Description

Essential oil composition for scalp care and application
Technical Field
The invention belongs to the field of cosmetics, and particularly relates to an essential oil composition for scalp care and application thereof.
Background
With the continuous improvement of the living standard and the living quality of people, the requirements of consumers on the beauty and skin care products necessary for daily life are higher and higher, and the influence of the plant skin care concept of returning to nature is emphasized more and more.
In recent years, scalp health problems are increasingly prominent due to factors such as fast pace of life, night stay, irregular diet, hair scalding and dyeing, environmental pollution and the like. Data from the scalp health research center of the Chinese health education Association show that the scalp of more than 60% of adults in our country have health problems of varying degrees. However, human perception of scalp care is still at a low level and there are still few scalp care products on the market.
The healthy scalp ecosystem is maintained by three major balances: oil, flora and metabolic balance. If the three major balance environments are unbalanced, the scalp is easy to produce oil and become greasy, a large amount of harmful bacteria are bred, and the phenomenon of itching occurs, and meanwhile, the scalp cuticle metabolism is too fast, and dandruff is formed. In addition, the scalp is the second thin skin of the human body and ages 6 times faster than the face. The hair is dried, easy to age and inflame due to wind blowing, solarization, dyeing and ironing, hot air drying and the like, and the hair is also dried, branched and fallen, scales of the hair are damaged, and the hair quality is weak.
In order to solve the problems of dry scalp, easy aging, dry hair, frizzy hair and damaged hair scales, currently, moisturizing agents such as glycerin and the like and conditioning agents such as cationic polymers or silicone oil and the like are added in washing-off type washing and protecting products on the market. However, the moisturizer in the washing and caring product has short retention time on hair and is not easy to be absorbed in time, and the cationic polymer or the silicone oil can be adsorbed on scalp and hair, so that the absorption of the scalp on functional components can be hindered after long-term use, the metabolism abnormality of the scalp microenvironment can be brought, the hair is easy to become greasy, and the like. The leave-on washing and protecting product can solve some hair problems, but often brings negative effects of greasy feeling, sticky hand feeling, white scurf and the like after use.
The essential oil has small molecular weight, is easy to permeate skin to be absorbed and enter deep layers of the skin, has very good effects of promoting blood circulation of scalp, activating hair roots and promoting hair growth, and is gradually favored by people.
CN201810008867X discloses a scalp care essential oil and its preparation process, which comprises the following components: tea tree seed, lavender, basil, turmeric, black sesame, fleece-flower root, ginseng and rosemary; the scalp care essential oil can effectively care the scalp, adopts plants and traditional Chinese medicines as the original formula, has the effects of astringing pores, promoting cell regeneration, promoting blood circulation and conditioning oily scalp, reduces the damage of the traditional chemical shampoo to the scalp, and protects the scalp.

Test example 1 zone of inhibition test
The testing method is an Oxford cup method (also called a pipe-disk method), which is a universal method for measuring the potency of antibiotics at home and abroad and is also a method specified by the multi-national pharmacopoeia. The inhibition effect of the sample to be tested on Malassezia (Malassezia furfur, Pityrosporum orbiculare), purchase unit: Guangdong province microorganism culture collection, GIM2.181, is judged through the bacteriostasis zone test.
Test samples: the products prepared in examples 1 to 4 and comparative examples 1 to 6 were diluted 5 times with olive oil and used as samples to be tested.
Pouring the culture medium subjected to high-temperature sterilization into sterile culture dishes with the diameter of 9cm by aseptic operation, enabling each culture dish to have 15-20 mL of the culture medium, cooling, adding 0.2mL of malassezia suspension, uniformly coating the surface of the culture medium by using a coating rod, vertically placing an Oxford cup on the surface of the corresponding culture medium by using tweezers, slightly pressurizing to enable the Oxford cup to be in contact with the culture medium without gaps, placing 3 small tubes on each flat plate, respectively dropwise adding 0.1mL of various samples to be detected into each small tube, and marking without overflowing. Malassezia: and (5) placing the culture box at 37 ℃ for anaerobic culture for 2-5 days, and observing the result.
During culture, on one hand, test bacteria start to grow, and on the other hand, a sample to be tested diffuses towards the periphery by taking the Oxford cup as an original point to form a circular area where bacterial colonies cannot grow, and the circular area is a 'bacteriostatic zone'. The bigger the zone of inhibition, the better the acne removing effect.
The result of the drug sensitivity test is that the sensitivity is judged according to the diameter of the inhibition zone, and the judgment standard refers to the following table 2.
TABLE 2 drug sensitivity test criteria
Diameter of antibacterial ring (millimeter) Sensitivity of the device
More than 20 Extreme sensitivity

