Abstract
本发明公开了一种营养护肝的组合物及其制备方法,灵芝提取物6~14份;维生素C(VC)2~10份;还原型谷胱甘肽(GSH)4~10份;滇黄精提取物等补充剂0~10份,制备为常用口服制剂。本发明能够发挥保肝、护肝、抗氧化、排铅、美白作用,舒缓神经提高睡眠质量,缓解疲劳,辅助降血脂。

本发明涉及领域,尤其涉及一种营养护肝的组合物及其制备方法。
背景技术
近年来由于我国经济的迅速发展,国民生活水平的提高,饮食结构的变化,过量饮酒和因为食物中过量蛋白、脂类、糖类摄入带来的疾病和潜在危害越来越严重,如:酒精肝、脂肪肝、药物肝等。另一方面生活节奏的日益加快,人们在生活、学习、工作中的竞争越来越激烈,生活不规律,精神状况不佳,过大的压力易造成急躁、焦虑、郁闷等情绪问题,在困扰包括学生、白领等在内的脑力工作者的同时也很容易影响到肝。因此越来越多的人开始关注自身的亚健康问题,保肝护肝、缓解精神压力的食品和保健食品就越来越受青睐,目前市场上的保健食品以增强免疫力、抗疲劳、抗氧化、护肝等为主,且大多为单一功能的产品,并无可同时具有缓解压力和护肝的保健食品,难以满足人们养生保健的需求。

本发明还公开了一种制备所述的营养护肝的组合物的方法,包括下列步骤:
1)分别称取灵芝与滇黄精提取物,将其粉碎成粗颗粒,加入溶剂,进行提取,分别获得提取物;
2)称取适量的植物油,加热到40~65℃,然后加入适量蜂蜡进行混合,搅拌均匀后,加入6~14重量份的灵芝提取物、2~10重量份的维生素C、4~10重量份的还原型谷胱甘肽与0~10重量份的滇黄精提取物等补充剂,搅拌混合,通过80目筛过滤,抽真空-0.08~-0.06Mpa脱气泡,获得组合物;
3)通过组合物制备口服制剂。
作为优选的技术方案,所述步骤1)中将其粉碎成粗颗粒,加入10倍量水,煎煮二次,每次煎煮2小时,合并煎液,过滤,滤液浓缩,干燥,粉碎后即得提取物;所述干燥为常压干燥、减压干燥、喷雾干燥其中的一种。

由于采用了上述技术方案一种营养护肝的组合物及其制备方法,1)分别制备将灵芝与滇黄精提取物,将其粉碎成粗颗粒,加入溶剂,进行提取,分别获得提取物;2)称取适量的植物油,加热到40~65℃,然后加入适量蜂蜡进行混合,搅拌均匀后,加入6~14重量份的灵芝提取物、0.1~8重量份的滇黄精提取物、2~10重量份的维生素C、4~10重量份的还原型谷胱甘肽与0~0.5重量份的补充剂,搅拌混合,通过80目筛过滤,抽真空-0.08~-0.06Mpa脱气泡,获得组合物;3)通过组合物制备口服制剂。
本发明的优点:
该组合物能够发挥保肝、护肝、抗氧化、排铅美白作用,舒缓神经提高睡眠质量,缓解疲劳,辅助降血脂。
本品由药食两用的灵芝、滇黄精药粉或其提取物,以及谷胱甘肽、维生素C组成,具有较好的保肝、护肝、抗氧化、排铅、美白等作用,尤其对由于肝损伤引起的脸色发黑发黄等情况的改善作用明显。
灵芝是常用的传统名贵中药之一,有“仙草”、“瑞草”之称。2020版《中国药典》收载为多孔菌科真菌赤芝Ganoderma lucidum(Leyss.exFr.)Karst.或紫芝Ganoderma sinenseZhao,Xu et Zhang的干燥子实体。其性味与归经:甘,平。归心、肺、肝、肾经。功能与主治为:补气安神,止咳平喘。用于心神不宁,失眠心悸,肺虚咳喘,虚劳短气,不思饮食。现代医学研究表明灵芝及其提取物具有多种药理作用,如:抗肿瘤、增强免疫功能、降血糖、保肝、抗衰老、抗炎、抗凝血等。灵芝可用于药品、保健食品、化妆品、生物敷料等。
灵芝提取物主要成分有灵芝多糖、糖肽类化合物、三萜类化合物、甾醇类化合物、核苷类化合物、生物碱类等,其中灵芝多糖具有增强免疫系统的机能、降低血压、护肝、预防心血管疫病的产生、加速血液微循环、提高血液供氧能力、降低机体静止状态下的无效耗氧量等。灵芝三萜类化合物具有保肝、护肝、抗炎、镇痛、镇静、抗衰老、抑制肿瘤细胞、抗缺氧等作用。
滇黄精,为百合科植物滇黄精Polygonatum kingianum Coll.et Hemsl.、黄精Polygonatum sibiricum Red.或多花黄精Polygonatum cyrtonema Hua的干燥根茎。其主要含黄精多糖糖A、B、C和黄精低聚糖糖A、B、C和甾体皂苷(主要是呋甾烯醇型皂苷和两个螺甾烯醇型皂苷),以及尚含有赖氨酸等10多种氨基酸及锌、铁、硒、菸酸、粘液质、淀粉,以及醌类等成分。
黄精多糖可有效提高人体内细胞的SOD活性和机体免疫力,SOD可清除过氧化阴离子、降低体内过氧化物沉积,抗癌,抗衰老,降血脂等。
现代药理研究表明,滇黄精具有保护肝脏、降血糖、降血脂、增加耐缺氧、抗疲劳、抗氧化、延缓衰老、增强免疫、增加冠脉血流量、止血、抗菌、抗病毒等作用。常用于治疗高血压、冠心病、白细胞减少症、再生障碍性贫血、药物中毒性耳聋等。
谷胱甘肽(还原型)是一种既可以由人体自己产生,也可以从食物获得的,能帮助身体预防各种疾病的营养素,在医药、保健品和化妆品等行业中有着广泛应用。谷胱甘肽是由谷氨酸、半胱氨酸和甘氨酸通过肽键缩合而成的三肽化合物,是哺乳动物细胞中抗氧化胁迫最重要的低分子硫醇。具有较好的保护肝脏、解毒、清除自由基、抗衰老、抗氧化等作用。
维生素C是一种存在于食物中的高等灵长类动物与其他少数生物的必需营养素。用于防治坏血病、改善铁、钙和叶酸的利用;改善脂肪和类脂特别是胆固醇的代谢,预防心血管病,对化学性肝损伤有预防性保护作用。其对谷胱甘肽的稳定性也有一定保护作用。

