的一种能自我复制的DNA分子,包括细菌质粒、噬菌体和动植物病毒。本发明中所述的载体特指质粒载体,因此在本发明中使用术语“载体”或“质粒载体”均是可以的,均表达一个意思。本发明中所述的pUC-RNAi载体为本领域常用pUC载体、pLGNe载体为本领域常用超量表达载体,本发明背景技术中的现有技术文献中也记载了这些基本载体,只是转入的干扰基因片段不同;T-克隆载体为常用的商业化载体pGEM-T,购自Promega公司。在本发明中,pUC-RNAi载体和干扰序列的连接产物被称为“正向干扰序列载体”;在“正向干扰序列载体”上再次连接干扰序列得到的pUC-RNAi载体被称为“pUC-RNAi-CsEXPA8干扰载体”;最后通过该中间载体将正向和反向干扰序列连接到pLGNe载体上形成最终使用的干扰载体,被称为“CsEXPA8干扰表达载体”。。由于pLGNe载体载体上带有花椰菜花叶病毒启动子(CaMV 35S启动子),故由其构建的CsEXPA8干扰表达载体也具有CaMV 35S启动子,其具有如SEQ ID NO:10所示的核苷酸序列。
本发明中提及的“活体植株”是在正常生长的植物,未从植物中剥离出的植物本身。
实施例1、克隆柑橘CsEXPA8的干扰序列
1.柑橘晚锦橙的RNA提取及cDNA合成
选取柑橘晚锦橙的嫩叶0.05g,用EASYspin植物RNA快速提取试剂盒(艾德莱,CAT:RN09)提取叶片总RNA,用琼脂糖凝胶电泳验证RNA质量,用Nanodrop 2000 Thermo测定RNA的浓度。
使用500 ng的RNA利用iScript™ cDNA Synthesis Kit (Bio-Rad, Hercules,CA, USA)合成10 μl的cDNA,将cDNA稀释5倍后于-20℃保存备用。
CsEXPA8干扰序列的获得
使用分别如SEQ ID NO:2和SEQ ID NO:3所示的引物rCsEXPA8-F和rCsEXPA8-R从实施例1所得cDNA中扩增获得CsEXPA8基因干扰片段,片段长度为372 bp,其序列如SEQ IDNO:1所示,即为本发明所述的干扰序列。
将扩增的DNA片段经琼脂糖凝胶电泳后,用Biospin胶回收试剂盒(波尔公司,BSC02M1)回收,将回收产物与T-克隆载体连接,使用分别如SEQ ID NO:2和SEQ ID NO:3所示的引物rCsEXPA8-F和rCsEXPA8-R扩增所得连接产物,扩增出相应片段的菌液进行测序,获得正确的CsEXPA8干扰序列。扩增体系:2 × PCR mix(TaRaKa):25 μL;引物rCsEXPA8-F和rCsEXPA8-F(100 μmol/L):各1 μL;cDNA约60 ng;加ddH2O至50 μL。扩增程序:94℃,5min;94℃,30 s,60℃,30 s,72℃,30 s,32个循环;72℃延伸5 min。
实施例2、构建CsEXPA8干扰表达载体并转化农杆菌
载体构建流程图如图1所示,所有限制性内切酶购自THERMO公司,按照使用说明操作。
利用GenElute™质粒提取试剂盒(Sigma,PLN350)提取含有CsEXPA8干扰序列的T-克隆质粒和pUC-RNAi质粒,利用Asc 和Swa/>分别对上述两个质粒进行双酶切,酶切产物经琼脂糖凝胶电泳后回收,其中,T-克隆质粒回收CsEXPA8干扰片段,pUC-RNAi质粒回收大片段,将回收产物利用T4-DNA酶连接,将连接产物转化大肠杆菌DH5α并涂布在附加50 mg/L卡那霉素的LB固体培养基上。挑起LB培养基上长出的单菌落放入附加50 mg/L 卡那霉素的LB液体培养基中过夜;利用质粒提取试剂盒提取菌液的质粒,利用Asc/>和Swa/>对质粒进行双酶切,对酶切产物进行琼脂糖凝胶电泳,电泳条带中含有372 bp的质粒确定为正向干扰序列质粒。
对上述正向干扰质粒和含有CsEXPA8干扰片段的T-克隆质粒分别用Xba 和BamH/>进行双酶切,酶切产物经琼脂糖凝胶电泳后,T-克隆质粒回收372 bp的片段(即CsEXPA8干扰片段),正向干扰质粒回收大片段;将回收的干扰片段与线性化的正向干扰质粒按上述的方法进行连接,用连接产物转化大肠杆菌DH5α;提取菌液的质粒,用Xba/>和BamH/>酶切,酶切产物含有372 bp片段的质粒确定为pUC-RNAi-CsEXPA8质粒。
