Description

Bionic sebum composition for skin barrier repair and preparation method and application thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a bionic sebum composition for skin barrier repair, and a preparation method and application thereof.
Background
The skin barrier structure is a brick wall structure and is composed of a sebum membrane, horny layer keratin, lipid, dermal mucopolysaccharide, mucopolysaccharide and the like. The skin can not only prevent the invasion of harmful factors such as physical, chemical, biological and the like from the outside; and can avoid the loss of water, lipid and other substances in the epidermis and the dermis of the human body in daily life, and play the role of the skin barrier function through the skin. Skin sensitivity such as dry skin, skin aging, pigmentation, solar dermatitis, etc., all belong to impaired skin barrier. After the skin barrier is damaged, the loss of substances such as water, lipid and the like can be further accelerated, the skin barrier is more easily damaged by external stimulation and environmental change, and red swelling, local redness or other pathological changes and the like can be caused in severe cases.
At present, a variety of skin barrier function repairing cosmetics are available, and inflammation of damaged skin is reduced by enhancing the moisture retention of the surface layer of the skin or adding soothing components. For example, trehalose, hyaluronic acid, chitosan and other components are added to supplement water and moisturize, so that the effect of repairing the damaged skin barrier is achieved. This approach does not fundamentally address the problem of impaired skin barrier function. The normal function of the skin can be recovered through the effects of various plant essential oils, but many essential oils are easily absorbed by the skin, but the essential oils cannot play a role in repairing barriers and reducing the loss of water and lipid substances, and have the risk of causing skin allergy.
Therefore, there is a need to provide a biomimetic sebum composition for skin barrier repair that can promote absorption, repair skin barrier, improve skin oxidation resistance, and reduce water and lipid loss.

Product effectiveness testing
1. Acute skin irritation test
34 SPF-grade white rabbits (divided into 17 groups) were selected as test animals (purchased from the center of laboratory animals of Sichuan university) and the back hairs of the test animals were cut off to a range of 3 cm × 3 cm. The bionic sebum compositions prepared in examples 1 to 7 and comparative examples 6 to 10 and 0.5g of each of the single components in comparative examples 1 to 5 were applied to the unhaired skin of the test animal, and fixed with a non-irritating adhesive tape for 2 hours. The test composition was removed from the skin of the test animal and the skin reaction was continuously observed at the site of contact of the test animal with the composition.
The test results show that after 72 hours, no erythema or redness appeared on the skin of each test animal, and it was preliminarily judged that under the test conditions, the biomimetic sebum compositions prepared in examples 1-7 and comparative examples 6-10, and the single components in comparative examples 1-5 were all non-irritating according to the skin irritation intensity classification.
2. Oxidation resistance test
(1) Determination of superoxide anion radical scavenging ability
The biomimetic sebum compositions of examples 1-5 and comparative examples 6-10 were taken and tested on the individual components of comparative examples 1-5 (i.e., the samples to be tested). The specific determination method is as follows:
0.05mol/L of Tris-HCl buffer (4.5 mL) with pH 8.2 was preheated in a water bath at 25 ℃ for 20 min. Then adding 1mL of sample to be detected and 0.4mL of 25mmol/L pyrogallol solution, mixing uniformly, reacting for 5min in a water bath at 25 ℃, and adding 1.0mL of 8mol/L hydrochloric acid to terminate the reaction. Absorbance at 299nm with Tris-HCl buffer as referenceValue A 2
Blank control experiment: only 1mL of ethanol was used in place of the test sample, and the other steps were not changed. Absorbance A at 299nm 1
The superoxide anion radical scavenging ratio was calculated as follows:
for superoxide anion radical clearance (%) - [1- (A) 2 /A 1 )]×100%
(2) Determination of the scavenging ability of hydroxyl radicals
The biomimetic sebum compositions of examples 1-5 and comparative examples 6-10 were taken and tested on the individual components of comparative examples 1-5 (i.e., the samples to be tested). The specific test method is as follows:
3mL of FeSO with the concentration of 2mmol/L are sequentially added into a 25mL colorimetric tube 4 3mL of H with a concentration of 1mmol/L 2 O 2 Shaking, adding 3mL of salicylic acid 6mmol/L, shaking, heating in 37 deg.C water bath for 15min, taking out, and measuring absorbance A 1
Adding samples to be tested with certain concentration, shaking, heating in water bath for 15min, taking out, and measuring absorbance A 2
Blank control experiment: only 1mL of ethanol is used for replacing the sample to be tested, and other steps do not change A 3
The clearance rate of the hydroxyl free radical is calculated according to the following formula:
(A) clearance of hydroxyl radical (%) - 3 -A 2 )/A 1 ×100%。
The test results are shown in table 2.
TABLE 2
Figure BDA0003596007890000111
Figure BDA0003596007890000121
As can be seen from table 2, compared with the comparative example, the biomimetic sebum composition provided by the invention has higher superoxide anion radical clearance rate and hydroxyl radical clearance rate, has excellent oxidation resistance, and can repair skin barrier and improve skin oxidation resistance.
3. Skin hydration test
The creams prepared in application examples 1 to 7 and comparative application examples 1 to 5 were subjected to a moisture content test of the horny layer of cheek skin, and the test instrument was a moisture content meter (Corneometer) of the horny layer of skin. The specific test method comprises the following steps: each group of face cream was tried by 35 healthy women (age 28-35 years). The cream is applied to cheek within 3 min after washing face every day without using other skin care products. Before and after the cream is used for four weeks, the moisture content of the horny layer of the cheek skin is tested by an instrument for 3 times in parallel, and the moisture content value of the horny layer is recorded by taking an average value.
Wherein, the change rate is relative to the change rate before use, and the calculation formula is as follows:
delta after four weeks is T 4 -T 0
Water content change rate of horny layer (T) 4 -T 0 )/T 0 ×100%;
In the formula, T 0 -stratum corneum moisture content value before application of cream to the test area;
T 4 the stratum corneum moisture content value after four weeks of application of the cream to the test area.
The higher the skin moisture content, the better the skin moisturizing properties and the better the skin barrier function. The comparative example is obviously different from the embodiment, and meanwhile, the skin barrier repairing composition has a good moisturizing effect. The test results are shown in Table 3.
TABLE 3
Figure BDA0003596007890000122
Figure BDA0003596007890000131
As can be seen from table 3, compared with the comparative application example, the cream provided by the application example of the present invention can effectively repair the skin barrier, and has excellent moisturizing and moisturizing effects. The cream of application example 7 is the best in effect, because the components in application example 7 are the most, and squalane, lecithin, cholesterol, ceramide, hydrogenated polyisobutene, vitamin E and meadowfoam seed oil are also contained on the basis of application example 1. It is more similar in composition to sebum of the skin. However, as is clear from application example 1, the cream in application example 1 does not contain these components, but the rate of change in moisture content of the stratum corneum after use is not so great as in application example 7, and therefore, in the present invention, the grass-cotton seed oil, the glass-chicory seed oil, the evening primrose oil, the tea seed oil, and the pepper fruit oil play a more important role in repairing the skin barrier and improving the skin moisture retention. The cream of comparative application example 5 is the worst in effect, and the main reason is that the cream does not contain pepper fruit oil, has weak penetration effect, and the rest components cannot well penetrate into the skin, so that the repair effect is weak.