为了弥补以上不足,本发明提供了一种营养护肝的组合物及其制备方法以解决上述背景技术中的问题。

为了使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,下面结合具体实施例,进一步阐述本发明。
实施例一:
称取配方量的灵芝、滇黄精粉碎成粗颗粒,加入10倍量水,煎煮二次,每次煎煮2小时,合并煎液,过滤,滤液浓缩,减压干燥,粉碎后包装,检验,该提取物含量要求总多糖≥8.0%,以及符合食品相关要求,即得提取物。
按重量比,称取75重量份的大豆油,40~65℃,加入4重量份的蜂蜡混合,搅拌均匀,加入上述9重量份的灵芝提取物、0.1重量份的滇黄精提取物、6重量份的谷胱甘肽、6重量份的维生素C,搅拌混合,80目筛过滤,抽真空(-0.08~-0.06Mpa)脱尽气泡,备用。
囊壳配制:按重量称取300重量份的明胶、130重量份的甘油,300重量份的水,15重量份的焦糖色,投入化胶罐中60~80℃溶胶,抽真空(-0.08~-0.06Mpa)脱尽气泡,100目筛过滤,60±5℃保温,备用。
制丸,将制好的的软胶囊内容物和胶液分别输送到制丸机中,控制温度18-26℃,相对湿度小于45%,胶囊内容物0.65g/丸,制备软胶囊,干燥,包装,检验合格即得。
实施例二:
称取配方量的灵芝、滇黄精,粉碎成粗颗粒,加入10倍量50%乙醇,加热回流二次,每次1.5小时,合并煎液,静置10h以上,过滤,滤液浓缩,喷雾干燥,包装,检验,合格后即得提取物。
按重量比,称取71重量份的亚麻籽油,40~65℃,加入5重量份的蜂蜡混合,搅拌均匀,加入上述11重量份的灵芝提取物、3重量份的滇黄精提取物、7重量份的谷胱甘肽、6重量份的维生素C,搅拌混合,80目过滤,抽真空(-0.08~-0.06Mpa)脱尽气泡,备用。
囊壳配制:按重量称取300重量份的明胶、132重量份的甘油,298重量份的水,10重量份的食用色素,投入化胶罐中60~80℃溶胶,抽真空(-0.08~-0.06Mpa)脱尽气泡,100目过滤,60±5℃保温,备用。
制丸,将制好的的软胶囊内容物和胶液分别输送到制丸机中,控制温度18-26℃,相对湿度小于45%,胶囊内容物0.25g/丸,制备软胶囊,干燥,包装,检验合格即得。
实施例三:
按重量比,称取灵芝提取物为9份、滇黄精提取物2份、谷胱甘肽8份、维生素C 5份,甘露醇8份、微晶纤维素10份、羧甲基淀粉钠5份、微粉硅胶0.5份混合,70%乙醇适量制粒,干燥,压片,每片0.5g,包装,检验合格即得。
实施例四
分别称取配方量的灵芝、滇黄精粉碎成粗颗粒,分别加入10倍量水,煎煮二次,每次煎煮2小时,合并煎液,过滤,滤液浓缩,减压干燥,粉碎后包装,检验,该提取物含量要求总多糖≥8.0%,以及符合食品相关要求,即得各自提取物。
按重量比,称取大豆油75份,40~65℃,加入蜂蜡4份混合,搅拌均匀,加入上述饮片6重量份灵芝提取物、4重量份的谷胱甘肽、2重量份的维生素、1重量份的滇黄精提取物、0.1重量份其他补充剂,搅拌混合,80目筛过滤,抽真空(-0.08~-0.06Mpa)脱尽气泡,备用。
囊壳配制:按重量称取明胶300份、甘油130份,水300份,焦糖色13份,投入化胶罐中60~80℃溶胶,抽真空(-0.08~-0.06Mpa)脱尽气泡,100目筛过滤,60±5℃保温,备用。
制丸,将制好的的软胶囊内容物和胶液分别输送到制丸机中,控制温度18-26℃,相对湿度小于45%,胶囊内容物0.65g/丸,制备软胶囊,干燥,包装,检验合格即得。
所述补充剂包括0.05重量份的薤白多糖;2重量份的荷花花蕊粉末;0.1重量份的郁李仁分离蛋白粉;1重量份的香薷粉。
实施例五
分别称取配方量的灵芝、滇黄精,粉碎成粗颗粒,分别加入10倍量50%乙醇,均加热回流二次,每次1.5小时,合并煎液,静置10h以上,过滤,滤液浓缩,喷雾干燥,包装,检验,合格后即各自得提取物。
按重量比,称取71重量份的亚麻籽油,40~65℃,加入8重量份的蜂蜡混合,搅拌均匀,加入上述14重量份的灵芝提取物、10重量份的谷胱甘肽、10重量份的维生素C、8重量份的滇黄精提取物、0.5重量份的补充剂,搅拌混合,80目过滤,抽真空(-0.08~-0.06Mpa)脱尽气泡,备用。

试验方法:采用酒精肝损伤模型法,小鼠灌胃实验(SPF级昆明种雄性小鼠),七个试验组给予不同的样品,空白对照组和模型对照组给予同体积纯水,按20mL/kg.BW每天经口灌胃一次,每周称两次体重,按体重调整受试样品剂量。连续给予受试样品30天,结束时将模型对照组和七个试验组,一次经口灌胃给予50%乙醇溶液,灌胃量12mL/kg.BW,空白对照组给予同体积的纯水,禁食16h后颈椎脱臼处死动物,取肝左叶作组织病理学检查,剩余肝脏用0.9%氯化钠注射液冲洗、拭干后准确称取组织重量,剪碎置于匀浆器中,按重量g:体积mL=1:9的比例,加入0.9%氯化钠注射液,在冰水浴条件下以20000r/min机械匀浆10S,间歇30S,反复3次制备成10%的肝匀浆,均以2500r/min离心10min,取上清液进行各项指标(丙二醛MDA、还原型GSH、甘油三酯TG)的检测和动物肝脏病理学检查。
试验结果
注1:各剂量组与模型对照组比较,*表示P<0.05,**表示P<0.01;模型对照组与空白对照组比较,#表示P<0.05;##表示P<0.01。
注2:配方1维生素C、配方2灵芝提取物+滇黄精提取物、配方3GSH、配方4GSH+VC、配方5滇黄精提取物+GSH+VC、配方6灵芝提取物+GSH+VC、配方7灵芝提取物+GSH+VC+补充剂。
由上表可见,与模型对照组比较,各配方试验组MDA值降低,其中配方5、配方6、配方7有显著性差异(P<0.05);GSH值配方5、配方6、配方7组均升高,其中配方7有显著性差异(P<0.05);TG值各配方组均降低,其中配方6、配方7均有显著性差异(P<0.01)。说明本品有明显的保护肝脏作用。