将pUC-RNAi-CsEXPA8质粒和pLGNe质粒分别利用KpnI和SalI进行双酶切,酶切产物经琼脂糖凝胶电泳后回收,pUC-RNAi-CsEXPA8质粒回收含有干扰序列的条带,pLGNe质粒回收大片段,两个片段回收后按上述的方法进行连接,用连接产物转化大肠杆菌DH5α;提取菌液的质粒,用KpnI和SalI酶切,酶切产物含有干扰片段的质粒为CsEXPA8干扰表达质粒。利用冻融法将CsEXPA8干扰表达质粒导入农杆菌菌株EHA105中,菌液保存在‒80℃的低温冰箱中。
实施例3、晚锦橙遗传转化
1.柑橘实生苗上胚轴的获得
将晚锦橙果实洗净,用75%酒精表面消毒,在无菌的条件下取出种子,剥掉种皮,接种在添加30g/L蔗糖和8g/L琼脂的MS培养基(PhytoTechnology Laboratories™,M519)上,28℃下暗培养2周,然后在16h光照/8h黑暗的光周期下培养1周。无菌条件下取萌发幼苗的上胚轴切成1 cm左右的茎段,用于根癌农杆菌介导的柑橘遗传转化。
根癌农杆菌的制备
转化前2d,将含有CsEXPA8干扰表达载体的农杆菌菌液涂布在添加50 mg/L卡那霉素的LB固体培养基上。挑取农杆菌单菌落,接种于10 mL含有相同抗生素的LB液体培养基中,28℃、220rmp的条件下震荡培养过夜。利用分光光度计测量菌液的OD值,将菌液用上述LB液体培养基稀释成OD值为0.1的菌液,在上述相同的条件下继续震荡培养,监测菌液的OD值,待其达到0.5时,用50 mL的无菌离心管收集菌液,在5000 r/min的条件下离心10 min,弃上清,用pH 5.4的MS液体培养基重新悬浮后用于柑橘遗传转化。
柑橘上胚轴茎段的转化
将切成1 cm左右的晚锦橙上胚轴茎段在农杆菌菌液中浸泡12 min,期间轻微晃动。取出茎段后将表面的菌液用无菌滤纸吸干;将茎段转移到附加1mg/L的N6-异戊烯基腺嘌呤(2-ip)、0.5 mg/L吲哚乙酸(IAA)、1 mg/L的2,4-二氯苯氧乙酸(2,4-D)、100 μM 乙酰丁香酮(AS)、30 g/L蔗糖和8 g/L琼脂的MS固体培养基上;在26℃的黑暗条件下共培养3d。
转化子的筛选
共培养完成后,将上胚轴转移到添加2 mg/L 6-苄氨基嘌呤(6-BA)、0.5 mg/LIAA、50 mg/L 卡那霉素、500 mg/L头孢霉素、30 g/L蔗糖和8 g/L琼脂的MS固体培养基中,28℃暗培养7 d后,转移至28℃、16 h光照/8 h黑暗的光周期下培养,每两周继代一次。
对上胚轴茎段两端伤口上萌发的不定芽利用β-葡萄糖苷酸酶(GUS)染液进行GUS组织化学染色;GUS染色呈蓝色的,确定为GUS阳性芽。
GUS染液含有如下成分:100 mM NaH2PO4,100 mM Na2HPO4,0.5 mM K4[Fe(CN)6],0.5 mM K3[Fe(CN)6],10 mM依地酸二钠(EDTA-Na2),1 mM的5-溴-4-氯-3-吲哚-β-葡萄糖苷酸(X-gluc),0.1%叠氮化钠(Sodium azide),0.1% TritonX-100。
转化子的成苗培养
待GUS阳性芽的茎长约0.5cm时,水平切下,在无菌的条件下微嫁接在枳橙砧木上;待嫁接口充分愈合后,从枳橙砧木的基部水平切下GUS阳性芽,按照田间切接的方法将其嫁接在田间枳橙砧木上,用塑料袋保湿2周,待其成活后去除塑料袋。
实施例4、转基因植株的PCR检测
GUS阳性芽嫁接到田间3个月后,采取叶片100 mg,使用DNA提取试剂盒(艾德莱,CAT:DN15)提取基因组DNA,PCR检测GUS基因的整合。PCR反应条件:94℃ 3 min;94℃ 30 s,60℃ 30 s,72℃ 30 s,32次循环;72℃5 min。检测引物为GUS-F和GUS-R,其核苷酸序列分别如SEQ ID NO:4和SEQID NO:5所示。阳性植株可以得到500 bp左右的扩增片段,而野生型晚锦橙植株无相应的扩增条带。
实施例5、CsEXPA8基因的表达分析
