Description

Dandruff-removing and itching-relieving shampoo composition containing multi-element zinc
Technical Field
The invention belongs to the field of daily chemicals, and particularly relates to an anti-dandruff and itching-relieving shampoo composition containing multi-element zinc.
Background
Dandruff is medically known as pityriasis capitis, a skin disease caused by malassezia (one of the fungi). The proliferation of malassezia on the scalp causes the hyperproliferation of the stratum corneum of the scalp, thereby promoting the abnormal desquamation of stratum corneum cells in the form of white dandruff, namely dandruff.
The research results of Lafang and the scalp microecological research project group of the institute of microbiology of Chinese academy of sciences show that the healthy scalp ecological environment is kept balanced by pH value, grease, flora, metabolic balance and the like. When the secretion of oil and fat of scalp is unbalanced, the oil of scalp is greasy; when the scalp flora is unbalanced in environment and lipophilous bacteria are bred in large quantity, the phenomenon of itching can occur; the scalp stratum corneum is metabolized too fast, so that the fungus clusters form dandruff, the scalp is not only itching and even the white objects in blocks fall off, and in addition, the distribution of scalp microorganisms of people in different areas is not the same.
In daily life, people select anti-dandruff shampoo, which is a widely adopted method for solving the dandruff problem. Salicylic acid, Zinc Pyrithione (ZPT), ketorolac ethanolamine salt (OCT), climbazole and ketoconazole are main anti-dandruff agents in the market for anti-dandruff shampoos and other anti-dandruff products, and Zinc Pyrithione (ZPT) has been widely used as an anti-dandruff agent in various hair washing products.
Zinc pyrithione is an organic zinc salt, which is named: the zinc bis (N-pyridine-2-thiol oxide anion) or the zinc N-2-pyrithione salt is a complex formed by coordination of N-2-Pyrithione (PT) serving as a ligand and zinc ions. In the solution, the complex is easy to decompose to form ligand and zinc ions, the zinc ions can adsorb and destroy the functions of cell membranes and enter the cell membranes to enable certain cell components to escape, interfere the cell metabolic process or interfere the action of various enzymes, so that the cells lose the due biological functions, finally the cells die, and finally the sterilization effect is achieved, so that the zinc plays an important role.
Zinc also has the effect of inhibiting 5-a reductase, thereby reducing excessive sebum secretion, supplementing zinc element to skin, and helping to maintain normal metabolism of skin. The zinc complex and the zinc-containing compound comprise organic zinc salt and inorganic zinc salt. The zinc pyrithione as an organic zinc salt has good oil control and bacteriostasis effects, but has certain degreasing property, and the scalp and hair are dry after use. The organic zinc salt also comprises organic zinc such as zinc gluconate, zinc lactate, zinc pyrrolidone carboxylate and the like, has good solubility, and has the effects of synergistically moisturizing, controlling oil, inhibiting bacteria and synergistically removing dandruff; in addition, inorganic zinc salts include zinc oxide, zinc chloride, zinc sulfate, zinc carbonate, and the like,
zinc oxide is insoluble in water like zinc pyrithione. The application of common zinc oxide (such as pharmaceutical grade zinc oxide) is that the zinc oxide ointment sold in the market has certain functions of astringency, antibiosis and antiphlogosis. The research finds that the antibacterial effect of the zinc oxide depends on the specific surface area of the zinc oxide. The zinc oxide has rich raw material sources and low price, and simultaneously, the zinc is also a mineral element necessary for human bodies. Zinc oxide, especially with a large specific surface area, has been one of the hot spots in the research of inorganic antibacterial agents.
The zinc oxide or nano zinc oxide with small particle size generally has large specific surface area and surface effect, is easy to generate affinity with contacted microorganisms to have bactericidal capacity, the mechanism of the zinc oxide or nano zinc oxide is similar to that of zinc pyrithione, ZnO slowly releases zinc ions in an aqueous medium, the zinc ions are gradually released, and due to the redox property of the zinc ions, when the zinc ions are combined with bacterial cell membranes, the zinc ions react with organic matters in the zinc ions to destroy the structure of membrane proteins, so that the activity of the membrane proteins is lost, and the aim of sterilization is fulfilled. Meanwhile, holes on the surface of the nano zinc oxide can generate electrons to directly participate in the reaction, more holes can generate more electrons, and the sterilization capability of the nano zinc oxide is improved. According to the contact type sterilization mechanism, zinc ions directly contact with bacteria to kill the bacteria, and when the bacteria are killed, the zinc ions are dissociated from thalli and then contact with other bacteria to complete a new sterilization task, so that good sterilization activity is displayed.
The invention aims to provide a shampoo composition, which achieves good effects of nourishing, astringing, controlling oil, inhibiting bacteria and relieving itching of scalp by using multi-zinc (organic zinc salt and inorganic zinc salt) and active ingredients which have the functions of relieving scalp, sterilizing and diminishing inflammation and are derived from plants.