注2:配方1维生素C、配方2灵芝提取物+滇黄精提取物、配方3GSH、配方4GSH+VC、配方5滇黄精提取物有+GSH+VC、配方6灵芝提取物+GSH+VC、配方7灵芝提取物+GSH+VC+补充剂。
上表可以看出,与模型对照组比较,各配方组肝细胞脂肪变性程度均减轻,其中配方6、配方7组与模型对照组差异有显著(P<0.01),病理组织学指标可判断有明显保肝作用。
结论:依据《允许保健食品声称的保健功能目录非营养素补充剂(2022年版)(征求意见稿)》中“对化学性肝损伤有辅助保护功能检验方法”可以判定,本组合物对酒精性肝损伤有辅助保护作用。
人体食用改善皮肤颜色观察
选取2022年1月-2022年10月社区医院招募的志愿者。纳入标准:①对肤色不满意,非皮肤病引起的面部偏黑、皮肤干燥及脱屑者;②入组前3个月未使用过皮肤美白产品;③女性,年龄18~45岁;④知情同意并签署同意书。排除标准:①有银屑病、单纯疱疹等皮肤病;②有化妆品过敏史;③妊娠或哺乳期女性;④有系统性疾病。采用随机数字表法,将志愿者随机分为3个观察组和1个对照组。
干预方法:
对照组:给予面部清洁保湿,志愿者平躺,采用皮肤蒸汽喷雾仪器对其进行面部熏蒸,每次5~10min,1次/d,使用一次性毛巾将脸部残余水分清除,连续熏蒸美白1个月;观察组:在对照组基础上给予口服配方5、配方6、配方7,3粒/次,2次/d,连续应用1个月。
盲法美白效果评价:干预前皮肤肤色与肤色色度卡进行比对,试验后比对志愿者皮肤颜色。显效:干预后肤色较干预前明显白了两个度;有效:干预后皮肤肤色较干预前明显白了一个度;无效:干预前后皮肤肤色变化不明显。总有效率以显效+有效计。
结果见表3

结果表明本组合物对人体皮肤颜色有明显改善作用。

Claims (7)
3.一种制备如权利要求1至2中任意一项所述的营养护肝的组合物的方法,其特征在于,包括下列步骤:
1)分别称取灵芝与滇黄精,将其粉碎成粗颗粒,加入溶剂,进行提取,分别获得提取物;
2)称取适量的植物油,加热到40~65℃,然后加入适量蜂蜡进行混合,搅拌均匀后,加入6~14重量份的灵芝提取物、0.1~8重量份的滇黄精提取物、2~10重量份的维生素C、4~10重量份的还原型谷胱甘肽与0~0.5重量份的补充剂,搅拌混合,通过80目筛过滤,抽真空-0.08~-0.06Mpa脱气泡,获得组合物;
3)通过组合物制备口服制剂。
4.如权利要求3所述的营养护肝的组合物的制备方法,其特征在于:所述步骤1)中将其粉碎成粗颗粒,加入10倍量水,煎煮二次,每次煎煮2小时,合并煎液,过滤,滤液浓缩,干燥,粉碎后即得提取物;所述干燥为常压干燥、减压干燥、喷雾干燥其中的一种。
5.如权利要求3所述的营养护肝的组合物的方法,其特征在于:所述步骤1)中将其粉碎成粗颗粒,加入10倍量的50%~80%乙醇,加热回流提取二次,每次煎煮1.5~2小时,合并煎液,静置,过滤,滤液回收乙醇、浓缩,干燥,粉碎后即得提取物;所述干燥为减压干燥、喷雾干燥其中的一种。
6.如权利要求4或5所述的营养护肝的组合物的方法,其特征在于:所述提取物中总多糖含量≥8.0%。
7.如权利要求3所述的营养护肝的组合物的制备方法,其特征在于:所述口服制剂为胶囊、软胶囊剂、片剂、颗粒剂、粉剂其中的一种。