提取按照实施例4验证为PCR阳性植株的叶片,使用EASYspin植物RNA快速提取试剂盒提取叶片总RNA,用琼脂糖凝胶电泳验证RNA质量,用NanoDrop 2000 Thermo浓度计测定其浓度。使用500ng的RNA利用iScript™ cDNA Synthesis Kit (Bio-Rad, Hercules,CA, USA)合成10 μL cDNA,将cDNA稀释5倍。
利用实时荧光定量PCR进行CsEXPA8基因表达量的检测,采用的引物为qCsEXPA8-f和qCsEXPA8-r,其核苷酸序列分别如SEQ ID NO:6和SEQ ID NO:7所示。定量PCR内参为柑橘Actin基因,采用引物为Actin-F和Actin-R,其核苷酸序列分别如SEQ ID NO:8和SEQ IDNO:9所示。反应体系:iTaqTMUniversal SYBR®6 μL,100 μmol/L引物各0.3 μL,cDNA 50 ng,加ddH2O至12 μL。反应条件:95℃ 3 min,94℃ 10 s;56℃ 10 s,72℃ 10 s,40次循环;72℃ 10 min。实验重复三次。以WT植株为参照,采用2-△△Ct法计算转基因植株中CsEXPA8基因的相对表达量。
检测结果见图2,其检测结果显示,CsEXPA8基因在WT转基因植株中的表达水平为1.02,CsEXPA8基因在RNAi-5和RNAi-6转基因植株中的表达水平分别为0.34和0.33。
因此,RNAi-5和RNAi-6转基因植株与WT植株相比(即把WT植株的表达水平当作1),RNAi-5和RNAi-6表达量分别下调至现有柑橘的33.0%和32.7%。
实施例6、转基因植株的溃疡病抗性评价
采取上述转基因植株和野生型晚锦橙的枝条,将枝条上的芽嫁接在田间枳壳砧木上;待芽萌发3个月后,将转基因植株和野生型晚锦橙放置在28℃,湿度为85%,光周期为16h光照/8 h黑暗的培养箱中;用1mL去针头的注射器向叶片注射溃疡病菌0.3 mL(菌种为XccYN1 株系,由西南大学柑桔研究所胡军华博士馈送),菌液浓度为5 ×105CFU/mL。注射12d后拍照记录。拍照后立即用直径为0.5cm的打孔器将注射部位的叶片取下,3个叶圆片为一组,放入1.5 mL的离心管中,加入200 µL的无菌水,捣碎,定容成1000 µL,连续梯度稀释,取50 µL的菌液涂布LB平板,28℃暗培养2 d,统计菌斑的个数,分析转基因植株受溃疡病菌诱导后的细菌生长情况。试验重复3次。
如图3所示,活体接种溃疡病菌12d后,观察发现野生型植株(WT植株)发病较为严重,而CsEXPA8干扰表达的转基因植株(RNAi-5和RNAi-6转基因植株)虽均有不同程度发病,但病斑大小与野生型植株(WT植株)的病斑大小相比存在肉眼可见的明显差异。
如图4所示,对活体接种溃疡病菌的叶片中的细菌细胞进行统计,发现2个CsEXPA8干扰表达的转基因植株RNAi-5和RNAi-6的叶片接种部位溃疡病菌细胞的数量均显著小于野生型植株(WT植株),在受溃疡病菌侵染12d后,WT的叶片接种部位细菌细胞数量分别为3.28×108/cm2,RNAi-5和RNAi-6的叶片接种部位细菌细胞数量分别为5.47×107/cm2和8.46×107/cm2。其中RNAi-5叶片接种部位细菌细胞的数量仅为野生型(WT植株)对照组的16.9%。
综上所述,通过上述图3或4所示结果可知,活体植株接种溃疡病菌后,CsEXPA8干扰表达的转基因植株与WT植株的病斑大小相比存在肉眼可见的明显差异;转基因柑橘的活体叶片接种溃疡细菌部位的病菌细胞的数量降低至现有野生型对照柑橘的16.9%;由此可以证明,CsEXPA8基因表达显著抑制了活体植株的转基因柑橘中溃疡细菌入侵部位溃疡病菌的生长,从而大大减轻了柑橘溃疡病的症状。
此外,应当理解,虽然本说明书按照实施方式加以描述,但并非每个实施方式仅包含一个独立的技术方案,说明书的这种叙述方式仅仅是为清楚起见,本领域技术人员应当将说明书作为一个整体,各实施例中的技术方案也可以经适当组合,形成本领域技术人员可以理解的其他实施方式。
Claims (10)
