Abstract
The invention discloses a plant anti-dandruff composition and a preparation method and application thereof, wherein the composition consists of glycolipid and plant extract, wherein the mass ratio of the glycolipid to the plant extract is (1-4) to (2-5); the plant extract consists of rosemary leaf water and Chinese arborvitae twig extract; the glycolipid is sophorolipid. The composition has synergistic effect in inhibiting Malassezia. The preparation process is simple to operate, is applied to shampoo and scalp care solution products, and has the effect of removing dandruff.
Description
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a plant anti-dandruff composition, and a preparation method and application thereof.
Background
As early as 70 s, chemical dandruff-removing antipruritic agents were widely used in hair-washing and hair-care products. Such as early sulfur and salicylic acid, although having bactericidal effect, the irritation is very large; the subsequently developed selenium sulfide product has unstable performance and complex production process, and can cause sebaceous gland atrophy, accelerate surface cell aging and damage hair quality after long-time use; the dandruff removing and itching relieving agents adopted by the dandruff removing product are basically chemical substances artificially synthesized by ZPT, climbazole and the like, the irritation problem can not be solved properly, the quality is uneven, the long-time use can generate drug dependence, the scalp antibacterial, anti-virus and anti-disease immunity is reduced, and the hair quality damage problems such as alopecia, hair falling and the like are caused. Although hair can be cleaned by the chemical synthetic product, the problem of dandruff generation of scalp cannot be fundamentally solved. With the ban of ZPT by the european union, the development of novel safe and efficient anti-dandruff ingredients has been a major concern in the industry.
The development of mild and non-irritating anti-dandruff products is therefore a technical problem to be solved.
Disclosure of Invention

In one aspect of the invention, the invention provides a plant anti-dandruff composition, which consists of glycolipid and plant extract, wherein the mass ratio of the glycolipid to the plant extract is (1-4) to (2-5); the plant extract comprises rosemary leaf water and cacumen biotae extract; the glycolipid is sophorolipid.
As a preferred embodiment of the plant dandruff removing composition of the present invention: the mass ratio of the rosemary leaf water to the cacumen biotae extract is (90-98) to (2-10).
As a preferred embodiment of the plant dandruff-removing composition of the present invention: the mass ratio of the rosemary leaf extract to the cacumen biotae extract is 96: 4.
As another aspect of the present invention, the present invention also provides a method for preparing the plant anti-dandruff composition, the method for preparing the sophorolipid comprising: performing plate culture on candida, selecting a single colony to a liquid culture medium for fermentation, standing and layering after the fermentation is finished, and collecting bottom sophorolipid.
Preferably, the sophorolipid includes an acid-type sophorolipid and a lactone-type sophorolipid.
Preferably, the preparation method of the plant extract comprises: mixing rosemary leaf and cacumen biotae, adding water, heating, refluxing, extracting, filtering, and collecting filtrate to obtain the plant extract.
As another aspect of the invention, the invention also provides the application of the plant anti-dandruff composition in cosmetics with anti-dandruff efficacy.
The invention has the beneficial effects that: the invention discloses a plant anti-dandruff composition and a preparation method thereof (comprising a composition product), wherein the composition has the effect of synergistically inhibiting malassezia. The preparation process is simple to operate, is applied to shampoo and scalp care solution products, and has the effect of removing dandruff.
Detailed Description
In order to make the aforementioned objects, features and advantages of the present invention more comprehensible, embodiments accompanying specific embodiments of the present invention are described in detail below.
In the following description, numerous specific details are set forth in order to provide a thorough understanding of the present invention, but the present invention may be practiced in other ways than those specifically described and will be readily apparent to those of ordinary skill in the art without departing from the spirit of the present invention, and therefore the present invention is not limited to the specific embodiments disclosed below.
Furthermore, reference herein to "one embodiment" or "an embodiment" means that a particular feature, structure, or characteristic described in connection with the embodiment is included in at least one implementation of the invention. The appearances of the phrase "in one embodiment" in various places in the specification are not necessarily all referring to the same embodiment, nor are separate or alternative embodiments mutually exclusive of other embodiments.
The invention comprises the following raw materials:
raw material 1: INCI name: glycolipids;
raw material 2: INCI name: rosemary (Rosmarinus OFFICINALIS) leaf water; raw material 3: INCI name: leaf extract of Thuja ORIENTALIS (BIOTA ORIENTALIS).
Sophorolipid is a glycolipid surfactant generated by metabolism of saccharomycetes, has the biological characteristics of good surface activity, biodegradability, environmental friendliness to human bodies, bacteriostasis, antivirus and the like, is one of the most promising biological surfactants at present, and can be applied to the fields of agriculture, pharmacy, cosmetics, food and the like.
Rosemary: chinese medicinal plant identification by picture: rosemary is a tonic, a diaphoretic, a stomach-invigorating and tranquillizing drug and can treat various headache diseases. Mixing with borax to make into infusion, and is excellent shampoo and can prevent premature alopecia. In modern medical analysis, rosemary mainly contains phenolic acids and terpenes, such as carnosic acid, rosmarinic acid, 1, 8-cineole, camphor, alpha-pinene, etc. The method has the advantages of destroying bacterial cell walls, damaging bacterial inner and outer membranes, destroying permeability and integrity of cell membranes, causing exosmosis of intracellular substances (K, nucleic acid and protein), destroying intracellular environment stability and having strong inhibition effect on gram-negative bacteria.
Cacumen biotae: the book Ben Cao Hui Yan (treatise on materia Medica): ce Bai Ye, also can stop bleeding and remove wind-damp. It is taken immediately after pounded into juice for hematemesis, epistaxis, metrorrhagia, hematochezia and blood heat overflowing to the meridians. It is used for treating wind-heat rash and pain caused by the general injection and inability to rotate. But the two diseases are specialized for blood damage due to heat and tendon damage due to wind-damp. The cacumen Platycladi contains volatile oil, and the main ingredients of the oil are cacumen Platycladi, cacumene Platycladi, fenchone, etc.; other ingredients further comprise flavonoids such as fructus Citri Junoris essence, quercetin, myricetin, hinokiflavone, etc.; the leaves also contain trace elements such as potassium, sodium, nitrogen, phosphorus, calcium, magnesium, manganese, zinc and the like. Quercetin and tannin are effective components in stopping bleeding, and can obviously shorten bleeding and blood coagulation time. The volatile oil has obvious inhibiting effect on Staphylococcus aureus, tetracoccus tetragonorrhoeae, escherichia coli and fungi. The flavone has strong antiinflammatory effect, and can inhibit biosynthesis of LTB4, 5-HETE and release of B-glucuronidase as metabolite of arachidonic acid of neutrophils.
(1) Preparation of glycolipids:
the sophorolipid used by the invention is sophorolipid which is a microbial secondary metabolite produced by candida by taking sugar, vegetable oil and the like as carbon sources through a fermentation process under certain conditions.
Fermentation medium: 100g/L glucose, 5g/L yeast powder and KH 2 PO 4 1g/L,MgSO 4 ·7H 2 O0.5g/L,NaCl 0.1g/L,CaCl 2 ·2H 2 0.1g/L of O, 0.7g/L of peptone, 50g/L of rapeseed oil and sterile water as a solvent.
Fermentation of natural candida: candida strains (ATCC 22214 as an example) were picked.
Activating the strain, selecting strain YPD solid culture medium, streaking and activating/test tube slant, and culturing at 30 deg.C for 48 hr to obtain single colony. And (4) tube preservation: stored at 4 ℃ for later use.
Single colonies were picked from the plate and inoculated into 50ml YPD liquid medium, shake-cultured at 30 ℃ and 220rpm for 48h. After fermentation, the fermentation tank is heated to 60 ℃ and maintained for 20-30 minutes to inactivate the yeast.
Separating to collect brown yellow oily viscous glycolipid at the bottom, adding warm distilled water with the same volume of 55-65 deg.C, mixing to remove residual hydrophilic impurities, and heating again (55-65 deg.C) to promote separation between water and glycolipid. Standing for layering, taking the oily product at the lower layer, and combining the upper aqueous phase with the bacterial liquid layer for treatment. The water washing is repeated for 2-3 times to obtain the clarified brown yellow viscous glycolipid.
(2) Plant extracts: an example of a preparation method of a rosemary leaf water and cacumen biotae extract compound raw material comprises the following steps: the compound raw materials of the rosemary leaf water and the cacumen biotae extract are water extracts. An example of an extraction process: putting rosemary leaf and cacumen biotae into a reflux tank according to the mass ratio of 96: 4, adding water with the mass being 4 times that of the medicinal materials, heating and refluxing at 95 ℃, calculating the time from the beginning of water reflux, wherein the extraction time is 2.5 hours, filtering and collecting the extracting solution after the reflux is finished, and combining the extracting solutions for later use.
Mixing glycolipid, rosemary leaf water and cacumen Platycladi extract at a certain ratio, adding deionized water to 100%, stirring and dissolving uniformly to obtain the dandruff removing composition.
Mixing the glycolipid obtained in the step (1) and the rosemary and biota orientalis extract compound raw material obtained in the step (2) according to a certain weight ratio, adding deionized water to 100%, and stirring and dissolving uniformly to obtain the plant anti-dandruff composition.
The plant anti-dandruff composition consists of the components in the following mass percentage in the table 1, wherein the table 1 comprises the following components:

Verifying the bacteriostatic effect of the anti-dandruff composition:
and (3) testing the malassezia bacteriostasis rate:
1) Preparing malassezia furfur suspension;
2) Malassezia (taking GDMCC 2.181 as an example) suspension is appropriately diluted with PBS (0.03 mol/L phosphate buffer) at the following concentrations: 0.1ml of the sample was dropped into 5.0ml of the control sample PBS, and the number of recovered bacteria was 1X 10 4 ~9×10 4 cfu/ml;
3) Diluting a test sample to a specified concentration with sterile water;
4) Sucking 5.0ml of test sample stock solution, placing into a sterilized test tube, and keeping the temperature at 20 ℃ for 5min;
5) Sucking 0.1ml of malassezia liquid, adding into a test tube containing 5.0ml of sample, quickly stirring, and immediately timing;
6) After 5min of action, taking 0.5ml of malassezia and sample mixed solution, adding into a PBS test tube containing 4.5ml of sterilized solution, and mixing well;
7) Placing for 10min, sucking 1ml of sample liquid, placing in a sterilization plate, inoculating two sterilization plates for each sample liquid, pouring 15ml of Leeming-Notman culture medium at 40-45 ℃, rotating the plates to ensure that the sample liquid is fully and uniformly, turning the plates after the Leeming-Notman culture medium is solidified, and counting viable bacterial colonies after culturing for 7d at 28 ℃;
8) Replacing a test sample with PBS, and simultaneously operating according to the steps to serve as a control sample;
9) The experiment was repeated three times and averaged.

Calculating the CI indexes of the formulas of group I and group II in the table 1 according to a calculation formula to obtain: CI I =0.435、CI II = 0.425. It is known that the glycolipid and the plant extract are compounded and used according to a certain proportion, and the compound has the effect of synergistically inhibiting malassezia furfur. The glycolipid and the plant extract can generate synergistic effect in the mass ratio range of 10: 50 to 40: 20.
The plant anti-dandruff compositions of examples 1 to 4 and comparative examples 1 to 2 were prepared according to Table 4.

the preparation method of the shampoo containing the plant anti-dandruff composition comprises the following steps:
(1) Mixing sodium laureth sulfate, sodium lauroamphoacetate, cocamidomethyl MEA, a suspending agent, a chelating agent and deionized water, heating to 80 x 85 ℃, adding a cation conditioner, and uniformly stirring and dissolving a pearling agent;
(2) Cooling to 60 deg.C, sequentially adding sodium benzoate and citric acid, mixing and stirring;
(3) And continuously cooling to 40 ℃, adding the plant anti-dandruff composition, a proper amount of polydimethylsiloxane and phenoxyethanol, uniformly stirring, sampling and discharging to obtain a finished product.
To verify the efficacy of the shampoo containing the plant dandruff-removing composition, comparative experiments of examples and comparative examples were conducted, as shown in table 5:

The frequency of use is once every two days, and no other hair washing and caring products are used, and no hair beautifying action is carried out. The anti-dandruff effect was evaluated before and after 4 weeks of use of the test product.

(1) Mixing polyethylene glycol-90M, aminomethyl propanol, a chelating agent and deionized water, and stirring to dissolve uniformly;
(2) Adding plant dandruff removing composition, appropriate amount of citric acid, phenoxyethanol and ethylhexylglycerin, stirring, sampling, testing, and discharging to obtain the final product.

A plant anti-dandruff composition characterized by: the composition consists of glycolipid and plant extract, wherein the mass ratio of the glycolipid to the plant extract is (1-4) to (2-5); the plant extract comprises rosemary leaf water and cacumen biotae extract; the glycolipid is sophorolipid.
2. A plant antidandruff composition according to claim 1, characterized in that: the mass ratio of the rosemary leaf extract to the cacumen biotae extract is (90-98) to (2-10).
3. A plant antidandruff composition according to claim 2, characterized in that: the mass ratio of the rosemary leaf water to the cacumen biotae extract is 96: 4.

A method of preparing a plant dandruff-removing composition according to claim 3, wherein: the preparation method of the sophorolipid comprises the following steps: performing plate culture on candida, selecting a single colony to a liquid culture medium for fermentation, standing and layering after the fermentation is finished, and collecting bottom sophorolipid.

A method of preparing a plant anti-dandruff composition according to claim 4, characterized in that: the preparation method of the plant extract comprises the following steps: mixing rosemary leaf and cacumen biotae, adding water, heating, refluxing, extracting, filtering, and collecting filtrate to obtain the plant extract.
6. Use of the plant anti-dandruff composition according to claim 1 in cosmetics having anti-dandruff effect.
7. Use according to claim 6, characterized in that: the cosmetic comprises shampoo and scalp care solution.
8. Use of the plant dandruff-removing composition of claim 1 for the preparation of a product having the effect of inhibiting malassezia furfur.