Zinc oxide bacteriostasis experiment
Bacteriostatic test and experimental method
The method is a universal method for testing the potency of antibiotics at home and abroad, and is also a method specified by pharmacopoeias of various countries. And placing the oxford cup containing quantitative bacteriostatic drugs on an agar plate inoculated with the bacteria to be detected, wherein the drugs contained in the oxford cup continuously diffuse around the oxford cup to form a decreasing gradient concentration. The growth of the bacteria to be detected is inhibited within the bacteriostatic concentration range around the oxford cup, and a transparent zone (i.e. bacteriostatic zone) with sterile growth is formed. The size of the inhibition zone reflects the sensitivity of the bacteria to be detected to the determination drug, and is in a negative correlation with the minimum (or Minimum) Inhibitory Concentration (MIC) of the drug to the bacteria to be detected, i.e., the larger the inhibition zone is, the smaller the MIC is, and the stronger the inhibitory effect is.
The strain is as follows: malassezia furfur ATCC 44344. The strains are purchased from Guangdong culture Collection of microorganisms.
Preparing a culture medium: the culture medium is weighed according to the formula and then dissolved in a proper amount of deionized water, the mixture is shaken up, and water is added to the required volume.
Determination of bacteriostatic efficacy: the medium was shaken well and poured into plates, and the plates were cooled to solidify, about 15mL each. The bacterial suspension with a certain proportion is dripped on a flat plate, and a coating rod is used for uniformly coating the surface of the flat plate for 3 times. The plate was covered and dried at room temperature for 5 min. An Oxford cup having an inner diameter of 6mm was gently placed on the solidified medium with tweezers. Samples were aspirated with a 1mL disposable syringe needle and added to the Oxford cups, approximately 250. mu.L of sample per cup. After culturing for a certain time at a proper temperature, observing the size of the inhibition zone or measuring the diameter of the inhibition zone by using a vernier caliper. When the inhibition zone is measured, the inhibition zone which is uniform and completely aseptically grows is selected for measurement. The diameter of the catheter is measured by taking the outer edge of the bacteriostatic ring as a boundary.
Parameters of the zinc oxide sample with large specific surface area: specific surface area of 80m or more2(g) a particle diameter of 0.05 μm or more.
Experimental methods
(1) Samples of zinc oxide (large specific surface area) were dispersed in acetone (ZnO 25% content) and then diluted to 0.16-2.5% concentration zinc oxide dispersion solutions for use, with blank (no ZnO) samples being water and acetone, respectively.
(2) The culture medium is prepared (see table 6), the spore-forming bacteria are placed in the culture medium, water, acetone dispersion solutions with the content of less than 0.16%, 0.16%, 0.3%, 0.6%, 1.25%, 2.5% and acetone dispersion solution containing 1.5% ZPT and the like are respectively placed in the culture medium containing the rhodochrous furfuryl (spore-forming bacteria), and then the mixture is cultured for 3 days at 37 ℃ under aerobic condition, and the growth condition of the spore-forming bacteria is observed (see table 7).

Table 6, the medium was weighed according to the recipe and dissolved in a suitable amount of deionized water, shaken up and added with water to the desired volume. Placing the conical flask on an electromagnetic oven, heating and stirring until the culture medium solution boils slightly, and keeping the state for 5-10 min. Sterilizing at 121 deg.C for 20 min, cooling to 60 deg.C, and taking out if there is no contamination.
Bacteriostatic experiments-Table 7
The results in table 7 show that no formation of inhibition zones was observed for both the blank control sample (containing no zinc oxide) and the sample with a large specific surface area zinc oxide content of < 0.16%, indicating that the three samples did not have the effect of inhibiting the growth of malassezia furfur (spore dander bacteria); the zinc oxide with large specific surface area contains 0.16 percent, 0.3 percent, 0.6 percent, 1.25 percent and 2.5 percent of zinc oxide, the growth of the bacteria to be detected is inhibited within the range of the bacteriostatic concentration around the oxford cup, a transparent zone (i.e. bacteriostatic zone) with aseptic growth is formed, and the bacteriostatic zone shows a trend from small to large; the formation of an obvious inhibition zone can be observed by 1.5 percent ZPT, and the size of the inhibition zone is similar to that of the inhibition zone formed by zinc oxide with the content of 2.5 percent and large specific surface area.
Minimum Inhibitory Concentration (MIC) of zinc oxide
The medical grade zinc oxide and the zinc oxide with large specific surface area are respectively suspended in acetone to respectively prepare suspensions with the concentration from the lowest concentration of 90PPM to 6000PPM, the suspensions are divided into equal parts and put into a culture medium for culture, and finally the lowest concentration MIC of inhibited malassezia is observed, which is shown in Table 8.
Table 8.
Figure BDA0002232399510000141
Table 8 above can derive: the zinc oxide with large specific surface area has lower bacteriostatic (dandruff spore bacteria) concentration than the medical grade zinc oxide, which shows that the bacteriostatic effect of the zinc oxide with large specific surface area is stronger.
Clinical experiment on dandruff removal and itching relieving of shampoo composition
The test method comprises the following steps: 60 volunteers with moderate dandruff and itching were selected, healthy scalp patients without seborrheic dermatitis, allergy and wound were asked, before trial, sensory and hair image evaluations were performed on the patients, and the patients were allowed to use the shampoo of the preferred embodiment of the present invention and the shampoo of the blank embodiment of the present invention, continuously (samples were used according to daily use frequency), after 14 days (two weeks), the patients were stopped, and after 12 hours, 24 hours and 48 hours of stopping, the dandruff and itching status were observed, respectively, and scored, and the comprehensive evaluations are shown in table 9.
The score before and after the use of the volunteer skin was 1, 2, 3, 4, 5, the higher the score the better the effect: 1-2: invalid; 2-3: medium; 3-4: good; 4-5: very good.
Sensory evaluation of the hair of the volunteers before and after the use of the product. TABLE 9
Figure BDA0002232399510000151
In Table 9, the percentage of inefficiency in the preferred embodiment of the invention is significantly lower than in the blank embodiment of the invention, as seen by the statistics of reduced dandruff and reduced head itch.