1.一种利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:所述方法为降低柑橘属植物中CsEXPA8基因的转录水平,其中降低柑橘属植物中CsEXPA8基因的转录水平是通过RNA干扰(RNAi)实现的。
2.根据权利要求1所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:所述方法包括以下步骤:
(1)克隆柑橘CsEXPA8的干扰序列;
(2)进行包含干扰序列的CsEXPA8干扰表达载体构建;
(3)CsEXPA8干扰表达载体转化柑橘得到转基因柑橘植株。
3.根据权利要求2所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:所述干扰序列的核苷酸序列如SEQ ID NO:1所示。
4.根据权利要求2或3所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:步骤(1)中,所述干扰序列的克隆方法为:提取柑橘总RNA,然后反转录为cDNA,以cDNA为模板采用PCR扩增得到干扰序列。
5.根据权利要求4所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:所述PCR扩增所采用的引物为rCsEXPA8-F和rCsEXPA8-R,其核苷酸序列分别如SEQ IDNO:2和SEQ ID NO:3所示。
6.根据权利要求2-5任一所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:步骤(2)中,CsEXPA8干扰表达载体构建方法为:
A.构建出正向干扰序列载体:利用AscI和SwaI分别双酶切pUC-RNAi载体和含有所述干扰序列的T-克隆载体,用T4-DNA连接酶将酶切后的pUC-RNAi载体大片段和所述干扰序列的酶切片段连接;
B.构建CsEXPA8干扰表达载体:将步骤A得到的正向干扰序列载体和含有所述干扰序列的T-克隆载体再分别利用XbaI和BamHI进行双酶切,用T4-DNA连接酶将酶切后的正向干扰序列载体大片段和干扰序列片段连接,获得同时包含正向和反向干扰序列片段的pUC-RNAi-CsEXPA8干扰载体;最后,利用KpnI和SalI分别双酶切所述pUC-RNAi-CsEXPA8干扰载体和pLGNe载体,利用T4-DNA连接酶将酶切后的所述正向和反向干扰序列片段和线性化的pLGNe载体连接。
7.根据权利要求2-6任一所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:步骤(3)中,用所述CsEXPA8干扰表达载体转化柑橘的方法为:通过冻融法将所述CsEXPA8干扰表达载体导入农杆菌菌株EHA105中,再用根癌农杆菌介导法转化柑橘上胚轴茎段,经离体培养后、染色鉴定和嫁接得到转基因柑橘植株。
8.根据权利要求2-7任一所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:步骤(3)得到转基因柑橘植株后,对转基因柑橘植株进行抗性评价,通过活体植株接种溃疡病菌,判定活体植株中CsEXPA8基因干扰与柑橘溃疡病抗性的相关性。
9.根据权利要求8所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:对转基因柑橘植株进行抗性评价前,通过PCR验证转基因柑橘植株,采用的引物为GUS-F和GUS-R,其核苷酸序列分别如SEQ ID NO:4和SEQ ID NO:5所示。
10.根据权利要求9所述的利用CsEXPA8基因表达提高柑橘对溃疡病抗性的方法,其特征在于:PCR验证后,用qRT-PCR验证转基因柑橘植株中CsEXPA8基因的转录水平,检测所采用的引物为qCsEXPA8-f和qCsEXPA8-r,其核苷酸序列分别如SEQ ID NO:6和SEQ ID NO:7所示;内参基因为柑橘Actin基因,采用引物为Actin-F和Actin-R,其核苷酸序列分别如SEQID NO:8和SEQ ID NO:9所示。