The test results are shown in Table 3.
TABLE 3 test results of zone of inhibition test
Figure GDA0002976139740000071
The results show that the malassezia inhibition effects of the embodiments 1-4 in the invention are very obvious, which shows that the essential oil composition has very good antibacterial and anti-dandruff effects; and the effect is better than that of single-component essential oil, which shows that the essential oil has synergistic effect on inhibiting malassezia.
Test example 2 scalp moisturizing Effect test
Experimental materials: the products of example 1 and comparative examples 1-6 were diluted to 5 times with olive oil and used as samples to compare the change in moisture content of the skin of the test subjects after the samples were used.
The test refers to the evaluation guideline of moisturizing efficacy of cosmetics in the light industry standard to test the moisturizing efficacy of the sample. The principle of the method is as follows: the capacitance method is used for measuring the moisture content of the human skin horny layer, and is based on the obvious difference of dielectric constants of water and other substances, the capacitance values of the measured skin are different according to the moisture content of the human skin horny layer, and the parameters can represent the moisture content of the scalp.
40 subjects stained with permum were selected, all subjects had no history of any scalp allergy or scalp affection, and the test sites had no abnormality. A female subject; the age is 21-45 years. The whole testing process is carried out in a constant temperature and humidity laboratory, the room temperature is 20-22 ℃, and the humidity is 40-60%. The moisture content of each experimental site was determined using a Corneometer CM825 (Courage + Khazaka, germany, available from hokkiso). The larger the value, the higher the water content of the stratum corneum. The posterior side of the head was selected as the test site (5 cm. times.5 cm). Before testing, the testee washes hair with the same shampoo, dries, and then keeps a certain body position for 30 min. Example 1 and comparative examples 1 to 6 were each separately preparedThe sample is applied to the test site. The test time points are respectively before sample application (0h), after sample application for 1h and 2 h. The sample coating amount is 2.0 +/-0.1 mg/cm2
And then calculating the average hydration rate of the volunteers at different time intervals, and calculating the hydration rate by using a formula (1):
Figure GDA0002976139740000081
the results are shown in Table 4.
TABLE 4
Figure GDA0002976139740000082
As can be seen from Table 4, the skin of the sample of example 1 of the present invention still has a high moisture content 2 hours after application, indicating that the essential oil composition prepared by the present invention has a good moisturizing effect. The samples of comparative examples 1 to 6, however, had lower hydration rates per time period than the samples of example 1 of the present invention. Therefore, after the essential oils are scientifically combined, the moisturizing effect is improved under the condition of consistent dosage, and the essential oils have good synergistic moisturizing and synergistic effects.
Test example 3 scavenging of free radicals test
1. Preparation of the test specimens
The products prepared in example 1 and comparative examples 1 to 6 were prepared into 1% ethanol solutions, and the following experiments were performed.
2. Evaluation of superoxide anion radical scavenging ability
4.5mL of Tris-HCl buffer (0.05 mol/L, pH 8.2) was preheated in a water bath at 25 ℃ for 20 min. Then adding 1mL of sample ethanol solution and 0.4mL of 25mmol/L pyrogallol solution, mixing uniformly, reacting for 5min in a water bath at 25 ℃, and adding 1.0mL of 8mol/L HCl to terminate the reaction. Absorbance values were measured at 299nm with Tris-HCl buffer as a reference. Blank control the sample was replaced with 1mL of solvent of the sample ethanol solution.
Superoxide anion radical scavenging rate (%) [ 1- (A2/A1) ] x 100%
Wherein A1 is the absorbance value of the blank control; a2 is the absorbance value of the sample.
3. Evaluation of hydroxyl radical scavenging ability
Sequentially adding 2mmol/L FeSO into a 25mL colorimetric tube4 3mL,1mmol/L H2O23mL, shaking up, then adding 3mL of salicylic acid with the concentration of 6mmol/L, shaking up, heating in a water bath at 37 ℃ for 15min, taking out, and measuring the absorbance; adding the solutions to be tested with certain concentrations, shaking, heating in water bath for 15min, and taking out to test their absorbance. The following formula is the clearance rate of hydroxyl radical (. OH) by the liquid to be tested:
hydroxyl radical clearance (%) ([ A3-A4- (A3-A5) ]/A1X 100%
Wherein A3 is the absorbance value of the reaction system before adding the sample; a4 is the absorbance value of the system after removing OH from the sample; a5 is the absorbance value of the system after blank control elimination of OH.
4. Comparative testing of samples against free radicals
TABLE 5 evaluation results of radical scavenging function
Figure GDA0002976139740000091
Experiments show that the scavenging rate of the essential oil composition in the embodiment 1 of the invention on superoxide anions is up to more than 85%, and the scavenging rate on hydroxyl radicals is about 85%, which shows that the composition has an obvious effect of scavenging free radicals, and the effect of scavenging free radicals is much better than that of single essential oil in the comparison ratios 1-5 and base oil in the comparison ratio 6, and shows that the compositions have a synergistic effect after scientific compounding.
Test example 4 skin irritation test
Experimental materials: the product of example 1 and comparative examples 1-6 was diluted 5 times with olive oil and used as a sample.
Subject: a total of 30 volunteers, of which women: 20 men and 10 men, the age is 20-40 years old.
The test method comprises the following steps: 0.03mL of the sample was taken by a syringe and placed in a chamber of a spot test tape, and the spot test tape with the sample was immediately stuck to the normal skin of the left forearm from the lower part in the longitudinal direction while gently pressing the chamber one by one to expel air and uniformly distribute the test substance. The test site is marked for easy observation. Every 24 hours, and half an hour after the patch was removed, the examination was performed according to the terms of the CTFA guidelines (1981). The patch test was performed 6 times in total, and on day 6 after completion of the skin application, additional examination was performed to observe delayed reactions. The data is compared using the last examination. The identification was carried out using the following table 6 as a standard:
